RESUMO

The body color of Neocaridina denticulate sinensis is a compelling phenotypic trait, in which a cascade of carotenoid metabolic processes plays an important role. The study was conducted to compare the transcriptome of cephalothoraxes among three pigmentation phenotypes (red, blue, and chocolate) of N. denticulate sinensis. The purpose of this study was to explore the candidate genes associated with different colors of N. denticulate sinensis. Nine cDNA libraries in three groups were constructed from the cephalothoraxes of shrimps. After assembly, 75022 unigenes were obtained in total with an average length of 1026 bp and N50 length of 1876 bp. There were 45977, 25284, 23605, 21913 unigenes annotated in the Nr, Swissprot, KOG, and KEGG databases, respectively. Differential expression analysis revealed that there were 829, 554, and 3194 differentially expressed genes (DEGs) in RD vs BL, RD vs CH, and BL vs CH, respectively. These DEGs may play roles in the absorption, transport, and metabolism of carotenoids. We also emphasized that electron transfer across the inner mitochondrial membrane (IMM) was a key process in pigment metabolism. In addition, a total of 6328 simple sequence repeats (SSRs) were also detected in N. denticulate sinensis. The results laid a solid foundation for further research on the molecular mechanism of integument pigmentation in the crustacean and contributed to developing more attractive aquatic animals.

RESUMO

A rapid increase in the number of end-of-life (EoL) passenger vehicles has led to a large amount of waste plastics in China. However, the scale and efficiency of recycling resources from EoL vehicles still restricts the sustainable and healthy development of the automotive industry. The current behavior of automotive/recycling industry entities, as well as the strategy of waste management policymakers, may depend on the potential of total recyclable resources. To reveal such recycling potential of various plastic materials in EoL passenger vehicles, we predicted total EoL passenger vehicles in China from 2021 to 2030 (used the Weibull distribution) considering passenger vehicle ownership (estimated by the Gompertz model), quantified the demand for new passenger vehicles (estimated using its non-linear relationship with income level and passenger vehicle ownership), and assessed the recyclable plastics by categories and by provinces. The results show that (i) the annual average recycled plastic resources from EoL vehicles would exceed 2400 thousand t in 2030, more than 2.5 times in 2021, showing a great recycling potential; (ii) the differences among the three scenarios are relatively small, indicating that no matter the saturation level of passenger vehicles in China would be high or low, a rapid increase of recyclable plastic resources can be expected from 2021 to 2030; (iii) at the provincial level, a considerable gap between the potential of recycling plastic from EoL passenger vehicles and the regional processing capacity. Given such great potential and regional differences, the recycling policies should be applied in stages and consider the development level and recovery pressure in each region.

The rice bran polysaccharide was extracted from defatted rice bran, and the optimal conditions of the experiment were determined. The chemical structure of rice bran polysaccharide was analyzed by IR and 13 C NMR. Three rice bran polysaccharide derivatives were prepared and characterized, namely carboxymethylated rice bran polysaccharide, phosphorylated rice bran polysaccharide, and acetylated rice bran polysaccharide. It showed that the phosphorylated rice bran polysaccharide had good activity.

Penaeus vannamei is the principle cultured shrimp species in China. However, with the increase of culture density, the growth difference between individuals is also expanding. Here, we make use of RNA-seq to study the growth mechanisms of P. vannamei. After 120 days, we examined the transcriptomes of rapid-growing individuals (RG) and slow-growing individuals (SG). A total of 2116 and 176 differentially expressed genes (DEGs) were found in SG and RG, respectively. Moreover, the main DEGs are opsin, heat shock protein (HSP), actin, myosin, superoxide dismutase (SOD), cuticle protein, and chitinase. GO analysis further revealed that the DEGs were enriched in biological processes significantly, such as "sensory perception," "sensory perception of light stimulus," "response to stimulus," and "response to stress." Additionally, KEGG enrichment analysis showed that the DEGs were mainly enriched in "pentose and glucuronate interconversions," "amino sugar and nucleotide sugar metabolism," "glycophospholipid biosynthesis," and "glutathione metabolism." Interestingly, the upstream genes in the ecdysone signaling pathway, including molting inhibition hormone (MIH) and crustacean hyperglycemic hormone (CHH), did not differ significantly between RG and SG, which suggests that the cause for the inconsistent growth performance is due to the stress levels rather than the ecdysone signal pathway. In summary, this work provides data that will be useful for future studies on shrimp growth and development.

RESUMO

The Epstein-Barr virus (EBV) is endemic in humans and can efficiently transform infected B cells under some circumstances. If an EBV carrier experiences immune suppression, EBV+ B cells can turn into lymphoblasts and exhibit growth expansion that may cause lymphoproliferative diseases which often develop into lymphoma. Our immune system conducts surveillance for EBV+ B cells in order to block spontaneous tumor formation. Here, we summarize the EBV products involved in tumorigenesis, EBV-associated lymphomas, and pathologically relevant mouse models. Preclinical mouse models for a range of EBV-associated diseases not only clear the path to new therapeutic approaches but also aid in our understanding of the nature of lymphomagenesis and immune surveillance.

Rice bran polysaccharide has a wide range of physiological functions, which has aroused great interest of scientists at home and abroad. It is an ideal healthcare product. The polysaccharide was extracted from defatted rice bran with water. It was subjected to protein removal with the Sevag reagent. The chemical structure of rice bran polysaccharide was analyzed by infrared spectroscopy and nuclear magnetic resonance spectroscopy. The polysaccharide significantly increased the content of superoxide dismutase and catalase in the serum, liver and spleen of mice, and decreased the content of malondialdehyde in the serum, liver and spleen to a certain extent. The polysaccharide could upregulate the expression of nuclear factor E2-related factor 2 and its downstream antioxidant factors NQ01 and HO-1, and enhance the activity of the antioxidant response element luciferase.

The purpose of this research was conducted to determine anthocyanin constituent from the grains of Purple black rice No. 6. Moreover, the in vitro antioxidant activity of black rice pigment (BRP) was evaluated. The crude extract of black rice was isolated and purified by silica gel thin layer chromatography. Anthocyanins were elucidated using ultraviolet visible spectroscopy, infrared spectroscopy, nuclear magnetic resonance spectroscopy, and high-performance liquid chromatography. Meanwhile, salicylic acid Fenton system, superoxide radical system, H2 O2 scavenging system, DPPH-free radical scavenging system, and Prussian blue method were used to determine the free radical scavenging ability and total reducing ability of BRP. Overall results suggest that the main component of BRP is cyanidin-3-glucoside, which is a kind of anthocyanins. It also had significant scavenging capacity to OH, O2 - , H2 O2 , DPPH, and its scavenging rate and reducing ability increased with the increase in pigment concentration.

In recent years, glucan has become one of the hotspots in life science research. It is widely concerned because of its advantages, such as good immune regulation, antitumor, antivirus, hypoglycemic and lipid-lowering effects, antioxidation, antiaging, low toxicity, and high efficiency. At present, there are still two problems in the pharmacodynamics of glucans. Firstly, compared with other drugs used in clinic, most of the glucans still lack information about the mechanism of action between glucans and corresponding ligands in vivo. Secondly, the research on the structure-activity relationship of glucans is still slow. Herein, the structure-activity relationships of glucans were reviewed. The relationship between structure and biological activity is very important for the development of new glucan drugs.

Selenium polysaccharide is an organic selenium compound formed by the combination of polysaccharide and selenium. The biological activity of selenium polysaccharide from plant such as Grifola frondosa is generally higher than that of selenium and polysaccharide, and it is more easily absorbed and utilized by human body. Therefore, selenium polysaccharide has a wide range of applications in immune regulation, anti-tumor, anti-oxidation, anti-aging and so on. Due to the unique pharmacological activity of selenium polysaccharide, the research of selenium polysaccharide has gradually become a hot spot. At present, there are few kinds of selenium polysaccharide, but its structure is very complex. The chemical structure and mechanism of selenium polysaccharide in vivo are not completely clear and need further study. Herein, the preparation and biological activities of selenium polysaccharides were introduced systematically, which provided theoretical basis for further research and application of selenium polysaccharides.

Polysaccharides have various biological activities, such as antioxidant, anti-tumor, antiviral, anti-aging, anti-inflammatory, anti-radiation, hypoglycemic and lipid lowering, immune regulation, etc. Although many polysaccharides have been proved to have various biological activities, in the research process, it has been found that more polysaccharides in nature do not have biological activities or have low activities, and some polysaccharides have poor activities due to lack of certain groups. While others are due to polysaccharide with too large molecular weight, because its molecular weight is too large to smoothly enter cells and thus cannot exert its activity. For the former, we need to adopt molecular grafting modification. This kind of modification method is mainly chemical modification method, which increases the functional groups and molecular weight of various polysaccharides to improve their biological activity. For the latter, we need to adopt molecular degradation modification, which includes biodegradation, physical degradation, chemical degradation and other means to improve the solubility of polysaccharide in water phase by reducing its molecular weight, thus improving its activity. This article mainly introduces polysaccharide structure modification methods that have been widely used in recent years.

In soybean (Glycine max)-rhizobium interactions, the type III secretion system (T3SS) of rhizobium plays a key role in regulating host specificity. However, the lack of information on the role of T3SS in signaling networks limits our understanding of symbiosis. Here, we conducted an RNA sequencing analysis of three soybean chromosome segment substituted lines, one female parent and two derived lines with different chromosome-substituted segments of wild soybean and opposite nodulation patterns. By analyzing chromosome-linked differentially expressed genes in the substituted segments and quantitative trait loci (QTL)-assisted selection in the substituted-segment region, genes that may respond to type III effectors to mediate plant immunity-related signaling were identified. To narrow down the number of candidate genes, QTL assistant was used to identify the candidate region consistent with the substituted segments. Furthermore, one candidate gene, GmDRR1, was identified in the substituted segment. To investigate the role of GmDRR1 in symbiosis establishment, GmDRR1-overexpression and RNA interference soybean lines were constructed. The nodule number increased in the former compared with wild-type soybean. Additionally, the T3SS-regulated effectors appeared to interact with the GmDDR1 signaling pathway. This finding will allow the detection of T3SS-regulated effectors involved in legume-rhizobium interactions.

Glucose-6-phosphate dehydrogenase (G6PDH) is known as a critical enzyme responsible for nicotinamide adenine dinucleotide phosphate (NADPH) generation in the pentose phosphate pathway (PPP), and has an essential function in modulating redox homeostasis and stress responsiveness. In the present work, we characterized the nine members of the G6PDH gene family in soybean. Phylogenic analysis and transit peptide prediction showed that these soybean G6PDHs are divided into plastidic (P) and cytosolic (Cy) isoforms. The subcellular locations of five GmG6PDHs were further verified by confocal microscopy in Arabidopsis mesophyll protoplasts. The respective GmG6PDH genes had distinct expression patterns in various soybean tissues and at different times during seed development. Among them, the Cy-G6PDHs were strongly expressed in roots, developing seeds and nodules, while the transcripts of P-G6PDHs were mainly detected in green tissues. In addition, the activities and transcripts of GmG6PDHs were dramatically stimulated by different stress treatments, including salt, osmotic and alkali. Notably, the expression levels of a cytosolic isoform (GmG6PDH2) were extraordinarily high under salt stress and correlated well with the G6PDH enzyme activities, possibly implying a crucial factor for soybean responses to salinity. Enzymatic assay of recombinant GmG6PDH2 proteins expressed in Escherichia coli showed that the enzyme encoded by GmG6PDH2 had functional NADP+-dependent G6PDH activity. Further analysis indicated overexpression of GmG6PDH2 gene could significantly enhance the resistance of transgenic soybean to salt stress by coordinating with the redox states of ascorbic acid and glutathione pool to suppress reactive oxygen species generation. Together, these results indicate that GmG6PDH2 might be the major isoform for NADPH production in PPP, which is involved in the modulation of cellular AsA-GSH cycle to prevent the oxidative damage induced by high salinity.

Dextran has become a hot research topic in drug vehicle material because of its biodegradable, nonspecific cell adhesion, resistance to protein adsorption, low price and ease of structural modification. The fate and changes of dextran in vivo are not fully understood. It is helpful to guide the design and modification of dextran drug vehicles to clarify the changes in the morphology, metabolism and function of drug targets. With the deep understanding of dextran and the emergence of new functional dextran derivatives, its application in nanodrug delivery systems will be more and more, clinically applicable delivery systems may also be available.

Soybeans are an important cash crop and are widely used as a source of vegetable protein and edible oil. MicroRNAs (miRNA) are endogenous small RNA that play an important regulatory role in the evolutionarily conserved system of gene expression. In this study, we selected four lines with extreme phenotypes, as well as high or low protein and oil content, from the chromosome segment substitution line (CSSL) constructed from suinong (SN14) and ZYD00006, and planted and sampled at three stages of grain development for small RNA sequencing and expression analysis. The sequencing results revealed the expression pattern of miRNA in the materials, and predicted miRNA-targeted regulatory genes, including 1967 pairs of corresponding relationships between known-miRNA and their target genes, as well as 597 pairs of corresponding relationships between novel-miRNA and their target genes. After screening and annotating genes that were targeted for regulation, five specific genes were identified to be differentially expressed during seed development and subsequently analyzed for their regulatory relationship with miRNAs. The expression pattern of the targeted gene was verified by Real-time Quantitative PCR (RT-qPCR). Our research provides more information about the miRNA regulatory network in soybeans and further identifies useful genes that regulate storage during soy grain development, providing a theoretical basis for the regulation of soybean quality traits.

Flexible perovskite solar cells (PSCs) have been quickly developed as the most promising candidates for low-cost photovoltaic technology. However, the bendable and foldable properties of PSCs induce the decrease of their efficiencies. In this paper, we report the design of a new kind of flexible PSCs with a sandwiched structure. The critical layer of the flexible device is designed at a neutral layer of the sandwiched structure, which is stress-free, no matter how the device bending is. During the bending test, sandwich-structured flexible PSCs showed extremely long bending lifetime, which is at least 5-8 times higher than that of generally reported devices. At the same time, the sandwiched structure works as the encapsulation effect. The flexible device with a sandwiched structure greatly improves the device's long-term stability. Therefore, the designed sandwiched structure significantly promotes the bending ability and stability of flexible PSCs.

Dextran as a drug carrier for inhibiting cancer cells effectively reduces the toxic and side effects of the drug in the biological body. Targeting improves the concentration of active substance around the target tissue, which reduces damage to other heavy organs and other normal tissues. Dextran will be a potential carrier for the delivery of antitumor drugs in the future, which provides the possibility of slow-release chemotherapy and targeted drug delivery. Herein, the preparation and drug delivery of dextran-drug complex were summarized and discussed in detail.

Pulmonary embolism and cardiac tamponade are potentially fatal acute conditions that rarely present concomitantly in the emergency department (ED). Both require early diagnosis and urgent intervention, and are usually observed as separate easily identifiable diseases. However, in a patient exhibiting a concomitant presentation of pulmonary embolism with cardiac tamponade, diagnosis and therapeutic intervention are extremely challenging. A 48-year-old woman presented with cardiac tamponade as an initial symptom of an underlying lung adenocarcinoma and masked massive pulmonary embolism (MPE), which led to the development of sudden cardiac arrest after successful pericardiocentesis. She presented with a high index of suspicion for a diagnosis of MPE using echocardiography after successful pericardiocentesis, and this diagnosis was confirmed using computed tomography. Extracorporeal membrane oxygenation and adjusted-dose unfractionated intravenous heparin administration were performed; unfortunately, they were unsuccessful. This report would help ED physicians because this case demonstrates that lung cancer can initially present as pulmonary embolism with cardiac tamponade and pulmonary embolism can be misdiagnosed in the presence of concomitant cardiac tamponade. Bedside echocardiography may fail to diagnose life-threatening MPE with coexisting cardiac tamponade. MPE can also lead to the development of sudden cardiac arrest after successful pericardiocentesis. Thrombolytic and anticoagulant use in MPE with coexisting hemorrhagic cardiac tamponade is a controversial issue. The risk-benefit ratio of both therapies needs to be considered on a case-by-case basis for improved clinical outcomes.

RESUMO

Epothilone, the macrolide compound produced by Sorangium cellulosum, has antitumor activity. Its anti-tumor mechanism is similar to that of paclitaxel, which promotes the polymerization of tubulin and induces apoptosis. Herein, 7-O-ß-d-galactosyl-polyethylene glycol-epothilone B 6 was synthesized. It showed that the toxicity of the synthesized compound was 1/182 of the epothilone B. In addition, compound 6 also had significant anticancer activity under the action of enzyme.
