ABSTRACT

The objective was to quantify the effects of age and ractopamine (RAC) on whole body oxygen consumption and Leu flux, and oxygen flux and metabolism of nitrogenous compounds by the portal-drained viscera (PDV), liver, and hindquarters (HQ) of steers. Multicatheterized steers were fed a high energy diet every 2 h in 12 equal portions. Five younger steers (body weight, [BW] = 223 ±â€…10.1 kg) were 6 mo old and five older steers (BW = 464 ±â€…16.3 kg) were 14 mo old. Treatments were control (Cont) or 80 mg RAC per kg diet in a crossover design. Nitrogen (N) balance was measured on day 9 to 13. Whole body oxygen consumption and net flux were measured on day 11 and day 13, and net flux of N variables, Phe and Leu kinetics were measured on day 13. Whole body oxygen consumption increased (P < 0.05) in response to RAC in older but not younger steers. Retained N was greater (P = 0.009) for younger than older steers and increased (P = 0.010) with RAC in both ages of steers. Nitrogen retained as a percentage of N apparently absorbed increased (P < 0.05) in the older steers but not the younger steers in response to RAC. Oxygen uptake was greater (P < 0.05) in PDV, liver, and total splanchnic tissues in the younger steers and there was no response to RAC. In contrast, oxygen uptake in HQ increased (P < 0.05) with RAC in the older but not the younger steers. Concentration and net PDV release of α-amino N (AAN) were not affected by age or RAC. Uptake of AAN by liver decreased with RAC (P = 0.001). Splanchnic release of AAN was greater in younger steers (P = 0.020) and increased (P = 0.024) in response to RAC. For HQ tissues, uptake (P = 0.005) and extraction (P = 0.005) of AAN were lesser in older than younger steers and both increased (P = 0.001) in response to RAC. Based on Phe kinetics in HQ, RAC increased (P < 0.05) protein synthesis in older steers but not in younger steers. In contrast, protein breakdown decreased (P < 0.05) in response to RAC in younger steers. In response to RAC, protein degradation was less (P < 0.05) in younger than older steers. Based on Leu kinetics, whole body protein synthesis was greater in the younger steers (P = 0.022) but not altered in response to RAC. Ractopamine enhanced lean tissue growth by increasing supply of AAN to peripheral tissues and altering protein metabolism in HQ. These metabolic responses are consistent with established responses to RAC in production situations.


Ractopamine (RAC) is a feed additive that stimulates rate of gain, feed efficiency, and carcass leanness in finishing cattle. The objective of the present study was to quantify the effects of age and RAC on whole body metabolism and nutrient use by tissues of the portal-drained viscera, liver, and hindquarters of steers. Whole body oxygen consumption and oxygen uptake in hindquarters increased in response to RAC in older but not younger steers. Retained nitrogen was greater for younger than older steers and increased with RAC in both ages of steers. Uptake of α-amino nitrogen by liver decreased with RAC. ABSTRACT

Fescue toxicosis is a syndrome that results when cattle consume toxic endophyte-infected tall fescue. The objective of this study was to compare the response in physiological variables, sweat gland function, hair follicle cycling, and gene expression to feeding a total mixed ration that included tall fescue haylage and tall fescue seed containing a toxic endophyte (EI) or tall fescue haylage containing a nontoxic novel endophyte (EN) in beef heifers (Angus × Senepol heifers, n = 31) with 2 different hair genotypes. Numbers in each subgroup were as follows: novel endophyte, heterozygous slick (EN-S; n = 8), novel endophyte, homozygous hairy (wild type, EN-W; n = 7), endophyte-infected, heterozygous slick (EI-S; n = 10), and endophyte-infected, homozygous hairy (wild type, EI-W; n = 6). Physiological measurements were taken weekly for 7 wk. Data were analyzed using the MIXED procedure of SAS including dietary fescue treatment (EN vs. EI) and hair genotype (S vs. W) as main effects, day as a repeated measure, and temperature-humidity index (THI) as a covariate. Skin biopsies were taken before treatment initiation and on day 37 of treatment. Average surface temperature (ST) increased as the THI increased (P < 0.0001). Average ST was greater (P < 0.01) for animals fed EI than for animals fed the EN fescue diet, and greater (P < 0.01) for animals with the W genotype compared with animals with the S genotype. The difference between heifers with the S and W genotype was greater at greater THI (genotype × day interaction, P < 0.01). Transepidermal water loss (TEWL) was greater (P < 0.05) for animals with the S genotype compared with the W genotype and greater (P < 0.05) for heifers with the S genotype than for heifers with the W genotype when fed EI (36.7, 38.5, 30.0, and 38.7 g/m2 per hour for EN-W, EN-S, EI-W, and EI-S, respectively). The fraction of follicles in telogen in plucked hair samples for heifers fed EI was greater for animals with the S genotype than the W genotype (fraction in telogen: 0.456, 0.565, 0.297, 0.702 for EN-W, EN-S, EI-W, and EI-S, respectively; diet × genotype interaction, P < 0.05). Fraction of follicles in anagen was the opposite. EI fescue resulted in increased ST, changes in hair follicle cycling that support greater hair growth, and decreased TEWL for heifers with the W genotype compared with S genotype, suggesting greater heat stress in response to EI.

ABSTRACT

Fescue toxicosis is a multifaceted syndrome common in cattle grazing endophyte-infected tall fescue. The objective of this study was to evaluate the impact of the slick hair trait on physiological and reproductive parameters in heifers experiencing fescue toxicosis. Angus × Senepol heifers (n = 31) were blocked by weight (393.5 ± 17.3 kg) and phenotype relative to hair coat at birth, and randomly fed novel endophyte fescue (EN) or endophyte-infected fescue (EI) haylage in a total mixed ration for 91 d. Weekly measurements were collected to monitor heifer growth and response during ergot alkaloids exposure. Following 28 d of treatment, estrus was synchronized and heifers were inseminated. Ovary mapping and AI pregnancy rate were examined via transrectal ultrasonography. Blood samples were taken for genotyping: slick (S) or wildtype (W). Data were analyzed using repeated measures in PROC MIXED of SAS including fescue treatment (EN vs. EI), genotype (S vs. W), and sample collection time as main effects. Body condition scores were decreased for W heifers compared with S heifers (5.48 vs. 5.66, respectively; P < 0.0001). Surface temperature was greater for EI-W heifers (37.2 °C) compared with other groups (36.4, 36.6, 36.7 °C for EN-S, EN-W, EI-S, respectively; P < 0.05). Serum PRL concentrations were reduced for EI heifers compared with EN heifers (133.5 vs. 163.1 ng/mL, respectively; P < 0.05). The average number of 2 to 4 mm follicles were greater in EI-W heifers (13.8 follicles) compared with other groups (12.2, 10.6, and 11.1 for EN-S, EN-W, and EI-S, respectively; P < 0.0001). However, the average number of preovulatory follicles (≥9 mm) were reduced in EI-W heifers (0.52 follicles) compared with other heifer groups (0.94, 0.88, and 0.85 ± 0.04 for EN-S, EN-W, and EI-S, respectively; P < 0.05). Ovulatory follicle size was smaller in EI-W heifers compared with EN-W heifers (9.14 vs. 11.57 mm, respectively; P = 0.05). Corpus luteum area was reduced in EI-W heifers (235.1 mm2) compared with other heifer groups (297.2, 272.7, and 276.8 mm2 for EN-S, EN-W, and EI-S, respectively; P < 0.05). Concentrations of P4 were greater for EN heifers compared with EI heifers (2.7 vs. 1.8 ng/mL, respectively; P < 0.05). Pregnancy was not established in EI-W heifers (0%) compared with other heifer groups (37.5%, 57.1%, and 62.5% for EN-S, EN-W, and EI-S, respectively; P < 0.05). Overall, the slick hair mutation appears to aid in offsetting the physiological symptoms associated with fescue toxicosis and helps to improve reproductive performance.

ABSTRACT

Two studies separated effects of dietary ergot alkaloids from effects of feed intake or ambient temperature on respiration rate (RR), heart rate (HR), surface temperature (ST), rectal temperature (RT), blood pressure (BP), serum hormone, and plasma metabolite concentrations in beef steers. The balanced, single reversal design for each experiment used 8 beef steers fed tall fescue seed [2.5 g/kg body weight (BW)] with (E+) or without (E-) ergot alkaloids as part of a 60:40 switchgrass hay: supplement diet. Periods were 35 days with 21 days of preliminary phase and 14 days of feeding fescue seed once daily. Measures of dependent variables were collected on d 20, 25, 29, and 35 of each period at 0730 (before feeding), 1230 and 1530. In Experiment 1 steers weighed 286 kg, gained 0.61 kg BW/day, E+ supplied 2.72 mg ergot alkaloids including 1.60 mg ergovaline per steer daily, and mean minimum and maximum daily ambient temperatures were 23.6 and 32.3°C. In Experiment 2 steers weighed 348 kg, gained 1.03 kg BW/day, E+ supplied 3.06 mg ergot alkaloids including 2.00 mg ergovaline daily, and mean minimum and maximum daily ambient temperatures were 11.9 and 17.4°C. Dry matter intake was not affected by fescue seed treatment (P < 0.20) in either experiment. In both experiments, E+ reduced HR (P < 0.01) and increased insulin (P = 0.07). Systolic BP minus diastolic BP decreased (P < 0.05) for E+ in both experiments, due to increased diastolic BP in Experiment 1 (P < 0.03) and decreased systolic BP in Experiment 2 (P < 0.07). In Experiment 1, above the thermoneutral zone, E+ increased (P < 0.05) RR, RT, and left side ST in comparison to E-, but in Experiment 2, within the thermoneutral zone, E+ and E- did not differ (P < 0.18). Ergot alkaloids from fescue seed affect the cardiovascular system of steers separately from effects of feed intake or environmental temperature. Ergot alkaloids interact with ambient temperatures above the steers' thermoneutral zone to exacerbate the symptoms of hyperthermic stress.

ABSTRACT

During the differentiation of muscle satellite cells, committed myoblasts respond to specific signalling cues by exiting the cell cycle, migrating, aligning, expressing muscle-specific genes and finally fusing to form multinucleated myotubes. The predominant foetal growth factor, IGF-2, initiates important signals in myogenesis. The aim of this study was to investigate whether ERK5 and its upstream MKK activator, MEK5, were important in the pro-myogenic actions of IGF-2. ERK5 protein levels, specific phosphorylation and kinase activity increased in differentiating C2 myoblasts. ERK5-GFP translocated from the cytoplasm to the nucleus after activation by upstream MEK5, whereas phospho-acceptor site mutated (dominant-negative) ERK5AEF-GFP remained cytoplasmic. Exogenous IGF-2 increased MHC levels, myogenic E box promoter-reporter activity, ERK5 phosphorylation and kinase activity, and rapidly induced nuclear localisation of ERK5. Transfection with antisense Igf2 decreased markers of myogenesis, and reduced ERK5 phosphorylation, kinase and transactivation activity. These negative effects of antisense Igf2 were rescued by constitutively active MEK5, whereas transfection of myoblasts with dominant-negative MEK5 blocked the pro-myogenic action of IGF-2. Our findings suggest that the MEK5-ERK5 pathway is a novel key mediator of IGF-2 action in myoblast differentiation.

ABSTRACT

OBJECTIVE: To develop an endoscopic technique for use in monitoring devlopment of gastric ulcers via a gastric cannula during withholding of feed and administration of a finely ground diet to pigs. ANIMALS: 6 pigs weighing between 60 and 70 kg. PROCEDURE: A gastric cannula was surgically inserted adjacent to the pars esophagea in each pig. Pigs were fed a finely ground diet for two 7-day periods that were separated by a 48-hour period during which feed was withheld. Endoscopic examination via the gastric cannula was used to monitor development of ulcers in the pars esophageal region of the pigs during the 48-hour period of feed withhold and subsequent 7-day feeding period. An ulcer score was assigned during each endoscopic examination. A final examination was performed during necropsy and compared with results for the final endoscopic examination. RESULTS: Consumption of a finely ground diet for 7 days resulted in progressive erosive damage to the pars esophageal region of the stomach. Further significant increases in ulcerative damage were detected after 24 and 48 hours of withholding of feed. Final examination during necropsy did not reveal significant differences from results obtained during the final endoscopic examination. CONCLUSIONS AND CLINICAL RELEVANCE: Endoscopic examination via a gastric cannula was an effective means of monitoring ulcer development in the pars esophagea of pigs. Feeding a finely ground diet and withholding of feed induced endoscopically observable ulcers in the stratified squamous epithelial region of the stomach. Direct visual examination during necropsy confirmed the accuracy of endoscopic examination.

ABSTRACT

OBJECTIVE: To determine whether withholding feed from pigs prior to slaughter had any effects on meat quality, percentage of pigs with Salmonella spp in cecal contents during slaughter, or percentage of pigs with lacerations of the gastrointestinal tract during slaughter. DESIGN: Split-plot design. ANIMALS: 873 pigs. PROCEDURES: At the finishing barn, pigs were assigned to 30 pens. Feed withdrawal times were assigned to pens at random, and pigs in each pen were marketed in 3 groups. The first marketing group consisted of the 10 heaviest pigs in each pen, the second consisted of the next 10 heaviest pigs, and the third consisted of all remaining pigs. RESULTS: Withdrawing feed improved the redness score assigned to the meat but did not have any other significant effects on carcass composition or meat quality. The percentage of pigs with Salmonella spp in the cecal contents decreased from the first (73%) to the second (64%) to the third (52%) marketing group. However, isolation of Salmonella spp from cecal contents was not associated with feed withdrawal time or with pen prevalence of Salmonella shedding during the 2 months prior to slaughter. Feed withdrawal time and marketing group did not have any significant effects on overall prevalence of gastrointestinal tract lacerations. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggest that withdrawal of feed from pigs prior to slaughter does not increase the prevalence of Salmonella colonization or the risk of carcass contamination associated with gastrointestinal tract lacerations during slaughter but only slightly enhances meat quality.

OBJECTIVE: To determine whether withdrawing feed from pigs prior to slaughter had any effects on prevalence or severity of gastric ulcers. DESIGN: Split-plot design. ANIMALS: 873 pigs. PROCEDURES: At the finishing barn, pigs were assigned to 30 pens. Feed withdrawal times (0, 12, or 24 hours) were assigned to pens at random, and pigs in each pen were marketed in 3 groups over a period of 4 weeks. The first marketing group consisted of the 10 heaviest pigs in each pen, the second consisted of the next 10 heaviest, and the third consisted of all remaining pigs. Feed was withheld from all pigs in each pen prior to removal of each marketing group. Thus, feed was withheld once, twice, or 3 times for pigs in the first, second, and third marketing groups, respectively. RESULTS: Feed withdrawal time was not significantly associated with ulcer score at the time of slaughter. Ulcer scores and prevalence of chronic damage were higher in the third marketing group, regardless of feed withdrawal time. Prevalence of severe damage, prevalence of chronic damage, and prevalence of esophageal constriction increased as carcass weight decreased. No pigs died of gastric ulceration. CONCLUSION AND CLINICAL RELEVANCE: Results suggest that withdrawal of feed from pigs prior to slaughter does not increase damage to the stomach and that repeated feed withdrawal does not result in fatal gastric ulceration.
