RESUMO

16S rRNA gene sequence analysis showed that the type strain Brevibacterium frigoritolerans DSM 8801T had the highest similarity (99.7%) with the reference strain Bacillus simplex NBRC 15720T, followed by Bacillus muralis DSM 16288T (99.6%), Bacillus butanolivorans DSM 18926T (99.5%), and Bacillus loiseleuriae FJAT-27997T (97.9%). This relationship is confirmed by the phylogenetic analysis indicating that Bre. frigoritolerans DSM 8801T fell in the genus Bacillus group and formed a clade with the closely related Bacillus species. Average nucleotide identity and in silico DNA-DNA hybridization (isDDH) values between strain DSM 8801T and the most closed reference strain Bac. simplex NBRC 15720T were both much lower than species definition threshold values of 95% and 70%, respectively, which indicated strain DSM 8801T should not be affiliated to one of the validly named Bacillus species. The percentage of conserved protein (POCP) values between the strain DSM 8801T and the type strains of the above species were 80.7%, 69.2%, 72.2%, 53.6%, and 50.0%, respectively, higher than the genus definition threshold value of 50%. The main isoprenoid quinone of strain DSM 8801T was MK-7, the main polar lipids were diphosphatidylglycerol (DPG), phosphatidylglycerol (PG), and phosphatidyl ethanolamine (PE), and the major fatty acids were anteiso-C15:0 and iso-C15:0. On the basis of the phenotypic, chemotaxonomic, phylogenetic and genomic characteristics, Bre. frigoritolerans DSM 8801T should belong to the genus Bacillus, and be proposed to reclassify as Bacillus frigoritolerans comb. nov., with the type strain DSM 8801T (=ATCC 25097T  = CCUG 43489T  = CIP 67.20T  = JCM 11681T).

Boron forms a rich variety of low-dimensional nanosystems, including the newly discovered helix Be6 B10 2- (1) and Be6 B11 - (2) clusters. We report herein on the elucidation of chemical bonding in clusters 1/2, using the modern quantum chemistry tools of canonical molecular orbital analyses and adaptive natural density partitioning (AdNDP). It is shown that clusters 1/2 contain a chiral helix Be2 B10 Be2 or Be2 B11 Be2 skeleton with a total of 11 and 12 segments, respectively, which effectively curve into "helical pseudo rings" and chemically consist of two "quasicircles" as defined by their anchoring Be centers. The helix skeleton is connected via Lewis-type B-B and Be-B-Be σ bonds, being further stabilized by island π/σ bonds and a loose π bond at the junction. The Be6 component in 1/2 assumes a distorted prism shape only physically, and it is fragmented into four parts: two terminal Be2 dimers and two isolated Be centers. A Be2 dimer at the far end manages to bend over and cap a quasicircle from one side of B plane. Consequently, each quasicircle of a helical pseudo ring is capped from opposite sides by two Be2 /Be units, facilitating intramolecular charge-transfers of 5 electrons from Be to B. Overall, the folding of B helix involves as many as 10 electrons. The enormous electrostatics offers the ultimate driving forces for B helix formation.

Three-dimensional (3D) organic-inorganic lead halides represented by [CH3NH3]PbI3 perovskite have attracted great interest for their diverse functional properties and promising optoelectronic applications. However, 3D lead halides are still very rare and their ferroelectricity remains controversial. Here, we report an unprecedented 3D lead halide perovskite-related ferroelectric [2-trimethylammonioethylammonium]Pb2Cl6 ([TMAEA]Pb2Cl6), which contains a 3D lead chloride framework of corner- and edge-sharing PbCl6 octahedral, with the [TMAEA]+ cations occupying the voids of the framework. [TMAEA]Pb2Cl6 shows a ferroelectric-to-paraelectric phase transition with the Curie temperature as high as 412 K, a typical ferroelectric hysteresis loop at 293 K with a spontaneous polarization of 1 µC/cm2, and a clear ferroelectric domain switching. To the best of our knowledge, [TMAEA]Pb2Cl6 is the first 3D lead halide showing such an excellent ferroelectricity. Additionally, it also exhibits a semiconducting property with a direct band gap of 3.43 eV. This finding enriches the family of 3D hybrid lead halides and inspires the exploration of 3D lead halide ferroelectrics.

BACKGROUND: Traumatic brain injury (TBI) has increased in rank among traumatic injuries worldwide. Traumatic brain injury is a serious obstacle given that its complex pathology represents a long-term process. Recently, systems biology strategies such as metabolomics to investigate the multifactorial nature of TBI have facilitated attempts to find biomarkers and probe molecular pathways for its diagnosis and therapy. METHODS: This study included a group of 20 rats with controlled cortical impact and a group of 20 sham rats. We utilized mNSS tests to investigate neurological metabolic impairments on day 1 and day 3. Furthermore, we applied metabolomics and bioinformatics to determine the metabolic perturbation caused by TBI during the acute period in the hippocampus tissue of controlled cortical impact (CCI) rats. Notably, TBI-protein-metabolite subnetworks identified from a database were assessed for associations between metabolites and TBI by the dysregulation of related enzymes and transporters. RESULTS: Our results identified 7 and 8 biomarkers on day 1 and day 3, respectively. Additionally, related pathway disorders showed effects on arginine and proline metabolism as well as taurine and hypotaurine metabolism on day 3 in acute TBI. Furthermore, according to metabolite-protein database searches, 25 metabolite-protein pairs were established as causally associated with TBI. Further, bioinformation indicated that these TBI-associated proteins mainly take part in 5'-nucleotidase activity and carboxylic acid transmembrane transport. In addition, interweaved networks were constructed to show that the development of TBI might be affected by metabolite-related proteins and their protein pathways. CONCLUSION: The overall results show that acute TBI is susceptible to metabolic disorders, and the joint metabolite-protein network analysis provides a favorable prediction of TBI pathogenesis mechanisms in the brain. The signatures in the hippocampus might be promising for the development of biomarkers and pathways relevant to acute TBI and could further guide testable predictions of the underlying mechanism of TBI.

OBJECTIVE: To assess the therapeutic effect of a novel tension band using 3.0 mm cannulated screw combined with a titanium cable and specific shims comparatively with patellotibial tubercle cerclage in comminuted fractures of the inferior pole of the patella. METHODS: The retrospective study from March 2012 to July 2017 was conducted in Peking University Third Hospital and comprised 63 patients with comminuted fractures of the inferior pole of the patella: 41 treated with new tension band using 3.0 mm cannulated screw combined with a titanium cable and specific shims (new tension band group) and 22 with patellotibial tubercle cerclage (tubercle cerclage group). Gender, age, AO/OTA fracture type, injury mechanism, inter-fragmentary gap, and follow-up time of patients were recorded. Two groups were compared regarding: operation time, blood loss, partial weight-bearing time, fracture-healed time, Bostman score and knee mobility at 12-month follow-up, and postoperative complications. Continuous and categorical parameters were analyzed by Mann-Whitney U test and the chi-squared test, respectively. Fisher's exact test was used for small data subsets. RESULTS: Between the two groups, no statistically significant difference was found in mean age, gender, AO/OTA fracture type, injury mechanism, mean inter-fragmentary gap, or mean follow-up time (P > 0.05). The mean operation time of new tension band group was significantly longer than that of tubercle cerclage group (76.4 min vs 64.2 min, P = 0.006), while there was no significant difference in blood loss. After surgery, new tension band group had a significantly earlier mean partial weight-bearing time (5.2 weeks vs 7.4 weeks, P < 0.001) and fracture-healed time (9.6 weeks vs 11.6 weeks, P < 0.001). At 12-month follow-up, patients of new tension band group had a significantly higher mean Bostman score (28.5 vs 25.8, P < 0.001) and knee mobility (126.7 vs 117.3, P < 0.001). Ten complications related with internal fixation were found in tubercle cerclage group including two cases of loose internal fixation, two cases of cerclage breakage, and six cases of low patella position who undertook secondary operation. No complications were found in new tension band group (0 in 41 vs 10 in 22, P < 0.001). CONCLUSION: Patients with comminuted fractures of the inferior pole of the patella treated with a novel tension band experienced a longer operation time, but earlier partial-weight-bearing and fracture-healed time, better clinical outcomes at 12-month follow-up, and less complications. It should be considered an alternative therapy for the treatment of distal pole patellar fractures.

In recent years, some studies suggested that the pretreatment C-reactive protein to albumin ratio (CAR) may be predictive for prognosis of esophageal cancer (EC), but their results were inconsistent. Therefore, the current meta-analysis was preformed to better determine the prognostic value of pretreatment CAR in EC. The PubMed, EMBASE, Web of Science, Cochrane Library, and PubMed Central databases were searched up to January 10, 2019 to identify studies evaluating the correlation between CAR and prognosis of EC. The primary outcome was the overall survival (OS) and secondary outcomes were disease-free survival (DFS) and cancer-specific survival (CSS). Pooled hazard ratios (HRs) and corresponding 95% confidence intervals (95% CIs) were used to estimate the predictive role of CAR for prognosis in EC. Based on the results of this research, a total of 11 studies with 2930 patients diagnosed as EC were included. Pooled results suggested that elevated pretreatment CAR was significantly associated with poor OS (HR = 1.80, 95% CI: 1.31-2.47, P < .001) with high heterogeneity (I2 = 86.5%, P < .001) and poor CSS (HR = 1.72, 95% CI: 1.33-2.22, P < .001) without heterogeneity (I2 = 0.0%, P < .323); however, no significant association was observed between pretreatment CAR and DFS (HR = 1.41, 95% CI: 0.60-3.34, P = .429) with high heterogeneity (I2 = 76.7%, P < .038). Subgroup analyses further manifested that EC patients with higher CAR had worse OS. An elevated pretreatment CAR may indicate poor survival in patients with EC. Thus, pretreatment CAR may serve as a promising biomarker in EC and could be used to predict prognosis and help decision-making in clinical work.

The mechanisms involved in the regulation of gene expression in response to phosphate (Pi) deficiency have been extensively studied, but their chromatin-level regulation remains poorly understood. We examined the role of histone acetylation in response to Pi deficiency by using the histone deacetylase complex1 (hdc1) mutant. Genes involved in root system architecture (RSA) remodeling were analyzed by quantitative real-time polymerase chain reaction (qPCR) and chromatin immunoprecipitation qPCR. We demonstrate that histone H3 acetylation increased under Pi deficiency, and the hdc1 mutant was hypersensitive to Pi deficiency, with primary root growth inhibition and increases in root hair number. Concomitantly, Pi deficiency repressed HDC1 protein abundances. Under Pi deficiency, hdc1 accumulated higher concentrations of Fe3+ in the root tips and had higher expression of genes involved in RSA remodeling, such as ALUMINUM-ACTIVATED MALATE TRANSPORTER1 (ALMT1), LOW PHOSPHATE ROOT1 (LPR1), and LPR2 compared with wild-type plants. Furthermore, Pi deficiency enriched the histone H3 acetylation of ALMT1 and LPR1. Finally, genetic evidence showed that LPR1/2 was epistatic to HDC1 in regulating RSA remodeling. Our results suggest a chromatin-level control of Pi starvation responses in which HDC1-mediated histone H3 deacetylation represses the transcriptional activation of genes involved in RSA remodeling in Arabidopsis.

Molecular ferroelectrics have attracted considerable interests because of their easy and environmentally friendly processing, low acoustical impedance and mechanical flexibility. Herein, a molecular thermochromic ferroelectric, N,N'-dimethyl-1,4-diazoniabicyclo[2.2.2]octonium tetrachlorocuprate(II) ([DMe-DABCO]CuCl4 ) is reported, which shows both excellent ferroelectricity and intriguing thermochromism. [DMe-DABCO]CuCl4 undergoes a ferroelectric phase transition from Pca21 to Pbcm at a significantly high Curie temperature of 413 K, accompanied by a color change from yellow to red that is due to the remarkable deformation of [CuCl4 ]2- tetrahedron, where the ferroelectric and paraelectric phases correspond to yellow and red, respectively. Combined with multiple bistable physical properties, [DMe-DABCO]CuCl4 would be a promising candidate for next-generation smart devices, and should inspire further exploration of multifunctional molecular ferroelectrics.

Piezoelectric sensors that can work under various conditions with superior performance are highly desirable with the arrival of the Internet of Things. For practical applications, a large piezoelectric voltage coefficient g and a high Curie temperature Tc are critical to the performance of piezoelectric sensors. Here, we report a two-dimensional perovskite ferroelectric (4-aminotetrahydropyran)2PbBr4 [(ATHP)2PbBr4] with a saturated polarization of 5.6 µC cm-2, high Tc of 503 K [above that of BaTiO3 (BTO, 393 K)], and extremely large g33 of 660.3 × 10-3 V m N-1 [much beyond that of Pb(Zr,Ti)O3 (PZT) ceramics (20 to 40 × 10-3 V m N-1), more than 2 times higher than that of poly(vinylidene fluoride) (PVDF, about 286.7 × 10-3 V m N-1)]. Combined with the advantages of molecular ferroelectrics, such as light weight, easy and environmentally friendly processing, and mechanical flexibility, (ATHP)2PbBr4 would be a competitive candidate for next-generation smart piezoelectric sensors in flexible devices, soft robotics, and biomedical devices.

BACKGROUND: Squamous cell carcinoma (SCC) of the nail bed is a poorly reported malignant subungual tumor. Because it presents with nonspecific symptoms and signs, it is frequently misdiagnosed by dermatologists or surgeons. A delay in diagnosis and/or wrong treatment might increase the possibility of disease progression. Thus, new perspectives are needed to assist dermatologists and surgeons with diagnosing and treating SCC. This rare case presented with a 2-year delay in the diagnosis of SCC teaches a valuable lesson. CASE SUMMARY: A 62-year-old female presented with a non-healing subungual growth in the nail bed of the right middle finger for 2 years. The lesion was first medicated with iodine by the patient herself without any relief. Twenty months later, a dermatologist diagnosed the lesion as paronychia and treated it with nail avulsions repeatedly with no obvious alleviation. A lesionectomy confirmed the lesion was SCC. An extended excision of the tumor with amputation of the distal interphalangeal joint was subsequently performed. A biopsy of sentinel lymph nodes was negative. Due to the result of preoperative positron emission tomography-computed tomography scanning, sweeping of axillary lymph nodes was considered dispensable and was skipped. At the 2-year follow-up, the patient showed a quick recovery and no sign of recurrence. CONCLUSION: Our successful diagnosis and treatment of the case highlights the need for additional attention to long-standing non-healing lesions of the nail bed and the necessity for discreet evaluation and customization of surgical interventions.

Two-dimensional (2D) organic-inorganic perovskites (OIPs), with improved material stability over their 3D counterparts, are highly desirable for device applications. It is their considerable structural diversity that offers an unprecedented opportunity to engineer materials with fine-tuning functionalities. The isosteric substitution of hydrogen by an electronegative fluorine atom has been proposed as a useful route to improve the photovoltaic performance of 2D OIPs, whereas its valuable role in developing ferroelectricity is still waiting for further exploration. Herein, for the first time we applied fluorinated aromatic cations in extending the family of 2D OIP ferroelectrics, and successfully obtained [2-fluorobenzylammonium]2PbCl4 as a high-performance ferroelectric semiconductor. The failures in the nonferroelectric [4-fluorobenzylammonium]2PbCl4 and [3-fluorobenzylammonium]2PbCl4 demonstrate that the selective introduction of fluorine in correct structural positions is particularly essential. This work represents an unprecedented proof-of-concept in the use of fluorinated aromatic cations for the targeted design of excellent 2D OIP ferroelectrics, and is believed to inspire the future development of low-cost, high-efficiency, and stable device applications.

Background: Inherited medullary thyroid carcinoma (MTC) is primarily caused by RET mutations that are commonly localized in exons 5, 8, 10, 11, and 13-16. In this study, we report pedigrees for individuals with MTC that harbor a germline S409Y variant within exon 6 of the RET proto-oncogene. Methods: Targeted sequencing was used to diagnose four apparently sporadic MTC index cases carrying the germline RET S409Y (c.1226 C>A) variant. Subsequently, 27 relatives of these individuals underwent clinical and genetic assessments and/or thyroid surgery. Furthermore, in silico analyses and in vitro assays were performed to predict or verify the potential oncogenic activity of the S409Y variant. Results: Overall, 15 of 31 participants were found to carry the RET S409Y variant. Of these, 6 presented with isolated MTC (mean age 50.2 years; range 41-75 years), of which 3 presented with neck lymph node metastases and 2 presented with distant liver or lung metastases. Among the remaining 9 carriers, 3 (mean age 56 years; range 41-76 years) had elevated serum calcium-stimulated calcitonin (sCtn) or concurrent marginally elevated serum calcitonin (Ctn) levels, whereas the other 6 (mean age 37.5 years; range 14-52 years) exhibited typical Ctn/sCtn levels (p < 0.05). None of the 15 carriers in these 4 families presented clinical evidence of pheochromocytoma, hyperparathyroidism, or Hirschsprung's disease. In silico analyses revealed that S409Y was a "possibly damaging" mutation that could affect the RET protein inter-domain interface. An in vitro assay revealed that the phosphorylation level of RET tyrosine 905 was relatively higher in the RET S409Y mutant than in wild-type (WT) RET. Moreover, transfection of HEK 293 cells with S409Y enhanced the phosphorylation activity of AKT, ERK pathways, and it increased cell proliferation compared with WT RET, but to a lesser degree than that for the RET C618Y and C634Y mutations. Conclusions: This study demonstrates that the novel germline RET S409Y variant is likely pathogenic and is associated with lower penetrance of MTC than that for the C618Y and C634Y mutations. Individuals with S409Y should be managed using a personalized approach, and additionally, "at-risk" family members should be evaluated. Additional studies are needed to elucidate the correlation between the S409Y mutation and multiple endocrine neoplasia type 2-specific tumors.

The nuclear localization signals (NLS) were usually composed of basic residues (K and R) and played an important role in delivery of genomes and structural protein into nucleus. In this research, we identified that 3Dpol/3CD entered into nucleus during viral propagation of duck hepatitis A virus type 1 (DHAV-1). To investigate the reason that 3Dpol/3CD entered into nucleus, the amino acid sequence of 3CD was analyzed through NLS Mapper program. The basic region 17PRKTAYMRS25 was subsequently proved to be a functional NLS to guide 3Dpol/3CD into nucleus. 18R, 19K and 24R were found essential for maintaining the nuclear targeting activity, and exchange between 24R and 24K had no impact on cellular localization of 3Dpol. Since the entry of 3Dpol/3CD into nucleus was essential for shutoff of host cell transcription and maintaining the viral propagation of picornavirus numbers, our study provided new insights into the mechanism of DHAV-1 propagation.

A ferroelectric with a high phase-transition temperature (Tc ) is an indispensable condition for practical applications. Over the past decades, both strain engineering and the isotope effect have been found to effectively improve the Tc within ferroelectric material systems. However, the former strategy seems to prefer working in inorganic ferroelectric thin films, while the latter is also limited to some certain systems, such as hydrogen-bonded ferroelectrics. It is noted that a mono-fluorinated molecule is geometrically very similar to its parent molecule and the substitution of H by an F atom can introduce a chiral center on the molecule to template or stabilize polar structures. Significantly, the barrier of rotation of the fluorinated organic molecules is raised, resulting in a remarkable increase in Tc . Herein, by applying the molecular design strategy of H/F substitution to the organic-inorganic perovskite ferroelectric (pyrrolidinium)CdCl3 with a low Tc of 240 K, two high-Tc chiral perovskite ferroelectrics, (R)- and (S)-3-F-(pyrrolidinium)CdCl3 are successfully synthesized, for which the Tc reaches 303 K. The significant enhancement of 63 K in Tc extends the ferroelectric working temperature range to room temperature. This finding provides a new effective way to regulate the Tc in ferroelectrics and to design high-Tc molecular ferroelectrics.

Plantago asiatica L. seeds is a common folk medicine with a long history of medical use in China because of its antipyretic, diuretic, and expectorant properties. It has been applied to treat hypertension clinically due to its diuresis, however, its efficacy and mechanisms on anti-hypertension has not been reported yet to our knowledge. In this study, we investigated the antihypertensive effect and underlying mechanisms of P. asiatica L. seeds extract (PASE) in spontaneously hypertensive rat (SHR). Male SHRs were treated with 2.5 mg/kg of fosinopril (FOS) and 400 mg/kg of PASE orally per day for once or 12 weeks. SHR or Wistar-Kyoto rats (WKY) receiving vehicle (distilled water) was used as control. The results demonstrated systolic, diastolic, and mean blood pressures (SBP, DBP, and MBP) were significantly lowered after single and long-term intragastric administration of PASE. The cardiac and aortic index and collagen accumulation were improved in the PASE group compared with the SHRs group. Meanwhile, PASE treatment remarkably reduced urine total protein, the ratio of serum urea nitrogen to serum creatinine, and increased serum potassium. The levels of serum angiotensin I (Ang I), angiotensin II (Ang II), the ratio of Ang II to Ang I, and aldosterone (ALD) were lowered after treatment of PASE. Besides, PASE and its major active constituents of phenylethanoid glycosides, including isoacteoside, plantamajoside and acteoside, were found to effectively inhibit angiotensin-converting enzyme (ACE) activation in vitro. These findings suggest that PASE has the antihypertensive effect that may involve a mechanism of ACE inhibition and simultaneously protect organ damage against hypertension.

In order to solve the problem of odor pollution in drinking water effectively, the typical odorants 2-methylisoborneol (2-MIB) and Geosmin (GSM) were selected as target pollutants. The degradation effect and factors influencing these two typical odors by ultrasonically (US) activated persulfate (PS) advanced oxidation technology were systematically investigated in this work. When the sample was treated for 15 min, the application of US/PS combination technology increased the removal rates of 2-MIB and GSM by 57.0% and 63.6%, respectively, compared with sonication alone. Furthermore, the optimum removal rates of 2-MIB and GSM were 88.7% and 93.3% when the 2-MIB and GSM concentrations were 100 ng·L-1. Moreover, the degradation rates of 2-MIB and GSM increased with increasing PS concentration (from 0.25 mmol·L-1 to 2 mmol·L-1) and US intensity (from 0.33 W·cm-2 to 0.53 W·cm-2). The presence of humic acid competes to consume free radicals to inhibit the degradation of odor. The addition of the radical scavengers methanol and tert-butanol can reduce the removal rates of 2-MIB and GSM. The presence of methanol had a stronger inhibition effect on odor degradation than tert-butanol, indicating that the odor removal was mainly caused by sulfate radicals and hydroxyl radicals in the combination US/PS technology.

Three new isoquinoline alkaloids, including two aporphines (1 and 2) and one oxoaporphine (3), were isolated from the 90% EtOH extract of the whole plants of Thalictrum tenue Franch. The structures of the isolated compounds were established based on 1D and 2D (1H-1H COSY, HMQC, and HMBC) NMR spectroscopy, in addition to high resolution mass spectrometry, respectively. The isolated alkaloids were tested in vitro for cytotoxic activity against six esophageal carcinoma cell lines. Consequently, alkaloids 1-3 exhibited some cytotoxic activities against all the tested tumor cell lines with IC50 values less than 20 µM.

Transcatheter arterial chemoembolization (TACE) has become one of the first-line standard treatments for intermediate-advanced hepatocellular carcinoma, as well as an effective treatment for metastatic hepatic carcinoma. The majority of TACE-related complications are mild and acceptable to patients. Compared with conventional (C)-TACE, drug-eluting bead (DEB)-TACE allows permanent embolization of blood vessels, a slow continuous release of anti-tumour drugs in a locally targeted manner, and reduction of the systemic release of anti-tumour drugs, so that their adverse effects are significantly reduced. The general consensus is that DEB-TACE is safer and better tolerated by patients than C-TACE because serious complications after DEB-TACE are rarely reported. This current case report describes a rare case of diffuse biliary peritonitis secondary to rupture of a hepatic tumour after DEB-TACE. After the procedure, the patient presented with progressively worsening upper abdominal pain. As conventional management methods for the suspected tumour rupture failed, an emergency laparotomy was performed to remove the metastatic mass of differentiated hepatic adenocarcinoma. The patient remains under surveillance with no further complications. In our opinion, although DEB-TACE is safe and rarely has serious complications, caution should be exercised when this method is used to treat tumours that are located close to the liver surface.

A novel organic-inorganic ABX3 perovskite-type material with specific hydrogen bonding interactions, N,N-dimethylethanolammonium trichlorocadmate ([DMEA]CdCl3), has been synthesized as a phase transition material. It is notable that the DMEA cations are arranged to form one-dimensional chains connected by hydrogen bonds at room temperature, which are very sparse in other perovskite-type compounds. The strong intermolecular interactions have made the phase transition temperature of the material reach up to 429 K, as confirmed by differential scanning calorimetry measurements, variable-temperature structural analyses, and dielectric measurements. The origin of the symmetry-breaking phase transition is associated with the motion or reorientation of the DMEA cations, accompanied by the crystal structures from orthorhombic Pnma to monoclinic P21/c with the temperature decreases. The finding of [DMEA]CdCl3 with unprecedented hydrogen bonding interactions has opened a new avenue to design novel phase transition materials with higher transition temperatures.

OBJECTIVE: To investigate the correlation of E-cadherin expression level with the clinical characterastics in children with acute leukemia (AL), and to explore the possible regulatory mechanism. METHODS: Real-time quantitative RT-PCR was applied to detect the expression level of E-cadherin in bone marrow samples from 135 child patients diagnosed as AL, and its relevance with clinical indicators was statistically analyzed. The expression levels of E-cadherin, ß-catenin, and Akt/p-Akt were detected by using Western blot. The bone marrow samples from 22 children with non-malignant hematological diseases were used as controls. RESULTS: The expression level of E-cadherin significantly decreased in newly diagnosed patients with all 3 types of AL as compared with bone marrow samples from control group (P<0.01). In B-ALL group, compared with standard risk group, E-cadherin expression level significantly decreased in intermediate risk group (P<0.05). Moreover,the expression level of E-cadherin mRNA was also reduced in splenomegaly group (P<0.01). However, the correlation of E-cadherin level with clinical characteristics was not found in T-ALL and AML (P>0.05). The expression level of E-cadherin in the patients from Common-B-ALL group was higher than B-ALL patients with other immunophenotypes (P<0.01), while no significant difference was found among patients grouped by FAB classification. By the correlation analysis of measured data, lower E-cadherin expression level was found to be related with high WBC count and serum lactic dehydrogenase level (LDH) (r=-0.419, r=-0.269), but with low blood platelet count in B-ALL (r=0.335). In T-ALL, expression of E-cadherin was found to be negatively correlated with LDH and percentage of immature cells in the bone marrow (r=-0.567, r=-0.557). In addition, the lower expression of E-cadherin was also found to be related with WBC count and percentage of immature cells in the bone marrow in newly diagnosed AML patients (r=-0.368, r=-0.391). Compared with control group, the expression of E-cadherin was down-regulated significantly (P<0.01), while ß-catenin, Akt significantly was up-regulated in 3 types of AL patients (P<0.01). The expression of p-Akt and p-Akt/Akt was up-regulated significantly in T-ALL (P<0.01). CONCLUSION: Lower expression of E-cadherin is related factor of unfavourable prognosis in children with acute leukemia. The expression deficiency or down-regulation of E-cadherin may activate Wnt/ß-catenin and PI3K/ Akt signaling pathways to promote the genesis and progress of haematological malignancies, thus resulting in a series of malignant biological behaviors in cells. E-cadherin may be a new prognostic indicator for pediatric acute leukemia, thus to guide individualized hemotherapy.
