BACKGROUND: In multiple myeloma (MM), impact of specific chromosomal translocations involving IgH (14q21 locus, including t(4;14), t(11;14), and t(14;16) etc.) has been explored extensively. However, over 15% MM patients harboring IgH translocation with undefined partners have long been ignored. METHODS: A prospective non-randomized cohort study with a total of 715 newly-diagnosed MM cases was conducted, 13.6% of whom were t(14;undefined) positive. The whole cohort was divided into four groups: no IgH split (47.7%); t(14;undefined) (13.6%); t(11;14) (17.6%); and t(4;14) or t(14;16) group (21.1%). RESULTS: Median OS for the four groups were 84.2, not reached (NR), 58.7, and 44.2 months respectively, with p values for t(14;undefined) vs. no IgH split, t(11;14), and t(4;14)/t(14;16) groups of 0.197, 0.022 and 0.001, respectively. In bortezomib-based group, the survival advantage gained by t(14;undefined) group was much more significant compared to t(11;14) and t(4;14)/t(14;16) groups. Importantly, t(14;undefined) turned out to be an independent predictive factor for longer OS of MM patients in multivariate analysis, especially in the context of bortezomib-treatment. Similar results were also observed in the PUMCH external validation cohort. CONCLUSION: Collectively, our data confirmed and externally validated the favorable prognosis of the t(14;undefined) groups, especially in the era of novel agents.

Highly efficient Ru-catalyzed selective C-C or C-O bond cleavage of polyols (e.g., crude glycerol) for N-hydroxyethylation or N-acetonylation of amines was achieved through the hydrogen-borrowing approach. A variety of amines were transformed to the desired amino alcohols/ketones in moderate-to-excellent yields, opening up new avenues for generation of oxygenated pharmaceuticals and fine chemicals from renewable raw materials. The use of new redox-active catalysts containing bisphosphine/thienylmethylamine ligands allows this hydrogen-borrowing system to be operated selectively under both basic and acidic conditions.

For vitellogenin (Vtg) absorption to occur, there needs to binding of the glycolipophosphoproteins to the oocyte membrane in oviparous species, including teleosts. The cDNAs encoding homologous Vtg receptors (VgRs) LR8- and Lrp13 were cloned from ovaries of the large yellow croaker (Larimichthys crocea), an economically important species in Chinese aquaculture. The full-length Lc-lr8-/lrp13 cDNAs contained 4266/3760 base pairs (bp) and the deduced Lc-LR8-/Lrp13 proteins had 844/1218 amino acids, respectively. The VgRs comprised a ligand-binding domain, an epidermal growth factor precursor homology domain, YWXD motifs forming a ß-propeller structure, and transmembrane and cytoplasmic domains. There was a marked relative abundance of Lc-lr8-/lrp13 transcripts in the tissues that were evaluated, with the largest abundance in the ovaries at Stage II of development. Furthermore, there was a lesser relative abundance of Lc-lr8-/lrp13 mRNA transcript during ovarian development (Stages II to IV). In situ hybridization technology was used to analyze decreasing relative abundance pattern of Lc-lr8-/lrp13 mRNA transcript during oogenesis in Stage II to IV of ovarian development. By combining mRNA relative abundance with morphological results, a model was developed to explain the reduction in Lc-lr8-/lrp13 mRNA transcript relative abundance during ovarian development. During the early developmental stages, transcription, translation, and differential accumulation of VgRs in previtellogenic and vitellogenic oocytes may occur and result in Vtg absorption in teleost oocytes. Overall, there is preliminary evidence indicating that at least two VgRs (Lc-LR8-/Lrp13) are present in the large yellow croaker and may be important for Vtg transport from the blood into the oocyte during ovarian development.

Isolated complex III (CIII) deficiencies are among the least frequently diagnosed mitochondrial disorders. Clinical symptoms range from isolated myopathy to severe multi-systemic disorders with early death and disability. To date, we know of pathogenic variants in genes encoding five out of 10 subunits and five out of 13 assembly factors of CIII. Here we describe rare bi-allelic variants in the gene of a catalytic subunit of CIII, UQCRFS1, which encodes the Rieske iron-sulfur protein, in two unrelated individuals. Affected children presented with low CIII activity in fibroblasts, lactic acidosis, fetal bradycardia, hypertrophic cardiomyopathy, and alopecia totalis. Studies in proband-derived fibroblasts showed a deleterious effect of the variants on UQCRFS1 protein abundance, mitochondrial import, CIII assembly, and cellular respiration. Complementation studies via lentiviral transduction and overexpression of wild-type UQCRFS1 restored mitochondrial function and rescued the cellular phenotype, confirming UQCRFS1 variants as causative for CIII deficiency. We demonstrate that mutations in UQCRFS1 can cause mitochondrial disease, and our results thereby expand the clinical and mutational spectrum of CIII deficiencies.

Larimichthys crocea is an economically important marine fish in China. To date, the molecular mechanisms underlying testicular development and spermatogenesis in L. crocea have not been thoroughly elucidated. In this study, we conducted a comparative transcriptome analysis between testes (TES) and pooled multiple tissues (PMT) (liver, spleen, heart, and kidney) from six male individuals. More than 54 million clean reads were yielded from TES and PMT libraries. After mapping to the draft genome of L. crocea, we acquired 25,787 genes from the transcriptome dataset. Expression analyses identified a total of 3853 differentially expressed genes (DEGs), including 2194 testes-biased genes (highly expressed in the TES) and 1659 somatic-biased genes (highly expressed in the PMT). The dataset was further annotated by blasting with multi-databases. Functional genes and enrichment pathways involved in spermatogenesis and testicular development were analyzed, such as the neuroactive ligand-receptor interaction pathway, gonadotropin-releasing hormone (GnRH) and mitogen-activated protein kinase (MAPK) signaling pathways, cell cycle pathway, and dynein, kinesin, myosin, actin, heat shock protein (hsp), synaptonemal complex protein 2 (sycp2), doublesex- and mab-3-related transcription factor 1 (dmrt1), spermatogenesis-associated genes (spata), DEAD-Box Helicases (ddx), tudor domain-containing protein (tdrd), and piwi genes. The candidate genes identified by this study lay the foundation for further studies into the molecular mechanisms underlying testicular development and spermatogenesis in L. crocea.

Phenotypic and functional heterogeneity is one of the hallmarks of human cancers. Tumor genotype variations among tumors within different patients are known as interpatient heterogeneity, and variability among multiple tumors of the same type arising in the same patient is referred to as intra-patient heterogeneity. Subpopulations of cancer cells with distinct phenotypic and molecular features within a tumor are called intratumor heterogeneity (ITH). Since Nowell proposed the clonal evolution of tumor cell populations in 1976, tumor heterogeneity, especially ITH, was actively studied. Research has focused on the genetic basis of cancer, particularly mutational activation of oncogenes or inactivation of tumor-suppressor genes (TSGs). The phenomenon of ITH is commonly explained by Darwinian-like clonal evolution of a single tumor. Despite the monoclonal origin of most cancers, new clones arise during tumor progression due to the continuous acquisition of mutations. It is clear that disruption of the "epigenetic machinery" plays an important role in cancer development. Aberrant epigenetic changes occur more frequently than gene mutations in human cancers. The epigenome is at the intersection of the environment and genome. Epigenetic dysregulation occurs in the earliest stage of cancer. The current trend of epigenetic therapy is to use epigenetic drugs to reverse and/or delay future resistance to cancer therapies. A majority of cancer therapies fail to achieve durable responses, which is often attributed to ITH. Epigenetic therapy may reverse drug resistance in heterogeneous cancer. Complete understanding of genetic and epigenetic heterogeneity may assist in designing combinations of targeted therapies based on molecular information extracted from individual tumors.

BACKGROUND: Synergistic therapy of tumor is a promising way in curing cancer and in order to achieve effective tumor therapy with real-time drug release monitoring, dynamic cellular imaging and antitumor activity. RESULTS: In this work, a polymeric nanoparticle with Forster resonance energy transfer (FRET) effect and chemo-photodynamic properties was fabricated as the drug vehicle. An amphiphilic polymer of cyclo(RGDfCSH) (cRGD)-poly(ethylene glycol) (PEG)-Poly(L-histidine) (PH)-poly(ε-caprolactone) (PCL)-Protoporphyrin (Por)-acting as both a photosensitizer for photodynamic therapy (PDT) and absorption of acceptor in FRET was synthesized and self-assembled into polymeric nanoparticles with epirubicin (EPI)-acting as an antitumor drug for chemotherapy and fluorescence of donor in FRET. Spherical EPI-loaded nanoparticles with the average size of 150 ± 2.4 nm was procured with negatively charged surface, pH sensitivity and high drug loading content (14.9 ± 1.5%). The cellular uptake of EPI-loaded cRGD-PEG-PH-PCL-Por was monitored in real time by the FRET effect between EPI and cRGD-PEG-PH-PCL-Por. The polymeric nanoparticles combined PDT and chemotherapy showed significant anticancer activity both in vitro (IC50 = 0.47 µg/mL) and better therapeutic efficacy than that of free EPI in vivo. CONCLUSIONS: This work provided a versatile strategy to fabricate nanoassemblies for intracellular tracking of drug release and synergistic chemo-photodynamic therapy.

An atom-precise Cu0-containing copper cluster, Cu14(C2B10H10S2)6(CH3CN)8 (abbreviated as Cu14-8CH3CN) is reported, which is synthesized via a simultaneous reduction strategy and fully characterized by single-crystal X-ray diffraction, ESI-TOF-MS, and X-ray photoelectron spectroscopy. Cu14-8CH3CN is the only copper cluster that has a virtually identical silver structural analog, i.e., Ag14(C2B10H10S2)6(CH3CN)8 (hereafter as Ag14-8CH3CN). Nevertheless, density functional theory calculations reveal that the electronic structure of Cu14-8CH3CN differs significantly from the superatom electronic configuration of Ag14-8CH3CN. Moreover, Cu14-8CH3CN shows room-temperature luminescence and good electrocatalytic activities in the ethanol oxidation reaction and detection of H2O2. This pair of unprecedented analogous molecular nanoscale systems offer an ideal platform to investigate the fundamental differences between copper and silver in terms of catalytic activity and optical properties.

Ziziphi Spinosae Semen (ZSS) has been used for treatment of insomnia in China for centuries. To reveal the influence of insomnia on the levels of the neurotransmitters including serotonin (5-HT), glutamic acid (Glu), γ-aminobutyric acid (GABA), noradrenaline (NE) and dopamine (DA), and to study the role of ZSS aqueous extract in the treatment of insomnia, an UPLC-ESI- MS/MS method was developed and validated for simultaneous determination of five neurotransmitters in the rat brain. The brain samples were pretreated by one-step direct protein precipitation with acetonitrile. The analytes were detected in positive mode with multiple reaction monitoring (MRM) and the procedure was completed in less than 10 min. The method showed a good linearity (R2 > 0.9967) with the other validation parameters were within acceptance range. The results indicated that the concentration of 5-HT, GABA and DA is significantly lower (P < 0.01) in para-chlorophenylalanine (PCPA)-induced insomnia rat model group, while Glu and NE significantly higher than those in control group (P < 0.01). Treatment with ZSS aqueous extract (4 or 8 g·kg-1·d-1 for seven days) could ameliorate the symptoms of insomnia by significantly changing the levels of the neurotransmitter parameters mentioned above. The data obtained in this study demonstrate that ZSS aqueous extract could ameliorate the symptoms of insomnia by modulating the levels of monoamines and amino acid neurotransmitters in the brain.

Recently, increasing evidence has suggested the association between gut dysbiosis and Alzheimer's disease (AD) progression, yet the role of gut microbiota in AD pathogenesis remains obscure. Herein, we provide a potential mechanistic link between gut microbiota dysbiosis and neuroinflammation in AD progression. Using AD mouse models, we discovered that, during AD progression, the alteration of gut microbiota composition leads to the peripheral accumulation of phenylalanine and isoleucine, which stimulates the differentiation and proliferation of pro-inflammatory T helper 1 (Th1) cells. The brain-infiltrated peripheral Th1 immune cells are associated with the M1 microglia activation, contributing to AD-associated neuroinflammation. Importantly, the elevation of phenylalanine and isoleucine concentrations and the increase of Th1 cell frequency in the blood were also observed in two small independent cohorts of patients with mild cognitive impairment (MCI) due to AD. Furthermore, GV-971, a sodium oligomannate that has demonstrated solid and consistent cognition improvement in a phase 3 clinical trial in China, suppresses gut dysbiosis and the associated phenylalanine/isoleucine accumulation, harnesses neuroinflammation and reverses the cognition impairment. Together, our findings highlight the role of gut dysbiosis-promoted neuroinflammation in AD progression and suggest a novel strategy for AD therapy by remodelling the gut microbiota.

With the increasing requirement of environmental protection, the development of lubricating materials with non-toxicity and good biodegradability becomes more and more significant. As the novel green nanomaterial derived from natural cellulose, cellulose nanocrystals (CNCs) in the present work were prepared from native cotton and added into polyalphaolefin (PAO) base oil as the lubricant additive. To improve the compatibility of CNCs with PAO, the surface of CNCs were grafted by stearoyl chains, which entangled with polyolefin chains and led to a good dispersibility and stability of the colloidal solution. This hybrid oil with the elevated viscosity improved the formation of lubricant film in the boundary lubrication regime. Combining with the mending effect of CNC particles on the surface roughness and scars, both the friction and the wear were dramatically reduced. Specifically, the introduction of 2 wt% modified nanocrystals (mCNC) in PAO base oil reduced the coefficient of friction (COF) by 30%. The results of this study suggest that cellulose nanocrystal is a promising ecofriendly and effective lubricant additive.

INTRODUCTION: In our previous study, we demonstrated that there might be correlations between laryngopharyngeal reflux disorder and the hue value of laryngoscopic images. And we found that different regions of larynx have different hue values. It was hypothesized that the degree of inflammation varies between different laryngeal regions, due to an acid reflux pattern. OBJECTIVE: The objective of this study was to compare the changing trends of hue values of different laryngeal regions in patients with laryngopharyngeal reflux (LPR) disease. METHODS: Ninety-seven patients, including 20 pH-positive, 19 pH-negative were tested for LPR through multichannel intraluminal impedance 24-hour pH monitoring, and 58 controls with reflux symptom index less than 13. Laryngoscopic images of all patients were obtained. The hue values of 7 areas of interest, including both sides of the true vocal folds, the false vocal folds, the arytenoids, and the interarytenoid space, were quantified using a hue calculation. The analysis of variance analysis was applied to find if there was significant difference between different groups within each region. RESULTS: (1) In the regions of both sides of the true vocal folds and interarytenoid, there was no significant difference between positive group and negative group; (2) in the regions of both sides of false vocal folds, there was no significant difference between negative group and control group; (3) in the regions of both sides of arytenoids, there was neither significant difference between positive group and negative group nor between negative group and control group; (4) in other comparisons, there were significant differences. CONCLUSION: Hue values of separate laryngeal regions are different. In negative group, the hue values of interarytenoid region are similar with positive group, and the sensitivity to the acid are different from different regions. The sensitivity in the true vocal folds may be present.

Prodrug self-assembled nanomedicines with cleavable moieties sensitive to intracellular stimuli have attracted great interest to pharmacists. In this paper, a docetaxel-hyaluronic acid (DTX-HA) conjugate with peptide, hydrazone bonds and disulfide in sequence, which were cleavable catalyzed by metalloproteinase (MMP), weak acidity and glutathione (GSH), respectively, were involved in a moiety as the linker to immobilize DTX on HA chains, the prodrugs were self-assembled into nanoparticles and utilized for cancer chemotherapy. The synthesis of conjugate was characterized by 1H NMR, ESI-TOF and GPC. The self-assembly of the conjugates was investigated via DLS and TEM. The release profiles revealed that the nanomedicine was disassociated to trigger the drug release in the simulated intracellular conditions of MMP, pH value and GSH, respectively. The in vitro anticancer activity of the nanomedicine with IC50 test, cytometry and confocal laser scanning microscopy exhibited efficient cellular uptake and apoptosis of cancer cells. The in vivo anticancer study of the nanomedicine in tumor-bearing nude mice showed promising therapeutic efficacy in magnificent inhibition tumor growth, long circulation time in pharmacokinetic and low toxicity to organs.

Plasminogen activator, tissue type (PLAT) and its inhibitor serpin family E member 1 (SERPINE1) cooperatively regulate PLAT activity in various reproductive processes. However, it is unknown whether this includes bovine oocyte maturation. We addressed this question in the present study by evaluating PLAT and SERPINE1 protein localization in immature cumulus-oocyte complexes (COCs), as well as PLAT mRNA and protein expression in cultured COCs after 0, 8, 16, and 24 h of in vitro maturation (IVM). We also examined the effects of PLAT and SERPINE1 on germinal vesicle breakdown (GVBD) and oocyte cyclic 3' 5' adenosine monophosphate (cAMP) levels, cumulus expansion index, and expansion-related gene expression in oocytes derived from bovine COCs cultured for 4, 8, and 12 h and in COCs cultured for 16 h. Both PLAT and SERPINE1 localized in cumulus cells but only the latter was detected in oocytes. PLAT and SERPINE1 transcript levels increased during IVM; however, from 8 to 16 h, the levels of PLAT remained stable whereas those of SERPINE1 increased, resulting in a decline in PLAT concentration. Additionally, PLAT delayed GVBD, increased oocyte cAMP levels, and blocked cumulus expansion and associated gene expression, which was reversed by SERPINE1 supplemented. Thus, PLAT delays bovine oocyte GVBD by enhancing oocyte cAMP levels during the first 8 h of IVM; suppression of PLAT activity via accumulation of SERPINE1 in COCs results in cumulus expansion from 8 to 16 h of IVM. These findings provide novel insights into the molecular mechanisms underlying in vitro bovine oocyte maturation.

The synthesis of atom-precise analogues of homometallic nanoclusters remains a great challenge. Herein we report the first pair of atom-precise copper/silver-thiolate halide cluster analogues, namely [Cu17/Ag17I3S(C2B10H10S2)6(CH3CN)11] (Cu17 and Ag17), obtained by bottom-up self-assembly and complete-metal-exchange-induced cluster-to-cluster transformation, respectively. The differences in optical absorption and emission of these analogues were fully elucidated by experimental and theoretical methods.

Two amino functionalized Metal-Organic Frameworks (MOFs), {[Zn(Py2TTz)(2-NH2-BDC)]·(DMF)}n (1) and {[Cd(Py2TTz)(2-NH2-BDC)]·(DMF)·0.5(H2O)}n (2) (where Py2TTz = 2,5-bis(4-pyridyl)thiazolo[5,4-d]thiazole, 2-NH2-BDC = 2-amino-1,4-benzenedicarboxylate, and DMF = N,N-dimethylformamide), were synthesized and characterized using the primary ligand 2-amino-1,4-benzenedicarboxylic acid (2-NH2-H2BDC) and the auxiliary ligand 2,5-bis(4-pyridyl)thiazolo[5,4-d]thiazole (Py2TTz). They possess similar 2-fold interpenetrated three-dimensional bipillared-layer framework structures composed of typical binuclear metal nodes, 2-NH2-BDC two-dimensional layers and Py2TTz bipillars. Notably, thiazole nitrogen atoms and pendant -NH2 groups are present in channels in the two frameworks. Given their good chemical stabilities, high thermal stabilities, and exposed nitrogen sites, gas adsorption and catalytic experiments of the two MOFs were performed. The results demonstrate that MOF 2 can selectively adsorb carbon dioxide gas; moreover, the two MOFs can be employed as recyclable heterogeneous catalysts for Knoevenagel condensation reactions under solvent-free conditions.

Although endoscopic submucosal dissection (ESD) is gradually becoming a first-line treatment for superficial esophageal neoplasms (SEN), strictures occur in almost 100% of cases after circumferential ESD. A standard method to prevent stricture has not been established. Thus, we propose a novel self-help inflatable balloon to prevent stricture. The new balloon was used by the patients themselves at home (4-5 times a day, duration of each procedure was approximately 15-20 min), and was removed when the defects were almost healed. From January 2018 to September 2018, eight patients who received circumferential ESD for SEN and underwent a novel self-help inflatable balloon to prevent stricture were enrolled. Median size of the mucosal defects was 76.3 mm (range: 50-90 mm). Median time for removing the self-help inflatable balloon was 94.6 days (range, 71-119 days). Only one (12.5%) patient experienced stricture, and three endoscopic balloon dilation sessions were carried out for this patient. All patients tolerated the balloon well, and none experienced perforation or delayed bleeding. The self-help inflatable balloon seems to show a high preventive effect against stricture in patients whose mucosal defect was no longer than 100 mm in length after esophageal circumferential ESD. This method is economic, feasible, and safe.

RESUMO

Functionalization of amines by using CO2 is of fundamental importance considering the abundance of amines and CO2 . In this context, the catalytic formylation and methylation of amines represent convenient and successful protocols for selective CO2 utilization as a C1 building block. This study represents the first example of selective catalytic double N-formylation of aryl amines by using a dinuclear Mn complex in the presence of phenylsilane. This robust system also allows for selective formylation and methylation of amines under a range of conditions.
