RESUMO

Notch signaling and epigenetic factors are known to play critical roles in regulating tissue homeostasis in most multicellular organisms, but how Notch signaling coordinates with epigenetic modulators to control differentiation remains poorly understood. Here, we identify heterochromatin protein 1c (HP1c) as an essential epigenetic regulator of gut homeostasis in Drosophila. Specifically, we observe that HP1c loss-of-function phenotypes resemble those observed after Notch signaling perturbation and that HP1c interacts genetically with components of the Notch pathway. HP1c represses the transcription of Notch target genes by directly interacting with Suppressor of Hairless (Su(H)), the key transcription factor of Notch signaling. Moreover, phenotypes caused by depletion of HP1c in Drosophila can be rescued by expressing human HP1γ, suggesting that HP1γ functions similar to HP1c in Drosophila. Taken together, our findings reveal an essential role of HP1c in normal development and gut homeostasis by suppressing Notch signaling.

Mycotoxins are a great potential threat to human health, and the progress in the development of mycotoxin detection methods is of an escalating importance with the increasing emphasis on food safety. Aptamer, performing the same function as antibody in specific binding with targets, exhibits profound potential in biosensing since its debut in 1990. Recent years have witnessed the rapid development of aptasensors for mycotoxin detection with the achievement of ultralow limit of detection and high sensitivity in the lab. However, there is still no officially approved aptasensing methods in mycotoxin detection application. In order to provide researchers with inspirations in the design and development of aptasensors for mycotoxin detection, we divide these aptasensors into two types, namely "on the surface" and "in the colloid", according to the location where the key sensing reaction occurs. We also systematically review aptasensors reported in the past 5 years under the abovementioned criterion of classification, and compare the advantages and disadvantages of each kind of aptasensors. Finally, we discuss prospective directions in the development of aptasensors for mycotoxin detection. This paper will offer insight and motivation to practitioners working on the research and practical application of aptasensors in the detection of mycotoxins and other substances.

The residue of phthalate acid esters (PAEs) in plastic film greenhouses had become a global concern environmental pollution problem. However, few studies have focused on the occurrence and fate of PAEs in the soil-air interface of the greenhouses. In this study, the occurrence, health risks, and soil-air exchange of PAEs from ten soil samples and four air samples of different greenhouses were investigated by a case study. The concentrations of total PAEs and individual PAEs congeners between two seasons were significantly different in the greenhouse soil and atmosphere. Di-(2-ethylhexyl) phthalate (1.04 mg kg-1) and diisobutyl phthalate (0.16 mg kg-1) were the predominant PAE congeners of soil in spring and autumn, respectively. Di-(2-ethylhexyl) phthalate was the major PAE congeners in the greenhouse atmosphere both of spring (0.03 µg m-3) and autumn (1.32 µg m-3). Health risks assessment showed that the predominant exposure route for adults was dietary intake, and the total carcinogenic risk level of PAEs was acceptable. Besides, fugacity model analysis indicated that di-(2-ethylhexyl) phthalate tended to deposit from air to soil with mean deposition flux of 175.1 kg (h⋅km2)-1 in autumn and 11.9 kg (h⋅km2)-1 in spring. On the contrary, diethyl phthalate escaped from soil with mean volatilization flux of 0.005 kg (h⋅km2)-1 in autumn and 0.025 kg (h⋅km2)-1 in spring. Other PAE congeners have tried to establish an equilibrium status through recycling continuously between the soil and air, and the source-sink relationships depended on their concentrations and hydrophobicity. This study showed that the distribution and movement of PAEs in the soil-air interface might be principally caused by temperature and their chemical properties.

Interferon (IFN)-inducible 44 like (IFI44L) is an IFN-stimulated gene (ISG), which is located on the same chromosome as the known antiviral ISG IFI44. Expression of IFI44L is induced by IFN and HIV-1 infection. However, the mechanism by which IFN-I induces IFI44L production has not yet been determined. In this study, we analyzed transcriptional regulation of IFI44L via cloning of the IFI44L promoter. We found that IFI44L has two IFN-stimulated response elements (ISRE), which are necessary for the basal level of IFI44L transcription. IFN-I and IFN-II can activate the IFI44L promoter through one of the two ISREs. IFN regulatory factor (IRF)-1 can activate transcription of IFI44L by binding to one of the ISREs. Additionally, co-transfection of the IFI44L promoter with an HIV-1 infectious clone or HIV-1 infection activated IFI44L promoter transcription, but did not upregulate IFI44L expression via ISREs. These findings will help to understand the interaction between IFI44L and HIV-1, and aid in elucidation of the role of IFI44L in the antiviral innate immune response.

The flySAM/CRISPRa system has recently emerged as a powerful tool for gain-of-function studies in Drosophila melanogaster This system includes Gal4/UAS-driven dCas9 activators and U6 promoter-controlled sgRNA. Having established dCas9 activators superior to other combinations, to further enhance the efficiency of the targeting activators we systematically optimized the parameters of the sgRNA. Interestingly, the most efficient sgRNAs were found to accumulate in the region from -150bp to -450bp upstream of the transcription start site (TSS), and the activation efficiency showed a strong positive correlation with the GC content of the sgRNA targeting sequence. In addition, the target region is dominant to the GC content, as sgRNAs targeting areas beyond -600bp from the TSS lose efficiency even when containing 75% GC. Surprisingly, when comparing the activities of sgRNAs targeting to either DNA strand, sgRNAs targeting to the non-template strand outperform those complementary to the template strand, both in cells and in vivo In summary, we define criteria for sgRNA design which will greatly facilitate the application of CRISPRa in gain-of-function studies.

BACKGROUND: Diet affects the human gastrointestinal microbiota. Blood and urine samples have been used to determine nutritional biomarkers. However, there is a dearth of knowledge on the utility of fecal biomarkers, including microbes, as biomarkers of food intake. OBJECTIVES: This study aimed to identify a compact set of fecal microbial biomarkers of food intake with high predictive accuracy. METHODS: Data were aggregated from 5 controlled feeding studies in metabolically healthy adults (n = 285; 21-75 y; BMI 19-59 kg/m2; 340 data observations) that studied the impact of specific foods (almonds, avocados, broccoli, walnuts, and whole-grain barley and whole-grain oats) on the human gastrointestinal microbiota. Fecal DNA was sequenced using 16S ribosomal RNA gene sequencing. Marginal screening was performed on all species-level taxa to examine the differences between the 6 foods and their respective controls. The top 20 species were selected and pooled together to predict study food consumption using a random forest model and out-of-bag estimation. The number of taxa was further decreased based on variable importance scores to determine the most compact, yet accurate feature set. RESULTS: Using the change in relative abundance of the 22 taxa remaining after feature selection, the overall model classification accuracy of all 6 foods was 70%. Collapsing barley and oats into 1 grains category increased the model accuracy to 77% with 23 unique taxa. Overall model accuracy was 85% using 15 unique taxa when classifying almonds (76% accurate), avocados (88% accurate), walnuts (72% accurate), and whole grains (96% accurate). Additional statistical validation was conducted to confirm that the model was predictive of specific food intake and not the studies themselves. CONCLUSIONS: Food consumption by healthy adults can be predicted using fecal bacteria as biomarkers. The fecal microbiota may provide useful fidelity measures to ascertain nutrition study compliance.

Intense particle beams generated from the interaction of ultrahigh intensity lasers with sample foils provide options in radiography, high-yield neutron sources, high-energy-density-matter generation, and ion fast ignition. An accurate understanding of beam transportation behavior in dense matter is crucial for all these applications. Here we report the experimental evidence on one order of magnitude enhancement of intense laser-accelerated proton beam stopping in dense ionized matter, in comparison with the current-widely used models describing individual ion stopping in matter. Supported by particle-in-cell (PIC) simulations, we attribute the enhancement to the strong decelerating electric field approaching 1 GV/m that can be created by the beam-driven return current. This collective effect plays the dominant role in the stopping of laser-accelerated intense proton beams in dense ionized matter. This finding is essential for the optimum design of ion driven fast ignition and inertial confinement fusion.

Cognitive dysfunction caused by chronic cerebral hypoperfusion is a common underlying cause of many cognition-related neurodegenerative diseases. The mechanisms of cognitive dysfunction caused by CCH are not clear. Long non-coding RNA is involved in synaptic plasticity and cognitive function, but whether lncRNA is involved in cognitive dysfunction caused by CCH has not yet been reported. In the present study, we identified the altered lncRNAs and mRNAs by deep RNA sequencing. A total of 128 mRNAs and 91 lncRNAs were up-regulated, and 108 mRNAs and 98 lncRNAs were down-regulated. Real-time reverse transcription-polymerase chain reaction verified the reliability of the lncRNA and mRNA sequencing. Gene Ontology and KEGG pathway analyses showed that differentially-expressed mRNAs were related to peptide antigen binding, the extracellular space, the monocarboxylic acid transport, and tryptophan metabolism. The co-expression analysis showed that 161 differentially expressed lncRNAs were correlated with DE mRNAs. By predicting the miRNA in which both DE lncRNAs and DE mRNAs bind together, we constructed a competitive endogenous RNA network. In this lncRNAs-miRNAs-mRNAs network, 559 lncRNA-miRNA-mRNA targeted pairs were identified, including 83 lncRNAs, 67 miRNAs, and 108 mRNAs. Through GO and KEGG pathway analysis, we further analyzed and predicted the regulatory function and potential mechanism of ceRNA network regulation. Our results are helpful for understanding the pathogenesis of cognitive dysfunction caused by CCH and provide direction for further research.

Nexus of runoff and sediment in watercourses is important for sustainable watershed management, especially in rivers with hyper-concentrated sediments. However, complex relationships between runoff and sediment were not fully understood. In this research, Mann-Kendall (MK) and moving t-test methods were used to confirm the suitable temporal range based on time series data of precipitation in the Anning River Basin from 1971 to 2017. At the same time, the month of typical precipitation (i.e., the largest annual average precipitation) was identified. A model of runoff and sediment over two stations from 2010 to 2015 were constructed respectively based on copula analysis method. Synchronous and asynchronous probabilities of runoff and sediment were analysed from the temporal and spatial aspects separately. The encounter probabilities of runoff and sediment over multiple periods and at two stations were analysed. The sediment distribution was illustrated under multiple runoff distribution conditions. The encounter probability of runoff and sediment distribution changed greatly, which was principally due to employment of reservoir operation, soil and water conservation measures. The variability of the joint probability was much obvious in tributary regions than that of the mainstream areas of Anning River basin. The results could be adopted to improve the understanding of interactive relationships between runoff and sediment and provide a significant references for regional management of soil and water.

Guanine-rich repeat sequences are known to adopt diverse G-quadruplex (G4) topologies. Determining the unfolding rates of individual G4 species is challenging due to the coexistence of multiple G4 conformations in a solution. Here, using single-molecule magnetic tweezers, we systematically measured the unfolding force distributions of 4 oncogene promoter G4s, 12 model sequences with two 1-nucleotide (nt) thymine loops that predominantly adopt parallel-stranded G4 structures, and 6 sequences forming multiple G4 structures. All parallel-stranded G4s reveal an unfolding force peak at 40-60 pN, which is associated with extremely slow unfolding rates on the order of 10-5-10-7 s-1. In contrast, nonparallel G4s and partially folded intermediate states reveal an unfolding force peak <40 pN. These results suggest a strong correlation between the parallel-stranded G4s folding topology and the slow unfolding rates and provide important insights into the mechanism that govern the stability and the transition kinetics of G4s.

This study investigated the effects of participants' sex and the facial trustworthiness of proposers on third-party decision-making. Sixty-three participants participated in this study. The participant's task was to act as an interest-free third party who observed a dictator game in which proposers with either trustworthy or untrustworthy faces made offers to recipients, to evaluate the reasonableness of the offers, and to express their intentions to punish the proposers. The results showed that offers from trustworthy proposers were rated as more reasonable than offers from untrustworthy proposers. Similarly, the participants were more likely to punish untrustworthy proposers than trustworthy proposers. In addition, there was a trend that male participants punished proposers more severely than female participants when proposers made unfair offers. These results demonstrate that an individual's facial trustworthiness affects other people's judgment and decisions.

INTRODUCTION: Autism spectrum disorder (ASD) is a complex and prevalent neurodevelopmental disorder characterized by deficits in social communication and social interaction as well as repetitive behaviors. Alterations in function connectivity are widely recognized in recent electroencephalogram (EEG) studies. However, most studies have not reached consistent conclusions, which could be due to the developmental nature and the heterogeneity of ASD. METHODS: Here, EEG coherence analysis was used in a cohort of children with ASD (n = 13) and matched typically developing controls (TD, n = 15) to examine the functional connectivity characteristics in long-distance and short-distance electrode pairs. Subsequently, we explore the association between the connectivity strength of coherence and symptom severity in children with ASD. RESULTS: Compared with TD group, individuals with ASD showed increased coherence in short-distance electrode pairs in the right temporal-parietal region (delta, alpha, beta bands), left temporal-parietal region (all frequency bands), occipital region (theta, alpha, beta bands), right central-parietal region (delta, alpha, beta bands), and the prefrontal region (only beta band). In the long-distance coherence analysis, the ASD group showed increased coherence in bilateral frontal region, temporal region, parietal region, and frontal-occipital region in alpha and beta bands. The strength of such connections was associated with symptom severity. DISCUSSION: Our study indicates that abnormal connectivity patterns in neuroelectrophysiology may be of critical importance to acknowledge the underlying brain mechanism.

Background: The treatment of ketamine-induced bladder contractures remains poorly studied. We therefore evaluated the efficacy of cystectasia with a sodium hyaluronate balanced solution in this kind of bladder contracture. Methods: Eighteen patients presenting with ketamine-induced bladder contracture between July 2010 and February 2018 were selected and analysed. Ketamine was discontinued in all patients, who were then treated with weekly cystectasia (0.09% sodium hyaluronate balanced solution) 3 times. The volume of the first perfusion was twice the preoperatively measured bladder capacity, and the volume of the subsequent two perfusions was increased by 100 mL each time. The Pelvic Pain and Urgency/Frequency (PUF) symptom score, O'Leary-Sant Interstitial Cystitis (IC) Symptom Index (ICSI), IC Problem Index (ICPI), Quality of Life (QOL) score, and bladder capacity were recorded before surgery and 3 and 12 months after the 3rd expansion. Results: No significant complications were observed during the 3 expansions. Fourteen patients completed the full follow-up schedule. Preoperatively and at the 3- and 12-month follow-up evaluations performed after the 3rd expansion, the PUF symptom scores were 20.4±3.6, 11.5±3.1, and 13.2±3.3, respectively; the mean ICSI was 13.6±2.8, 7.7±2.3, and 8.2±2.5, respectively; the mean ICPI was 10.6±2.6, 7.3±2.1, and 7.7±2.5, respectively; and the mean QOL scores were 6.0±0, 2.1±0.5, and 2.7±0.8, respectively; and the mean bladder catheter volume was 83±27, 234±56, and 228±52 mL, respectively. There were significant differences between all preoperative and postoperative values. Conclusions: Cystectasia with a sodium hyaluronate balanced solution is an effective treatment modality for ketamine-induced bladder contracture.

In the present study, the effects of Zn-3Cu-xFe (x = 0, 0.2, 0.5 wt%) alloys on endothelial cells (EA.hy926) and smooth muscle cells (A7r5), the hemocompatibility and antibacterial properties were also evaluated. The cell viability of EA.hy926 cells and A7r5 cells decreased with the increasing of extract concentration. At the same Zn2+ concentration (over 6 ppm), the cell viability of EA.hy926 cells increased with the addition of Cu or Cu and Fe content, but no significant effect on A7r5 cells was observed. The hemolysis rate of Zn-3Cu-xFe alloys samples was about 1%, and there was no adversely affected on platelets adhering to the surface of the Zn alloys. As Fe content increases in the Zn-Cu-Fe alloys, the antibacterial lower concentrations against Staphylococcus aureus and Escherichia coli was improved due to the higher degradation rate and more Zn2+ and Cu2+ released. Our previous study already showed that the Zn-Cu-Fe alloy exhibited excellent mechanical properties and moderate degradation rate. Based on the above results, the in vitro biocompatibilities and antibacterial properties of Zn-3Cu alloy are significantly improved by the alloying of trace Fe, and the hemocompatibility is not adversely affected, which indicated that Zn-Cu-Fe alloy is a promising vascular stents candidate material.

Zinc (Zn) is an essential micronutrient for most organisms including humans, and Zn deficiency is widespread in human populations, particularly in underdeveloped regions. Cereals such as rice (Oryza sativa) are the major dietary source of Zn for most people. However, the molecular mechanism underlying Zn uptake in rice is still not fully understood. Here, we report that a member of the ZIP (ZRT, IRT-like protein) family, OsZIP9, contributes to Zn uptake in rice. It was expressed in the epidermal and exodermal cells of lateral roots, localized in the plasma membrane and induced during Zn deficiency. Yeast-expressed OsZIP9 showed much higher Zn influx transport activity than other rice ZIP proteins in a wide range of Zn concentrations. OsZIP9 knockout rice plants showed a significant reduction in growth at low Zn concentrations, but could be rescued by a high Zn supply. Compared with the wild type, accumulation of Zn in root, shoot and grain was much lower in knockout lines, particularly with a low supply of Zn under both hydroponic and paddy soil conditions. OsZIP9 also showed Co uptake activity. Natural variation of OsZIP9 expression level is highly associated with Zn content in milled grain among rice varieties in the germplasm collection. Taken together, these results show that OsZIP9 is an important influx transporter responsible for the take up of Zn and Co from external media into root cells.

People often accompany gestures in the process of speaking, but the individual's gesture style is not the same. The present study uses the ambiguity resolution paradigm to explore the influence of two kinds of gesture styles on gesture-verbal comprehension. The study manipulated the gesture styles and meaning types of target words and recorded N400 amplitude. This study found that (1) in the non-grooming condition, compared with the situation where the gesture and semantics are inconsistent, a smaller N400 appears under the consistent condition; (2) in the grooming condition, the grooming gesture will reduce the effect of the iconic gesture on speech understanding. N400 amplitude increased only in the case of matching between dominant meaning gesture and subordinate meaning target word. These results suggest that the communication styles of gestures of different speakers, in the process of speech comprehension, affect how well listeners integrate gestures and language.

Using a Dazzler system and tilting a compressor grating, we provide an effective way of using the laser group delay dispersion to continuously steer the electron beam accelerated by an asymmetric laser wakefield. The deviation angle of the electron beam was the same as that of the angularly chirped laser pulse from its initial optical axis, which is determined by the laser pulse-front-tilt (PFT). This method can be utilized to continuously control over the pointing direction of electron bunches to the requisite trajectories, especially for practical applications in highly sensitive alignment devices such as electron-positron colliders or undulators. Additionally, we investigate the effect of PFT on the properties of the electron beam.

Citrate synthase (CS), the rate-limiting enzyme in the tricarboxylic acid (TCA) cycle catalyzes the first step of the cycle, namely, the condensation of oxaloacetate and acetyl-CoA to produce citrate. The expression and enzymatic activity of CS are altered in cancers, but posttranslational modification (PTM) of CS and its regulation in tumorigenesis remain largely obscure. SIRT5 belongs to the nicotinamide adenine dinucleotide (NAD)+-dependent deacetylase sirtuin family and plays vital roles in multiple biological processes via modulating various substrates. Here, we show that SIRT5 interacts with CS and that SIRT5 desuccinylates CS at the evolutionarily conserved residues K393 and K395. Moreover, hypersuccinylation of CS at K393 and K395 dramatically reduces its enzymatic activity and suppresses colon cancer cell proliferation and migration. These results provide experimental evidence in support of a potential therapeutic approach for colon cancer.

The absolute response of the GE Amersham Typhoontm imaging plate scanner is studied in this paper. The sensitivity function of the scanner with different photomultiplier tube voltages was obtained by using a pre-calibrated Cu Kα x-ray tube. The results showed that the sensitivity function decreases exponentially with higher voltage and is also affected by the scanning pixel size. The spatial resolution and the fading effect of the imaging plate system on x rays were also investigated and compared with the previous scanner models.

It is reported that dihydroartemisinin could reduce the expression of phosphorylated adhesion kinase and matrix metalloproteinase-2, inhibit the growth, migration and invasion of ovarian cancer cells, promote the formation of Treg cells through TGF-beta/Smad signaling pathway, and play an immunosuppressive role; dihydroartemisinin could also inhibit the growth of lung cancer cells by inhibiting the expression of vascular endothelial growth factor(VEGF) receptor KDR. However, there are few studies on dihydroartemisinin in hepatocellular carcinoma cells. In order to preliminarily explore the effect of dihydroartemisinin on invasion and metastasis of hepatocellular carcinoma cells, CCK-8 method and crystal violet staining were used to detect the effect of dihydroartemisinin on the growth of hepatocellular carcinoma cell 7402 and highly metastatic hepatocellular carcinoma cell MHCC97 H. The effects of dihydroartemisinin on the invasion and metastasis of hepatocellular carcinoma cell 7402 and highly metastatic hepatocellular carcinoma cell MHCC97 H were studied by using cell wound healing and Transwell. Western blot was used to detect the protein expression of epidermal growth factor receptor(EGFR) and its downstream signaling pathway in cells treated with dihydroartemisinin for 48 hours. The results showed that dihydroartemisinin could inhibit the growth of hepatocellular carcinoma cell 7402 and highly metastatic hepatocellular carcinoma cell MHCC97 H at 25 µmol·L~(-1). As compared with the control group, the number of cell clones was significantly reduced, and the ability of cell migration and invasion was weakened. Western blot results showed that as compared with the control group, dihydroartemisinin group could down-regulate the protein expression of EGFR and its downstream signaling pathways p-AKT, p-ERK, N-cadherin, Snail and Slug, and up-regulate the expression of E-cadherin protein, thus affecting the migration, invasion and metastasis of hepatocellular carcinoma cells 7402 and MHCC97 H.
