ABSTRACT

Banana Fusarium oxysporum f. sp. cubense tropical race 4 (Foc-TR4) is a highly destructive pathogen that infects nearly all major banana cultivars and has a tendency to spread further. Secreted proteins play a crucial role in the process of Fusarium wilt infection in bananas. In this study, we analyzed the codon usage bias (CUB) of the Foc-TR4 classical secretory protein genome for the first time and observed a strong bias toward codons ending with C. We found that 572 out of the 14,543 amino acid sequences in the Foc-TR4 genome exhibited characteristics of classical secretory proteins. The CUB was largely influenced by selection optimization pressure, as indicated by the ENC value and neutral plot analysis. Among the identified codons, such as UCC and CCC, 11 were found to be optimal for Foc-TR4 gene expression. Codons with higher GC content and a C base in the third position showed greater selectivity. The CUB in the secretory proteins encoded by Foc-TR4 provides insights into their evolutionary patterns, contributing to the development and screening of novel and effective antifungal drugs.

ABSTRACT

Citrus reticulata Blanco 'Orah' is grown throughout southern China and provides enormous economic value. However, the agricultural industry has suffered substantial losses during recent years due to marbled fruit disease. The present study focuses on the soil bacterial communities associated with marbled fruit in 'Orah'. The agronomic traits and microbiomes of plants with normal and marbled fruit from three different orchards were compared. No significant differences were found in agronomic traits between the groups, except for higher fruit yields and higher quality of fruits in normal fruit group. Additionally, a total of 2,106,050 16S rRNA gene sequences were generated via the NovoSeq 6000. The alpha diversity index (including the Shannon and Simpson indices), Bray-Curtis similarity, and principal component analyses indicated no significant differences in microbiome diversity between normal and marbled fruit groups. For the healthy 'Orah', the most abundant associated phyla were Bacteroidetes, Firmicutes, and Proteobacteria. In comparison, Burkholderiaceae and Acidobacteria were the most abundant taxa with the marbled fruit group. In addition, the family Xanthomonadaceae and the genus Candidatus Nitrosotalea were prevalent with this group. Analysis using the Kyoto Encyclopedia of Genes and Genomes pathways showed that several pathways related to metabolism significantly differed between the groups. Thus, the present study provides valuable information regarding soil bacterial communities associated with marbled fruit in 'Orah'.

ABSTRACT

During the forming process of carbon fiber composite pressure vessels, the parameters of the curing and forming processes become one of the critical factors affecting the production cost and forming quality. The curing temperature of 4251 A4/B2 epoxy resin is measured in this research, and the effect of curing temperature on the mechanical properties of composite materials for winding is studied, which is finally verified in the test of pressure vessels. First, the actual curing temperature of the epoxy resin is tested and analyzed using differential scanning calorimetry (DSC). Second, under two different curing regimes, the tensile and flexural properties are tested by making pure epoxy resin matrix test pieces, Naval Ordnance Laboratory (NOL) rings, and carbon fiber composite unidirectional plates that affect the overall performance of composite pressure vessels. At the same time, the test results provide reliable process parameters for numerical simulation and manufacturing of pressure vessels. Finally, the filament-wound 35 MPa type III pressure vessel is cured and carried out using a hydraulic burst test. The results show the resin matrix has good fluidity and excellent interface bonding with carbon fiber when the curing temperature is 112 °C. Compared with the results in curing temperature of 100 °C, the tensile strength of the NOL ring reaches 2260.8 MPa, up by 22%. In the 90° direction, the tensile and flexural strengths of the unidirectional plates increase by 68.86% and 37.42%, respectively. In the 0° direction, the tensile and flexural strengths of the unidirectional plates increase by 5.82% and 1.16%, respectively. The pressure vessel bursting form is reasonable and meets the CGH2R standard. The bursting pressure of the vessel is up to 104.4 MPa, which verifies the rationality of the curing regime used in the curing process of the pressure vessel. Based on the results of this paper, the curing temperature affects the fluidity of the epoxy resin, which in turn affects the interfacial bonding properties of the composite, and the forming quality of the wound components and the pressure vessel, ultimately. When using 4251A4/B2 epoxy resin for wet winding pressure vessels, the choice of a 112 °C curing temperature will help improve the vessel's overall performance. This work could provide reliable experience and insight into the curing process analysis of pressure vessel manufacturing.
