RESUMO

Identification of immunogenic targets against hepatitis B virus (HBV)-encoded proteins will provide crucial advances in developing potential antibody therapies. In this study, 63 treatment-naïve patients with chronic HBV infection and 46 patients who achieved hepatitis B surface antigen loss (sAg loss) following antiviral treatment were recruited. Moreover, six patients who transitioned from the hepatitis B e antigen-positive chronic infection phase (eAg+CInf) to the hepatitis phase (eAg+CHep) were enrolled from real-life clinical practice. Additionally, telbivudine-treated eAg+CHep patients and relapsers or responders from an off-treatment cohort were longitudinally studied. The frequencies and function of B cells were assessed by flow cytometry. We devised a peptide array composed of 15-mer overlapping peptides of HBV-encoded surface (S), core (C), and polymerase (P) proteins and performed a screening on B-cell linear epitopes with sera. Naïve B cells and plasmablasts were increased, whereas total memory, activated memory (AM), and atypical memory (AtM) B cells were reduced in sAg- patients compared with sAg+ patients. Importantly, longitudinal observations found that AtM B cells were associated with successful treatment withdrawal. Interestingly, we identified six S-specific dominant epitopes (S33, S34, S45, S76, S78, and S89) and one C-specific dominant epitope (C37) that reacted with the majority of sera from sAg- patients. Of note, more B-cell linear epitopes were detected in CHep patients with alanine aminotransferase (ALT) flares than in nonflare CInf patients, and five B-cell linear epitopes (S4, S5, S10, S11, and S68) were overwhelmingly recognized by ALT flare patients. The recognition rates of epitopes on C and P proteins were significantly increased in CHep patients relative to CInf patients. Strikingly, a statistically significant elevation in the number of positive epitopes was observed when ALT nonflare patients shifted into the flare phase. Moreover, S76 identified at baseline was confirmed to be associated with a complete response after 48 weeks of telbivudine therapy. Taken together, we identified several functional cure-related B-cell linear epitopes of chronic HBV infection, and these epitopes may serve as vaccine candidates to elicit neutralizing antibodies to treat HBV infection.

Background: Cytogenetics at diagnosis is the most important prognostic factor for adult acute myeloid leukemia (AML), but nearly 50% of AML patients who exhibit cytogenetically normal AML (CN-AML) do not undergo effective risk stratification. Therefore, the development of potential biomarkers to further define risk stratification for CN-AML patients is worth exploring. Methods: Transcriptome data from 163 cases in the GSE12417-GPL96 dataset and 104 CN-AML patient cases in the GSE71014-GPL10558 dataset were downloaded from the Gene Expression Omnibus database for overall survival (OS) analysis and validation. Results: The combination of Wilms tumor 1 (WT1) and cluster of diffraction 58 (CD58) can predict the prognosis of CN-AML patients. High expression of WT1 and low expression of CD58 were associated with poor OS in CN-AML. Notably, when WT1 and CD58 were used to concurrently predict OS, CN-AML patients were divided into three groups: low risk, WT1low CD58high; intermediate risk, WT1highCD58high or WT1lowCD58low; and high risk, WT1high CD58low. Compared with low-risk patients, intermediate- and high-risk patients had shorter survival time and worse OS. Furthermore, a nomogram model constructed with WT1 and CD58 may personalize and reveal the 1-, 2-, 3-, 4-, and 5-year OS rate of CN-AML patients. Both time-dependent receiver operating characteristics and calibration curves suggested that the nomogram model demonstrated good performance. Conclusion: Higher expression of WT1 with lower CD58 expression may be a potential biomarker for risk stratification of CN-AML patients. Moreover, a nomogram model constructed with WT1 and CD58 may personalize and reveal the 1-, 2-, 3-, 4-, and 5-year OS rates of CN-AML patients.

The incidence rate of cerebrovascular diseases is increasing year by year, but the accuracy of clinical diagnosis is not high enough to cause disease. Many patients cannot effectively diagnose and treat the disease at the early stage. CT perfusion imaging (CTP) and CT angiography (CTA) were used to diagnose cerebrovascular diseases. In this paper, 26 cases of patients with acute cerebrovascular disease in our hospital from June to August 2020 were selected as the research objects. According to the diagnosis method voluntarily chosen by patients, 6 cases were only detected by CT perfusion imaging, 8 cases were only detected by CT angiography, and the remaining 12 cases were diagnosed by CT perfusion imaging and CT angiography. Then, according to the diagnosis results of the CT image of the patients with cerebrovascular disease, these samples were followed up after 3 months, 6 months, and 9 months, and more accurate diagnosis results were obtained. The study showed that, after 3 months of CT detection, the sensitivity of patients with CTP detection was 67%, that of patients with CTA detection was 72%, and that of patients with combined detection was 83%. After 6 months of CT detection, the sensitivity of patients with CTP detection was 75%, that of patients with CTA detection was 79%, and that of patients with combined detection was 93%. After 9 months of CT detection, the sensitivity of patients with CTP detection was 86%, that of patients with CTA detection was 89%, and that of patients with combined detection was 99%. Therefore, the use of CTA combined with CTP joint detection of acute cerebrovascular disease imaging technology can effectively improve the diagnostic accuracy of patients.

A novel fluorescent probe 2-(4-(diethylamino)-2-hydroxybenzylidene)-N-(2,3,3-trimethylbicyclo[2.2.1]heptan-2-yl)hydrazinecarbothioamide (HT) was prepared in this study by a condensation reaction. HT has been confirmed to possess high specificity toward Ga3+ over other metal ions (including Al3+ and In3+) via a distinct fluorescence light-up response. Moreover, HT exhibited good detection performances for Ga3+ including high selectivity, excellent anti-interference ability, a wide working pH range, and good reversibility. The association constant and limit of detection (LOD) were calculated to be 5.34 × 103 M-1 and 1.18 × 10-6 M, respectively. The detection mechanism of HT toward Ga3+ was proposed and confirmed by 1H NMR analysis, HRMS analysis, and DFT calculations. A simple test strip-based portable detecting device and a molecular INHIBIT logic circuit were established for improving its practical applicability. Furthermore, the desirable sensing performance of HT for Ga3+ was successfully reconfirmed in MCF-7 cells and zebrafish.

BACKGROUND: Hemorrhoids are a common and seriously disruptive condition that seriously affects people's lives in terms of treatment. Injection therapy is an effective minimally invasive scheme for the treatment of grade II-III hemorrhoids, but its clinical application is limited by the adverse reactions caused by injection drugs. Some clinical studies have confirmed the efficacy and safety of Shaobei injection as a traditional Chinese medicine extract. However, there is no standard randomized controlled study to verify its efficacy and explore its potential mechanism. METHODS: This is a prospective, randomized, single blind, parallel controlled trial to study the efficacy of Shaobei injection in the treatment of grade II-III hemorrhoids and its effect on the expression of fibulin-3 and fibulin-5 in fibulin protein family. The patients will be randomly divided into a treatment group and control group. The treatment group will be treated with Shaobei injection, and the control group will be treated with rubber band ligation. The observation indexes include: visual analysis scale, postoperative hospital stay, total use of painkillers, fibulin-3 and fibulin-5, hemorrhoids recurrence, and adverse events. Finally, the data will be statistically analyzed by SPASS 18.0 software. DISCUSSION: This study will compare the efficacy of Shaobei injection with the rubber band ligation method in the treatment of grade II-III haemorrhoids and investigate its effect on the expression of fibulin-3 and fibulin-5 in the fibulin protein family. The results of this study will provide a basis for the clinical use of Paeoniflora injection as an alternative to traditional sclerosing agent in the treatment of grade II-III haemorrhoids.Trial registration: OSF Registration number:DOI 10.17605/OSF.IO/MKVDB.

OBJECTIVE: Disaster scenes produce long-term negative feelings in those who experience them. Previous studies have focused on mitigating disaster impacts through directed forgetting or conscious suppression. However, the initial emotional processing of disaster scenes is not fully understood, hindering the comprehension of long-term disaster impacts. This study aims to investigate how pictures of disaster scenes evoking disgust and sadness are processed via cortical electrical activity. METHODS: Pictures of grief and mutilation from disasters were used to evoke sadness and disgust, respectively. Event-related desynchronization (ERD) and event-related potentials (ERPs) were used to quantify the intensity and time-course of emotional processing. RESULTS: The information processing of emotional pictures was stronger than neutral pictures, represented by greater declines of alpha ERD. In the posterior ERP components of N1 and EPN, amplitudes for emotional pictures were larger than those for neutral pictures, which reflected the effects of arousal on visual perception. In the anterior ERP components of P2, P3, and LPP, disgust pictures showed higher attention attraction and enhanced encoding memory processing. CONCLUSIONS: Disgust disaster scenarios induced long-term prominent LPP, which may correspond with the long-term negative impacts of the disaster.

In nature, the normal growth, development, and quality of plants are significantly affected by many abiotic stresses, such as drought, salinity, low temperature, and heavy metals. Among heavy metals, copper is an essential element for plant growth and development but also has a toxic effect on plants when its concentration is excessive. Therefore, plants have evolved a complex regulatory network to regulate the balance of copper ions in cells. Heavy metal ATPases (HMAs), which transport heavy metals to intracellular compartments or detoxify heavy metals present at excessive concentrations, have been extensively studied in model plant species. However, no comprehensive and systematic surveys of members of the HMA gene family have been conducted in the model legume species Medicago truncatula. Here, nine putative MtHMAs were identified in the M. truncatula genome. These MtHMAs were phylogenetically divided into two distinct groups. The members in each group had a relatively conserved gene structure and motif composition. The number of introns in the MtHMAs varied from 5 to 16, with the majority of these genes containing 8 introns. The expression patterns showed that MtHMAs exhibit preferential or distinct expression patterns among different tissues. Finally, the expression patterns of the members of this gene family were verified in the leaves and roots of plants under Cu stress. Our findings will be valuable for the functional investigation and application of members of this gene family in M. truncatula and other related legume species.

The rs58542926C >T (E167K) variant of the transmembrane 6 superfamily member 2 gene (TM6SF2) is associated with increased risks for nonalcoholic fatty liver disease (NAFLD) and type 2 diabetes (T2D). Nevertheless, the role of the TM6SF2 rs58542926 variant in glucose metabolism is poorly understood. We performed a sex-stratified analysis of the association between the rs58542926C >T variant and T2D in multiple cohorts. The E167K variant was significantly associated with T2D, especially in males. Using an E167K knockin (KI) mouse model, we found that male but not the female KI mice exhibited impaired glucose tolerance. As an ER membrane protein, TM6SF2 was found to interact with inositol-requiring enzyme 1 α (IRE1α), a primary ER stress sensor. The male Tm6sf2 KI mice exhibited impaired IRE1α signaling in the liver. In conclusion, the E167K variant of TM6SF2 is associated with glucose intolerance primarily in males, both in humans and mice.

Nano-modification of polyester has become a research hotspot due to the growing demand for high-performance polyester. As a functional carrier, silica nanoparticles show large potential in improving crystalline properties, enhancing strength of polyester, and fabricating fluorescent polyester. Herein, we briefly traced the latest literature on synthesis of silica modifiers and the resultant polyester nanocomposites and presented a review. Firstly, we investigated synthesis approaches of silica nanoparticles for modifying polyester including sol-gel and reverse microemulsion technology, and their surface modification methods such as grafting silane coupling agent or polymer. Then, we summarized processing technics of silica-polyester nanocomposites, like physical blending, sol-gel processes, and in situ polymerization. Finally, we explored the application of silica nanoparticles in improving crystalline, mechanical, and fluorescent properties of composite materials. We hope the work provides a guideline for the readers working in the fields of silica nanoparticles as well as modifying polyester.

The need for a precise regulation of the properties of chiral nematic structures in response to external stimuli is addressed. Self-assembled iridescent coatings are produced under the effect of electrostatic interactions between cellulose nanocrystals and poly(acrylic acid), endowing a high anisotropic dissymmetry (>0.3) and sensitivity to environmental humidity (13.1 nm/1% at 68-75% relative humidity, RH). The phenomena associated with shifts in selective light reflection (green to orange) and polarization, facilitate tunable transmitted colors (blue to orange) at given rotation angles (RA). Such properties are conveniently integrated into a "RH-RA-color" ternary code that is introduced as an anticounterfeiting technology, taking advantage of multicolor patterns that conveniently track with changes in RH and RA. The proposed charge-driven assembly opens new opportunities for chiral nematic materials that enable precise optical sensing and information encryption.

BACKGROUND & AIMS: Either activation of mTORC1 due to loss of Tsc1 (Tuberous Sclerosis Complex 1) or defective hepatic autophagy due to loss of Atg5 leads to spontaneous liver tumorigenesis in mice. The purpose of this study was to investigate the mechanisms of how autophagy impacts mTORC1 activation-mediated liver metabolic changes and tumorigenesis. METHODS: Atg5 Flox/Flox (Atg5F/F) and Tsc1F/F mice were crossed with albumin Cre mice to generate liver-specific Atg5 knockout (L-Atg5 KO), L-Tsc1 KO and L-Atg5/Tsc1 double KO (DKO) mice. These mice were crossed with p62/Sqstm1F/F (p62) and whole body Nrf2 KO mice to generate L-Atg5/Tsc1/p62 and L-Atg5/Tsc1, Nrf2 triple KO (TKO) mice. These mice were housed for various time points up to 12 months, and blood and liver tissues were harvested for biochemical and histological analysis RESULTS: Deletion of Atg5 in L-Tsc1 KO mice inhibited liver tumorigenesis, but increased mortality of L-Tsc1 KO mice accompanied by drastically enhanced hepatic ductular reaction (DR), hepatocyte degeneration and metabolic reprogramming. Deletion of p62 reversed DR, hepatocyte degeneration and metabolic reprogramming as well as the mortality of L-Atg5/Tsc1 DKO mice, but unexpectedly promoted liver tumorigenesis via activation of a group of oncogenic signaling pathways. Nrf2 ablation markedly improved DR with increased hepatocyte population and improved metabolic reprogramming and survival of the L- Atg5/Tsc1 DKO mice without tumor formation. Decreased p62 and increased mTOR activity was also found in a subset of human hepatocellular carcinoma. CONCLUSIONS: These results reveal previously undescribed functions of hepatic p62 in suppressing tumorigenesis and regulating liver cell repopulation and metabolic reprogramming resulting from persistent mTORC1 activation and defective autophagy. LAY SUMMARY: Metabolic liver disease and viral hepatitis are common chronic liver diseases and risk factors of hepatocellular carcinoma, which are often associated with impaired hepatic autophagy with increased mTOR activation. Using multiple genetic engineered mice that are defective of hepatic autophagy with persistent mTOR activation, we dissected the complex interplay among mTOR, autophagy, p62 and Nrf2 on liver cell repopulation, metabolic reprogramming and redox homeostasis in liver tumorigenesis. Our results uncovered an unexpected novel tumor suppressor function of p62/Sqstm1 by regulating liver cell repopulation, ductular reaction and metabolic reprogramming in liver tumorigenesis.

This paper presents a novel two-dimensional (2-D) partial Maxwell fish-eye (PMFE) lens with the capability of wide-angle beam scanning inspired by the Gutman lens and Eaton lens, which is obtained by cutting a part from the 2-D Maxwell fish-eye (MFE) lens along a straight line. In terms of the refractive index profile, the MFE lens is similar to the Gutman lens near the center and the Eaton lens near the edge, respectively. We demonstrate the potential of the PMFE lens in wide-angle beam scanning based on its Gutman-like focusing and Eaton-like rotating characteristics corresponding to different feed points. As an example, a fully metallic PMFE lens antenna in the Ka-band composed of a bed of nails and a series of linearly arranged waveguide feeders is designed and experimentally verified. The measured results reveal wide-angle scanning ranges, especially about ±90° at 36 GHz, low reflections and low mutual couplings. The frequency scanning due to the dispersion of the lens is also discussed.

Wuliangye baijiu is one of the most famous baijiu in China, with a rich, harmonic aroma profile highly appreciated by consumers. Thousands of volatiles have been identified for the unique aroma profile. Among them, fatty acid esters have been identified as the main contributors to the aroma profile. In addition, many non-ester minor compounds, many of which are more polar than the esters, have been identified to contribute to the characteristic aroma unique to Wuliangye baijiu. The analysis of these minor compounds has been challenging due to the dominance of esters in the sample. Thus, it is desirable to fractionate the aroma extract into subgroups based on functional group or polarity to simplify the analysis. This study attempts a new approach to achieve simultaneous volatile extraction and fractionation using tandem LiChrolut EN and silica gel solid-phase extraction (SPE) columns. A baijiu sample (10 mL, diluted in 40 mL of water) was first passed through the LiChrolut EN (1.0 g) column. The loaded LiChrolut EN column was then dried with air and coupled with a silica gel (5.0 g) SPE column with anhydrous Na2SO4 (10.0 g) in between. The volatile compounds were eluted from the LiChrolut EN column and simultaneously fractionated on the silica gel column based on polarity. The simultaneous extraction and fractionation technique enabled the fractionations of all fatty acid esters into less polar fractions. Fatty acids, alcohols, pyrazines, furans, phenols, hydroxy esters, and other polar compounds were collected in more polar fractions. This technique was used to study the volatile compounds in Wuliangye, Moutai, and Fengjiu baijiu. In addition to fatty acid esters, many minor polar compounds, including 2,6-dimethylpyrazine, 2-ethyl-6-methylpyrazine, 2-ethyl-3,5-dimethylpyrazine, p-cresol, and 2-acetylpyrrole, were unequivocally identified in the samples. The procedure is fast and straightforward, with low solvent consumption.

CircPRKDC has been disclosed to participate in the tumorigenesis of serval tumors, but the regulatory mechanisms of circPRKDC in GC are still unknown. CircPRKDC, miR-493-5p, and insulin receptor substrate 2 (IRS2) levels were tested by RT-qPCR. The epithelial-mesenchymal transition (EMT)-related protein levels were evaluated via western blot. The cell viability, migration and invasion were evaluated through CCK-8 and Transwell assays. Luciferase reporter and RIP assays were employed to confirm the binding ability between miR-493-5p and circPRKDC or IRS2. CircPRKDC was upregulated in GC samples, and circPRKDC silencing restrained GC cell viability, metastasis, and EMT and suppressed GC tumor growth. Besides, miR-493-5p was a target of circPRKDC, and the repressive impact of circPRKDC knockdown on GC development was neutralized by miR-493-5p inhibition. Moreover, miR-493-5p targeted IRS2 and IRS2 addition rescued the effects of circPRKDC depletion on GC progression. Finally, circPRKDC knockdown could regulate IRS2 expression by targeting miR-493-5p. These results elaborated that circPRKDC accelerated GC development via sponging miR-493-5p and increasing IRS2, which might provide novel potential targets for GC treatment.

BACKGROUND: Associations of dietary or supplementary intake of several unsaturated fatty acids and mortality have been widely studied but the results were still hitherto inconsistent or limited. It is still need to explore the effects of these fatty acids by using the objective biomarkers. OBJECTIVE: We aimed to investigate the relevancy of several serum n-3 and n-6 fatty acids with all-cause and disease-specific mortality to confirm their health effects and effects on the associations between dietary quality and all-cause mortality. METHODS: A total of 4132 people from NHANES 2003-2004 and 2011-2012 and the mortality information was confirmed from the NDI. CPH models adjusted for known risk factors were conducted to explore the associations between circulating n-3 and n-6 fatty acids and all-cause or CVD or cancer mortality under complex sampling. We further evaluated their effects on association between dietary quality and all-cause mortality. RESULTS: A total of 437 deaths occurred during the mean follow-up of 83.34 months, including 157 CVD death and 100 cancer death. Serum LA, ALA, EPA and DHA were associated with all-cause mortality (HR in quintile5: LA:0.584, 95%CI: 0.387-0.882, Ptrend = 0.011; ALA:0.626, 95%CI: 0.432-0.907, Ptrend = 0.008; EPA:0.535, 95%CI: 0.375-0.764, Ptrend = 0.001; DHA:0.669, 95%CI: 0.468-0.955, Ptrend = 0.031). Additionally, serum EPA and ALA were respectively related to CVD and cancer mortality (Q5 HR: EPA:0.450, 95%CI: 0.23-0.854, Ptrend = 0.009; ALA:0.387, 95%CI: 0.167-0.900, Ptrend = 0.022). Serum AA, GLA, DGLA and SDA were not associated with any risk of mortality. The effect on all-cause mortality of the lower AHEI scores can be improved by adherence to a higher serum LA, EPA and DHA (in the lowest AHEI strata, LA in tertile3 compared to tertile1 HR:0.596, 95%CI: 0.366-0.970; EPA:0.660, 95%CI: 0.454-0.959; DHA:0.666, 95%CI; 0.444-1.000). CONCLUSIONS: Our results support the recent dietary recommendations to increase the intake of plant-derived and marine-derived n-6 and n-3 to improve the ability of primary and secondary prevention.

The suspended monorail (SM) vehicle-bridge system has been considered a promising modern transit mode due to its clear advantages: low pollution, high safety, convenient construction, and low cost. The wind-induced response can significantly affect the running safety and comfort of this type of vehicle due to its special suspended position from a fixed track. This study is the first to systematically investigate its aerodynamic characteristics and interference effects under various spacing ratios using wind tunnel tests and numerical simulations. A high level of agreement between the wind tunnel test and CFD (computational fluid dynamics) results was obtained, and the aerodynamic interference mechanism can be well explained using the CFD technique from a flow field perspective. A wireless wind pressure acquisition system is proposed to achieve synchronization acquisition for multi wind pressure test taps. The paper confirms that (1) the proposed wireless wind pressure acquisition system performed well; (2) the aerodynamic coefficients of the upstream vehicle and bridge were nearly unchanged for vehicle-bridge combinations with varying spacing ratios; (3) the aerodynamic interference effects were amplified when two vehicles meet, but the effects decrease as the spacing ratio increases; (4) the aerodynamic force coefficients, mean, and root mean square (RMS) wind pressure coefficients for the downstream vehicle and bridge are readily affected by the upstream vehicle; (5) the vortex shedding frequencies of vehicles and bridges can be readily obtained from the lift force spectra, and they decrease as the spacing ratio increases; and (6) a spacing ratio of 3.5 is suggested in the field applications to ensure the running safety and stability of the SM vehicle-bridge system under exposure to crosswinds.

OBJECTIVE: The non-coding RNAs have been identified as potential biomarkers in cancers. This study aimed to investigate the effect of long intergenic non-protein coding RNA 467 (LINC00467)/microRNA (miR-125a-3p)/sirtuin 6 (SIRT6) axis on non-small cell lung cancer (NSCLC). METHODS: Expression of LINC00467, miR-125a-3p, SIRT6, and ERK1/2 signaling pathway-related factor in NSCLC was assessed. A549 and H1299 cells were induced by cisplatin (DDP) to establish DDP-resistant cell lines, and the drug resistance was detected. DDP-resistant cells were transfected with relative oligonucleotides and/or plasmids to observe the effects of LINC00467 and miR-125a-3p on NSCLC cells. The interaction among LINC00467, miR-125a-3p, and SIRT6 was detected using dual luciferase reporter gene assay and RNA pull-down assay. A tumor xenograft model was established, and Ki67 expression was detected in tumors. RESULTS: LINC00467 and SIRT6 were upregulated, miR-125a-3p was suppressed, and the ERK1/2 pathway was activated in NSCLC. LINC00467 or SIRT6 inhibition or miR-125a-3p elevation restricted malignancy and DDP resistance of NSCLC cells. Reduced miR-125a-3p reversed the effect of inhibited LINC00467 on NSCLC cells. LINC00467 competitively bound to miR-125a-3p, and miR-125a-3p targeted SIRT6. Inhibiting LINC00467 and SIRT6 or upregulating miR-125a-3p repressed tumor formation in vivo. CONCLUSION: LINC00467 silencing upregulates miR-125a-3p to decrease DDP resistance in NSCLC cells via inhibiting SIRT6 and inactivating the ERK1/2 signaling pathway.

These silaarenes possess a particular aggregation-induced emission behavior. Mechanistic investigations demonstrate that the relay trimetallic transmetalation plays a pivotal role in governing this transformation.

BACKGROUND: Alfalfa, the "queen of forage", is the most extensively cultivated forage legume in the world. The development and yield of alfalfa are seriously limited by abiotic stress. MADS-box transcription factors are one of the largest gene families and play a pivotal role in plant development and abiotic stress. However, little is known regarding the MADS-box transcription factors in autotetraploid cultivated alfalfa. RESULTS: In the present study, we identified 120 MsMADS-box genes in the alfalfa genome. Phylogenetic analysis indicated that 75 type-I MsMADS-box genes were classified into the Mα, Mß, and Mγ subgroups, and 45 type-II MsMADS-box genes were classified into 11 subgroups. The promoter region of MsMADS-box genes containing several hormone and stress related elements. Chromosomal location analysis revealed that 117 MsMADS-box genes were unevenly distributed on 32 chromosomes, and the remaining three genes were located on unmapped scaffolds. A total of nine pairs of segmental duplications and four groups of tandem duplications were found. Expression analysis showed that MsMADS-box genes were differentially expressed in various tissues and under abiotic stresses. qRT-PCR analysis revealed that the expression profiles of eight selected MsMADS-box genes were distinct under various stresses. CONCLUSIONS: In this study, MsMADS-box genes were identified in the cultivated alfalfa genome based on autotetraploid level, and further confirmed by Gene Ontology (GO) analysis, phylogenetic analysis, sequence features and expression analysis. Taken together, these findings will provide clues for further study of MsMADS-box functions and alfalfa molecular breeding. Our study is the first to systematically identify and characterize the MADS-box transcription factors in autotetraploid cultivated alfalfa (Medicago sativa L.), and eight MsMADS-box genes were significantly involved in response to various stresses.

A number of applications require x rays of both high flux and narrow bandwidth. In this work, we experimentally demonstrate the high-efficiency generation of narrowband soft x rays from carbon nanotube foams irradiated by a femtosecond laser pulse at an intensity of 1019W/cm2. The building blocks of the foam, single-walled carbon nanotube bundles with diameters smaller than the laser skin length can be volumetrically heated and fully ionized on a femtosecond time scale. The three-dimensional network structure of the foam permits deep penetration and drastic absorption of the laser pulse, and results in bright line emissions without prominent Stark broadening. A single-shot yield of 3×1014photons in the carbon Lyα line at 3.37 nm was measured with a bandwidth of 0.013 nm.
