Using anti-neurofilament H non-phosphorylated antibodies (SMI-32) as markers for the neuronal maturation level and Y channel responsible for motion processing, we investigated early postnatal development of the primary visual areas 17 and 18 in cats aged 0, 10, 14, and 34 days and in adults. Two analyzed parameters of SMI-32-immunolabeling were used: the total proportion of SMI-32-labeling and the density of labeled neurons. (i) The developmental time course of the total proportion of SMI-32-labeling shows the general increase in the accumulation of heavy-chain neurofilaments. This parameter showed a different time course for cortical layer development; the maximal increment in the total labeling in layer V occurred between the second and fifth postnatal weeks and in layers II-III and VI after the fifth postnatal week. In addition, the delay in accumulation of SMI-32-labeling was shown in layer V of the area 17 periphery representation during the first two postnatal weeks. (ii) The density of SMI-32-labeled neurons decreased in all layers of area 18, but was increased, decreased, or had a transient peak in layers II-III, V, and VI of area 17, respectively. The transient peak is in good correspondence with some transient neurochemical features previously revealed for different classes of cortical and thalamic neurons and reflects the time course of the early development of the thalamocortical circuitry. Some similarities between the time courses for the development of SMI-32-labeling in areas 17/18 and in A- and C-laminae of the LGNd allow us to propose heterochronous postnatal development of two Y sub-channels.

The paper presents a method of analysis of cell populations that combines the use of normalized spatial coordinates of the neurons with the morphometric criteria of their evaluation. These algorithms were applied to check the heterogeneity of apopulation of neurons Y-conducting channel in cat at the level of the lateral geniculate body (LGB). As a specific marker of Y-neurons, SMI-32 antibodies were used. Evaluated The dynamics of the distribution of the number of cells and the orientation of their soma within each layer and mediolaterally along the length of LGB dorsal nucleus (LGBDN). Among the SMI-32-positive neurons, the existence of at least two populations was detected differing in number, orientation and distribution of the soma in different layers of LGBDN. The heterogeneity of Y-neuron population in LGBDN detected in this study is consistent with the earlier electrophysiological data. We believe that the described algorithm for neuronal analysis may be successfully applied to study not only LGB, but also other extensive structures of the brain, including those having laminar organization.

RESUMO

Using the technique of histochemical detection of enzyme acetylcholinesterase (AChE), the existence of the loci with high enzyme activity in the lateral nucleus of the lateral posterior thalamic complex was demonstrated in 2-week-old kittens (n = 4) in contrast to the kittens aged 14 weeks (n = 4). These loci were located opposite similar loci in the medial nucleus of the lateral posterior thalamic complex. Possible link between identified AChE-positive loci and the organization of thalamo-cortical and cortico-thalamic projections is discussed.

To examine the effect of rhythmical light stimulation on postnatal development of the visual system, the formation of Meynert cells was studied in area 17 and posteromedial suprasylvian area (PMLS) of kittens reared under the conditions of flickering light stimulation (15 Hz frequency). Profile area of neuronal cell bodies and cytochrome oxidase (CO) activity level were measured in frontal sections of the visual cortex in control (n = 6) and stimulated (n = 6) kittens. Meynert cells located in area 17 and PMLS demonstrated an approximately 37% increase in CO activity in the stimulated animals. At the same time, the profile area of Meynert cell bodies in PMLS was decreased by 20% as compared to that in normal animals. The presence of functional disturbances of Y-conducting visual channel and the possibility of binocular vision suppression in the stimulated animals are discussed.

To examine the effect of an artificial rhythmic light stimulation on the development of structural and functional organization of Y neurons of cat visual system in the ontogenesis, the distribution of the neurons immunopositive to SMI-32 antibodies was studied in lateral geniculate nucleus (LGN) and posteromedial suprasylvian area (PMLS). Laminar distribution of SMI-32-positive neurons and neuronal body profile area were analyzed in intact animals (n = 4) and in kittens (n = 4) grown under conditions of rhythmic light stimulation with 15 Hz frequency for 4 months. In light-stimulated animals, changes in laminar distribution of immunopositive neurons were detected in both LGN (decline in the percentage of the immunopositive cells in C(M) layer) and in PMLS area (decrease in cell count in layer V). Morphometric analysis has shown the significant reduction of cell body profile area in immunopositive neurons in light-stimulated kittens only in layers III and V of PMLS area. The data obtained suggest that Y channel functional disturbances in light-stimulated animals are caused by the structural and metabolic changes detected in Y neurons.

RESUMO

The aim of the present investigation was to develop an algorithm of superposition of retinotopic map of PMLS (posterior medial part of lateral suprasylvian sulcus) visual area on a 3-D model of brain frontal slices. A step by step description of the algorithm is presented. The suggested algorithm may be used for the solution of the various tasks of the analysis of the organization of interneuronal connections.

Development of axonal connections between cat primary visual area 17 and visual motion processing center was studied to investigate cortico-cortical connection plasticity in ontogenesis as affected by an experimental modification of visual environment (flickering light stimulation). By using a retrograde axonal labeling by horseradish peroxidase, a distribution of initial neurons in area 17 that send afferent projections to PMLS (posterior medial part of lateral suprasylvian sulcus) was analyzed. Sixteen 5-week-old and 12-14-week-old kittens, than were reared in normal visual environment or were subjected to a flickering light of 15 Hz frequency, were examined. It was shown that session stimulation by flickering light led to an impairment of normal development of regular organization of the connections between these visual areas including the decrease of labeled surface area and labeled initial neuron density in area 17. The data obtained elucidate the structural bases of cortical mechanisms that underlie motion processing disturbances in kittens stimulated by a flickering light.
