RESUMO

The ability to arrange distinct cells in specific, predefined patterns at single-cell resolution can have broad applications in cell-based assays and play an important role in facilitating interdisciplinary research for researchers in various fields. However, most existing methods for single-cell patterning are based on the complicated lithography-based microfabrication process, and require professional skills. Thus, exploiting convenient and universal strategies of single-cell preparation while maintaining high-throughput single-cell patterning remains a challenge. Here, we describe a simple approach for rapid and high-efficiency single-cell patterning using an ultrathin metal microstencil (UTmS) and common tools available in any laboratory. In this work, ultrathin steel microstencil plates with only 5 µm thickness could be fabricated with laser drilling and achieve single-cell prototyping on an arbitrary planar substrate under gravity-induced natural sedimentation without requiring additional fixation, reaction pools, and centrifugation procedures. In this method, the UTmS is reusable and single-cell occupancy could easily reach approximately 88% within 30 min on fibronectin-modified substrates under gravity-induced natural sedimentation, and no significant effect on cell viability was observed. To verify this method, the real-time and heterogeneous study of calcium release and apoptosis behaviors of single cells was carried out based on this new strategy. To our knowledge, it is the first time that a UTmS with 5 µm thickness is directly applied to facilitate the micropatterning of high-resolution single cells, which is valuable for researchers in different fields owing to its user-friendly operation.

RESUMO

Chronic HBV infection, the main cause of liver cirrhosis and hepatocellular carcinoma, has become a global health concern. Machine learning algorithms are particularly adept at analyzing medical phenomenon by capturing complex and nonlinear relationships in clinical data. Our study proposed a predictive model on the basis of 55 routine laboratory and clinical parameters by machine learning algorithms as a novel non-invasive method for liver fibrosis diagnosis. The model was further evaluated on the accuracy and rationality and proved to be highly accurate and efficient for the prediction of HBV-related fibrosis. In conclusion, we suggested a potential combination of high-dimensional clinical data and machine learning predictive algorithms for the liver fibrosis diagnosis.

Patients with anti-interferon (IFN)-γ autoantibodies have weakened immune defenses against intracellular pathogens. Because of its low incidence and non-specific symptoms, diagnosis of anti-IFN-γ autoantibody syndrome is difficult to establish during the early stages of infection. Here, we report a patient with high titers of serum anti-IFN-γ autoantibodies suffering from opportunistic infections. The patient presented with intermittent fever for 2 weeks. During his first hospitalization, he was diagnosed with Talaromyces marneffei pulmonary infection and successfully treated with antifungal therapy. However, multiple cervical lymph nodes subsequently became progressively enlarged. Mycobacterium abscessus infection was confirmed by positive cervical lymph node tissue cultures. High-titer serum anti-IFN-γ antibodies were also detected. Following anti-M. abscessus therapy, both his symptoms and lymph node lymphadenitis gradually improved. Anti-IFN-γ autoantibody syndrome should be considered in adult patients with severe opportunistic coinfections in the absence of other known risk factors.

BACKGROUND: To explore adverse drug reactions (ADRs) and the effects of nursing interventions after Aidi injection for the treatment of non-Hodgkin's lymphoma (NHL). METHODS: A total of 104 NHL patients treated in our hospital from March 2019 to March 2020 were selected. All patients underwent conventional chemotherapy, with a concomitant Aidi injection administered at different doses (40, 60, 80, and 100 mL). ADRs were analyzed for all patients, and the clinical outcomes of ADRs were recorded after specific nursing interventions were performed. RESULTS: A total of 17 NHL patients had ADRs, with a total incidence rate of 15.89% (total of 30 ADRs). In terms of different types of ADRs, inappetence accounted for the largest proportion, followed by skin pruritus and fever, phlebitis, nausea and vomiting, and chest distress and/or palpitation, the last of which shared the same proportion as chills and/or low fever and urticaria. The incidence rate of ADRs (total of 18 ADRs, 60.00%) was higher in NHL patients who were given 100 mL of Aidi injection. In terms of age and gender distribution of ADRs, there were 10 males (55.56%) and 7 females (44.44%), and ADRs were more common in patients aged 46-60 years old (total of 6 ADRs, 35.29%). Aidi injection mainly induced mild ADRs (total of 22 ADRs, 73.33%), and the resulting ADRs mostly occurred for <2 hours (total of 19 ADRs, 60.00%). After the specific nursing interventions were performed, no deaths due to ADRs occurred, and 12 (64.71%) cases were cured, 5 (29.41%) cases improved, and 1 (5.88%) case had no progression. CONCLUSIONS: Particular attention should be paid to ADRs in the treatment of NHL patients with Aidi injection. After ADRs occur, specific nursing interventions can aid in recovery and lead to improvements in prognosis.

BACKGROUND: Xinyin Tablet (XYT) has been widely used in the treatment of CHF, Which helping to improve the clinical symptoms, enhance exercise, and even may improve the long-term prognosis of patients. However, the exact effectiveness and safety of XYT for CHF has not be comprehensively researched, so we want to generalize the effectiveness and safety of XYT for CHF through the meta-analysis, which may benefit the design of future clinical trials and provide valuable references. METHODS: This protocol complies with the Preferred Reporting Items for Systematic Review and Meta-Analysis Protocols. From the inception until September 2020, a systematic and comprehensive electronic search about Relevant randomized controlled trials will be conducted in 4 English literature databases and 4 Chinese literature databases. The registration number: INPLASY2020100015. 2 investigators will be arranged to deal with the study selection and data extraction independently. The New York Heart Function Classification, traditional Chinese medicine (TCM) symptom scores, the scores of quality of life, 6-min walk distance (6MWD), etc. will be systematically measured as outcomes. At last, the data will be handled by Review Manager 5.3 and Stata 15.0. RESULTS AND CONCLUSION: This study is hoping to provide a high-level evidence to prove the therapeutic effect of XYT on CHF, which may enhance the application of Chinese medicine.

PURPOSE: To compare the diagnostic accuracy of parameters derived from the histogram analysis of precontrast, 10-min hepatobiliary phase (HBP) and 20-min HBP T1 maps for staging liver fibrosis (LF). METHODS: LF was induced in New Zealand white rabbits by subcutaneous injections of carbon tetrachloride for 4-16 weeks (n = 120), and 20 rabbits injected with saline served as controls. Precontrast, 10-min and 20-min HBP modified Look-Locker inversion recovery (MOLLI) T1 mapping was performed. Histogram analysis of T1 maps was performed, and the mean, median, skewness, kurtosis, entropy, inhomogeneity and 10th/25th/75th/90th percentiles of T1native, T110min and T120min were derived. Quantitative histogram parameters were compared. For significant parameters, further receiver operating characteristic (ROC) analyses were performed to evaluate the potential diagnostic performance in differentiating LF stages. RESULTS: Finally, 17, 20, 21, 21 and 20 rabbits were included for the F0, F1, F2, F3, and F4 pathological grades of fibrosis, respectively. The mean/75th of T1native, entropy of T110min and entropy/mean/median/10th of T120min demonstrated a significant good correlation with the LF stage (|r| = 0.543-0.866, all P < 0.05). The 75th of T1native, entropy10min, and entropy20min were the three most reliable imaging markers in reflecting the stage of LF. The area under the ROC curve of entropy20min was larger than that of entropy10min (P < 0.05 for LF ≥ F2, ≥F3, and ≥ F4) and the 75th of T1native (P < 0.05 for LF ≥ F2 and ≥ F3) for staging LF. CONCLUSION: Magnetic resonance histogram analysis of T1 maps, particularly the entropy derived from 20-min HBP T1 mapping, is promising for predicting the LF stage.

Acute lung injury (ALI) severely impairs gas exchange and results in high mortality. Insulin-like growth factor binding protein 3 (IGFBP-3) plays a crucial role in diverse lung diseases;however, the expression and function of IGFBP-3 in ALI remain unclear. In the present study, mice were injected with lipopolysaccharide to establish ALI, and IGF and IGFBP3 expression was measured using ELISA, western blotting, and immunohistochemical staining. Mice with ALI were then treated with recombinant human IGFBP-3 (rhIGFBP-3), and treatment was evaluated using survival analysis, histological staining analysis, and inflammatory cytokine expression in lung tissues and bronchoalveolar lavage fluid (BALF). The expression of NF-κB and VEGF was also measured using western blotting and ELISA in ALI mice. Our results demonstrated the upregulation of IGF expression in lung tissues and BALF of ALI mice, accompanied by downregulation of IGFBP-3. Administration of rhIGFBP-3 prolonged the survival time and attenuated LPS-induced lung injury. The expression of TNF-α, IL-6, IL-1ß, and IFN-γ both in lung tissues and BALF decreased after rhIGFBP-3 treatment, whereas IL-10 expression increased. These results suggest that rhIGFBP-3 inhibits the expression of NF-κB and VEGF in lung tissues. Collectively, our study demonstrates a protective role of rhIGFBP-3 in ALI by regulation of lung inflammation.

OBJECTIVES: Heart failure (HF) is a common and potentially fatal condition. In 2015, HF affected approximately 40 million people globally. Evidence showing that the use of nitrates can improve clinical outcomes in patients with HF is limited. This study aimed to assess the effect of nitrates on functional capacity and exercise time in patients with HF. METHODS: PubMed, Cochrane Library, and Embase databases were reviewed for articles on the use of nitrates and other treatments for patients with HF. The primary endpoints were the 6-minute walk test distance, exercise time, and quality of life. Secondary endpoints were all-cause mortality, arrhythmia, hospitalization, and worsening HF. The weighted mean difference, risk ratio, and 95% confidence interval were calculated. RESULTS: A total of 14 related studies that comprised 26,321 patients were included. No significant differences were found in the 6-minute walk test distance, exercise time, and quality of life between the nitrate and control treatment groups. There were also no differences in all-cause mortality, the incidence of arrhythmia, hospitalization, and worsening HF between these two groups. CONCLUSION: Patients with HF who receive nitrate treatment do not have better quality of life or exercise capacity compared with controls.

This study investigates the roles of trust in citizens and compliance with agency policies in mediating the direct and indirect relationships between internal procedural justice and external procedural justice among Chinese and Taiwanese police officers. Based on survey data collected from 1,253 police officers, this study comparatively analyzes whether supervisory treatment of officers is predictive of trust in citizens and willingness to follow agency policies, which in turn is linked to their willingness to act fairly and justly toward citizens on the street. The results indicate that officer trust in citizens mediates the relationship between internal and external procedural justice in both China and Taiwan, but compliance with agency policies does not. Internal procedural justice directly predicts external procedural justice among Taiwanese officers, but such a connection is not found among Chinese officers. Implications for future research and policy are discussed.

INTRODUCTION: Unstable angina (UA), referred to as acute coronary syndrome (ACS), causes unexpected chest pain. Xueshuantong injection (lyophilised) (XST) is a traditional Chinese herbal injection having the potential to treat ACS. However, no clinical trial has been performed in this field. This clinical trial aims to examine the efficacy and safety of XST. METHODS AND ANALYSIS: This is a randomised, parallel-arm, controlled, double-blind and multicentre clinical trial. A total of 1200 participants with UA will be enrolled in a 1:1 ratio, with 600 patients included in the XST treatment group and 600 with 1/20th dose in the control group. The efficacy assessment and major adverse cardiovascular events will be observed, and the frequency of angina attack, angina pectoris will be examined at the start and end of the run-in period. All adverse events will be recorded, regardless of the severity, to assess the safety of XST. The baseline characteristics of patients will be summarised and compared using the t test or non-parametric statistical test. Qualitative data will be analysed using the χ2 or Fisher exact tests, Cochran-Mantel-Hasenszel test and Wilcoxon test. ETHICS AND DISSEMINATION: This trial has been approved by the Research Ethics Committee of The First Affiliated Hospital of Guangzhou University of Chinese Medicine, China (approval number: ZYYEC [2017] 0021). Written informed consent will be obtained from all participants. The results of this trial will be disseminated to the public through academic conferences and peer-reviewed journals. TRIAL REGISTRATION: This study was registered on the Chinese Clinical Trial Registry (http://www.chictr.org.cn/) with the ID ChiCTR1800015911.

Bottom-up construction of transition copper-nitrogen-carbon (Cu-N-C) electrocatalysts with high-performance and long-term durability for the oxygen reduction reaction (ORR) still remains a great challenge. Herein, we propose a temperature-controlled synthesis strategy with confinement effect for fabrication of a novel two-dimensional dual-metal (Cu/Zn) zeolitic imidazolate framework material, which presents an ultrathin nanosheet morphology after high-temperature thermal treatment (denoted as Cu-N-UNS). By controlling the reaction temperature as well as regulating the ratio of metal ions and taking advantages of the confinement effect of surfactants, the rationally designed ultra-thin carbon layer not only prevents aggregation of transition Cu particles and avoids direct contact with reactants and electrolyte solutions to enhance the durability of electrocatalysts, but also shortens the electronic transmission path between the active transition metal species and carbon surface. Therefore, the electrocatalyst exhibits excellent electrocatalytic performance for the ORR (E1/2 ≈ 0.898 V), which is superior to those of state-of-the-art benchmark noble-metal electrocatalysts. Moreover, the even distribution of Cu-N-C and existence of N-Cu2+-Cu0 active sites make a great contribution to the electrocatalyst activity. Notably, the Cu-N-UNS used as air electrodes for Zn-air batteries also exhibits a high peak power density of ≈134.7 mW cm-2 at a current density of ≈231.9 mA cm-2 with remarkable durability.

Previous density-functional theory (DFT) calculations show that sub-nanometric Cu clusters (i.e., 13 atoms) favorably generate CH4 from the CO2 reduction reaction (CO2 RR), but experimental evidence is lacking. Herein, a facile impregnation-calcination route towards Cu clusters, having a diameter of about 1.0 nm with about 10 atoms, was developed by double confinement of carbon defects and micropores. These Cu clusters enable high selectivity for the CO2 RR with a maximum Faraday efficiency of 81.7 % for CH4 . Calculations and experimental results show that the Cu clusters enhance the adsorption of *H and *CO intermediates, thus promoting generation of CH4 rather than H2 and CO. The strong interactions between the Cu clusters and defective carbon optimize the electronic structure of the Cu clusters for selectivity and stability towards generation of CH4 . Provided here is the first experimental evidence that sub-nanometric Cu clusters facilitate the production of CH4 from the CO2 RR.

Effective features can improve the performance of a model and help us understand the characteristics and underlying structure of complex data. Previously proposed feature selection methods usually cannot retain more discriminative information. To address this shortcoming, we propose a novel supervised orthogonal least square regression model with feature weighting for feature selection. The optimization problem of the objective function can be solved by employing generalized power iteration and augmented Lagrangian multiplier methods. Experimental results show that the proposed method can more effectively reduce feature dimensionality and obtain better classification results than traditional feature selection methods. The convergence of our iterative method is also proved. Consequently, the effectiveness and superiority of the proposed method are verified both theoretically and experimentally.

This study presents low-crystalline and non-stoichiometric cobalt oxide (Co3-xO4-δ) porous nanosheet arrays (PNAs) grown on carbon fiber cloth (CFC) (Co3-xO4-δ PNAs/CFC) by a facile in situ anodic oxidation strategy. We firstly verified that the above prepared low crystalline cobalt oxide contained tetrahedral CoO4 vacancies, resulting in the creation of O vacancies at adjacent octahedral CoO6 sites, allowing the generation of tetragonal-pyramidal CoO5 sites which were regarded as active sites and being accessible for the oxygen evolution reaction (OER) with different reaction mechanisms compared to that of traditional CoO6 sites in high-crystalline and stoichiometric Co3O4, thus endowing Co3-xO4-δ PNAs/CFC with significantly improved OER activity and superior stability compared to their crystalline counterparts (Co3O4 PNAs/CFC), as illustrated by experiments and density functional theory (DFT) calculations. This study will open up a new approach for the synthesis of defect-rich materials and provide new insight into the structure-property relationship of OER catalysts.

The widely used route of high-temperature pyrolysis for transformation of Prussian blue analogs (PBAs) to functional nanomaterials leads to the fast removal of CN- ligands, and thus the formation of large metal aggregates and the loss of porous structures inside PBAs. Here, a controllable pyrolysis route at low temperature is reported for retaining the confined effect of CN- ligands to metal cations during the whole pyrolysis process, thereby preparing high-surface-area cubes comprising disordered bimetallic oxides (i.e., Co3 O4 and Fe2 O3 ) nanoparticles. The disordered structure of Co3 O4 enables the exposure of abundant oxygen vacancies. Notably, for the first time, it is found that the in situ generated CoOOH during the oxygen evolution reaction (OER) can inherit the oxygen vacancies of pristine Co3 O4 (i.e., before OER), and such CoOOH with abundant oxygen vacancies adsorbs two - OH in the following Co3+ to Co4+ for markedly promoting OER. However, during the similar step, the ordered Co3 O4 with less oxygen vacancies only involves one - OH, resulting in the additional overpotentials for adsorbing - OH. Consequently, with high surface area and disordered Co3 O4 , the as-synthesized electrocatalysts have a low potential of 237 mV at 10 mA cm-2 , surpassing most of reported electrocatalysts.

Carbon dioxide electroreduction provides a useful source of carbon monoxide, but comparatively few catalysts could be sustained at current densities of industry level. Herein, we construct a high-yield, flexible and self-supported single-atom nickel-decorated porous carbon membrane catalyst. This membrane possesses interconnected nanofibers and hierarchical pores, affording abundant effective nickel single atoms that participate in carbon dioxide reduction. Moreover, the excellent mechanical strength and well-distributed nickel atoms of this membrane combines gas-diffusion and catalyst layers into one architecture. This integrated membrane could be directly used as a gas diffusion electrode to establish an extremely stable three-phase interface for high-performance carbon dioxide electroreduction, producing carbon monoxide with a 308.4 mA cm-2 partial current density and 88% Faradaic efficiency for up to 120 h. We hope this work will provide guidance for the design and application of carbon dioxide electro-catalysts at the potential industrial scale.

Stellera chamaejasme is one of the most serious weeds in Qinhai-Tibetan Plateau, the rapid expansion of which exerts an increasing effect on the alpine meadow ecosystem. With high-throughput sequencing technology, geostatistics and GIS method, the spatial heterogeneity of soil fungal diversity in Stellera occurrence area and the spatial correlation between Stellera coverage and soil fungal diversity were investigated in a typical degraded alpine meadow of the Qilian Mountain. Compared to no-Stellera area, the fungi richness in Stellera area decreased, the dominance increased, and the α-diversity reduced. The difference of fungal species composition enhanced and ß-diversity significantly increased. The spatial pattern of soil fungal diversity was affected by the invasion of Stellera, resulting in higher fragmentation in occurrence area. Spatial heterogeneity of species composition increased remarkably, and spatial stability of α-diversity and ß-diversity decreased. The portion of positive correlation and negative correlation interlaced, indicating no clear spatial correlation between Stellera coverage and soil fungal diversity. Our results indicate that the spatial pattern of soil fungal diversity was affected by the interaction of soil and vegetation in Stellera invaded meadows.

BACKGROUND: Emerging reports demonstrated that PIWI-interacting RNAs (piRNAs) played an indispensable role in tumorigenesis. However, it still remains elusive whether piR-1245 in gastric juice specific in stomach could be employed as a biomarker for gastric cancer (GC). The present work is aiming at exploring the possibility of piR-1245 in gastric juice as a potential marker to judge for diagnosis and prognosis of gastric cancer. METHODS: Gastric juice was collected from 66 GC patients and 66 healthy individuals. Quantitative real-time reverse transcriptase polymerase chain reaction (qRT-PCR) was employed to measure the levels of piR-1245 expression. Then, the pattern of piR-1245 expression in gastric juice was determined between GC patients and healthy individuals. A receiver operating characteristic (ROC) curve was constructed for distinguishing GC from healthy individuals. RESULTS: Gastric juice piR-1245 levels in GC were higher than those of controls (P < .0001). The value of area under ROC (AUC) was 0.885 (sensitivity, 90.9%; specificity, 74.2%; 95% confidence interval, 0.8286 to 0.9414). High gastric juice piR-1245 expression was signally correlated with tumor size (P = .013) and TNM stage (P = .001). GC patients with high piR-1245 expression in gastric juice exerted a poorer overall survival (OS) (P = .0152) and progression-free survival (PFS) (P = .013). COX regression analysis verified that gastric juice piR-1245 expression was an independent prognostic risk variable for OS (P < .05). CONCLUSIONS: The current study suggested that piR-1245 in gastric juice had the potential to be a useful biomarker for GC detection and prognosis prediction.

Cryogenic soft X-ray tomography (Cryo-SXT) is ideally suitable to image the 3D sub-cellular architecture and organization of cells with high resolution in the near-native preservation state. Cryogenic fluorescence microscopy (Cryo-FM) can determine the location of a molecule of interest that has been labeled with a fluorescent tag, thus revealing the function of the cells. To understand the relations between the sub-cellular architecture and the function of cells, correlative Cryo-SXT and Cryo-FM was applied. This method required the matching of images of different modalities, and the accuracy of the matching is important. Here, a precise correlative method of Cryo-SXT and Cryo-FM is introduced. The capability of matching images of different modalities with high resolution was verified by simulations and practical experiments, and the method was used to identify vacuoles and mitochondria.
