Background: Reports on the association between HTLV-2 infection and the development of diseases in the human host are rare, which has led the scientific community to believe that HTLV-2 is not an important etiological agent of lymphoproliferative or neurodegenerative disorders, which is the case for HTLV-1. In the present study, we demonstrated cases of fibromyalgia in HTLV-1 carriers and, in an unprecedented finding, in two patients with confirmed HTLV-2 infection. Methods: A total of 957 individuals visited the Virology Laboratory at the Federal University of Pará for screening and confirmation tests for HTLV-1/2 infection. Individuals with confirmed HTLV-1 and HTLV-2 infection were clinically evaluated for signs and symptoms associated with infection. Results: Sixty-nine individuals (7.2%) were identified as positive for HTLV infection, with 56 confirmed cases of HTLV-1 infection (5.9%), 12 cases of HTLV-2 infection (1.2%) and one case classified as undetermined (0.1%). Sixteen (23.2%) of these patients presented with rheumatological signs and complained of diffuse pain throughout the body; 12 of whom were infected by HTLV-1 (75%) and 4 were infected by HTLV-2 (25%). After anamnesis and careful evaluation, four patients were diagnosed with fibromyalgia, two of whom were infected by HTLV-1 (16.7%; 2/12) and two by HTLV-2 (50%; 2/4). The clinical follow-up and laboratory analysis results are reported in detail in this paper. Conclusion: Considering the clinical cases presented herein as the first reports of patients with HTLV-2 infection with clinical symptoms of fibromyalgia, the importance of further studies on the pathogenicity of HTLV-2, similar to what have already been performed for HTLV-1, is highlighted. Our results also confirm previous evidence of an association between HTLV-1 infection and fibromyalgia.

This study aimed to describe the prevalence of HTLV-1/2 in quilombola communities in the state of Pará and investigate the possible sociodemographic risk factors associated with the infection, as well as to trace the occurrence of the familial transmission of the virus. A total of 310 individuals living in eight quilombos located in the state of Pará (northern Brazil) were investigated for the presence of anti-HTLV-1/2 antibodies using an enzyme-linked immunosorbent assay (ELISA), and positive samples were confirmed using Western blot and/or real-time quantitative polymerase chain reaction (qPCR). Participants answered a questionnaire about sociodemographic aspects and risk factors for infection. Anti-HTLV-1/2 antibodies were detected in two individuals (one man and one woman), for an overall seroprevalence of 0.65%. Both individuals belonged to the community of São José de Icatú. The search for intrafamilial infection identified two other infected women, which increased the general prevalence of HTLV-1 among the Icatú to 6.25% (4/64). Western blot and qPCR confirmed their HTLV-1 infection, and phylogenetic analysis demonstrated that the isolates were of the cosmopolitan subtype and transcontinental subgroup. Epidemiological investigation of the cases revealed that the three women, at some point in their lives, had a relationship with the infected male individual. HTLV-1 is transmitted silently between individuals in the community of São José de Icatú with a present or past family relationship, stressing the need for screening and laboratory diagnosis to prevent further dissemination of the virus and surveillance of disease emergence.

Patients with focal segmental glomerulosclerosis (FSGS) who are refractory to drug treatment may present progressive loss of kidney function, leading to chronic kidney disease stage 5 under dialysis treatment. The safety of systemic administration of bone marrow-derived mononuclear cells (BMDMCs) has been shown in different preclinical models of kidney diseases. However, to date, no study has evaluated the safety and biodistribution of BMDMCs after infusion in renal arteries in patients with FSGS. We used a prospective, non-randomized, single-center longitudinal design to investigate this approach. Five patients with refractory FSGS and an estimated glomerular filtration rate (eGFR) between 20 and 40 ml/min/1.73 m2 underwent bone marrow aspiration and received an arterial infusion of autologous BMDMCs (5 × 107) for each kidney. In addition, BMDMCs labeled with technetium-99m (99mTc-BMDMCs) were used to assess the biodistribution by scintigraphy. All patients completed the 270-day follow-up protocol with no serious adverse events. A transient increase in creatinine was observed after the cell therapy, with improvement on day 30. 99mTc-BMDMCs were detected in both kidneys and counts were higher after 2 hr compared with 24 hr. The arterial infusion of BMDMCs in both kidneys of patients with FSGS was considered safe with stable eGFR at the end of follow-up. This trial is registered with NCT02693366.

This study introduces a novel method for the quantification of malachite green (MG), a pervasive cationic dye, in surface water by synergizing multiphase electroextraction (MPEE) with digital image analysis (DIA) and partial least square discriminant analysis. Aimed at addressing the limitations of conventional DIA methods in terms of quantitation limits and selectivity, this study achieves a significant breakthrough in the preconcentration of MG using magnesium silicate as a novel sorbent. Demonstrating exceptional processing efficiency, the method allows for the analysis of 10 samples within 20 min, exhibiting remarkable sensitivity and specificity (over 0.95 and 0.90, respectively) across 156 samples in both training and test sets. Notably, the method detects MG at low concentrations (0.2 µg L-1) in complex matrices, highlighting its potential for broader application in environmental monitoring. This approach not only underscores the method's cost-effectiveness and simplicity but also its precision, making it a valuable tool for the preliminary testing of MG in surface waters. This study underscores the synergy among MPEE, DIA, and chemometric tools, presenting a cost-efficient and reliable alternative for the sensitive detection of water contaminants.

Since hop secondary metabolites have a direct correlation with the quality of beer and other hop-based beverages, and the volatile fraction of hop has a complex composition, requiring effective separation, here we explore the application of headspace solid-phase microextraction as a sample preparation method, coupled with comprehensive two-dimensional gas chromatography-mass spectrometry (GC×GC-MS) analysis. The methodology involved the use of a DVB/PDMS fibre with 500 mg of hop cone powder, extracted for 40 min at 50 °C, for both GC-MS and GC×GC-MS. The varieties Azacca, Cascade, Enigma, Loral, and Zappa were studied comprehensively. The results demonstrate that GC×GC-MS increases the number of peaks by over 300% compared to classical GC-MS. Overall, 137 compounds were identified or tentatively identified and categorised into 10 classes, representing between 87.6% and 96.9% of the total peak area. The composition revealed the highest concentration of sesquiterpene hydrocarbons for Enigma, whilst Zappa showed a relatively significant concentration of monoterpene hydrocarbons. Principal component analysis for all compounds and classes, along with hierarchical cluster analysis, indicated similarities between Zappa and Cascade, and Azacca and Loral. In conclusion, this method presents an optimistic advancement in hop metabolite studies with a simple and established sample preparation procedure in combination with an effective separation technique.

Objective: The aim of this study was to assess the effects of accelerated tooth movement (ATM) methods: corticopuncture (CP), photobiomodulation (PBM), and their combination (CP + PBM) by evaluating tooth displacement, alveolar bone changes, and molecular and cellular response compared with conventional induced tooth movement. Materials and methods: Tooth movement and bone changes were evaluated on days 1, 3, and 7 (9 animals per time point) using microtomography, histological, and immunohistochemical evaluation, at compression and tension sites. CP groups received two perforations in the palate and one mesial to the molars. PBM was performed using GaAlAs diode laser applied every other day for 7 days (λ = 808 nm, 100 mW) in two points for 15 sec/point and total energy of 3 J. Results: Tooth movement was significantly increased in all three ATM groups after 7 days compared with the control group (mean 0.24 mm) by 27% PBM (0.31 mm), 45% CP (0.35 mm), and 57% CP + PBM (0.38 mm) (p < 0.05). At the compression side, all ATM groups showed significant decrease in bone density on day 3 (p < 0.05) and significant less bone volume on day 7 compared with Control (p < 0.05). At the tension side, PBM group showed a significant increase in bone density and volume on day 3 (p < 0.05). Immunohistochemistry analysis showed that at the compression side, tartrate-resistant acid phosphatase-positive cells, RANKL, and tumor necrosis factor-alpha expression were highly marked of the PBM and the combined method groups (p < 0.05). PBM and CP + PBM groups showed a significant increase in expression Runt-related transcription factor 2 and osterix (p < 0.05) at the tension side. Conclusions: All ATM groups showed increase on tooth displacement with CP + PBM group showing greatest tooth displacement. CP method appears to stimulate bone catabolism, PBM has more effect on bone formation, and the combined method showing a synergistic effect on bone remodeling.

Chronic kidney disease (CKD) has long been recognized as a state of progressive decline in renal function. Morbidity and mortality are well correlated to the stage of renal function decline. Approximately one million deaths are estimated to be related to CKD worldwide. They are mostly associated with cardiovascular disease as a result of concurrent hypertension, accelerated atherosclerosis, and volume overload. Even with the best current treatment, disease progression is the general rule with a small fraction who reach CKD stage 5 requiring kidney transplantation or dialysis. Transplant patients show substantial reductions in mortality and cardiovascular events, as well as improvements in quality of life. However, the capacity of health systems to deliver kidney transplantation varies worldwide with worse indicators in low-income countries. Consequently, exploring novel and better therapeutic options for CKD is mandatory. Cell-based therapy is a promising strategy for treating CKD in preclinical models, and several clinical trials involving kidney disease exhibit a favorable safety profile. This chapter aims to provide an overview of CKD and the recent results of clinical trials of cell therapy in kidney diseases.

Gas chromatography-mass spectrometry (GC-MS), physicochemical and microbiological analyses, sensory descriptive evaluation, and multivariate analyses were applied to evaluate the efficiencies of microfiltration and pasteurization processes during the shelf life of beer. Samples of microfiltered and pasteurised beer were divided into fresh and aged groups. A forced ageing process, which consisted of storing fresh samples at 55° C for 6 days in an incubator and then keeping them under ambient conditions prior to analysis, was applied. Physicochemical analysis showed that both microfiltered and pasteurised samples had a slight variation in apparent extract, pH, and bitterness. The samples that underwent heat treatment had lower colour values compared with those that were microfiltered. Chromatographic peak areas of vicinal diketones increased in both fresh and aged samples. The results of the microbiological analysis revealed spoilage lactic acid bacteria (Lactobacillus) and yeasts (Saccharomyces and non-Saccharomyces) in fresh microfiltered samples. In the GC-MS analysis, furfural, considered by many authors as a heat indicator, was detected only in samples that underwent forced ageing and not in samples that were subjected to thermal pasteurisation. Finally, sensory analysis found differences in the organoleptic properties of fresh microfiltered samples compared with the rest of the samples.

In Brazil, the coronavirus disease 2019 (COVID-19) epidemic spread rapidly in a heterogeneous way, mainly due to the different socioeconomic and behavioral characteristics of different regional populations and different evaluation periods. We performed a cross-sectional study including 1,337 individuals (first wave = 736/second wave = 601) after the first two waves of COVID-19 in the city of Belém, the capital of the state of Pará. The detection of IgG anti-SARS-CoV-2 antibodies was performed using an enzyme-linked immunosorbent assay test followed by statistical analysis using the RStudio program. Our results showed an increase in the seroprevalence (first wave= 39.1%/second wave= 50.1%) of anti-severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) IgG antibodies in the population of Belém from the first to the second pandemic wave. Advanced age, primary or secondary education level, lack of social isolation, and a low frequency of protective mask use were considered risk factors for SARS-CoV-2 infection during the first wave compared to the second wave. This study is one of the firsts to provide important information about the dynamics of virus circulation and the groups vulnerable to exposure in the two major periods. Our data emphasize the socioeconomic characteristics of the affected population and that nonpharmacological prevention measures are crucial for combating the pandemic.

The Brazilian cerrado is considered one of the most critical biomes in the world. Araticum (Annona crassiflora Mart.) is a native plant from the Brazilian cerrado, abundant in nutrients and highly energetic. This study aimed to obtain chemical fingerprints of different parts of the araticum fruit, i.e. pulp, peel, and seed. Extracts from these three parts were prepared using different solvents (ethanol, water, and mixtures of ethanol and water) and later analyzed by paper spray ionization mass spectrometry. In general, ethanol extracted more metabolites than the other solvents. The chemical profiles varied according to the fruit part, geographic location, and extractor solvent. Among the metabolites, acetogenins (62.3%) and alkaloids (20.7%) predominated. Principal component analyses revealed that the samples were grouped according to the fruit part, regardless of the extractor solvent used. Araticum shows remarkable potential due to the beneficial properties of the metabolites for human health. The insertion of araticum in the human diet is still underexplored but is a promising alternative.

The Specialty Coffee Association (SCA) sensory analysis protocol is the methodology that is used to classify specialty coffees. However, because the sensory analysis is sensitive to the taster's training, cognitive psychology, and physiology, among other parameters, the feasibility of instrumental approaches has been recently studied for complementing such analyses. Spectroscopic methods, mainly near infrared (NIR) and mid infrared (FTIR-Fourier Transform Infrared), have been extensively employed for food quality authentication. In view of the aforementioned, we compared NIR and FTIR to distinguish different qualities and sensory characteristics of specialty coffee samples in the present study. Twenty-eight green coffee beans samples were roasted (in duplicate), with roasting conditions following the SCA protocol for sensory analysis. FTIR and NIR were used to analyze the ground and roasted coffee samples, and the data then submitted to statistical analysis to build up PLS models in order to confirm the quality classifications. The PLS models provided good predictability and classification of the samples. The models were able to accurately predict the scores of specialty coffees. In addition, the NIR spectra provided relevant information on chemical bonds that define specialty coffee in association with sensory aspects, such as the cleanliness of the beverage.

Genetic variations in components of the immune response seem to be an important factor that contributes to the manifestation of symptoms of some diseases related to HTLV-1 infection. Nerve growth factor (NGF) and the p75 neurotrophin receptor (p75NTR) are related to the maintenance of neurons and the activation of the immune response. In this study, we evaluated the association of the NGF -198C/T, NGF Ala35Val, and p75NTR Ser205Leu polymorphisms with HTLV-1 infection and plasma cytokine levels in 166 samples from individuals infected with HTLV-1 (59 symptomatic and 107 asymptomatic). The genotyping and quantification of the proviral load were performed by real-time PCR, and cytokine levels were measured by ELISA. The NGF -198C/T and NGF Ala35Val polymorphisms were not associated with HTLV-1 infection. The frequency of the Ser/Leu genotype of p75NTR Ser205Leu was more frequent in the control group (p = 0.0385), and the Ser/Leu genotype and allele Leu were more frequent among the asymptomatic (p < 0.05), especially with respect to the HTLV-1-associated myelopathy (HAM) group (p < 0.05). The symptomatic showed a higher proviral load and higher TNF-α and IL-10 levels (p < 0.05). Asymptomatic carriers of the Ser/Leu genotype (p = 0.0797) had lower levels of proviral load and higher levels of TNF-α (p = 0.0507). Based on the results obtained, we conclude that the p75NTR Ser205Leu polymorphism may be associated with reduced susceptibility to HTLV-1 infection, a lower risk of developing symptoms, including HAM, and better infection control.

A recent case of contamination of some batches of a Brazilian beer brand with diethylene glycol (DEG) had great repercussion, resulting in at least seven deaths. In this article, a direct method was developed for the rapid detection of DEG in beer samples based on portable near-infrared spectroscopy combined with partial least squares discriminant analysis (PLS-DA). The discriminant model was built with 100 uncontaminated beer samples and 100 samples containing DEG in a concentration range between 10 and 1000 mg L-1, totalizing 200 samples of different brands and styles. The method was validated by estimating figures of merit, such as false positive and false negative rates, sensitivity, specificity, accuracy, accordance, and concordance. The decision limit (CCα) of the method was 52 mg L-1 and the detection capability (CCß) was 106 mg L-1. This method does not consume reagents/solvents and can be suitable for the beer industry quality control or forensic investigations.

The devastating effect of health system overload was observed after cases of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) peaked in April 2020 in Belém, a capital metropolis in the Brazilian Amazon. Our results showed a high seroprevalence (39.24%) of anti-SARS-CoV-2 IgG antibodies among the population of the capital of the state of Pará after the first wave of the pandemic. Old age, mixed race, a high school education level, low income and contact with infected individuals are risk factors and may be driving seropositivity and exposure to SARS-CoV-2 in this population. This study is one of the first to provide important information to understand the socioeconomic inequalities, behavioral characteristics and viral transmission dynamics associated with the risk of SARS-CoV-2 infection in the capital of the state of Pará, northern Brazil.

OBJECTIVES: The emergence of SARS-CoV-2 and its pandemic spread generated serious concern about the impact of the infection on vulnerable indigenous populations of the Brazilian Amazon. Thus, this study aimed to perform a seroepidemiological survey of anti-SARS-CoV-2 antibodies in those populations. SETTING: Six indigenous ethnic groups living in the State of Pará (Northern Brazil) were investigated. The villages of Xikrin do Bacajá, Assurini, Araweté, Parakanã, Munduruku and Kararaô were visited from October 2020 to January 2021. DESIGN AND PARTICIPANTS: We performed a cross-sectional study to investigate the prevalence of anti-spike (S1) IgG antibodies. Plasma was tested for the presence of anti-SARS-CoV-2 IgM and IgG antibodies using two assays (a lateral flow rapid test and an ELISA). A total of 1185 individuals of both sexes were enrolled in the study. RESULTS: The prevalences of IgM and IgG antibodies were 6.9% and 68.1%, respectively, ranging from 0% to 79.6%, with significant differences (p<0.001) between age groups in three communities (Araweté, Xikrin and Munduruku) and a virulence rate of 0.86%. The overall IgG prevalence obtained by rapid tests and ELISAs were similar, and the agreement of the results between the two tests was 80%, which was classified as good (kappa=0.4987; p<0.001; sensitivity of 82.1% and specificity of 71.6%). Herd immunity was probably attained, similar to that found in other communities of the Amazon. CONCLUSIONS: SARS-CoV-2 spread rapidly among the indigenous populations investigated, but it had a low mortality rate. It is necessary to expand serological investigations to other communities in the Amazon region of Brazil.

Spectrofluorimetry combined with multiway chemometric tools were applied to discriminate pure Aroeira honey samples from samples adulterated with corn syrup, sugar cane molasses and polyfloral honey. Excitation emission spectra were acquired for 232 honey samples by recording excitation from 250 to 500 nm and emission from 270 to 640 nm. Parallel factor analysis (PARAFAC), partial least squares discriminant analysis (PLS-DA), unfolded PLS-DA (UPLS-DA) and multilinear PLS-DA (NPLS-DA) methods were used to decompose the spectral data and build classification models. PLS-DA models presented poor classification rates, demonstrating the limitation of the traditional two-way methods for this dataset, and leading to the development of three-way classification models. Overall, UPLS-DA provided the best classification results with misclassification rates of 4% and 8% for the training and test sets, respectively. These results showed the potential of the proposed method for routine laboratory analysis as a simple, reliable, and affordable tool.

HTLV-1/2 infection is endemic in Indigenous peoples of the Americas. Its origin is attributed to the migratory flow of Amerindian ancestral peoples. The present study aimed to investigate the seroprevalence of HTLV-1/2 infection in Indigenous peoples of the Brazilian Amazon. A total of 3350 Indigenous people belonging to 15 communities were investigated. The investigation was performed using serological (ELISA), molecular (qPCR) and confirmatory (Western blot and/or Inno-Lia) tests to detect and differentiate the infection. The seroprevalence was 8.3% for HTLV-1/2 infection, with 0.1% of individuals seropositive for HTLV-1 and 8.1% for HTLV-2. The prevalence of infection was statistically higher in women (10.1%) than in men (6.5%) (p = 0.0002). This female predominance was observed in all age groups; in females the prevalence was significant from 41 years old (p < 0.0001) and in males from 51 years old (p < 0.0001). Here, we present a prevalence of HTLV-1/2 among Indigenous peoples of the Brazilian Amazon. The endemic infection in these groups must reflect the different epidemiological profiles observed in these peoples, such as sexual transmission through rejection of condom use, breastfeeding, especially in cases of cross-breastfeeding, and the high rate of pregnancy in the villages.

In this work, a new large-volume multiphase, multi-sample electroextraction device was developed and applied to selectively extract malachite green (MG) from water samples. This device was easily constructed with ordinary materials and capable of extracting ten samples simultaneously, obtaining MG preconcentrated on a solid support, to fit into a pipette tip. A multi-well plate was applied to extract MG from aquaculture water samples, and the extracts containing the desorbed MG were analysed by LC-DAD and LC-MS/MS. The signals from both detectors were used in two independent validation procedures. Linearity, matrix effect, selectivity, precision, trueness, and limits of detection and quantification were all evaluated. For both detectors, linearity was demonstrated in the range of 0.5-5 µg L-1 (R2 > 0.98). Matrix effect was insignificant for LC-DAD only, and the average preconcentration factor was about 60 times. Recoveries ranged from 94 to 113% for LC-DAD and 95-115% for LC-MS/MS analysis. ANOVA was applied to estimate the standard deviation under repeatability (6.96-8.61% for LC-DAD and 5.98-7.41% for LC-MS/MS) and within-reproducibility (6.96-8.61% for LC-DAD and 6.56-7.41% for LC-MS/MS) conditions. The limits of detection and quantification for LC-MS/MS analysis were 4.29 and 28.74 ng L-1, respectively, while, for LC-DAD, these limits were 14.29 and 95.81 ng L-1, respectively. The results demonstrated that the developed method was suitable for determining MG in water samples, and the large-volume multiphase, multi-sample electroextraction device proved to be a powerful sample preparation technique to obtain high clean-up and large preconcentration levels, which are of paramount importance for environmental applications.

Este artigo apresenta processos formativos que constituíram estratégias para a implementação da Política Nacional de Educação Popular em Saúde (PNEPS) no Sistema Único de Saúde (SUS) e foram desenvolvidos com a perspectiva pedagógica da Educação Popular (EP), conforme fundamentada por Paulo Freire. São eles: o Curso de Educação Popular em Saúde para Agentes Comunitários e de Vigilância em Saúde (EdPopSUS), o Projeto de Pesquisa e Extensão "Vivências de Extensão em Educação Popular e Saúde no SUS" (Vepop-SUS) e o Curso de Formação Histórica e Política para Estudantes das Áreas da Saúde (FHP). Apontaram-se contextualizações e detalhamentos metodológicos dessas experiências, bem como aproximações, complementariedades e distanciamentos entre elas. Por meio das dimensões e dos enfoques explicitados, indica-se que a EP configura um caminho teórico e metodológico potente para a formação em saúde na perspectiva do bem viver e da emancipação humana. This article presents the educational processes that constituted strategies for the implementation of the National Policy for Popular Education in Health in the Brazilian National Health System (SUS) and were developed with the pedagogical perspective of Popular Education (PE), as substantiated by Paulo Freire. These are namely: the Popular Education Course on Health for Community and Health Surveillance Agents (EDPOPSUS), the Research and Extension Project "Extension Experiences in Popular Education and Health in SUS" (VEPOP-SUS) and the Historical and Political Education Course for Students of the Health Care Area (FHP). Contextualization and methodological details of these experiences were pointed out, as well as approximations, complementarities and differences between them. Through explicit dimensions and approaches, it is indicated that the PE constitutes a powerful theoretical and methodological path for health education from the perspective of good living and human emancipation. Objetivo: apresentar à organização e funcionamento dos Conselhos Distritais de Saúde (CDS) de João Pessoa-Paraíba-Brasil, a participação social em saúde, suas dificuldades e potencialidades, particularmente aquelas observadas em espaços institucionalizados de controle social. Método: estudo qualitativo e descritivo utilizando observação participante, pesquisa documental e entrevistas individuais. Resultado: identificou-se o número, distribuição e organização dos CDS; número e periodicidade de reuniões; pautas abordadas; estratégias de mobilização; e composição; dificuldades e potencialidades. Conclusão: os resultados mostram que há diferentes dinâmicas de funcionamento, resultantes de níveis distintos de responsabilização e envolvimento de gestores, usuários e trabalhadores. Todos os CDS apresentam dificuldades e potencialidades.


Objective: To present to the organization and functioning of the District Health Councils (CDS) of João Pessoa­Paraíba­Brazil, social participation in health, its difficulties and potentialities, particularly those observed in institutionalized spaces of social control. Method: qualitative and descriptive study using participant observation, documentary research and individual interviews. Result: the number, distribution and organization of CDS were identified; number and frequency of meetings; guidelines addressed; mobilization strategies; and composition; difficulties and potentialities. Conclusion: the results show that there are different dynamics of functioning, resulting from different levels of accountability and involvement of managers, users and workers. All CDS present difficulties and potentialities.
