RESUMO

Signal transducer and activator of transcription 3 (STAT3) is a transcription factor that contributes to cancer progression through multiple processes of cancer development, which makes it an attractive target for cancer therapy. The IL-6/STAT3 pathway is associated with an advanced stage in colorectal cancer patients. In this study, we identified trichothecin (TCN) as a novel STAT3 inhibitor. TCN was found to bind to the SH2 domain of STAT3 and inhibit STAT3 activation and dimerization, thereby blocking STAT3 nuclear translocation and transcriptional activity. TCN did not affect phosphorylation levels of STAT1. TCN significantly inhibited cell growth, arrested cell cycle at the G0/G1 phase, and induced apoptosis in HCT 116 cells. In addition, the capacities of colony formation, migration, and invasion of HCT 116 cells were impaired upon exposure to TCN with or without IL-6 stimulation. In addition, TCN treatment abolished the tube formation of HUVEC cells in vitro. Taken together, these results highlight that TCN inhibits various cancer-related features in colorectal cancer development in vitro by targeting STAT3, indicating that TCN is a promising STAT3 inhibitor that deserves further exploration in the future.

Objective: The present study aimed to examine the relationship between physical fitness and academic performance, after controlling for potential confounds.Participants: The sample comprised 2,324 college students representing three Chinese universities.Methods: Physical fitness was measured using the Chinese National Student Physical Fitness Standard; academic performance (ie, Grade Point Average) and lifestyle behaviors were assessed using questionnaires. Academic performance and overall physical fitness were used to divide participants into three categories: low, moderate, and high.Results: Several physical fitness indicators systematically differed across the three academic performance categories. Additionally, overall physical fitness was positively associated with academic performance, even after controlling for the effects of lifestyle behaviors; those who belonged to the high overall physical fitness group had lower odds ratios of low academic performance than those belonging to the low overall physical fitness group.Conclusions: Poor academic performance was related to low overall physical fitness . The probability of having poor academic performance was significantly lower among students with high physical fitness than those with low physical fitness.

Remodeling of energy-storing white fat into energy-consuming beige fat has led to a promising new approach to alleviate adiposity. Several studies have shown adipokines can induce white adipose tissue (WAT) beiging through autocrine or paracrine actions. Betatrophin, a novel adipokine, has been linked to energy expenditure and lipolysis but not clearly clarified. Here, we using high-fat diet-induced obesity to determine how betatrophin modulate beiging and adiposity. We found that betatrophin-knockdown mice displayed less white fat mass and decreased plasma TG and NEFA levels. Consistently, inhibition of betatrophin leads to the phenotype change of adipocytes characterized by increased mitochondria contents, beige adipocytes and mitochondria biogenesis-specific markers both in vivo and in vitro. Of note, blocking AMP-activated protein kinase (AMPK) signaling pathway is able to abolish enhanced beige-like characteristics in betatrophin-knockdown adipocytes. Collectively, downregulation of betatrophin induces beiging in white adipocytes through activation of AMPK signaling pathway. These processes suggest betatrophin as a latent therapeutic target for obesity.

Self-powered smart windows are desirable with the expectations of their energy-saving, weather-independent, user-controllable, and miniature performance. Recently developed solar- or thermal-powered smart windows largely depend on the weather conditions and have an extremely slow response, and only a certain portion of the saved energy can be utilized by the external circuit for mode conversion. In this work, a self-powered normally transparent smart window was developed by the conjunction of a rotary freestanding sliding triboelectric nanogenerator (RFS-TENG) and a polymer network liquid crystal (PNLC) cell. To fabricate the PNLC cell, the alignment layer with randomly distributed microdomains was constructed to encapsulate a mixture of LC polymers and nematic LCs. The opacity of the smart window exposed to an alternating electric field was considerably improved owing to the embedded microdomains and a dense web of LC polymers. The ultrahigh haziness greatly alleviates the charge density required for the LC actuation and thus enables the driving by the TENG where the charge amount is usually limited. The RFS-TENG was elaborately designed with six periodic bent triboelectric films and Ag electrodes, which presented an ultralow friction wear and met the frequency requirement to achieve the steady opacity. By harvesting the mechanical energies from ambient environments, the tribo-induced smart window can benefit a wide variety of fields, such as self-powered sunroofs, wind-driven smart farming systems etc.

Ginsenoside compound K (CK), a major metabolite of protopanaxadiol ginsenosides, exhibits significant anticancer activities against various cancer cells. However, CK has poor water solubility and low bioavailability, which have limited its application. In this study, A54 peptide was utilized to fabricate CK-loaded micelles (APD-CK) for liver targeting, using deoxycholic acid-O-carboxymethyl chitosan as the vehicle. The average particle size of APD-CK micelles was about 171.4 nm by dynamic light scattering in the hydrated state and their morphology were spherical with good dispersion. An in vitro release assay indicated pH-responsive and sustained release behavior through a mechanism of non-Fickian diffusion. Moreover, the in vitro cytotoxicity of the APD-CK micelles against HepG2 and Huh-7 cells was significantly stronger than that of CK up to 20 µg/mL. Enhanced cellular uptake of micelles in both cell types was established using confocal fluorescence scanning microscopy and flow cytometry. In addition, western blot analysis revealed that APD-CK micelles could promote the protein expression levels of caspase-3, caspase-9, and poly (ADP-ribose) polymerase. Therefore, APD-CK micelles are a potential vehicle for delivering hydrophobic drugs in liver cancer therapy, enhancing drug targeting and anticancer activity.

Thrombotic disease is a serious threat to human health. The rapid and accurate detection of thrombosis is still a clinical challenge. To achieve the accurate diagnosis of thrombosis with magnetic resonance imaging (MRI), nanomaterials-based contrast agents have been developed in recent years. In this study, cyclic RGD functionalized liposomes targeted to the activated platelets are developed for thrombosis dual-mode MRI. The cyclic RGD functionalized liposomes (cRGD@MLP-Gd) encapsulated with gadolinium diethylenetriamine penta-acetic acid (Gd-DTPA) and superparamagnetic iron oxide (SPIO) are prepared, and their thrombus-targeted T1 and T2 MRI potential is evaluated in vitro and in vivo. Results show that cRGD@MLP-Gd could actively bind to the activated platelets and gradually accumulate at the thrombus site with a T1 - T2 contrast enhancement imaging effect in vitro. In in vivo MRI experiments, cRGD@MLP-Gd exhibits a T2 contrast enhancement at 1 h after intravenous administration, followed by a visibly larger T1 contrast enhancement at the thrombus site. This dynamic property showed that cRGD@MLP-Gd could actively bind to thrombus and possessed an enhanced T1 and T2 dual-mode MRI effect in vivo. Our results establish the characterization, feasibility and superiority of cRGD@MLP-Gd for the rapid identification of thrombosis, showing great potential to improve diagnostic accuracy and sensitivity to thrombosis of the MRI technique.

One new ß,γ-butenoate derivative phenylbutenote (1), and one new α-pyrone nocapyrone T (2) were isolated from the deep-sea derived actinomycete Nocardiopsis sp. HDN 17-237. Their structures were elucidated by extensive HRMS, IR and NMR analyses. Among them, compound 1 is the first microbial natural products bearing a rare ß,γ-butenoate moiety, and compound 2 is the first α-pyrone isolated from strain of Mariana Trench. Compounds 1 and 2 were tested for antioxidant and antibacterial activities, while none of them showed significant activity.

Background: Exercise modes can be divided into open skill exercise (OSE) and closed skill exercise (CSE). While research has shown that these two exercise modes may have different effects on cognitive function, this possibility has not been systematically reviewed. Objective: The purpose of the present review was to objectively evaluate the research literature regarding the effects of OSE versus CSE on cognitive function. Methods: Six electronic databases (Web of Science, EMBASE, Google Scholar, PubMed, PsycINFO, and SPORTDiscus) were searched from inception dates to December 2018 for studies examining the associations of OSE and CSE with cognitive function. The literature searches were conducted using the combinations of two groups of relevant search items related to exercise modes (i.e., OSE and CSE) and cognitive function. Articles were limited to human studies in all age groups. Both intervention and observational studies with full text published in English-language peer-reviewed journals were considered eligible. The search process, study selection, data extraction, and study quality assessment were carried out independently by two researchers. Results: A total of 1,573 articles were identified. Fourteen observational and five intervention studies met the inclusion criteria. Twelve of the 14 observational studies found that OSE benefits cognitive function, and seven of these 14 observational studies supported superior effects of OSE compared with CSE for enhancing cognitive function. Three of the five intervention studies found that OSE (versus CSE) led to greater improvements in cognitive function in both children and older adults. Conclusion: Although the majority of studies in this review were observational cross-sectional designs, the review tends to support that OSE is more effective for improving some aspects of cognitive function compared with CSE. More rigorous randomized control trials with long-term follow-ups are needed in order to confirm these differential cognitive effects of the two exercise modes.

OBJECTIVE: To determine whether the Bundled Payments for Care Improvement (BPCI) initiative affected patient-reported measures of quality. DATA SOURCES: Surveys of Medicare fee-for-service beneficiaries discharged from acute care hospitals participating in BPCI Model 2 and comparison hospitals between October 2014 and June 2017. Variables from Medicare administrative data and the Provider of Services file were used for sampling and risk adjustment. STUDY DESIGN: We estimated risk-adjusted differences in patient-reported measures of care experience and changes in functional status, for beneficiaries treated by BPCI and comparison hospitals. DATA COLLECTION: We selected a stratified random sample of BPCI and matched comparison beneficiaries. We fielded nine waves of surveys using a mail and phone protocol, yielding 29 193 BPCI and 29 913 comparison respondents. PRINCIPAL FINDINGS: Most BPCI and comparison survey respondents reported a positive care experience and high satisfaction. BPCI respondents were slightly less likely than comparison respondents to report positive care experience or high satisfaction. Despite these differences in care experience, there was no difference between BPCI and comparison respondents in self-reported functional status approximately 90 days after hospital discharge. CONCLUSIONS: These findings reduce concerns that BPCI may have unintentionally harmed patient health but suggest room for improvement in patient care experience.

BACKGROUND: This study aimed to investigate the effects of self-paced cycling at an active workstation on executive functions and cortical activity. METHODS: In a crossover study design, 37 young adults (45.9% females) were randomly assigned to the following two task conditions: (1) performing cognitive tests during sitting, (2) performing cognitive tests while cycling at an active workstation. Executive functions were assessed by the Stroop color and word test and the task-switching paradigm. Cortical activity was monitored using a multi-channel functional near-infrared spectroscopy (fNIRS) system. RESULTS: The behavioral results showed that there were no significant differences on the Stroop interference effects (P = 0.66) between the sitting and the cycling conditions. In all probability, no differences on the global switch costs (P = 0.90) and local switch costs (P = 0.67) were observed between the sitting and the cycling conditions. For the fNIRS results, the oxygenated hemoglobin (oxy-Hb) in response to the Stroop interference in channels 5, 10, and 12 were decreased during the cycling condition (all Ps < 0.05, FDR-corrected). Conversely, the oxy-Hb associated with the global switch costs in channels 3, 29, and 31 were increased during the cycling condition (all Ps < 0.05, FDR-corrected). CONCLUSIONS: The findings indicated that behavioral performances on executive functions were not affected by cycling at an active workstation, while cognitive resources were reallocated during cycling at an active workstation.

BACKGROUND: Species diversity is determined by both local environmental conditions that control differentiation and extinction and the outcome of large-scale processes that affect migration. The latter primarily comprises climatic change and dynamic landscape alteration. In the past few million years, both Southeast Asia and Eastern Africa experienced drastic climatic and geological oscillations: in Southeast Asia, especially in China, the Tibetan Plateau significantly rose up, and the flow of the Yangtze River was reversed. In East Africa, lakes and rivers experienced frequent range expansions and regressions due to the African mega-droughts. To test how such climatic and geological histories of both regions relate to their respective regional species and genetic diversity, a large scale comparative phylogeographic study is essential. Bellamya, a species rich freshwater snail genus that is widely distributed across China and East Africa, represents a suitable model system to address this question. We sequenced mitochondrial and nuclear DNA for members of the genus from China and used published sequences from Africa and some other locations in Asia to investigate their phylogeny and distribution of genetic diversity. RESULTS: Our phylogenetic analysis revealed two monophyletic groups, one in China and one in East Africa. Within the Chinese group, Bellamya species show little genetic differentiation. In contrast, we observe fairly deep divergence among the East African lakes with almost every lake possessing its unique clade. Our results show that strong divergence does not necessarily depend on intrinsic characteristics of a species, but rather is related to the landscape dynamics of a region. CONCLUSION: Our phylogenetic results suggest that the Bellamya in China and East Africa are independent phylogenetic clades with different evolutionary trajectories. The different climate and geological histories likely contributed to the diverging evolutionary patterns. Repeated range expansions and regressions of lakes likely contributed to the great divergence of Bellamya in East Africa, while reversal of the river courses and intermingling of different lineages had an opposite effect on Bellamya diversification in China.

Purpose: The expression of microRNA-505 (miR-505) has been investigated in various cancers; however, its effect and mechanism in relation to gastric cancer (GC) are yet to be determined. Thus, the current evaluation aimed to examine the expression and potential role of miR-505 in GC. Materials and methods: Quantitative real-time PCR was carried out to analyze miR-505 expression in GC cells and tissues. We observed that miR-505 is differentially expressed in GC cells following transfection of its mimics or inhibitors. Changes in cell invasion, cell proliferation, and epithelial-mesenchymal transition markers were measured. Results: These findings indicated that miR-505 expression is downregulated in both GC cell lines and GC tissues. In addition, knockdown miR-505 induced the invasion and proliferation of GC cells. Transfection of miR-505 mimics led to an elevation in N-cadherin expression but a decrease in E-cadherin expression. Furthermore, we have shown that miR-505 binds to the 3'-UTR region of Polo-like kinase-1. Conclusion: Our results indicated that miR-505 suppresses GC cell proliferation and invasion; it may be a valuable candidate gene for seeking therapy strategy for GC.

RESUMO

A new polyene compound (1) and a new diketopiperazine (2), as well as three known compounds (3-5), were isolated from the Antarctic marine-derived fungus Penicillium crustosum HDN153086. The structures of 1-5 were deduced based on MS, NMR and TD-DFT calculations of specific ECD spectra. These compounds were evaluated for their cytotoxic activities against K562 cell line and only compound 2 exhibited cytotoxicity against K562 cell, with IC50 value of 12.7 µM.

Ginsenoside compound K (CK) has been shown to exhibit anticancer properties. In this study, chitosan nanoparticles loaded with ginsenoside compound K (CK-NPs) were prepared as a delivery system using a self-assembly technique with amphipathic deoxycholic acid-O carboxymethyl chitosan as the carrier, which improved the water solubility of CK. By evaluating drug loading, entrapment efficiency, and in vitro release behavior, the feasibility of CK-NPs as a drug carrier nanoparticle for the treatment of human hepatic carcinoma cells (HepG2) was investigated. Result revealed that CK and CK-NPs showed a dose-dependent inhibitory effect on HepG2 cells with IC50 values of 23.33 and 16.58 µg/mL, respectively. Furthermore, fluorescence imaging demonstrated that CK-NPs promoted cellular uptake in vitro. Therefore, all results indicated that CK-NPs might be a novel drug delivery system to improve the solubility and enhance the cytotoxic and apoptotic potentials of CK for effective liver cancer chemotherapy.

OBJECTIVE: To enhance proxy survey responses for deceased cancer patients. DATA SOURCES: Two waves of surveys about care experiences of cancer chemotherapy patients (used for a value-based purchasing initiative), collected November 2016 through March 2017. STUDY DESIGN: Surveys were mailed to deceased patients (for proxy response) in the untailored first survey wave, and using a tailored strategy in the second wave. We compared proxy responses for deceased patients with and without the tailored approach. PRINCIPAL FINDINGS: A tailored survey approach improved the proxy response rate from 17.6 percent to 39.0 percent. CONCLUSIONS: Simple tailoring strategies can significantly enhance proxy response rates when surveying patients in a high mortality population. However, we were unable to address the impact of higher proxy response rates on response bias since we surveyed different patients in each wave.

An asymmetric organocatalytic direct arylation approach to construct arylated quaternary stereogenic centers with a catalyst loading of 1 mol % is reported. The formation of the hemiketal moiety in stabilizing the hydroquinone intermediate proves to be important in leading to hydroquinone products instead of oxidation quinone products obtained in previously reported methods. A series of structurally and stereochemically complex heterocyclic frameworks are obtained, including spiro-, dispiro-, fused, and bridged heterocycles.

In 1937, Drs. Moritz and Oldt described arteriolar injuries in the kidneys (and other viscera) in hypertension, across the age range, in both sexes, and, in different races. This hypothesis proposes that injuries to vasomotor nerves cause the arteriolar injury in the kidney in hypertension, (as well as that in the uterus in preeclampsia). Different patterns of perivascular hyalinisation in different viscera are clues to the varying causes and consequences of arteriolar injury. In the uterus there is a symmetrical, perivascular "halo of hyalinisation" that marks the lines of extension of regenerating, injured nerves to the placental bed, whereas in the kidney there is a disordered and asymmetrical "halo of hyalinisation" where persistent, and recurrent, increases in intravascular pressures interrupt development of regenerating nerves. Consequences of injuries to vasomotor nerves include releasing a "soup" of cytokines that cause regeneration of "new" nerves expressing primitive, pain and stretch receptors including TRPV-1 and P2X3 purinergic "stretch" receptors that may be significant in the afferent mechanism in preeclampsia. There is also concurrent, "background" hyperplasia of denervated tunica media and intima leading to narrowing of the arterioles and a further drive to hypertension through renal ischaemia (Goldblatt, 1942). These observations require support from animal studies and other investigations to establish causation. This hypothesis may provide a number of potential mechanisms that reinforce, or accelerate, the physiological processes that contribute to hypertension.

Metallic nanoparticle based disinfection represents a promising approach for microbial pollution control in drinking water and thus, biosynthesis of non precious metal nanoparticles is of considerable interest. Herein, an original and efficient route for directly microbial synthesis of copper nanoparticles (Cu-NPs) by Shewanella loihica PV-4 is described and their satisfactorily antimicrobial activities are established. Cu-NPs were successfully synthesized and most of them attaching on the bacterial cell surfaces suggested extracellular Cu(II) bioreduction mainly contributed to this biosynthesis. Using a suite of characterization methods, polycrystalline nature and face centered cubic lattice of Cu-NPs were revealed, with size in the range of 10-16 nm. With Cu-NPs dosage of 100 µg/mL and 105 CFU/mL fresh Escherichia coli suspension, the obtained antibacterial efficiency reached as high as 86.3 ±â€¯0.2% within 12 h. Cell damages were primarily caused by the generated reactive oxygen species with H2O2 playing significant roles. Both cell membrane and cytoplasm components were destroyed, while the key inactivation mechanisms were lipid peroxidation and DNA damage as concluded through correlation analysis. The cost-effective and eco-friendly biosynthesis of Cu-NPs with high antibacterial activities make them particularly attractive for drinking water disinfection.
