ABSTRACT

Nisin (Ni) is a polypeptide bacteriocin produced by lactic streptococci (probiotics) that can inhibit the majority of gram-positive bacteria, and improve the growth performance of broilers, and exert antioxidative and anti-inflammatory properties. The present study investigated the potential preventive effect of Nisin on necrotic enteritis induced by Clostridium perfringens (Cp) challenge. A total of 288 Arbor Acres broiler chickens of 1-d-olds were allocated using 2 × 2 factorial arrangement into four groups with six replicates (12 chickens per replicate), including: (1) control group (Con, basal diet), (2) Cp challenge group (Cp, basal diet + 1.0 × 108 CFU/mL Cp), (3) Ni group (Ni, basal diet + 100 mg/kg Ni), and (4) Ni + Cp group (Ni + Cp, basal diet + 100 mg/kg Ni + 1.0 × 108 CFU/mL Cp). The results showed that Cp challenge decreased the average daily gain (ADG) of days 15 to 21 (P<0.05) and increased interleukin-6 (IL-6) content in the serum (P < 0.05), as well as a significant reduction in villus height (VH) and the ratio of VH to crypt depth (VCR) (P<0.05) and a significant increase in crypt depth (CD) of jejunum (P<0.05). Furthermore, the mRNA expressions of Occludin and Claudin-1 were downregulated (P<0.05), while the mRNA expressions of Caspase3, Caspase9, Bax, and Bax/Bcl-2 were upregulated (P<0.05) in the jejunum. However, the inclusion of dietary Ni supplementation significantly improved body weight (BW) on days 21 and 28, ADG of days 15 to 21 (P<0.05), decreased CD in the jejunum, and reduced tumor necrosis factor-α (TNF-α) content in the serum (P<0.05). Ni addition upregulated the mRNA levels of Claudin-1 expression and downregulated the mRNA expression levels of Caspase9 in the jejunum (P<0.05). Moreover, Cp challenge and Ni altered the cecal microbiota composition, which manifested that Cp challenge decreased the relative abundance of phylum Fusobacteriota and increased Shannon index (P<0.05) and the trend of phylum Proteobacteria (0.05

Necrotic enteritis (NE), a severe digestive disorder in broiler chickens caused by Clostridium perfringens (Cp), a gram-positive bacterium, is a widespread issue in the global poultry industry, leading to significant economic losses. Nisin (Ni), a polypeptide bacteriocin produced by probiotic lactic streptococci, has been found to enhance daily weight gain and feed intake, while also exhibiting inhibitory effects on gram-positive bacteria and anti-inflammatory properties. In this study, a NE infection model in broilers was established to examine the potential preventive effects of Ni. These results demonstrated that Cp challenge reduced growth performance, caused inflammatory responses and intestinal apoptosis, damaged intestinal morphology and barrier function, and was accompanied by changes in the composition of the gut microbiota. Dietary supplementation with Ni improved growth performance and protected intestine against Cp challenge-induced damage in broilers. As a result, Ni may be a potential safe and effective additive for NE prevention in broiler production.

ABSTRACT

The effects of maternal dietary energy and arginine level on embryonic development and serum lipid metabolism were investigated in this study. A 2 × 3 factorial experiment was conducted with six treatments represented by 10 replicates of eight Arbor Acre broiler breeder hens each. Diets fed from 40 to 50 weeks of age were formulated to contain two digestible arginine levels (9.6 g/kg and 14.5 g/kg) and three metabolic energy levels (10.08 MJ ME/kg, 11.88 MJ ME/kg, and 13.68 MJ ME/kg). Artificial insemination was used, and eggs collected from 50 weeks of hens' age were hatched. Embryonic growth, biochemical and endocrine indexes of embryonic serum and allantoic fluid were measured on different embryonic days (E). The results were as follows: Egg weight (E0, E11, E13) and embryonic weight (E12, E15) in the high-energy group (13.68 MJ ME/kg) were significantly decreased (p < 0.01), as were embryonic breast rate (E13, E15, E21), thigh rate (E13-E21) and liver rate (E15-E21). The reciprocal effects of arginine and energy were significant on breast rate (E11, E13, E17), thigh rate (E19, E21) and liver rate (E11, E19) of the embryo (p < 0.05). CHO (E13-E19), high-density lipoprotein (E13, E15, E21), low-density lipoprotein (E15, E19, E21), and blood glucose (E13) levels in embryonic serum decreased with the increase in maternal dietary energy level (p < 0.05), but triglyceride levels (E19, E21) showed the opposite result (p < 0.05). The levels of cholesterol and blood glucose in embryonic serum at E11 and urea nitrogen in allantoic fluid at E11-E15 were significantly decreased in the 14.5 g/kg arginine group (p < 0.01). With the increase in maternal dietary energy and arginine levels, embryonic serum nitric oxide synthases levels (E11, E15, E19) increased significantly (p < 0.01). The reciprocal effect of arginine and energy in maternal diets was significant on the embryonic serum high-density lipoprotein level at E21 (p < 0.05). Embryonic serum insulin levels at E13 were significantly elevated in the high-energy group (13.68 MJ ME/kg). The reciprocal effect of arginine and energy was significant on the embryonic serum growth hormone level (p < 0.01). Embryonic serum growth hormone levels were significantly reduced in the 14.5 g/kg arginine and 13.68 MJ/kg metabolic energy group (p < 0.01). In conclusion, maternal restricted feeding improved embryonic development and regulated lipid metabolism-related indices in embryonic serum. Maternal dietary addition of digestible arginine had a significant effect on lipid metabolism indices in embryos. There was a maternal effect of maternal dietary energy and arginine levels on embryo growth and development. The deposition of maternal nutrients affects the development of embryos.

ABSTRACT

OBJECTIVE: The objective of this study was to investigate the effects of different degrees of maternal dietary energy restriction on lipid deposition in embryonic tissues during the medium laying period (37 to 39 weeks) in Arbor Acres (AA) broiler breeders. METHODS: A single factor design was adopted, and 400 AA broiler breeders (20 weeks of age) with a similar weight were randomly allocated into four groups. The birds in the control group were fed a corn-soybean meal based diet, and those in trial groups were fed diets with 80%, 70%, and 50% energy levels of the basal diet. Incubated eggs from the medium laying period were collected. Samples of developing embryos at various stages were prepared for composition analysis. RESULTS: The embryo weight in the 80% energy group was higher than those of the other groups on embryonic day (E) 13, but at 21 E, they were significantly decreased with decreasing energy intake of the broiler breeders (p<0.05). Additionally, the levels of crude fat in tissues in the restriction groups were significantly decreased (p<0.05). The long axis and area of adipocytes in breast muscle, thigh muscle and the liver were significantly decreased (p<0.05) at 21 E in the 80%, 70%, and 50% energy groups. CONCLUSION: The effects of the 80% maternal dietary energy restriction energy affects egg production performance, egg quality, and nutrient deposition in egg weights, which then directly impacts on the developmental process of embryos, especially on fat utilization and deposition.

ABSTRACT

OBJECTIVE: The effects of maternal dietary energy levels on breast muscle fibre development in offspring of broiler breeders were investigated. METHODS: A total of 480 20-week-old Arbor Acres (AA) healthy female broiler breeders, with an average body weight of 2.33±0.01 kg, were randomly divided into 4 treatment groups with 6 replicates and 20 broiler breeders for each replicate and fed a corn and soybean meal diet with 100%, 80%, 70%, and 50% energy levels, respectively. Approximately 300 eggs per treatment were collected for incubation for 6 days. Then, 120 0-day-old female broilers at each energy level were randomly selected and divided into 6 replicates with 20 broilers for each replicate, with this experimental phase with the offspring lasting for 49 days. RESULTS: Compared with the 100% energy group, the breast muscle fibre diameter at embryonic day 21 in the 80% energy group was significantly reduced (p<0.05). In the 80% energy group, the muscle fibre density of the breast increased significantly (p<0.05) at embryonic days 15 and 21. The breast muscle fibre diameter of the offspring in each group was significantly decreased (p<0.05) on the 1st day. The breast muscle sarcomere length of the embryos in the 80% energy group was significantly higher (p<0.05) than those in the 70% and 50% energy groups. Compared with the 100% energy group, the expression of the myostatin gene in the offspring was significantly decreased (p<0.05). CONCLUSION: In conclusion, the effects of a maternal dietary energy level of 80% in this study were found to be optimal for breast muscle fibre development in offspring, which indicated that the metabolic energy level of AA broilers of 9.36 MJ/kg for the mid-term diet for laying eggs has a more practical significance.

ABSTRACT

OBJECTIVES: To assess the readability of printed education materials (PEMs) for patients with systemic lupus erythematosus (SLE) and to explore the perceptions of patients with SLE with different health literacy regarding the readability of PEMs. DESIGN: A mixed-methods study, including a cross-sectional survey and semistructured interviews. SETTING: The SLE PEMs were collected from 13 hospitals in China. The interviews were conducted in the Department of Rheumatology of a hospital in Hefei, China. PARTICIPANTS: In the cross-sectional survey, convenience sampling was used to select the Chinese SLE PEMs, with 20 PEMs included. In the qualitative study, the patients with SLE were divided into two groups based on their health literacy. Then, purposive sampling was used to select participants in each group, with 18 patients recruited. OUTCOME MEASURES: The readability of PEMs was assessed by the language analysis technology and the Chinese version of the Suitability Assessment of Materials (SAM-C) instrument. RESULTS: For text factors of readability, the mean Chinese language difficulty coefficient was 67.09±8.03, which indicates that the text of PEMs was difficult to read. For non-text factors, the mean SAM-C score was 45.62±9.51. Eight PEMs were rated not suitable, 12 were adequate and none were superior. In the interviews, eight categories were identified: information source, content, actionability, plain language, pictures, tables, numbers and layout. Patients with different health literacy had discrepant views on the detail of basic information, the necessity of question list, the location of functional pictures and the application of mathematical symbols. CONCLUSIONS: The readability of Chinese SLE PEMs does not perform well, and it is necessary to reduce the difficulty of words, shorten the length of sentences and improve the picture design and actionability. To develop PEMs tailored to patients' level of health literacy, patients' unique view of readability should be integrated into the design of PEMs.

ABSTRACT

The goal of this experiment was to examine effects of diets supplemented with exogenous inosine monophosphate (IMP) on the growth performance, flavor compounds, enzyme activity and gene expression of chicken. A total of 1,500 healthy, 1-day-old male 3-yellow chickens were used for a 52-d experimental period. Individuals were randomly divided into 5 groups (group I, II, III, IV, V) with 6 replicates per group, and fed a basal diet supplemented with 0.0, 0.05, 0.1, 0.2, and 0.3% IMP, respectively. There was no significant response to the increasing dietary IMP level in average daily feed intake (ADFI), average daily gain (ADG), and feed:gain ratio (F/G) (P ≥ 0.05). IMP content of the breast and thigh muscle showed an exponential and linear response to the increasing dietary IMP level (P < 0.05), the highest IMP content was obtained when the diet with 0.3% and 0.2% exogenous IMP was fed. There were significant effects of IMP level in diet on free amino acids (FAA) (exponential, linear and quadratic effect, P < 0.05) and delicious amino acids (DAA) (quadratic effect, P < 0.01) content in breast muscle. FAA and DAA content in thigh muscle showed an exponential and linear response (P < 0.05), and quadratic response (P < 0.01) to the increasing dietary IMP level, the highest FAA and DAA content was obtained when the diet with 0.2% exogenous IMP was fed. Dietary IMP supplementation had a quadratic effect on 5΄-NT and the alkaline phosphatase (ALP) enzyme activity in the breast muscle (P < 0.05), and the adenosine triphosphate (ATP) enzyme activity in the thigh muscles increased exponentially and linearly with increasing IMP level in diet (exponential effect, P = 0.061; linear effect, P = 0.059). Cyclohydrolase (ATIC) gene expression in thigh muscle had a quadratic response to the increasing dietary IMP level (P < 0.05), 0.2% exogenous IMP group had the highest (AMPD1) gene expression of the breast muscle and ATIC gene expression of the thigh muscle. These results indicate that dietary IMP did not affect the growth performance of chicken, the diet with 0.2 to 0.3% exogenous IMP is optimal to improve the meat flavor quality in chicken.

ABSTRACT

Interferons have attracted considerable attention due to their vital roles in the host immune response and low induction of antibiotic resistance. In this study, total RNA was extracted from spleen cells of chicken embryos inoculated with Newcastle disease vaccine, and the full-length chicken interferon-γ (ChIFN-γ) gene was amplified by RT-PCR. The full complementary DNA sequence of the ChIFN-γ gene was 495 bp long and was cloned into the prokaryotic expression vector pProEX™HTb . The plasmid was transformed into Escherichia coli DH5α and the expression of ChIFN-γ was induced by isopropyl ß-D-1-thiogalactopyranoside. Sodium dodecyl sulfate - polyacrylamide gel electrophoresis and Western blot results showed the expressed fusion protein had a molecular weight of approximately 18 kDa and was recognized by an anti-His mAb. Moreover, ChIFN-γ was found to demonstrate anti-viral activity in vitro. To test the in vivo function of ChIFN-γ in broilers under heat stress, a total of 100 broilers were randomly assigned to either a control group or a treated group, in which they were hypodermically injected with recombinant ChIFN-γ. Results demonstrated ChIFN-γ affects the messenger RNA expression levels of heat shock protein 70 (HSP70) in the heart and lung tissues, and decreases the concentration of HSP70 in serum. Therefore, we conclude recombinant ChIFN-γ can reduce heat stress to some extent in vivo.

ABSTRACT

The effects of maternal L-arginine supplementation on laying performance and the antioxidant capacity of broiler breeder hens, egg yolk, and their one-day-old offspring were investigated. In a 9 wk experiment, 210 60-week-old Arbor Acres healthy female broiler breeders were randomly divided into 5 treatments with 6 replicates of 7 females and fed a corn and soybean meal diet with 5 arginine levels (0.96%, 1.16%, 1.36%, 1.56%, and 1.76% digestible arginine). Laying performance and anti-oxidant capacity of broiler breeder hens, eggs, and offspring were evaluated. Digestible arginine level in the broiler breeder diet had a significant effect on the laying rate (linear and quadratic effect, P<0.0001). The highest laying rate was obtained when the diet with 1.36% digestible arginine was fed. There was a significant effect of digestible arginine level in the broiler breeder diet on the total antioxidant capacity (T-AOC) levels and methane dicarboxylic aldehyde (MDA) concentration in the broiler breeder serum, egg yolk and serum, and liver and breast of one-day-old offspring (linear and quadratic effect, P<0.05). The T-AOC level was highest and the MDA concentration lowest in all tissues when a diet with 1.36% digestible arginine was fed. No difference in glutathione peroxidase (GSH-PX) activity in the broiler breeder serum was observed. There were significant effects of digestible arginine level in the broiler breeder diet on the GSH-PX activity of the egg yolk (linear effect, P<0.01; quadratic effect, P<0.05) and serum, liver, and breast of one-day-old offspring (linear and quadratic effect, P≤0.01). The GSH-PX activity in all tissues measured in this experiment was highest when the dietary digestible arginine was 1.36%. These results indicate that the diet with 1.36% digestible arginine (1,972 mg/d) is optimal to satisfy the nutritional needs of a female broiler breeder during the late laying period.

ABSTRACT

This study was designed to determine the nutritional lipid requirement of Manchurian trout and to investigate the effects of lipid concentrations on the antioxidant status in larvae with experimental diets with different lipid levels. Oxidative stress differences between different organs and tissues were also assessed. Manchurian trout larvae were fed for 35 days and, during that period, growth and survival, the activity of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPX) and content of malondialdehyde (MDA) in viscera, muscle, gill and brain of four diets, lipid levels from 15 to 30%, and control treatment were measured. Growth rates were similar, but survival was low, between high and low dietary lipid levels. SOD activity was stimulated in viscera, muscle and brain in high lipid diets, but reduced in gills with increased lipid content. SOD was kept lower in the control group. GPX activity was inhibited in viscera and stimulated in gill, muscle and brain. CAT activities were enhanced by all treatments and showed the lowest values in the control. Lipid peroxidation of the diet was promoted in all organs, excluding the gill which showed no regular pattern. MDA content increased with increased dietary lipid levels in viscera, muscle and brain. Our results indicate that the most appropriate lipid requirement is probably 20-25% and a higher dietary level of lipids might induce oxidative stress in Manchurian trout larvae. The brain and gill were probably the most sensitive organs to oxidative damage.
