RESUMO

Cadmium (Cd) is a major heavy metal toxicant found in industrial zones. Humans and animals are exposed to it through their diet, which results in various physiological problems. In the current study, the toxic effects of Cd on the liver were investigated by whole-transcriptome sequencing (RNA-seq) of the livers of Xiangxi heifers fed a diet with excess Cd. We randomly divided six healthy heifers into two groups. The first group received a control diet, whereas the second group received Cd-exceeding diets for 100 days. After 100 days, the livers were collected. A total of 551 differentially expressed mRNAs, 24 differentially expressed miRNAs, and 169 differentially expressed lncRNAs were identified (p < 0.05, |log2FC| >1). Differentially expressed genes (DEGs) were analyzed by gene ontology and Kyoto Encyclopedia of Genes and Genomes enrichment analyses. We found that under Cd exposure, DEGs were enriched in the adenosine 5'-monophosphate-activated protein kinase pathway, which is involved in autophagy regulation, and the peroxisome proliferator-activated receptor pathway, which is involved in lipid metabolism. In addition, the apolipoprotein A4 gene, which has anti-inflammatory and antioxidant effects, the anti-apoptotic gene ATPase H+/K+ transporting the nongastric alpha2 subunit, and the cholesterol metabolism-associated gene endothelial lipase gene were significantly downregulated. C-X-C motif chemokine ligand 3, cholesterol 7α-hydroxylase, and stearoyl-CoA desaturase, which are involved in the development of fatty liver, were significantly upregulated. These genes revealed the main effects of Cd on the liver of Xiangxi yellow heifers. The current study provides insightful information regarding the DEGs involved in autophagy regulation, apoptosis, lipid metabolism, anti-inflammation, and antioxidant enzyme activity. These may serve as useful biomarkers for predicting and treating Cd-related diseases in the future.

The purpose of the present study was to investigate the expression of α-SMA and SM22α in airway smooth muscle (ASM) of bronchioles from children younger than 14 years who died of acute interstitial pneumonia (AIP). This is based upon the hypothesis that as contractile marker proteins α-SMA and SM22α can serve as an index of the overcontractile phenotype of ASM that is seen in AIP. Lung tissue samples of children were obtained from autopsies and divided into the AIP group (55.9% male and 44.1% female, between 0.4 and 132 months old, n = 34) and the control group (60% male and 40% female, between 2 and 156 months old, n = 10). We recorded the post-mortem interval (PMI), height, clinical symptoms and abdominal fat thickness (AFT) of each case. Haematoxylin-and-eosin-stained sections were used to examine the luminal area and observe the morphological changes in the bronchioles. Immunohistochemistry and Masson's trichrome staining were used to detect the expression of contractile marker proteins and the degree of pulmonary fibrosis respectively. Compared with the control group, the luminal areas of bronchioles in the AIP group were smaller (p < .001). The expression differences in α-SMA and SM22α between the two groups were statistically significant (p = .01 and p = .02 respectively). Also, there was no significant correlation of the contractile marker proteins expression with PMI, height, clinical symptoms and AFT. The collagen deposition difference in lung between the two groups was not statistically significant (p = .224). These findings suggest that enhancement of ASM contractile function appears to be involved in the death mechanism of children with AIP, which affords more insights into the understanding of AIP.

While partial nephrectomy (PN) is generally preferred for localized renal cell carcinoma (RCC), radical nephrectomy (RN) is occasionally required. A new-baseline glomerular filtration rate (NBGFR) >45 ml/min/1.73 m2 after kidney cancer surgery is associated with strong survival outcomes. If NBGFR after RN will be above this threshold and the tumor has increased oncologic potential, RN may be a relevant consideration. Predicting NBGFR, defined as the GFR at 3-12 mo after RN, has been challenging owing to omission of two important parameters: split renal function (SRF) and renal function compensation (RFC). Our objective was to evaluate a simple SRF-based model in comparison to five published non-SRF-based models using data from a retrospective cohort of 445 RN patients. SRF was obtained via readily available semiautomated software (FUJIFILM Medical Systems) that provides differential parenchymal volume analysis on the basis of preoperative imaging. Our conceptually simple and clinically implementable SRF-based model more accurately predicts NBGFR after RN than five published non-SRF-based models (all p < 0.01). The SRF-based model also improved prediction of the clinically relevant threshold of NBGFR >45 ml/min/1.73 m2 (all p < 0.05). Patient summary: We validated a novel approach for more accurate prediction of kidney function after removal of one kidney. Our approach can be used in clinical and practice and will help in making decisions on full or partial removal of a kidney for kidney cancer.

Triptolide (TP), a compound isolated from a Chinese medicinal herb, possesses potent anti-tumor, immunosuppressive, and anti-inflammatory properties, but was clinically limited due to its poor solubility, bioavailability, and toxicity. Considering the environment-friendly, low-cost mechanochemical techniques and potential dissolution enhancement ability of Na2GA, an amorphous solid dispersion (Na2GA&TP-BM) consisting of TP and Na2GA were well-prepared to address these issues. The performance of Na2GA&TP-BM was improved through ball milling, such as from crystalline state to an amorphous solid dispersion, suitable nano micelle size and surface potential, and increased solubility. This change had a significant improvement of pharmacokinetic behavior in mice and could be able to extend the blood circulation time of the antitumor drug. Moreover, in vitro and in vivo anti-tumor study showed that Na2GA&TP-BM displayed more potent cytotoxicity to tumor cells. The work illustrated an environment-friendly and safe preparation of the TP formulation, which was promising to enhance the oral bioavailability and antitumor ability of TP, might be considered for efficient anticancer therapy.

INTRODUCTION: A number of case and case series descriptions established an association between Legionella infection and hyperCKemia, but the frequency and severity of hyperCKemia in Legionella infection remain unknown. This study aims to investigate the incidence, extent, and consequences of hyperCKemia in a large group of patients with Legionella infection. METHODS: This retrospective study included patients with confirmed Legionella infection during a ten-year period who received creatine kinase (CK) testing. Comparisons were performed between groups of Legionella patients with and without hyperCKemia. RESULTS: A total of 267 patients were included. HyperCKemia was present in 144 (53.9%) patients. The mean peak CK value was 9598 IU/L (range: 226 to 462,000 IU/L) while peak CK exceeded 1000 IU/L in 82 (56.9%) patients and 5000 IU/L in 33 (22.9%). When compared to patients without hyperCKemia, patients with hyperCKemia had higher incidences of neurologic symptoms (p = 0.036), acute renal failure (p = 0.028), dialysis requirement (p = 0.009), and need for ICU care (p = 0.016). HyperCKemia resolved in most Legionella patients by 7 days from CK peaking. DISCUSSION: Legionella infection requiring hospitalization appears to be associated with increased incidence of hyperCKemia and rhabdomyolysis. Greater awareness of the high incidence and the possible severity of hyperCKemia is needed when treating patients with Legionella infection.

Objective: To investigate the correlation between intravoxel incoherent motion diffusion-weighted imaging (IVIM-DWI) and the pathological classification of idiopathic orbital inflammatory pseudotumors (IOIPs). Methods: Nineteen patients who were diagnosed with IOIPs (a total of 24 affected eyes) between November 2018 and December 2020 were included in the study. All the patients underwent magnetic resonance imaging orbital plain scans and IVIM-DWI multiparameter scans before an operation. The true diffusion coefficient (D), pseudodiffusion coefficient (D*), and perfusion fraction (f) values were obtained. Based on histopathology, the lesions were divided into three types: lymphocytic infiltration, fibrosclerotic, and mixed. The correlation between IVIM-DWI parameters and pathological classification was tested with the histopathological results as the gold standard. The data were analyzed using SPSS version 17.0, with P < 0.05 defined as significant. Results: Among the 19 patients (24 eyes) affected by IOIP, there were no significant differences between IOIP pathological classification and gender or age (P > 0.05). There were statistically significant differences between the D and f values for different pathological types of IOIP and IVIM parameters (P < 0.05), and there was no significant difference in D* value between the different pathological types (P > 0.05). Conclusion: The D and f values showed correlation with different types of IOIP, and the sensitivity of the D value was higher than that of the f value. The D* value showed no significant distinction between pathological types of IOIP.

A variety of inbred mouse strains have been used for research in metabolic disorders. Despite being inbred, they display large inter-individual variability for many traits like food intake and body weight. However, the relationship between dietary macronutrients and inter-individual variation in body weight and food intake of different mouse strains is still unclear. We investigated the association between macronutrient content of the diet and variations in food intake, body composition, and glucose tolerance by exposing five different mouse strains (C57BL/6, BALB/c, C3H, DBA/2, and FVB) to 24 different diets with variable protein, fat, and carbohydrate contents. We found only increasing dietary fat, but not protein or carbohydrate had a significant association (positive) with variation in both food intake and body weight. The highest variation in both body weight and food intake occurred with 50% dietary fat. However, there were no significant relationships between the variation in fat and lean mass with dietary protein, fat, or carbohydrate levels. In addition, none of the dietary macronutrients had significant impacts on the variation in glucose tolerance ability in C57BL/6 mice. In conclusion, the variations in food intake and body weight changes increased with the elevation of dietary fat levels.

Aqueous zinc (Zn) metal batteries have been regarded as the most promising aqueous batteries due to their low redox potential, high theoretical capacity, and abundant Zn resources. Unfortunately, Zn dendrite growth and serious side reactions drastically curtail the cycle life, severely affecting their large-scale application. Herein, a multifunctional ordered Zn-aminotrimethylene phosphonic acid (Zn-ATMP) film is in situ modified on the surface of metal Zn via a facile etching process. The modified layer can not only retard the side reactions and suppress the corrosion rate, but also lower the Zn nucleation overpotential and accelerate diffusion and homogenize deposition of Zn2+ due to the strong Zn affinity. Consequently, the as-prepared Zn-ATMP@Zn anode in the symmetric cell enables long-term lifespan (over 1000 h) at 10.0 mA cm-2 with a high areal capacity of 5 mAh cm-2 . Furthermore, when assembled with a SeS2 -based cathode, a long lifespan for over 280 cycles at 2 C can be achieved for the aqueous Zn-SeS2 battery. This work provides a reliable strategy for constructing stabilized Zn anode and accelerating the development of an aqueous energy storage system.

The microenvironment in the stomach is different from other digestive tracts, mainly because of the secretion of gastric acid and digestive enzymes, bile reflux, special mucus barrier, gastric peristalsis, and so on, which all contribute to the formation of antibacterial environment. Microecological disorders can lead to gastric immune disorders or lead to the decrease of dominant bacteria and the increase of the abundance and virulence of pathogenic microorganisms and then promote the occurrence of diseases. The body performs its immune function through innate and adaptive immunity and maintains microbial balance through the mechanism of immune homeostasis. Microecological imbalance can lead to the invasion of pathogenic microorganisms and damage mucosal barrier and immune system. The coexistence of gastric microorganisms (including viruses and fungi) may play a synergistic or antagonistic role in the pathogenesis of gastric diseases. Probiotics have the ability to compete with intestinal pathogens, increase the secretion of immunoglobulin A (IgA), stimulate the production of mucin, bacteriocin, and lactic acid, regulate the expression and secretion of cytokines, and regulate the growth of microbiota, which all have beneficial effects on the host microbial environment. At present, most studies focused on Helicobacter pylori, ignoring other stomach microbes and the overall stomach microecology. So, in this article, we reviewed advances in human gastric microecology, the relationship between gastric microecology and immunity or gastric diseases, and the treatment of probiotics in gastric diseases, in order to explore new area for further study of gastric microorganisms and treatment of gastric diseases.

Dietary macronutrient composition influences both hepatic function and aging. Previous work suggested that longevity and hepatic gene expression levels were highly responsive to dietary protein, but almost unaffected by other macronutrients. In contrast, we found expression of 4005, 4232, and 4292 genes in the livers of mice were significantly associated with changes in dietary protein (5%-30%), fat (20%-60%), and carbohydrate (10%-75%), respectively. More genes in aging-related pathways (notably mTOR, IGF-1, and NF-kappaB) had significant correlations with dietary fat intake than protein and carbohydrate intake, and the pattern of gene expression changes in relation to dietary fat intake was in the opposite direction to the effect of graded levels of caloric restriction consistent with dietary fat having a negative impact on aging. We found 732, 808, and 995 serum metabolites were significantly correlated with dietary protein (5%-30%), fat (8.3%-80%), and carbohydrate (10%-80%) contents, respectively. Metabolomics pathway analysis revealed sphingosine-1-phosphate signaling was the significantly affected pathway by dietary fat content which has also been identified as significant changed metabolic pathway in the previous caloric restriction study. Our results suggest dietary fat has major impact on aging-related gene and metabolic pathways compared with other macronutrients.

The transition from the geometry to the mesh can be rather difficult, manual and time-consuming, especially for the large scale complex structures. The procedure of mesh generation needs massive human intervention making the automatic engineering analyses of structures from CAD geometry models hardly possible. This paper focuses on implementing a polyhedron element with arbitrary convex topology based on the Scaled Boundary Finite Element Method (SBFEM) in ABAQUS on the strength of the interface of UEL (the subroutine to define a general user-defined element) for automatic dynamic analyses of three-dimensional structures. This implementation empowers ABAQUS to analyze any model with arbitrary polyhedron elements. When the geometry of a structure is obtained from CAD, the dynamic analyses can be launched seamlessly and automatically. Cases of a cantilever subjected to a dynamic harmonic excitation with the traditional hexahedron element and this polyhedron element are compared to verify the accuracy of the UEL. Taking a practical example of the Soil-Structure Interaction analysis of a Nuclear Power Plant, the applicability and performance of this implementation are tested. The results of the two examples confirm that this polyhedron element based on SBFEM can be more accurate using much less degrees of freedom and its implementation in ABAQUS is robust and compatible.

In birds, male gonads form on both sides whereas most females develop asymmetric gonads. Multiple early lines of evidence suggested that the right gonad fails to develop into a functional ovary, mainly due to differential expression of PITX2 in the gonadal epithelium. Despite some advances in recent years, the molecular mechanisms underlying asymmetric gonadal development remain unclear. Here, using bulk analysis of whole gonads, we established a relatively detailed profile of four representative stages of chicken gonadal development at the transcriptional and chromatin levels. We revealed that many candidate genes were significantly enriched in morphogenesis, meiosis and subcellular structure formation, which may be responsible for asymmetric gonadal development. Further chromatin accessibility analysis suggested that the transcriptional activities of the candidate genes might be regulated by nearby open chromatin regions, which may act as transcription factor (TF) binding sites and potential cis-regulatory elements. We found that LHX9 was a promising TF that bound to the left-biased peaks of many cell cycle-related genes. In summary, this study provides distinctive insights into the potential molecular basis underlying the asymmetric development of chicken gonads.

We propose a scheme to generate ultra-strong four-wave mixing (FWM) signal based on a suspended monolayer graphene nanoribbon nanomechanical resonator (NR) coupled to an Au nanoparticle (NP). It is shown that, the FWM spectrum can switch among two-peaked, three-peaked, four-peaked or five-peaked via the modulation of exciton-phonon and exciton-plasmon couplings. This is mainly attributed to the vibrational properties of NR related to the exciton-phonon coupling, and the energy-level splitting of the localized exciton correlated to three classes of resonances consisting of three-photon resonance, Rayleigh Resonance, and AC-Stark atomic resonance. Especially, in a dual-strong coupling regime, the gains for these peaks can be as high as nine orders of magnitude (∼ 109) around the lower bistable threshold due to a combined effect of two couplings. Our findings not only offer an efficient way to measure the vibrational frequency of NR and the exciton-phonon coupling strength but also provide a possibility to fabricate high-performance optoelectronic nanodevices.

Exosome has been demonstrated to be secreted from cells and seized by targeted cells. Exosome could transmit signals and exert biological functions in cancer progression. Nevertheless, the underlying mechanisms of exosome in ovarian cancer (OC) have not been fully explored. In this study, we wanted to explore whether Fibroblast growth factor 9 (FGF9), as an exosome-associated gene, was importantly essential in OC progression and prognosis. Firstly, comprehensive bioinformatics platforms were applied to find that FGF9 expression was lower in OC tissues compared to normal ovarian tissues. Meanwhile, downregulated FGF9 displayed favorable prognostic values in OC patients. The gene enrichment of biological functions indicated that abnormally expressed FGF9 could be involved in the OC-related immune signatures, such as immunoinhibitors and chemokine receptors. Taken together, these findings could provide a novel insight into the significance of FGF9 in OC progress and supply a new destination of FGF9-related immunotherapy in clinical treatment.

Magnetic sensors actuated by bulk acoustic wave (BAW) have attracted extensive attention due to the fact of their high sensitivity, GHz-level high frequency, and small size. Different from previous studies, suppression of energy loss and improvement in energy conversion efficiency of the BAW magnetoelectric (ME) sensor were systematically considered during the device design in this work. Finite element analysis models of material (magnetic composite), structure (ME heterostructure), and device (BAW ME magnetic sensor) were established and analyzed in COMSOL software. Additionally, the magnetic composite was prepared by radio frequency magnetron sputtering, and its soft magnetism was characterized by magnetic hysteresis loop and surface roughness. The research results demonstrate that after inserting four layers of 5 nm Al2O3 films, a performance of 86.7% eddy current loss suppression rate, a less than 1.1% magnetostriction degradation rate, and better soft magnetism were achieved in 600 nm FeGaB. Furthermore, compared with other structures, the two-layer piezomagnetic/piezoelectric heterostructure had a better ME coupling performance. Eventually, the design of the BAW ME magnetic sensor was optimized by the resonance-enhanced ME coupling to match the resonance frequency between the magnetic composite and the BAW resonator. When a 54,500 A/m direct current bias magnetic field was applied, the sensor worked at the first-order resonance frequency and showed good performance. Its linearity was better than 1.30%, the sensitivity was as high as 2.33 µmV/A, and the measurement range covered 0-5000 A/m.

Implantable medical devices have been facing the severe challenge of wireless communication for a long time. Acoustically actuated magnetoelectric (ME) transducer antennas have attracted lots of attention due to their miniaturization, high radiation efficiency and easy integration. Here, we fully demonstrate the possibility of using only one bulk acoustic wave (BAW) actuated ME transducer antenna (BAW ME antenna) for communication by describing the correspondence between the BAW ME antenna and components of the traditional transmitter in detail. Specifically, we first demonstrate that the signal could be modulated by applying a direct current (DC) magnetic bias and exciting different resonance modes of the BAW ME antenna with frequencies ranging from medium frequency (MF) (1.5 MHz) to medium frequency (UHF) (2 GHz). Then, two methods of adjusting the radiation power of the BAW ME antenna are proposed to realize signal amplification, including increasing the input voltage and using higher order resonance. Finally, a method based on electromagnetic (EM) perturbation is presented to simulate the transmission process of the BAW ME antenna via the finite element analysis (FEA) model. The simulation results match the radiation pattern of magnetic dipoles perfectly, which verifies both the model and our purpose.

As a desirable candidate for lithium-ion batteries, potassium-ion batteries (PIBs) have aroused great interest because of their low cost and high power and energy densities. However, the insertion/extraction of K+ with a large radius (1.38 Å) usually bring about the destruction of the electrode materials. Here, ultrafine Fe7S8 nanocrystals are successfully implanted into hollow carbon nanospheres (Fe7S8@HCSs) via a facile solvothermal method and subsequent novel low-temperature sulfurization, which avoid the aggregation of Fe7S8 nanoparticles produced during high-temperature sulfidation. The ultrafine Fe7S8 nanoparticles and hollow carbon spheres can not only buffer the severe expansion/shrinkage of electrode materials caused by the repeated insertion/extraction of K+, but also provide additional accessible pathways for the high-rate K+ transmission. When tested as an anode material for PIBs, Fe7S8@HCSs achieve impressive K+ storage capacity of 523.2 mAh g-1 at 0.1 A g-1 after 100 cycles and remarkable rate capacity of 176.3 mAh g-1 at 5 A g-1. Further, assembling this anode with a K2NiFe(CN)6 cathode yields stable cycling performance, revealing its great potential for large-scale energy storage applications.

Thinopyrum intermedium (JJJsJsStSt, 2n = 6x = 42), a wild relative of common wheat, possesses many desirable agronomic genes for wheat improvement. The production of wheat-Th. intermedium introgression lines is a key step for transferring these beneficial genes into wheat. In this study, we characterized three wheat-Th. intermedium introgression lines TA3681, TA5566, and TA5567 using non-denaturing fluorescence in situ hybridization (ND-FISH), genomic in situ hybridization (GISH), polymerase chain reaction (PCR)-based landmark unique gene (PLUG) and intron targeting (IT) markers. Our results showed that TA3681 is a wheat-Th. intermedium 1St disomic addition line, TA5566 is a wheat-Th. intermedium non-Robertsonian translocation line carrying two pairs of 3A-7Js translocation chromosomes, and TA5567 is a wheat-Th. intermedium non-Robertsonian translocation line carrying a pair of 3A-7Js translocation chromosomes. We developed 13, 36, and 15 Th. intermedium chromosome-specific markers for detecting the introgressed Thinopyrum chromosomes in TA3681, TA5566, and TA5567, respectively. Stem rust assessment revealed that TA3681 exhibited high level of seedling resistance to Chinese prevalent Puccinia graminis f. sp. tritici (Pgt) pathotypes, and both TA5566 and TA5567 were highly resistant to Australian Pgt pathotypes, indicating that Th. intermedium chromosomes 1St and 7Js might carry new stem rust resistance genes. Therefore, the new identified introgression lines may be useful for improving wheat stem rust resistance.

RATIONALE & OBJECTIVE: Previous studies showing poor cardiopulmonary resuscitation (CPR) outcomes in the dialysis population have largely been derived from claims data and are somewhat limited by a lack of detailed characterization of CPR events. We aimed to analyze CPR-related outcomes in individuals receiving maintenance dialysis. STUDY DESIGN: Retrospective chart review. SETTING & PARTICIPANTS: Using electronic medical records from a single academic health care system, we identified all hospitalized adult patients receiving maintenance dialysis who had undergone in-hospital CPR between 2006 and 2014. EXPOSURE: Initial in-hospital CPR. OUTCOMES: Overall survival, predictors of unsuccessful CPR, predictors of death during the same hospitalization among initial survivors, predictors of discharge-to-home status. ANALYTICAL APPROACH: We provide descriptive statistics for the study variables and used t tests, χ2 tests, or Fisher exact tests to compare differences between the groups. We built multivariable logistic regression models to examine the CPR-related outcomes. RESULTS: A total of 184 patients received in-hospital CPR: 51 (28%) did not survive the initial CPR event, and 77 CPR survivors died (additional 42%) later during the same hospitalization (overall mortality 70%). Only 18 (10%) were discharged home, with the remaining 32 (17%) discharged to a rehabilitation facility or a nursing home. In the multivariable model, the only predictor of unsuccessful CPR was CPR duration (OR, 1.41; 95% CI, 1.24-1.61; P < 0.001). Predictors of death during the same hospitalization after surviving the initial CPR event were CPR duration (OR, 1.15; 95% CI 1.04-1.27; P = 0.007) and older age (OR, 1.64; 95% CI, 1.23-2.2; P < 0.001). Older people also had lower odds of discharge-to-home status (OR, 0.25; 95% CI, 0.11-0.54; P < 0.001). LIMITATIONS: Retrospective study design, single-center study, no information on functional status. CONCLUSIONS: Patients receiving maintenance dialysis experience high mortality following in-hospital CPR and only 10% are discharged home. These data may help clinicians provide useful prognostic information while engaging in goals of care conversations.

RESUMO

Sarcophaga polystylata (Ho, 1934) remains to be of medical and epidemiological significance. Here, we report the complete mitochondrial genome (mitogenome) of S. polystylata (GenBank accession no. MW592361). The length of this mitogenome was 15,233 bp with 39.4% A, 9.5% G, 14.3% C, and 36.8% T. The mitogenome of S. polystylata was composed of 13 protein-coding genes (PCGs), two ribosomal RNAs (rRNAs), 22 transfer RNAs (tRNAs), and a non-coding control region. Phylogenetic tree indicated that S. polystylata and Sarcophaga peregrina clustered together closely, both of which separated clearly from other species. This study provides the mitochondrial genetic data of S. polystylata for further understanding the phylogenetic relationship of sarcophagids species.
