RESUMO

The effect of vitamins (B1, B2, B3, B6, B12, H, PP and folic acid) and amino acids (glutamic and aspartic acids) on glucose isomerase biosynthesis was studied in Streptomyces albogriseolus. These compounds were added either alone or in combinations to different growth media (synthetic and complex). The results were processed using mathematical methods, and the following mixture of vitamins and amino acids was proposed to be added to the complex fermentation medium: B2, 10 micrograms/L; B6, 10 micrograms/L; H, 1 microgram/L; aspartic acid, 0.01 microM. The production of glucose isomerase rose more than 1.5 times after such additions.

RESUMO

The role of systems for glucose transport in the manifestation of carbon catabolite repression of glucoamylase synthesis was studied in the yeast Endomycopsis fibuligera. Experimentas were conducted with its mutant AB-192 defective in the system of transport universal for glucose and 2-deoxy-D-glucose (2-DG). The nature of the mutation was established from the following data: (1) transport of labeled glucose into the mutant cells was twice as low in comparison with the parent culture 20-9; (2) transport of labeled 2-DG was suppressed almost entirely; (3) no competition was found between glucose and 2-DG for penetration into the mutant cells. Glucoamylase synthesis in the mutant AB-192 was not sensitive to catabolite repression by glucose. This was confirmed by the resistance of the AB-192 cells to the inhibition by glucose and their complete resistance to the repression by 2-DG. Moreover, an addition of cAMP did not stimulate glucoamylase synthesis by the mutant culture in the presence of glucose and 2-DG. It can be concluded therefore that the resistance of the yeast to catabolite repression by the glucose is caused by the mutation in the system for carbohydrate transport. The results suggest that the system of glucose transport plays an important role in the manifestation of carbon catabolite repression in the yeast Endomycopsis fibuligera.

The effect of glucose, sucrose, fructose, maltose, alpha-methyl glucoside, glycerol, nonmetabolizing glucose analog--2-deoxy-D-glucose, and cyclic 3',5'-adenosine monophosphate (cAMP) on the glucoamylase biosynthesis by the yeast Endomycopsis fibuligera 20-9 was investigated. The sugars tested induced repression of the enzyme synthesis. The repressive effect of glucose, sucrose and maltose was reversed partially or completely by cAMP. The strongest derepressive effect of cAMP was noted in the presence of 2-deoxy-D-glucose. The transport of glucose and 2-deoxy-D-glucose in yeast cells was also investigated. Those compounds were found to compete for the entry into the cell. It is concluded that glucoamylase synthesis in Endomycopsis fibuligera 20-9 was susceptible to catabolite repression. Its possible mechanism discussed.

RESUMO

The growth cycle and the accumulation of glucoamylase by the yeast-like fungus Endomycopsis fibuligera 20-9 were studied in the conditions of submerged cultivation. The enzyme was accumulated by the end of the logarithmic phase and particularly at the stationary phase of the cultural growth. A correlation has been established between the accumulation of the glucoamylase in the cultural broth and the production of blastospores. Possible systems involved in the control of the enzyme accumulation are discussed.

RESUMO

The effect of nitrosoguanidine (NG) and ultra-violet light (UV) on the survival and the activity of glucoamylase (GIA) was studied with Endomycopsis fibuligera. Variants which strongly differed from the control cultures in their activity of Gia were sported only by the action of NG, particularly at a concentration of 500 mcg/ml, during 90 min. In this case, about 5 per cent of variants with the activity of GIA being 160-200 per cent of the control value appeared in the population.

RESUMO

According to the author's data postoperatively in patients receiving cyclopropane a much more early restoration of the renal excretory function was noted, postoperative intestinal paresis is liquidated more rapidly, the number of complications on the part of the respiratory and cardiovascular systems is diminished up to a minimum. There was less degree of postoperative metabolic alkalosis under the influence of cyclopropane, and it was also more rapidly eliminated.

The effect of glucose and cyclic 3',5'-adenosine monophosphate (cAMP) on the synthesis of glucoamylase and alpha-amylase by the yeast Endomycopsis fibuligera str. 20-9 was investigated. The glucoamylase synthesis was inhibited by every concentration (0.05--3.0%) of glucose. The synthesis of alpha-amylase was significantly reduced by high concentrations (2.0-3.0%) of carbohydrate only. An addition of cAMP (1 X 10(-5) M) reversed compeltely the inhibitory effect of 0.5-2.0% glucose on the alpha-amylase synthesis and partially restored the glucoamylase synthesis. It is concluded that catabolite repression may be involved in the regulation of alpha-amylase and glucoamylase synthesis by Endomycopsis fibuligera str. 20-9 and that this type of regulation shows varying effectiveness in the enzyme synthesis.
