RESUMO

There has been an increasing interest in studying microbial necromasses and their contribution to soil organic carbon (SOC) accumulation. However, it remains unclear how the interaction among climate, plants, and soil influence the microbial anabolism and how microbial necromass contribute to SOC formation. Here, we assessed the relative contribution of microbial residues to SOC pool across a subtropical elevation gradient (ranged from 630 to 2130 m a.s.l.) representing a subtropical ecosystem on Wuyi Mountain in China, by using amino sugars as tracers. Analysis of topsoil (0-10 cm) amino sugars and the composition of microbial community across this gradient revealed that the soil total amino sugars accounting for 12.2-25.7% of the SOC pool, decreased with increasing elevation. Moreover, the linear reduction in the bacterial-derived carbon (C) and an increase in the ratio of fungal- to bacterial-derived C with increasing elevation suggested the reduction in the contribution of bacterial-derived C to SOC pool across this elevation gradient. The divergent changes in the contribution of the microbial residues to SOC infer a potential change in SOC composition and stability. The microbial-derived SOC formation and its climatic responses are influenced by the interaction of vegetation types and soil properties, with soil amorphous Fe being the determiner of soil amino sugar accrual. Our work highlights the importance of understanding ecosystem type and mineral composition in regulating microbial-mediated SOC formation and accumulation in responses to climate change in subtropical ecosystems.

Nylon 5T is one of the bio-based nylons, its raw material 1,5-pentanediamine is derived from biomass resources and produced by biological methods. 1,5-pentanediamine-terephthalate (PDA-TPA) is the monomeric salt for nylon 5T polymerization, and its own product quality has a significant impact on the performance of nylon 5T. PDA-TPA was prepared by anti-solvent crystallization in this study. It exists in two solid forms, a monohydrate [form (I)] and an anhydrous phase [form (II)]. The transition temperature of the two phases was around 65°C in the given ethanol-water binary (7:1) mixture. The characterization of monohydrate and anhydrate phases regarding structures and stabilities was carefully carried out using powder X-ray diffraction, single crystal X-ray diffraction, differential scanning calorimetry, thermogravimetric analysis, hot-stage microscopy and Fourier transform infrared spectroscopy. The relationship between the molecular interactions of monohydrate and anhydrate phases under different packing architectures and their thermal behaviours was analysed and established. In addition, the relationships between the structures and thermal behaviours for the two solid forms were analysed and established. In addition, the effect of solvent on phase conversion, the relationships between the temperature and water activity, as well as the relative stability of monohydrate and anhydrate phases under different thermodynamic conditions, were investigated by solid-solid transformation and solvent-mediated transformation experiments. It was obvious that the transition temperature of monohydrate and anhydrate phases of PDA-TPA was significantly influenced by water activity, and the larger the value of water activity is, the higher is the transition temperature. These studies give insight into the transformation of nylon 5T monomer salt and contribute to the control of target crystal preparation.

The blood vessel growth inhibitor bevacizumab targets vascular endothelial growth factor (VEGF), a crucial regulator of angiogenesis. Recently, small extracellular vesicles (sEVs) have been demonstrated to be important vehicles in the transport of growth factors to target cells. In this study, we isolated primary carcinoma-associated fibroblasts (CAFs) from four human oral squamous cell carcinoma (OSCC) specimens. Compared with other non-extracellular vesicle components, CAF-derived sEVs were found to be the main regulators of angiogenesis. The ability of CAF sEVs to activate VEGF receptor 2 (VEGFR2) signaling in human umbilical vein endothelial cells (HUVEC) was dependent on the association between sEVs and VEGF. In addition, sEV-bound VEGF secreted by CAFs further activated VEGFR2 signaling in HUVEC in a bevacizumab-resistant manner. VEGF was found to interact with heparan sulfate proteoglycans on the CAF sEV surface and could be released by heparinase I/III. The bioactivity of the dissociated VEGF was retained in vitro and in vivo and could be neutralized by bevacizumab. These findings suggest that the combined use of heparinase and bevacizumab might inhibit angiogenesis in patients with high levels of sEV-bound VEGF.

ß-d-glucan is a natural non-digestible polysaccharide that can be selectively recognized by recognition receptors such as Dectin-1 receptors, resulting in an emerging interest on exploring its capacity for carrying biological information to desired organs or cells. CpG oligodeoxynucleotide (ODN) has the potentiality to initiate an immune-stimulatory cascade via activating B cells inducing proinflammatory cytokines, which is conducive to immunotherapy and nucleic acid vaccine. Herein, we developed a pH-sensitive delivery system loading with CpG ODN by introducing poly-ethylenimine (PEI) to a hyperbranched ß-d-glucan (HBB) and coating with poly-ethylene glycol (PEG) shell via acidic liable Schiff bond. This delivery system exhibited a favorable biocompatibility and facilitated the cellular uptake of CpG ODN at pH 6.8 with the possibility of having higher accumulation in acidic cancer microenvironment. Furthermore, this carrier together with class B CpG ODN could enhance the secretion of cytokines including interleukin-6 and interferon-α as well as capable of interferon-α induction.

Early detection has the potential to reduce cancer mortality, but an effective screening test must demonstrate asymptomatic cancer detection years before conventional diagnosis in a longitudinal study. In the Taizhou Longitudinal Study (TZL), 123,115 healthy subjects provided plasma samples for long-term storage and were then monitored for cancer occurrence. Here we report the preliminary results of PanSeer, a noninvasive blood test based on circulating tumor DNA methylation, on TZL plasma samples from 605 asymptomatic individuals, 191 of whom were later diagnosed with stomach, esophageal, colorectal, lung or liver cancer within four years of blood draw. We also assay plasma samples from an additional 223 cancer patients, plus 200 primary tumor and normal tissues. We show that PanSeer detects five common types of cancer in 88% (95% CI: 80-93%) of post-diagnosis patients with a specificity of 96% (95% CI: 93-98%), We also demonstrate that PanSeer detects cancer in 95% (95% CI: 89-98%) of asymptomatic individuals who were later diagnosed, though future longitudinal studies are required to confirm this result. These results demonstrate that cancer can be non-invasively detected up to four years before current standard of care.

PURPOSE: Primary ciliary dyskinesia (PCD), which commonly causes male infertility, is an inherited autosomal recessive disorder. This study aimed to investigate the clinical manifestations and screen mutations associated with the dynein axonemal assembly factor 2 (DNAAF2) gene in a Han Chinese family with PCD. METHODS: A three-generation family with PCD was recruited in this study. Eight family members underwent comprehensive medical examinations. Genomic DNA was extracted from the participants' peripheral blood, and targeted next-generation sequencing technology was used to perform the mutation screening. The DNAAF2 expression was analyzed by immunostaining and Western blot. RESULTS: The proband exhibited the typical clinical features of PCD. Spermatozoa from the proband showed complete immotility but relatively high viability. Two novel compound heterozygous mutations in the DNAAF2 gene, c.C156A [p.Y52X] and c.C26A [p.S9X], were identified. Both nonsense mutations were detected in the proband, whereas the other unaffected family members carried either none or only one of the two mutations. The two nonsense heterozygous mutations were not detected in the 600 ethnically matched normal controls or in the Genome Aggregation Database. The defect of the DNAAF2 and the outer dynein arms and inner dynein arms were notably observed in the spermatozoa from the proband by immunostaining. CONCLUSION: This study identified two novel compound heterozygous mutations of DNAAF2 leading to male infertility as a result of PCD in a Han Chinese family. The findings may enhance the understanding of the pathogenesis of PCD and improve reproductive genetic counseling in China.

Generation of hydroxyl radicals in the Fenton system (FeII /H2 O2 ) is seriously limited by the sluggish kinetics of FeIII reduction and fast FeIII precipitation. Here, boron crystals (C-Boron) remarkably accelerate the FeIII /FeII circulation in Fenton-like systems (C-Boron/FeIII /H2 O2 ) to produce a myriad of hydroxyl radicals with excellent efficiencies in oxidative degradation of various pollutants. The surface B-B bonds and interfacial suboxide boron in the surface B12 icosahedra are the active sites to donate electrons to promote fast FeIII reduction to FeII and further enhance hydroxyl radical production via Fenton chemistry. The C-Boron/FeIII /H2 O2 system outperforms the benchmark Fenton (FeII /H2 O2 ) and FeIII -based sulfate radical systems. The reactivity and stability of crystalline boron is much higher than the popular molecular reducing agents, nanocarbons, and other metal/metal-free nanomaterials.

RATIONALE: The orexin projection system includes the lateral hypothalamus, reticular activating structure, and ventrolateral preoptic nucleus, and this system is related to the pathogenesis of narcolepsy. Here, we report a case of narcolepsy type 2 caused by hippocampal glioma of the right temporal lobe. PATIENT CONCERNS: A 44-year-old male farmer complained of excessive daytime sleepiness (EDS) over the past 3 months and more. INTERVENTIONS: The lesion of the right anteromedial temporal lobe was removed and its pathological examination was carried out. OUTCOMES: General examination showed no abnormalities of his heart, lungs, or abdomen. Neurological examination showed no positive sign. The blood routine and biochemical examination were normal. He scored 7 on the Pittsburg sleep quality index, 16 on the Epworth sleepiness scale, 52 on the self-rating anxiety scale, and 48 on the self-rating depression scale. The multiple sleep latency test data showed 2 periods of sleep-onset rapid eyes movement period across 4 successive tests; the average sleep latency was under 8 minutes, and the rapid eyes movement latency was under 7 minutes. Lesion of glioma in hippocampus area of the right anteromedial temporal lobe was confirmed through magnetic resonance imaging, magnetic resonance spectroscopy, and histological examination. After surgical removal of the glioma from the hippocampus area of the right anteromedial temporal lobe, the patient's EDS symptoms disappeared immediately. He scored 3 on the Epworth sleepiness scale. During our follow-up three months later, he remained well with no complications. DIAGNOSIS: We diagnosed the patient with narcolepsy type 2 according to the 3rd Edition of International Classification of Sleep Disorders (ICSD-3). CONCLUSION: The patient suffered from EDS and was diagnosed with narcolepsy type 2. The narcolepsy type 2 was linked to glioma of the hippocampus area. The hippocampus might be another part of regulating the sleep-arousal pathway, and the glioma secretion might interact with the orexin projection system.

BACKGROUND: The long-term renal outcome for IgA nephropathy (IgAN) in large cohorts of children remains unclear. IgAN is a progressive disease, to explore novel biomarkers is necessary for predicting the disease activity and progression of IgAN. In addition, there is a hot debate on when to treat with immunosuppression in children. We aimed to confirm the long-term renal survival, find some undetected risk factors and investigate when to treat with immunosuppression can benefit for renal outcome in Chinese children. METHODS: 1243 Children with IgAN were enrolled and a follow-up of at least 1 year after a biopsy from 2000 to 2017. Long-term renal survival, undetected risk factors and the renal survival of immunosuppressive and non-immunosuppressive therapy were evaluated. The primary endpoint of the study was a combined outcome of either ≥50% reduction in estimated glomerular filtration rate (eGFR) or end-stage renal disease (ESRD) or death. RESULTS: The median follow-up time were 86.8 months (interquartile range 54.7-140.2 months). The 5-, 10- and 15-year renal survival rates were 95.3%, 90.3% and 84%, respectively. Cox multivariate regression and Kaplan-Meier analysis showed that hypertension, hyperuricemia, high 24 h urine protein (24 h-UP) levels, lower initial eGFR, high urine C3 levels, high retinol-binding protein (RBP) levels, segmental glomerulosclerosis (S) and tubular atrophy and interstitial fibrosis (T) were associated with renal outcome. The statistically significant predictive perfect power for renal outcome was RBP ≥ 0.7µg/ml (AUC = 0.899, sensitivity = 84.00%, specificity = 86.00%), 24 h-UP ≥ 1 g/24 h (AUC = 0.722, sensitivity = 84.20%, specificity = 52.70%), eGFR < 60 ml/min/1.73 m2 (AUC = 0.718, sensitivity = 81.30%, specificity = 39.20%) and S1 lesion (AUC = 0.703, sensitivity = 75.50%, specificity = 65.10%).Children with urinary RBP ≥ 0.7µg/ml were associated with a 2.513-fold risk than patients with urinary RBP < 0.7µg/ml (P = 0.003). Our study suggested that immunosuppressive therapy may reduce the risk of progression in IgAN children had both eGFR > 50 ml/min/1.73 m2 and proteinuria of at least 1 g/day. CONCLUSIONS: This is the first report that the 15-year renal survival rate of children with IgAN in China was 84%. At the same time, this is the first study to reveal that urinary RBP ≥ 0.7µg/ml may indicate a poor renal outcome. In addition, this study supports immunosuppressive therapy for IgAN children had both proteinuria ≥1 g/day and initial eGFR > 50 ml/min/1.73m2.

METHOD: The data of 32 hemodialysis patients with COVID-19, including epidemiological, demographic, clinical, laboratory, and radiological, were collected from the Blood Purification Department of Wuhan Fourth Hospital from February 3 to 16, 2020. RESULTS: Of the 32 patients, 23 were male, and the median age was 58 years; the median dialysis vintage was 33 months. Two groups were divided according to the patient's primary renal disease: group 1 (16 patients with diabetic nephropathy), group 2 (12 patients with primary glomerulonephritis, 2 with obstructive kidney disease, 1 with hypertensive renal damage, and 1 with polycystic kidney). No significant differences were observed between the two groups in epidemiological characteristics, blood cell counts, and radiological performance. Hemodialysis patients are susceptible to COVID-19 at all ages, and patients with diabetes may be a high-risk population (50%). Common symptoms included fever (15 cases), cough (21 cases), and fatigue (7 cases). The blood lymphocyte count decreased in 84.6% of the patients (median: 0.765 × 109/L). Chest CT revealed ground-glass-like lesions in 18 cases, unilateral lung patchiness in 7 cases, bilateral lung patchiness in 7 cases, and pleural effusion in 2 cases. CONCLUSION: Only 46.875% of the hemodialysis patients with COVID-19 had fever in the early stage; and diabetics may be the most susceptible population. A decrease in blood lymphocyte count and ground-glass opacity on chest CT scan is beneficial in identifying the high-risk population.

Signal regulatory protein α (SIRPα), a transmembrane protein that is predominantly expressed in dendritic cells (DCs) or macrophages, interacts with CD47 that is overexpressed in almost all types of tumor cells. The interaction between SIRPα and CD47 leads to a negative signal that prevents the phenotypic and functional maturation of DC and inhibits phagocytosis. The SIRPα knockdown in DCs that were pulsed with a modified HPV16E7 (HPV16mE7) protein with enhanced antigenicity and reduced transformation activity results in increased cytokine (TNF-α/IL-12/IL-6) secretion, IFN-γ secretion by T lymphocytes, and in vitro/in vivo tumoricidal activity against cervical cancer cells. Taken together, these results suggest that SIRPα-silenced DC vaccination presented potential therapeutic implications against cervical cancer.

Saccharina japonica is a brown macroalga that has been commercially cultivated in China for almost a century. As a natural raw material, it is widely used in the food and pharmaceutical industries, and it may potentially be useful for biofuel production. However, little is known about the genes involved in carbohydrate biosynthesis, and their regulation is less understood. In this study, the analysis of growth traits and alginate and mannitol contents suggested that sporophyte development could be divided into four stages. Accordingly, we performed transcriptome analysis of the S. japonica sporophyte. In total, 589 million clean reads were generated, and 4,514 novel genes were identified. Gene expression analysis revealed that 2,542 genes were differentially expressed. Kyoto Encyclopedia of Genes and Genomes pathway enrichment analysis indicated that these genes were significantly enriched in "Carbon metabolism," "Photosynthesis," and "Photosynthesis-antenna proteins" pathways, which are important for metabolism of various carbohydrates during sporophyte development. Systematic analysis identified the genes encoding enzymes for the biosynthesis of cell wall carbohydrates (including alginate, fucoidan, and cellulose) and cytoplasm storage carbohydrates (mannitol, laminarin, and trehalose). Among them, some key genes associated with carbohydrate content were further identified based on detailed expression profiling, representing good candidates for further functional studies. This study provides a global view of the carbohydrate metabolism process and an important resource for functional genomics studies in S. japonica. The results obtained lay the basis for elucidating the molecular mechanism of carbohydrate biosynthesis and for genetic breeding of carbohydrates-related traits in kelp.

To improve lipid and astaxanthin productivity without compromising biomass during the whole cultivation period, carbon-dependent kinetics involving nitrogen stress was applied under excess light to elevate intracellular carbon availability and metabolic activity of Chromochloris zofingiensis. Results suggested that fed-batch strategy proposed could increase lipid and astaxanthin productivity to 457.1 and 2.0 mg L-1 d-1, respectively. Biomass productivity at 1084.3 mg L-1 d-1 was comparable with that under suitable condition. Then 13C tracer-based metabolic flux analysis (13C-MFA) demonstrated that central carbon metabolism provided ATP, NADPH and carbon availability for lipid biosynthesis during the strategy. In combination with targeted metabolite analysis, 13C-MFA revealed that the strategy improved precursor content for lipid biosynthesis and elevated path rate to synthesize C16:0 and C18:0. The enhanced lipid content potentially accounted for the high biomass productivity. Therefore, comprehensively understanding relationships between carbon availability and carbon conversion could precisely design strategy for productivity improvements during cultivation.

Accumulation of high-value products in microalgae is not conducive with rapid cell growth, which is the potential conflict in microalgal production. Overcoming such conflict faces numerous challenges in comprehensively understanding cell behavior and metabolism. Here, we show a fully integrated interaction between cell behavior, carbon partitioning, carbon availability and path rate of central carbon metabolism, and have practically overcome the production conflict of Chromochloris zofingiensis. We demonstrate that elevated carbon availability and active path rate of precursors are determinants for product biosynthesis, and the former exhibits a superior potential. As protein content reaches a threshold value to confer survival advantages, carbon availability becomes the major limiting factor for product biosynthesis and cell reproduction. Based on integrated interaction, regulating the C/N balance by feeding carbon source under excess light increases content of high-value products without inhibiting cell growth. Our findings provide a new orientation to achieve great productivity improvements in microalgal production.

With the integration of the global economy and increased international trade, CO2 emissions embodied in trade have attracted broad attention. In this study, we examined Taiwan's CO2 emissions embodied in its imports and exports with its 167 trade partners from 1970 to 2015 using the emissions embodied in bilateral trade (EEBT) method. The results show that since the mid-1990s, Taiwan has shifted to being a net CO2 emissions exporter. The share of Taiwan's CO2 emissions exported to developed countries has decreased. However, the share exported to Mainland China, Hong Kong special administrative region and many developing countries in Southeast Asia has increased. The results of a structural decomposition analysis show that emissions intensity changes were the main driving force for Taiwan's exported CO2 emissions during 1970-1996, while increased export volumes were the largest driving force for Taiwan's exported CO2 emissions during 1996-2015. Emissions intensity changes and population growth are the main driving factors for Taiwan's CO2 emissions produced and consumed by itself increases. Based on the SDA results, environmentally friendly policies, such as energy efficiency and industrial structure optimization, are suggested to help Taiwan shift towards a sustainable economy.

Periodontal ligament stem cells (PDLSCs) from beagle dogs had the characteristics of multi-directional differentiation and had great application potential in tissue engineering and cell regenerative medicine. In this study, we analysed the odontogenesis and neuronal differentiation characteristics of PDLSCs in vitro. Results showed that the calcined tooth powder (CTP) and silver nanoparticles (AgNPs) additives could induce the PDLSCs into odontogenesis differentiation; besides, the immunofluorescence staining identified that the high dosage calcined tooth powder (400 µg/mL) significantly facilitated the odontogenesis associated with BMP4 expression. While the nutritional factor (L-glutamine, NGF (nerve growth factor), bFGF (basic fibroblast growth factor), IGF-1 (insulin-like growth factor-1) and EGF (epidermal growth factor)) additives were prior to induce the PDLSCs into neuronal differentiation. Simultaneously, PDLSCs had high proliferation ability with the different supplemented additives. Importantly, the Western blot results also proved the BMP4 and SMAD1 proteins were highly expressed in the induced odontoblast, while the SOX1, NCAM1, GFAP and VEGFA proteins were all obviously expressed in the induced neurons. Hence, PDLSCs had characteristics of both odontogenesis and neuronal differentiation.

BACKGROUND: This study was aimed to investigate the regulatory role of microRNA-210 (miRNA-210) on the progression of liver cancer and Hepatitis B virus (HBV)-associated liver cancer. METHODS: The expression of miRNA-210 was detected in liver tissues of HBV-associated cirrhosis and liver cancer, and in HepG2 and HepG2.2.15 cells by qRT-PCR. MiRNA-210 was silenced in HepG2 and HepG2.2.15 cells by the transfection of miRNA-210 inhibitor. The cell viability and apoptosis was detected by MTT assay and Annexin V-fluorescein isothiocyanate/propidium iodide staining, respectively. The protein expression of EGR3 was detected by Western blot. The regulatory relationship between EGR3 and miRNA-210 was predicted by TargetScan and identified by Dual luciferase reporter gene assay. RESULTS: MiRNA-210 was overexpressed in the liver tissues of HBV-associated cirrhosis and liver cancer, and in HepG2 and HepG2.2.15 cells (P < 0.05). Silencing of miRNA-210 inhibited the viability and promoted the apoptosis of HepG2 and HepG2.2.15 cells (P < 0.05). EGR3 was a target of miRNA-210, which was down-regulated in the liver tissues of HBV-associated cirrhosis and liver cancer, and in HepG2 and HepG2.2.15 cells (P < 0.05). Silencing of miRNA-210 increased the mRNA and protein expression of EGR3 (P < 0.05). Silencing of EGR3 reversed the anti-tumor effect of miRNA-210 inhibitor on HepG2 and HepG2.2.15 cells (P < 0.05). CONCLUSIONS: Silencing of miRNA-210 inhibits the progression of liver cancer and HBV-associated liver cancer via up-regulating EGR3.

As a hazardous waste, the disposal of chromium enriched tannery sludge has attracted increasing public concern due to its potential adverse risks towards the environment. And incineration is considered to be an effective method to stabilize heavy metals, like Cr, in solid phase during tannery sludge treatment. In this study, real chromium enriched tannery sludge without pre-treatment was incinerated at 300°C-1200 °C under air atmosphere to investigate the transformation of chromium speciation. Here detailed thermal behavior, phase transformation and chromium speciation were characterized by TG-DSC, XRD and XPS, respectively. Experimental results show that content of Cr(VI) increases gradually with the increase of temperature from 300 °C to 500 °C and reaches a maximal level of 46% total Cr at 500 °C, with different Cr(VI) species of CaCrO4, MgCrO4 and Cr5O12. However, the content of Cr(VI) decreases gradually with the further increase of temperature, with only about 5% Cr(VI) at high temperature of 1200 °C, due to formation of Cr(III) species of Cr2O3 crystallite and MgCr2O4 spinel. Besides, a growing number of hexagonally shaped flake-like crystallite Cr2O3 can be discovered from characterization results of XRD and SEM. Finally, the reduction of CaCrO4 to Cr2O3 in the presence of SiO2 is thermodynamically feasible over 700 °C, indicating possible transformation of Cr(VI) to Cr(III) through controlled incineration temperature.

PURPOSE: This research aimed to explore the correlation between miR-34a expression in peripheral blood and clinical characteristics of patients with chronic hepatitis C (CHC) as well as the diagnostic and prognostic values of serum miR-34a in CHC. METHODS: Serum samples of 41 CHC patients and 18 normal participants were collected to examine the expression levels of miR-34a using qRT-PCR. The changes of serum TBA, liver enzyme AST and ALT were also determined by enzyme colorimetry and rate method. The levels of serum fibrotic markers hyaluronic acid (HA), type III procollagen (PCIII), type IV collagen (IV-C) and laminin (LN) were detected by radioimmunoassay. Degree of liver fibrosis was examined by liver biopsy. Western blot analysis was used to investigate the expression of ac-p53, p53 and Sirt1 in the liver tissues of CHC patients. RESULTS: MiR-34a was significantly increased in the serum of CHC patients than that in healthy participants, and serum miR-34a was correlated with liver fibrosis index. Serum TBA, AST and ALT levels, and AST/ALT ratios in patients with CHC were increased with increasing degree of fibrosis, and were positively associated with serum miR-34a. Furthermore, the liver tissues of CHC patients showed low Sirt1 protein expression and highly ac-p53 protein expression. CONCLUSIONS: Serum miR-34a in patients with CHC could promote liver fibrosis through mediating the Sirt1/p53 pathway and might function as pivotal biomarker on the prognosis and diagnosis of CHC patients.
