ABSTRACT

Introduction: Emotion dysregulation is commonly reported among autistic individuals. Prior work investigating the neurofunctional mechanisms of emotion regulation (ER) in autistic adults has illustrated alterations in dorsolateral prefrontal cortex (dlPFC) activity, as well as concurrent atypical patterns of activation in subcortical regions related to affect during cognitive reappraisal of social images. Whereas most research examining ER in autism has focused on regulation of negative emotions, the effects of regulating positive emotions has been generally understudied. This is surprising given the relevance of positive motivational states to understanding circumscribed interests (CI) in autism. Methods: Accordingly, the purpose of this study was to use fMRI with simultaneous eye-tracking and pupillometry to investigate the neural mechanisms of ER during passive viewing and cognitive reappraisal of a standardized set of nonsocial images and personalized (self-selected) CI images. Results: The autistic group demonstrated comparatively reduced modulation of posterior cingulate cortex (PCC) activation during cognitive reappraisal of CI images compared to viewing of CI, although no eye-tracking/pupillometry differences emerged between-groups. Further, the autistic group demonstrated increased PCC connectivity with left lateral occipital and right supramarginal areas when engaging in cognitive reappraisal vs. viewing CI. Discussion: In autistic adults, CI may be differentially modulated via PCC. Considering the documented role of the PCC as a core hub of the default mode network, we further postulate that ER of CI could potentially be related to self-referential cognition.

ABSTRACT

The purpose of the current study was to assess meaningful variability in visual-perceptual skills using a standardized assessment of visual perception, the Test of Visual Perceptual Skills (TVPS), across children with and without autism spectrum disorder (ASD). In addition to assessing overall accuracy across subtests of the TVPS, we also assessed response variability at the item-level, and the linear relationship between quantitative measures of ASD symptoms, task performance, and item-level variance. We report a significant linear relationship between ASD features and performance on the TVPS Figure Ground subtest. Additionally, results of an item-level analysis point to a significant relationship between within-task variability on the Figure Ground subtest and quantitative ASD traits, with a less variable response pattern being associated with increased ASD symptoms. Findings presented here suggest variability in perceptual processing across ASD may be influenced by individual differences in trait distribution.

ABSTRACT

The current study examined the relationship between quantitative measures of reward and punishment sensitivity, features of autism spectrum disorder (ASD), and resting and functional pupil response metrics across a clinically heterogeneous sample. Scores on a parent-report measure of punishment and reward sensitivity were correlated with ASD features. We also assessed whether pupil measurements could be used as a physiologic correlate of reward sensitivity and predictor of ASD diagnosis. In a logistic regression model, pupil dilation metrics, sex, and IQ, correctly classified 86.3% of participants as having an ASD diagnosis versus not. This research highlights individual differences of reward sensitivity associated with ASD features. Results support the use of pupil metrics and other patient-level variables as predictors of ASD diagnostic status.

ABSTRACT

We assessed the association between dynamic changes in pupil response in the context of visual perception and quantitative measures of the autism phenotype in healthy adults. Using Navon stimuli in a task-switching paradigm, participants were instructed to identify global or local information based on a cue. Multiple pupil response trajectories across conditions were identified. We combined trajectory patterns for global and local conditions and used data-driven methods to identify three distinct pupil trajectory sub-groups. We report higher scores on quantitative measures of autism features in individuals who demonstrated an increased change in pupil diameter across both conditions. Results demonstrate the use of individualized pupil response trajectories in order to quantitatively characterize visual perception associated with the broader autism phenotype (BAP).

ABSTRACT

We applied a trajectory-based analysis to eye tracking data in order to quantify individualized patterns of pupil response in the context of global-local processing that may be associated with autism spectrum disorder (ASD) features. Multiple pupil response trajectories across both global and local conditions were identified. Using the combined trajectory patterns for global and local conditions for each individual, we were able to identify three groups based on trajectory group membership that were thought to reflect perceptual strategy. Results indicated that the proportion of children with ASD was significantly greater in the group demonstrating a local-focus response. This research presents a novel analytic approach to the objective characterization of individualized pupil response patterns that are associated with ASD features.

ABSTRACT

The pupil is known to reflect a range of psychological and physiological variables, including cognitive effort, arousal, attention, and even learning. Within autism spectrum disorder (ASD), some work has used pupil physiology to successfully classify patients with or without autism. As we have come to understand the heterogeneity of ASD and other neurodevelopmental disorders, the relationship between quantitative traits and physiological markers has become increasingly more important, as this may lead us closer to the underlying biological basis for atypical responses and behaviors. We implemented a novel paradigm designed to capture patterns of pupil adaptation during sustained periods of dark and light conditions in a pediatric sample that varied in intellectual ability and clinical features. We also investigate the relationship between pupil metrics derived from this novel task and quantitative behavioral traits associated with the autism phenotype. We show that pupil metrics of constriction and dilation are distinct from baseline metrics. Pupil dilation metrics correlate with individual differences measured by the Social Responsiveness Scale (SRS), a quantitative measure of autism traits. These results suggest that using a novel, yet simple, paradigm can result in meaningful pupil metrics that correlate with individual differences in autism traits, as measured by the SRS.

ABSTRACT

Prosaccade and antisaccade errors in the context of social and nonsocial stimuli were investigated in youth with autism spectrum disorder (ASD; n = 19) a matched control sample (n = 19), and a small sample of youth with obsessive compulsive disorder (n = 9). Groups did not differ in error rates in the prosaccade condition for any stimulus category. In the antisaccade condition, the ASD group demonstrated more errors than the control group for nonsocial stimuli related to circumscribed interests, but not for other nonsocial stimuli or for social stimuli. Additionally, antisaccade error rates were predictive of core ASD symptom severity. Results indicate that the cognitive control of visual attention in ASD is impaired specifically in the context of nonsocial stimuli related to circumscribed interests.
