ABSTRACT

Cholangiocytes play a crucial role in bile formation. Cholangiocyte injury causes cholestasis, including primary biliary cholangitis (PBC). However, the etiology of PBC remains unclear despite being characterized as an autoimmune disease. Using single-cell RNA sequencing (scRNA-seq), fluorescence-activated-cell-sorting, multiplex immunofluorescence (IF) and RNAscope analyses, we identified unique DUOX2+ACE2+ small cholangiocytes in human and mouse livers. Their selective decrease in PBC patients was associated with the severity of disease. Moreover, proteomics, scRNA-seq, and qPCR analyses indicated that polymeric immunoglobulin receptor (pIgR) was highly expressed in DUOX2+ACE2+ cholangiocytes. Serum anti-pIgR autoantibody levels were significantly increased in PBC patients, regardless of positive and negative AMA-M2. Spatial transcriptomics and multiplex IF revealed that CD27+ memory B and plasma cells accumulated in the hepatic portal tracts of PBC patients. Collectively, DUOX2+ACE2+ small cholangiocytes are pathogenic targets in PBC, and preservation of DUOX2+ACE2+ cholangiocytes and targeting anti-pIgR autoantibodies may be valuable strategies for therapeutic interventions in PBC.

ABSTRACT

ABSTRACT This study is to investigate the most efficient extractives of extracting oil recipe for stroke treatment and the protective effects on an oxygen and glucose deprivation model in PC12 cells. An orthogonal experimental design L9 (34) was carried out for oil recipe's optimization with supercritical CO2 fluid extraction. 2-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide and enzyme-linked immunosorbent assay were conducted to evaluate cell activity and indexes in the cell lysate. The result showed that the optimum extraction condition was 30 Mpa, 50 ºC, 100 min, the extracts were analyzed by gas chromatography-mass spectrometry and among forty detected compounds 27 were identified, representing 80.86% of the total oil content. trans-Cinnamaldehyde (14.14%), piperine (9.32%), β-amyrin (6.79%), lupenone (6.28%), longifolene (6.07%), β-caryophyllene (5.21%), α-bisabolol (4.11%), and β-bisabolene (2.56%) were high mass fraction. Oil recipe could significantly attenuate PC12 cell damage, the lactate dehydrogenase release and decreased the malondialdehyde levels, glutathione peroxidase and nicotinamide adenine dinucleotide phosphate oxidase activity, glutathione and nitric oxide content (p < 0.01) and increased the level of superoxide dismutase after oxygen and glucose deprivation. The protective mechanism may be related to oil recipe's antioxidant effect by scavenging free radicals.

ABSTRACT

PURPOSE:: To investigate the hepatotoxicity and nephrotoxicity of 3-Bromopyruvate (3BP) in mice. METHODS:: Fifteen nude mice were grafted subcutaneously in the left flank with MDA-MB-231 cells, then all mice were divided into control group (PBS), 3BP group (8 mg/kg), positive group (DNR: 0.8 mg/kg) when tumor volume reached approximately 100 mm3. 28 days later, tumors, livers and kidneys were stored in 4 % formalin solution and stained with hematoxylin and eosin staining. The Kunming mice experiment included control group (PBS), 3BP group (4mg/kg; 8mg/kg; 16mg/kg), positive group (DNR: 0.8 mg/kg). 24 hours later, the blood were used for the determination of hepatic damage serum biomarkers. Livers were stored in 4 % formalin solution for the later detection. RESULTS:: 3BP at the dose of 8mg/kg had a good effect on inhibiting tumor growth in nude mice and did not damage liver and kidney tissues. Kunming mice experiment showed 3BP at the dose of 16mg/kg did damage to liver tissues. CONCLUSION:: 3-Bromopyruvate at the dose of suppressing tumor growth did not exhibit hepatotoxicity and nephrotoxicity in nude mice, and the effect on liver was confirmed in Kunming mice.
