ABSTRACT

L-arabinose isomerase (L-AI) is a functional enzyme for the isomerizing of D-galactose to produce D-tagatose. In this study, L-AI-C6-encoding gene from the probiotic Lactobacillus fermentum C6 was cloned and expressed in Bacillus subtilis WB600 for investigating enzymatic characteristics and bioconverting D-tagatose by means of whole-cell catalysis. Results showed that the engineered B. subtilis WB600-pMA5-LAI achieved a maximum specific activity of L-AI-C6 (232.65 ±â€¯15.54 U/mg protein) under cultivation in LB medium at 28 °C for 40 h. The recombinant L-AI-C6 was purified, and enzymatic characteristics test showed its optimum reaction temperature and pH at 60 °C and 8.0, respectively. In addition, L-AI-C6 exhibited good stability within the pH range of 5.5-9.0. By using B. subtilis WB600-pMA5-LAI cells as whole-cell catalyst, the highest D-tagatose yield reached 42.91 ±â€¯0.28 % with D-galactose as substrate, which was 2.41 times that of L. fermentum C6 (17.79 ±â€¯0.11 %). This suggested that the cloning and heterologous expression of L-AI-C6 was an effective strategy for improving D-tagatose conversion by whole-cell catalysis. In brief, the present study demonstrated that the reaction temperature, pH, and stability of L-AI-C6 from L. fermentum C6 meet the demands of industrial application, and the constructed B. subtilis WB600-pMA5-LAI shows promising potential for the whole-cell biotransformation of D-tagatose.

ABSTRACT

The online monitoring of GIS equipment can be realized through detecting SF6 decomposition gasses. Metal oxide heterojunctions are widely used as gas-sensing materials. In this study, the structural and electrical properties of In2O3-ZnO and TiO2-ZnO heterojunctions were analyzed based on density functional theory calculations. After heterojunction structural optimization, the electrical conductivity of these two heterojunctions was enhanced compared to each intrinsic model, and the electrical conductivity is ranked as follows: In2O3-ZnO heterojunction > TiO2-ZnO heterojunction. The gas-sensing response of these two heterojunctions to four SF6 decomposition gasses, H2S, SO2, SOF2, and SO2F2, was investigated. For gas adsorption systems, the adsorption energy, charge transfer, density of states, charge difference density, and frontier molecular orbitals were calculated to analyze the adsorption and gas-sensing performance. For gas adsorption on the In2O3-ZnO heterojunction surface, the induced conductivity changes are in the following order: H2S > SO2F2 > SOF2 > SO2. For gas adsorption on the TiO2-ZnO heterojunction surface, H2S and SOF2 increase conductivity, and SO2 and SO2F2 decrease conductivity.

ABSTRACT

In this study, we report the successful synthesis of few-layer parallel PtSe2ribbons on an Au foil employing a surface melting strategyviathe chemical vapor deposition growth method at 650 °C. The controlled formation of parallel ribbons was directed by the Au steps generated through antimony treatment. These ribbons exhibit an average length of exceeding 100µm and a width of approximately 100 nm across a substantial area. Electrocatalysis measurements showcase the catalytic performance of PtSe2ribbons grown on Au foil, which can be further augmented through subsequent oxidation treatment. This investigation introduces an effective growth method for few-layer ribbons at low temperatures and broadens the scope of employing the substrate-guided strategies for the synthesis of one-dimensional materials. Additionally, it underscores the potential of PtSe2ribbons as an electrocatalyst for hydrogen evolution.

ABSTRACT

Chinese wild rice (CWR) is a nutritious and healthy whole grain, worth developing. To develop and use its value, a new type of huangjiu was brewed with CWR, and the flavour characteristics, sensory quality, functional and bioactive components were evaluated. CWR (67 flavour substances) and glutinous rice (GR)-CWR huangjiu (62 flavour substances) had a better flavour than GR huangjiu (54 flavour substances), and the overall style of GR-CWR huangjiu was more skewed towards GR. The fruity, honey, caramel-like, herb and smoky aroma attributes of CWR huangjiu were higher than those of GR huangjiu (P < 0.05), while only the alcoholic was weaker (P < 0.05) due to the lower alcohol content. The huangjiu brewed using CWR had a better taste than that brewed using only GR. Furthermore, CWR huangjiu had the highest content of total dietary fiber (732.0 ± 15.2 mg/100 g), followed by GR-CWR (307.0 ± 8.5 mg/100 g), and GR (127.0 ± 2.3 mg/100 g). CWR huangjiu also had the highest total phenolic compounds (3.32 ± 0.05 mg/100 g/%vol) and total saponins (2.46 ± 0.03 mg/100 g/%vol) contents, followed by GR-CWR and GR. This study provides guidance for exploring further possibilities for CWR in the future.

ABSTRACT

An oil-immersed transformer is a critical electrical device for power delivery. Online monitoring of transformer operation is the key to ensuring the regular operation of power systems. This paper proposes Au/ZnO, Pd/ZnO, and Pt/ZnO heterojunctions as new gas-sensitive materials and investigates their gas-sensitive performance to dissolved gases (C2H4, CO, and H2) in transformer oil. Upon theoretical density functional theory (DFT) calculations, the analysis of the total density of states (TDOS), partial density of states (PDOS), molecular orbital theory, and charge deformation density reveals that Au, Pd, and Pt form heterojunctions with ZnO, which enhance the electrical conductivity of the system. Meanwhile, intrinsic ZnO is unsuitable for gas detection and adsorption, while the Au/ZnO heterojunction suits C2H4 detection. In contrast, the Pd/ZnO heterojunction is suitable for H2 detection, and the Pt/ZnO heterojunction is suitable for C2H4 and CO detection. The electrical conductivity of the adsorption models is changed to varying degrees after gas adsorption. The different change rate electrical conductivity just serves as a theoretical foundation for determining the type and concentration of dissolved gases in transformer oil. The research results act as a theoretical foundation for constructing gas sensors with a ZnO-based material.

ABSTRACT

Daqu is a saccharification agent required for fermenting Baijiu, a popular Chinese liquor. Our objective was to investigate the relationships between physicochemical indices, microbial community diversity, and metabolite profiles of strong-flavor Jinhui Daqu during different storage periods. During different storage periods of Jinhui Daqu, we combined Illumina MiSeq sequencing and non-target sequencing techniques to analyze dynamic changes of the microbial community and metabolite composition, established a symbiotic network and explored the correlation between dominant microorganisms and differential metabolites in Daqu. Fungal community diversity in 8d_Daqu was higher than that in 45d_Daqu and 90d_Daqu, whereas bacterial community diversity was higher in 90d_Daqu. Twelve bacterial and four fungal genera were dominant during storage of Daqu. Bacillus, Leuconostoc, Kroppenstedtia, Lactococcus, Thermomyces and Wickerhamomyces decreased as the storage period increased. Differences of microbiota structure led to various metabolic pathways, and 993 differential metabolites were found in all Daqu samples. Differential microorganisms were significantly related to key metabolites. Major metabolic pathways involved in the formation of amino acids and lipids, such as l-arogenate and hydroxyproline, were identified. Interactions between moisture, acidity, and microbes may drive the succession of the microbial community, which further affects the formation of metabolites.

ABSTRACT

KerJY-23 was a novel keratinase from feather-degrading Ectobacillus sp. JY-23, but its enzymatic characterization and structure are still unclear. In this study, the KerJY-23 was obtained by heterologous expression in Escherichia coli BL21(DE3), and enzymatic properties indicated that KerJY-23 was optimal at 60 °C and pH 9.0 and could be promoted by divalent metal ions or reducing agents. Furthermore, KerJY-23 had a broad substrate specificity towards casein, soluble keratin, and expanded feather powder, but its in vitro degradation against chicken feathers required an additional reducing agent. Homology modeling indicated that KerJY-23 contained a highly conserved zinc-binding HELTH motif and a His-Asp-Ser catalytic triad that belonged to the typical characteristics of M4-family metallo-keratinase and serine-keratinase, respectively. Molecular docking revealed that KerJY-23 achieved a reinforced binding on feather keratin via abundant hydrogen bonding interactions. This work not only deepened understanding of the novel and interesting metallo-serine keratinase KerJY-23, but also provided a theoretical basis for realizing the efficient use of waste feather keratin.

ABSTRACT

Wheat Qu plays the role of saccharification fermentation, providing microorganisms and flavor in the fermentation of huangjiu, and the use of functional microorganisms to fortify wheat Qu is becoming increasingly popular. Yet, the mechanisms promoting microbial successions of wheat Qu remain unclear. In this study, we first correlated microbial community succession with physicochemical factors (moisture, temperature, acidity, glucoamylase and amylase) in inoculated raw wheat Qu (IRWQ) with Saccharopolyspora rosea. The Mantel test was performed to investigate the significance and found that temperature (r = 0.759, P = 0.001), moisture (r = 0.732, P = 0.006), and acidity (r = 0.712, P = 0.017) correlated significantly with the bacterial community in phase 1 (0-40 h). Meanwhile, temperature correlated significantly with the fungal community in phases 1 and 2 (40-120 h). To confirm the effect of temperature on microbial communities, the artificial reduction of bio-heat (37°C) in IRWQ also reduced the relative abundance of heat-resistant microorganisms including Bacillus and Saccharopolyspora. A higher abundance of Saccharopolyspora (87%) in IRWQ was observed following biofortified inoculation of S. rosea, in which glucoamylase activity increased by 40% compared to non-inoculated raw wheat Qu (NIRWQ) (1086 U/g vs 776 U/g). Finally, the IRWQ was employed to mechanized huangjiu fermentation and it was found to reduce the bitter amino acid and higher alcohol content by 27% and 8%, respectively, improving the drinking comfort and quality of huangjiu.

ABSTRACT

Titanium thin films are particularly important as electrode layers, barrier layers, or intermediate buffer layers in the semiconductor industry. In order to improve the quality of Ti thin films and the adhesion and diffraction abilities of irregular parts, this paper used high-power pulsed magnetron sputtering (HPPMS/HiPIMS) to prepare titanium thin films. The effects of different trigger voltages (700 V, 800 V, and 900 V) on plasma properties were studied, and the microstructure, mechanical properties and corrosion resistance of the films were also studied. The results showed that as the voltage increased, the grain size of the thin films gradually increased. The residual stress of the titanium films changed from compressive stress (-333 MPa) to tensile stress (55 MPa) and then to low compressive stress (-178 MPa). The hardness values were 13 GPa, 9.45 GPa and 6.62 GPa, respectively. The wear resistance of the films gradually decreased, while the toughness gradually increased. The corrosion resistance of the films decreased as well.

ABSTRACT

In a concentrating photovoltaic (CPV) system, realizing an illumination spot consistent with the photosensitive area of the CPV cell while maintaining high optical performances is a challenging but rewarding issue. For instance, uniform irradiance distribution not only reduces the risk of damage to CPV cells due to the local heat accumulation, but also improves photoelectric conversion efficiency; free chromatic aberration ensures that a wide solar spectrum can be regulated; a high concentration ratio helps reduce the usage of CPV cells, and so on. Here, we propose a method to design a freeform mirror array to concentrate sunlight and generate a square light pattern with uniform irradiance distribution on the photovoltaic cell. Each freeform mirror can produce a uniform illumination pattern on the CPV cell. We demonstrate the effectiveness of the proposed method with several design examples, and a series of factors affecting the optical performance is also analyzed.

ABSTRACT

Phase generated carrier (PGC) is widely applied in interferometric phase estimation for distance, vibration and velocity measurements. However, traditional PGC methods suffer from nonlinear effects, causing limitations to demodulation of signal. Modified PGC methods, such as ellipse fitting algorithm (EFA), resolves these issues, but usually requires additional phase shift. With our proposed method in this paper, only one period of signal and one test point is required to attain accurate depth of phase modulation and phase. We use a photodiode to calibrate light intensity in data acquisition, and develop a Levenburg-Marquadt algorithm to estimate values of PGC parameters. An improved algorithm is also proposed to avoid local optimization based on prior information to ensure measurement stability. Less than 5 × 10-3 rad phase measurement uncertainty and over 55 dB Signal to Noise and Distortion Ratio (SINAD) is obtained in experiment.

ABSTRACT

The service life of an artificial hip joint is limited to 10-15 years, which is not ideal for young patients. To extend the lifespan of these prostheses, the coefficient of friction and wear resistance of metallic femoral heads must be improved. In this study, a Cu-doped titanium nitride (TiNX -Cu) film with "autoantifriction" properties is deposited on a CoCrMo alloy via magnetron sputtering. When delivered in a protein-containing lubricating medium, the Cu in TiNX -Cu quickly and consistently binds to the protein molecules in the microenvironment, resulting in the formation of a stable protein layer. The proteins adsorbed on the TiNX -Cu surface decompose into hydrocarbon fragments owing to the shear stress between the Al2 O3 /TiNX -Cu tribopair. The synergistic effect of the catalysis of Cu and shear stress between the Al2 O3 /TiNX -Cu tribopair transforms these fragments into graphite-like carbon tribofilms with an antifriction property. These tribofilms can simultaneously reduce the friction coefficient of the Al2 O3 /TiNX -Cu tribopair and enhance the wear resistance of the TiNX -Cu film. Based on these findings, it is believed that the autoantifriction film can drive the generation of antifriction tribofilms for lubricating and increasing the wear resistance of prosthetic devices, thereby prolonging their lifespan.

ABSTRACT

The freeform imaging system is playing a significant role in developing an optical system for the automotive heads-up display (HUD), which is a typical application of augmented reality (AR) technology. There exists a strong necessity to develop automated design algorithms for automotive HUDs due to its high complexity of multi-configuration caused by movable eyeballs as well as various drivers' heights, correcting additional aberrations introduced by the windshield, variable structure constraints originated from automobile types, which, however, is lacking in current research community. In this paper, we propose an automated design method for the automotive AR-HUD optical systems with two freeform surfaces as well as an arbitrary type of windshield. With optical specifications of sagittal and tangential focal lengths, and required structure constraints, our given design method can generate initial structures with different optical structures with high image quality automatically for adjusting the mechanical constructions of different types of cars. And then the final system can be realized by our proposed iterative optimization algorithms with superior performances due to the extraordinary starting point. We first present the design of a common two-mirror HUD system with longitudinal and lateral structures with high optical performances. Moreover, several typical double mirror off-axis layouts for HUDs were analyzed from the aspects of imaging performances and volumes. The most suitable layout scheme for a future two-mirror HUD is selected. The optical performance of all the proposed AR-HUD designs for an eye-box of 130 mm × 50 mm and a field of view of 13° × 5° is superior, demonstrating the feasibility and superiority of the proposed design framework. The flexibility of the proposed work for generating different optical configurations can largely reduce the efforts for the HUD design of different automotive types.

OBJECTIVE: To analyze the factors related to postoperative recurrence in patients with localized laryngeal amyloidosis (LocLA) and to construct a nomogram prediction model (NPM). METHODS: We collected the data for LocLA patients diagnosed from March 2000 to May 2019 and clinical characteristics data were extracted. Factors related to recurrence were analyzed using multivariate logistic regression. The NPM was constructed for predicting the recurrence risk of LocLA. The receiver operating characteristic (ROC) curve evaluated the distinguishing ability using the area under curve (AUC). The calibration curve was created to evaluate the consistency of the NPM. RESULTS: A total of 226 confirmed LocLA cases were included. One hundred seventy-five cases (77.4%) had localized single nodule, and 51 cases had more than one lesions. Sixty-three (27.9%) cases had no multinucleated giant cell (MGC) around amyloid, and 163 (72.1%) cases had MGC around amyloid. Multivariate logistic regression analysis showed that more than one lesions (odds ratio [OR] = 3.206 and 95% confidence interval [CI]: 1.492-6.888; P value: .003), subglottic involvement (OR = 2.926 and 95% CI: 1.300-6.585; P = .010), and no multinucleated giant cell (MGC) around amyloid (OR = 2.503 and 95% CI: 1.173-5.342; P = .018) had a statistically significant effect on postoperative LocLA recurrence (P < .05). The AUC of the ROC curve was 0.753 (95% CI: 0.667-0.832). The bias-corrected curve approached the ideal curve, with an average absolute error of 0.037. CONCLUSIONS: More than one lesions, subglottic involvement, and no MGC around amyloid are risk factors for postoperative recurrence of LocLA. The NPM constructed has good applicability.

ABSTRACT

Saccharopolyspora is an important microorganism in the fermentation process of wheat qu and huangjiu, yet the mechanisms by which it performs specific functions in huangjiu remain unclear. A strain with high amylase and glucoamylase activities was isolated from wheat qu and identified as Saccharopolyspora rosea (S. rosea) A22. We initially reported the whole genome sequence of S. rosea A22, which comprised a circular chromosome 6,562,638 bp in size with a GC content of 71.71%, and 6,118 protein-coding genes. A functional genomic analysis highlighted regulatory genes involved in adaptive mechanisms to harsh conditions, and in vitro experiments revealed that the growth of S. rosea A22 could be regulated in response to the stress condition. Based on whole-genome sequencing, the first genome-scale metabolic model of S. rosea A22 named iSR1310 was constructed to predict the growth ability on different media with 91% accuracy. Finally, S. rosea A22 was applied to huangjiu fermentation by inoculating raw wheat qu, and the results showed that the total higher alcohol content was reduced by 12.64% compared with the control group. This study has elucidated the tolerance mechanisms and enzyme-producing properties of S. rosea A22 at the genetic level, providing new insights into its application to huangjiu.

ABSTRACT

In this paper, a method is proposed to solve the initial optical structure of an off-axis multimirror system for an extreme ultraviolet lithography (EUVL) application. By tracing the characteristic rays, the primary aberration can be expressed as a function of the distance and curvature based on the Seidel aberration theory. The initial structure with a favorable aberration performance is calculated when the function value is 0. We solve two different initial structures with an off-axis, six-mirror configuration with different optical powers. The NA of the finally optimized optical system is 0.25, the wavefront aberration rms value is less than 0.04λ, and the absolute distortion is below 1.2 nm.

ABSTRACT

Secretory carcinoma (SC) is a recently recognized type of salivary gland tumor characterized by t(12;15) (p13;q25) translocation resulting in an ETV6-NTRK3 gene fusion. Most SCs are located in a main salivary gland, and primary sinonasal secretary carcinoma is rare. We describe three cases of primary SC in the sinonasal cavity with high-grade transformation (HGT) in one case, and the first case in the pharynx. All tumors comprised slightly atypical cells with solid, tubular, microcystic growth patterns. The case with HGT included two components with distinct sharp boundaries and comedo necrosis, high mitotic figures and obvious cellular atypia. Tumor cells were positive for vimentin, S100, and Gata-3 and negative for p63 and DOG-1. Three cases showed nuclear staining of pan-TRK and one showed cytoplasmic staining. All cases harbored ETV6 gene rearrangement, and ETV6-NTRK3 gene fusion was detected in three cases. Most patients were treated with radical resection and adjuvant therapy. After excision, all remained tumor-free for 65-164 months (medium 98.5 months). SC in the sinonasal cavity and pharynx is a low-grade malignant tumor with histologic features overlapping those of other salivary gland tumors. Immunohistochemical analysis and fluorescence in situ hybridization are useful techniques for its differential diagnosis.

BACKGROUND: Mechanized Huangjiu is a stable product, is not subject to seasonal production restrictions, and markedly reduces labor intensity compared to traditional manual Huangjiu. However, the bitterness of mechanized Huangjiu impedes its further development. RESULTS: Based on process optimization, when the fermentation temperature was 45 °C and the fermentation time was 122 h, the inoculation amount of Saccharopolyspora was 5%, the amount of added water was 26%, and the glucoamylase and amylase activities of wheat Qu increased by 27% and 40% respectively, compared with those before optimization. Huangjiu fermented by raw wheat Qu inoculated with Saccharopolyspora rosea F2014 showed a significant (P < 0.05) decrease in bitter amino acid content (1.24 vs. 2.86 g L-1 , a decrease of 56%), which attenuated its bitterness. CONCLUSION: An innovative fermentation process of inoculating Saccharopolyspora into raw wheat Qu was developed for the first time. ABSTRACT

Freeform illumination optics design with extended light sources to realize an accurate light control is very important, but still remains a challenging issue. Here, we propose a new method to design compact and efficient freeform lenses for extended sources. We employ a localized surface control strategy to directly modify the freeform surface to redistribute the light rays emitted from the extended LED source in a desired manner. By the combination of basic radiometry calculation and backward ray tracing, we obtain the irradiance distribution on the target plane and estimate the localized freeform surface to be modified. The optimization function with a Gaussian form is adopted to modify the localized surface. The smoothness of the freeform surface is taken into account in the optimization process to guarantee the processability of the freeform optics. We demonstrate the effectiveness of the proposed method with three design examples.

ABSTRACT

Sodium chlorides in non-1:1 stoichiometry are counterintuitive but recently their existence has been found under the high pressure condition or in the confined space between graphene sheets. Here the direct observation of the formation of Na3 Cl nanoclusters, a stable magic-number structure, is reported on an Ir(111) surface using scanning tunneling microscopy and noncontact atomic force microscopy. The stability of Na3 Cl nanoclusters in the free and adsorbed state is corroborated by density functional theory calculations. It is also found that a density of nanoclusters together with Cl adatoms may further aggregate and self-assemble into a Na3 Cl4 monolayer, forming a novel metastable phase of NaCl(111) with a honeycomb lattice. Further calculations suggest that charge transfer between the polar nanoclusters and the metal substrate stabilizes NaCl of non-1:1 stoichiometry. The work exhibits the possibility of exploring unconventional ionic crystals on the surface with atomically precise control of structure and composition.
