There is a growing quest for an ideal biomaterial that shows appropriate cellular response and is not susceptible to microbial adhesion. In this study, commercial grade II titanium was submitted to RF/DC plasma surface modification at 2.2 mbar, using gas mixtures of argon, nitrogen, and oxygen at proportions 4:1:2 and 4:1:3. The surfaces were physically and chemically characterized. In order to evaluate bacterial response, the surfaces were exposed to Staphylococcus epidermidis. Oxynitrided samples, although having a higher roughness as compared with untreated samples, exhibited lower bacterial growth. This observation is probably due to the formation of different crystalline phases of nitrides and oxides caused by plasma treatment. The surface with highest contact angle and highest surface tension showed lower bacterial adhesion. These results were confirmed by scanning electron microscopy. The role of nitrogen in reducing bacterial adhesion is clear when this material is compared with untreated titanium, on which only an oxide film is present.

Chronic wounds are those that remain in a chronic inflammatory state and fail to follow normal healing process. Infection is one of the most important causes of chronicity. A frequent pathogen isolated from chronic infections is Pseudomonas aeruginosa; refractory to therapy and host immune attack in its biofilm phenotype. Lactobacillus plantarum cultures supernatants (LAPS) interfere with its pathogenic capacity. In addition, LAPS showed bacteriostatic and bactericide properties and is neither cytotoxic nor an inductor of necrosis-apoptosis. LAPSs chemical composition was determined; allowing us to propose a correlation between its constituents and their biological activity. This article shows a pharmaceutical dosage form designed by using LAPS as an API with pro-healing activity and its quality control. Pharmacotechnical and anti-microbial assays were adapted to demonstrate that the vehicle used does not modify LAPS activities. Selected formulation (F100) showed fair mechanical and technological properties. From the in vitro release assays was found an adequate release from the carrier matrix and maintains its anti-pathogenic activity for 6 months. We propose F100 for chronic wounds treatment. The use of harmless bacteria by-products, such as LAPS, to antagonize infectious pathogens that have ability to form biofilm is an efficient and economic approach to treat infected chronic wounds.

CONTEXT: The local communities living in the Brazilian Caatinga biome have a significant body of traditional knowledge on a considerable number of medicinal plants used to heal several maladies. OBJECTIVE: Based on ethnopharmacological data, this study screened 23 aqueous plant extracts against two well-known models of biofilm-forming bacteria: Staphylococcus epidermidis and Pseudomonas aeruginosa. MATERIALS AND METHODS: Crystal violet assay and scanning electron microscopy (SEM) were used to evaluate the effect of extracts on biofilm formation and measurements of the absorbance at 600 nm to assess bacterial growth. Selected extracts were investigated regarding the cytotoxicity by MTT assay using mammal cells and the qualitative phytochemical fingerprint by thin layer chromatography. RESULTS: Harpochilus neesianus Mart. ex Nees. (Acanthaceae) leaves, Apuleia leiocarpa Vogel J. F. Macbr. (Fabaceae), and Poincianella microphylla Mart. ex G. Don L. P. Queiroz (Fabaceae) fruits showed non-biocidal antibiofilm action against S. epidermidis with activities of 69, 52, and 63%, respectively. SEM confirmed that biofilm structure was strongly prevented and that extracts promoted overproduction of the matrix and/or bacterial morphology modification. Poincianella microphylla demonstrated toxicity at 4.0 mg/mL and 2.0 mg/mL, A. leiocarpa presented toxicity only at 4.0 mg/mL, whereas H. neesianus presented the absence of toxicity against Vero cell line. Preliminary phytochemical analysis revealed the presence of flavonoids, terpenoids, steroids, amines, and polyphenols. DISCUSSION AND CONCLUSIONS: This work provides a scientific basis which may justify the ethnopharmacological use of the plants herein studied, indicating extracts that possess limited mammal cytotoxicity in vitro and a high potential as a source of antibiofilm drugs prototypes.

INTRODUCTION: Ilex paraguariensis A. St. Hil. (mate) is known in several South American countries because of the use of its leaves in stimulant herbal beverages. High saponin contents were reported in mate leaves and unripe fruits that possess a dissimilar composition. Two LC-UV methods previously reported for mate saponins assay focused on mate leaves and the quantification of the less polar saponin fraction in mate fruits. OBJECTIVE: To develop and validate a LC-UV method to assay the total content of saponins in unripe mate fruits and characterise the chemical structure of triterpenic saponins by UPLC/Q-TOF-MS. METHODOLOGY: From unripe fruits of mate a crude ethanolic extract was prepared (EX40) and the mate saponin fraction (MSF) purified by solid phase extraction. The LC-UV method was validated using ilexoside II as external standard. UPLC/Q-TOF-MS was adjusted from the LC-UV method to obtain the fragmentation patterns of the main saponins present in unripe fruits. RESULTS: Both LC-UV and UPLC/Q-TOF-MS methods indicate a wide range of Ilex saponins polarity. The ilexoside II and total saponin content of EX40 were 8.20% (w/w) and 47.60% (w/w), respectively. The total saponin content in unripe fruits was 7.28% (w/w). The saponins present in MSF characterised by UPLC/Q-TOF-MS are derived mainly from ursolic/oleanolic, acetyl ursolic or pomolic acid. CONCLUSION: The validated LC-UV method was shown to be linear, precise, accurate and to cover several saponins previously isolated from Ilex species and could be applied for the quality control of unripe fruit saponins.

This work reports the physico-chemical characterisation of the micellar structures formed by a saponin fraction obtained from an important South American species, Ilex paraguariensis (mate). The mate saponin-enriched fraction (MSF) mainly comprises triterpenic glycosides and was obtained from mate green fruits through solid-phase extraction. The physico-chemical studies focused on the determination of the critical micellar concentration (CMC), the size and shape of the micelles, using conventional transmission electronic microscopy (TEM), as well as Cryo-TEM, light scattering and small-angle neutron scattering. The rheological behaviour of the solutions up to 4 wt% was also determined using a controlled-strain rheometer. Finally, the MSF ability to solubilise poorly water-soluble drugs was assayed using carbamazepine and flurbiprofen as basic and weak acidic drug models. Small spherical micelles of around 20 Å radius were observed in the presence of elongated structures with lengths of more than 500 nm, possessing a well-defined CMC of 0.41 g/L. MSF solutions ranging from 0.25 to 4% (w/v) demonstrated a viscoelastic behaviour independent of the concentration. MSF could improve the solubility of carbamazepine in the range of 0.13 to 1.5% (w/v).

Saponins are natural compounds able to form abundant foam, a desirable quality required in some chemical, foods, cosmetic and pharmaceutical processes. Ilex paraguariensis A. St. Hil. (Aquifoliaceae) known as mate, is a South American widely cultivated specie due to the preparation of a tea-like beverage from its leaves. Moreover, its green fruits are a rich source of non-toxic and very low haemolytic saponins. In this study, mate saponin fraction (MSF) was evaluated as a foam former, focusing on its foamability, foam lifetime, and film drainage in the presence of different electrolytes (ionic strength I = 0.024 M). Sodium dodecyl sulphate (SDS) and polysorbate 80 (Poly-80) were used as reference surfactants. The critical micelle concentration (CMC), the minimum attainable surface tension (γCMC), surface excess concentration (Γ) and cross-section molecular surface (A) values of MSF were comparable to those of Poly-80. The foamability of MSF and both reference surfactants was equivalent. The addition of MgCl2 resulted in a negative effect on MSF foamability. The salts NaCl, KBr, and KNO3 exhibited a negative influence on MSF foam lifetime. Similar behavior was observed for MSF film drainage (order of activity: Na2HPO4 > MgCl2 ≈ NaCl ≈ KNO3 > KBr), where a primary fast film drainage rate was followed by film thinning stabilization after around 5 min. The behavior described above seems to be uncorrelated to the solutions' zeta potential.


As saponinas são compostos naturais capazes de formar espuma abundante, qualidade desejável exigida em alguns processos químicos, alimentícios, cosméticos e farmacêuticos. Ilex paraguariensis A. St. Hil. (Aquifoliaceae), mais conhecida como mate, é uma espécie largamente cultivada devido ao consumo do chimarrão. Além disso, seus frutos verdes representam uma fonte rica de saponinas pouco tóxicas e pouco hemolíticas. Nesse trabalho avaliou-se a fração de saponinas de mate (MSF) como formadora de espuma, com ênfase na espumabilidade, permanência da espuma e drenagem do filme na presença de diferentes eletrólitos (I = 0,024 M). Dodecil sulfato de sódio (SDS) e polissorbato 80 (Poly-80) foram utilizados como tensoativos-referência. Os valores de concentração micelar crítica (CMC), tensão superficial na CMC (γCMC), concentração de excesso de superfície (Γ) e área do grupamento polar na interface (A) referente à MSF foram comparáveis ao Poly-80. A espumabilidade da MSF e de ambos os tensoativos-referência foram equivalentes. A adição de MgCl2 resultou em um efeito negativo sobre a espumabilidade de MSF. Os sais NaCl, KBr e KNO3 tiveram influência negativa sobre a estabilidade da espuma. Em relação à drenagem do filme de MSF, após 60 min. seguiu-se a ordem Na2HPO4 > MgCl2 ≈ NaCl ≈ KNO3 > KBr, caracterizando uma drenagem inicial acelerada seguida pelo equilíbrio hidrodinâmico do filme após 5 min. O comportamento observado parece não estar relacionado com o potencial zeta das soluções.
