ABSTRACT

The systemic action of certain psychotropic drugs (haloperidol, penfluridol, trifluoperazine pimozide and verapamil) inhibitors of calcium-calmodulin-system, some biochemical and morphological parameters during the sexual differentiation of hypothalamus in neonatal (72 hours) male and androgenized female rats were studied. The hypothalamic weight in normal adult rats of both sexes not changed (39.7 +/- 3.79 vs 40.57 +/- 7.60 mg); however, verapamil increased the hypothalamic weight (table II), haloperidol and pimozide were the most effective (1.50 +/- 0.49 g and 2.00 +/- 0.11 g respectively). The testosterone administered to newborn females decreased the ovary development (46.3 +/- 4.5 mg. vs 23.1 +/- 2.7 mg), the haloperidol and pimozide partially reverted this effect.

ABSTRACT

Twenty schizophrenic patients were treated for a year with long term neuroleptic Penfluridol to test its antipsychotic and re-socializing effect. They underwent a clinical follow up and an evaluation of psychosocial aspects was also performed by a psychiatric social worker. Their social behaviour and reinsertion to work and family were studied by the use of specially designed scales. Clinical improvement and significant changes in all of the studied aspects of social behaviour were observed after a year of treatment. Also a reduction of treatment abandonment and of frequency of relapse through family handling was detected. Finally a number of preliminary observations on family structure and dynamics as well as changes determined on it by the treatment of the psychotic patient are summarized.

ABSTRACT

Con el objeto de evaluar el efecto antipsicotico y resocializador del neuroleptico de accion prolongada penfluridol, veinte pacientes esquizofrenicos fueron tratados durante un ano, y paralelamente al seguimiento clinico se efectuo una evaluacion de los aspectos psicosociales a cargo de una asistente social psiquiatrica. Se analizo la reinsercion del paciente en su medio familiar, su comportamiento social y su reincorporacion laboral, para lo que se disenaron instrumentos especificos. Despues de un ano de tratamiento se observo que junto con la mejoria clinica, se produjeron cambios muy significativos en todos los aspectos del comportamiento social estudiados. Se senala ademas, que el manejo familiar reduce el abandono del tratamiento y con ello la frecuencia de recaida.Se resumen, por ultimo,algumas observaciones preliminares sobre la estructura y dinamica y los cambios que ocurren en esta con el tratamiento del paciente psicotico

ABSTRACT

The difficulties of regular drug intake in long term treatments for psychotic patients gave rise to the need of using medicines at the longest possible intervals, facilitating thus adequate control and regularity. Penfluridol is a neuroleptic meeting that requirement: it is necessary only one dose per week. This paper reviews the results of Penfluridol in 26 patients (20 inpatients and 6 outpatients), ages between 17 and 54 with a mean of 36.8, sex feminine, and the following diagnoses: schizophrenia, paranoid: 14, simple: 8, hebephrenic: 3, catatonic: 1. The patients, divided in two groups of 13 each, had one oral dose a week, of between 10 and 100 mg, during 90 days. The first group took only Penfluridol, suppressing any other medicaments. The other group added Penfluridol to the prescriptions already in use. The results, as described in tables I and II, were evaluated according to 36 items. The general evaluation was positive with no negative biases. The side effects were scarce and temporary: insomnia in 7 cases during the first week, and extra-pyramidal symptoms in another 7 cases, that were controlled with antiparkinsonians. The conclusion is that Penfluridol is a valuable contribution to longterm treatments in psychoses.

ABSTRACT

Thirty female patients were studied while taking penfluridol, continuous neuroleptic antipsychotic medication given orally. These patients were hospitalized making observation and control adequate in 73.3% of the cases. Colateral effects that we observed were minimum and variable in nearly all the patients, and many of these effects are of little significance as they are observed with other neuroleptics.
