Constipation is a common gastrointestinal disorder characterized by changes in intestinal habits. Increasing evidence indicates that long-term use of irritant laxatives causes serious side effects. Meanwhile, more than 50% of patients are dissatisfied with sense of use of non-prescriptional laxatives. ß-Glucans are natural polysaccharides widely found in yeast, fungus, and plants, which have been reported to exhibit various pharmacological effects. The aim of this study was to characterize the effect of ß-glucans extracted from the bread yeast cell wall on loperamide-induced constipation mice. Forty mice were fed with loperamide (10 mg/kg) to make the constipation model and a diet supplemented with 2.5, 5, and 10 mg/kg ß-glucan. We assessed the defecation frequency, intestinal transit function of mice, as well as used high-throughput sequencing to analyze the intestinal microbiota composition and functional biological profiles data. Meanwhile, we detected expression of neurotransmitters including acetylcholinesterase, substance P, and serotonin (5-HT) and expression of tight junction protein (TJP) including zonula occludens-1 and mucin-2 in distal colon to characterize the possible molecular mechanisms. ß-Glucans significantly enhanced intestinal motility and provided a possibility to regulate the expression of neurotransmitters and TJP in mice. The intestinal microecological portion of the treatment group partially recovered and was closer to the normal group. This study showed that ß-glucans can influence the intestinal microbiota and restore microecological balance to regulate the express of neurotransmitters and TJP to recover intestinal epithelial mechanical barrier. We suggested that ß-glucans could be used as an active nutritional supplement to protect the damaged intestinal barrier and help patients who have constipation complications and dysbiosis.

Lead-free potassium and sodium niobate (KNN) nanofiber webs with random and aligned configurations were prepared by the electrospinning process from polymer-modified chemical solution. The crystallization process, structure, composition, dielectric, ferroelectric, and piezoelectric properties of the nanofibers and nanofiber webs were investigated. Theoretical analysis and experimental results showed that the surface-induced heterogeneous nucleation resulted in the remarkable lower crystallization temperature for the KNN nanofibers with the {100} orientation of the perovskite phase in contrast to the bulk KNN gel and thus well-controlled chemical stoichiometry. Low dielectric loss, large electric polarization, and high piezoelectric performance were obtained in the nanofiber webs. In particular, the aligned nanofiber web exhibited further improved piezoelectric strain and voltage coefficients and higher FOM than their thin film counterparts and is promising for high-performance electromechanical sensor and transducer applications.

OBJECTIVE: To build a CT-based radiomics predictive mode to evaluate the differentiation degree of the esophageal squamous carcinoma.
 Methods: A total of 160 patients with surgical pathology, complete clinical data and chest CT scanning before operation were retrospectively collected from January 2008 to August 2016. All patients were assigned randomly to a primary data set and an independent validation. Texture analysis was performed on CT images, while the carcinomas were performed by manual segmentation to extract the radiomics features. Radiomics features were extracted and 9 radiomics signatures were finally selected after dimension reduction. Radiomics features were extracted and established via Matlab. Multivariable logistic regression analysis was performed to build the predictive model. A 10-fold cross-validation was used for selecting parameters in the least absolute shrinkage and selection operator (LASSO) model by minimum criteria. The receiver operating characteristic (ROC) curves and areas under ROC curve (AUC) were used to compare the model performance in the primary validation and the independent validation for evaluating the differentiation degree of esophageal squamous carcinoma.
 Results: Radiomics signature showed great effect in discriminating primary data set and independent validation. The predictive model had a good performance in primary data set. The AUC was 0.791, the sensitivity was 81.6%, and specificity was 72.3%. In the independent validation, the AUC was 0.757, the sensitivity was 70.0%, and the specificity was 73.0%.
 Conclusion: The predictive model can be used for evaluating the differentiation degree of esophageal squamous carcinoma efficiently, which can be helpful to clinicians in diagnosis and choice of treatment for esophageal squamous carcinoma.

BACKGROUND: Despite the recommendations of statins treatment for secondary prevention of atherosclerotic cardiovascular disease (ASCVD), treatment adherence and persistence are still a concern. This study examined the real world practice of long-term adherence and persistence to statins treatment initiated after hospital discharge for ASCVD, and their associated factors in a nationwide cohort. METHODS: Post discharge statin prescriptions between 2006 and 2012 were extracted from the Taiwan National Health Insurance claims database. Good adherence, defined as proportion of days covered (PDC) ≥0.8 and mean medication possession ratio (MPR), was measured every 180-day period. Non-persistence was defined on the date patients failed to refill statin for 90 days after the end of the last prescription. Their associations with influential factors were analyzed using a generalized estimating equation and Cox's proportional hazard model. RESULTS: There was a total of 185,252 post-discharge statin initiations (from 169,624 patients) and followed for 467,398 patient-years in the study cohort. Percentage of good adherence (mean MPR) was 71% (0.87) at 6-months; declined to 54% (0.68), 47% (0.59), and 42% (0.50) at end of year 1, 2, and 7, respectively. Persistence in statin treatment was 86, 67, 50, and 25% at 6-month, 1-, 2-, and 7-year, respectively. Comparing the statin-cohort initiated from year 2006 to 2012, 1-year persistence increased from 58 to 73%, and 1-year good adherence improved from 45 to 61%. Factors associated with sub-optimal adherence and non-persistence included: prescription by primary care clinics or non-cardiology specialties; patients' age > 75 years; no history of previous statin use; ASCVD events with ischemic stroke diagnosis; comorbidities of renal disease, liver disease, depression, and chronic obstructive pulmonary disease. CONCLUSIONS: Despite the improving trends, long-term adherence and persistence of statin treatment were suboptimal in Taiwan. Strategies to maintain statin treatment adherence and persistence need to be implemented to further enhance the positive trend.

In this work, using a modified Stöber process, we synthesized ordered mesoporous silica cubic particles (OMS-C) and prepared a nanocatalyst (Ag-OMS-C) based on OMS-C with a high surface area via an in situ auto-reduction strategy. The as-prepared Ag-OMS-C nanocomposites demonstrated open mesopores (3.51 nm), a large specific surface area (540 m2 g-1) and a high pore volume (0.88 cm3 g-1). The catalytic reduction of 4-nitrophenol (4-NP) over the Ag-OMS-C nanocatalyst was almost complete within 150 s without stirring and the rate constant k (30 × 10-3 s-1) is much higher than those of other substrate-supported Ag nanocatalysts. Moreover, the Ag-OMS-C nanocomposites hold a stable catalytic efficiency over five reaction cycles. The results indicate that the Ag-OMS-C nanocatalyst exhibited high catalytic activity and good reusability toward the reduction of 4-NP, which might be attributed to the large specific surface area, the pore structure of the nanocatalyst, as well as the synergistic effect between OMS-C and AgNPs.

The chemical structures of Ru (II) complexes are known to affect their cellular behavior and toxicity. In this study, three new luminescent Ru (II) complexes, [Ru(bpy)2(HIPMP)](ClO4)2 (Ru1, bpy = 2,2'-bipyridine, HIPMP = 2-(1H-imidazo-[4,5-f] [1,10] phenanthrolin-2-yl)-4-methylphenol), [Ru(phen)2(HIPMP)](ClO4)2 (Ru2, phen = 1,10-phenanthroline), [Ru(dmb)2(HIPMP)](ClO4)2 (Ru3, dmb = 4,4'-dimethyl-2,2'-bipyridine), were synthesized, and their anticancer activities were examined. All three complexes displayed anticancer activities against various cancer cells, with Ru2 exhibiting the highest cytotoxic activities. Ru2 was shown to accumulate specifically in the endoplasmic reticulum (ER) and induce ER stress-mediated apoptosis. In addition, Ru2 could generate reactive oxygen species (ROS) and trigger mitochondrial membrane potential depolarization. These results demonstrated that Ru2 induced apoptosis in HeLa cells through ER stress and ROS production.

OBJECTIVE: Self-rated health represents a reliable and important health measure related to general health and quality of life. This study aimed to identify the differences of health states of rural residents in a lower middle income setting in China and its associated factors. METHODS: A descriptive study of a stratified random sample of 3870 individuals was conducted in rural Anhui during 2015. We investigated the influence of five independent variables: individual demographic characteristics, family factors, social capital traits, physical health conditions and healthy lifestyle habits of participants who self-related their health as good. A chi-square test and ordinal logistic regression analyses were used to identify the relationship of these variables and self-rated health. RESULTS: The study found that respondents who negatively rated their health often were female, elderly, poor, lived alone, had low levels of education, inadequate social support, poor physical health, used healthcare services and lived in the lower economic regions. We found no significant correlations between self-rated health and employment, marital status, medical insurance, or exercise frequency. Surprisingly, smoking and drinking also seemed to be unrelated to poor self-reported health. CONCLUSION: Health differences based on region were apparent in rural China. We highlighted the possible impacts of income, age, physical health, education, advanced age, and social support on health. The results from this study could inform the delivery of appropriate health and social healthcare interventions to promote rural residents' health and quality of life.

Thymocyte-expressed molecule involved in selection (Themis) has been shown to be important for T cell selection by setting the threshold for positive versus negative selection. Themis interacts with the protein tyrosine phosphatase (PTP) Src-homology domain containing phosphatase-1 (Shp1), a negative regulator of the T cell receptor (TCR) signaling cascade. However, how Themis regulates Shp1 is still not clear. Here, using a very sensitive phosphatase assay on ex vivo thymocytes, we have found that Themis enhances Shp1 phosphatase activity by increasing its phosphorylation. This positive regulation of Shp1 activity by Themis is found in thymocytes, but not in peripheral T cells. Shp1 activity is modulated by different affinity peptide MHC ligand binding in thymocytes. Themis is also associated with phosphatase activity, due to its constitutive interaction with Shp1. In the absence of Shp1 in thymocytes, Themis interacts with Shp2, which leads to almost normal thymic development in Shp1 conditional knockout (cKO) mice. Double deletion of both Themis and Shp1 leads to a thymic phenotype similar to that of Themis KO. These findings demonstrate unequivocally that Themis positively regulates Shp1 phosphatase activity in TCR-mediated signaling in developing thymocytes.

The maintenance of autoreactive B cells in a quiescent state is crucial for preventing autoimmunity. Here we identify a variant of human immunoglobulin G1 (IgG1) with a Gly396→Arg substitution (hIgG1-G396R), which positively correlates with systemic lupus erythematosus. In induced lupus models, murine homolog Gly390→Arg (G390R) knockin mice generate excessive numbers of plasma cells, leading to a burst of broad-spectrum autoantibodies. This enhanced production of antibodies is also observed in hapten-immunized G390R mice, as well as in influenza-vaccinated human G396R homozygous carriers. This variant potentiates the phosphorylation of the IgG1 immunoglobulin tail tyrosine (ITT) motif. This, in turn, alters the availability of phospho-ITT to trigger longer adaptor protein Grb2 dwell times in immunological synapses, leading to hyper-Grb2-Bruton's tyrosine kinase (Btk) signaling upon antigen binding. Thus, the hIgG1-G396R variant is important for both lupus pathogenesis and antibody responses after vaccination.

Twisted light has recently gained enormous interest in communication systems ranging from fiber-optic to radio frequency regimes. Thus far, the light-emitting diode (LED) has not yet been exploited for orbital angular momentum (OAM) encoding to transmit data, which, however, could open up an opportunity towards a new model of secure indoor communication. Here, by multiplexing and demultiplexing red, green and blue (RGB) twisted beams derived from a white light emitting diode, we build a new visible light communication system with RGB colors serving as independent channels and with OAM superposition modes encoding the information. At the sender, by means of theta-modulation, we use a computer-controlled spatial light modulator to generate two-dimensional holographic gratings to encode a large alphabet with 16 different OAM superposition modes in each RGB channel. At the receiver, based on supervised machine learning, we develop a pattern recognition method to identify the characteristic mode patterns recorded by CCD cameras, and therefore, decoding the information. We succeed in demonstrating the transmission of color images and a piece of audio over a 6-meter indoor link with the fidelity over 96%.

Corrosion in internal cavity is one of the most common problems occurs in many hollow metallic components, such as pipes containing corrosive fluids and high temperature turbines in aircraft. It is highly demanded to non-destructively detect the corrosion inside hollow components and determine the corrosion extent from the external side. In this work, we present two high-frequency ultrasonic non-destructive testing (NDT) technologies, including piezoelectric pulse-echo and laser-ultrasonic methods, for detecting corrosion of Ni superalloy from the opposite side. The determination of corrosion layer thickness below ∼100 µm has been demonstrated by both methods, in comparison with X-CT and SEM. With electron microscopic examination, it is found that with multilayer corrosion structure formed over a prolonged corrosion time, the ultrasonic NDT methods can only reliably reveal outer corrosion layer thickness because of the resulting acoustic contrast among the multiple layers due to their respective different mechanical parameters. A time-frequency signal analysis algorithm is employed to effectively enhance the high frequency ultrasonic signal contrast for the piezoelectric pulse-echo method. Finally, a blind test on a Ni superalloy turbine blade with internal corrosion is conducted with the high frequency piezoelectric pulser-receiver method.

Piezoelectric materials are vital in determining ultrasonic transducer and imaging performance as they offer the function for conversion between mechanical and electrical energy. Ultrasonic transducers with high-frequency operation suffer from performance degradation and fabrication difficulty of the demanded piezoelectric materials. Hence, we propose 1-D polymeric piezoelectric nanostructure with controlled nanoscale features to overcome the technical limitations of high-frequency ultrasonic transducers. For the first time, we demonstrate the integration of a well-aligned piezoelectric nanotube array to produce a high-frequency ultrasonic transducer with outstanding performance. We find that nanoconfinement-induced polarization orientation and unique nanotube structure lead to significantly improved piezoelectric and ultrasonic transducing performance over the conventional piezoelectric thin film. A large bandwidth, 126% (-6 dB), is achieved at high center frequency, 108 MHz. Transmission sensitivity of nanotube array is found to be 46% higher than that of the monolithic thin film transducer attributed to the improved electromechanical coupling effectiveness and impedance match. We further demonstrate high-resolution scanning, ultrasonic imaging, and photoacoustic imaging using the obtained nanotube array transducers, which is valuable for biomedical imaging applications in the future.

The chemokine receptor 4 (CXCR4) has been an attractive molecular target for tumor imaging, because it is overexpressed in many tumor types and involved in tumor progression and metastasis. The purpose of this study is to examine the CXCR4 targeting properties of 99m Tc-labeled AMD3465, a small molecule antagonist of CXCR4. 99m Tc-AMD3465 was prepared in high yield (>95%) and stable in mice serum at least for 4 hours. In vitro cell binding experiments were performed with Chinese hamster ovary (CHO), MCF-7 (breast cancer), and CHO-CXCR4 (CHO stably transfected to express CXCR4) cell lines. Small animal single photon emission computed tomography/computed tomography imaging studies in nude mice bearing MCF-7 and CHO xenografts showed that the uptakes of the radiotracer in MCF-7 tumors were significantly higher than those in the CXCR4-negative CHO tumors (P < 0.05), and the MCF-7 tumors uptake could be blocked with an excess of unlabeled AMD3465 (P < 0.05). These results suggested that 99m Tc-AMD3465 could be a potential single photon emission computed tomography radiotracer for CXCR4 imaging.

The detection of plastic deformation of metallic alloy materials with second-harmonic Rayleigh ultrasonic wave is first investigated using direct-write piezoelectric ultrasonic transducers, in which piezoelectric poly(vinylidenefluoride/ trifluoroethylene) [P(VDF/TrFE)] polymer coatings and electrodes are directly deposited, processed, and patterned on the alloy to be evaluated. Rayleigh ultrasonic signals, generated by the direct-write transducers on titanium alloy specimens, are characterized by a laser scanning vibrometer. The results show that acoustic nonlinearity increases with plastic strain, and an increase of ~40% in the acoustic nonlinearity corresponding to a plastic strain of 5.1%. The measurement data and technical features with the use of the direct-write transducers are compared with the conventional discrete angle beam piezoelectric transducer. The results and analyses show that compared with the conventional discrete angle beam piezoelectric transducers, implementation of the direct-write piezoelectric transducers has significant technical advantages and is promising for applications in determining nonlinear ultrasonic waves and plastic strain of structural materials.

Maleic acid (MA), an intermediate reagent used in many industrial products, instigated public health concerns in Taiwan when it was used to adulterate an array of starch-based delicacies to improve texture and storage time. Established studies reported that exposure to high concentrations of MA induce renal injury; little is known whether oxidative stress is induced at a relative low dose. This study aims to investigate the effect of oral single dose exposure of MA on the status of oxidative stress and inflammation. Single dose of MA at 0, 6 and 60 mg/kg (control, low- and high-dose groups, respectively) were orally administered to adult male and female rats. Urine samples were collected and analyzed to measure 8-hydroxy-2'-deoxyguanosine (8-OHdG), 8-iso-prostaglandin F2α (8-IsoPGF2α), 8-nitroguanine (8-NO2Gua) and N-acetyl-S-(tetrahydro-5-hydroxy-2-pentyl-3-furanyl)-L-cysteine (HNE-MA) using LC-MS/MS. Results revealed that oral consumption of MA induced oxidative DNA damage and lipid peroxidation, as demonstrated by the statistically significant increases in urinary levels of 8-NO2Gua, 8-OHdG, and 8-isoPGF2α, in high-dosed male rats within 12 h of oral gavage (p < 0.05). Additionally, increases in concentration of these biomarkers persist for days after consumption; male rats appear to be more sensitive to oxidative burden compared to their counterparts. The aforementioned findings could help elucidate the mechanisms through which nephrotoxicity occur.

This paper starts from a review on the progress in fabrication of piezoelectric ceramic coatings by thermal spray method. For our experimental work, two types of lead-free piezoelectric ceramic coatings, including potassium-sodium niobate-based and bismuth sodium titanate-based, are fabricated by thermal spray process, and their structure, morphology, and piezoelectric properties are characterized. Our obtained lead-free ceramic coatings exhibit single phase of perovskite structure, relatively dense morphology, and competitive piezoelectric coefficients. The mechanism of forming the piezoelectric perovskite crystalline phase by thermal spray involving melting-recrystallization process is analyzed in comparison to that of ceramic synthesis through solid-state reaction. Suppression of volatile loss and decomposition at high temperature due to the extremely high melting and cooling rate in the thermal spray process, and the impact on the resulting structure are discussed. Significant advantages of the thermal spray method over alternative processing methods for forming piezoelectric ceramic coatings are summarized. The combination of environmentally friendly lead-free compositions and the scalable thermal spray processing method will promote more applications of piezoelectric ceramic coatings for producing distributive sensors and transducers, and forming advanced smart structures and systems.

Maleic acid (MA), a chemical intermediate used in many consumer and industrial products, was intentionally adulterated in a variety of starch-based foods and instigated food safety incidents in Asia. We aim to elucidate possible mechanisms of MA toxicity after repeated exposure by (1) determining the changes of metabolic profile using 1 H nuclear magnetic resonance spectroscopy and multivariate analysis, and (2) investigating the occurrence of oxidative stress using liquid chromatography tandem mass spectrometry by using Sprague-Dawley rat urine samples. Adult male rats were subjected to a 28 day subchronic study (0, 6, 20 and 60 mg kg-1 ) via oral gavage. Urine was collected twice a day on days 0, 7, 14, 21 and 28; organs underwent histopathological examination. Changes in body weight and relative kidney weights in medium- and high-dose groups were significantly different compared to controls. Morphological alterations were evident in the kidneys and liver. Metabolomic results demonstrated that MA exposure increases the urinary concentrations of 8-hydroxy-2'-deoxyguanosine, 8-nitroguanine and 8-iso-prostaglandin F2α ; levels of acetoacetate, hippurate, alanine and acetate demonstrated time- and dose-dependent variations in the treatment groups. Findings suggest that MA consumption escalates oxidative damage, membrane lipid destruction and disrupt energy metabolism. These aforementioned changes in biomarkers and endogenous metabolites elucidate and assist in characterizing the possible mechanisms by which MA induces nephro- and hepatotoxicity.

OBJECTIVE: To assess the prognostic value of texture analysis for single hepatocellular carcinomas (HCCs) after hepatectomy. MATERIALS AND METHODS: A total of 61 HCC patients were enrolled in this retrospective study. Textural characteristics of the computed tomography (CT) images were quantified. The differences between the hepatic arterial phase and the portal venous phase were obtained (the Dif.). The receiver operating characteristic (ROC) curves were used for data screening. Cox regression analyses were performed to determine independent factors adjusted with the derived clinical and radiological variables. Model identifications were based on Akaike information criteria. Kaplan-Meier and log-rank tests were performed for overall survival (OS) and disease-free survival (DFS). RESULTS: ROC and Cox regression analyses identified five parameters. Filter 1.0 achieved the best performance, in which the Dif.Scale 1.2 was a superior indicative independent marker for OS (p=0.05). Kaplan-Meier analyses further demonstrated that the Dif.Scale2.2 at filter 0 (p=0.001), Dif.Scale1.2 (p=0.006), Dif.Scale3.2 (p=0.005) at filter 1.0, Dif.Wavelet 8 at filter 1.5 (p<0.001), and corona (p=0.032) were associated with OS. Moreover, Dif.Scale 2.2 at filter 0 (p=0.039), Dif.Scale1.2 at filter 1.0 (p=0.001), and Dif.Wavelet 8 at filter 1.5 (p=0.007) were associated with DFS, while the Barcelona-Clínic Liver Cancer (BCLC) parameters showed no statistical correlation with OS (p=0.057). CONCLUSIONS: For patients with a single HCC treated by hepatectomy, the textural features for Gabor and Wavelet, especially the varying Dif., potentially provided prognostic information beyond traditional indicators such as those of the BCLC.
