RESUMO

Psoriasis and chronic ulcers not only significantly impair quality of life but also pose a challenge in dermatological treatment. This study aimed to identify new therapeutic targets and biomarkers for psoriasis and chronic ulcers by comparing their gene expression profiles. The gene expression profiles of psoriatic, wound and chronic ulcer patients, as well as healthy controls, were determined via RNA extraction and next-generation sequencing of biopsies. In order to identify biomarkers, functional enrichment, differential expression analysis and machine learning algorithms were implemented. It is worth mentioning that the genes IL17A, TNF, KRT16, MMP9, and CD44 exhibited substantial correlations with the pathogenesis of the conditions being studied. As evidenced by their AUC-ROC values approaching 0.90, machine learning models accurately identified these biomarkers. The differential gene expression was consequently validated via qRT-PCR, which highlighted the increased expression of matrix remodelling enzymes and inflammatory cytokines. Additionally, genes essential for maintaining epidermis integrity and facilitating wound healing exhibited downregulation. These insights into the molecular mechanisms of psoriasis and chronic ulcers pave the way for the development of targeted therapies, offering hope for improved treatment strategies.

RESUMO

Objective To discuss effects of Wugong Baidu Drink on expressions of Bcl-2, NF-κB mRNA genes related to the apoptosis of HaCaT in psoriasis. Methods Wistar rats were randomly divided into 6 groups:blank group, Western medicine control group, TCM control group, and Wugong Baidu Drink high, medium, and low dose groups. They were given normal saline, methotrexate suspension, compound natural indigo capsule suspension, and the different doses of Wugong Baidu Drink for gavage, respectively. Drug-containing serum of each group was prepared, and was put in the HaCaT respectively to subculture. The intervention effect of different Drug-containing serums on expressions of objective genes were detected through PR-PCR method. Results Drug-containing serum of each group could decrease the expressions of Bcl-2 mRNA and NF-κB mRNA. The most obvious effects were shown in the groups of Western medicine control group and Wugong Baidu Drink high dose group. All Wugong Baidu Drink groups showed a dose-response relationship. Conclusion Wugong Baidu Drink probably can promote the apoptosis of HaCaT by reducing the gene expressions of Bcl-2 and NF-κB. It can also decrease immune response by regulating serum factors related to T cells to play the role of anti-psoriasis.

RESUMO

Objective To evaluate the efficacy and safety of butyl flufenamate ointment for the treatment of solar dermatitis.Methods Sixty-four patients with solar dermatitis were randomly divided into the observation group (n =32) and control group (n =32) to topically apply butyl flufenamate ointment and diclofenac sodium cream,respectively,for 3 weeks.The efficacy was evaluated by lesion scores and a visual analog scale for pruritus.Results At the end of the treatment,4 patients in the observation group were clinically cured,19 markedly improved,17 improved,and 2 showed no improvement; meanwhile,2 patients in the control group were clinically cured,11 markedly improved,14 improved,and 5 showed no improvement.The total response rate was significantly higher (93.8% vs.84.4%,P < 0.05),while the pruritus score was significantly lower (P < 0.05) in the observation group than in the control group.Conclusion Butyl flufenamate ointment is superior to dicofenac sodium cream in terms of both efficacy and anti-pruritic effect.
