Accurate mechanical feedback systems are critical to the successful implementation of virtual and robotic surgical assistant systems. Experimental measurements of reaming force could further our understanding of the cancellous bone reaming process during hip arthroplasty to help develop surgical simulators with realistic force effects and improve the protection mechanism of robot-assisted surgical systems. In this study, reaming experiments with natural bone (porcine femur) and a bone substitute (polyurethane blocks) were performed on a CNC lathe. This paper proposes using the maximum reaming force of the steady reaming stage to represent the force characteristic. The reaming force is biased to one side in the overlap direction and the maximum reaming force will vary when the reamer is not coincident with the long axis of the bone. The diameter of the reamer has the greatest influence on reaming force, which clearly increases with increasing reamer diameter. During operation, a medium rotation speed and high feed speed can reduce the reaming force. After cutting, the morphology of the cut surface is not flat, but arc-shaped, which will have a significant impact on implantation of the femoral prosthesis. In in vitro cutting experiments, polyurethane blocks can be used as a substitute for cancellous bone.

Objective: To develop and validate ubiquitination-related molecular subtypes and a novel prognostic index using ubiquitination-related genes (URGs) for patients with bladder cancer (BCa). Materials and Methods: We downloaded the clinical data and transcriptome data of BCa from TCGA and GEO database. Consensus clustering analysis was conducted to identify ubiquitination-related molecular subtypes for BCa. Besides, we performed univariate and multivariate Cox regression analysis to develop a novel prognostic URGs-related index for BCa. We conducted internal and external verification in TCGA cohort and GEO cohort, respectively. Furthermore, the associations of ubiquitination-related molecular subtypes and prognostic index with tumor immune environment were also investigated. Results: A total of four ubiquitination-related molecular subtypes of BCa were finally identified. These four molecular subtypes had significantly different clinical characteristics, prognosis, PD-L1 expression level and tumor microenvironment. Besides, we developed a novel prognostic index using six URGs (including HLA-A, TMEM129, UBE2D1, UBE2N, UBE2T and USP5). The difference in OS between high and low-risk group was statistically significant in training cohort, testing cohort, and validating cohort. The area under ROC curve (AUC) for OS prediction was 0.736, 0.723, and 0.683 in training cohort, testing cohort, and validating cohort, respectively. Multivariate survival analysis showed that this index was an independent predictor for OS. This prognostic index was especially suitable for subtype 1 and 3, older, male, high grade, AJCC stage III-IV, stage N0, stage T3-4 BCa patients. Conclusions: This study identified a total of four ubiquitination-related molecular subtypes with significantly different tumor microenvironment, prognosis, clinical characteristics and PD-L1 expression level. Besides, a novel ubiquitination-related prognostic index for BCa patients was developed and successfully verified, which performed well in predicting prognosis of BCa.

The cytokine storm is a marker of severity of various diseases and increased mortality. The altered metabolic profile and energy generation of immune cells affects their activation, exacerbating the cytokine storm. Currently, the emerging field of immunometabolism has highlighted the importance of specific metabolic pathways in immune regulation. The glycolytic enzyme pyruvate kinase M2 (PKM2) is a key regulator of immunometabolism and bridges metabolic and inflammatory dysfunction. This enzyme changes its conformation thus walks in different fields including metabolism and inflammation and associates with various transcription factors. This review summarizes the vital role of PKM2 in mediating immunometabolic reprogramming and its role in inducing cytokine storm, with a focus on providing references for further understanding of its pathological functions and for proposing new targets for the treatment of related diseases.

OBJECTIVE: To investigate the predictors of response to intravesical Bacillus Calmette-Guerin (BCG) immunotherapy for intermediate and high-risk non-muscle invasive bladder cancer (NMIBC) patients. MATERIALS AND METHODS: We retrospectively analyzed the clinicopathological data of 184 intermediate and high risk NMIBC cases receiving transurethral resection of bladder tumor (TURBT) and intravesical BCG immunotherapy from December 2014 to April 2021 at our center. All patients were divided into BCG responders and non-responders. Multivariate Logistic regression analysis was performed to identify the independent predictors of response to intravesical BCG immunotherapy. Univariate and multivariate Cox regression analyses were applied to explore the independent prognostic factors of recurrence-free survival (RFS). Receiver operating characteristic (ROC) curve and Kaplan-Meier survival analysis were also utilized. RESULTS: The RFS of BCG responders was significantly increased compared with BCG non-responders. Multivariate Cox regression analysis demonstrated that low grade, pTa stage, non-CIS, lower relative visceral fat area (rVFA) and lower systemic immune inflammation index (SII) were independent prognostic factors of increased RFS after intravesical BCG immunotherapy. Multivariate Logistic regression analysis demonstrated that pTa stage, low grade, non-CIS, low rVFA, and low SII were independent predictors of response to intravesical BCG immunotherapy. Kaplan-Meier survival analysis indicated that the RFS of patients in low rVFA group or low SII group was significantly increased in comparison with those in high rVFA group or high SII group. ROC curve analysis showed that the area under ROC (AUC) of including SII and rVFA was significantly increased, indicating that the inclusion of preoperative SII and rVFA could significantly improve the predictive efficiency. CONCLUSIONS: Low grade, pTa stage, non-CIS, preoperative lower rVFA and lower SII were vital independent predictors of response to intravesical BCG immunotherapy and were associated with preferable prognosis in NMIBC patients. The inclusion of preoperative SII and rVFA could significantly improve the predictive efficiency.

In this work, oxygen-defect-rich, three-dimensional (3D) cobalt-gallium layered double hydroxides (Co0.50-Ga0.50-LDH) assembled by porous and ultrathin nanosheets are prepared by a simple one-step strategy. Briefly, an aqueous solution containing Co2+ and Ga3+ is quickly pouring into the aqueous solution of hexamethylenetetramine, the state-of-the-art LDH was obtained followed by a mild and fast hydrothermal reaction. This mild and rapid synthesis strategy introduces a large number of pores into the ultrathin LDH nanosheets, resulting in a high concentration of oxygen vacancies in the Co0.50-Ga0.50-LDH, and the concentration of oxygen vacancies can be arbitrarily modulated, which has been corroborated by X-ray photoelectron spectroscopy and electron spin resonance measurements. The synergistic effect of the oxygen vacancy and the introduced Ga ions in the LDH nanosheets enhances the adsorption of the LDH nanosheets on OH-, endowing Co0.50-Ga0.50-LDH with outstanding performance for the supercapacitor application. Co0.50-Ga0.50-LDH offers a high specific capacity (0.62C·cm-2) at 10 mV·s-1 and extraordinary cycling stability. An aqueous asymmetric supercapacitor (ASC) constructed with Co0.50-Ga0.50-LDH and activated carbon (AC) materials exhibits high energy density and a long lifespan. This result encourages the wide application of porous ultrathin LDH nanosheets in energy storage, catalysis and light response.

BACKGROUND: The risk factors for complete atrioventricular block (CAVB) after device closure of perimembranous ventricular septal defect (pmVSD) remain unclear. OBJECTIVE: The objective of this study is to analyze the incidence and risk factors for CAVB after device closure for pmVSD. METHODS: We reviewed 1884 patients with pmVSD undergoing successful device occlusion between June 2005 and January 2020. Permanent CAVB was defined as CAVB requiring implantation of permanent pacemaker (PPM) or extraction of occluder. RESULTS: In total, 14 (0.7%) patients developed permanent CAVB. Of these patients, 10 (0.5%) required PPM implantation. Four permanent CAVBs occurred within 7 days after the procedure (acute), 2 between 7 and 30 days (subacute), 3 between 30 days and 1 year (late), and 5 more than 1 year (very late). None of the subacute, late, and very late CAVB recovered normal conduction with medication and eventually required device removal or PPM implantation. Four patients with acute CAVB and 1 with subacute CAVB underwent device removal, and 4 (80%) of them recovered normal conduction. Multivariate regression revealed that the ratio of device to defect size was the only independent risk factor for permanent CAVB (odds ratio, 3.027; 95% confidence interval, 1.476-6.209; P=0.003). CONCLUSION: The incidences of permanent CAVB after occlusion for pmVSD and PPM implantation were 0.7% and 0.5%, respectively. The ratio of device to defect size was the only independent risk factor for permanent CAVB. Device removal is an effective therapeutic modality for recovering normal conduction in acute and subacute CAVB patients.

Lac insects include astonishing species responsible for lac production. Lac is composed of resins, dyes, and shellac wax with significant economic importance. Previously, 11 species of the genus Kerria were reported from China, with the highest species diversity in Yunnan province. Another lac-producing species of the genus Kerria, namely Kerrianepalensis Varshney, is recorded for the first time in Yunnan province, China, on Dalbergiacochinchinensis Pierre ex Laness. (Fabaceae), a new host plant. In addition, a key to the 12 Kerria species recorded in China is also given.

Optical metasurfaces hold great potential for near-eye display applications with high optical efficiency, light-weight and compactness. Taking advantage of optical resonances in subwavelength features, metasurfaces can diffract optical beams at RGB primary colors efficiently, forming superimposed virtual images over a real-world scene with merely a single glass substrate in augmented realities (AR) applications. We report for the first time a metasurface with double or triple nano-beams in each period for high angle diffraction with a uniform efficiency at RGB wavelengths. An efficiency as high as 30-40% of the first diffraction order is obtained across field of view, allowing a single piece of AR glass for light in-coupling to deliver image uniformly. The underlying physics is investigated through systematic full-vector numerical simulations. It is found that strong resonances inside the nano-beams with different sizes are the main reason for the deflection of wavefront as well as the Poynting vectors, leading to an efficient coupling of the incident light into the first-order diffraction. The resonances also manipulate the light absorption among the RGB colors for uniform efficiency. This work provides a new understanding of optimal metasurface structure for waveguide couplers using multiple nano-beams.

An asymmetric cryptosystem is presented for encrypting multiple images in gyrator transform domains. In the encryption approach, the devil's spiral Fresnel lens variable pure phase mask is first designed for each image band to be encrypted by using devil' mask, random spiral phase and Fresnel mask, respectively. Subsequently, a novel random devil' spiral Fresnel transform in optical gyrator transform is implemented to achieved the intermediate output. Then, the intermediate data is divided into two masks by employing random modulus decomposition in the asymmetric process. Finally, a random permutation matrix is utilized to obtain the ciphertext of the intact algorithm. For the decryption approach, two divided masks (private key and ciphertext) need to be imported into the optical gyrator input plane simultaneously. Some numerical experiments are given to verify the effectiveness and capability of this asymmetric cryptosystem.

The prognosis of pancreatic ductal adenocarcinoma (PDAC) is poor despite diagnostic progress and new chemotherapeutic regimens. Constitutive activation of NF-κB is frequently observed in PDAC. In this study, we found that YEATS2, a scaffolding protein of ATAC complex, was highly expressed in human PDAC. Depletion of YEATS2 reduced the growth, survival, and tumorigenesis of PDAC cells. The binding of YEATS2 is crucial for maintaining TAK1 activation and NF-κB transcriptional activity. Of importance, our results reveal that YEATS2 promotes NF-κB transcriptional activity through modulating TAK1 abundance and directly interacting with NF-κB as a co-transcriptional factor.

It has been reported that long non-coding RNAs (lncRNAs) play a crucial role in the progression of various types of cancer. The role of numerous lncRNAs in a variety of cancer types has been investigated. However, the underlying mechanisms of the majority of lncRNAs in bladder cancer (BCa) remain to be elucidated. In the present study, abnormally expressed lncRNAs in BCa and para-carcinoma tissues were identified through screening the Cancer RNA-Seq Nexus database and were validated using reverse transcription-quantitative PCR. It was found that LOC339524 expression levels were markedly downregulated in BCa tissues and cells (J82, T24, UM-UC-3 and 5637). LOC339524 overexpression was revealed to suppress the proliferation of BCa cells. LOC339524 was also discovered to act as a sponge for microRNA (miR)-875-5p, as identified using dual luciferase reporter assays and biotin pull-down analysis. LOC339524 downregulated the expression of miR-875-5p and knockdown of miR-875-5p expression inhibited the proliferation of bladder cancer cells. In addition, COP9 signalosome subunit 7A (COPS7A) was identified to be the target gene of miR-875-5p and COPS7A expression level was upregulated following LOC339524 overexpression. lncRNA LOC339524 was proposed to function as a competitive endogenous RNA to facilitate the expression of COPS7A by binding to miR-875-5p. In conclusion, the findings of the present study suggested that LOC339524 may inhibit cell proliferation in BCa by targeting the miR-875-5p/COPS7A signaling axis.

Graphene/carbon nanotube (CNT)-based adsorbents were fabricated on a kilogram scale by extrusion processing (where graphene is used as the major adsorption material and CNTs make up the backbone to enhance the mechanical strength) and then mixed and bonded with poly(tetrafluoroethylene). Kilogram-scale adsorbents were used to treat the content of o-cresol in wastewater to be <1.12 mg/kg in a continuous and reversible adsorption-desorption apparatus, which could last for 99 h with a space velocity of 30 h-1 and a total wastewater capacity of 5 tons per day. Brunauer-Emmett-Teller (BET), Fourier transform infrared spectroscopy (FT-IR), X-ray photoelectron spectroscopy (XPS), thermogravimetric analysis (TGA), and Raman spectroscopy all suggested that the surface properties and pore structure of the spent adsorbents remain unchanged after recycling at both low-temperature adsorption and high-temperature desorption in vacuum. These results provided an effective reversible adsorbent system for removing aromatic organics and prompted the scaled-up applications of carbon nanomaterials in the treatment of wastewater.

Ten types of Tf2O/TTBP-mediated amide transformation reactions were investigated. The results showed that compared with pyridine derivatives 2,6-di-tert-butyl-4-methylpyridine (DTBMP) and 2-fluoropyridine (2-F-Pyr.), TTBP can serve as an alternative amide activation system for the direct transformation of both secondary and tertiary amides. For most surveyed examples, higher or comparable yields were generally obtained. In addition, Tf2O/TTBP combination was used to promote the condensation reactions of 2-(tert-butyldimethylsilyloxy)furan (TBSOF) with both tertiary and secondary amides, the one-pot reductive Bischler-Napieralski-type reaction of tertiary lactams, and Movassaghi and Hill's modern version of the Bischler-Napieralski reaction. The value of the Tf2O/TTBP-based methodology was further demonstrated by the concise and high-yielding syntheses of several natural products.

Cynaroside is the primary flavonoid component of honeysuckle which has been widely used as Chinese traditional medicine given its anti-inflammation properties. Overactive systemic inflammatory response and multi-organ injury are the leading causes of life-threatening sepsis. Regulation of macrophage polarization balance may act as a promising strategy for its treatment. In the present study, we aimed to investigate whether cynaroside exerted protective effects against sepsis and its potential mechanism. Building upon a sepsis mouse model, we observed cynaroside alleviated serum levels of inflammatory factors including IL-1ß and TNF-α at 5 and 10 mg/kg. The pathological injury of heart, kidney and lung was remarkedly attenuated as the levels of blood urea nitrogen, creatinine, creatine kinase-MB and lactate dehydrogenase were reduced nearly 2.8-, 2.7-, 2.4-, and 2.5-fold as compared with the sepsis mice, respectively. We further demonstrated cynaroside suppressed the biomarker of pro-inflammatory macrophage M1 phenotype (iNOS+) and promotes the anti-inflammatory M2 polarization (CD206+) in the injury organs of septic mice. Mechanistic research verified cynaroside inhibited LPS-induced polarization of macrophage into M1 phenotype, which can be highly blocked by Nrf2 inhibitor. Expectedly, Nrf2 and its downstream (Heme oxygenase-1 (HO-1)) was upregulated in injury organs after treating with cynaroside, indicating the involvement of Nrf2 signaling. Taken together, the data claims cynaroside ameliorated systematic inflammation and multi-organ injury dependent on Nrf2/HO-1 pathway in septic mice.

Background: The preoperative distinguishment of lymph nodes with reactive hyperplasia or tumor metastasis plays a pivotal role in guiding the surgical extension for papillary thyroid carcinoma (PTC) with Hashimoto's thyroiditis (HT), especially in terms of the central lymph node (CLN) dissection. We aim to identify the preparative risk factors for CLN metastasis in PTC patients concurrent with HT. Materials and Methods: We retrospectively reviewed and analyzed the data including the basic information, preoperative sonographic characteristics, and thyroid function of consecutive PTC patients with HT in our medical center between Jan 2019 and Apr 2021. The Chi-square and Fisher's exact tests were used for comparison of qualitative variables among patients with or without CLN metastasis. Univariate and multivariate logistic regression analyses were used to determine the risk factors for CLN metastasis. The nomogram was constructed and further evaluated by two cohorts produced by 1,000 resampling bootstrap analysis. Results: A total of 98 in 214 (45.8%) PTC patients were identified with CLN metastasis. In multivariate analysis, four variables including high serum thyroglobulin antibody (TgAb) level (>1,150 IU/ml), lower tumor location, irregular margin of CLN, and micro-calcification in the CLN were determined to be significantly associated with the CLN metastasis in PTC patients with HT. An individualized nomogram was consequently established with a favorable C-index of 0.815 and verified via two internal validation cohorts. Conclusions: Our results indicated that preoperatively sonographic characteristics of the tumor and lymph node condition combined with serum TgAb level can significantly predict the CLN in PTC patients with HT and the novel nomogram may further help surgeons to manage the CLN in this subpopulation.

Agricultural works alter earth's surface at the largest scale among human-driven activities. Previous studies have focused more on the reclamation of natural land, however, farmland marginalization (FM), emerging as an important mean of land use changes in mountainous and hilly areas (MHAs) has always been overlooked in the background of production efficiency improvement along with urbanization and population migration. This paper examined the characteristics of the spatial-temporal distribution and conversion of marginalized farmland in the MHAs of China at county level (excluding Hong Kong, Macau, and Taiwan) from 1990 to 2020, regarding farmland in MHAs converted into non-built-up land as FM. The results showed that: (1) The total area of marginalized farmland in the MHAs was 1.03 × 106 km2. The counties with larger area of marginalized farmland were concentrated around the Hu Line, and those with higher ratio were distributed in southern mountainous areas. (2) The area of marginalized farmland in each stage exhibited a fluctuating trend from 1990 to 2020. Forests and grasslands were prioritized as the desirable types in land conversion, and had prominent spatial agglomeration. (3) The influence of FM in MHAs on land use changes at county level demonstrated significant spatial-temporal heterogeneity, with wide range and low intensity from 1990 to 2000 and 2015 to 2020, and narrow range and high intensity from 2000 to 2015, and the counties with high intensity were distributed in the Loess Plateau and Sichuan-Chongqing hilly region. (4) The slope of marginalized farmland exhibited a prominent rule of spatial distribution, but an insignificant temporal trend under the influence of governmental policies. The larger the slope was, the higher the degree of marginalization was, but not necessarily earlier it occurred. The results can provide a reference for the formulation and implementation of farmland protection policies.

The tumor immune microenvironment of lung cancer is associated with prognosis and immunotherapy efficacy. Long noncoding RNAs are identified as prognostic biomarkers associated with immune functions. We constructed a signature comprising differentially expressed immune-related lncRNAs to predict the prognosis of patients with lung adenocarcinoma. We established the immune-related lncRNA signature by pairing immune-related lncRNAs regardless of expression level and lung adenocarcinoma patients were divided into high- and low-risk groups. The prognosis of patients in the two groups was significantly different; The immune-related lncRNA signature could serve as an independent lung adenocarcinoma prognostic indicator. The signature correlated negatively with B cell, CD4+ T cell, M2 macrophage, neutrophil, and monocyte immune infiltration. Patients with low risk scores had a higher abundance of immune cells and stromal cells around the tumor. Gene set enrichment analysis showed that samples from low-risk group were more active in the IgA production in intestinal immune network and the T and B cell receptor signaling pathway. High-risk groups had significant involvement of the cell cycle, DNA replication, adherens junction, actin cytoskeleton regulation, pathways in cancer, and TGF-ß signaling pathways. High risk scores correlated significantly negatively with high CTLA-4 and HAVCR2 expression and higher median inhibitory concentration of common anti-tumor chemotherapeutics (e.g., cisplatin, paclitaxel, gemcitabine) and targeted therapy (e.g., erlotinib and gefitinib). We identified a reliable immune-related lncRNA lung adenocarcinoma prognosis model, and the immune-related lncRNA signature showed promising clinical prediction value.

BACKGROUND: Adolescent differentiated thyroid carcinoma (DTC) is a rare type of thyroid cancer that represents a special entity of all endocrine-related cancer. This study aims to establish the first nomogram for predicting the regional (central and lateral) lymph node metastasis (LNM) in the adolescent population for better surgical management. METHOD: We retrospectively reviewed the clinicopathology characteristics of adolescent patients with DTC in the Surveillance, Epidemiology, and End Results database between 2010 and 2015. RESULTS: A total of 1,930 adolescent patients between the ages of 10 and 24 years from the SEER database were enrolled in this study. Six predictive factors including age, race, histology, multifocality, extrathyroidal invasion (EI) and tumor size were identified to be significantly associated with the regional LNM via univariate and multivariate logistic regression analyses. These indicators were used to construct a nomogram for predicting the regional LNM in adolescent patients with DTC. Moreover, a satisfied predictive ability of the model was determined with a C-index of 0.794, supported by an internal validation group with a C-index of 0.776. The Decision Curve Analysis and calibration curve further conducted a great agreement in our model. CONCLUSION: The first predictive model containing multiple factors has been successfully established with good discrimination for predicting the regional LNM in adolescent patients with DTC. This nomogram could effectively help surgeons to make better individualized surgical decision intraoperatively, especially in terms of whether cervical lymph node dissection (LND) is warranted.

The control of single atoms offers fundamental insight into understanding the charge transport through single clusters, and the atomic precision of the clusters provides the opportunity to manipulate the charge transport even at the single-atom level. Herein, we designed and investigated the electrical conductance and thermopower of Anderson-type polyoxometalate (POM) clusters with single-atom variation using the scanning tunneling microscopy break-junction (STM-BJ) technique. Our results show the electrical conductance of single clusters can be changed by an order of magnitude by substituting different center-metal atoms, and the electrical conductance of clusters shows different bias-dependence. Furthermore, the Seebeck coefficients of the POM clusters also can be significantly changed by the center-metal atoms. The non-equilibrium quantum transport calculations reveal that the electrostatic potential profile is non-uniformly dependent on the center-metal atoms. This leads to gating of electrical conductance by bias voltage. This supports the tuning of thermopower and bias dependent transmission spectra. This work provides the fundamental understanding of single-atom control of charge transport in POM single-cluster junctions.
