Objective: Reductions in placental weight and size have been associated with reduced fetal growth. However, few studies have examined the association of placental weight and size with the risk of fetal growth restriction (FGR). Methods: We enrolled 121 mother-newborn pairs, including 54 FGR cases and 67 healthy controls, from our previous case-control study. The weight, surface area, and thickness of the placenta were measured by medical professionals. Results: Reduced placental weight and surface area were found to be associated with decreased birth weight. A 10-unit decrement in placental weight (g) and surface area (cm2) was associated with 33.9 (ß = 33.9, 95% CI, 22.1-45.7) and 24.3 (ß = 24.3, 95% CI, 11.2-37.5) g decrease in birth weight, respectively. Those associations varied by infant gender and the magnitudes of effect were larger among male fetuses. Moreover, reduced placental weight and surface area were associated with increased odds of FGR. A 10-unit decrease in placental weight and surface area were associated with 21% (OR = 1.21, 95% CI, 1.08-1.44) and 19% (OR = 1.19, 95% CI, 1.06-1.41) increase in the odds of FGR. Conclusions: Our results suggest that fetuses with lower placental weight and smaller surface area are at higher risk of developing FGR.

Introduction: The treatment strategy for low-grade gliomas (LGGs) is still controversial, and there are no standardized criteria to predict the prognosis of patients with LGGs. Magnetic resonance imaging (MRI) is a routine test for preoperative diagnosis for LGG and can reflect the destructive features for the tumor. In the present study, we aimed to explore the relationship between the MRI features and prognosis in patients with LGG.Methods: Clinical data of 80 patients with pathologically proved LGGs between January 2010 and December 2016 were analyzed retrospectively. MRI features were classified as contrast enhancement pattern (focal enhancement, diffuse enhancement and ring-like enhancement), necrosis and cysts based on the preoperative MR images. Kaplan-Meier method and multivariate analysis were performed on the data by SPSS software to explore the prognostic significance of MRI features.Results: Patients with cystic LGG had a significantly longer 5-year progression-free survival (PFS) than that with no cyst (90.9 ± 8.7 vs 65.7 ± 9.1%, P=0.045). Multivariate analysis further verified cyst as an independent prognosis factor for PFS (P=0.027, hazard ratio [HR] = 0.084). Additionally, patients with ring-like enhancement exhibited significantly longer 5-year PFS time in the Kaplan-Meier survival curves (100 vs 67.2 ± 7.7%, P=0.049). There was no significant difference in PFS and overall survival (OS) between patients with or without necrosis.Conclusion: Our study suggests that cyst formation and ring-like enhancement on preoperative MR images can be useful to predict a favorable prognosis in patients with LGGs.

CONTEXT: No studies have assessed the association between air pollution exposure and vitamin D status in pregnant women. OBJECTIVE: To examine the association between particulate air pollution exposure and circulating 25-hydroxyvitamin [25(OH)D] levels in pregnant women. DESIGN: A longitudinal cohort study. PARTICIPANTS: A total of 3285 pregnant women were recruited at a maternal and child health hospital. INTERVENTIONS: None. MAIN OUTCOME MEASURES: Serum 25(OH)D levels. RESULTS: We observed trimester-specific associations between particulate air pollution exposure and circulating 25(OH)D levels. The associations were most pronounced for the periods of the third trimester and the entire pregnancy. A 10 µg/m3 increase in fine particulate matter (PM2.5) and particulate matter with aerodynamic diameter ≤10 µm (PM10) exposure during the entire pregnancy was associated with a 4.62% (95% CI, -6.31% to -2.93%) and 5.06% (95% CI, -6.50% to -3.62%) decrease in 25(OH)D levels, respectively. Particulate air pollution exposure was also associated with elevated odds of maternal vitamin D deficiency. A 10 µg/m3 increase in PM2.5 and PM10 exposure during the entire pregnancy was associated with a 45% (OR = 1.45, 95% CI, 1.29 to 1.63) and 48% (OR = 1.48, 95% CI, 1.33 to 1.64) increase in the odds of maternal vitamin D deficiency. Mediation analysis estimated that decreased solar UV-B radiation mediated 69.5% and 66.4% of the inverse association between PM2.5 and PM10 exposure and circulating 25(OH)D levels. CONCLUSION: Our results suggest that prenatal exposure to particulate air pollution may play an independent role in maternal vitamin D deficiency. The role of air pollution should be incorporated into future guidelines for the prevention of maternal vitamin D deficiency.

Previous studies have demonstrated that fibroblast growth factor 8b (FGF8b) is up-regulated in a large proportion of prostate cancer patients, and plays a key role in the aggressive progress of prostate cancer. Herein, we investigated the effects of a short peptide derived from the gN helix domain of FGF8b on the metastatic behaviors of prostate cancer cells. The results demonstrated that the synthetic peptide might reverse the effects of FGF8b on cell proliferation, migration and invasion by suppressing the activation of MAPK and Akt signaling cascades, and reducing the expressions of the metastasis-related proteins, resulting in suppression of the aggressive phenotype of the prostate cancer cells. Collectively, these results underline the therapeutic potential of the FGF8b mimic peptide in advanced prostate cancer.

Distinct enterotypes have been observed in the human gut but little is known about the genetic basis of the microbiome. Moreover, it is not clear how many genetic differences exist between enterotypes within or between populations. In this study, both the 16S rRNA gene and the metagenomes of the gut microbiota were sequenced from 48 Han Chinese, 48 Kazaks, and 96 Uyghurs, and taxonomies were assigned after de novo assembly. Single nucleotide polymorphisms were also identified by referring to data from the Human Microbiome Project. Systematic analysis of the gut communities in terms of their abundance and genetic composition was also performed, together with a genome-wide association study of the host genomes. The gut microbiota of 192 subjects was clearly classified into two enterotypes (Bacteroides and Prevotella). Interestingly, both enterotypes showed a clear genetic differentiation in terms of their functional catalogue of genes, especially for genes involved in amino acid and carbohydrate metabolism. In addition, several differentiated genera and genes were found among the three populations. Notably, one human variant (rs878394) was identified that showed significant association with the abundance of Prevotella, which is linked to LYPLAL1, a gene associated with body fat distribution, the waist-hip ratio and insulin sensitivity. Taken together, considerable differentiation was observed in gut microbes between enterotypes and among populations that was reflected in both the taxonomic composition and the genetic makeup of their functional genes, which could have been influenced by a variety of factors, such as diet and host genetic variation.

OBJECTIVES: To enlarge the diversity of the desaturases associated with PUFA biosynthesis and to better understand the transcriptional regulation of desaturases, a Δ6-desaturase gene (Md6) from Mucor sp. and its 5'-upstream sequence was functionally identified in Saccharomyces cerevisiae. RESULTS: Expression of the Δ6-fatty acid desaturase (Md6) in S. cerevisiae showed that Md6 could convert linolenic acid to γ-linolenic acid. Computational analysis of the promoter of Md6 suggested it contains several eukaryotic fundamental transcription regulatory elements. In vivo functional analysis of the promoter showed the 5'-upstream sequence of Md6 could initiate expression of GFP and Md6 itself in S. cerevisiae. A series deletion analysis of the promoter suggested that sequence between -919 to -784 bp (relative to start site) named as eMd6 is the key factor for high activity of Δ6-desaturase. The activity of Δ6-desaturase was increased by 2.8-fold and 2.5-fold when the eMd6 sequence was placed upstream of -434 with forward or reverse orientations respectively. CONCLUSION: To our best knowledge, the native promoter of Md6 from Mucor is the strongest promoter for Δ6-desaturase reported so far and the sequence between -919 to -784 bp is an enhancer for Δ6-desaturase activity.

In this paper, a three-phase hollow fiber liquid-phase microextraction (HF-LPME) method combined with high-performance liquid chromatography (HPLC) was developed for the determination of hypoxanthine (HX), xanthine (Xan) and adenine (A) and then for the first time successfully applied to the analysis of HX, Xan and A in Alysicarpus vaginalis (L.) DC. medicinal materials. Different factors affecting the HF-LPME procedure were investigated and optimized. Under optimal extraction conditions (1-octanol as organic solvent, pH of the donor and acceptor phase 10.0 and 3.5, respectively, extraction time 40 min, stirring rate 800 rpm and salt addition 10%, w/v), HX, Xan and A could be determined within the test ranges with a good correlation coefficient (r(2) > 0.9992). The limit of detection for HX, Xan and A was 153, 173 and 97 ng/mL, respectively, and the intra- and inter-day relative standard deviations were no more than 9.8%. The content of HX, Xan and A in Alysicarpus vaginalis (L.) DC. medicinal materials was 120.40, 18.37 and 62.75 µg/g, respectively. This procedure afforded a convenient, sensitive, accurate and inexpensive method with a high extraction efficiency for determination of HX, Xan and A.

In this paper, a sensitive, rapid and reproducible high-performance liquid chromatography-tandem mass spectrometry method was developed to analyze 16α-hydro-ent-kauran-17,19-dioic acid in rat plasma. First, this study compared the pharmacokinetics of 16α-hydro-ent-kauran-17,19-dioic acid after oral administration of monomer and Siegesbeckiae pubescens Makino extract in rat plasma with approximately the same dosage of 6.0 mg/kg. Second, chromatographic separation was performed on a Waters Symmetry C18 column (2.1 × 100 mm, 3.5 µm) with isocratic elution using methanol-water containing 5 mmol/L ammonium acetate (70:30, v/v) as mobile phase at a flow rate of 0.2 mL/min. The calibration curves were linear over the range of 30-12000 ng/mL for monomer. At different time points (0, 0.083, 0.25, 0.75, 1, 2, 4, 6, 8, 12, 18, 24, 36, 48, 60 and 72 h) after administration, the concentrations of monomer in rat plasma were determined and main pharmacokinetic parameters were estimated. The double absorption presented in this study indicates that the pharmacokinetics of monomer in rat plasma have significant differences between different groups.

A novel chiral ionic liquid functionalized ß-cyclodextrin, 6-O-2-hydroxpropyltrimethylammonium-ß-cyclodextrin tetrafluoroborate ([HPTMA-ß-CD][BF4]), was synthesized and used as a chiral selector in capillary electrophoresis. [HPTMA-ß-CD][BF4] not only increased the solubility in aqueous buffer in comparison with the parent compound, but also provided a stable reversal electroosmotic flow, and the enantioseparation of eight chiral drugs was examined in phosphate buffer containing [HPTMA-ß-CD][BF4] as the chiral selector. The effects of the [HPTMA-ß-CD][BF4] concentration and the background electrolyte pH were studied. Moreover, the chiral separation abilities of ß-CD and [HPTMA-ß-CD][BF4] were compared and possible mechanisms for the chiral recognition of [HPTMA-ß-CD][BF4] are discussed.

RESUMO

3'-Deoxy-4-O-methylepisappanol, a new 3-benzylchroman derivative, was isolated from Sappan Lignum, together with thirteen known chemical compounds identified as protosappanin A, sappanchalcone, sappanone B, palmitic acid, (+)-(8S,8'S)-bisdihydrosiringenin, brazilein, 3-deoxysappanchalcone, (+)-lyoniresinol, 3-deoxysappanone B, protosappanin B, isoprotosappanin B, 3'-O-methylbrazilin and brazilin, respectively. Among these known compounds, this is the first time that (+)-(8S,8'S)-bisdihydrosiringenin was obtained from the family Caesalpiniaceae.

OBJECTIVE: To observe clinical therapeutic effect of catgut implantation at Shu- and Mu-acupoints of liver, spleen and kidney on premature ovarian failure. METHODS: One hundred and thirty-two cases of premature ovarian failure were randomly divided into a catgut implantation group and a medication group, 66 cases in each group. The catgut implantation group were treated by catgut implantation at She- and Mu-acupoints of liver, spleen and kidney, and the medication group were treated with oral administration of Estradiol Valerate 2 mg, qd, for 20 days, and 10 days later, Medroxyprogesterone Acetate 4 mg was added, b. i. d, for 10 days, averaging 6 months of medication. The therapeutic effects and changes of serum follicle-stimulating hormone (FSH) and estradiol (E2) were observed in the two groups. RESULTS: After treatment, serum FSH and E2 significantly improved in the two groups (P<0.01), with the serum E2 in the catgut implantation group increased more significantly than that in the medication group (P<0.01). The cured rate and the total effective rate were 84.9% and 97.0% in the catgut implantation group and 31.8% and 84.8% in the medication group, the cured rate in the catgut implantation group being better than that in the medication group (P<0.05). Ten month later, the therapeutic effect in the catgut implantation group was kept. CONCLUSION: Catgut implantation at Shu- and Me-acupoints of liver, spleen and kidney has a good therapeutic effect on premature ovarian failure with no side effect.

OBJECTIVE: To compare the therapeutic effects of acupuncture and western medicine on minimal brain dysfunction (MBD) and to search for a clinically effective therapy for MBD. METHODS: Sixty-eight cases were randomly divided into an acupuncture group and a western medicine group, 34 cases in each group. The acupuncture group were treated by acupuncture at Dazhui (GV 14) and Shenque (CV 8), and the western medicine group by taking Haloperidol orally. One month constituted one course. After treatment, the total effective rate and scores of Connell's scale for diagnosis and behavior of MBD were compared between the two groups. RESULTS: The total effective rate and the score after treatment were 97.1% and 10 +/- 0.37 in the acupuncture group and 82.4% and 15 +/- 0.93 in the western medicine group, with a very significant difference between the two groups (P < 0.01, P < 0.000 5), the acupuncture group being better than the western medicine group. Follow-up survey for 2-10 months showed the effects of the acupuncture group still were kept. CONCLUSION: Acupuncture at Dazhui (GV 14) and Shenque (CV 8) can effectively cure MBD.
