RESUMO

Enterovirus 71 (EV71) is a causative agent of hand, foot and, mouth disease (HFMD) in young children. It is valuable for virologists to develop a fast method to rescue infectious virus from a viral cDNA clone. Here, we report a method for rapid rescue of infectious EV71 by using cells expressing T7 polymerase. The full-length EV71 genome was amplified in one step by long-distance PCR with a T7 promoter at the 5' end, and the T7 polymerase gene was cloned into a lentivirus vector for construction of a stable cell line expressing T7 polymerase. The infectious virus was rapidly and efficiently rescued by single transfection of cells with the infectious cDNA clone. Further experiments showed that the rescued virus had characteristics similar to those of the parental virus. This method circumvented the difficulty in performing in vitro transcription of a long linear DNA to obtain high-quality RNA. The construction of the viral cDNA clone and the fast rescue of the infectious virus will greatly benefit future investigations.

To explore the biological properties of rosin derivatives, two dehydroabietic acid derivatives N-(5-dehydroabietyl-1,3,4-thiadiazole)-yl-pyridine-2-carboxamide (DTPC) and di-N-(5-dehydroabietyl-1,3,4-thiadiazole)-yl-pyridine-2,6-carboxamide (DDTPC) with 1,3,4-thiadiazole, pyridine and amide moieties were designed and synthesized according to superposition principle of activity group. They interact with calf thymus DNA (CT DNA) via intercalation based on the results of circular dichroism (CD) and fluorescence spectroscopy, DNA denaturation and viscosity studies. Fluorescence and CD spectral experiments indicate that they might be transported and stored by protein like bovine serum albumin (BSA). MTT assay was further carried out to examine their cytotoxicity, they both showed selective cytotoxicity and DTPC exhibited better cytotoxicity. The antiproliferative effect of DTPC toward A431 cell line was stronger than that of clinically used cisplatin and oxaliplatin. In addition, the cytotoxicity of DTPC and DDTPC was closely related with their DNA binding ability.

Planktonic microorganisms play a key role in the biogeochemical processes of the aquatic system, and they may be affected by many factors. High-throughput sequencing technology was used in this study to investigate and study the bacterioplankton community of water bodies in the upper reaches of the Heihe River Basin in Qinghai Plateau. Results showed that Proteobacteria, Firmicutes, Bacteroidetes and Actinobacteria are the predominant phyla in this river section, while the main genera are Thiomonas, Acidibacillus, Acidocella, Rhodanobacter, Acidithiobacter and Gallionella, which are autochthonous in the acid-mine drainage. Additionally, total nitrogen, total phosphorus, permanganate index and pH are significantly correlated with the bacterioplankton abundance and are the main limiting factors for the spatial distribution of the bacterioplankton. PICRUSt inferred that the mainstream microbial assemblages had a higher abundance of KOs belong to metabolism of terpenoids and polyketides, while the tributary had higher abundance of KOs belong to the immune system. The relationship between bacterioplankton community composition and environmental factors in the Heihe River basin was discussed for the first time in this study, which provides a theoretical basis for the healthy, orderly development of the water environment in the Heihe River Basin.

As an indispensable part of the Maolan karst forest soil microorganisms, eukaryotic microbes play an important role in ecosystem material and energy flow. To investigate the composition and diversity of soil eukaryotic microbial communities under different succession stages in the Maolan karst forest, we explored the diversity and composition of soil eukaryotic microbes under four succession stages of primary forests (YSL), shrub forests (GML), bushes (GMC), and grassland (CD) via high-throughput sequencing (HTS) of the 18S rDNA. The results showed that the composition of soil eukaryotic microbial communities was similar in different succession stages under different classification levels. There was a significant difference in α diversity, Shannon, and Simpson's indices at different stages of succession as follows: YSL > GMC > GML > CD. The non-metric multidimensional scaling analysis showed that there were differences in the soil eukaryotic microbial community structure at different succession stages. The linear discriminant analysis effect size (LEfSe) analysis showed that the number of differential indicator species in YSL was higher than in GML, GML, and CD. The results of our study provide a theoretical basis for further research on soil eukaryotic microorganisms in different succession stages.

In order to clarify the effects of chelate tetrasodium glutamate diacetate (GLDA) on the remediation of cadmium (Cd) contaminated farmland by Pennisetum purpureum Schum, GLDA was applied in different methods within 60 days:total application doses of 585, 1170, and 2340 kg·hm-2 were equally divided into 1-4 applications, respectively, and the time intervals of 2-4 applications were 30, 20, and 15 days, respectively. The biomass, Cd content, and amount of Cd extracted from aboveground parts of Pennisetum purpureum Schum, in addition to the pH, dissolved organic carbon (DOC) mass concentration, and other indicators in soil were analyzed. The results showed that the biomass and Cd content of the aboveground parts of Pennisetum purpureum Schum increased significantly when GLDA was applied many times at a low application dose. The number of applications and the total dose were the key factors influencing the biomass and Cd content, respectively. The highest amount of Cd extracted was 16.78 g·hm-2 at 585 kg·hm-2 (applied four times), which was 275.39% higher than the CK treatment (i.e., no GLDA was applied). There was significant positive correlations between the pH, mass concentration of DOC, and content of DTPA-Cd, and the total dose and number of applications. The mass concentration of DOC was the main factor affecting the DTPA-Cd content. The total Cd content in the soil (after at 585 kg·hm-2 was applied four times) decreased by 3.23% compared with that of the soil before planting Pennisetum purpureum Schum. Therefore, the application of GLDA is of great significance for the remediation of Cd contaminated farmland by Pennisetum purpureum Schum, and the application method should be selected reasonably.

Due to the unsatisfactory robustness of current predictive biomarkers in many cases, application of immunotherapy in advanced cancers with limited treatment options, such as stage IV intrahepatic cholangiocarcinoma (ICC), was quite common. Hence, strategies to enhance the therapeutic effect of immunotherapy or to extend the scope of potential beneficial patients were urgently needed. Combination of radiotherapy and anti-programmed death receptor-1 (PD-1) immunotherapy was a promising one, since they were found to have a synergistic anti-tumor effect in animal models and a couple of patients. We here present a 68-years-old male with chemotherapy-intolerable stage IV ICC, whose primary tumor had low PD-L1 expression level, scarce CD8+ cells in tumor microenvironment, high microsatellite instability (MSI), and high tumor mutation burden (TMB). These biomarkers showed a conflicting prediction of the treatment response and clinical benefit of anti-PD-1 immunotherapy. Combination therapy of anti-PD-1 immunotherapy and radiotherapy was adopted as first-line treatment for the patient. After six cycles of immunotherapy, shrinkage of the primary liver tumor and metastatic lymph nodes happened, alongside with new lung metastasis, which indicated a mixed response. Radiotherapy was then administered to both the liver and lung lesions, accompanied with continued immunotherapy. The combined therapy eventually led to a complete response for both the primary tumor and all metastases without treatment-related adverse effects. The patient has survived for 26 months after the combined therapy and remains tumor-free currently. This case demonstrates the high inconsistency between immunotherapy response biomarkers and the synergetic anti-tumor effect of immunotherapy and radiotherapy in ICC.

Dietary restriction has been well-described to improve health metrics, but whether it could benefit pathophysiological adaptation to extreme environment, for example, microgravity, remains unknown. Here, we investigated the effects of a daily rhythm of fasting and feeding without reducing caloric intake on cardiac function and metabolism against simulated microgravity. Male rats under ad libitum feeding or time-restricted feeding (TRF; food access limited to 8 hours every day) were subjected to hindlimb unloading (HU) to simulate microgravity. HU for 6 weeks led to left ventricular dyssynchrony and declined cardiac function. HU also lowered pyruvate dehydrogenase (PDH) activity and impaired glucose utilization in the heart. All these were largely preserved by TRF. TRF showed no effects on HU-induced loss of cardiac mass, but significantly improved contractile function of cardiomyocytes. Interestingly, TRF raised liver-derived fibroblast growth factor 21 (FGF21) level and enhanced cardiac FGF21 signaling as manifested by upregulation of FGF receptor-1 (FGFR1) expression and its downstream markers in HU rats. In isolated cardiomyocytes, FGF21 treatment improved PDH activity and glucose utilization, consequently enhancing cell contractile function. Finally, both liver-specific knockdown (KD) of FGF21 and cardiac-specific FGFR1 KD abrogated the cardioprotective effects of TRF in HU rats. These data demonstrate that TRF improves cardiac glucose utilization and ameliorates cardiac dysfunction induced by simulated microgravity, at least partially, through restoring cardiac FGF21 signaling, suggesting TRF as a potential countermeasure for cardioprotection in long-term spaceflight.

Sishen Pill (SSP) is a typical prescription in the pharmacopeia of traditional Chinese medicine (TCM), and is usually used to treat inflammatory bowel disease (IBD). It is known that inflammatory dendritic cells (DCs) and imbalance of gut microbiota play significant roles in the pathogenesis of IBD. However, it is not clear whether SSP can treat IBD by regulating interaction of DCs and gut microbiota. In the present study, the levels of inflammatory DCs and gut microbiota were analyzed by flow cytometry and 16S rDNA analysis. SSP relieved the pathological damage to the colon of mice with colitis induced by dextran sodium sulfate (DSS). As typical indicators of inflammatory DCs, the levels of CD11c+CD103+E-cadherin+ cells and pro-inflammatory cytokines [interleukin (IL)-1ß, -4, -9, and -17A] were decreased in mice with colitis treated by SSP for 10 days. Simultaneously, the gut microbiota composition was regulated, and beneficial bacteria were increased and pathogenic bacteria were reduced. The results indicated that SSP regulated the interaction between inflammatory DCs and gut microbiota to treat DSS-induced colitis.

Immune memory has a protective effect on the human body, but abnormal immune memory is closely related to the occurrence and development of autoimmune diseases including inflammatory bowel disease (IBD). Sishen Pill (SSP) is a classic prescription of traditional Chinese medicine, which is often used to treat chronic colitis, but it is not clear whether SSP can alleviate experimental colitis by remodeling immune memory. In the present study, the therapeutic effect of SSP on chronic colitis induced by dextran sulfate sodium (DSS) was evaluated by colonic length, colonic weight index, macroscopic and microscopic scores, and pathological observation. The cytokine levels were tested by enzyme-linked immunosorbent assay (ELISA); the percentages of central memory T (Tcm) and effector memory T (Tem) cells were analyze\d by flow cytometry; and activation of phosphoinositide 3-kinase (PI3K)/Akt signaling proteins was measured by western blotting. After 7-days' treatment, SSP alleviated DSS-induced colitis, which was demonstrated by decreased colonic weight index, colonic weight, histopathological injury scores, restored colonic length, gradual recovery of colonic mucosa, and lower levels of interleukin (IL)-2, IL-7, IL-12, and IL-15, while SSP increased IL-10 expression. SSP obviously regulated the quantity and subpopulation of Tcm and Tem cells. Furthermore, SSP markedly inhibited activation of PI3K, Akt, phospho-Akt, Id2, T-bet, forkhead box O3a, Noxa, and C-myc proteins in the PI3K/Akt signaling pathway and activated Rictor, Raptor, tuberous sclerosis complex (TSC)1, TSC2, phospho-AMP-activated kinase (AMPK)-α, AMPK-α, eukaryotic translation initiation factor 4E-binding protein 2, kinesin family member 2a, and 70-kDa ribosomal protein S6 kinase. These results indicate that SSP effectively controls Tem cells in the peripheral blood to relieve experimental colitis induced by DSS, which were potentially related with inhibiting the PI3K/Akt signaling pathway.

Objective: To reveal the risk factors, the symptom distribution characteristics, the clinical values of white blood cell counts (WBC counts), red blood cell distribution width (RDW), neutrophil-to-lymphocyte ratio (NLR), platelet-to-lymphocyte ratio (PLR) and monocyte-to-lymphocyte ratio (MLR) in hospitalized patients with acute exacerbation of chronic obstructive pulmonary disease (AECOPD) combined with depression and/or anxiety. Methods: The study included prospective cross-sectional and case-control studies, and was executed in the Affiliated Hospital of Zunyi Medical University, Guizhou, China. Previously diagnosed chronic obstructive pulmonary disease (COPD) patients who admitted to the hospital with AECOPD, patients with depression and/or anxiety, and healthy people were enrolled in the study. The Hamilton Rating Scales were used to assess all subjects, and the complete blood counts (CBC) were collected. Baseline data and clinical measurement data [spirometry, arterial blood gas analysis, and COPD evaluation test (the CAT scale)] from patients with AECOPD were collected. Results: Of the 307 patients with AECOPD included, 63.5% (N=195) had depressive and/or anxiety symptoms, and 36.5% (N=112) had no symptoms. Sex, respiratory failure, number of comorbidities, number of acute exacerbations in the previous year and the CAT score were closely related to AECOPD combined with depression and/or anxiety (p<0.05). The CAT scale score were the independent risk factor (OR=6.576, 95% CI 3.812-11.342) and significant predictor of AECOPD with depression and/or anxiety (AUC=0.790,95% CI 0.740-0.834); the patients with depression and/or anxiety were more severe and characteristic than the patients with AECOPD combined with depression and/or anxiety; RDW was associated with AECOPD with depression and/or anxiety (p=0.020, OR1.212,95% CI1.03-1.426), and had certain clinical diagnostic value (AUC=0.570,95% CI 0.531-0.626). Conclusion: Depression and anxiety should not be ignored in patients with AECOPD. The severity and quality of life of COPD were closely related to the occurrence of depression and/or anxiety symptoms. In most cases, perhaps depression and anxiety in AECOPD are only symptoms and not to the extents of the diseases. RDW had clinical diagnostic value in AECOPD combined with depression and/or anxiety. NLR, PLR, MLR, and RDW may become the novel indicators for evaluating the degree of inflammation of AECOPD and deserve further research.

To explore the effects of water management mode on Cd environmental behavior in different parent-material-developed paddy soils, two parent-material-developed paddy soils (yellow clayey soil and granitic sandy soil) under three exogenous Cd levels (0.5, 2.0, and 5.0 mg·kg-1) with different water management modes (long-term flooding, moistening irrigation, and wet-dry rotation) were cultured in this study. The soil redox potential (Eh value), pH value, Cd concentration in soil solution, and Cd fractionation were also determined. The results showed that water management mode had different effects on the pH and Eh values of soils developed from different parent materials. The change rates of soil pH value were as follows:long-term flooding:-2.61% (yellow clayey soil), 2.25% (granitic sandy soil); alternation of dry and wet:-1.96% (yellow clayey soil); 0.52% (granitic sandy soil); wet irrigation:-4.08% (yellow clayey soil) and -0.52% (granitic sandy soil). The Eh value of the soils was negatively correlated with the pH value. The influence pattern of water management model on Cd mass concentration of soil solutions in two parent-material soils was consistent. The Cd mass concentration of soil solutions in granitic sandy soil was higher than that in yellow clayey soil. The mean values of Cd concentration were 1.03 µg·L-1 for yellow clayey soil and 1.07 µg·L-1 for granitic sandy soil. Water management mode had no significant effect on the proportions of organic bound Cd or Fe-Mn bound Cd in two different parent-material-developed soils. The long-term flooding mode promoted the transformation of exogenous Cd to residual Cd, and this promotion in yellow clayey soil was higher than that in granitic sandy soil. In conclusion, during the process of regulating soil Cd bioavailability through water management, the role of soil parent materials needs to be considered.

The T follicular helper T (Tfh) cells play a significant role in the pathogenesis of inflammatory bowel disease (IBD), which is regulated by the Bcl-6/Blimp-1 pathway. Some studies have suggested that regulating activation of the Bcl-6/Blimp-1 pathway should be an effective method to treat IBD. Sishen Pill (SSP) has been used frequently to treat chronic colitis. Its mechanism is related to the downstream proteins in the Bcl-6/Blimp-1 pathway. However, it is unknown whether SSP regulates the function and differentiation of Tfh cells to treat IBD. In the present study, chronic colitis was induced by dextran sodium sulfate and treated with SSP for 7 days. SSP effectively treated chronic colitis, regulated the balance between Tfh10, Tfh17 and T follicular regulatory cells, while SSP increased the Blimp-1 level, inhibited expressions of Bcl-6, T-cell costimulator, programmed death (PD)-1 and PD-ligand 1 on the surface of Tfh cells. SSP inhibited activation of BcL-6, phosphorylated signal transducer and activator of transcription (p-STAT)3, signal lymphocyte activation molecule (SLAM)-associated protein but improved Blimp-1 and STAT3 expression in colonic tissues. The results indicated that SSP regulated the differentiation and function of Tfh cells to treat IBD, which was potentially related with inhibiting the Bcl-6/Blimp-1 pathway.

Long intergenic non-protein-coding RNA 00205 (LINC00205) has been found to play crucial roles in hepatocellular carcinoma progression. In this study, we aimed to determine the expression pattern of LINC00205 in retinoblastoma (RB), to identify its functions in RB progression in detail, and to reveal the underlying mechanisms. Herein, we showed that LINC00205 is highly expressed in RB tissues and cell lines. The LINC00205 upregulation correlated with adverse clinicopathological parameters and shorter overall survival in patients with RB. LINC00205 depletion decreased the proliferative, migratory, and invasive abilities; promoted the apoptosis of RB cells in vitro; and impeded the tumor growth of RB cells in vivo. Mechanism investigation revealed that LINC00205 can act as a competing endogenous RNA by sponging microRNA-665 (miR-665) in RB cells, thereby upregulating miR-665's target: high-mobility group box 1 (HMGB1). Finally, rescue experiments confirmed that enhancing the miR-665-HMGB1 axis output attenuated the influence of the LINC00205 knockdown on RB cells. To sum up, the newly identified LINC00205-miR-665-HMGB1 pathway was systematically studied and may be validated as a potential target for RB diagnosis, prognosis, and therapy.

The effect of the growth of ambient pressures on the penetration of laser welded molybdenum (Mo) alloy was explored. It was found that when ambient pressure rose from 0.1 MPa to 1.8 MPa, the penetration of base metal (BM) was significantly reduced, which was only 17% of that obtained under ambient pressure of 0.1 MPa. Moreover, the mechanism underlying the significant reduction of the penetration of BM was analyzed. At first, by using a high-resolution scanning electron microscope (SEM), the size and the number of nano-sized metallic particles generated during laser welding under different ambient pressures were surveyed. Furthermore, the scattering and absorption of the nano-sized metallic particles for laser energy under different ambient pressures were investigated; afterwards, by applying a high-speed camera and a spectrometer, the transient behaviors and spectral signals of plasmas during fiber laser spot welding under different ambient pressures were monitored. On this basis, the inverse bremsstrahlung absorption of plasmas for laser energy under different ambient pressures was explored; finally, fiber laser spot welding test was carried out on glass/metal composite samples under different ambient pressures to survey the influence of the change of ambient pressure on dynamic behaviors of the molten pool during the welding.

PURPOSE: To test the technical reproducibility of acquisition and scanners of CT image-based radiomics model for early recurrent hepatocellular carcinoma (HCC). METHODS: We included primary HCC patient undergone curative therapies, using early recurrence as endpoint. Four datasets were constructed: 109 images from hospital #1 for training (set 1: 1-mm image slice thickness), 47 images from hospital #1 for internal validation (sets 2 and 3: 1-mm and 10-mm image slice thicknesses, respectively), and 47 images from hospital #2 for external validation (set 4: vastly different from training dataset). A radiomics model was constructed. Radiomics technical reproducibility was measured by overfitting and calibration deviation in external validation dataset. The influence of slice thickness on reproducibility was evaluated in two internal validation datasets. RESULTS: Compared with set 1, the model in set 2 indicated favorable prediction efficiency (the area under the curve 0.79 vs. 0.80, P = 0.47) and good calibration (unreliability statistic U: P = 0.33). However, in set 4, significant overfitting (0.63 vs. 0.80, P < 0.01) and calibration deviation (U: P < 0.01) were observed. Similar poor performance was also observed in set 3 (0.56 vs. 0.80, P = 0.02; U: P < 0.01). CONCLUSIONS: CT-based radiomics has poor reproducibility between centers. Image heterogeneity, such as slice thickness, can be a significant influencing factor.

Eukaryotic chromatin is tightly packed into hierarchical structures, allowing appropriate gene transcription in response to environmental and developmental cues. Here, we provide a chromosome-scale de novo genome assembly of sesame with a total length of 292.3 Mb and a scaffold N50 of 20.5 Mb, containing estimated 28,406 coding genes using Pacific Biosciences long reads combined with a genome-wide chromosome conformation capture (Hi-C) approach. Based on this high-quality reference genome, we detected changes in chromatin architectures between normal growth and dark-treated sesame seedlings. Gene expression level was significantly higher in 'A' compartment and topologically associated domain (TAD) boundary regions than in 'B' compartment and TAD interior regions, which is coincident with the enrichment of H4K3me3 modification in these regions. Moreover, differentially expressed genes (DEGs) induced by dark treated were enriched in the changed TAD-related regions and genomic differential contact regions. Gene Ontology (GO) enrichment analysis of DEGs showed that genes related to 'response to stress' and 'photosynthesis' functional categories were enriched, which corresponds to dark treatment. These results suggested that chromatin organization is associated with gene transcription in response to dark treatment in sesame. Our results will facilitate the understanding of regulatory mechanisms in response to environmental cues in plants.

By conducting field positioning experiments, we studied the development trend of Cd pollution in a typical paddy system. The samples of atmospheric deposition and irrigation water were collected monthly from November 2015 to November 2018 during which fertilizer, soil, and rice samples were also collected. The Cd concentration in the samples was monitored and analyzed to conduct research on the balance between Cd inputs and outputs in a typical paddy system in Hunan Province. The results suggest that through irrigation water, atmospheric deposition and fertilizer, the average annual input of Cd in the paddy field system is 8.735 g·(hm2·a)-1, of which atmospheric deposition, the major source, accounts for 69.15%-82.04% of the total input, with an average of 76.61%. This is followed by irrigation water and fertilizer, respectively, accounting for 12.62%-23.66% and 5.34%-7.19%, with an average of 16.94% and 6.45%, respectively. Through surface runoff, soil infiltration and the rice harvest of the aboveground portion, the annual average output of Cd contained in the paddy system is 7.093 g·(hm2·a)-1. Rice harvest accounts for 85.27%-95.02% of the total output, with an average of 89.69%; surface runoff accounted for 4.57%-13.96% of the total output, with an average of 9.41%; and soil infiltration accounted for 0.41%-1.51% of the total output, with an average of 0.90%. The study indicates that Cd contained in paddy systems in Central Hunan exhibits a net input, and the soil Cd pollution is increasing as a result. Straw returning and straw removal have an important impact on the soil Cd balance, and straw removal can slow the trend of soil Cd pollution accumulation.

Research of bacterial communities and metabolism potential of paddy soils contaminated by antimony (Sb) and arsenic (As) are vital to acquire understanding for their bioremediation. Here, the relative abundance of Sb and As metabolism genes, the diversity and composition of the bacterial community, and the influences of geochemical properties and the bacterial community and metabolism potential have been researched by Tax4Fun2 prediction and high-throughput sequencing. LEfSe (linear discriminant analysis effect size) analysis shown different taxa were enriched in dissimilar soil layers. RDA (Redundancy analysis) and relative importance analysis indicated the main properties including total sulfur (TS), total organic carbon (TOC), pH, and the bioavailable fractions of Sb and As affects the bacterial community, which Sbrec, Astot, and Asrec had greater impact on the bacterial taxonomic community. For example, Asrec, Astot, and Sbrec had a positive correlation with Chloroflexi and Rokubacteria, but negatively correlated with Proteobacteria and Actinobacteria. Obtaining metabolic function genes by using the tax prediction method. RDA, relative importance analysis, and co-occurrence network analysis showed the geochemical properties and bacterial community affected Sb and As related bacterial functions. The partial least squares path model (PLS-PM) analysis indicated Sb and As contamination fractions had negative effects on ecological function, bacterial community structure had positive influences on ecological function, and the direct effects of geochemical properties on ecological function was greater than community structure. The direct impact of As contamination fractions on bacterial community structure was greater than Sb, while the direct impact of Sb contamination fractions on bacterial function was more remarkable than As. Obviously, this study provides a scientific basis for the potential of biochemical remediation of Sb and As contamination in paddy soils profile.
