Microbiologically influenced corrosion (MIC) of copper by Desulfovibrio vulgaris, a sulfate reducing bacterium (SRB), was investigated in anaerobic vials with a fixed broth volume of 40 mL but varied headspace volumes (10 mL, 85 mL 160 mL). It was found that the headspace volume variation had a very large effect on the dissolved [H2S] in the broth and the cell counts of planktonic and sessile cells, as well as Cu corrosion severity. A 16× smaller headspace led to a 1.6-fold increase in the dissolved [H2S], a 13-fold decrease in sessile SRB cell count, a 32-fold decrease in planktonic cell count and a 3.7-fold increase of Cu weight loss. SEM images revealed that different headspace volumes caused different corrosion patterns on the immersed coupons. With a lower headspace volume, pitting corrosion was observed, while with a higher headspace volume, intergranular corrosion was seen. The results confirmed that SRB MIC of Cu belongs to metabolite-MIC (M-MIC) by H2S, unlike SRB MIC of carbon steel that belongs to extracellular electron transfer-MIC (EET-MIC) that is directly correlated with sessile cell counts rather than dissolved [H2S]. .

RESUMO

This white paper presents principles for validating proarrhythmia risk prediction models for regulatory use as discussed at the In Silico Breakout Session of a Cardiac Safety Research Consortium/Health and Environmental Sciences Institute/US Food and Drug Administration-sponsored Think Tank Meeting on May 22, 2018. The meeting was convened to evaluate the progress in the development of a new cardiac safety paradigm, the Comprehensive in Vitro Proarrhythmia Assay (CiPA). The opinions regarding these principles reflect the collective views of those who participated in the discussion of this topic both at and after the breakout session. Although primarily discussed in the context of in silico models, these principles describe the interface between experimental input and model-based interpretation and are intended to be general enough to be applied to other types of nonclinical models for proarrhythmia assessment. This document was developed with the intention of providing a foundation for more consistency and harmonization in developing and validating different models for proarrhythmia risk prediction using the example of the CiPA paradigm.

BACKGROUND: Early diagnosis decreases the mortality of hepatocellular carcinoma (HCC). We aimed to investigate the usefulness of PIVKA-II, AFP, AFP-L3, CEA, and their combinations in the diagnosis of primary and metastatic HCC. METHODS: One hundred and twenty patients with primary HCC (PHC), 115 with metastatic HCC (MHC), 89 with chronic liver disease (CLD), and 116 healthy volunteers were included. The diagnostic values of each marker and their combinations for HCC diagnosis were represented by ROC curve analyses. RESULTS: PIVKA-II, AFP, and AFP-L3 levels in PHC group were higher than that in normal control, CLD, and MHC groups. CEA levels in MHC group were higher than that in the other three groups. When the four markers were analyzed individually, PIVKA-II showed the highest positive rate in PHC group (76.7%) and CEA showed the highest positive rate in MHC group (69.6%). PIVKA- II showed the largest area under ROC curve (AUC = 0.835) to discriminate PHC group from CLD group. Combined PIVKA-II with AFP-L3 increased the AUC to 0.910. CEA showed the highest AUC (0.849) to discriminate MHC group from CLD group. Combined CEA with PIVKA-II increased the AUC to 0.866. AFP-L3 alone showed the highest AUC (0.890) to discriminate MHC group from PHC group. Combined PIVKA-II with AFP-L3, and CEA increased the AUC to 0.957. CONCLUSION: PIVKA-II, AFP-L3, AFP, and CEA are effective biomarkers for the diagnosis of PHC and MHC. Their combinations could improve the diagnostic performance compared with each marker used alone in detecting PHC and MHC.

The International Council on Harmonization (ICH) S7B and E14 regulatory guidelines are sensitive but not specific for predicting which drugs are pro-arrhythmic. In response, the Comprehensive In Vitro Proarrhythmia Assay (CiPA) was proposed that integrates multi-ion channel pharmacology data in vitro into a human cardiomyocyte model in silico for proarrhythmia risk assessment. Previously, we reported the model optimization and proarrhythmia metric selection based on CiPA training drugs. In this study, we report the application of the prespecified model and metric to independent CiPA validation drugs. Over two validation datasets, the CiPA model performance meets all pre-specified measures for ranking and classifying validation drugs, and outperforms alternatives, despite some in vitro data differences between the two datasets due to different experimental conditions and quality control procedures. This suggests that the current CiPA model/metric may be fit for regulatory use, and standardization of experimental protocols and quality control criteria could increase the model prediction accuracy even further.

BACKGROUND: Scaffold proteins play a critical role in an increasing number of biological signaling processes, including simple tethering mechanism, regulating selectivity in pathways, shaping cellular behaviors. While many databases document the signaling pathways, few databases are devoted to the scaffold proteins that medicate signal transduction. RESULTS: Here, we have developed a user-friendly database, ScaPD, to describe computationally predicted, experimentally validated scaffold proteins and associated signaling pathways. It currently contains 273 scaffold proteins and 1118 associated signaling pathways. The database allows users to search, navigate and download the scaffold protein-mediated signaling networks. CONCLUSIONS: Manually curated and predicted scaffold protein data will be a foundation for further investigation of the scaffold protein in the signal transduction. With maintained up-to-date data, ScaPD ( http://bioinfo.wilmer.jhu.edu/ScaPD ) will be a valuable resource for understanding how individual signaling pathways are regulated.

BACKGROUND AND AIM: Examination of top-cited articles is a tool that can help to identify and monitor outstanding scientific researches and landmark papers. We aimed to identify the 100 most cited published papers in peer-reviewed biomedical journals in the field of digestive diseases and to examine their characteristics. METHODS: The Web of Science (including Science Citation Index) was searched for the most cited papers related to digestive diseases, published from 1955 to the present. The top 100 most cited articles were identified. The number of citations, countries, and institutions of origin, year of publication, study design, topic, and levels of evidence of the articles were noted and analyzed. RESULTS: The most top-cited articles had a mean of 1375 citations. These articles were published between 1978 and 2009 in 29 high-impact journals, with the New England Journal of Medicine (n = 22) topping the list. Of the 100 articles, 34 were clinical studies, 15 were review articles, and 34 were concerned basic science. These articles came from 18 countries, with the USA contributing most of the top-cited articles (n = 53). Eighty-seven institutions produced these 100 top-cited articles, led by the University of Barcelona (n = 4). Seven persons authored two or more of these top-cited articles. The mostly represented specialty was gastrointestinal oncology (n = 49). CONCLUSIONS: Our study can give a historical perspective on the scientific progress of digestive diseases, as well as allow for recognition of most important advances in this area and provide useful information to guide future researches.

RATIONALE: Synthetic and natural coumarin derivatives possess a wide range of biological activities. Fragmentation pathway studies are important in identifying both naturally occurring coumarins and synthetic coumarins with novel structures and properties. METHODS: The fragmentation pathways of eleven coumarin derivatives are investigated by electrospray ionization (ESI) ion-trap mass spectrometry (ESI-ITMSn ) and ESI quadrupole time-of-flight mass spectrometry (QTOFMS) in positive mode. Compounds 1-9 in this study were newly synthesized in our laboratory. Compounds 10 and 11 were isolated from the root of Zanthoxylum armatum. RESULTS: The major fragmentation pathways for 11 coumarin derivatives are greatly affected by the heterocyclic ring structures and the side-chain substituents. Typical losses of small neutral molecules, such as CH3 CH2 OH, CH2 =CH2 , CO, and H2 O, are observed for compounds 1-5. Compounds 6-9 share similar fragmentation pathways through losses of CO, aromatic rings, and the coumarin skeleton. The main product ions at m/z 205, 219, and 220 observed for compounds 10 and 11 are produced by the loss of C5 H12 O2 , C4 H10 O2 , and the C4 H9 O2 radical, respectively. CONCLUSIONS: The fragmentation pathways of 11 coumarin derivatives are elucidated based on ITMSn and QTOFMS spectral data. Differences in the structures of the heterocyclic rings and side-chain substituents strongly affect the fragmentation pathways of the coumarins. The present results will facilitate further research into the fragmentation pathways and structural characterization of these classes of compounds with diverse structures. Tumor-associated antigens (TAAs) trigger a TAA-specific immune response, thus they are the crux of antitumor immunosurveillance. A major advance in tumor immunology in the last 20 years was marked by the verification that CTL or B-cell epitopes rather than integral TAAs induce immunoreactivity. Previous studies on the correlation between heparanase (Hpa) expression and clinical or pathological features have generally used commercial antibodies against full-length Hpa protein rather than the functional epitopes, and the antigen determinants of such antibodies have not yet been defined. In our investigation of Hpa peptide expression in gastric cancer tissues and its association with tumor invasion, metastasis and prognosis, we analyzed Hpa expression in the tissues of 132 patients with gastric cancer using tissue microarray (TMA) technology and immunohistochemical staining. Three self-developed rabbit polyclonal antibodies against Hpa multiple antigenic peptides (MAP) and one commercial polyclonal rabbit antibody against the 50-8 kDa Hpa heterodimer were used. Clinical and pathological significance was evaluated using the Chi-square test and Kaplan-Meier survival curve analysis. The results demonstrated that the positivity rates using the antibody against MAP2 and the commercial antibody were 60.6% (80/132) and 65.2% (86/132), respectively. No expression of either MAP1 or MAP3 was noted in the cancer tissues of the 132 cases. MAP2 behaved in a similar manner to the commercial antibody in that a higher Hpa expression was observed in the cancer tissues with vessel invasion, serosal involvement, distant metastasis, poor differentiation and TNM stages III and IV. Moreover, the patients with a positive Hpa expression had a far poorer prognosis, with lower one-year and five-year survival rates. Our results demonstrate that in a similar manner to full-length Hpa proteins, MAP2 expression is closely associated with the invasion, metastasis and prognosis of gastric cancer. This finding may be of potential use in clinical therapy and in estimating the prognosis of a tumor.

OBJECTIVE: To establish the basis for Chinese dietary exposure assessment database by classifying and coding the data from the national dietary survey and pollutant surveillance. METHODS: The method, which combined CODEX food classifying and coding of Codex Alimentarius Commission (CAC) with Chinese food classification of food composition table, was applied to classify and code the data of 1 810 703 Chinese dietary consumption and 487 819 pollutant surveillance. The coding system was according to the first two letters of the respective food group that represent the type or source of foods, the last four digits represent the serial number of the food in the CAC food classification. If the foods can be found in CAC food code system, its original food code is used. The new codes corresponding with the foods which are not exist in CAC food code system, is added according to CAC coding methods. RESULTS: Dietary consumption data were divided into 6 major categories, 19 types, 75 groups, the agricultural products of pollutant surveillance corresponding to 499 codes. Comparing with CAC food coding system, Chinese dietary consumption data have added F (candy snacks) and G (beverages) 2 major categories, 4 types, 33 groups, 302 new codes. The additional groups most were the processing food groups with Chinese characteristics, such as canned, beverages, candy, meat products. CONCLUSION: The foundation of data communication to dietary exposure assessment has been established, and the connection of Chinese food classifying and coding with CAC data have been achieved.

OBJECTIVE: To investigate the effects of the self-developed anti-heparanase polypeptide antibodies on growth and invasion of human hepatocellular carcinoma HCCLM6 cells. METHODS: Using MTT, flow cytometry, plate clone formation, transwell invasion and heparan degrading enzyme assay, the growth and invasion changes of human hepatocellular carcinoma HCCLM6 cells by co-culture with each of three self-developed rabbit anti-heparanase polyclonal antibodies were detected. RESULTS: Compared with normal rabbit IgG, in the presence of each anti-heparanase polypeptide antibody, the growth, cell cycle and clone formation remained unchanged, and under the P1 or P2 anti-heparanase polypeptide antibody (with final concentration 100 microg/ml), the cell invasiveness was inhibited by 52.5% and 36.6%, respectively, and the heparanase activity was inhibited by 42.9% and 39.1%, respectively. CONCLUSION: The P1 and P2 anti-heparanase polypeptide antibodies can effectively inhibit the invasion ability and heparanase activity of liver cancer HCCLM6 cells. However, All the three antibodies have no effects on its growth, cell cycle and clone formation.

BACKGROUND: The aim of this study was to screen and identify novel B cell epitopes within the human heparanase protein and to investigate the impact of self-developed anti-heparanase polypeptide antibodies on growth and invasion of HCCLM6 human hepatocellular carcinoma cells in vitro. METHODS: The flexible regions of secondary structure and the B cell epitopes of the human heparanase amino acid sequence were predicted by DNAStar and Bcepred software.The multiple antigenic peptides (MAP) of the epitopes were synthesized in eight-branched form. Rabbits were immunized with the eight-branched MAPs mixed with the universal T-helper epitope human IL-1beta peptide (VQGEESNDK, amino acid 163-171). The immunogenicity of the synthesized peptides was evaluated by ELISA, western blot and immunohistochemistry. The impact of the self-developed rabbit anti-heparanase polyclonal antibodies on growth and invasion ability of HCCMLM6 cells were analyzed in a cell culture model. The cells were first treated with one of the three antibodies, respectively, and then measured by using MTT, flow cytometry, plate clone formation, invasion assay and heparan sulfate degrading enzyme assay. RESULTS: The three amino acid sequences 1-15 (MAP1), 279-293 (MAP2), and 175-189 (MAP3) in the large subunit of the human heparanase protein were predicted as its most potential epitopes. ELISA, western blot and immunohistochemistry analysis showed that all three MAPs were capable to induce high titer of serum antibodies. Antibodies induced by MAP1 and MAP2 were high specific. Furthermore, anti-MAP2 antibodies showed the strongest avidity towards liver cancer tissues. Under the treatment with the three anti-heparanase antibodies, respectively, the growth, cell cycle and clone formation of the cells remained unchanged when compared with a treatment with normal rabbit IgG. However, an inhibition of cell invasiveness and heparanase activity could be detected under the treatment with anti-MAP1- or anti-MAP2-antibody (with a terminal concentration of 100 mug/ml). The cell invasiveness was decreased by 54 and 38%, respectively, the heparanase activity by 43 and 39%, respectively. CONCLUSION: The multiple antigenic peptides MAP1 (AC 1-15) and MAP2 (AC 279-293) may be the dominant B cell epitopes in the human heparanase protein. The induced polypeptide antibodies can effectively inhibit the heparanase activity of HCCLM6 liver cancer cells and therefore influence their invasion ability, which provides a theoretic basis for the development of anti-heparanase antibodies and their clinical use as vaccine.

OBJECTIVE: To explore the construction of bridge table of Chinese dietary exposure estimation model. METHODS: Bridge table can be structured by SAS procedure based on the matching relationship of foodcode in the structural of dietary survey data and the CAC (Codex Alimentarius Commission) code in the structure of pollutant monitoring data. RESULTS: According to the relationship of foodcode and CAC code, bridge table comprising the information of processing methods, processing factor and variability factor could be established. CONCLUSION: Actually, bridge table links the primary information which connect the dietary survey data with pollutant monitoring data. It could be an important foundation of constructing dietary exposure estimation model.

OBJECTIVE: To study the morphology differences of ocular rectus muscles pulley between preschool children and adults. METHODS: It was an experimental research. Ten fresh cadaver orbits (five children and five adults) were fixed in situ and serially sectioned at 4 microm thickness. Every section was stained with Masson's trichrome and picrosirius red methods. After being stained, extraocular muscle pulley in typical sections was examined through gross anatomy and light microscopy. Two-factor analysis of variance for quantitative data was used in statistical treatment. RESULTS: Near the equator of the globe, the children and adults' extraocular muscle pulleys were all at the points where the rectus penetrate posterior Tenon's fascia, encircling the rectus orbit side by fiber ring structure. There were pulley bands among rectus pulleys. The rectus pulley length and the origin distance from the corneoscleral limbus of the children were significantly less than those from the adult group (P < 0.05). The main composition of the children rectus pulley was similar to that of the adults, but the smooth muscle and collagen type III fibers contents of the medial rectus pulley in children were significantly greater than those in the adults group (P < 0.05). Collagen type I fibers in the children were significantly less than that of the adults group (P < 0.05). CONCLUSION: There are morphological differences in rectus pulley between preschool children and adults, which provides an anatomic basis for the better understanding of the pathogenesis of strabismus.

OBJECTIVE: To establish 3-D visible human model of the rectus EOM Pulley. METHODS: The pictures of Masson stained adjacent coronal sections of a whole orbit were imaged by digital camera, which were entered into computer continuously after removing spottiness through PhotoShop 7.0 software. 3D-Doctor software was used to register and split the image. Finally three-dimensional computerized reconstructions of the rectus EOM pulley and eye ball were achieved. At the same time pulley system structure was analyzed using the 3D-Doctor software. RESULTS: Three-dimensional model of the rectus EOM pulley displayed directly the three-dimensional relationship of pulley and the eye ball, extraocular muscles and orbit. This three-dimensional model could be rotated randomly, providing convenience for multi-angle observation. The demonstration of the three-dimensional model selectively provides a approach to get rid of disturbance and lock the aim for observation. The length of pulley was about 4.6 to 5.8 mm. The origin of the four rectus pulleys was 13.8 to 18.0 mm from the corneoscleral limbus. Media rectus pulley was thickest among four rectus pulley. The ratio between media rectus pulley thickness and corresponding media rectus thickness could reach 1.11. CONCLUSIONS: Three-dimensional model of the rectus EOM pulley provided direct picture for clinic doctors to understand the structure of pulley, which also provided three-dimensional anatomic basis for iconography research of the rectus EOM pulley. The data of pulley could be helpful to establish Chinese rectus EOM pulleys data bank.

The objective in this paper was to determine the amount of metal elements in the Plant Bupleurum scorzoneri folium Willd. by flame atomic absorption spectrophotometry (FAAS). FAAS method was established for the determination of Ca, Mg, Fe, Cu, Zn and Pb in the Plant Bupleurum scorzoneri folium Willd. after digestion with HNO3-HClO4 (V : V= 4 : 1) at 90-95 degrees C and at normal pressure. In the meantime, the optimum parameters of FAAS and the effects of solution medium on the results were studied. The analytical results of Ca, Mg, Fe, Cu and Zn were 5,588.9, 1,790.5, 869.3, 78.4, 44.3 microg x g(-1) respectively, and Pb was not checked out in the samples. The results showed that the recovery of standard addition was 99.57%-102.10%, the relative standard deviation (n= 9) was 0.18%-2.26%. The method is quick, simple and convenient, and the results are satisfactory.

RESUMO

OBJECTIVE: To study the structure and three-dimensional distribution of extraocular muscle Pulley in orbit. METHODS: 5 fresh cadaver orbits were fixed in situ and serially sectioned at 4 microm thickness. Every nearby sections were stained using Masson's trichrome, Van Gieson'stain and immunohistochemical staining, the stained tissue was examined by gross anatomy and light microscopy. RESULTS: Near the equator of the globe, extraocular muscle Pulleys were at the points where rectus penetrate posterior Tenon's fascia, encircling the rectus orbit side by fiber ring structure. There was Pulley Band among extraocular muscle Pulleys. The main composition of the Pulley and Pulley Band were collagen (I and III type), elastic and smooth muscle. CONCLUSION: Constitution of Pulley provides dissection basis as the functional origins of the extraocular muscle.

OBJECTIVE: To study the maternal blood glucose management of gestational diabetes mellitus during the labor and relationship of maternal blood glucose in labor and neonatal blood sugar. METHODS: A prospective study on maternal blood glucose monitoring and control during labor was carried in 40 pregnant women with abnormal glucose metabolism. We adjusted maternal blood glucose by low-dosage constant insulin drop during labor. RESULTS: Maternal blood glucose in labor arranged between 3.8 approximately 11.2 mmol/L, and low-dosage constant insulin drop was used in 17 cases (42.5%). The neonatal blood glucose was (4.0 +/- 1.5) mmol/L following delivery and (3.9 +/- 1.0) mmol/L at 24-hour postpartum. There were 2 cases with neonatal hypoglycemia. The neonatal blood glucose of the rest 23 cases was (4.2 +/- 1.5) mmol/L, and (3.9 +/- 1.0) mmol/L at 24-hour postpartum, and 1 case with neonatal hypoglycemia. There was no the difference of the blood glucose level of newborns between mothers with abnormal glucose metabolism and the normal pregnant women. CONCLUSION: It is necessary to monitor and control the blood glucose level by low dose constant insulin during labor in pregnant women with abnormal glucose metabolism.
