RÉSUMÉ

Human schistosomiasis is a parasitic disease caused by an infection with trematodes of the genus Schistosoma. The disease mainly affects impoverished populations. Around 800 million people are exposed to the infection, which is a public health problem in the tropical and subtropical regions of Africa, Asia, the Caribbean and South America. In Brazil, Schistosoma mansoni is the only species that causes schistosomiasis and the disease is widely distributed. Conventional diagnosis of the disease is carried out by detecting eggs using parasitological methods, such as the Kato-Katz test. Schistosomiasis has been reported in all regions of Brazil and is characterized as endemic in seven states in the Northeast Region and two states in the Southeast Region. In 2015, 78,7% of all cases reported in Brazil occurred in the Northeast Region. It is estimated that 1,5 million people is infected with this disease in Brazil and more than 25 millions live in areas with a high risk of transmission. Despite the reduction in mortality and morbidity, schistosomiasis was responsible for 8,756 deaths between 2000 and 2011 and 2,517 deaths between 2015 and 2019 in Brazil and it remains an important public health problem. In the state of Rio de Janeiro, some areas have low endemicity or isolated foci of Schistosoma mansoni and the majority of infected individuals have mild infections. The last survey of the disease in the state of Rio de Janeiro was carried out between 2010 and 2015 in students aged 7 to 17.Schistosomiasis was reported in 10 of the 21 municipalities studied. Of the 5,111 school children screened for S. mansoni infection, 46 (1,65%) were tested positive. Studies carried out in areas of low endemicity in Rio de Janeiro showed that among the 205 patients infected by S. mansoni in Sumidouro, around 84% were aged 14 or over and all, except one individual, had the intestinal form (91,2%) or hepato-intestinal (8,3%) of schistosomiasis. Another study carried out in Sumidouro showed that with tests based on patent Schistosoma egg infection determined by the Kato-Katz test, active infections were diagnosed in eight (8/108) individuals. The intensity of infection expressed by parasite loads ranged from 6 to 72 eggs per gram of feces/individual. The results showed DNA amplification in 32 of the 100 individuals tested by real-time PCR. All individuals with patent ovo infection showed positive DNA amplification. These studies showed that if we only analyzed school-age children using the Kato-Katz test, the majority of the infected population would never be diagnosed with S. mansoni infection. In situations of low endemicity, with low intensities of infection, with low severity in the population and in the most affected age groups, schistosomiasis requires a more sensitive diagnostic approach (e.g. screening by PCR rather than Kato test), otherwise many infected individuals will remain invisible to the healthcare system.

A doença afeta principalmente populaçoes empobrecidas.

Multiple microbial detections in stool samples of indigenous individuals suffering from chronic gastroenteric disorder of a likely infectious origin, characterized by recurring diarrhea of variable intensity, in the rural north-east of Colombia are common findings, making the assignment of etiological relevance to individual pathogens challenging. In a population of 773 indigenous people from either the tribe Wiwa or Kogui, collider bias analysis was conducted comprising 32 assessed microorganisms including 10 bacteria (Aeromonas spp., Campylobacter spp., enteroaggregative Escherichia coli (EAEC), enteropathogenic Escherichia coli (EPEC), enterotoxigenic Escherichia coli (ETEC), Salmonella spp., Shiga toxin-producing Escherichia coli (STEC), Shigella spp./enteroinvasive Escherichia coli (EIEC), Tropheryma whipplei and Yersinia spp.), 11 protozoa (Blastocystis spp., Cryptosporidium spp., Cyclospora spp., Dientamoeba fragilis, Entamoeba coli, Entamoeba bangladeshi/dispar/histolytica/moshkovskii complex, Entamoeba histolytica, Endolimax nana, Giardia duodenalis, Iodamoeba buetschlii and Pentatrichomonas hominis), 8 helminths (Ascaris spp., Enterobius vermicularis, Hymenolepis spp., Necator americanus, Schistosoma spp., Strongyloides spp., Taenia spp. and Trichuris spp.), microsporidia (Encephalocytozoon spp.) and fungal elements (microscopically observed conidia and pseudoconidia). The main results indicated that negative associations potentially pointing towards collider bias were infrequent events (n = 14), while positive associations indicating increased likelihood of co-occurrence of microorganisms quantitatively dominated (n = 88). Microorganisms showing the most frequent negative associations were EPEC (n = 6) and Blastocystis spp. (n = 3), while positive associations were most common for Trichuris spp. (n = 16), Dientamoeba fragilis (n = 15), Shigella spp./EIEC (n = 12), Ascaris spp. (n = 11) and Blastocystis spp. (n = 10). Of note, positive associations quantitively dominated for Blastocystis spp. In conclusion, collider bias assessment did not allow clear-cut assignment of etiological relevance for detected enteric microorganisms within the assessed Colombian indigenous population. Instead, the results suggested complex microbial interactions with potential summative effects. Future studies applying alternative biostatistical approaches should be considered to further delineate respective interactions.

RÉSUMÉ

BACKGROUND: Control interventions recommended by the World Health Organization have successfully resulted in low-intensity schistosomiasis transmission areas. To achieve elimination of transmission, new diagnostic screening tools are needed to overcome less than adequate sensitivity of the currently used Kato-Katz faecal thick smear method. Ideally, in-house serological tests should be avoided due to not having a continuous supply of kits as would be necessary for large population studies. Quality assurance provided by manufacturers and proper performance evaluations are also needed. We evaluated the accuracy of two commercially available serology tests as screening methods for detecting light schistosomiasis infections. METHODS: Serum samples were collected in 2015 from individuals living in a low-endemicity locality in northeastern Brazil and deposited in a biorepository. We evaluated immunoglobulin G (IgG) and IgM enzyme-linked immunosorbent assays (ELISAs) and an immunochromatographic test (ICT). The Helmintex method was used to define true-positive samples. RESULTS: Overall sensitivity was close to 90% for both the IgG ELISA and ICT, yet specificity was 28% and 18%, respectively. For the IgM ELISA, the values were estimated to be 55% and 43%, respectively. CONCLUSIONS: Poor specificity and positive predictive values prevent these tests from being recommended for screening populations in low-intensity schistosomiasis-endemic areas.

RÉSUMÉ

The river basins of Brazil contain a highly diverse ichthyofauna of remarkable endemism, including several threatened species. Accordingly, Lignobrycon myersi is a fish species distributed only in a few rivers from the state of Bahia, northeastern Brazil. Since this species is classified as Near Threatened and is poorly studied, efforts to understand the genetic structure of populations and putative cryptic forms should help define efficient strategies of management and conservation. Herein, the molecular identification and the population genetic diversity of specimens of L. myersi across their range (Almada, Contas, and Cachoeira river basins) were assessed using mitochondrial markers (16S rDNA and D-Loop, respectively). The inferences based on phylogenetics, genetic distance, and species delimitation methods invariably identified all samples as L. myersi. In addition, sequencing of D-loop fragments revealed significant haplotype diversity and a considerable level of population genetic structure. Despite their geographic isolation, these data suggested that populations from Almada and Contas rivers represent a single evolutionary lineage that could be managed as a whole. In contrast, the population from Cachoeira River was highly differentiated from the others and should be managed separately as a unique and endemic unit, particularly focused on the conservation of native habitats.

RÉSUMÉ

Previous studies about the genetic diversity, connectivity and demographic history in Lutjanidae fishes have reported a common pattern of genetic homogeneity and expansion in populations from Western South Atlantic. In the present work, we inferred the population structure, the levels of genetic diversity and the demographic history of the Brazilian snapper Lutjanus alexandrei, a recently described and endemic species from Northeastern coast of Brazil. Five different fragments, including mitochondrial DNA (Control Region, Cyt b and ND4) and nuclear DNA (Myostatin and S7) regions were analyzed in 120 specimens of L. alexandrei from four localities in Northeastern Brazil, representing the first study of population genetics in this species. High levels of genetic diversity were observed following a panmictic pattern, probably related to the larval dispersal by the current tides along the Brazilian coast. In addition, both demographic history and neutrality tests indicated that L. alexandrei has undergone population expansion during Pleistocene. In this sense, the sea level variation from this period could have increased the available resources and suitable habitats for the Brazilian snapper.

RÉSUMÉ

Objective: to evaluate the seasonal and endemic characteristics of conjunctivitis at the ophthalmology service of the Leiria de Andrade Foundation (FLA) in the last ten years to trace the epidemiological profile of conjunctivitis in Fortaleza - CE. Methods: this was a descriptive and epidemiological study based on quantitative and qualitative analysis, retrospectively, from January to December 2012 to 2019, with a projection for the years 2020 and 2021. Results: 107,778 medical records were analysed, with endemic and seasonal fluctuation, being the months of October to December more frequent in the intervals of the highest incidence of the disease. Two peaks were notorious, one with epidemic characteristics, from July 2013 to November 2014, and the other with outbreak characteristics, probably due to a national-level epidemic, between January and April 2018. The most affected age group was between 19 and 59 years, covering about 72% of cases, with no statistical difference between genders. Conclusion: according to the study data, it was possible to infer that the epidemiological scenario of Fortaleza - CE is in line with the literature regarding age range and sex. The endemicity of the disease reinforces its relevance in the scenario of the Unified Health System (SUS) of the region.

BACKGROUND: Pseudalcantarea grandis (Schltdl.) Pinzón & Barfuss is a tank bromeliad that grows on cliffs in the southernmost portion of the Chihuahuan desert. Phytotelmata are water bodies formed by plants that function as micro-ecosystems where bacteria, algae, protists, insects, fungi, and some vertebrates can develop. We hypothesized that the bacterial diversity contained in the phytotelma formed in a bromeliad from an arid zone would differ in sites with and without surrounding vegetation. Our study aimed to characterize the bacterial composition and putative metabolic functions in P. grandis phytotelmata collected in vegetated and non-vegetated sites. METHODS: Water from 10 individuals was sampled. Five individuals had abundant surrounding vegetation, and five had little or no vegetation. We extracted DNA and amplified seven hypervariable regions of the 16S gene (V2, V4, V8, V3-6, 7-9). Metabarcoding sequencing was performed on the Ion Torrent PGM platform. Taxonomic identity was assigned by the binning reads and coverage between hit and query from the reference database of at least 90%. Putative metabolic functions of the bacterial families were assigned mainly using the FAPROTAX database. The dominance patterns in each site were visualized with rank/abundance curves using the number of Operational Taxonomic Units (OTUs) per family. A percentage similarity analysis (SIMPER) was used to estimate dissimilarity between the sites. Relationships among bacterial families (identified by the dominance analysis and SIMPER), sites, and their respective putative functions were analyzed with shade plots. RESULTS: A total of 1.5 million useful bacterial sequences were obtained. Sequences were clustered into OTUs, and taxonomic assignment was conducted using BLAST in the Greengenes databases. Bacterial diversity was 23 phyla, 52 classes, 98 orders, 218 families, and 297 genera. Proteobacteria (37%), Actinobacteria (19%), and Firmicutes (15%) comprised the highest percentage (71%). There was a 68.3% similarity between the two sites at family level, with 149 families shared. Aerobic chemoheterotrophy and fermentation were the main metabolic functions in both sites, followed by ureolysis, nitrate reduction, aromatic compound degradation, and nitrogen fixation. The dominant bacteria shared most of the metabolic functions between sites. Some functions were recorded for one site only and were related to families with the lowest OTUs richness. Bacterial diversity in the P. grandis tanks included dominant phyla and families present at low percentage that could be considered part of a rare biosphere. A rare biosphere can form genetic reservoirs, the local abundance of which depends on external abiotic and biotic factors, while their interactions could favor micro-ecosystem resilience and resistance.

RÉSUMÉ

This study aimed to evaluate the performance of the point-of-care circulating cathodic antigen (POC-CCA) test in a highly endemic area in Brazil, comparing it to the Kato-Katz (KK) technique for sensitivity, specificity and the intensity of the reaction of the test in relation to the parasitic load. The community in Sergipe, Brazil, participated in the study, providing three stool samples, one of urine (POC-CCA) and fingers tick blood sample was tested by enzyme-linked immunosorbent assay (ELISA). Sensitivity, specificity, positive predictive value, negative predictive value, accuracy, kappa coefficient and Spearman's correlation were calculated for the POC-CCA test using the KK as the reference. The prevalence of schistosomiasis by KK testing was 48.82%; POC-CCA (t+) 66.14%; POC-CCA (t-) 45.24%. ELISA results showed 100% agreement in individuals with high and moderate eggs per gram (EPG). POC-CCA presented good diagnostic performance in individuals with medium and high EPG, but there were a high number of false negatives in individuals with low intensity infections. As observed, POC-CCA-filter test improves accuracy and sensitivity compared to a conventional test.

Neotropical páramos as biogeographic units. Introduction: Páramo has been defined from various points of view, which take into account different factors that are easy to recognize or measure, nevertheless at the biogeographic level it has been evaluated with criticized methods used to identify historical units. The analysis of endemicity, despite its importance and wide recognition, has not been used as a tool to evaluate Páramo. Objective: Determine whether the neotropical Páramo is one or several biogeographic units. Methods: We included distributional records from Aves, Amphibia, Mammalia, Reptilia, Marchantiophyta, and Spermatophyta. We found 7 025 species with 193 250 suitable occurrences obtained from the GBIF. We used each taxonomic group as an independent partition or as a component of a larger partition, such as total plants (Plants-T: Marchantiophyta + Spermatophyta), or total animals (Animals-T: Aves + Amphibia + Mammalia + Reptilia), or total evidence (Plants-T + Animals-T). In order to identify areas of endemism, we used the optimality criterion (NDM/VNDM) with grids of 0.5° or 0.25°. We calculated the intersection among polygons of previous definitions and the areas recovered in our analyses. Results: Both grid sizes, 0.25° and 0.5°, identified areas of endemism in different sectors along the Andean and Central American cordilleras, but only the 0.25° size allowed us to recognize areas/sectors with a higher resolution. We recovered eight areas, which were considered as subprovinces (Santa Marta-Perijá, Mérida, Santanderes-Boyacá, Cundinamarca, Central-Western Cordillera, Northern Ecuador, Central-South Ecuador, and Talamanca). These areas were between 4 and 66 % consistent with previous definitions. Conclusions: Páramo has been considered a single biogeographic unit, however, given our analyses we identified it as a unit composed of eight biogeographic subprovinces, which is consistent with some published studies.

Introduction: Toxoplasmosis acquired by women of childbearing age is a problem still not understood by the community and decision makers from health systems. Few consistent data are available in Ecuador about the circulation of this parasitic infection. Therefore, it is of great interest to broaden knowledge about the variables leading to its acquisition. Objective: Determine the seroprevalence of toxoplasmosis among young women and its relationship to epidemiological variables. Methods: A study was conducted of 403 serum samples from students attending the School of Health Sciences of the National University of Chimborazo. Serological behavior of Toxoplasma gondii was evaluated by determination of anti-Toxoplasma gondii IgG using an enzyme immunoassay. Identification was made of its relationship to eating habits, keeping animals at home, and knowledge about the risk of being seronegative when deciding to have children. Results: Of the total sample, 26.8 percent (108/403) were seropositive for Toxoplasma gondii. The oral route was the most significant mode of acquisition of the disease. A relationship was found between seroprevalence and keeping yard animals and pets. 73.9 percent (298/403) of the respondents were not aware that seronegative results during pregnancy constitute a potential risk. Conclusions: Anti-Toxoplasma gondii antibodies were found in blood of the samples studied, pointing to the existence of previous Toxoplasma gondii infections and the influence of the variables analyzed(AU)

RÉSUMÉ

Melioidosis has been detected in the Caribbean, and an increasing number of cases has been reported in the past few decades, but only 2 cases were reported in Guadeloupe during the past 20 years. We describe 3 more cases that occurred during 2016-2017 and examine arguments for increasing endemicity.

RÉSUMÉ

Abstract INTRODUCTION: Brazil's southernmost state, Rio Grande do Sul (RGS), was considered schistosomiasis-free until 1998 when a low endemic focus was identified in Esteio, a city located next to the capital of RGS. In the last two decades, the control interventions applied in the region have been apparently successful, and the absence of new cases indicated the possibility of interrupted schistosomiasis transmission. The objective of this study was to update the clinical and epidemiological data of schistosomiasis in Esteio. METHODS: We reviewed all 28 individuals diagnosed with the infection since 1997 and a survey was applied to a group of 29 school-aged children residing in Vila Pedreira, one of the most affected neighborhoods. RESULTS No eggs were detected in fecal samples using the Helmintex method, and all samples were negative for serum antibodies on examination by the western blot technique using the Schistosoma mansoni microsomal antigen (MAMA- WB). In contrast, 23 individuals (79%) tested positive for the cathodic circulating antigen with the point-of-care immunochromatographic test (POC-CCA) on urine samples. Of the 28 formerly infected individuals, only eight were located, of which four tested positive, and four tested negative for serum antibodies using the MAMA-WB technique. CONCLUSIONS: Current adverse conditions for S. mansoni transmission in Esteio and the absence of a confirmed diagnosis suggests that there is (i) a lack of specificity of the POC-CCA test in low endemic settings, and (ii) a high probability that interruption of schistosomiasis has been achieved in Esteio.

RÉSUMÉ

Our goals were to determine whether the pattern of geographical distribution of dung beetles in the Brazilian Atlantic Forest (BAF), Southeast Brazil, supports the identification of the areas of endemism in this biome, how the number and boundaries of the areas of endemism vary with the spatial scale used in the analysis, if the areas of endemism identified here are consistent with previous proposals described in the literature. We used the analysis of endemicity (NDM) on the distributional patterns of 198 species and subspecies of dung beetles in order to identify areas of endemism into grids of 0.25°x0.25°, 0.5°x0.5° and 1°x1°. A total of 15 consensus areas (CA) were identified with the different grid sizes employed (two of 0.25°, three of 0.5° and 10 of 1°). Our results support the assumptions about the influence of the grid size, since a smaller number of CA were identified for the 0.25° grids (02 CA), whereas by increasing the size of the units, there was an increase in the number of the areas of endemism (10 CA of 1°). Despite different levels of geographical congruence, the areas of endemism located in the BAF described elsewhere are corroborate, all or part of them. The overlap of these CA allowed the definition of three generalized areas of endemism: Southeast, Central and Southwest. These generalized areas encompassed habitats with great environmental variety, potentially influencing the levels of endemism of species of dung beetles. Furthermore, they were corroborated by other areas of endemism described in the literature, allowing us to argue that processes forming these regions have influenced dung beetle species as described for other groups of organisms.(AU)

Knowledge on the hydrological evolution of the Neotropical region was used along with the current distribution of the anablepid species to investigate the historical biogeography of this family. Areas of endemism were delimited by endemicity analysis resulting in seven individual areas of endemism and three consensus areas located in northwestern Argentina, southern Brazil, and northern South America. These areas were discussed in the context of anablepid species diversification, especially for the genus Anableps and the subgenera Jenynsia and Plesiojenynsia. The existence of areas of endemism for the family analyzed reveals an intimate association with historical events that occurred in the geological evolution of South America, which can be associated with the main diversification patterns and historical hypotheses in the context of Neotropical freshwater fishes biogeography.

RÉSUMÉ

We reviewed Giemsa-stained thick blood smears, obtained through the national malaria surveillance program in the Amazon region of Ecuador, by light microscopy for Mansonella spp. microfilariae. Of 2,756 slides examined, 566 (20.5%) were positive. Nested PCR confirmed that the microfilariae were those of M. ozzardi nematodes, indicating that this parasite is endemic to this region.

RÉSUMÉ

Information on the biogeography of arbuscular mycorrhizal fungi (AMF) is important because this group of obligately symbiotic soil microbes is a ubiquitous and functionally critical component of terrestrial ecosystems. In this paper, we utilize a biogeography database summarizing data on AMF species distribution linked to geographic and environmental conditions to describe global distribution patterns and interpret these patterns within a phylogenetic perspective. The data were obtained from accessions in living culture collections (INVAM, CICG), species descriptions, and other published literature from 1960 to 2012. The database contains 7105 records, 6396 of them from 768 published papers and the remaining 709 from culture accessions. Glomeromycotan species were recorded in all seven continents, 87 countries, 11 biogeographical realms, and 14 biomes. The distribution of families differed among climatic zones and continents, but they, together with all genera, appear to be cosmopolitan. Distribution of AMF species shows a slight decrease from low to high latitudes, but this decrease is steeper in the southern than in the northern hemisphere. A total of 189 species is shared between ancient supercontinents Gondwana and Laurasia and 78 species are common to all climatic zones. Ninety-five species (43% of the total) have known cosmopolitan distribution, including members of all genera except Redeckera. Some species have disjunct distribution and 26% of species have been registered from only one continent. Data on AMF distribution challenge the "Everything is everywhere" hypothesis in favor of the "moderate endemicity model" for species distribution. Data from this study provide a foundation to formulate and test hypotheses of biogeographic patterns and processes in Glomeromycota.

Results: Of the study population, 38 students (36.0 percent) were found to be positive for antibodies to T. gondii, indicating contact with the parasite at some point in their life. The demonstrated seropositivity may be due to the lack of knowledge about the main routes of infection, the risk of being seronegative during pregnancy and the high endemicity of the parasite. Conclusions : The seroprevalence of T. gondii infection in the studied population were demonstrated(AU)
