RESUMO

Diesel exhaust (DE) had been associated with cardiopulmonary toxicity and developmental toxicity. However, neonatal very early-in-life exposure had not been extensively studied previously. To investigate the potential effects of neonatal very early-in-life exposure to DE, a brand-new chicken embryo in ovo exposure model had been established, with which the cardiopulmonary effects of DE exposure via air cell infusion at embryonic day 18/19 (ED18/19) were assessed in hatchling chicks post-hatch 0-, 1-, or 2-weeks. Heart rates were assessed with electrocardiography. Cardiac and pulmonary morphologies were investigated with histopathological methods. Cardiopulmonary effects were explored with immunohistochemistry for alpha smooth muscle actin (alpha-SMA). In further investigations, the expression levels of phosphorylated AhR, serum levels of TGF-ß1, phosphorylated SMAD2/3 and phosphorylated p38MAPK were assessed in the lung tissues. Significantly elevated heart rates, increased right ventricular wall thickness and cardiac collagen deposition were observed in the hearts of exposed hatchling chicks. Significantly increased collagen deposition as well as increased vascular alpha-SMA layer thickness/decreased cavity area were observed in exposed animal lungs. These effects persisted up to two weeks post-hatch. Mechanistic studies revealed elevated phosphorylated AhR expression levels in 0-week and 1-week chicken lungs, while phosphorylated SMAD2/3 levels significantly increased in 0-week chicken lungs but decreased in 2-week chicken lungs following DE exposure. Phosphorylation of p38MAPK did not remarkably increase until 2-week post-hatch. In summary, the novel chicken neonatal very early-in-life exposure model effectively exposed the chicken embryos during the neonatal initial breathing, resulting in cardiopulmonary toxicity, which is associated with AHR, TGF-ß1 and MAPK signaling.

Carrier recombination is a crucial process governing the optical properties of a semiconductor. Although various theoretical approaches have been utilized to describe carrier behaviors, a quantitative understanding of the impact of defects and interfaces in low dimensional semiconductor systems is still elusive. Here, we develop a model system consisting of chemically tunable, highly luminescent halide perovskite nanocrystals to illustrate the role of carrier diffusion and material dimensionality on the carrier recombination kinetics and luminescence efficiency. Our advanced synthetic methods provide a well-controlled colloidal system consisting of nanocrystals with different aspect ratios, halide compositions, and surface conditions. Using this system, we reveal the scaling laws of photoluminescence quantum yield and radiative lifetime with respect to the aspect ratio of nanocrystals. The scaling laws derived herein are not only a phenomenological observation but proved a powerful tool disentangling the carrier dynamics of microscopic systems in a quantitative and interpretable manner. The investigation of our model system and theoretical formulation bring to light the dimensionality, as a hidden constraint on carrier dynamics, and identify the diffusion length as an important parameter that distinguishes nanoscale and macroscale carrier behaviors. The conceptual distinction in carrier dynamics in different dimensionality regimes informs new design rules for optical devices where complex microstructures are involved.

Pt-based alloy catalysts are promising candidates for fuel-cell applications, especially for cathodic oxygen reduction reaction (ORR) and anodic methanol oxidation reaction (MOR). The rational design of composition and morphology is crucial to promoting catalytic performances. Here, we report the synthesis of Pt-Co nanoframes via chemical etching of Co from solid rhombic dodecahedra. The obtained Pt-Co nanoframes exhibit excellent ORR mass activity in acidic electrolyte, which is as high as 0.40 A mgPt-1 initially and 0.34 A mgPt-1 after 10 000 potential cycles at 0.95 VRHE. Furthermore, their MOR mass activity in alkaline media is up to 4.28 A mgPt-1 and is 4-fold higher than that of commercial Pt/C catalyst. Experimental studies indicate that the weakened binding of intermediate carbonaceous poison contributes to the enhanced MOR behavior. More impressively, the Pt-Co nanoframes also demonstrate remarkable stability under long-term testing, which could be attributed to the negligible electrochemical Co dissolution.

Children's psychological and physiological responses to interparental conflict have received considerable attention due to their implications for later adjustment, yet limited research has investigated the interplay between these two response systems. This study investigates patterns of association between children's psychological responses (e.g., emotional distress) and cortisol reactivity to interparental conflict, including possible moderations by negative caregiving environment. Participants included 193 families (mother, father, and child). Parents completed questionnaires relating to their caregiving behaviors toward the child (107 girls and 86 boys, M age = 7.99 years, SD = 0.53 years) and children's psychological responses to interparental conflict. Children provided three saliva samples over the course of watching videos depicting conflicts between two adults, whom children were asked to pretend were their parents. Based on a series of Latent Growth Curve Models, only children's emotional responses to interparental conflict (indicated by increased distress) were associated with greater cortisol reactivity. Additionally, fathers' harsh parenting behavior moderated the relation between children's emotional reactivity and cortisol reactivity, yet the moderation effect was not found for mothers' parenting. Findings are discussed in terms of the importance of exploring both psychological and physiological reactivity to conflict and the possible moderating role of harsh parenting.

BACKGROUND: The challenge of using bioengineered liver lies in sustaining the quantity of high-quality hepatocytes and the vasculature for blood perfusion. We characterized the heparinization of a porcine decellularized liver scaffold (DLS) as a carrier to support hepatocyte angiogenesis, thereby developing functional and vascularized hepatic tissue useful to treat liver injury. METHOD: The porcine DLS was obtained by the removal of cellular components and then subjected to heparinization by the end-point attachment technique. The heparinized DLSs were recellularized with rat hepatocyte spheroids to construct engineered hepatic tissue. The hepatic tissue was heterotopically implanted in the omentum majus of a rat model with liver injury induced by carbon tetrachloride (CCl4 ). RESULTS: Hepatocyte spheroids in the heparinized DLS remained viable for at least 10 weeks in vivo. The entire scaffold was populated with hepatocytes and arranged well. The volume of the heparinized DLS group was expanded over 400-fold. Liver-specific functions such as albumin synthesis, glycogen storage and cytochrome P 3A4 activity were highly expressed in the hepatic tissue. In addition, endothelial cells were recruited, as shown by CD31 staining, and new blood vessels formed, as visualized by fluorescein isothiocyanate-labelled dextran intravital confocal microscopy. The heparinized bioengineered hepatic tissue alleviated CCl4 -induced liver injury by regulating the deposition and degradation of the extracellular matrix. CONCLUSION: Primary hepatocyte spheroids survived for an extended time on the heparinized DLS and expanded to generate vascularized and functional bioengineered hepatic tissue that can alleviate liver injury in rats.

The self-assembly of nanoparticles, a process whereby nanocrystal building blocks organize into even more ordered superstructures, is of great interest to nanoscience. Here we report the layer-by-layer assembly of 2D perovskite nanosheet building blocks. Structural analysis reveals that the assembled superlattice nanocrystals match with the layered Ruddlesden-Popper perovskite phase. This assembly proves reversible, as these superlattice nanocrystals can be reversibly exfoliated back into their building blocks via sonication. This study demonstrates the opportunity to further understand and exploit thermodynamics to increase order in a system of nanoparticles and to study emergent optical properties of a superlattice from 2D, weakly attracted, perovskite building blocks.

Perfluorooctanoic acid (PFOA) is a widespread persistent organic pollutant and may induce developmental toxicities, including developmental cardiotoxicity. To explore the potential mechanism of developmental cardiotoxicity induced by PFOA exposure, chicken embryo primary cardiomyocytes were extracted either from chicken embryos pretreated with PFOA (2 mg/kg), or from untreated embryos and then directly exposed cells to PFOA (1, 10, 30 or 100 µg/ml) in culture. Additionally, peroxisome proliferator activated receptor alpha (PPAR alpha) silencing lentivirus was applied to the embryos on embryonic day (ED2). Cell viability was measured with CCK-8 kit, morphology was assessed with hematoxylin and eosin staining, and intracellular Ca2+ concentrations were determined with Fluo-4 AM probe. Western blotting was utilized to confirm PPAR alpha silencing efficiency and the protein abundance of Wnt5a and Frizzled2. The results indicated that both PFOA pretreatment and direct exposure decreased primary cardiomyocyte viability, altered cell morphology and increased intracellular Ca2+ concentrations. While l-carnitine co-treatment effectively abolished such changes, PPAR alpha silencing only abolished most of the changes in PFOA pretreatment group, but not in cells directly exposed to relatively high doses of PFOA. The protein abundance of Wnt5a and Frizzled2 was increased by PFOA pretreatment, while direct exposure to PFOA increased Frizzled2 abundance but decreased Wnt5a abundance. PPAR alpha silencing resulted in over 50% decrease of PPAR alpha expression level, which abolished the Wnt5a/Frizzled2 expression alterations following PFOA exposure. In conclusion, PFOA-induced primary cardiomyocyte toxicity is associated with PPAR alpha and Wnt5a/Frizzled2, in which PPAR alpha seems to play regulatory roles towards Wnt5a/Frizzled2.

Orientation selectivity is a fundamental property of visual cortical neurons and plays a crucial role in pattern perception. Although many studies have dedicated to explain how the orientation selectivity emerged, the mechanism underlying orientation selectivity is still not clear. In this work, we investigated the synchronization between spikes and local field potentials (LFP) in gamma band, with the aim of providing a new avenue to analyze the orientation selectivity. The experimental data were recorded by utilizing two chronically implanted multi-electrode arrays, where each array consisted of 48 electrodes and was placed over V1 and V4, respectively, in two macaques performing a selective visual attention task. An unbiased and robust measure for quantifying the synchronization between spikes and LFP was employed in the analysis process, which is termed as spike-triggered correlation matrix synchronization (SCMS) and performs reliably for limited samples of data. We observed the spike-LFP synchronization in three cases, i.e., spikes and LFP in V1, spikes and LFP in V4, spikes in V4 and LFP in V1. From the orientation tuning curves based on the spike-LFP synchronization, it is found that there is a strong correlation between the synchronization and grating orientation. The neurons in both V1 and V4 exhibit orientation selectivity, but V1 is stronger. In addition, the spike-LFP synchronization strength between V1 and V4 also shows orientation selectivity to drifting gratings. It means that the synchronization not only reflects the basic features of visual stimulation, but also describes the orientation tuning characteristics of neurons in different regions. Our results suggest that the spike-LFP synchronization can be used as an alternative and effective method to study the mechanism for generating orientation selectivity of visual neurons.

Little is known regarding the dynamic interactions between fathers and mothers in families of children with autism spectrum disorder (ASD) during the parenting process. This study used an actor-partner interdependence mediation (APIMeM) model to investigate the intrapersonal and interpersonal effects of emotion dysregulation and parenting stress on parenting behaviors among 211 pairs (total N = 422) of Chinese parents of children with ASD. The results indicated that for both fathers and mothers, there were significant indirect actor effects of parental emotion dysregulation on parents' own parenting behaviors through their own parenting stress. However, no significant direct or indirect partner effect was found in the analyses. These findings suggest that the emotional parenting dynamics occurred on the individual rather than the dyadic level in these families.

Ion exchange, as a postsynthetic transformation strategy, offers more flexibilities in controlling material compositions and structures beyond direct synthetic methodology. Observation of such transformation kinetics on the single-particle level with rich spatial and spectroscopic information has never been achieved. We report the quantitative imaging of anion exchange kinetics in individual single-crystalline halide perovskite nanoplates using confocal photoluminescence microscopy. We have systematically observed a symmetrical anion exchange pathway on the nanoplates with dependence on reaction time and plate thickness, which is governed by the crystal structure and the diffusion-limited transformation mechanism. Based on a reaction-diffusion model, the halide diffusion coefficient was estimated to be on the order of [Formula: see text] This diffusion-controlled mechanism leads to the formation of 2D perovskite heterostructures with spatially resolved coherent interface through the precisely controlled anion exchange reaction, offering a design protocol for tailoring functionalities of semiconductors at the nano-/microscale.

Objective: To assess the effects of marital conflict on parenting practices for mothers and fathers and to examine whether these effects differ for within-person and cross-person links in parental dyads. Background: Existing findings are mixed regarding the nature and magnitude of the association between marital conflict and childrearing behaviors. Little is known about parental role differences in this regard between fathers and mothers and the mutual influence on the other's responding. Method: A sample of 235 families (fathers, mothers, and their kindergarten children) participated in the study over a 2-year period. Fathers and mothers independently reported on constructive and destructive marital conflict tactics, as well as on their parenting behaviors in scenarios of children experiencing negative emotions. Results: Results indicated cross-person and within-person relations. For example, fathers' destructive conflict predicted mothers' distress reactions to children's negative emotions, supporting a spillover hypothesis. Mothers' destructive conflict behaviors predicted less unsupportive maternal parenting, supporting a compensatory hypothesis. Conclusion: Fathers' and mothers' marital conflict behaviors may have different implications for their own and their spouse' parenting. Implications: Intervention and prevention programs that target improving marital conflict interactions may also help promote positive parenting. The findings also support that both fathers and mothers should be included in these programs to increase the beneficial effects on parenting practices.

The shortage of human hepatocytes continues to be a significant limitation for the widespread application of hepatocyte transplantation and bioartificial liver (BAL) support therapy. Recombinant activation gene 2 (Rag2) and fumarylacetoacetate hydrolase (Fah)-deficient mice could be highly repopulated with human hepatocytes. However, Fah/Rag2-deficient mice can only produce up to 1 × 108 human hepatocytes per mouse. We hypothesized that 2-10 × 1010 human hepatocytes can be produced per Fah/Rag2-deficient pig, which is an adequate supply for hepatocyte transplantation and BAL therapy. In a novel approach, we used stably transfected Cas9 cells and single-guide RNA adenoviruses containing fluorescent reporters to enrich porcine cells with Fah/Rag2 dual gene mutations. This resulted in the construction of Fah/Rag2 double knockout porcine iliac artery endothelial cells, which were subsequently used for generating Fah/Rag2-deficient pigs.

A Ni-catalyzed reductive cross-coupling of α-hydroxycarbonyl compounds modified with oxalyl groups and aryl halides has been developed that furnishes α-aryl esters under mild conditions and tolerates a variety of functionalized aryl halides bearing electron-withdrawing and -donating groups. This work highlights C-O bond fragmentation on secondary alkyl carbon centers that generates α-carbonyl radicals.

Toward advancing the understanding of relations among family relationships when children transition into adolescence, this study investigated whether parent-child relationship (PCR) quality assessed at the daily level changed developmentally and/or fluctuated due to daily experiences. Specifically, this study examined (a) whether parents' daily perceptions of marital relationship (MR) quality were associated with their own and/or their partners' PCR on the same day and the following day, and (b) whether relations among these daily influences changed over a 2-year period as children developed. Participants recruited included 237 2-parent families with preadolescent or adolescent children (52% girls). Both fathers and mothers completed daily diaries about MR and PCR for 15 consecutive days each year for 3 consecutive years. Results indicated that daily PCR did not change developmentally but was subject to day-to-day variations based on parents' daily MR: parents' distressed MR was related to their own lower emotional quality in PCR on the same day (supporting the spillover hypothesis), but higher emotional quality in PCR on the next day (supporting the compensatory hypothesis). Compensatory association was also found between father-reported average MR and mother-reported daily PCR. Furthermore, the same-day spillover and cross-day compensatory effects tended to decrease developmentally, as children transitioned into adolescence. The findings illustrated the interdependent, changing, and dynamic patterns of family relationships and underscored the importance of differentiating between father-child and mother-child relationship. The natural liver extracellular matrix (ECM) achieved by decellularization holds great potential in the fields of tissue engineering and regenerative medicine. Additionally, the use of crosslinking agents on the ECM to stabilize its ultrastructure and enhance scaffold durability is gaining interest in tissue engineering. The objective of this study was to compare the scaffold properties of porcine liver ECM crosslinked with different agents (glutaraldehyde, genipin, and quercetin) to find the best strategy for producing a decellularized matrix with optimal and stable characteristics for transplantation and regeneration. The properties examined include mechanical properties, material stability, immunogenicity, and angiogenic capacity. Scaffolds were implanted into the greater omentum of rats, and their abilities to induce immune cell subpopulation invasion and neovascularization were evaluated. The results show that genipin crosslinking of decellularized liver matrices increased the mechanical and proangiogenic properties and reduced the inflammatory response in vivo.

This study aims to evaluate the effectiveness and safety of the spheroid reservoir bioartificial liver (SRBAL) with porcine hepatocyte organoids in a preclinical nonhuman primate model of acute liver failure (ALF). Methods: Thirty healthy rhesus monkeys were infused with α-amanitin and lipopolysaccharide and randomized into five groups (ALF alone control group; sham no-cell SRBAL treatment group; groups A, B and C with SRBAL treatment started at 12 h, 24 h and 36 h after induction of ALF, respectively). Animals were continuously treated with the SRBAL device for 6 h and followed for up to 336 h. Results: Survival of ALF monkeys improved with hepatocyte SRBAL treatment compared to control groups. Blood ammonia and total bilirubin were lower, and albumin levels were higher in all hepatocyte SRBAL treatment groups. No evidence of porcine endogenous retrovirus was identified in monkey liver or blood after SRBAL treatment. Titers of monkey antibody (IgG, IgM) did not rise after SRBAL treatment. In survival cases, the proportion of necrotic and apoptotic hepatocytes was lower in SRBAL-treated groups, with earlier liver regeneration leading to recovery. Cytokines TNF-α, IL-6, IL-12, IL-1ß, IL-8, IFN-γ and IL-2 were ameliorated by the SRBAL treatment, while levels of M-CSF; HGF, EGF and VEGF; IL-1RA and MIF rose on priming, proliferation and the late phase of liver regeneration. Conclusions: The benefit of SRBAL therapy included preventive effects and therapeutic effects. SRBAL improved survival rate and prolonged median survival time in a nonhuman primate model of drug-induced ALF, and these benefits declined with a delay in the initiation of therapy. Improved survival and recovery of ALF monkeys was associated with a reduction in blood ammonia levels, inhibition of the pro-inflammatory response of ALF, and provided a microenvironment more suitable for regeneration of the injured liver.

As pigs are similar to humans in anatomy, physiology and pathology, nutrition metabolism and disease characteristics, genetically modified pigs are already used for the studies of disease mechanism, pathology and toxicology and the evaluation of drugs. But the production of large modified animals is difficult, cumbersome, time-consuming and costly. With the breakthrough of gene editing technology, clustered regularly interspersed short palindromic repeat (CRISPR)/CRISPR-associated 9( Cas9)(CRISPR/Cas9) technology has greatly improved the mutation efficiency, reduced the cost and simplified the steps, and promoted the widespread application of genetically modified pigs. In this paper, the production methods of genetically modified pigs and the research progress of genetically modified pigs by CRISPR/Cas9 in the medical field were reviewed.

Pt-based alloys have shown great promise as cathodic catalysts for cost-effective proton-exchange membrane fuel cells. Post-synthesis treatment has been recognized as a critical step to improve the catalytic performance of Pt-based alloys. Here, we present the effects of catalyst processing on the catalytic behavior of Pt-Ni nanoframe electrocatalysts in oxygen reduction reaction. The Pt-Ni nanoframes were made by corroding the Ni-rich phase from solid rhombic dodecahedral particles. A total of three different corrosion procedures were compared. Among them, electrochemical corrosion led to the highest initial specific activity (1.35 mA cm-2 at 0.95 V versus reversible hydrogen electrode) by retaining more Ni in the nanoframes. However, the high activity gradually went down in a subsequent stability test due to continuous Ni loss and concomitant surface reconstruction. On the other hand, the best stability was achieved by a more-aggressive corrosion using oxidative nitric acid. Although the initial activity was compromised, this procedure imparted a less-defective surface, and thus, the specific activity dropped by only 7% over 30 000 potential cycles. These results indicate a delicate trade-off between the activity and stability of Pt-Ni nanoframe electrocatalysts. The obtained understanding of how to balance the activity-stability trade-off via catalyst processing can be generalized to other Pt-based alloys.

Ever since its development, the Difficulties in Emotion Regulation Scale (DERS) has been widely adopted as a comprehensive measure to assess emotion regulation problems among English-speaking adults. To assess emotion regulation in adults from Chinese-speaking societies and to promote future cross-cultural examination of the emotion regulation processes, the authors aimed to develop a Chinese version of the DERS and provide an initial validation of this instrument. For the purpose of the current study, we recruited 862 Chinese adults from universities and local companies. The results indicated a similar six-factor solution in the Chinese version to the original version. Internal consistency and test-retest reliability were good. Concurrent validity was assessed by examining the correlations of the DERS and its subscales with measures of psychopathological symptoms and self-regulation of negative mood. The results demonstrated strong correlations of the DERS subscales with the Symptom Checklist-90 (SCL-90) and the Generalized Expectancy for Negative Mood Regulation Scale, except for that between the awareness subscale and the SCL-90. For the convergent validity, most DERS subscales were significantly correlated with personality traits, emotional intelligence, and self-control ability, with several exceptions. Bismuth selenide (Bi2Se3), with a wide bulk band gap and single massless Dirac cone at the surface, is a promising three-dimensional topological insulator. Bi2Se3 possesses gapless surface states and an insulator-like bulk band gap as a new type of quantum matter. Different Bi2Se3 nanostructures were prepared using electron beam evaporation with high production efficiency. Structural investigations by energy-dispersive X-ray analysis, scanning electron microscopy, and X-ray diffraction revealed the sample stoichiometries and the structural transition mechanism from nanocrystals to nanoflakes. The optical properties systematically probed and analyzed by spectroscopic ellipsometry showed strong dependence on the nanostructures and were also predicted to have structure-modifiable technological prospects. The optical parameters, plasma frequencies, scattering rates of the free electrons, and optical band gaps were related to the topological properties of the Bi2Se3 nanostructures via light-matter interactions, offering new opportunities and approaches for studies on topological insulators and spintronics. The high-quality Bi2Se3 nanostructures provide advantages in exploring novel physics and exploiting prospective applications.
