RESUMO

Visible-light-induced site-selective C(sp3)-H pyridylation of amides has been accomplished using N-amidopyridinium salts. The N-centered radicals generated by the single-electron reduction of N-amidopyridinium substrates undergo 1,5-hydrogen atom transfer to form alkyl radical intermediates. Excellent C4-selectivity in radical trapping with pyridinium salts is observed for the alkyl radicals to provide δ-pyridyl sulfonamides and γ-pyridyl carboxamides. The utility is demonstrated by offering a practical approach for the late-stage functionalization of complex amide derivatives.

RESUMO

INTRODUCTION: This study is a prospective, assessor-blinded, parallel-group, randomized controlled pilot trial to explore the effectiveness of 12-week adjuvant moxibustion therapy for arthralgia in menopausal females at stage I to III breast cancer on aromatase inhibitor (AI) administration, compared with those receiving usual care. METHODS/DESIGN: Forty-six menopausal female patients with breast cancer who completed cancer therapy will be randomly allocated to either adjuvant moxibustion or usual care groups with a 1:1 allocation ratio. The intervention group will undergo 24 sessions of adjuvant moxibustion therapy with usual care for 12 weeks, whereas the control group will receive only usual care during the same period. The usual care consists of acetaminophen administration on demand and self-directed exercise education to manage AI-related joint pain. The primary outcome is the mean change of the worst pain level according to the Brief Pain Inventory-Short Form between the initial visit and the endpoint. The mean changes in depression, fatigue, and quality of life will also be compared between groups. Safety and pharmacoeconomic evaluations will also be included. DISCUSSION: Continuous variables will be compared by an independent t test or Wilcoxon rank-sum test between the adjuvant moxibustion and usual care groups. Adverse events will be analyzed using the chi-square or Fisher exact test. The statistical analysis will be performed by a 2-tailed test at a significance level of .05.

Three Apiaceae species Ledebouriella seseloides, Peucedanum japonicum, and Glehnia littoralis are used as Asian herbal medicines, with the confusingly similar common name "Bang-poong". We characterized the complete chloroplast (cp) genomes and 45S nuclear ribosomal DNA (45S nrDNA) sequences of two accessions for each species. The complete cp genomes of G. littoralis, L. seseloides, and P. japonicum were 147,467, 147,830, and 164,633 bp, respectively. Compared to the other species, the P. japonicum cp genome had a huge inverted repeat expansion and a segmental inversion. The 45S nrDNA cistron sequences of the three species were almost identical in size and structure. Despite the structural variation in the P. japonicum cp genome, phylogenetic analysis revealed that G. littoralis diverged 5-6 million years ago (Mya), while P. japonicum diverged from L. seseloides only 2-3 Mya. Abundant copy number variations including tandem repeats, insertion/deletions, and single nucleotide polymorphisms, were found at the interspecies level. Intraspecies-level polymorphism was also found for L. seseloides and G. littoralis. We developed nine PCR barcode markers to authenticate all three species. This study characterizes the genomic differences between L. seseloides, P. japonicum, and G. littoralis; provides a method of species identification; and sheds light on the evolutionary history of these three species.

Sensors and electronic devices based on semiconductors in their two-dimensional forms have many advantages. In this paper, we studied micro-Hall sensors based on two-dimensional molybdenum diselenide for the first time. The micro-Hall sensor based on a Ti/MoSe2/Ti structure clearly showed a linear dependence of the Hall voltage as a function of the magnetic field, with a magnetic sensitivity of ∼16 V/AT. The magnetic sensitivity was higher in the Au/MoSe2/Au structure, with a maximum value of ∼120 V/AT at a bias current of 100 mA; the minimum detectable magnetic field was found to be 1.45 µT/Hz1/2 at the same current value, making our new micro-Hall sensor a very good candidate for magnetic sensing applications.

Pancreatic metastasis is extremely rare, particularly from small-cell lung cancer (SCLC). Studies on the role of endoscopic ultrasound-guided fine needle aspiration (EUS-FNA) with liquid-based cytology (LBC) in the diagnosis of metastatic small-cell carcinoma in the pancreas have been rarely conducted. We report herein a case of pancreatic metastasis from SCLC diagnosed using EUS-FNA with LBC (ThinPrep). A 71-year-old man presented with chief complaints of hemoptysis and jaundice over the past 1 month. Lung & pancreas tumors with multiple liver nodules were detected on computed tomography. The aspirated material from the pancreas using EUS-FNA was prepared as a cytologic specimen with ThinPrep method, which revealed scattered and clustered "small blue cells" with scant cytoplasm and stippled chromatin with frequent apoptotic bodies. Immunocytochemical staining of the cellblock material revealed strong positivity for CD56 and thyroid transcription factor-1. Endobronchial biopsy for lung mass revealed nests of small, round, blue tumor cells with hyperchromatic nuclei showing salt and pepper chromatin, scant cytoplasm, and brisk mitotic activity. Therefore, a diagnosis of metastatic small-cell carcinoma to the pancreas with an extensive stage was finally made.

The characteristics of an estuary are determined by various factors as like as tide, wave, river discharge, etc. which also control the water quality of the estuary. Therefore, detecting the changes of characteristics is critical in managing the environmental qualities and pollution and so the locations of monitoring should be selected carefully. The present study proposes a framework to deploy the monitoring systems based on a graphical method of the spatial and temporal optimizations. With the well-validated numerical simulation results, the monitoring locations are determined to capture the changes of water qualities and pollutants depending on the variations of tide, current and freshwater discharge. The deployment strategy to find the appropriate monitoring locations is designed with the constrained optimization method, which finds solutions by constraining the objective function into the feasible regions. The objective and constrained functions are constructed with the interpolation technique such as objective analysis. Even with the smaller number of the monitoring locations, the present method performs well equivalently to the arbitrarily and evenly deployed monitoring system.

STATEMENT OF PROBLEM: Rapid prototyping, including stereolithography (SLA), is a more recent technique for fabricating metal frameworks than the conventional lost-wax technique. However, investigations of the marginal discrepancies and internal spacing of cobalt-chromium (Co-Cr) metal copings fabricated using SLA are lacking. PURPOSE: The purpose of this in vitro study was to evaluate the clinical acceptability of the marginal discrepancies and internal spacing of Co-Cr metal copings fabricated using the SLA technique. MATERIAL AND METHODS: A resin tooth of a maxillary right first premolar was prepared with a deep chamfer margin for a metal-ceramic crown. Titanium master dies were milled after scanning the prepared tooth (n=45). In conventional lost wax group (group LW), the conventional lost-wax technique was used to fabricate Co-Cr metal copings (n=15). In milling group (group MC), a computer-aided design (CAD) system was used to design the metal copings, which were milled from Co-Cr alloy (n=15). The CAD system was also used to design the metal copings in a 3D-printed group (group SL), and Co-Cr metal copings were cast from resin patterns fabricated using the SLA device (n=15). Marginal discrepancies and internal spaces were measured using an optical microscope at ×100 magnification at 11 reference points. The values were analyzed statistically with 1-way analysis of variance (α=.05). RESULTS: The mean (±SD) overall space was 63.2 ±16.6 µm for group LW, 70.2 ±15.5 µm for group SL, and 130.3 ±13.8 µm for group MC. The overall spaces differed significantly between group MC and the other 2 groups (P<.05). The marginal discrepancy and internal spaces were significantly larger in group MC than in groups LW and SL. (P<.05). Occlusal spaces differed significantly among the 3 study groups (P<.05). CONCLUSIONS: Co-Cr metal copings fabricated using an SLA technique showed clinically acceptable marginal discrepancies and internal spaces. These spaces did not differ significantly from those obtained with the conventional lost-wax technique.

Food-grade titanium dioxide (TiO2) is a common and widespread food additive in many processed foods, personal care products, and other industrial categories as it boosts the brightness and whiteness of colours. Although it is generally recognised as safe for humans, there is a growing interest in the health risks associated with its oral intake. This study quantified and identified TiO2 nanoparticles present in confectionery foods, which are children's favourite foods, with inductively coupled plasma optical emission spectrometry (ICP-OES) and transmission electron microscopy (TEM). A reliable digestion method using hot sulphuric acid and a digestion catalyst (K2SO4:CuSO4 = 9:1) was suggested for titanium analysis. Validations of the experimental method were quite acceptable in terms of linearity, recoveries, detection limits, and quantification limits. Of all the 88 analysed foods, TiO2 was detected in 19 products, all except three declared TiO2 in their labelling. The mean TiO2 content of candies, chewing gums, and chocolates were 0.36 mg g-1, 0.04 mg g-1, and 0.81 mg g-1, respectively. Whitish particles isolated from the confectionery foods were confirmed as TiO2 nanoparticles via TEM and energy dispersive X-ray spectroscopy (EDX), in which nanosized particles (<100 nm) were identified.

The hydrogen evolution reaction using semiconductor photocatalysts has been significantly improved by cocatalyst loading. However, there are still many speculations regarding the actual role of the cocatalyst. Now a photocatalytic hydrogen evolution reaction pathway is reported on a cocatalyst site using TiO2 nanosheets doped with Rh at Ti sites as one-atom cocatalysts. A hydride species adsorbed on the one-atom Rh dopant cocatalyst site was confirmed experimentally as the intermediate state for hydrogen evolution, which was consistent with the results of density functional theory (DFT) calculations. In this system, the role of the cocatalyst in photocatalytic hydrogen evolution is related to the withdrawal of photo-excited electrons and stabilization of the hydride intermediate species; the presence of oxygen vacancies induced by Rh facilitate the withdrawal of electrons and stabilization of the hydride.

Supercapacitors are promising energy storage devices due to their high power density, long cycling life, and short charging time. NiO is one of the alternative inorganic materials that could be used to replace the conventional RuO2 electrodes in these supercapacitors. In the present study, NiO thin film was prepared by radio frequency magnetron sputtering using a NiO alloy target. This process offeres several advantages such as the superior adhesion of the resulting thin films and the easy control of the deposition rate. Rapid thermal annealing (RTA) at different annealing temperatures was used to control the properties of the NiO thin films intended for supercapacitor applications. The lattice imperfections and interstitials/vacancies in the NiO thin films were influenced by the annealing temperature, and subsequently affected the bandgaps, optical transmittance, carrier concentration, and resistivity. Consequently, the the supercapacitive behavior was influenced by the surface area and the variation on the homogeneity of the crystallites in the NiO thin films with a change in the annealing temperature.

When mixtures of aggregates and water dredged from the seabed are discharged at the surface into the adjacent water from a barge, coarse sediments sink immediately and fine sediments are suspended forming a plume. Recently, elongated plumes of fine sediment were observed by satellites near a dredging location on the continental shelf. Such plume streaks were longer in certain conditions with seasonality than expected or reported previously. Therefore, the present work studied the appearance of sediment plume with field measurements and numerical simulations and explains the seasonally varying restoring force and thicknesses of the surface mixed layer resulting from the vertical density distribution near the surface, along with mixing by hydrodynamic process. The resulting mixtures, after vertical restoring and mixing with the surroundings, determine the horizontal transport of suspended sediments. A numerical model successfully reproduced and explained the results from field measurements and satellite images along with the seasonal variations.

Panax ginseng C. A. Meyer, reputed as the king of medicinal herbs, has slow growth, long generation time, low seed production and complicated genome structure that hamper its study. Here, we unveil the genomic architecture of tetraploid P. ginseng by de novo genome assembly, representing 2.98 Gbp with 59 352 annotated genes. Resequencing data indicated that diploid Panax species diverged in association with global warming in Southern Asia, and two North American species evolved via two intercontinental migrations. Two whole genome duplications (WGD) occurred in the family Araliaceae (including Panax) after divergence with the Apiaceae, the more recent one contributing to the ability of P. ginseng to overwinter, enabling it to spread broadly through the Northern Hemisphere. Functional and evolutionary analyses suggest that production of pharmacologically important dammarane-type ginsenosides originated in Panax and are produced largely in shoot tissues and transported to roots; that newly evolved P. ginseng fatty acid desaturases increase freezing tolerance; and that unprecedented retention of chlorophyll a/b binding protein genes enables efficient photosynthesis under low light. A genome-scale metabolic network provides a holistic view of Panax ginsenoside biosynthesis. This study provides valuable resources for improving medicinal values of ginseng either through genomics-assisted breeding or metabolic engineering.

BACKGROUND: The ginseng (Panax ginseng C.A. Meyer) is a perennial herbaceous plant that has been used in traditional oriental medicine for thousands of years. Ginsenosides, which have significant pharmacological effects on human health, are the foremost bioactive constituents in this plant. Having realized the importance of this plant to humans, an integrated omics resource becomes indispensable to facilitate genomic research, molecular breeding and pharmacological study of this herb. DESCRIPTION: The first draft genome sequences of P. ginseng cultivar "Chunpoong" were reported recently. Here, using the draft genome, transcriptome, and functional annotation datasets of P. ginseng, we have constructed the Ginseng Genome Database http://ginsengdb.snu.ac.kr /, the first open-access platform to provide comprehensive genomic resources of P. ginseng. The current version of this database provides the most up-to-date draft genome sequence (of approximately 3000 Mbp of scaffold sequences) along with the structural and functional annotations for 59,352 genes and digital expression of genes based on transcriptome data from different tissues, growth stages and treatments. In addition, tools for visualization and the genomic data from various analyses are provided. All data in the database were manually curated and integrated within a user-friendly query page. CONCLUSION: This database provides valuable resources for a range of research fields related to P. ginseng and other species belonging to the Apiales order as well as for plant research communities in general. Ginseng genome database can be accessed at http://ginsengdb.snu.ac.kr /.

Autoimmune pancreatitis (AIP) is a distinct subtype of pancreatitis, which is classified into type 1 and 2 based on the clinicopathological features. According to the international consensus diagnostic criteria, pancreas resection or core biopsy specimens are recommended to make an accurate histological evaluation. However, the usefulness of endoscopic ultrasonography (EUS) guided fine needle aspiration (FNA) for histological evaluation has also been reported. Furthermore, the simultaneous presentation of type 2 AIP and Crohn's disease (CD) is very rare, especially in the Asian population. Therefore, we herein report a case of type 2 AIP with CD, which was diagnosed using EUS guided FNA with a 22-gauge needle.

A mandibular molar with a thick buccal bone plate is a challenging problem in endodontic surgery despite the increase in the success rate of endodontic surgery nowadays. This report describes the application of a surgical template to guide osteotomy and facilitate apex localization in a mandibular molar with a thick buccal bone plate. A 57-year-old woman visited the authors' clinic for pain in tooth 19 and was diagnosed with symptomatic apical periodontitis in this previously treated tooth. Nonsurgical retreatment was performed; however, 2 years later, the patient reported pain in the same tooth. A periapical lesion was confirmed using cone-beam computed tomographic (CBCT) imaging, and endodontic surgery on the mesial root of tooth 19 was planned. After CBCT imaging and cast scan data were transferred to implant surgical planning software, the data were superimposed. In the superimposed model, an anchor pin was designed to target the mesial root apex of tooth 19. The surgical template was then printed using a 3-dimensional printer. Endodontic microsurgery included application of this printed surgical template. A computer-aided design/computer-aided manufacturing (CAD/CAM)-guided surgical template minimized the extent of osteotomy and enabled precise targeting of the apex in this case. There were no postoperative complications. A CAD/CAM-guided surgical template is useful in endodontic surgery for complicated cases.

This study investigated the effect of paid sick leave on workers' decisions to obtain vaccinations for the seasonal flu. Our vaccination decision model suggested that the marginal effect of paid sick leave depended on the reduced cost of obtaining a vaccination now and the expected income benefit from claiming paid sick leave after flu infection. Our hypothesis was that these effects vary according to workers' income levels. To confirm this hypothesis, we examined 11,702 participants in the National H1N1 Flu Survey (NHFS) conducted in late 2009 to early 2010 and measured the marginal effect using a Bayesian endogenous covariates regression model. The results of our estimation indicate that having paid sick leave did affect workers' vaccination decisions differently based on their income levels. Low-income workers were willing to be vaccinated because of the positive expected income benefit. High-income workers were willing to be vaccinated because the positive cost effect dominated the negative expected income benefit.

BACKGROUND: Expression of caveolin-1 (Cav-1) is frequently altered in many human cancers and both tumor suppression and promotion functions of Cav-1 have been suggested based on its expression status. However, it remains unanswered how Cav-1 provokes opposite effects in different cancers or different phases of tumor progression. METHODS: To explore the implication of Cav-1 alteration in gastric tumorigenesis, the expression and mutational status of Cav-1 and its effects on tumor cell growth were characterized. RESULTS: A substantial fraction of primary tumors and cell lines displayed abnormally low or high Cav-1 mRNA expression, indicating the bidirectional alteration of Cav-1 in gastric cancers. While allelic imbalance and mutational alterations of the Cav-1 gene were rarely detected, aberrant promoter hyper- or hypo-methylation showed a tight correlation with bidirectional alteration of its expression. Abnormally low and high Cav-1 expression was more frequently observed in early and advanced cancers, respectively, suggesting the oncogenic switch of its function in tumor progression. Cell cycle progression, DNA synthesis, and colony forming ability were markedly decreased by Cav-1 transfection in low-expressing tumor cells but by its depletion in high-expressing cells. Interestingly, Cav-1 exerted opposite effects on MEK-ERK signaling in these two cell types through the reciprocal regulation of the RAF-ERK negative feedback loop. A feedback inhibition of RAF by ERK was stimulated by restoration of Cav-1 expression in low-expressing cells but by it depletion in high-expressing cells. As predicted, the opposite effects of Cav-1 on both tumor cell growth and inhibitory RAF phosphorylation were abolished if ERK is depleted. CONCLUSION: Bidirectional alteration of Cav-1 is linked to its opposite effects on gastric tumor cell growth, which stem from the reciprocal control on the RAF-ERK negative feedback loop.

BACKGROUND: Panax ginseng Meyer (Asian ginseng) has a large nuclear genome size of > 3.5 Gbp in haploid genome equivalent of 24 chromosomes. Tandem repeats (TRs) occupy significant portions of the genome in many plants and are often found in specific genomic loci, making them a valuable molecular cytogenetic tool in discriminating chromosomes. In an effort to understand the P. ginseng genome structure, we characterized an ultrahigh copy 167-bp TR (Pg167TR) and explored its chromosomal distribution as well as its utility for chromosome identification. METHODS: Polymerase chain reaction amplicons of Pg167TR were labeled, along with 5S and 45S rDNA amplicons, using a direct nick-translation method. Direct fluorescence in situ hybridization (FISH) was used to analyze the chromosomal distribution of Pg167TR. RESULTS: Recently, we reported a method of karyotyping the 24 chromosome pairs of P. ginseng using rDNA and DAPI (4',6-diamidino-2-phenylindole) bands. Here, a unique distribution of Pg167TR in all 24 P. ginseng chromosomes was observed, allowing easy identification of individual homologous chromosomes. Additionally, direct labeling of 5S and 45S rDNA probes allowed the identification of two additional 5S rDNA loci not previously reported, enabling the refinement of the P. ginseng karyotype. CONCLUSION: Identification of individual P. ginseng chromosomes was achieved using Pg167TR-FISH. Chromosome identification is important in understanding the P. ginseng genome structure, and our method will be useful for future integration of genetic linkage maps and genome scaffold anchoring. Additionally, it is a good tool for comparative studies with related species in efforts to understand the evolution of P. ginseng.

Genome duplication and repeat multiplication contribute to genome evolution in plants. Our previous work identified a recent allotetraploidization event and five high-copy LTR retrotransposon (LTR-RT) families PgDel, PgTat, PgAthila, PgTork, and PgOryco in Panax ginseng. Here, using whole-genome sequences, we quantified major repeats in five Panax species and investigated their role in genome evolution. The diploids P. japonicus, P. vietnamensis, and P. notoginseng and the tetraploids P. ginseng and P. quinquefolius were analyzed alongside their relative Aralia elata. These species possess 0.8-4.9 Gb haploid genomes. The PgDel, PgTat, PgAthila, and PgTork LTR-RT superfamilies accounted for 39-52% of the Panax species genomes and 17% of the A. elata genome. PgDel included six subfamily members, each with a distinct genome distribution. In particular, the PgDel1 subfamily occupied 23-35% of the Panax genomes and accounted for much of their genome size variation. PgDel1 occupied 22.6% (0.8 Gb of 3.6 Gb) and 34.5% (1.7 Gb of 4.9 Gb) of the P. ginseng and P. quinquefolius genomes, respectively. Our findings indicate that the P. quinquefolius genome may have expanded due to rapid PgDel1 amplification over the last million years as a result of environmental adaptation following migration from Asia to North America.

Pancreatic adenocarcinoma may account for more than 80% of all pancreatic neoplasms. Occasionally, other rare tumors such as lymphoma, metastatic tumor, and solid pseudopapillary neoplasm can be considered in the differential diagnosis. We report the case of an 82-year-old man with a pancreatic solid mass. This case suggests that endoscopic ultrasound (EUS)-guided fine-needle aspiration (FNA) with biopsy, that is, EUS-FNA is recommended in the differential diagnosis of the pancreatic solid mass apart from pancreatic adenocarcinoma. In particular, the histologic core obtained by EUS-guided biopsy is helpful for the immunostaining of molecular markers to confirm the final diagnosis.
