RESUMO

To systematically review the efficacy and safety of Huoxiang Zhengqi Pills combined with Western medicine in the treatment of acute gastroenteritis. Four Chinese databases( CNKI,VIP,Wan Fang,Sino Med) and three English databases( Cochrane Library,Medline,EMbase) were systematically and comprehensively searched from the database establishment to April 2019 to collect the randomized controlled trials( RCTs) about the treatment of acute gastroenteritis with Huoxiang Zhengqi Pills combined with Western medicine. Two investigators independently performed literature screening,data extraction and bias risk assessment. Rev Man 5. 3 software was used for data analysis. A total of 316 articles were retrieved and finally 44 studies were included in this study,involving 4153 patients. The overall quality of the included studies was generally low. Meta-analysis results showed that in the total clinical effective rate,Huoxiang Zhengqi Pills combined with conventional treatment or norfloxacin tablets was superior to conventional treatment or norfloxacin tablets alone. In terms of the time for improving clinical symptoms,Huoxiang Zhengqi Pills combined with conventional treatment or norfloxacin tablets could better relieve fever than conventional treatment or norfloxacin tablets alone. In terms of incidence of adverse reactions,there was no statistical difference between Huoxiang Zhengqi Pills combined with conventional treatment and conventional treatment alone. Other outcome measures were affected by various factors( such as inclusion of only 1 study or excessive heterogeneity among studies) and could not be concluded. Due to the limitations of the quality and quantity of included studies,this conclusion still needs to be verified by more high quality researches.

As a primary pioneering region in China's ongoing urbanization process, the Yangtze River Economic Belt's (YREB's) urbanization process is itself continually accelerating, causing increasing pressure on the area's water ecosystem. It is necessary to examine the coordination relationship between the urbanization system and the water ecosystem in the YREB for realizing sustainable urban development. To this purpose, we use two comprehensive index systems, along with an improved coupling coordination degree (CCD) model. This method is used to analyze the coordination between urbanization and the water ecosystem across spatial gradients and temporal scales in the YREB, from 2008 to 2017. The factors acting as obstacles were diagnosed by utilizing the obstacle degree model. The results show that: (1) the coordination state of each region gradually improved during the 2008-2017 period. In terms of spatial distribution, the coordination state between two systems gradually increased from east to west. Moreover, the spatial differences across the 11 analyzed regions gradually narrowed with the passage of time. (2) The coordination between the two systems, from 2008 to 2017, evolved from a state of serious imbalance to a state of good coordination. The two systems passed from an initial period of imbalance or antagonism, coupled with rapid growth (2008-2011), through a period of basic coordination with steady growth (2011-2014), and finally toward a period of good coordination with slow growth (2014-2017). (3) Spatial urbanization and pressures on subsystems are the key factors acting as obstacles in the urbanization system and water ecosystem, respectively. Facing the process of rapid urbanization in China, the coupling analysis of the coordination between urbanization and the water ecosystem can help the government to formulate a reasonable new-type urban development strategy. This strategy will play an important role in China's sustainable urban development and water environmental protection. The findings of this study provide important support for urban planning in the future.

RESUMO

An effective and simple solvent-controlled synthesis of thiocyanated enaminones and 2-aminothiazoles has been demonstrated from enaminones, potassium thiocyanate, and N-bromosuccinimide. This process features mild reaction conditions, simple and easy operation, short reaction time, and high yield and chemoselectivity and thereby provides an efficient protocol for the divergent synthesis of thiocyanated enaminones and substituted 2-aminothiazoles controlled by simply varying the solvent. All reaction components are commercially available or easily accessible at low cost. The potential utility of these methods in organic chemistry and medicinal chemistry applications is highlighted.

Osteoarthritis (OA) is a common joint degenerative disease that causes pain, joint damage, and dysfunction. External hyaluronic acid (HA) supplement is a common method for the management of osteoarthritis which requires multi-injections. It is demonstrated that biodegradable mesoporous silica nanoparticles successfully deliver an enzyme, hyaluronan synthase type 2 (HAS2), into synoviocytes from the temporomandibular joint (TMJ) and generate endogenous HA with high molecular weights. In a rat TMJ osteoarthritis inflammation model, this strategy promotes endogenous HA production and inhibits the synovial inflammation of OA for more than 3 weeks with one-shot administration. Such nanotherapy also helps repairing the bone defects in a rat OA bone defect model.

OBJECTIVES: This study aims to investigate the role of Cannabinoid receptor 2 (CB2) on osteogenesis of bone marrow-derived mesenchymal stem cells (BMSCs) under hypoxia. MATERIALS AND METHODS: BMSCs were isolated from Sprague-Dawley rats and cultured in the presence of cobalt chloride (CoCl2) to induce intracellular hypoxia. Cell proliferation was measured with MTT assay. Quantitative real-time PCR and western blot were applied to evaluate the mRNA and protein expressions of CB2 and osteogenic indicators including osteocalcin, RUNX2, collagen-1 and osterix (SP7). The osteogenic differentiation of BMSCs was further examined by ALP assay and alizarin red S (ARS) staining. Moreover, the activation of MAPKs signaling pathways was analyzed by western blot. RESULTS: CoCl2 dose-dependently increased hypoxia inducible factor while higher concentrations (200 and 400 µM) of CoCl2 markedly inhibited cell proliferation. CoCl2 induced hypoxia significantly increased the protein and mRNA expressions of osteocalcin, RUNX2, collagen-1 and osterix, along with enhanced ALP and ARS staining. Interestingly, such effects can be inhibited by the addition of CB2 inhibitor AM630. Moreover, AM630 partially inhibited hypoxia-induced p38 and ERK pathways, which may lead to a decrease in the osteogenic transcripts of RUNX2, collagen-1 and osterix. CONCLUSIONS: CoCl2 induced hypoxia could promote osteogenesis of rat BMSCs possibly through CB2.

Aim: To establish a web server that can mutually validate prognostic biomarkers of cervical cancer. Methods: Four datasets including expression profiling and relative clinical follow-up data were collected from Gene Expression Omnibus and The Cancer Genome Atlas. The web server was developed by R software. Results: The web server was named OScc including 690 patients and can be accessed at http://bioinfo.henu.edu.cn/CESC/CESCList.jsp. The Kaplan-Meier survival curves with log-rank p-value and hazard ratio will be generated of interested gene in OScc. Compared with previous predictive tools, OScc had the advantages of registration-free, larger sample size and subgroup analysis. Conclusion: The OScc is highly valuable to perform the preliminary assessment and validation of new or interested prognostic biomarkers for cervical cancer.

Background: Aging has often been linked to age-related vascular disorders. The elucidation of the putative genes and pathways underlying vascular aging likely provides useful insights into vascular diseases at advanced ages. Transcriptional regulatory network analysis is the key to describing genetic interactions between molecular regulators and their target gene transcriptionally changed during vascular aging.Results: A total of 469 differentially expressed genes were parsed into 6 modules. Among the incorporated sample traits, the most significant module related to vascular aging was associated with triglyceride and enriched with biological terms like proteolysis, blood circulation, and circulatory system process. The module associated with triglyceride was preserved in an independent microarray dataset, indicating the robustness of the identified vascular aging-related subnetwork. Additionally, Enpp5, Fez1, Kif1a, F3, H2-Q7, and their interacting miRNAs mmu-miR-449a, mmu-miR-449c, mmu-miR-34c, mmu-miR-34b-5p, mmu-miR-15a, and mmu-let-7, exhibited the most connectivity with external lipid-related traits. Transcriptional alterations of the hub genes Enpp5, Fez1, Kif1a, and F3, and the interacting microRNAs mmu-miR-34c, mmu-miR-34b-5p, mmu-let-7, mmu-miR-449a, and mmu-miR-449c were confirmed.Conclusion: Our findings demonstrate that triglyceride and free fatty acid-related genes are key regulators of age-related vascular dysfunction in mice and show that the hub genes for Enpp5, Fez1, Kif1a, and F3 as well as their interacting miRNAs mmu-miR-34c, mmu-miR-34b-5p, mmu-let-7, mmu-miR-449a, and mmu-miR-449c, could serve as potential biomarkers in vascular aging.Methods: The microarray gene expression profiles of aorta samples from 6-month old mice (n=6) and 20-month old mice (n=6) were processed to identify nominal differentially expressed genes. These nominal differentially expressed genes were subjected to a weighted gene co-expression network analysis. A network-driven integrative analysis with microRNAs and transcription factors was performed to define significant modules and underlying regulatory pathways associated with vascular aging, and module preservation test was conducted to validate the age-related modules based on an independent microarray gene expression dataset in mice aorta samples including three 32-week old wild-type mice (around 6-month old) and three 78-week old wild-type mice (around 20-month old). Gene ontology and protein-protein interaction analyses were conducted to determine the hub genes as potential biomarkers in the progress of vascular aging. The hub genes were further validated with quantitative real-time polymerase chain reaction in aorta samples from 20 young (6-month old) mice and 20 old (20-month old) mice.

Salidroside (Sal), the active ingredient of Rhodiola rosea L, has various pharmacological activities, including antioxidant, anti-inflammatory and anti-tumor activities. Recently, studies have shown that oxidative stress and apoptosis are related to the pathogenesis of inflammatory bowel disease. Therefore, we evaluated the effects of Sal on oxidative stress and apoptosis in colitis mice through the SIRT1/FoxOs pathway. To induce the colitis model, mice continuously consumed water containing 3% DSS for 7 days; some mice were also treated with Sal and the SIRT1/FoxOs pathway blocker selisistat (Ex527). Changes in body weight, DAI, colon length and colon tissue histology as well as SOD, GSH-Px and CAT activities were measured. The expression of SIRT1, FoxO1, FoxO3a, FoxO4, caspase-3, cleaved-caspase-3, Bax and Bcl-2 in colorectal tissues was detected by RT-PCR and Western blotting. The study showed that Sal decreased the DAI score, weight loss, colon shortening and colon tissue damage in colitis mice. Sal inhibited oxidative stress by upregulating SOD, GSH-Px and CAT while suppressing colonic apoptosis by downregulating the expression of Bax, caspase-3, and cleaved-caspase-3 and upregulating the expression of Bcl-2. Sal also activated SIRT1/FoxOs signaling, which increased the expression of SIRT1, FoxO1, FoxO3a and FoxO4 in colon tissue. Furthermore, SIRT1/FoxOs pathway inhibition using Ex527 partially eliminated the effect of Sal on colitis mice. The study manifested that Sal may protect colitis mice by activating the SIRT1/FoxOs pathway, which is related to oxidative stress and apoptosis in colon tissues.

STUDY DESIGN: A meta-analysis OBJECTIVE.: To investigate whether robot-assisted techniques are superior to conventional techniques in terms of the accuracy of pedicle screw placement and clinical indexes. SUMMARY OF BACKGROUND DATA: Robot-assisted techniques are increasingly applied to spine surgery to reduce the rate of screw misplacement. However, controversy about the superiority of robot-assisted techniques over conventional freehand techniques remains. METHODS: We conducted a comprehensive search of PubMed, EMBASE, and Cochrane Library for potentially eligible articles. The outcomes were evaluated in terms of risk ratio (RR) or standardized mean difference (SMD) and the associated 95% confidence intervals (CIs). Meta-analysis was performed using the RevMan 5.3 software and subgroup analyses were performed based on the robot type for the accuracy of pedicle screw placement. RESULTS: Nine randomized controlled trials with 696 patients were included in this meta-analysis. The results demonstrated that the robot-assisted technique was more accurate in pedicle screw placement than the freehand technique. Subgroup analyses showed that the TINAVI robot-assisted technique was more accurate in screw positions Grade A (RR, 1.10; 95% CI, 1.06-1.14), Grade B (RR, 0.46; 95% CI, 0.28-0.75), and Grades C + D + E (RR, 0.21; 95% CI, 0.09-0.45) than the freehand technique, whereas the Renaissance robot-assisted technique showed the same accuracy as the freehand technique in screw positions Grade A, Grade B, and Grades C + D + E. Furthermore, the robot-assisted techniques showed equivalent postoperative stay, visual analogue scale scores, and Oswestry disability index scores to those of the freehand technique and shorter intraoperative radiation exposure time, fewer radiation dose and proximal facet violations but longer surgical time than the freehand technique. CONCLUSION: The robot-assisted technique is more accurate in pedicle screw placement than the freehand technique. And TINAVI robot-assisted pedicle screw placement is a more accurate alternative to conventional techniques and the Renaissance robot-assisted procedure. LEVEL OF EVIDENCE: 1.

OBJECTIVE: To study the long-term efficacy and safety of CD19 chimeric antigen receptor T cells (CAR-T) in the treatment of relapsed patients with B-cell acute lymphoblastic leukemia (ALL) after allogeneic hematopoietic stem cell transplantation (allo-HSCT). METHODS: A total of 7 patients with B-cell ALL relapsed after allo-HSCT were treated with CD19 CAR-T cells from September 2015 to March 2018. Among them, 6 had hematological recurrence and 1 had positive of MRD. They all were treated with a single infusion of CAR-T cells. FC chemotherapy regimen was administered before transfusion. The median number of CAR-T cells transfused was 6.0 (range 4.0-8.6) )×106/kg. Long-term efficacy and toxicity were evaluated. RESULTS: Bone marrow examination performed at d 30 after CAR-T infusion showed that all 7 patients achieved complete remission and MRD negative, grade I CRS for 1 case and grade II CRS for 6 cases, two of them had mild neurotoxicity, which was controlled by treatment. Two patients presented grade VI intestinal GVHD after CAR-T infusion. The median follow-up time was 18 months (range 12-42). Follow-up showed that two patients relapsed at 9 months and 14 months after treatment, out of 2 patients one died of progressive disease and the other reachived the hematological remission, but MRD was positive after CD22 CAR-T cell therapy. At present, five patients are disease-free survival, moreover showed complete donor chimerism. One year after CAR-T cell therapy, the results of immune reconstitution showed that CD4 level was more than 300×106/L in 5 patients who disease-free survived. Among them, 3 patients had poor recovery of immunoglobulin and received gamma globulin replacement therapy. CONCLUSION: All patients are followed up for at least one year. The preliminary efficacy and safety are satisfactory. CAR-T cell infusion is an effective method for the treatment of B-ALL recurrence after allo-HSCT.

BACKGROUND: The incidence of diabetes mellitus (DM) is increasing year by year, and various complications can endanger the lives of patients. Type 2 diabetes mellitus (T2DM) accounts for more than 90% of DM, most of which is associated with insulin resistance (IR), and IR has been shown to be closely related to the onset of T2DM and the presence of DM complications. Berberine (BBR) has been shown to improve T2DM with IR in a number of ways. In this study, we aim to evaluate the efficacy and safety of BBR in the treatment of T2DM with IR to provide the newest evidence for clinical use. METHODS AND ANALYSIS: Literature research will be divided into 2 parts: electronic search and manual search. We will search PubMed, EMBASE, The Cochrane Library, the China National Knowledge Infrastructure, China Biology Medicine disc, the China Science and Technology Journal database, and the Wanfang database online. We will select the eligible studies published up to June 30, 2019. Dissertations, conference papers, ongoing trials, internal reports, etc., are searched by manual search methods. We use Homeostatic Model Assessment for IR (HOMA-IR) as the primary outcome of T2DM with IR, and we will also focus on the patient's blood glucose levels and all adverse reactions that occur during medication.Two reviewers will read the articles, extract the data information, and assess the risk of bias independently. Data analysis will use the software such as RevMan 5.3.5, ENDNOTE X7, and STATA 13.0. RESULTS: This study will provide a high-quality synthesis of current evidence of BBR for T2DM with IR from several aspects including HOMA-IR, blood glucose levels, and adverse events. CONCLUSION: This systematic review will provide evidence to assess the efficacy and safety of BBR in the treatment of T2DM with IR. ETHICS AND DISSEMINATION: Because all of the data used in this systematic review has been published, ethical approval is not required. TRIAL REGISTRATION NUMBER: PROSPERO CRD42019123225.

In this study, the morphology, ultrastructure, nutrient removal, metabolite levels, and interaction of an algal-bacterial consortium exposed to different concentrations of carboxylic single-walled carbon nanotubes (C-SWCNT) were investigated. At a C-SWCNT concentration of 0.05 mg·L-1, the removal rates of TN, NH3-N, PO43--P, and COD were 94.7%, 94.8%, 86.4% and 84.3%, respectively. When cells were exposed to 50 mg·L-1 C-SWCNT, its intracellular levels in individual algae and the algal-bacterial consortium were 23.6 µg·g-1 and 12.1 µg·g-1, respectively. C-SWCNT (0.05 mg·L-1) promoted the metabolism of fatty acids, amino acids, small molecules, and acid in the algal-bacterial consortium. The main response to the interaction of C-SWCNT and the consortium was the change in extracellular carbohydrate levels. C-SWCNT also increased chlorophyll a and glycine levels. These findings reveal new insights into our understanding of the biological responses and interactions between C-SWCNT and algal-bacterial consortium.

Two-dimensional carbon-based nanomaterials have demonstrated great promise as electrode materials for electrochemical energy storage. However, there is a trade-off relationship between energy storage and rate capability for carbon-based energy storage devices because of the incrementing ion diffusion limitations, especially for thick electrodes with high mass loading. Herein, we report the cross-linked microporous carbon nanosheets enabling high-energy and high-rate supercapacitors. The as-fabricated microporous carbon nanosheets exhibit an extraordinary thickness-independent electrochemical performance. With the thickness of 15 µm, the as-fabricated carbon nanosheet electrode possesses areal/volumetric/gravimetric capacitance of 895 mF cm-2/596 F cm-3/358 F g-1. Even at a high electrode thickness of 125 µm, the as-fabricated thick electrode presents an ultrahigh areal/volumetric/gravimetric capacitance of 4102 mF cm-2/328 F cm-3/328 F g-1. Furthermore, the as-assembled symmetric supercapacitor delivers an outstanding energy density of 19.2 W h kg-1 at a power density of 135 W kg-1 and ultralong cycling stability (capacitance retention of 95% after 180 000 charge/discharge cycles) in an alkaline electrolyte. This work not only provides a facile method for low-cost preparation of carbon nanostructures and high-value utilization of biomass wastes but also offers new insights into rational design and fabrication of advanced electrode materials for high-performance electrochemical energy storage.

BACKGROUND: Sleep-disordered breathing may precipitate angina pectoris (AP) in rapid eye movement (REM) sleep. The purpose of this study was to investigate whether REM sleep is associated with the incidence of AP in different categories of apnea-hypopnea index (AHI). METHODS: A total of 2710 participants from Sleep Heart Health Study (59.3% women; mean age 61.7 years), who had 6-8 h of sleep; experienced an almost 11-year follow-up period. Complete polysomnography data and medical records were available for all participants. Percentage of REM sleep time (REM%) was classified into grade 1 (<20%), grade 2 (20-25%) and grade 3 (>25%). Logistic regression analysis was used to characterize the independent association between REM sleep and prevalent AP in this multi-center community-based cohort study. RESULTS: Patients with REM% of grade 3 had a higher incidence of AP than those with grade 1 and grade 2 (50.3% vs 42.4% vs 42.1%, respectively; p = 0.003). After adjusting for age, gender, race, AHI, marital status, education level, body mass index, sleep duration, arousal index, diabetes, hypertension and smoking status, multivariate logistic regression analysis revealed that REM% >25% was independently associated with the occurrence of AP (odds ratio = 1.500; 95% confidence interval = 1.188-1.894; p = 0.001). CONCLUSIONS: The percentage of REM sleep time may affect the incidence of AP irrespective of AHI level. Therefore, the relationship between sleep structure and cardiovascular disease merits further exploration.

Thrombotic diseases are major cause of cardiovascular diseases. This study was designed to investigate the effect of tachyplesin I on platelet aggregation and thrombosis. Platelet aggregation was analysed with a whole blood aggregometer. The mice were employed to investigate the effect of tachyplesin I on thrombosis in vivo. Tachyplesin I inhibited thrombin-induced platelet aggregation in a dose-dependent manner. Furthermore, tachyplesin I significantly reduced thrombosis in carrageenan-induced tail thrombosis model by intraperitoneal injection (0.1, 0.2 or 0.4 mg/kg) or intragastric administration (15, 30 or 60 mg/kg). Tachyplesin I also prolonged the bleeding time (BT) and clotting time (CT). The results revealed that tachyplesin I inhibited platelet aggregation and thrombosis by interfering the PI3K/AKT pathway. Tachyplesin I did not show significantly toxicity to mice under 300 mg/kg via intravenous injection. The results show that tachyplesin I inhibits thrombosis and has low toxicity. It is suggested that tachyplesin I has the potential to develop a new anti-thrombotic drug.

The rapid development of the economy in China resulted in increasingly serious water pollution problem. A lot of water pollution treatment projects have been launched to improve the water environment quality. Water pollution treatment is a complex and long-term task. Considering the concept of water pollution with fuzziness and the factors affecting the effect of water pollution treatment (EWPT), this study constructs a multi-dimensional dynamic fuzzy comprehensive monitoring model. The model considers the vague boundaries in the representation of water pollution and various factors affecting the treatment effect, such as monitoring time, monitoring index, and monitoring location. In detail, firstly, existing methods for evaluating the EWPT are analyzed and reviewed. Then a multi-dimensional dynamic model is developed for monitoring the EWPT. Finally, the Yueya Lake of Henan Province in China is taken as an example to demonstrate the effectiveness and practicability of the proposed method. From the analysis of the results, to maintain the cleanliness of the water, efforts should still be made to eliminate and completely block the pollutants on the shore in order to fundamentally solve the problem.

BACKGROUND: The effects of oblique lateral interbody fusion (OLIF) and minimally invasive transforaminal lumbar interbody fusion (MI-TLIF) have not been compared by a meta-analysis. The present study aimed to summarize the radiographic and clinical outcomes of OLIF and MI-TLIF for degenerative lumbar disease. METHODS: We performed a systematic review of related studies and report the outcomes of OLIF and MI-TLIF for degenerative lumbar disease. The radiographic outcomes measures included disc height, segmental lordotic angle, lumbar lordotic angle, and fusion. The clinical and functional outcomes included operative blood loss, operative time, length of hospital stay, complications, visual analog scale, and Oswestry disability index. Data pooling and meta-analysis with the random effects model were performed to evaluate the results. RESULTS: A total of 47 studies met the inclusion criteria. Similar changes in terms of disc height, segmental lordotic angle, lumbar lordotic angle, length of hospital stay, visual analog scale, Oswestry disability index, and radiological evidence of fusion of >90% were observed between the 2 groups. The OLIF group showed less operative blood loss and operative time. The incidence of intraoperative and postoperative complications was 9.5% and 19.9% for the OLIF group and 3.5% and 8.5% for the MI-TLIF group, respectively. CONCLUSIONS: The radiographic and functional outcomes and length of hospital stay were similar between the 2 groups. The OLIF group showed advantages in operative blood loss and operative time; however, the incidence of complications in this technique was greater than that in the MI-TLIF group.

BACKGROUND: Cortical bone trajectory (CBT) has been well-known in spine surgery for obtaining improved fixation while minimizing soft tissue dissection. This study was designed to compare the bone mineral density (BMD) between the CBT and traditional trajectory (TT) by using Hounsfield unit (HU) values and identify the ideal decades of patients and the suitable lumbar segments using this CBT technology from a radiological standpoint. METHODS: Patients were selected randomly from an institutional database based on age (evenly distributed by a decade of life) and gender. A total of 240 healthy patients had a computed tomography (CT) scan of the chest, abdomen, and pelvis. For each patient, axial slices of every vertebra were cut in two planes: one horizontal to the pedicle representing the plane wherein pedicle screws were inserted using the TT and the other in a caudocranial plane representing the plane wherein pedicle screws were inserted using the CBT. For each trajectory, a region of interest (ROI) was selected within the area wherein the screws were inserted. A CT number (HU values) was then calculated within each ROI to represent bone density. RESULTS: HU values measured at the ROI of CBT were significantly greater than those of the traditional pedicle screw in all age groups, and the specific value (ratio of the HU values of CBT/the HU values of TT) between CBT and TT was 1.92. A significant difference was observed between male and female. The HU values of CBT and TT of males were generally higher than those of females (males: CBT/TT 1.89 ± 0.45; Females: CBT/TT 1.95 ± 0.47). The specific value in HU values significantly increased with increasing age (p = 0.000) and cauda lumbar level (p = 0.000) in males and females. CONCLUSION: BMD, as measured by HU values for the ROI of the CBT screw, was significantly greater than that of the traditional pedicle screw, especially in old patients and cauda lumbar segments.
