ABSTRACT

1. This study evaluated the minimal inhibitory concentration (MIC) of ciprofloxacin and the presence of plasmid-mediated quinolone resistance (PMQR) mechanisms in 97 nontyphoidal Salmonella spp. isolated from broilers and carcases from three different regions in Brazil. The presence of mutations in quinolone resistance determination regions (QRDRS) was investigated in the ciprofloxacin-resistant strain by DNA sequencing.2. Most of the Salmonella spp. (85.6%) had intermediate resistance to ciprofloxacin and only one isolate was resistant. MIC breakpoints ranged from ≤0.03 to 1 µg/ml and 67.0% of the strains had a MIC of 0.25 µg/ml (n=65). Thirteen strains (13.4%) were susceptible to ciprofloxacin with MIC ≤0.06 µg/ml. The qnrB gene was detected in eight isolates with intermediate resistance and in two susceptible strains. The other PMQR genes, qnrA, qnrC, qnrD, qnrS, qnrVC, aac(6')-Ib, qepA, oqxAB and mutations in QRDR were not detected in any strain.3. There was a high frequency of ciprofloxacin intermediate resistant Salmonella from broiler and broiler carcases from Brazil. The presence of these strains in poultry and derived products poses a risk to public health.

ABSTRACT

Enteropatogenic Escherichia coli (EPEC) and shigatoxigenic E. coli (STEC), are generally poultry and poultry product isolate and can cause serious human infections. Many strains may become resistant to various antimicrobials, which can hinder the treatment of bacterial diseases. Organic farming seeks to avoid the selection and frequency of antimicrobial-resistant bacteria. This study aims to verify the resistance of EPEC and STEC from organic and conventional (industrial) broiler isolates to antimicrobials. All isolates were submitted to disk diffusion test with tetracycline, gentamicin, enrofloxacin, ceftriaxone and amoxicillin + clavulanate (TET, GEN, ENO, CTX, AMC) and PCR to detect specific virulence genes for EPEC and STEC. A total of 297 E. coli strains were isolated, 213 from conventional. In organic broiler, 84 strains were isolated. The strains from the conventional broiler isolates were resistant to five antimicrobials tested: TET 48.82% (104/213), ENO 28.17% (60/213), CTX 15.49% (33/213), GEN 14.55% (31/213), and AMC 7.04% (15/213), and 9.86% (21/213) were considered multidrug-resistant. Organic chicken strains were resistant to four of the antimicrobials tested: TET 35.7% (30/84), ENO 9.5% (8/84), CTX 2.4% (2/84), GEN 4.8% (4/84). Of the strains from the organic broiler chicken isolates, only 1.2% (1/84) was considered multidrug-resistant. No EPEC and STEC were found in the organic chicken samples. The multidrug resistance was characterized in 9.52% (2/21) of the EPEC and 4.76% (1/21) of the STEC. The study demonstrated the absence of EPEC and STEC strains in organic broilers and carcasses and a lower frequency of multiresistant strains compared to conventional breeding.(AU)

Abstract This study aimed to analyze the socio-environmental characteristics, executive and nutritional functions in children aged 6 to 7 years, from public schools in Alagoas, Brazil. A quantitative, cross-sectional and descriptive study was performed involving 64 children of the mentioned age group, from public schools located in Alagoas: Maceió, on the coast; Major Isidoro, in the hinterland and Palmeira dos Índios, in the country. Such analyzes were made through the application of neuropsychological tests and anthropometric assessment with children and food and socio-environmental surveys with their parents. As for the type of housing, 100% were made of masonry, with a bathroom present in 98.4%. All children reported with the habit of bathing in the river / lagoon, presented some pathology. There was no significant difference between cities in terms of the sociodemographic characteristics evaluated, with the same result occurring with the factors associated with the occurrence of diseases in children. The subtests of WISC-IV, were below the average in all municipalities, and the TAC and SCC were classified within the average. However, even though the ranking were divided between below average and average, it is possible to identify from the subtests of WISC-IV, that the general IQ showed a cognitive level below the average. There was also no significant difference in the anthropometric assessment (weight, height, BMI and IMCI) between the evaluated students. The average weight was 23.3 kg to 25 kg, the height between 1.23 m to 1.24 m, the BMI between 16.4 to 17; the IMCI from 2.8 to 3.0. Children were classified within the average. Regarding micronutrients (Ca, Fe, K, Mg and Na, and vitamins A, C, D, B1, B9 and B12) and calories, there was also no significant difference between the cities evaluated. The same occurred with macronutrients (proteins, carbohydrates and lipids). This study showed that in general there was no difference between the students of the three municipalities. Probably, even though they are all public schools and from different cities, children have similar social conditions.

ABSTRACT

P34 is an antimicrobial peptide produced by Bacillus sp. P34, isolated from the intestinal contents of a fish from the Amazon basin. This peptide showed antibacterial properties against Gram-positive and Gram-negative bacteria and was characterized as a bacteriocin like substance. It was demonstrated that the peptide P34 exhibited antiviral activity against feline herpesvirus type 1 in vitro. The aim of this work was to evaluate P34 for its antiviral properties in vitro, using RK 13 cells, against the equine arteritis virus, since it has no specific treatment and a variable proportion of stallions may become persistently infected. The results obtained show that P34 exerts antiviral and virucidal activities against equine arteritis virus, probably in the viral envelope. The antiviral assays performed showed that P34 reduces significantly the viral titers of treated cell cultures. The mechanism of action of P34 seems to be time/temperature-dependent. This peptide tends to be a promising antiviral compound for the prevention and treatment of arteriviral infections since it has a high therapeutic index. However, more detailed studies must be performed to address the exact step of viral infection where P34 acts, in order to use this peptide as an antiviral drug in vivo in the future.(AU)


P34 é um peptídeo antimicrobiano produzido pelo Bacillus sp. P34, isolado do conteúdo intestinal de um peixe na Bacia Amazônica. Esse peptídeo demonstrou propriedades antibacterianas contra bactérias Gram-positivas e Gram-negativas e foi caracterizado como uma substância do tipo bacteriocina. Foi demonstrado que o peptídeo P34 exibiu atividade antiviral contra o herpesvírus felino tipo 1 in vitro. O objetivo deste trabalho foi avaliar o P34 in vitro, em cultivo de células RK 13, contra o vírus da arterite equina, uma vez que não há tratamento específico e uma variável proporção de garanhões pode permanecer persistentemente infectada. Os resultados obtidos mostram que o peptídeo exerce atividade antiviral e virucida contra o vírus da arterite equina, agindo provavelmente no envelope viral. Os ensaios antivirais realizados demonstraram que o P34 reduz significativamente os títulos do vírus nas células infectadas e tratadas. O mecanismo de ação do P34 parece ser tempo/temperatura dependente. Esse peptídeo tende a ser um antiviral promissor para o tratamento e a prevenção das infecções por arterivírus, tendo em vista que ele possui um alto índice terapêutico. Contudo, estudos mais detalhados devem ser realizados para precisar a etapa exata da infecção viral em que o P34 age, para que ele possa ser usado como antiviral in vivo no futuro.(AU)

ABSTRACT

Este estudo objetivou realizar atividades de extensão em fitoterapia, a partir de um levantamento de dados sobre as plantas medicinais, fitoterápicos e medicamentos convencionais utilizados por 292 idosos frequentadores de um programa de Atividades Físicas e Recreativas para a Terceira Idade (AFRID), na cidade de Uberlândia-MG, utilizando como instrumento de investigação, um questionário semiestruturado. Dentre os entrevistados verificamos que 88% utilizavam medicamentos prescritos, principalmente para o controle da hipertensão. O uso de plantas medicinais foi relatado por 76,7% dos idosos, sendo as mais citadas: Cymbopogon citratus, Mentha sp., Rosmarinus officinalis, Plectranthus barbatus, Ocimum gratissimum, e Matricaria chamomilla. Dezesseis (5,5%) idosos utilizavam fitoterápicos, principalmente preparados a partir de extratos de Ginkgo biloba, Aesculus hippocastanum e Passiflora incarnata em associação com Crataegus oxyacantha e Salix alba. O uso concomitante de plantas medicinais e fitoterápicos com medicamentos convencionais foi relatado por 86,2% e 81,3% dos idosos, respectivamente. Após a análise dos dados percebemos a necessidade do desenvolvimento de ações educativas para informar e conscientizar os idosos sobre o uso da fitoterapia. Elaboramos uma caderneta e uma cartilha para promoção da difusão dessas informações e o aprimoramento do uso da fitoterapia entre os idosos e, dessa forma, alcançar os profissionais de saúde sobre os riscos e benefícios dessa terapêutica; contribuindo assim para o uso seguro e racional da fitoterapia.


This study aimed to carry out extension activities in herbal medicine from a survey of data on medicinal plants, herbal and conventional medicines used by 292 elderly people who attended a program of physical activity called Physical and Recreational Activities for the Elderly, in Uberlândia-MG, using a semi-structured questionnaire as means of investigation. Among the respondents 88% used prescription drugs, primarily for control of hypertension. The use of medicinal plants was reported by 76.7% of the elderly, being the most cited ones: Cymbopogon citratus, Mentha sp., Rosmarinus officinalis, Plectranthus barbatus, Ocimum gratissimum. and Matricaria chamomilla. Sixteen respondents (5,5%) used herbal medicines, especially those prepared from extracts of Ginkgo biloba, Aesculus hippocastanum, and Passiflora incarnata L. in association with Crataegus oxyacantha L. and Salix alba. The concomitant use of medicinal plants and herbal medicines with conventional drugs was reported by 86.2% and 81.3% of participants, respectively. After analyzing the data, we detected the need to develop educational activities to inform and educate seniors about the use of herbal medicine, encompassing the development a book and a primer for initial dissemination of this information, improving herbal medicine use among the elderly. This course of action would allow a greater knowledge of health professionals about the risks and benefits of this therapy, thereby contributing to the safe and rational use of herbal medicine.

ABSTRACT

The most effective method to prevent yellow fever and control the disease is a vaccine made with attenuated live virus. Due to the neurological tropism of the virus, preventive vaccination is not recommended for infants under 6 months and for pregnant women. However there is a paucity of data regarding the safety for pregnant women and there are no experimental studies investigating adverse effects to the offspring after maternal exposure to the vaccine. This study aimed to investigate, in mice, the effects of maternal exposure to the yellow fever vaccine at three different gestational ages on the physical and behavioral development of the offspring. Pregnant Swiss mice received a single subcutaneous injection of water for injection (control groups) or 2 log Plaque Forming Units (vaccine-treated groups) of the yellow fever vaccine on gestational days (GD) 5, 10 or 15. Neither maternal signs of toxicity nor alterations in physical development and reflex ontogeny of the offspring were observed in any of the groups. Data from behavioral evaluation indicated that yellow fever vaccine exposure induced motor hypoactivity in 22-day-old females independent of the day of exposure; and in 60-day-old male and female pups exposed at GD 10. Moreover, 22-day-old females also presented with a deficit in habituation memory. Altogether, these results indicate that in utero exposure to the yellow fever vaccine may induce behavioral alterations in the pups that may persist to adulthood in the absence of observed maternal toxicity or disruption of physical development milestones or reflex ontogeny.

ABSTRACT

O patchouli possui óleo essencial nas folhas com utilização principalmente na indústria de perfumaria. O objetivo foi avaliar o melhor tempo de extração de óleo essencial de folhas secas de patchouli. Os tratamentos foram 1, 2, 3, 4, 5, 6, 7 e 8 horas de extração, através do método de hidrodestilação, com aparelho graduado do tipo Clevenger e balões com capacidade de 2 L.O delineamento foi inteiramente casualizado com três repetições. O material destilado foi seco à sombra até atingir aproximadamente 20% de umidade. Para cada tratamento, foram utilizadas amostras de 50 g de massa seca foliar. Foram avaliados o rendimento e a composição do óleo essencial. Não houve diferença entre os diferentes tempos de extração no rendimento de óleo essencial, podendo a extração do óleo essencial de patchouli ser realizada com uma hora de extração. O tempo de extração aumenta as porcentagens relativas do beta-guaieno (0,81%), beta-patchouleno (1,26%), alfa-selineno (1,37%), cariofileno (2,44%), alfa-patchouleno (3,08%) e gama-patchouleno (4,82%). O teor de pogostol (5,11%) reduz com o aumento do tempo de extração. O patchoulol, alfa-guaieno, alfa-bulneseno e seicheleno não sofrem influencia do tempo de extração.


Patchouli accumulatea an essential oil on its leaves, and it is mainly used by the perfume industry. The purpose of this work was to determine the distillation time for the extraction of essential oils from the leaves of patchouli. The treatments included 1, 2, 3, 4, 5, 6, 7 and 8 hours of extraction through the hydrodistillation method, using a Clevenger apparatus. The experimental design was completely randomized with three replications. The leaves were dried at room temperature until they reached 20% of humidity. The essential oil yield was measured from samples containing 50g of leaf dry mass. The essential oil production and composition were evaluated. No differences among the treatments were found on the essential oil yield, suggesting that the essential oil extraction can be carried out for 1 hour according to the experimental conditions. However, extending the extraction time, an increase in the relative percentage of beta-patchoulene (1.26%), cariofilene (2.44%), gamma-patchoulene (4.82%), alpha-patchoulene (3.08%), beta-guaiene (0.81%) and alpha-selinene (1.37%) was observed. The pogostol (5.11%) content was reduced with the increase of the extraction time. Patchoulol, alpha-guaieno, alpha-bulneseno and seicheleno contents were not affected by the evaluated extraction times.

The aim of this investigation was to describe the occurrence of bradycardia during the early postoperative period of liver transplantation in children. We retrospectively analyzed a cohort of 79 children with end-stage liver diseases who underwent liver transplantation. All children experienced >or=1 episode of a cardiac rate below the 2nd percentile of a 1-hour minimum duration, which was considered to be bradycardia. Patients <24 months were compared with older ones. The overall incidence of bradycardia was 37% (n = 31), including 25 patients who displayed bradycardia until postoperative day 3. In all cases, the electrocardiogram was normal, showing sinus rhythm. A comparison of the groups demonstrated an increased incidence of bradycardia among patients <24 months of age (P=.03). In all patients, there were no hemodynamic consequences; the cardiac rate returned to normal uneventfully. The explanations for bradycardia could not be applied to these patients because none of them had any volume change or electrolyte disturbances; liver function tests were not seriously altered. The mechanisms of this postoperative complications are unclear.

Amostras de Salmonella isoladas de esgotos do Rio de Janeiro foram examinadas para se verificar a presenca de especimes resistentes a drogas. Em contraste com um levantamento realizado em 1970, quando nao foram encontradas amostras resistentes, o presente trabalho revelou a ocorrencia de 31% (39/126) de amostras resistentes a pelo menos uma droga. Todas as linhagens multiresistentes foram capazes de transferir os marcadores respectivos para receptoras adequadas, Os isolados apresentaram o serotipo S. tryphimurium, con excecao de duas amostras, caracterizadas como S. agona. Duas dentre as 39 amostras resistentes e duas dentre 87 amostras sensiveis as drogas usadas eram colicinogenicas. Noventa por cento (29/32) dos determinantes transferiveis eram do tipo Fi
