RESUMO

Sleep problems among frontline medical staff during the COVID-19 epidemic require attention. A total of 249 frontline medical staff who were recruited to support Wuhan completed this cross-sectional study. A web-based questionnaire about insomnia, depression, anxiety, and fatigue was used to assess mental health status. The prevalence of sleep disorders among frontline medical staff was 50.6%. More time spent in Wuhan and a history of insomnia, depression, anxiety, and fatigue were associated with a higher risk of insomnia. People who stayed in Wuhan for a long time with a history of insomnia, depression, anxiety, and fatigue symptoms might be at high risk of insomnia.

RESUMO

The establishment of endometrial receptivity is a prerequisite for successful pregnancy. Women with adenomyosis possess a lower chance of clinical pregnancy after assisted reproductive technology, which is partially due to impaired endometrial receptivity. The establishment of endometrial receptivity requires the participation of multiple processes, and proper endometrial epithelial cell (EEC) proliferation is indispensable. Monoamine oxidase A (MAOA) is a key molecule that regulates neurotransmitter metabolism in the nervous system. In the present study, we demonstrated a novel role for MAOA in the establishment of endometrial receptivity in women with adenomyosis and in an adenomyotic mouse model. Attenuated MAOA impairs endometrial receptivity by promoting inappropriate proliferation of EECs via the downregulation of FOXO1 during the window of implantation. These results revealed that MAOA plays a vital role in endometrial receptivity in female reproduction.

Cyanidin-3-glucoside (C3G), an anthocyanin constituent of fruits and vegetables. It has been proven to possess numerous health benefits with no side effects. However, the poor stability of C3G is an intractable property that limits its application. Hence, the aim of this study is to improve the stability of C3G through the formation of well dispersed nanoparticles. In this study, C3G loaded ß-CD-EP-CMC nanoparticles exhibited nearly spherical with good disperse and homogeneous morphology. Results also indicated that the nanoparticles formation of grafting of C3G to ß-CD-EP-CMC could significantly improve the stability of C3G to against thermal or light degradation. Collectively, current results strongly aligned with the prospective purpose that the grafting of C3G to ß-CD-EP-CMC nanoparticles could be treated as an effective approach for improving the stability. This study opens a new avenue for the utilization and development of novel wall materials ß-CD-EP-CMC in C3G associated nutraceutical.

Although the use of omethoate (OMT) for pests control is enormously economically beneficial for agricultural production, the high toxicity of OMT to nontarget organisms has resulted in the contamination of soil, river water, and food materials. Developing sensitive and convenient techniques to detect OMT residues is vital to society. Electrochemiluminescence (ECL) is a powerful analytical tool and has been widely applied in biosensors. To boost the co-reaction efficiency and ECL intensity, we introduced defective ZIF-8 as the novel cathodic luminophore. At the same time, defect generated by the doping of MoTe2 nanoparticles into ZIF-8 could easily electrocatalytic reduce the co-reactor S2O82- to SO4•-. Hence, based on the catalysis of defective ZIF-8, the ECL intensity of MoTe2/ZIF-8 nanocomposites is much higher than both ZIF-8 and MoTe2 nanoparticles. By integration of as-prepared materials with specificity omethoate aptamer, the ECL sensor showed a broad linear range (10-10 g L-1 and 10-5 g L-1) and a comparatively low detection limit (3.3 × 10-11 g L-1). Besides, the ECL aptasensor appeared a good practical performance to detect potato and spinach extraction samples, which proposed a promising guideline for developing ECL aptasensors with high efficiency.

Mesenchymal progenitor cells are broadly distributed across perivascular niches-an observation conserved between species. One common histologic zone with a high frequency of mesenchymal progenitor cells within mammalian tissues is the tunica adventitia, the outer layer of blood vessel walls populated by cells with a fibroblastic morphology. The diversity and functions of (re)generative cells present in this outermost perivascular niche are under intense investigation; we have reviewed herein our current knowledge of adventitial cell potential with a somewhat narrow focus on bone formation. Antigens of interest to functionally segregate adventicytes are discussed, including CD10, CD107a, aldehyde dehydrogenase isoforms, and CD140a among others. Purified adventicytes (such as CD10+ , CD107alow , and CD140a+ cells) have stronger osteogenic potential and promote bone formation in vivo. Recent bone tissue engineering applications of adventitial cells are also presented. A better understanding of perivascular progenitor cell subsets may represent a beneficial advance for future efforts in tissue repair and bioengineering.

Although adequate periconceptional folic acid (FA) supplementation has reduced the occurrence of pregnancies affected by neural tube defects (NTDs), the mechanisms underlying FA-resistant NTDs are poorly understood, and thus NTDs still remain a global public health concern. A high level of Krüppel-like factor 12 (KLF12) exerts deleterious effects on heath in most cases, but evidence for its roles in development has not been published. We observed KLF12-overexpressing mice showed disturbed neural tube development. KLF12-overexpressing fetuses died in utero at approximately 10.5 days post-coitus, with 100% presenting cranial NTDs. Neither FA nor formate promoted normal neural tube closure in mutant fetuses. The RNA-seq results showed that a high level of KLF12 caused NTDs in mice via overactivating the sonic hedgehog (Shh) signaling pathway, leading to the upregulation of patched 1, GLI-Krüppel family member GLI1, hedgehog-interacting protein, etc., whereas FA metabolism-related enzymes did not express differently. PF-5274857, an antagonist of the Shh signaling pathway, significantly promoted dorsolateral hinge point formation and partially rescued the NTDs. The regulatory hierarchy between a high level of KLF12 and FA-resistant NTDs might provide new insights into the diagnosis and treatment of unexplained NTDs in the future.

AIMS: Increasing evidence has shown that hormone secretion is regulated by endocytosis. Eps15 homology domain-containing protein 3 (EHD3) is an endocytic-trafficking regulatory protein, but whether EHD3 is associated with testosterone secretion is not clear. This work aims to explore the role of EHD3 in testosterone synthesis. MAIN METHODS: Testosterone concentration was determined by ELISA. The effects of EHD3 on endocytosis were assessed by exosomes tracing assay and Immunofluorescence. Targeting relationship between EHD3 and NR5A1 was verified by chromatin immunoprecipitation (ChIP) and dual luciferase reporter gene assay in Leydig cells. For in vivo assessments, conditional NR5A1 knockout mouse model was established with CRISPR/Cas9 gene targeting technology. KEY FINDINGS: EHD3 overexpression significantly increased the concentration of testosterone. EHD3 knockdown markedly decreased testosterone synthesis by reducing endocytosis. The activity of the EHD3 promoter was positively regulated by NR5A1, which occupied the conserved sequence "AGGTCA" in the EHD3 promoter. Furthermore, mice with a Leydig cell-specific conditional NR5A1 knockout displayed the blunted levels of EHD3 and clathrin (a key factor for endocytosis), and serum testosterone concentration compared with NR5A1f/f mice. SIGNIFICANCE: This study suggests a potential molecular mechanism of testosterone synthesis to fully understand male reproductive health.

This paper applied mesoporous metal-organic frameworks (MOFs) of UiO-66 particles for pH-responsive doxorubicin (DOX) delivery and cancer treatment. Mesoporous structured UiO-66 MOFs were synthesized, and carboxymethylcellulose (CMC) was loaded for sensitive pH response and also as a linker to encapsulate the chemotherapeutic drug of DOX. The composite of UiO-66/CMC@DOX was synthesized, and the loading capacity was as high as 45µg DOX per mg of the carrier. The structure and crystalization of the UiO-66 MOFs were determined by the Transmitting Electron Microscope (TEM) and x-ray diffraction methods, while the loading of CMC and DOX was inspected by Fourier Transform InfraRed (FT-IR) and UV-vis spectroscopy. The DOX release from UiO-66/CMC@DOX was tested under different pH at 37 °C. The DOX accumulative release could reach 78% under the pH of 5. A lower pH was more favorable for DOX release due to the CMC shrinking and higher DOX solubility in an acidic environment. The cytotoxicity study indicated that, under the DOX concentration of 4µg ml-1, the A549 cell (Lung Carcinoma Cell Line) viability of UiO-66/CMC was 28%, which was lower than that from free DOX solution (47%). UiO-66 MOFs were demonstrated to be an efficient drug delivery carrier for chemotherapeutic drug and release.

BACKGROUND: Although exercise has beneficial effects on metabolic syndrome (Mets), the effects of exercise training on endothelial function and irisin level in the elderly with MetS remain controversial. The aim of this study is to evaluate the influence of exercise training on endothelial function and serum irisin level in the elderly with MetS. METHODS: Volunteers with MetS were recruited (n = 30). After 8 consecutive weeks of exercise training, flow-mediated dilation (FMD), colony-forming units (CFU) of endothelial progenitor cell (EPC), and serum irisin level were determined. Body mass index (BMI), Homeostasis Model of Assessment of Insulin Resistance (HOMA-IR), and inflammatory markers were detected. RESULTS: Exercise training program improved endothelial function, increased EPC-CFU, decreased HOMA-IR and inflammatory markers in patients with metabolic syndrome. However, exercise training had no influence on the serum irisin level. Multivariate linear regression analysis showed that change of EPC-CFU and HOMA-IR were independent prognostic risk factors for change of FMD. CONCLUSIONS: Exercise training improved endothelial function, alleviated insulin resistance and inflammation in the elderly with MetS. The findings suggest that exercise training program is an effective means for the treatment of MetS in the elderly.

Oocyte quality typically begins to decline with aging, which contributes to subfertility and infertility. However, there is still no effective treatment to restore the ovarian reserve and improve aged-oocyte quality. According to the present study, growth hormone (GH) secretion changes with maternal age in female mice. After intraperitoneal injection with GH (1 mg/kg body weight) every two days for two months, the 10-month-old mice showed a better ovarian reserve and oocyte quality than control mice. GH treatment decreased the occurrence rate of aneuploidy caused by spindle/chromosome defects. Additionally, the single oocyte transcriptome analysis indicated that GH decreased the expression of apoptosis-related genes in oocytes. It was also observed that GH treatment reduced the expression of γH2AX and apoptosis of aged oocytes via decreasing the activation of Fos and Jun. Collectively, our results indicate that GH treatment is an effective way to reverse the age-associated depletion of ovarian reserve and the decline of oocyte quality by decreasing apoptosis.

Family accommodation is a phenomenon that has been associated with worse treatment outcome of patients with obsessive-compulsive disorder (OCD) and greater severity of symptoms and levels of functional impairment. Yet, there are no Chinese scales to assess family accommodation in OCD among family members. The present study aimed to illustrate the steps of translation and cross-cultural adaptation of the Chinese versions of the Family Accommodation Scale (FAS). After obtaining authorization of the developers, the Chinese versions of the FAS were translated and adapted from the English versions based on a standard protocol, following six steps: forward translation, pilot administration, language adjustment and cultural adaptation, back-translation, review and minor edit, and final approval of the developer. Thirty-five pairs of patients and corresponding relatives with different education levels were administered the FAS in the pretest stage. This study found that the semantic, idiomatic, and conceptual equivalence were obtained between the Chinese versions and original English scales, and the Chinese versions of FAS were well translated and culturally adapted. We also found that the Chinese versions of the FAS can be easily understood by people of different socioeconomic statuses.

PROBLEM: The definition of chronic endometritis (CE) differs among studies, and currently, there is no accepted consensus. This study aimed to establish the minimum number of immunohistochemical analysis of CD138+ plasma cells to identify a clinically relevant CE. METHOD OF STUDY: We performed a retrospective study on 716 infertile patients who never did CE analysis and respective antibiotic treatment before. Samples were obtained by endometrial scratching in the mid-luteal phase before IVF-ET treatment. The number and distribution of CD138+ cells were analyzed by immunohistochemistry. Thirty high-power fields (HPF) were evaluated for each sample. Patients were classified in 2 main groups: (a) CD138low (<5 CD138+ cells in all HPFs), (b) CD138high (≥5 CD138+ cells in at least one HPF). Pregnancy outcome was compared among the groups. RESULTS: In the CD138high group, ß-hCG positive rate, clinical pregnancy rate and live birth rate were significantly decreased (P = .04, P = .01, P = .04, respectively). Also after adjusting for patient age, body mass index (BMI), and clinical characteristics, the ß-hCG positive rate (P = .05), clinical pregnancy rate (P = .01) and live birth rate (P = .02) were significantly lower in the CD138high than those in the CD138low group. Within the CD138low group, these parameters were not significantly different between patients without any plasma cells and patients with up to 4 plasma cells/HPF. CONCLUSION: We conclude that immunohistochemical analysis of CD138+ cells is a reliable method to detect CE which can be identified by the presence of ≥5 plasma cells in at least one out of 30 HPF.

STUDY QUESTION: What are the localization, characteristics and potential for tissue regeneration of two perivascular stem cells, namely CD34+ adventitial cells and CD146+ pericytes, in human endometrium? SUMMARY ANSWER: Human endometrial CD34+ adventitial cells (located in the outermost layer of blood vessels and mainly in the basal layer) and CD146+ pericytes showed mesenchymal stem cell (MSC) phenotypes in in vitro culture, but presented limited potential to regenerate endometrium. WHAT IS KNOWN ALREADY: Periodic endometrial regeneration is considered to be maintained by MSCs. Blood vessel wall, regarded as stem cell niche, harbors a large reserve of progenitor cells that may be integral to the origin of MSCs. However, a lack of validated markers has hampered the isolation of putative endometrial MSCs. Currently, CD146+ pericytes and Sushi Domain Containing 2 (SUSD2) positive cells have been identified in the endometrial perivascular region as sharing MSCs characteristics. STUDY DESIGN, SIZE, DURATION: The locations of adventitial cells and pericytes in the human endometrium were identified by immunofluorescence staining (n = 4). After CD34+CD146-CD45-CD56-CD144- adventitial cells and CD146+CD34-CD45-CD56-CD144- pericytes were isolated from the endometrium of normal women (n = 6) by fluorescence-activated cell sorting, their characteristics were investigated in culture. Adventitial cells and pericytes were induced to differentiate, respectively, into vascular endothelial-like cells or endometrial stromal-like cells in vitro, with their potential explored by in vivo xenotransplantation (n = 2 in each group) and eutopic transplantation (n = 2 in each group). PARTICIPANTS/MATERIALS, SETTING, METHODS: CD34+ adventitial cells and CD146+ pericytes were cultured in the inducing medium to differentiate into endothelial-like cells in vitro, and then analyzed for CD31, von Willebrand factor immunofluorescent staining and tube formation. They were also cultured to differentiate into endometrial stromal cells in vitro, with the expression of vimentin and CD13 being detected by western blot before and after induction, and the expression of prolactin and insulin-like growth factor-binding protein 1 being determined as well. Single dispersed CD34+ adventitial cells and CD146+ pericytes were respectively transplanted under the kidney capsule of NOG mice to investigate their differentiation potential in vivo. A eutopic transplantation model was constructed by grafting recellularized uterine matrix loaded up with CM-Dil labeled adventitial cells or pericytes into the injury region of nude rat's uterus. MAIN RESULTS AND THE ROLE OF CHANCE: CD34+ adventitial cells were mainly located at the outmost layer of endometrial large vessels, while CD146+ pericytes were found surrounding the inner endothelial cells of microvessels. A small proportion of CD34+ adventitial cells expressed SUSD2. The number of adventitial cells was ∼40 times higher than that of pericytes in the endometrium. Both adventitial cells and pericytes showed MSC phenotypes after in vitro culture. After in vitro induction into endometrial endothelial-like cells and stromal-like cells, adventitial cells showed higher plasticity than pericytes and a closer correlation with stromal-like cells. In the mouse xenotransplantation model, vimentin+ cells, CD31+ endothelial-like cells and CD146+ pericyte-like cells could be observed after adventitial cells were transplanted. CM-Dil-labeled adventitial cells or pericytes could survive in the immunocompromised nude rats after eutopic transplantation, and vimentin+ cells were detected. In addition, CM-Dil-labeled adventitial cells or pericytes did not express α-smooth muscle actin or E-cadherin after transplantation. LARGE SCALE DATA: N/A. LIMITATIONS, REASONS FOR CAUTION: CD34 was chosen as a novel marker to isolate adventitial cells from human endometrium according to previous literature. The association of endometrial CD34+ adventitial cells and SUSD2+ MSCs should be further investigated. WIDER IMPLICATIONS OF THE FINDINGS: The decellularized uterine matrix model might be useful in endometrial stem cell therapy. BACKGROUND: Family accommodation (FA) in obsessive-compulsive disorder (OCD) means that the relatives of patients are involved in, help or facilitate patients' ritual behaviors and avoidance, which is a frequent occurrence and underresearched phenomenon in China. Recent studies have suggested that FA is adversely associated with treatment response, contradictory to the goal of cognitive behavior therapy, a contributor to the maintenance of symptoms and increased symptom severity, and associated with low levels of family and social functioning. There is increasing interest and focus on establishing a family-based intervention for OCD treatment based on the inclusion of relatives by decreasing FA. The present study explored the psychometric properties of the Chinese version of the Family Accommodation Scale for OCD Interviewer-Rated (FAS-IR). METHOD: A total of 109 patients with OCD and 91 primary relatives were assessed in corresponding patient and family measures, and the FAS-IR was administered to relatives by trained interviewers. RESULTS: More than 90% of the relatives accommodated patients' symptoms with at least one kind of FA behavior over the previous week, and the incidence of extreme or everyday routines was as high as 59.3%. Exploratory factor analysis demonstrated two-factor structure for the whole scale, including (1) modification and facilitation, and (2) participation. Cronbach's alpha was 0.798 for the whole scale, and the interrater and test-retest reliability coefficients were 0.835 (95%CI: 0.603-0.937) and 0.882 (95%CI: 0.685-0.959), respectively. Convergent validity was supported in exploring FA and was associated with symptom severity, level of functional impairment and family functioning related to OCD. The FA was not significantly correlated with depressive symptoms rated by the patients, as evidence of acceptable divergent validity. There was no significant difference in FA total score based on patient gender, patient age, or relationship with patients. CONCLUSIONS: The Chinese version of the FAS-IR demonstrated excellent psychometric properties for assessing the degree of FA, suggesting that it is a useful and valuable instrument in clinical and research settings.

This invited commentary is the personal experience of a psychiatrist who assisted in Wuhan, China during the pandemic. From the personal perspective, it explains why psychiatrists need to go to Wuhan, discusses the psychological problems faced by the front-line medical staff and confirmed COVID-19 patients and the corresponding psychological interventions provided to them, describes the particularity and coping methods of psychological issues related to COVID-19 epidemic.

Dye sensitization is an alternative strategy to improve photoelectric activity of semiconductors and, particularly, to enhance the activity towards visible light domain. Herein, an artificial Z-scheme bipyridine ruthenium (Ru(bpy)32+) sensitizing narrow-gap bismuth oxy-iodide (BiOI) microspheres was constructed by a simple electrostatic interaction strategy for the first time. The electrochemical impedance spectroscopy (EIS) and photoluminescence (PL) analysis showed that this design of such Z-scheme structure was helpful to enhance the interfacial charge transfer and improve the photoelectric conversion efficiency. In addition, due to the sensitization of Ru(bpy)32+, the band gap was narrowed from 1.8 eV of BiOI microspheres to 1.3 eV of BiOI/Ru(bpy)32+ microspheres, leading to improve the utilization of visible light. So that, the photocurrent of the resulted BiOI/Ru(bpy)32+ was 13.0 times that of pure BiOI microspheres. In view of the outstanding photoelectrochemical (PEC) performance of BiOI/Ru(bpy)32+ and the high specificity of the aptamer, the PEC aptasensor for ampicillin (AMP) merits the excellent detection performance including a broad linear ranging from 1 × 10-7 nM to 100 nM as well as a low detection limit of 3.3 × 10-8 nM (S/N = 3). This work not only provides a novel way to construct and design highly efficient photoactive materials for PEC detection, but also broadens the application of Z-scheme in the field of sensing.

OBJECTIVE: To evaluate the mental health status of hospitalized patients with coronavirus disease 2019 (COVID-19) and to explore the related factors. METHOD: This was a cross-sectional survey among COVID-19 inpatients in two isolation wards of a designated hospital in Wuhan, China, from March 7, 2020, to March 24, 2020. Participants' demographic data, clinical data and levels of circulating inflammatory markers were collated. Mental health symptoms were evaluated with questionnaires, which included the Insomnia Severity Index (ISI) scale, the 9-item Patient Health Questionnaire (PHQ-9), the 7-item Generalized Anxiety Disorder (GAD-7) scale, and questions about patients' self-perceived illness severity. Multivariate linear regression analysis was performed to explore factors that associated with mental symptoms, and a structural equation model (SEM) was used to assess the possible relationships between those factors and the patients' mental health. RESULTS: Among the 85 participants, 45.9% had symptoms of depression (PHQ-9 ≥ 5), 38.8% had anxiety (GAD-7 ≥ 5), and 54.1% had insomnia (ISI ≥ 8). According to multivariate regression analysis, female sex, a higher level of interleukin (IL)-1ß and greater self-perceived illness severity were all significantly associated with a higher PHQ-9 score, higher GAD-7 score and higher ISI score. In addition, the disease duration and the neutrophil to lymphocyte ratio (NLR) were positively related to patients' self-perceived illness severity. The results of the SEM analyses suggested that sex (ß = 0.313, P < 0.001), self-perceived illness severity (ß = 0.411, P < 0.001) and levels of inflammatory markers (ß = 0.358, P = 0.002) had direct effects on patients' mental health. The disease duration (ß = 0.163, P = 0.003) and levels of inflammatory markers (ß = 0.101, P = 0.016) also indirectly affected patients' mental health, with self-perceived illness severity acting as a mediator. CONCLUSION: A majority of COVID-19 infected inpatients reported experiencing mental health disturbances. Female sex, disease duration, levels of inflammatory markers and self-perceived illness severity are factors that could be used to predict the severity of patients' mental symptoms.

Crabapple (Malus prunifolia Willd. Borkh) is a kind of wild apples with many health benefits. However, the utilization of crabapple fruit remains scarce, due to the poor stability of C3G. In this study, C3G loaded nanoparticles were established by chitosan (CS), chitosan oligosaccharides (CSO), and carboxymethyl chitosan (CMC) united with ionic crosslinking agent γ-Polyglutamic acid (PGA) or calcium chloride (CaCl2) to improve the stability of C3G. Results showed that C3G-loaded nanoparticles were exhibited nearly spherical with homogeneous morphology. Particularly, C3G-CMC-CaCl2 nanoparticles exhibited the highest encapsulation efficiency (53.88%) and loading efficiency (5.11%) with preferable particle size (180 nm), good stability (-19 mV) and blood compatibility. C3G-CMC-CaCl2 nanoparticles also revealed the highest releasing ratio (~75%) at pH 5.3 with stability. Present study established the chemical and cell biological basis for further application of C3G-loaded nanoparticles in nutraceutical and functional food fields, extending the application of crabapple in food processing with bioactive enhancement.

The weak antimicrobial property of multi-walled carbon nanotubes (MWCNTs) hampers their use as an antimicrobial material. In the report, Dihydromyricetin (DMY) coated multi-walled carbon nanotubes (MWCNTs-DMY) were prepared with poly(ethylene glycol) (PEG1000) as a linker and evaluated for inactivating pathogenic bacteria. UV-visible spectra, FT-IR, transmission electron microscopy (TEM), and thermal gravimetric analysis (TGA) were used to characterize the MWCNTs-DMY. It has been shown that PEG and DMY are covalently attached to MWCNTs with excellent stability. Vibrio parahaemolyticus was employed as the target bacterium. Comparing with the raw and the functionalized MWCNTs (r-MWCNTs and f-MWCNTs), MWCNTs-DMY exhibited stronger antibacterial ability. The MWCNTs-DMY composite showed up to 10-fold higher antimicrobial property than pristine MWCNTs. The deposited DMY was suggested to play an important role in the bactericidal action of MWCNTs-DMY. The results of this study demonstrated that MWCNTs-DMY composite can serve as an effective and economical antimicrobial material.
