The current study analyzes the contribution of 10 water quality parameters (including pH, turbidity, conductivity, total dissolved solids (TDS), hardness, total organic carbon (TOC), alkalinity, calcium ions, chlorides and sulfates) to corrosion extent of stainless steel valves taken from different locations in a reverse osmosis system of a reclaimed water plant. The valves were in service for 5 years. Raman spectroscopy and X-ray photoelectron spectroscopy analyses are conducted to quantify corrosion products on different valves under various water quality conditions. On that basis, bivariate and multivariate regression analyses between the 10 water quality parameters and the corrosion extent of valve specimens (represented by metal loss percentage (MLP) values) are carried out to check the contribution of those water quality parameters to MLP. The results indicate that the proportions of metal oxides as corrosion products vary according to the corrosion extent of the valves. Although no linear correlation is found, all 10 water quality parameters except for pH show a significant positive correlation with the MLP values of the valve specimens. Moreover, results of multivariate regression suggest that the variation of MLP can be explained by turbidity, TDS, TOC and sulfates. A positive contribution of turbidity, TDS and TOC to MLP is observed, whereas the contribution of sulfates is negative. The results from the current work help to identify the reasons for water quality-induced failure of stainless steel equipment in RO systems.

Background: As one of the most common cancers, breast carcinoma is the most common disease in women. Intersectin 1 (ITSN1) contributes to the actin cytoskeleton reconstruction in breast cancer. Purpose: The objective of this study to explore the functions of ITSN1 in breast carcinoma. Methods: We downloaded microarray datasets GSE8087, GSE50697, and GSE98238 from the Gene Expression Omnibus database. Differentially expressed genes (DEGs) were used to construct a protein-protein interaction (PPI) network using STRING database, and the modules from PPI network were verified by Cytoscape software. Gene ontology terms and Kyoto Encyclopedia of Gene and Genome pathway were used to analyze the biological functions using the DAVID database. ONCOMINE, GEPIA, UALCAN, and Human Protein Atlas databases were used to investigate the characteristics of ITSN1 for the prognosis of breast carcinoma. Cell counting kit-8, flow cytometry, and colony formation assays were used to detect cell viability, cell apoptosis, and cell proliferation. RT-PCR and Western blot assays were used to detect ITSN1, Ki67, and cleaved caspase-3 expressions. Results: Low expressions of ITSN1 were significantly associated with clinical cancer stages. RT-PCR and Western blot analysis showed low expression of ITSN1 in breast cancer tissues and cell lines. ITSN1 inhibition could promote cell proliferation and inhibit cell apoptosis, while ITSN1 overexpression could inhibit cell proliferation and increase cell apoptosis by regulating the levels of expression of Ki67 and cleaved-caspase-3. Conclusion: The results indicated that ITSN1 could be a prognostic biomarker for survivals of breast cancer patients.

The protective mechanism of chitosan oligosaccharide (COS) against lipopolysaccharides (LPS) -induced inflammatory responses in IPEC-J2 and in mice with DSS dextran sulfate sodium (DSS) -induced colitis is reported. Upon exposure to LPS, the proliferation rate of IPEC-J2 cells markedly decreased, and epithelial cell integrity was compromised. However, COS pretreatment significantly reduced these changes. Low-concentration (200 µg/mL) COS up-regulated Toll-like receptor 4 (TLR4) and nuclear p65 expression, but inhibited LPS-induced expression of nuclear p65, IL-6, and IL-8. Addition of the TLR4 inhibitor reduced nuclear p65, IL-6, and IL-8 expression in IPEC-J2 cells exposed to COS or LPS alone, and a slight up-regulation in nuclear p65 was observed in COS and LPS co-treated cells. Medium-dose COS (600 mg/kg/d) protected against DSS-induced colitis, in which TLR4 and nuclear p65 expression levels were decreased. We postulate that the prevention of both LPS- and DSS -induced inflammatory responses in IPEC-J2 cells and mice by COS are related to the inhibition of the TLR4/NF-κB signaling pathway.

Long non-coding RNAs (lncRNAs) are frequently dysregulated in cancer and their aberrant expression has been associated with cancer diagnosis and prognosis, which suggests that they may be promising molecular biomarkers. However, understanding of the expression pattern of lncRNAs and their prognostic roles in head and neck squamous cell carcinoma (HNSCC) is relatively limited. In the current study, the prognostic value of lncRNA expression profiles in predicting the OS of patients with HNSCC was investigated by integrating clinical and profiling data from The Cancer Genome Atlas. A total of ten lncRNAs closely associated with the prognosis of patients with HNSCC were identified and may serve as novel biomarkers. This 10-lncRNA signature was used to classify patients into 2 groups with significantly different overall survival (OS) times (median OS time, 1.65 vs. 13.04 years; P<0.0001). This lncRNA signature was validated in an independent testing cohort. The results of multivariable Cox regression and stratification analyses revealed that the prognostic value of the 10-lncRNA signature was independent of other clinical and pathological factors for the survival of patients with HNSCC. Functional analysis demonstrated that lncRNA expression-based risk scoring may reflect the basic status of the immune response in the tumor microenvironment. The presented study demonstrated the value of a lncRNA signature as a potential biomarker to improve the clinical prognosis of patients with HNSCC.

In this paper, we describe the discovery and optimization of a series of noncovalent reversible epidermal growth factor receptor inhibitors of EGFRL858R/T790M/C797S. One of the most promising compounds, 25g, inhibited the enzymatic activity of EGFRL858R/T790M/C797S with an IC50 value of 2.2 nM. Cell proliferation assays showed that 25g effectively and selectively inhibited the growth of EGFRL858R/T790M/C797S-dependent cells. This series of compounds, which occupy both the ATP binding site and the allosteric site of the EGFR kinase, may serve as a basis for the development of fourth-generation EGFR inhibitors for L858R/T790M/C797S mutants.

Metamaterial absorbers, consisting of assembling arrays of optical resonators with subwavelength dimensions and spacing, allow efficiently absorption electromagnetic radiation by leveraging the strong electrical and magnetic resonances. Beyond the enhanced absorption, there is a growing interest to realize multi-functional absorbers, for example, absorbers with extended bandwidth, strong polarization extinction ratio, to name a few. Traditionally, designing multi-functional absorbers require complex brute-force optimizations with sizable parameter space, which turn out to be rather inefficient. Here, using the particle swarm optimization algorithm, we design and experimentally demonstrate broadband and highly polarization selective mid-IR metal-insulator-metal absorbers, covering the technologically important 3-5 µm atmospheric transparency band. With spectrally averaged absorption exceeding 70%, a high polarization extinction ratio of 40.6 is concurrently achieved by the algorithm. We also investigate the incident angle dependence of the spectral absorption and clarify the origin of optical losses. By integrating with the growing range of mid-IR detectors and imagers, our devices can enable new applications such as mid-IR full Stokes imaging polarimetry for remote sensing.

Recent work shows that Pockels effect and optics pump-probe measurement could be utilized as a novel method for 511 keV ionizing radiation photon detection for positron emission tomography (PET) which could potentially overcome the inherent physical limitation for coincidence time resolution of around 100 ps (Tao et al 2016 Phys. Med. Biol. 61 7600-22). In this paper, we embrace this observation and introduce a two-crossed-polarizers based setup to achieve similar detection concept, which is a simpler and more compact setup with comparable ionizing radiation detection capability as the setup used in the previously proposed work. We evaluated the performance of our experimental setup with Lithium Niobate (LiNbO3) and Cadmium Telluride (CdTe) detector crystals, and the desired properties of an ideal detector crystal were discussed. The modulation signal induced by 511 keV photons in both LiNbO3 and CdTe can be detected with repeatable signal amplitude using two-crossed-polarizers based method, while CdTe could provide eight times higher detection sensitivity to 511 keV photons than LiNbO3 under the same bias voltage, suggesting high effective Z number and high density properties of CdTe, as well as a shorter carrier lifetime and lower carrier mobility of LiNbO3. In addition, the strength of modulation signal increased linearly with bias voltage before saturation. The modulation signal strength in LiNbO3 continued to increase after 2000 V due to its high resistivity which could reduce the dark current in the detector, while the modulation signal of CdTe with low resistivity tended to be saturated at a bias voltage higher than 1200 V. Therefore, further increasing the bias voltage for detector crystals (especially for LiNbO3) may enhance the modulation strength and improve the detection sensitivity for annihilation photons.

Tandem-junction organic solar cells require solar cells with visible light photo-response as front cells, in which an open-circuit voltage (Voc ) above 1.0 V is highly demanded. In this work, we are able to develop electron acceptors to fabricate non-fullerene organic solar cells (NFOSCs) with a very high Voc of 1.14 V. This was realized by designing perylene bisimide (PBI)-based conjugated materials fused with benzodithiophene, in which Cl and S atom were introduced into the molecules in order to lower the frontier energy levels. The fused structures can reduce the aggregation of PBI unit and meanwhile maintain a good charge transport property. The new electron acceptors were applied into NFOSCs by using Cl and S substituted conjugated polymers as electron donor, in which an initial power conversion efficiency of 6.63 % and a high Voc of 1.14 V could be obtained. The results demonstrate that the molecular design by incorporating Cl and S atom into electron acceptors has great potential to realize high performance NFOSCs.

BACKGROUND: Multimodal analgesia can improve postoperative pain and possibly accelerate functional recovery after surgery. Serratus plane block (SPB) is a novel, ultrasound-guided regional anaesthetic technique for complete analgesia of the anterolateral chest wall. But, the effect of SPB on the quality of recovery after breast cancer surgery has not been established. OBJECTIVE: To test the hypothesis that pre-operative SPB would enhance the quality of recovery following breast cancer surgery. DESIGN: A randomised, double-blind, parallel-group, placebo-controlled trial. SETTING: Single university teaching hospital, from March 2016 to June 2017. PATIENTS: Seventy-two women scheduled for breast cancer surgery. INTERVENTION: Participants were randomised in a 1 : 1 ratio to receive SPB with 25 ml of ropivacaine 0.5% or physiological saline. MAIN OUTCOME MEASURES: The primary endpoint was the 40-item Quality of Recovery questionnaire score 24 hours postoperatively hours. Secondary endpoints were postoperative pain intensity, cumulative opioid consumption, postoperative nausea and vomiting, dizziness, post anaesthesia care unit discharge time and patient satisfaction. RESULTS: The global median [IQR] 40-item Quality of Recovery questionnaire score at 24 postoperative hours was significantly higher in the SPB group (158 [153.8 to 159.3]) than the control group (141 [139 to 145.3]) with a median difference of 15 (95% confidence interval: 13 to 17, P < 0.001). Compared with the control group, postoperative pain scores at rest were significantly lower up to 24 h in the SPB group. Pre-operative SPB reduced postoperative cumulative opioid consumption, the incidence of postoperative nausea and vomiting and the post anaesthesia care unit discharge time. In addition, patient satisfaction scores were higher in the SPB group. CONCLUSION: Pre-operative administration of SPB with ropivacaine improved the quality of recovery, postoperative analgesia and patient satisfaction following breast cancer surgery. Purpose: Triple-negative breast cancer (TNBC) is a highly heterogeneous disease. It is very important to explore novel biomarkers to better clarify the characteristics of TNBC. It has been reported that polymorphisms in claudin 1 (CLDN1) are associated with risk of several cancers. But till now, there is no report about these polymorphisms and TNBC. Patients and methods: Between January 2004 and December 2013, 267 patients with stage I-III primary TNBC were included in our study. We investigated the association between polymorphisms in CLDN1 gene and clinicopathological characteristics or survival of these patients. We used Haploview 4.2 software to identify Tag single nucleotide polymorphisms (SNPs). MassARRAY MALDI-TOF System was used for genotyping. Results: We found that rs10513846 GA genotype was associated with older age [P=0.013, hazard ratios (HR) = 2.231, 95% confidence interval (CI): 1.186-4.195]. Rs10513846 AA genotype carriers were more likely to develop grade 3 tumors (P=0.005, HR = 2.889, 95% CI: 1.389-6.007). And rs9283658 genotypes were also related to grade, more patients with grade 3 tumors were rs9283658 CC genotype carriers (P=0.023, HR = 0.446, 95% CI: 0.222-0.894). There was no association between polymorphisms in CLDN1 and survival of TNBC patients. After multivariate analysis, tumor size (P=0.021, HR = 3.146, 95% CI: 1.185-8.354) and lymph node status (P<0.001, HR = 10.930, 95% CI: 3.276-36.470) were demonstrated to be independent prognostic factors. Conclusion: We first demonstrated that polymorphisms in CLDN1 gene were associated with age and differentiation of TNBC patients.

Risk of cross-connection is becoming higher due to greater construction of potable-reclaimed water dual distribution systems. Cross-connection events can result in serious health concerns and reduce public confidence in reclaimed water. Thus, reliable, cost-effective and real-time online detection methods for early warning are required. The current study carried out pilot-scale experiments to simulate potable-reclaimed water pipe cross-connection events for different mixing ratios (from 30% to 1%) using machine learning methods based on multiple conventional water quality parameters. The parameters included residual chlorine, pH, turbidity, temperature, conductivity, oxidation-reduction potential and chemical oxygen demand. The results showed that correlated variation occurred among water quality parameters at the time of the cross-connection event. A single parameter-based method can be effective at high mixing ratios, but not at low mixing ratios. The direct supporting vector machine (SVM)-based method managed to overcome this drawback, but coped poorly with abnormal readings of water parameter sensors. In that respect, a Pearson correlation coefficient (PCC)-SVM-based method was developed. It provided not only high detection performance under normal conditions, but also remained reliable when abnormal readings occurred. The detection accuracy and true positive rate of this method was still over 88%, and the false positive rate was below 12%, given a sudden variation of an individual water quality parameter. The receiver operating characteristic curves further confirmed the promising practical applicability of this PCC-SVM-based method for early detection of cross-connection events.

Lung cancer is one of the most common cancers and has been the most common cause of cancer deaths for several decades. Recently, lung cancer-associated lncRNA 1 (LCAL1) has been identified to be overexpressed in lung cancer tissues, while inhibiting LCAL1 expression has shown potential to inhibiting lung cancer growth. However, the molecular mechanism between LCAL1 and lung cancer cell survival remains poorly understood. In the present study, we provided the first evidence that LCAL1 may support lung cancer survival via inhibiting the activity of AMP-activated protein kinase (AMPK). According to our results, LCAL1 may physically interact with the catalytic subunit of tumor suppressor AMPK, prevent AMPK activation by upstream kinase (liver kinase B1), and thus inhibit the downstream AMPK signaling network. Our study revealed that overexpressed LCAL1 may induce aerobic glycolysis in lung cancer cells through AMPK/HIF1α axis, enhance protein synthesis through AMPK/mTOR/S6K axis, and suppress autophagic cell death through AMPK/ULK1 pathway. All these alterations supported rapid proliferation of lung cancer cells, while knockdown of LCAL1 expression demonstrated the potential of inhibiting lung cancer growth by reversing the tumorigenic phenotypes triggered by the loss of AMPK activity, and could become a promising therapeutic strategy for lung cancer treatment.

The aim of the present study was to discuss the design of a microfluidic chip consisting of columns, and its use for the enrichment of nasopharyngeal cancer (NPC) cells. A microfluidic chip experiment was simulated using FLUENT software. Within the microfluidic chip, aptamers were bound to the reaction chamber (consisting of columns) using a biotin-avidin system. Cell suspension was introduced into the reaction chamber to capture NPC cells. NPC cells were subsequently eluted, and the capture rate of the cells was calculated. The modified aptamer-bound microfluidic chip was able to capture NPC cells with a capture rate of ~90%. The modified aptamer-bound microfluidic chip has a wide range of potential applications for the diagnosis of NPC.

Heat stress (HS) and its associated pathologies are major challenges facing the pig industry in southern China, and are responsible for large economic losses. However, the molecular mechanisms governing the abnormal secretion of HS-responsive hormones, such as glucocorticoids, are not fully understood. The goal of this study was to investigate differentially expressed proteins (DEPs) in the adrenal glands of pigs, and to elucidate changes in the immune neuroendocrine system in pigs following HS. Through a functional proteomics approach, we identified 1202 peptides, corresponding to 415 proteins. Of these, we found 226 DEPs between heat-stressed and control porcine adrenal gland tissue; 99 of these were up-regulated and 127 were down-regulated in response to HS. These DEPs included proteins involved in substrate transport, cytoskeletal changes, and stress responses. Ingenuity Pathway Analysis was used to identify the subcellular characterization, functional pathway involvement, regulatory networks, and upstream regulators of the identified proteins. Functional network and pathway analyses may provide insights into the complexity and dynamics of HS-host interactions, and may accelerate our understanding of the mechanisms of HS.

Fast deposition of aligning ambipolar polymers for high-performance organic field-effect transistors (OFETs) and inverter circuits are highly desired for both scientific studies and industry applications. Here, large-area and ordered polymer films are prepared by a bar-coating method at a rate of 120 mm s-1 in air. Atomic force microscopy and grazing-incidence wide-angle X-ray scattering analysis indicate uniform edge-on poly(fluoroisoindigo-difluorobithiophene-fluoroisoindigo-bithiophene) (PFIBI-BT) in 11.7 ± 1 nm film (≈5 layers). The elongated, uniformly oriented grains can reduce the adverse effects of the grain boundaries and facilitate charge transport in polymers. Furthermore, OFETs based on parallel film show high hole/electron mobilities up to 5.5/4.5 cm2 V-1 s-1 , which are approximately nine times of the devices prepared by spin-coating. The gain of the inverter is as high as 174, which is one of the highest values in polymer inventers currently. These results demonstrate that the excellent bipolar performance of few-layer PFIBI-BT can be ensured while achieving the compatibility of the experimental process with industrial preparation.

Purpose: Triple-negative breast cancer (TNBC) is more than a single disease. Identifying biomarkers to further subdivide TNBC patients with distinct outcome is of great importance. It has been reported that single-nucleotide polymorphisms (SNPs) in Aurora kinase A (AURKA) or Aurora kinase B (AURKB) are associated with the risk and survival of several cancers. But till now, there is no research about these polymorphisms in TNBC patients. Materials and methods: In this study, we investigated the association between polymorphisms in AURKA or AURKB gene and prognosis of TNBC patients treated with taxane-based adjuvant chemotherapy. A total of 273 TNBC patients were enrolled. Haploview 4.2 software was used to identify Tag SNPs. Genotyping was conducted using the MassARRAY MALDI-TOF system. Results: We found that AURKA rs6099128 GG genotype carriers had significantly worse overall survival (OS) than TT+ TG genotype carriers (P = 0.003, HR = 12.499, 95% CI = 2.357-66.298). AURKB rs11651993 TT genotype carriers had better disease-free survival (DFS) than TC + CC genotype carriers (P = 0.018, HR = 1.876, 95% CI = 1.116-3.154). AURKB rs2289590 CC genotype carriers had worse DFS than CA + AA genotype carriers (P = 0.021, HR = 0.536, 95% CI = 0.315-0.912). After subgroup analysis, rs11651993 TC + CC genotype predicted worse DFS in subgroups of age ≤ 50, post-menopausal, grade unknown (UK), tumor size >2 cm, and lymph node negative. Rs2289590 CA + AA genotype could predict favorable DFS in pre-menopausal, grade 3 and lymph node-positive patients. Conclusion: We first demonstrated that polymorphisms in AURKA or AURKB gene might predict the OS or DFS of TNBC patients treated with taxane-based adjuvant chemotherapy.

A recent increase in the use of bisphenol A (BPA) alternatives to manufacture plastics has led to safety concerns. Here, we evaluated the estrogenic and anti-estrogenic activities of bisphenol AP (BPAP), a poorly studied BPA alternative, using in vitro, in vivo and in silico tools. BPAP exhibited weak estrogenicity but strong anti-estrogenicity (IC50 = 2.35 µM) in a GeneBLAzer™ ß-lactamase reporter gene assay. BPAP, when administered alone or in combination with E2 (50 µg kg-1 bw d-1) for 3 d, significantly decreased the uterine weights of post-weaning CD-1 mice at doses of 10 mg kg-1 bw d-1 and higher. When administered alone to prepubertal CD-1 mice for 10 d, BPAP significantly decreased the uterine weights at doses of 80 µg kg-1 bw d-1 and higher. Toxicogenomic analysis showed that BPAP regulated an opposite patterns of gene expression than that of E2 in mouse uteri. In a glucose tolerance test using male mice, BPAP was found to disrupt the blood glucose homeostasis at low doses relevant to human exposure (1 and 100 µg kg-1 bw d-1). Our results suggest that BPAP should be of great concern which might affect the sexual development in immature feminine and disrupt the blood glucose homeostasis at very low doses.

Here we numerically and experimentally explore the rich phenomena in the optical responses and local electromagnetic fields of a MIM absorber mediated by the dispersive spacer. We first show that the line shape of the spectral absorption is strongly modified by the interaction between the plasmonic resonances and the optical phonons in the silicon dioxide spacer. Importantly, broadening the spectral absorption in the long wave infrared range is achieved by tuning the strength of coupling. Modification to the local electromagnetic field distribution in the epsilon-near-zero region is also numerically studied. The incident-angle dependence and polarization dependence of the broadened absorption spectrum are evaluated. We also show that the spectral broadening mechanism can be generalized to other frequency bands by employing different spacing materials such as silicon nitride and polydimethylsiloxane. Our results can be useful for designing spectrally selective thermal detectors and thermal emitters.

RESUMO

A high IR-blocking cellulose film was designed based on an amorphous to anatase transition of TiO2 using atomic layer deposition (ALD). This transition was realized at 250 °C, at which the cellulose is thermal stable. Optimized ALD condition of 250 °C and 1200 cycles give us an excellent heat insulator, which could significantly reduce the enclosed space temperature from 59.2 to 51.9 °C after exposure to IR lamp for 5 min.

BACKGROUND: Polymorphisms in miRNA machinery genes have been proved to be related to risk or survival of several kinds of cancers, but the results are controversial and the role of these polymorphisms in gastric cancer remains uncertain. In our study, we investigated the association between five genetic variants in miRNA machinery genes ( DICER, RAN, XPO5 [name of the gene]) and clinical outcomes in Chinese gastric cancer patients. METHODS: A total of 96 patients with stage IB-III gastric cancer treated with radical gastrectomy and adjuvant chemotherapy of oxaliplatin and fluorouracils were analyzed. The MassARRAY MALDI-TOF system was used to determine the genotypes. RESULTS: DICER rs3742330 AG+GG genotype was associated with more advanced T stage compared to AA genotype ( P=0.009). More patients with XPO5 rs2257082 CC genotype had poorly differentiated tumors compared with CT+TT genotype carriers. After adjustment by age, sex, differentiation, T stage, and lymph node status, XPO5 rs2257082 CC genotype carriers were found to have worse disease-free survival than CT+TT genotype carriers (adjusted HR 3.099; 95% CI 1.270, 7.564; P=0.013), carriers of RAN rs14035 CC genotype had higher three-year OS rate than carriers of CT+TT genotype (adjusted HR 3.174; 95% CI 1.010, 9.973; P=0.048). CONCLUSIONS: These results indicated that genetic variants in miRNA machinery genes might be associated with the clinicopathological features and prognosis of completely resected gastric cancer patients.
