An on-line high pH-reversed phase liquid chromatography×low pH-reversed phase liquid chromatography tandem electrospray ionization mass spectrometry combined with pulse elution gradient in the first dimension was constructed to separate and identify alkaloids from M. cordata (willd.) R. Br.. The modulation was performed by using a dual second dimensional columns interface combined with a make-up dilution pump, which responsible for dilution and neutralization of the first dimensional effluent, and the dual second dimensional columns integrated the trapping and the separation function to reduce the second dimension system dead volume. Taking advantage of the dissociable characteristics of alkaloids, mobile phases with different pH values were applied in the first dimension (pH = 9.0) and the second dimension (pH = 2.6) to improve the orthogonality of two-dimension separation. Besides, the pulse elution gradient in first dimension and second dimensional gradient were carefully optimized and much better separation was achieved comparing to the separation with the traditional two-dimensional liquid chromatography approach. Finally, mass measurement for alkaloids in Macleaya cordata (willd.)R. Br. was performed by coupling proposed two-dimensional liquid chromatography system with triple quadrupole mass spectrometry, and 39 alkaloids were successfully identified by comparing the obtained result with the former reported results. OBJECTIVES: Identifying inflammation- or fibrosis-predominant strictures in Crohn's disease (CD) is crucial for treatment strategies. We evaluated the additive value of magnetisation transfer (MT) to conventional MRI for differentiating CD strictures using surgical histopathology as a reference standard. METHODS: Twenty-eight consecutive CD patients who underwent MRI preoperatively were recruited. MRI parameters included T2-weighted imaging (T2WI) hyperintensity, bowel wall thickness, enhancement pattern changes over time, enhancement pattern and gain ratio in dynamic contrast-enhanced phases, and MT ratio. Correlation analysis was performed using Spearman's rank test. Receiver operating characteristic curve analysis and Cohen's κ were used. A model with combined MRI variables characterising intestinal strictures was proposed and validated in 14 additional CD patients. RESULTS: Significant correlations with histological inflammation scores were shown for wall thickness (r = 0.361, p = 0.001) and T2WI hyperintensity (r = 0.396, p < 0.001), whereas histological fibrosis scores were significantly correlated with MT ratio (r = 0.681, p < 0.001) and wall thickness (r = 0.461, p < 0.001). T2WI hyperintensity could differentiate mild from moderate-to-severe inflammation with a sensitivity of 0.871 and a specificity of 0.800. MT ratio could discriminate mild from moderate-to-severe fibrosis with a sensitivity and a specificity of 0.913 and 0.923, respectively. Combining MT ratio and T2WI hyperintensity, the MRI classification moderately agreed with the pathological stricture classification (p < 0.01, κ = 0.549). In the validation set, the diagnostic accuracy of T2WI hyperintensity and MT ratio were 86% and 89%, with good agreement between MRI and histopathological classification (p < 0.01, κ = 0.665). CONCLUSIONS: MT ratio combined with conventional MRI improves the differentiation of fibrotic from inflammatory components of small-bowel strictures in CD patients. KEY POINTS: • MT ratio from magnetisation transfer imaging combined with T2WI from conventional MRI can simultaneously characterise bowel fibrosis and inflammation in adult Crohn's disease.

An in-situ approach is described for synthesis of poly(sulfobetaine-co-polyhedral oligomeric silsesquioxane) [poly(sulfobetaine-co-POSS)] that can be used in a hybrid monolithic column as a hydrophilic liquid chromatography (HILIC) stationary phase. Synthesis involves (a) radical polymerization of octa(propyl methacrylate)-polyhedral oligomeric silsesquioxane (MA-POSS) and organic monomers such as dimethylaminopropyl methacrylate or vinyl imidazole, and (b) in-situ ring-opening quaternization between 1,4-butane sultone and the organic monomers. The sulfobetaine groups are generated in-situ monolith. This obviates the need for synthesis of sulfobetaine monomer previously. The pore size and permeability of the material can be tuned by using a binary porogenic system (polyethyleneglycol 600 and acetonitrile) and via the composition of the polymerization mixture. The optimized hybrid monolith owns its merits to the presence of POSS and sulfobetaine groups with good mechanical stability, the lack of residual silanol groups, and adequate hydrophilicity. The column filled with the monoliths was evaluated as a stationary phase for HILIC. Several kinds of polar compounds (including nucleosides, bases, phenols, aromatic acids and amides) were separated by using mobile phases with high organic solvent fractions in capillary liquid chromatography. Detection of hydrogen peroxide (H2O2) produced by living cells is very significant to fully understand its roles in cellular physiology, as well as providing reliable diagnosis of pathological conditions. However, in situ detection of H2O2 released from adherent cells in cellular culture medium is still insufficiently achieved. Here, we report an electrochemical platform for in situ detection of H2O2 produced by adherent cells in cellular culture medium. It is based on the use of organic electrochemical transistor (OECT) fabricated on a flexible poly(ethylene terephthalate) substrate and Transwell support. A screen-printed carbon paste electrode was modified with carbon nanotubes and platinum nanoparticles and served as the gate of the device. Under optimal conditions, this device exhibits good modulation and sensitivity. It works in the 0.5 µM to 0.1 mM H2O2 concentration range and has a 0.2 µM detection limit. The cells were seeded and grew on the Transwell membrane. Upon being stimulated by N-formylmethionyl-leucyl-phenylalanine peptide, H2O2 produced by the adherent cells diffused into the bottom chamber of the Transwell and was in situ detected by OECT. Moreover, evaluating in vitro cytotoxicity of the nanomaterial using the OECT-Transwell platform was realized. This simple electrochemical platform would be of great interest for in vitro cytotoxicity, cellular physiology study, and diagnosis of pathological conditions.

Ecosystem service value (ESV) in 1997, 2005, 2013 and 2015 of Quanzhou Bay estuary wetland was evaluated dynamicly by market value, alternative value and opportunity cost methods, combined with classification of service indicators and deduplicate computing. The main driving forces for the changes of ESV and the pathway and intensity of their actions were identified using stepwise regression and path analysis methods. The results showed that the main ecosystem services of Quanzhou Bay estuary wetland were flood regulation, climate regulation, and food supply, which were directly driven by water supply, mariculture carrying capacity, and gross value of the regional production. Other drivers exerted indirect effect on the changes of main ESVs.

Obstructive sleep apnea (OSA) is closely associated with central nervous system diseases and could lead to autonomic nerve dysfunction, which is often seen in neurodegenerative diseases. Previous studies have shown that metoprolol prevents several chronic OSA-induced cardiovascular diseases through inhibiting autonomic nerve hyperactivity. It remains unclear whether chronic OSA can lead to dendritic remodeling in the brain, and whether metoprolol affects the dendritic remodeling. In this study we investigated the effect of metoprolol on dendrite morphology in a canine model of chronic OSA, which was established in beagles through clamping and reopening the endotracheal tube for 4 h every other day for 12 weeks. OSA beagles were administered metoprolol (5 mg· kg-1· d-1). The dendritic number, length, crossings and spine density of neurons in hippocampi and prefrontal cortices were assessed by Golgi staining. And the protein levels of hypoxia-inducible factor-1α (HIF-1α) and brain-derived neurotrophic factor (BDNF) were measured by Western blotting. We showed that chronic OSA successfully induced significant brain hypoxia evidenced by increased HIF-1α levels in CA1 region and dentate gyrus of hippocampi, as well as in prefrontal cortex. Furthermore, OSA led to markedly decreased dendrite number, length and intersections, spine loss as well as reduced BDNF levels. Administration of metoprolol effectively prevented the dendritic remodeling and spine loss induced by chronic OSA. In addition, administration of metoprolol reversed the decreased BDNF, which might be associated with the metoprolol-induced neuronal protection. In conclusion, metoprolol protects against neuronal dendritic remodeling in hippocampi and prefrontal cortices induced by chronic OSA in canine.

Background: Ulcerative colitis (UC) patients have an increased risk for the development of colorectal cancer (CRC). Our aim was to assess the risk of CRC in UC patients compared with disease extent, disease duration, and geographic variation. Methods: In this systematic review and meta-analysis, we searched PubMed, scientific meetings, and the bibliographies of identified articles, with English language restrictions for studies published from 1988 to 2018, and assessed the risk of CRC in UC patients. Patients with Crohn's disease, family history of CRC, and colorectal adenomatous polyp (CAP) were excluded from this research. The study was registered with PROSPERO, number CRD42018102213. Findings: We included 58 studies that included 267566 UC patients. Extensive UC and left-sided UC had a higher risk of CRC than proctitis UC. Geography also played a role in UC-associated CRC development. The time of malignant transformation in Asian UC patients started after 10-20 years of this disease duration. North American UC-associated CRC patients significantly increased in more than 30 years of this disease duration. Conclusion: In a systematic review of the literature, we found that disease extent, disease duration, and geography were strong, independent risk factors in UC-associated CRC development.

OBJECTIVE: To observe the effect of electroacupuncture (EA) on the expression of apolipoprotein E (ApoE) and related proteins of inflammation and anti-oxidative stress in spinal cord in mice with spinal cord injury (SCI), so as to explore its mechanisms underlying function repair. METHODS: Thirty-six female C57BL/6 mice were equally randomized into 3 groups: sham operation, model and EA. The SCI model was established by clamping the spinal cord for 25 s with a serrefine after laminectomy of the 1st lumbar vertebra (L1). EA (1.5 Hz/7.5 Hz, 1.0 mA) was applied to bila-teral "Zusanli" (ST36) and "Sanyinjiao" (SP6) for 10 min, once a day for 7 days. The hindlimb locomotor function was assessed according to the state of the range of motion, coordination, claw gesture of the hind leg ankle-joint, trunk stabi-lity and the tail posture by using Basso Mouse Scale(BMS). The histopathological changes of the injured area of the spinal cord were determined by H.E. staining. The expression levels of ApoE, phosphorylated nuclear transcription factor-κB(p-NF-κB), interleukin 1 beta(IL-1ß), phosphorylated extracellular regulatory protein kinase(p-ERK1/2), extracellular regulatory protein kinase(ERK1/2), nuclear factor erythroid 2-related factor 2(Nrf2) and heme oxidase-1(HO-1) in the spinal cord were detected by Western blot, and the glial fibrillary acidic protein (GFAP)-positive astrocytes were displayed by immunofluorescence staining. RESULTS: After modeling, the BMS scores were significantly decreased in the model group compared with the sham operation group (P<0.05). Following EA, the BMS scores were markedly increased in the EA group relevant to the model group (P<0.05), suggesting an improvement of the hindlimb locomotor function. H.E. stain showed structural disorder with lots of cavities, severe inflammatory infiltration with large quantity of inflammatory cells, and apparent reduction of normal neurons in the injured spinal cord tissue of model group, which was milder in the EA group. The expression levels of ApoE, p-NF-κB, IL-1ß, p-ERK1/2 (not ERK1/2), Nrf2 and HO-1 were significantly increased in the model group than those in the sham operation group (P<0.05). Compared with the model group, the expression levels of ApoE, p-ERK1/2, Nrf2 and HO-1 were further notably up-regulated (P<0.05), and those of p-NF-κB and IL-1ß proteins obviously down-regulated in the EA group (P<0.05). Immunoflorescence staining showed that the number of GFAP-positive cells was apparently increased in the model group compared with the sham operation group and observably decreased in the EA group relevant to the model group (P<0.05). CONCLUSION: EA can significantly improve locomotor function in SCI mice, which is associated with its effects in reducing inflammation, oxi-dative stress reactions and reactive astrocyte proliferation via up-regulating expression of ApoE, p-ERK1/2, and Nrf2/HO-1 (antioxidant pathway) and inhibiting IL-1ß and NF-κB expression.

The mechanism and effective treatment of bisphosphonate-related osteonecrosis of the jaw (BRONJ) are still uncertain. Our previous study revealed that zoledronate (ZOL) preferentially inhibited osteoclasts formation and platelet-derived growth factor-BB (PDGF-BB) secretion, causing suppression of angiogenesis and osteogenesis in vitro. The present study aimed to elucidate whether PDGF-BB had therapeutic effects on rat model of BRONJ by enhancing angiogenesis and angiogenesis. Firstly, rat model of BRONJ was established by ZOL and dexamethasone administration, followed by teeth extraction. The occurrence of BRONJ was confirmed and detected dead bone formation by maxillae examination, micro-CT scan and HE staining (10/10). Compared to control rats (0/10), both angiogenesis and mature bone formation were suppressed in BRONJ-like rats, evidenced by enzyme-linked immunosorbent assay (ELISA) for VEGF (P < 0.01), immunohistochemistry of CD31 (P < 0.05) and OCN (P < 0.01). Moreover, in the early stage of bone healing, the number of preosteoclasts (P < 0.001) and PDGF-BB secretion (P < 0.05) were significantly decreased in bisphosphonates-treated rats, along with the declined numbers of microvessels (P < 0.05) and osteoblasts (P < 0.05). In vitro study, CCK8 assay, alizarin red S staining and western blot assay showed that mandible-derived bone marrow mesenchymal stem cells (BMMSCs) in BRONJ-like rats presented suppressed functions of proliferation, osteogenesis and angiogenesis. Interestingly, recombinant PDGF-BB was able to rescue the impaired functions of BMMSCs derived from BRONJ-like rats at more than 10 ng/ml. Then fibrin sealant with or without recombinant PDGF-BB were tamped into the socket after debridement in BRONJ rats. After 8 weeks, fibrin sealant containing PDGF-BB showed significant therapeutic effects on BRONJ-like rats (bone healing: 8/10 vs 3/10, P < 0.05) with enhancing microvessels and mature bone formation. Our study suggested that the inhibition of angiogenesis and osteogenesis, the potential mechanisms of BRONJ, might partly result from suppression of PDGF-BB secretion in the early stage of bone healing. PDGF-BB local treatment after debridement might avail the healing of BRONJ by increasing angiogenesis and osteogenesis.

Background: Podocyte-derived microparticles (MPs) could be secreted from activated or apoptotic podocytes. An increased number of podocyte-derived MPs in the urine might reflect podocyte injury in renal diseases. This study aimed to observe the change of urinary podocyte-derived MP levels in patients with chronic kidney disease (CKD) and to further explore its correlation with the progression of CKD. Methods: A prospective, longitudinal study was conducted in eighty patients with biopsy-proven CKD. Podocyte-derived MPs (annexin V and podocalyxin positive) were detected by flow cytometry. The number of urinary podocyte-derived MPs was analyzed to evaluate the association with biochemical measurements and pathological glomerulosclerosis assessment. Patients with idiopathic membranous nephropathy (IMN) were followed up after the six-month treatment of prednisone combined with tacrolimus to evaluate the association of urinary podocyte-derived MP levels and the remission of IMN. Results: The CKD patients had higher urinary podocyte-derived MP levels compared with healthy controls (HCs). Baseline urinary levels of podocyte-derived MPs were positively correlated with 24-hour proteinuria, while were inversely correlated with the percentage of global glomerulosclerosis. The urinary podocyte-derived MPs levels had good discrimination for glomerulosclerosis [area under curve (AUC), 0.66]. The urinary podocyte-derived MPs levels in IMN patients were significantly decreased accompanied with the recovery of abnormal clinical parameters after six-month treatment. Conclusions: The urinary levels of podocyte-derived MPs were closely associated with podocyte injury and glomerulosclerosis, which could be useful for monitoring disease activity in CKD patients. Urinary podocyte-derived MPs might be a non-invasive biomarker for the evaluation of early CKD progression.

The atomistic simulation of supported metal catalysts has long been challenging due to the increased complexity of dual components. In order to determine the metal/support interface, efficient theoretical tools to map out the potential energy surface (PES) are generally required. This work represents the first attempt to apply the recently developed SSW-NN method, stochastic surface walking (SSW) global optimization based on global neural network potential (G-NN), to explore the PES of a highly controversial supported metal catalyst, Au/CeO2, system. By establishing the ternary Au-Ce-O G-NN potential based on first principles global dataset, we have searched for the global minima for a series of Au/CeO2 systems. The segregation and diffusion pathway for Au clusters on CeO2(111) are then explored by using enhanced molecular dynamics. Our results show that the ultrasmall cationic Au clusters, e.g., Au4O2, attaching to surface structural defects are the only stable structural pattern and the other clusters on different CeO2 surfaces all have a strong energy preference to grow into a bulky Au metal. Despite the thermodynamics tendency of sintering, Au clusters on CeO2 have a high kinetics barrier (>1.4 eV) in segregation and diffusion. The high thermodynamics stability of ultrasmall cationic Au clusters and the high kinetics stability for Au clusters on CeO2 are thus the origin for the high activity of Au/CeO2 catalysts in a range of low temperature catalytic reactions. We demonstrate that the global PES exploration is critical for understanding the morphology and kinetics of metal clusters on oxide support, which now can be realized via the SSW-NN method.

BACKGROUND: A validated histopathological tool to precisely evaluate bowel fibrosis in patients with Crohn's disease is lacking. We attempted to establish a new index to quantify the severity of bowel fibrosis in patients with Crohn's disease-associated fibrostenosis. METHODS: We analyzed the histopathological data of 31 patients with Crohn's disease strictures undergoing surgical resection. The most representative sections of resected strictured segments were stained with Masson trichrome to manifest bowel fibrosis. The collagen area fraction and histological fibrosis score were simultaneously calculated for the same section to evaluate the severity of bowel fibrosis. RESULTS: Collagen area fraction strongly correlated with histological fibrosis scores (r = 0.733, P < 0.001). It showed a stronger correlation (r = 0.561, P < 0.001) with the degree of bowel strictures than the histological fibrosis score did (r = 0.468, P < 0.001). It was also shown to be more accurate for diagnosing Crohn's disease strictures (area under the receiver operating characteristic curve = 0.815, P < 0.001) compared with the histological fibrosis score (area under the curve = 0.771, P < 0.001). High repeatability was observed for the collagen area fraction, with an intraclass correlation coefficient of 0.915 (P < 0.001). CONCLUSIONS: Collagen area fraction is a simple and reliable index to quantify the severity of bowel fibrosis in patients with Crohn's disease-associated fibrostenosis.

RESUMO

The little known species Nudina xizangensis Fang, 2000 is rediscovered from its type locality, Beibeng, Motuo County, Southeast Xizang, with genitalia of both sexes illustrated for the first time. Based on its unique genitalia structure, a new lichen moth genus, Paranudina Huang Wang gen. nov., is described with N. xizangensis as the type species. Adults and genitalia of Paranudina xizangensis (Fang, 2000) comb. nov. and Nudina artaxidia (Butler, 1881) are compared and illustrated.

RESUMO

Some new information on a poorly studied lepidopterous family, Immidae Heppner, 1977, is reported from Guangdong Province, in the southern part of mainland China. A new species, viz. Moca austrasinensis Huang Wang sp. nov. is described from Mt. Wutong, Shenzhen. Imma lathidora Meyrick, 1914 is recorded in Mt. Nankun, Huizhou, representing the first record from mainland China. Adults and male genitalia of the species mentioned are illustrated.

A healthy 52-year-old woman presented with 10-days history of back pain. Neurologic testing failed to detect any functional deficits in the upper and lower extremities, and the patient had a full range of cervical spine motion without associated pain. Spinal CT and MRI revealed a well-circumscribed intradural mass located at conus medullaris. The operation of L1 hemilaminectomy was performed, and pathological examination discovered dilatation of sweat ducts and suggested the diagnosis of ectopic sweat duct ectasia. IHC staining in epithelia immunophenotype showed: pan-cytokeratin (PCK)(+), epithelia membrane antigen (EMA)(+), P63(+), cytokeratin 5/6 (CK5/6)(+), gross cystic disease fluid protein 15 (GCDFP15)(-). Intraspinal ectopic sweat duct ectasia is extremely rare, which has not been reported in the literature to date.

The Barsine hypoprepioides (Walker, 1862) species-group is reviewed. Forty seven species and one subspecies belong to the species-group. Among them, fifteen species and one subspecies are new for science and described in the present paper: B. amoenissima sp. n. (Myanmar), B. selene sp. n. (Thailand and Laos), B. cao sp. n. (Thailand, Laos and Vietnam), B. speideli sp. n. (SE China, Thailand, Laos, Cambodia and Vietnam), B. mesomene sp. n. (NE India, Myanmar, China, Thailand, Laos and Vietnam), B. pandeia sp. n. B. takamukui (Matsumura, 1927), comb. n., B. karenkensis (Matsumura, 1930), comb. n., B. rhipiptera (Wileman West, 1928), comb. n., B. chi (Roepke, 1946), comb. nov., B. salakia (Schaus, 1922), comb. n., B. marginis (Fang, 1991), comb. nov. and B. dentata (Wileman, 1910), comb. nov. The lectotype is designated for Lithosia hypoprepioides Walker, 1862. Adults, male and female genitalia of all species are illustrated.

RESUMO

Barsine Walker, 1854 is a large genus of lichen moths (family Erebidae, subfamily Arctiinae, tribe Lithosiini) including about a hundred of valid species and widespread in Oriental tropics, mainland China, Taiwan, Japan, Korea and the southern part of the Russian Far East. During the studies of Lithosiini materials recently collected in the Chinese Province of Guangdong, we found one more, yet undescribed Barsine species. It is related to the recently described B. cacharensis Singh Kirti, 2016 by the male and female genitalia structure, but externally it is more similar to B. defecta Walker, 1854 and B. gratissima (de Joannis, 1930). The species is described below as new.

RESUMO

The epicopeiid moth genus Schistomitra Butler, 1881 is reported outside Japan for the first time, with a new species, Schistomitra joelmineti Huang & Wang, sp. nov., described from the southern part of Shaanxi and Gansu Province in China. Photographs of adults and genitalia are provided, and the distribution pattern of the genus is discussed.

Coumarins are widely present in a variety of plants and have a variety of pharmacological activities. In this study, we isolated a coumarin compound from Microsorium fortunei (Moore) Ching; the compound was identified as esculetin by hydrogen and carbon spectroscopy. Its anti-hepatitis B virus (HBV) activity was investigated in vitro and in vivo. In the human hepatocellular liver carcinoma 2.2.15 cell line (HepG2.2.15) transfected with HBV, esculetin effecting inhibited the expression of the HBV antigens and HBV DNA in vitro. Esculetin inhibited the expression of Hepatitis B virus X (HBx) protein in a dose-dependent manner. In the ducklings infected with duck hepatitis B virus (DHBV), the levels of DHBV DNA, duck hepatitis B surface antigen (DHBsAg), duck hepatitis B e-antigen (DHBeAg), alanine aminotransferase (ALT) and aspartate aminotransferase (AST) decreased significantly after esculetin treatment. Summing up the above, the results suggest that esculetin efficiently inhibits HBV replication both in vitro and in vivo, which provides an opportunity for further development of esculetin as antiviral drug.

A highly flexible electrochemical assay based on target-triggered DNAzyme spiders was proposed for the detection of telomerase. The DNAzyme-telomerase substrate primers (D-TSP) containing Cu2+-dependent DNAzymes serve as recognition elements, and primers of telomerase. Telomerase extracted from Hela cells recognize the D-TSP and elongated with DNA sequence repeats. A synthetic telomerase product hybridized with scaffold sequences of two DNAzyme-tethered probes on the basis of the mechanism of the proximity-ligation assay. The three-leg DNAzyme spiders has been assembled and initiated the autonomous hybridization/nicking/displacement cycles on substrate modified surface. The cleaved ferrocene-labeled fragements are adsorbed on gold surface leading to an increase in the electrochemical signal. As a result, the one input target, telomerase, release large amount of ferrocene-labeled DNA strands, achieving an exponential signal amplification and an excellent improvement in sensitivity over single molecule or two-component 'sandwich' binding complexes. Our proposed biosensor showed a nonlinear dependence with Hela cell numbers, ranging from 25 to 2000 with a detection limit of 10 cells. Telomerase activities from different cell lines were also successfully evaluated. Our electrochemical strategy based on target-triggered DNAzyme spiders was enzyme-free, PCR-free, simple in operation which indicated that it expected to expand the scope of DNA nanotechnology in the areas of clinical diagnosis.
