RESUMO

An ultrasensitive and versatile assay for biomarkers has been developed using graphene/gold nanoparticles (AuNPs) composites and single-particle inductively-coupled plasma/mass spectrometry (spICP-MS). Thrombin was chosen as a model biomarker for this study. AuNPs modified with thrombin aptamers were first non-selectively adsorbed onto the surface of graphene oxide (GO) to form GO/AuNPs composites. In the presence of thrombin, the AuNPs desorbed from the GO/AuNPs composites due to a conformation change of the thrombin aptamer after binding with thrombin. The desorbed AuNPs were proportional to the concentration of thrombin and could be quantified by spICP-MS. By counting the individual AuNPs in the spICP-MS measurement, the concentration of thrombin could be determined. This assay achieved an ultralow detection limit of 4.5 fM with a broad linear range from 10 fM to 100 pM. The method also showed excellent selectivity and reproducibility when a complex protein matrix was evaluated. Furthermore, the diversity and ready availability of ssDNA ligands make this method a versatile new technique for ultrasensitive detection of a wide variety of biomarkers in clinical diagnostics.

OBJECTIVE: To investigate the proliferation inhibition and pro-apoptotic effect of Huaier aqueous extract combined with routine chemotherapeutic drugs including Vincristine (VCR), Daunorubicin (DNR), L-aspartase (L-Asp) on human acute lymphoblastic leukemia cell lines Nalm-6 and Sup-B15. METHODS: Nalm-6 and Sup-B15 cell lines were treated with different concentrations of Huaier aqueous extract and chemotherapeutics including VCR, DNR, L-Asp alone or in combination for 48 h, and the growth inhibitory effect and IC50 values (the half maximal inhibitory concentration) were detected by CCK-8. Jin's formula was used to estimated the synergistic effect of these combinations. Apoptosis rates of Nalm-6 and Sup-B15 cells and expression of apoptosis-related proteins BAX, BCL-2, cleaved Caspase-3 were determined by flow cytometry and Western blot respectivcly. RESULTS: Huaier aqueous extract, VCR, DNR and L-Asp had inhibition effect on Nalm-6 and Sup-B15 cell lines. The inhibition rate of Huaier aqueous extract combined with VCR, DNR and L-Asp were all higher than those of each dug alone (P<0.05) and the combination index (q) was between 0.85 and 1.15 or greater than 1.15. The two kinds of drugs showed had additive or synergistic effects. The results of flow cytometry showed that the cell apoptosis rates in combined treatment group were higher than those of each drug alone (P<0.05). The results of Western blot revealed that Huaier aqueous extract and VCR all decreased protein expression of BCL-2 (P<0.05) and increase protein expression of BAX (P<0.05) and cleaved Caspase-3 (P<0.05) in Nalm-6 and Sup-B15 cells. Compared with Huaier aqueous extract or VCR alone, the effect of two drug combination were more significant. DNR down-regulated protein expression of BCL-2 (P<0.05) and up-regulated cleaved Caspase-3 (P<0.05). However, it had no effect on the expression of BAX in Nalm-6 and Sup-B15 cells. When it was combined with Huaier aqueous extract, the expression of cleaved Caspase-3 and BCL-2 showed more significant changes. The expression of BAX in combined treated group did not show significant difference, compared with group treated with Huaier aqueous extract in Nalm-6 and Sup-B15 cells. L-Asp did not show significant effect on the three apoptosis-related proteins and there was no significant difference between the combination group and the Huaier aqueous extract group. CONCLUSION: the combination of Huaier aqueous extract and VCR, DNR, L-Asp shows additive or synergistic effects on human acute lymphoblastic leukemia cell lines Nalm-6 and Sup-B15.

OBJECTIVE: To investigate the relationships among personality traits, resilience and depressive symptoms of primary and high school students. METHODS: Totally 6019 students aged 10-17 from 5 primary schools(grades 5-6), 3 junior middle schools(grades 7-9) and 2 senior high schools(grade 1) years were selected by cluster sampling in Wuhan, from September 2015 to January 2016. Among them, there were 2420 primary school students, 2912 junior high school students and 687 senior high school students. In addition, 3071 students were male, 2948 students were female. Participants were asked to complete self-report questionnaires, including demographic characteristic questionnaire, the center for epidemiological studies depression scale(CES-D), the connor davidson resilience scale(CD-RISC) and the NEO-five factor inventory(NEO-FFI). Multivariate Logistic regression analysis was used to analyze the influence factors of depressive symptoms in primary and high school students. RESULTS: The detection rate of depressive symptoms was 10. 5%(635/6019). Multivariate Logistic regression analysis showed that after adjusting for grade and family history of depression, neuroticism(OR=4. 53, 95% CI 3. 88-5. 28) and openness(OR=1. 33, 95% CI 1. 18-1. 50) were positively associated with depressive symptoms. But the higher level of extraversion(OR=0. 70, 95% CI 0. 62-0. 79) and conscientiousness(OR=0. 77, 95% CI 0. 67-0. 90) and resilience(OR=0. 77, 95% CI 0. 67-0. 88) were associated with lower risk of depressive symptoms in primary and high school students. CONCLUSION: Neuroticism and openness might be positively correlated with, whereas extraversion, conscientiousness and resilience might be negatively correlated with the onsets of depressive symptoms in primary and secondary school students. Thus, developing adaptive personality and improving resilience would contribute to the prevention and intervention of depression in primary and high school students.

Introducing an efficient method for the rapid conversion of starch into gluconic acid is desirable to solve the current problems existing in traditional gluconic acid preparation processes. In this study, a robust and easy-to-use multienzymatic cascade reaction system of coimmobilized GA@GOx hybrid nanoflowers with a specific spatial distribution of enzymes by compartmentalization was constructed and applied to catalyze starch to gluconic acid in one pot. In the preparation processes, the glucose oxidase (GOx) hybrid nanoflowers were first synthesized via a self-assembly mechanism, and then, glucoamylase (GA) was adsorbed on the surface of GOx hybrid nanoflowers through the interaction of Cu2+ and amino acids of GA. The optimum preparation conditions and reaction parameters of the GA@GOx hybrid nanoflowers had been investigated. In addition, the morphology, composition, and crystallization of the GA@GOx hybrid nanoflowers had been fully studied. Based on the lower Km, the GA@GOx hybrid nanoflowers with compartmentalization had a better effect of the substrate channeling on the catalytic efficiency. The final results indicated that the overall enzyme activity of the GA@GOx hybrid nanoflowers increased by 1.5 times, and the conversion efficiency was 92.12% within 80 min significantly superior to the free multienzyme system, which showed the outstanding conversion of starch into gluconic acid in one pot.

BACKGROUND: Previous gene-environment studies on depression have examined the interaction between FKBP5 gene and childhood trauma, but the results are inconsistent and few studies have focused on Asian adolescents. Psychological resilience may explain for the inconsistency. We examined the interaction between FKBP5 gene and childhood trauma on depressive symptoms in Chinese adolescents, and firstly explored the moderating role of resilience in the relationship. METHODS: This study comprised 942 participants (448 males, 47.6%) randomly recruited from four senior schools in Wuhan, Hubei of China. Depressive symptoms, childhood trauma, and resilience were respectively evaluated by the Center for Epidemiological Studies Depression Scale (CES-D), the Childhood Trauma Questionnaire (CTQ) and the Connor-Davidson Resilience Scale (CD-RISC). Three potentially functional FKBP5 polymorphisms were selected for genotyping. RESULTS: Participants carrying minor alleles of FKBP5 polymorphisms (rs3800373, rs1360780, and rs4713916) and a haplotype derived from these variants displayed higher CES-D scores when exposed to childhood physical abuse after adjusting for demographic characteristics and resilience (all P < 0.01). The three-way interactions of FKBP5 SNPs, physical abuse, and resilience on depressive symptoms all yielded statistical significance after adjusting for demographic characteristics (ß = -0.282 to -0.236; all P < 0.001). LIMITATIONS: Cross-sectional design, self- reported measurements and limited genotyped FKBP5 polymorphisms. CONCLUSION: FKBP5 variants in combination with childhood physical abuse may increase more pronounced depressive symptoms among Chinese adolescents, while resilience plays a moderating role in the associations. Future research to examine the exact mechanism of resilience in these associations is needed.

In this study, a new fluorescence probe HMAQ based on quinazoline and diaminomaleonitrile was constructed for sensing ClO- and Al3+. A fluorescence blue-shift with 102 nm together with a color change from golden-yellow to colorless was found by hypochlorite-induced hydrolysis of -CH=N- group to release the initial fluorophore. Besides, Al3+ could cause a 72-nm blue-shifted emission spectra and a color change from golden-yellow to brown. As expected, HMAQ exhibited a satisfactory selectivity and sensitivity to ClO-/Al3+ with a quick response. Most notably, the reversibility of the [HMAQ+Al3+] complex could be used to detect ClO- and Al3+ simultaneously without mutual interferences. The detection limits of HMAQ for ClO- and Al3+ were turned out to be 10.2 nM and 1.56 nM, respectively. The high-performance results of real-time detections demonstrated the enormous potential of HMAQ in real-water samples and living cells.

BACKGROUND: Various studies showed that personality traits and resilience might have impacts on depressive symptoms, separately. However, the relationships among personality traits, resilience and depressive symptoms are still undefined. Thus, this study tried to explore the potential effect of resilience on the associations between personality traits and depressive symptoms in Chinese adolescents. METHODS: Adolescents (n = 6019) aged 10-17 years were recruited from nine schools in Wuhan, China. Depressive symptoms, personality traits, and resilience were evaluated by the Center for Epidemiologic Studies Depression scale (CES-D), the NEO-Five Factor Inventory (NEO-FFI), and the Connor-Davidson Resilience Scale (CD-RISC), respectively. RESULTS: Neuroticism was positively associated with depressive symptoms, whereas extraversion, openness, agreeableness, and conscientiousness were negatively associated with depressive symptoms. Resilience separately moderated the associations of neuroticism, extraversion, agreeableness, and conscientiousness with depressive symptoms, and partly mediated the associations of all five personality traits with depressive symptoms. LIMITATIONS: This study is a cross sectional study and cannot ascertain the causal relationships between the variables. Also self-reported questionnaire instruments were used in the data collection. CONCLUSIONS: These findings suggested that resilience might play moderating and mediating roles in the associations of personality traits with depressive symptoms, and prompted that it was critical to improve resilience and develop adaptive personality traits in the prevention and intervention of depression in adolescents.

BACKGROUND & AIMS: Oxidative stress has been implicated in the pathogenesis of gestational diabetes mellitus (GDM). Vitamin C as natural antioxidant may help to increase the body's antioxidant capacity. The study is aimed to determine whether vitamin C intake during pregnancy is associated with lower risk of GDM. METHODS: Women with singleton pregnancy and without any history of diabetes were drawn from the ongoing Tongji Maternal and Child Health Cohort (TMCHC). Oral glucose tolerance tests (OGTT) were conducted during weeks 24-28 of gestation to screen for GDM. A validated food frequency questionnaire (FFQ) was used to assess dietary intake during mid pregnancy. Use of multivitamins and specific supplements of vitamin C was assessed by questionnaires. Odds ratios (ORs) of GDM risk were calculated by logistic regression models, adjusted for potential confounders. RESULTS: 344 (11.4%) of the 3009 women were diagnosed with GDM. Dietary vitamin C intake was inversely associated with the risk of GDM. Women with above adequate dietary vitamin C intake (more than 200 mg/day) experienced lower odds of GDM (OR 0.68, 95% CI: 0.49-0.95) than those with adequate intake (115-200 mg/day). There was no association between the total consumption of vitamin C and the risk of GDM (OR 1.04, 95% CI: 0.71-1.53). CONCLUSION: This data suggests that higher dietary consumption of vitamin C during pregnancy is independently associated with lower odds of GDM. Above 200 mg/day of dietary vitamin C intake may help reduce the odds of GDM. However, no such association between total vitamin C intake and the risk of GDM was found. Hence, sufficient vegetables and fruits rich in vitamin C should be recommended to protect pregnant women from developing gestational diabetes.

An aqueous two-phase system was used in conjunction with ultrasonic cell disruption to extract and separate solanine (mainly solasonine and solamargine) and Solanum nigrum polysaccharide from Solanum nigrum unripe fruit. The optimized conditions of the present study were determined by a single-factor experiment and a multifactor experiment. The concentration of ethanol was set at 60% and the duration of the ultrasonic cell disruption extraction was 50 min. In the ethanol-K2CO3 aqueous two-phase separation system, the concentration of ethanol was 36%, the concentration of K2CO3 was 0.21 mg·mL-1, and the temperature was 15 °C. The solasonine and solamargine were determined by high-performance liquid chromatography, and the Solanum nigrum polysaccharide was determined by an ultraviolet-visible spectrophotometer in accordance with the phenol-sulfuric acid method. xUnder optimized conditions, the average extraction efficiencies of solasonine, solamargine and Solanum nigrum polysaccharide were 95.86%, 95.95% and 96.95%, respectively, and the average separation efficiencies of solasonine, solamargine and Solanum nigrum polysaccharide were 2.07 mg·g-1, 2.05 mg·g-1 and 8.15 mg·g-1, respectively.

This work was to develop a cost-effective and sustainable method which included metal chelate ionic liquid-based aqueous two-phase flotation (IL-based ATPF) and a two-step precipitation process for purifying bromelain from pineapple. Firstly, the metal chelate IL-based ATPF with a copper chelate-functionalized thermosensitive block copolymer (L64-IDA-Cu(II)) as trapping agent was used as the primary purification to obtain the L64-IDA-Cu(II)-bromelain complex. Secondly, the two-step precipitation process based on the thermosensitive properties of the L64-IDA-Cu(II) was mainly carried out to achieve the further purification of bromelain. According to a series of optimal experiments, the enzymatic activity recovery of final bromelain was 95.22 ±â€¯0.04%, and the purification factor reached 6.56 ±â€¯0.03. The results of recycling of ILs and the trapping agent were satisfactory. Furthermore, the conclusions of comparison with other methods proved the superiority of this method. This novel recycling purification method has a goodindustrial prospect in future.

Enhancing the catalytic activity and stability of enzymes is of great importance in the development of green chemical and cost-effective application, with removal of bisphenol A (BPA) as a prominent example. Engineering immobilization carriers and immobilization methods of enzymes endows great potential to achieve above goal. Until now, these reports have focused on employing the metal-organic frameworks (MOFs) to increase the stability and reusability of enzymes, an enhancement in its catalytic activity has yet to be addressed. This work introduced a biomimetic mineralization process for facile synthesis of laccase@HKUST-1 biocomposite under mild condition. By exploiting the activity of laccase@HKUST-1, we demonstrated, for the first time, that the integration of laccase and HKUST-1 containing cofactor Cu2+ ions leaded to 1.5-fold enhancement in the catalytic activity compared with free laccase, which was due to the synergistic enhancement of substrate oxidation. Indeed, the laccase@HKUST-1 biocomposite could function as active biocatalysts under biologically challenging conditions, such as acidic condition, high temperature, organic solvent, and continuous operation. The oxidation of phenols, such as BPA, with laccase@HKUST-1 reached higher catalytic performance than free laccase, and gave 100% degradation efficiency within 4 h. This study provides a feasible method to improve the activity and stability of laccase, which enable completely remove of BPA from the environment.

Class I TGA transcription factors (TFs) are known to participate in plant resistance responses, however, their regulatory functions in the biosynthesis of secondary metabolites were rarely revealed. In this study, a class I TGA TF, TwTGA1, from Tripterygium wilfordii Hook.f. was cloned and characterized. Overexpression of TwTGA1 in T. wilfordii Hook.f. cells increased the production of triptolide and two sesquiterpene pyridine alkaloids, which was further enhanced by methyl jasmonate (MeJA) treatment. RNA interference of TwTGA1 showed no significant effects on the production of these metabolites, indicating the existence of other TGA partner(s) with overlapping functions. Heterologous expression of TwTGA1 in tobacco By-2 cells promoted the biosynthesis of pyridine alkaloids. Under the elicitation of MeJA, the contents of nonpyrrolidine alkaloids further increased but not for nicotine. TwTGA1 could induce the expression of Putrescine N-methyltransferase (PMT) and N-methylputrescine oxidase 1 (MPO1) through binding to their promoters. Finally, transient expression of TwTGA1 in leaves of Catharanthus roseus changed both the profiles of vinca alkaloids (increased contents of serpentine and catharanthine, but decreased that of vinblastine) and the expressions of biosynthesis-related genes. The metabolic and transcriptional data indicated a relationship between jasmonic acid signaling pathway and the functions of TwTGA1.

A 10-week feeding trial was conducted to investigate the effects of dietary bile acids (BA) on growth, glucose and lipid metabolism, liver histopathology, and the underlying regulation mechanism on AKT/FOXO1 (forkhead box O1) and cAMP/AMPK/SREBP1 (sterol regulatory element-binding protein 1) pathway in largemouth bass (Micropterus salmoides) fed with a high starch diet. Six experimental diets were prepared with BA levels at 0 (B0), 80 (B80), 160 (B160), 240 (B240), 300 (B300), and 600 (B600) mg/kg in a basal diet with 18.7% starch. Each diet was fed to six replicates with 30 fish (6.17 ± 0.03 g) in each tank. The highest weight gain rate (WGR) was observed in B300 group and the optimal level of BA was estimated at 475 mg/kg by a monistic cubic equation regression analysis. Dietary BA inclusion decreased hepatosomatic index (HSI) and hepatic lipid content significantly. The fish in B300 group clearly showed alleviated hepatic fibrosis, but more steatohepatitis symptoms diagnosed with various histopathological and immunofluorescence analysis. 10 out of 12 samples were observed hepatic fibrosis in B0 group while only two fibrosis samples in B300 group. The promoted liver histopathology by dietary BA was related to improved glucose and lipid metabolism. Dietary BA inhibited the expression of G6Pase by activating AKT and reducing FOXO1 transcription, which improved the regulation ability of gluconeogenesis, activated cAMP/AMPK and repressed SREBP1 transcription to inhibit hepatic lipogenesis, which prevented hepatic lipid accumulation. In conclusion, dietary BA enhanced the growth and alleviated liver fibrosis induced by a high starch diet to steatohepatitis/recovery symptom via improving glucose and lipid metabolism, which regulated by AKT/FOXO1 and cAMP/AMPK/SREBP1 pathway in largemouth bass.

The improvement of grain filling is the key issue for promoting wheat thousand grain weight and grain yield. The levels of polyamines (PAs) significantly affect grain filling in cereals, but the mechanism by which PAs affect grain filling in wheat is unclear. In the present study, six wheat cultivars whose grain filling differed were used, and their grain-filling characteristics and endogenous PA contents were measured. In addition, exogenous PAs were supplied during the wheat grain-filling period. The grain-filling characteristics, hormone levels, starch contents, and gene expression [based on RNA sequencing (RNA-seq)] in the grain were analyzed. The objective of the present study was to investigate the effects of PAs on grain filling in wheat. The results suggested that the direct synthetic pathway from putrescine (Put) to spermidine (Spd) in the grain was a key factor in promoting grain filling and thousand grain weight in wheat. Spd through regulates the grain-filling rate of inferior grain during the early grain-filling period to affecting the grain filling and thousand grain weight of wheat. The promotive effect of Spd on the grain filling of inferior wheat grain was notably related to carbohydrate metabolism in that grain. Spd significantly increased the zeatin (Z) + zeatin riboside (ZR) contents but reduced the ethylene (ETH) evolution rate in the inferior grain. In addition, Spd significantly increased the sucrose synthase (SS) and acid invertase (AI) activities in the inferior grain. These effects of Spd led to increased sucrose content in the inferior grain. These reasons might explain why Spd significantly promoted the filling and weight of inferior wheat grain.

The current usage and the existing problems in the implementability of clinical practice guidelines for acupuncture-moxibustion were investigated by questionnaire survey, aiming to provide reference for the development or update of clinical practice guidelines for acupuncture-moxibustion in the future. The results showed most of the acupuncture-moxibustion clinicians did not have a deep understanding of the guidelines, but they had a strong will of uniform standards and related guidelines. Although the published clinical practice guidelines for acupuncture-moxibustion achieved some success, they still had not got rid of the shackles of the previous textbook. The main existing problems in the guidelines included insufficient promotion, poor credibility, no evaluation criteria for curative effect, and lack of consideration for patients' will, etc. As the guidelines for acupuncture-moxibustion were based on the latest evidence of current clinical research, it reflected the low quality of current clinical research on acupuncture-moxibustion and lacking of evidence-based concept among acupuncture-moxibustion clinicians. The implementability of clinical practice guidelines is a key step in evidence-based translational medicine, while the research on the implementability of acupuncture-moxibustion guidelines is still blank. More attention should be paid to this field in the future.

RESUMO

To study the effects of combination of Aconiti Lateralis Radix Praeparata( Fuzi) with Trichosanthis Fructus( Gualou) on cardiac function,electrocardiogram,inflammatory response and myocardial fibrosis in pressure overload( PO) rats,and further explore the mechanism based on ß2-AR/PKA signaling. PO rat model was established by constricting the abdominal aorta. Twelve weeks after the operation,these rats were randomly divided into model goup( PO),low dose Fuzi group( FL,5. 4 g·kg-1·d-1),Gualou group( GL,5. 4 g·kg-1·d-1),Fuzi and Gualou combination group( FG,5. 4 g·kg-1·d-1+5. 4 g·kg-1·d-1) and high dose Fuzi group( FH,10. 8 g·kg-1·d-1). At the same time,sham operation group was set. After intervention for 6 weeks,carotid blood pressure,cardiac function,electrocardiogram and heart mass index were measured. HE staining was used to observe the inflammatory response in the rat heart and kidney. Masson staining was used to determine the myocardial fibrosis. Western blot was used to detect the protein expression of ß2-AR and PKA. As compared with sham operation group,the blood pressure and heart mass index were obviously increased in PO model group,but there was no significant difference in various treatment groups in the above indexes. As compared with PO model group,FH treatment significantly increased the ejection fraction( EF) and GL treatment effectively enhanced the cardiac output( CO),but other treatment groups had no significant effect on these parameters. Moreover,FG treatment can synergistically attenuate QT and QTc internal prolongation,but it also aggravated inflammatory response in the heart and kidney tissues and promoted myocardial fibrosis as compared to FZ or GL alone treatment,with toxic effects equivalent to FH treatment group. Following FG and FH treatment,simultaneously,ß2-AR and PKA protein levels were significantly elevated,indicating that the increasing toxicity of FG could be associated with activation of ß2-AR/PKA signaling. These results suggested that combination of FZ and GL could synergistically enhance toxicity of FZ in special pathological states such as pressure overload,and caution should be taken in clinical application.

An ultrasensitive assay is described for the detection and determination of Hg2+(aq) in water samples based on single-particle inductively-coupled plasma/mass spectrometry (spICP-MS). In the presence of Hg2+(aq), AuNPs modified with a segment of single-stranded DNA aggregate due to the formation of the well-known thymine (T)-Hg2+-T complex. Single particle (sp) ICP-MS is used quantify the degree of aggregation by the overall decrease in number of detected AuNPs or NP aggregates. Compared with most other Hg2+ assays that use the same principle of aggregation-dispersion with DNA modified AuNPs, this method has a much lower detection limit of (0.031 ng L-1, 155 fM) and a wider (10,000-fold) linear range (up to 1 µg L-1). The method also showed good practical potential because of its minimal interference from the water sample matrix. INTRODUCTION: The effect of N-acetylcysteine (NAC) treatment for patients with chronic bronchitis (CB) is controversial. To better understand the role of NAC in CB treatment, we performed a meta-analysis to provide a more accurate estimation of the importance of NAC treatment. METHODS: PubMed, Embase, and CNKI were systematically searched. The pooled relative risk (RR) and 95% confidence intervals (CI) were calculated using either fixed-effect model or random-effect model based on heterogeneity examination. Statistical analyses were performed using the STATA 12.0 and RevMan 5.2. RESULTS: A total of 11 publications with 775 patients who were taking NAC and 789 controls who were taking placebo were judged eligible regarding inclusion criteria. The pooled analysis demonstrated significant evidence that NAC reduced the frequency of CB exacerbations (RR = 0.81, 95% CI 0.69-0.93, P = 0.004). Patients treated with NAC had significant symptom improvement compared with controls (RR = 1.68, 95% CI 1.13-2.52, P = 0.01). NAC did not significantly increase the risk of adverse effects compared with placebo (RR 0.86, 95% CI 0.67-1.09, P = 0.22). Subgroup analysis was carried out to assess the stability of results. No publication bias was detected during analyses. CONCLUSION: There is a role for NAC treatment in the management of CB by reducing symptoms and exacerbations compared with placebo, without increasing the risk of adverse effects. A regular treatment of low dosage (< 1200 mg per day) and a duration of at least 3 months seems to be effective.

OBJECTIVE: To investigate the prevalence of early trauma and resilience among adolescents in Wuhan, and explore the relationship between early trauma and resilience. METHODS: Totally 4871 students aged 10-16 years were chosen by cluster sampling in Wuhan city from September to October 2015. All subjects completed self-report questionnaires, including general information, the Childhood Trauma Questionnaire( CTQ), and the Connor Davidson Resilience Scale( CD-RISC). RESULTS: The mean score of CD-RISC of the total sample was( 64. 70 ± 18. 34). Statistical significance in different gender( t = 5. 373, P<0. 001), age( F = 49. 401, P<0. 001), single child( t = 3. 529, P<0. 001), levels of mother's education( F = 36. 129, P< 0. 001), relationship between parents( F = 89. 831, P < 0. 001), family economic status( F = 36. 547, P<0. 001). The rate of early trauma was 30. 1%. Male( χ~2= 42. 272, P < 0. 001), lower levels of mother 's education( χ~2= 44. 345, P < 0. 001), poorer relationship between parents( χ~2= 133. 045, P < 0. 001), and worse family economic status( χ~2= 31. 231, P<0. 001) were associated with increased risk of early trauma. The scores of emotional abuse, physical abuse, sexual abuse, emotional neglect and physical neglect were negatively correlated with the scores of CD-RISC( r followed by-0. 256, -0. 107, -0. 053, -0. 355 and-0. 308, P<0. 01). Regression analysis implied female, older age, emotional abuse, emotional neglect, and physical neglect( B followed by-0. 156, -0. 117, -0. 109, -0. 214 and-0. 149, P < 0. 01) of primary and middle school students assumed predictive resilience. CONCLUSION: Emotional abuse, emotional neglect, and physical neglect are negatively associated with resilience among children and adolescent. The result suggest that reducing emotional abuse, emotional neglect and physical neglect experience may contribute to child resilience.

In this study, a novel type of multi-armed polymer (poyltehylene glycol, PEG) magnetic graphene oxide (GO) composite (GO@Fe3O4@6arm-PEG-NH2) has been synthesized as a support for immobilization of horseradish peroxidase (HRP) for the first time. The loading amount of HRP was relatively high (186.34 mg/g) due to the surface of carrier material containing a large amount of amino groups from 6arm-PEG-NH2, but degradation rate of phenols was also much higher (95.4%), which is attributed to the synergistic effect between the free HRP (45.4%) and the support material of GO@Fe3O4@6arm-PEG-NH2 (13.6%). Compared with the free enzyme, thermal, storage and operational stability of the immobilized HRP improved. The immobilized HRP still retained over 68.1% activity after being reused 8 times. These results suggest that the multi-armed magnetic composite has good application prospect for enzyme immobilization.
