RESUMO

The high chlorine (Cl) and lead (Pb) content characteristics of municipal solid waste incineration fly ash (MSWIFA) pose environmental risks and hinder resource utilization. Herein, an MSWIFA-based magnesium potassium phosphate cement (MKPC) preparation strategy was developed, which allowed the MSWIFA recycling and the Pb-Cl synergistic immobilization without the washing pretreatment. The compressive strength of the resulting 10 wt% MSWIFA-based MKPC was 28.44 MPa, with over 99.2% reduction in leaching toxicity of Pb and Cl. The high-angle annular dark field scanning transmission electron microscope (HAADF-STEM) and X-ray absorption spectroscopy (XAS) analyzes showed that Pb, phosphate and Cl- formed Pb5(PO4)3Cl in MKPC. In-situ X-ray diffraction (XRD) tests showed that Pb3(PO4)2 was gradually transformed to Pb5(PO4)3Cl through a dissolution-precipitation process. The formation energy, Bader charge, charge density difference and density of states (DOS) of Pb5(PO4)3Cl were analyzed by first-principles calculations, confirming that Pb5(PO4)3Cl was more thermodynamically stable than Pb3(PO4)2 and PbCl2 and that electronic interactions between Pb-p, O-p, P-p and Cl-p orbits were the origin of Pb-Cl synergistic immobilization. This work provides a new strategy for the resource utilization of MSWIFA without washing pretreatment, and provides an in-depth understanding of the Pb-Cl synergistic immobilization mechanism.

Polypropylene has been used for applications requiring high mechanical properties, good adhesion, chemical stability and insulation. Whereas, Polypropylene itself is flammable, and its limiting oxygen index (LOI) is low, which cannot pass the UL-94 combustion test. Therefore, extensive use will cause a serious threat to human life and property. With the wide application of thermoplastic polypropylene in industry, the development of environmentally friendly flame retardant materials has become an important research direction. For the past dozen years, researchers have been exploring flame retardants with high flame retardant efficiency, low toxicity, less smoke or other excellent performance flame retardants. This paper reviews the research progress of some phosphorus-containing flame retardants on the flame retardant properties of polypropylene in recent years. Phosphorus flame retardant is a flame retardant with high flame retardant efficiency, good stability and wide application. The types and flame retardant properties of phosphorus flame retardant will be introduced, and the future research of phosphorus flame retardant is summarized, direction and development opportunities.

Objective: We aimed to construct and validate machine learning models for endotracheal tube (ETT) size prediction in pediatric patients. Methods: Data of 990 pediatric patients underwent endotracheal intubation were retrospectively collected between November 2019 and October 2021, and separated into cuffed and uncuffed endotracheal tube subgroups. Six machine learning algorithms, including support vector regression (SVR), logistic regression (LR), random forest (RF), gradient boosting tree (GBR), decision tree (DTR) and extreme gradient boosting tree (XGBR), were selected to construct and validate models using ten-fold cross validation in training set. The optimal models were selected, and the performance were compared with traditional predictive formulas and clinicians. Furthermore, additional data of 71 pediatric patients were collected to perform external validation. Results: The optimal 7 uncuffed and 5 cuffed variables were screened out by feature selecting. The RF models had the best performance with minimizing prediction error for both uncuffed ETT size (MAE = 0.275 mm and RMSE = 0.349 mm) and cuffed ETT size (MAE = 0.243 mm and RMSE = 0.310 mm). The RF models were also superior in predicting power than formulas in both uncuffed and cuffed ETT size prediction. In addition, the RF models performed slightly better than senior clinicians, while they significantly outperformed junior clinicians. Based on SVR models, we proposed 3 novel linear formulas for uncuffed and cuffed ETT size respectively. Conclusion: We have developed machine learning models with excellent performance in predicting optimal ETT size in both cuffed and uncuffed endotracheal intubation in pediatric patients, which provides powerful decision support for clinicians to select proper ETT size. Novel formulas proposed based on machine learning models also have relatively better predictive performance. These models and formulas can serve as important clinical references for clinicians, especially for performers with rare experience or in remote areas.

Improving the level of autonomy during the landing phase helps promote the full-envelope autonomous flight capability of unmanned aerial vehicles (UAVs). Aiming at the identification of potential landing sites, an end-to-end state estimation method for the autonomous landing of carrier-based UAVs based on monocular vision is proposed in this paper, which allows them to discover landing sites in flight by using equipped optical sensors and avoid a crash or damage during normal and emergency landings. This scheme aims to solve two problems: the requirement of accuracy for runway detection and the requirement of precision for UAV state estimation. First, we design a robust runway detection framework on the basis of YOLOv5 (you only look once, ver. 5) with four modules: a data augmentation layer, a feature extraction layer, a feature aggregation layer and a target prediction layer. Then, the corner prediction method based on geometric features is introduced into the prediction model of the detection framework, which enables the landing field prediction to more precisely fit the runway appearance. In simulation experiments, we developed datasets applied to carrier-based UAV landing simulations based on monocular vision. In addition, our method was implemented with help of the PyTorch deep learning tool, which supports the dynamic and efficient construction of a detection network. Results showed that the proposed method achieved a higher precision and better performance on state estimation during carrier-based UAV landings.

Cell-cell communication and physical interactions play a vital role in cancer initiation, homeostasis, progression, and immune response. Here, we report a system that combines live capture of different cell types, co-incubation, time-lapse imaging, and gene expression profiling of doublets using a microfluidic integrated fluidic circuit that enables measurement of physical distances between cells and the associated transcriptional profiles due to cell-cell interactions. We track the temporal variations in natural killer-triple-negative breast cancer cell distances and compare them with terminal cellular transcriptome profiles. The results show the time-bound activities of regulatory modules and allude to the existence of transcriptional memory. Our experimental and bioinformatic approaches serve as a proof of concept for interrogating live-cell interactions at doublet resolution. Together, our findings highlight the use of our approach across different cancers and cell types.

OBJECTIVE: Previous observational studies have shown that the levels of tumor necrosis factor (TNF) increased in patients with schizophrenia. The present two-sample Mendelian randomization (MR) study aims to identify the causal link between TNF and schizophrenia. METHODS: To date, the largest genome-wide association study (GWAS) for TNF (n = 23 141) and for schizophrenia (53 386 cases and 77 258 controls) was used. All participants were of European ancestry. The MR-egger_intercept test and Cochran's Q statistic were used to determine the pleiotropy and heterogeneity, respectively. Weighted median and inverse variance weighted (IVW) were used to evaluate the causal association of TNF with schizophrenia. RESULTS: We found no significant pleiotropy or heterogeneity of all three selected plasma TNF genetic instrumental variants in breast cancer GWAS. Interestingly, the odds ratio (OR) = 1.517 with 95% confidence interval (CI), 1.006-2.288 and P = 0.047 of schizophrenia correspond to one unit increase in natural log-transformed TNF levels using IVW method. The increased trend was further proven using weighted median (OR = 1.585; 95% CI, 1.017-2.469; P = 0.042). Reverse MR analysis shows no causal effect of schizophrenia on plasma TNF levels. CONCLUSIONS: Our analysis suggested a causal association between genetically increased TNF signaling and increased risk of schizophrenia in the European population. Thus, TNF may be a potential risk for schizophrenia.

Inflammatory lung injury is a common respiratory disease with limited therapeutic effects. Increasing opinions approved that prevention is more important than drug treatment for inflammatory lung injury. Astragalus polysaccharides (APS) has multiple bioactivities including anti-inflammation and immunoregulation. However, its preventive effects on inflammatory lung injury remain unclear. In this study, mice were pretreated with APS via intragastric gavage and then were intratracheally instilled with lipopolysaccharides (LPS) to determine the role of APS in preventing lung injury. The results showed that APS pre-treatment improved the pathological changes of lung tissues, reduced the neutrophils infiltration, and inhibited the LPS-induced inflammation. Increasing evidence confirmed the close relationship between intestinal microbiota and lung inflammatory response. 16S rRNA analysis showed that APS treatment changed the microbiota composition in colon, increased the abundance of short-chain fatty acids (SCFAs)-producing genus such as Oscillospira, Akkermansia, and Coprococcus. Also, APS treatment significantly increased the serum concentrations of SCFAs including butyrate and propionate, and their anti-inflammation effects were demonstrated on mice primary alveolar macrophages. Our data confirmed the preventive effects of APS on LPS-induced lung injury, which were partly contributed by the alteration of intestinal microbiota composition and the resulting increase of serum SCFAs.

Hypoglycemia is a limiting factor for blood glucose management. Serious symptoms such as seizures, and coma may occur during severe hypoglycemia, and nocturnal hypoglycemia is particularly dangerous for patients with type 1 diabetes (T1D). An effective early alarm method is essential for hypoglycemia prevention but challenging, as blood glucose is affected by many factors and the hypoglycemia sequence patterns vary from person to person. In this paper, we proposed a hypoglycemia early alarm method for mining the hidden information in blood glucose based on multi-dimensional sequential pattern mining. The blood glucose, meal, and insulin time series information were used to construct a multi-dimensional database, then the UniSeq algorithm was used to extract multi-dimensional hypoglycemia sequence patterns. Hypoglycemia early alarm was realized through pattern matching with real-time blood glucose. The public OhioT1DM dataset was used for performance evaluation. The experiment results were: 75.76% Sensitivity, 75% Precision, 75.38% F1 score, and 25.17 minutes early alarm time. The result verified that multi-dimensional sequential pattern mining can extract more hidden information and demonstrate more potential significance in providing comprehensive diagnosis support for personalized treatment. Furthermore, early alarms for potential hypoglycemia can also reserve sufficient time for blood glucose management.

Non-alcoholic fatty liver disease (NAFLD) has become a growing public health problem. However, the complicated pathogenesis of NAFLD contributes to the deficiency of effective clinical treatment. Here, we demonstrated that liver-specific loss of Arid2 induced hepatic steatosis and this progression could be exacerbated by HFD. Mechanistic study revealed that ARID2 repressed JAK2-STAT5-PPARγ signaling pathway by promoting the ubiquitination of JAK2, which was mediated by NEDD4L, a novel E3 ligase for JAK2. ChIP assay revealed that ARID2 recruited CARM1 to increase H3R17me2a level at the NEDD4L promoter and activated the transcription of NEDD4L. Moreover, inhibition of Jak2 by Fedratinib in liver-specific Arid2 knockout mice alleviated HFD-induced hepatic steatosis. Downregulation of ARID2 and the reverse correlation between ARID2 and JAK2 were also observed in clinical samples. Therefore, our study has revealed an important role of ARID2 in the development of NAFLD and provided a potential therapeutic strategy for NAFLD.

Background: The role of serum uric acid (SUA) in affecting outcomes after endovascular treatment (EVT) in patients with ischemic stroke remains unclear. This study investigated the association of SUA with outcomes of patients with acute large vessel occlusion (LVO) who had received EVT. Methods: Patients with acute LVO stroke who underwent EVT within 24 hours were enrolled from a prospective, nationwide registry study. Baseline characteristics and SUA level within 24 hours of EVT were collected. The primary outcome was an excellent 90-day functional outcome [modified Rankin Scale (mRS) score 0-1]. Secondary outcomes included a favorable 90-day outcome (mRS score 0-2), symptomatic intracranial hemorrhage (sICH), and 90-day mortality. The SUA level was analyzed in quartiles and as a continuous variable. We investigated the independent association of SUA with the primary outcome using multivariable logistic regression. Results: Among 780 patients (mean age 64 years; 66.28% males), 230 (29.49%) had an excellent 90-day outcome. A higher SUA level was significantly associated with an excellent outcome in univariate logistic regression (P=0.045) and after adjusting for confounders in multivariate analysis [adjusted odds ratio (aOR), 0.998; 95% confidence interval (CI), 0.996-1.000; P=0.018]. Multivariate logistic regression analysis showed patients with SUA level in the fourth quartile had an excellent 90-day outcome (aOR, 0.367; 95% CI, 0.154-0.876; P=0.024). There was no significant association for SUA level with favorable 90-day outcome, sICH, or 90-day mortality (P>0.05). Conclusions: Among patients with acute LVO type of stroke who received EVT, baseline high SUA level may predict a better 90-day functional outcome.

Dithiothreitol (DTT) was adopted as a nucleophile to develop a new acid-catalyzed degradation method for grape seed proanthocyanidin extraction (GSPE). Backpropagation neural network and Box-Behnken design were employed and compared to establish the optimized degradation conditions. GSPE was reacted with DTT at a ratio of 1:1 under mild conditions with 0.14 M HCl at 40.8 °C for 60 min. Three monomeric proanthocyanidins and six novel flavan-3-ol-DTT conjugates consisting of three pairs of diastereomers were simultaneously obtained with a high yield (929 mg/g). All the degradation products showed protective effects against Aß25-35-induced neurotoxicity in PC-12 cells and prevented Aß25-35 aggregation based on the results from MTT and thioflavin T fluorescence assays, respectively. Detailed intermolecular interactions leading to the prevention of Aß25-35 aggregation were elucidated using molecular docking. This work would provide new compounds from functional foods that can be explored for their neuroprotective potential.

The hydrogels, because of their swelling properties in response to the environmental stimulus, are being widely considered for the design of controlled drug release systems. To meet the need for developing effective drug delivery methods, we developed special silica colloidal crystal (SCC)-embedded chitosan hydrogel films. The SCC films served as an interference substrate and drug storage layer, while the chitosan hydrogel served as a cover to regulate the drug release. The optical interferometry was performed to dynamically monitor the volume phase transition of chitosan hydrogel response to pH stimulation. Furthermore, the effects of crosslinking ratio and hydrogel thickness on the swelling properties of chitosan hydrogel were also evaluated. More importantly, the pH-responsive swelling of chitosan hydrogel was used to slowly release indomethacin. This system may provide support for drug delivery studies, therefore further expected to apply in the enhancement of the treatment efficiency of new drug therapies.

Leukocytes play an essential role in ontogeny, tissue regeneration, and innate and adaptive immunity. The migration of leukocytes to the infected or traumatized areas is necessary for their immune response function. As an adhesion molecule, CD99L2 is crucial in the extravasation of leukocytes, however, its role in the interstitial migration of leukocytes remains unclear. In this study, the cd99l2 gene was knock-out by TALEN (transcription activator-like effector nuclease) in zebrafish and discovered that the deletion had no effect on zebrafish development. The number of granulocytes and macrophages recruited to the wounded tissue was significantly reduced in the cd99l2 mutants following caudal fin damage. Further research revealed that the expression of mfap4 was drastically decreased in the cd99l2 mutants, which may be one of the reasons that affect the migration of macrophages to the wound site. Moreover,transgenic lines with labeled vasculature, neutrophils and macrophages demonstrated that neutrophils and macrophages migrate throughout the interstitial space to the wound tissue in both wild-type and mutant zebrafish at 60 hours post-fertilization, indicating that the cd99l2 gene is involved in the interstitial migration of leukocytes. Finally, RNA transcription, protein folding, and the P450 pathway were enriched in cd99l2 mutants by RNA-seq analysis. Previous research had demonstrated that the regulation of transcription and signal transduction could be affected by adhesion molecules, which may suggest that the cd99l2 gene is involved in the cascade signaling pathway of leukocyte migration as an adhesion molecule. In conclusion, this study uncovered a novel function of the cd99l2 gene in the process of leukocyte migration in zebrafish, which is expected to provide a theoretical foundation for inflammatory and immune-related diseases.

BACKGROUND: During the outbreak of COVID-19, online public opinion related to the epidemic was rapidly generated and developed rapidly. If some online public opinions cannot be effectively responded to and guided, it will bring risks to social order. The government should understand how to use information on social media to grasp public demands, provide useful information in a timely manner and take countermeasures. Studying the formation mechanism of online public opinion during the outbreak can help the government make scientific decisions and improve risk management capabilities. METHODS: The research selects the public opinion information of online platforms represented by WeChat, online communities, Sina Weibo and search engines, involving 75 relevant texts (1 January to 31 March 2022). According to the grounded theory method, using the QSR NVivo12 qualitative research software, the collected network texts were successively researched using open coding, axial coding and theoretical coding. RESULTS: The structure of online public opinion during the COVID-19 epidemic was obtained. The operation mechanism of the online public opinion system about COVID-19 was mainly affected by the interaction of online public opinion objects, online public opinion subjects, online public opinion intermediaries and government forces. It was based on social facts and citizens' appeals as the starting point, subject behaviors and prevention and control measures as the focus, government's governance as macro-control and citizens' evaluation as the guide. CONCLUSIONS: Scientific analysis of online public opinion is an important tool to identify and manage risks and improve the quality of government activities. Online public opinion has the function of assisting government decision-making, and the government can identify the important information reflected in it, especially the mainstream public opinion, as a reference for decision-making. By taking effective measures and properly responding to citizens' reasonable demands, the government can prevent social risks and avoid new negative public opinions. Contributions: According to the characteristics of the basic model of online public opinion, this study provides risk mitigation suggestions for Chinese public sectors to use online public opinion, optimize epidemic prevention policies and formulate strategic measures.

Sensitive determination of C-reactive protein (CRP) is of great significance because it is an early indicator of inflammation in cardiovascular disease and acute myocardial infarction. A disposable electrode with an integrated three-electrode system (working, reference, and counter electrodes) has great potential in the detection of biomarkers. In this work, an electrochemical immunosensing platform was fabricated on disposable and integrated screen-printed carbon electrode (SPCE) by introducing nanochannel arrays and gated electrochemical signals, which can achieve the sensitive detection of CRP in serum. To introduce active reactive groups for the fabrication of immuno-recognitive interface, vertically-ordered mesoporous silica-nanochannel film (VMSF) with rich amino groups (NH2-VMSF) was rapidly grown by electrochemical assisted self-assembly (EASA). The electrochemically reduced graphene oxide (ErGO) synthesized in situ during the growth of NH2-VMSF was used as a conductive adhesive glue to achieve stable bonding of the nanochannel array (NH2-VMSF/ErGO/SPCE). After the amino group on the outer surface of NH2-VMSF reacted with bifunctional glutaraldehyde (GA/NH2-VMSF/ErGO/SPCE), the converted aldehyde surface was applied for covalent immobilization of the recognitive antibody (Ab) followed with the blocking of the non-specific sites. The fabricated immunosensor, Ab/GA/NH2-VMSF/ErGO/SPCE, enables sensitive detection of CRP in the range from 10 pg/mL to 100 ng/mL with low limit of detection (LOD, 8 pg/mL, S/N = 3). The immunosensor possessed high selectivity and can realize reliable determination of CRP in human serum.

Two types of aquatic plants commonly used for the ecological restoration of rivers and lakes, Vallisneria natans (Lour.) Hara and Thalia dealbata Fraser ex Roscoe, were selected and grouped by plant parts (root, stem and foliage), and decomposing release experiments were conducted. The influence of the released substances on the water quality was analyzed, as well as the amount of nutrients released by each part of these two plants. The calculated maximum chemical oxygen demand releases from the foliage of V. natans and the foliage of T. dealbata were approximately 5.4 g/kg and 22.65 g/kg, respectively. Through three-dimensional fluorescence spectrum and parallel factor analyses, the different material compositions of the decomposing liquids from the plants were determined, and the main dissolved organic components of the decomposing liquid of V. natans were amino-acid-like and microbially derived humics, and those T. dealbata were soluble microbial by-product-like substances. The carbon-to-nitrogen ratio and humification index of each experimental group were compared. The experimental results showed that different parts of V. natans and T. dealbata had different rates of nutrient release. The dissolved organic matter in the decomposed solution can be utilized by microorganisms, which have the potential to become additional carbon sources. This study provides a new method for the treatment of aquatic plant litter. Different plant species can be used in combination according to their characteristics to ensure that better results are achieved during water treatment processes that use plant decomposing liquids as additional carbon sources.

Organophosphorus (OPPs) residues in dairy products are a potential threat to human health. To extract trace amounts of OPPs in dairy products, a graphitic carbon nitride (g-C3N4) was synthesized and combined with OPPs-based molecularly imprinted microspheres (MIM) to create a composite material (MIM/g-C3N4). Then, the MIM/g-C3N4 was used to prepare a solid phase extraction (SPE) cartridge to detect the OPPs in dairy products with UPLC method. The specific surface area of MIM/g-C3N4 was 172.208 m2/g, good thermal stability under 300℃, and could reuse up to 15 times. The four OPPs had good linear relationship within the range of 1-10000 ng/mL (r 2 > 0.999). The limits of detection were 0.7-2.6 ng/mL, and recoveries from blank dairy samples were 86.4 to 95.3 %. In this study, MIM combined with g-C3N4 was firstly utilized for the detection of OPPs in dairy products, which indicated it might be an ideal adsorbent for dairy products pretreatment.

Clonorchis sinensis (C. sinensis) infection is a risk factor for cholangiocarcinoma. Whether it also contributes to the development of hepatocellular carcinoma (HCC) is still unclear. This study explored the potential relationship between C. sinensis infection and HCC. A total of 110 Sprague-Dawley rats were divided into four treatment groups, the negative control group (NC) received intragastric (i.g.) administration of saline, while the clonorchiasis group (CS) received i.g. administration of 150 C. sinensis metacercariae. The diethylnitrosamine-induced group (DEN) received intraperitoneal (i.p.) administration of DEN. The clonorchiasis DEN-induced group (CSDEN) received i.g. administration of 150 C. sinensis metacercariae followed by i.p. administration of DEN. Hematoxylin and eosin staining, immunohistochemistry, and Masson's trichrome staining were performed for histopathological analysis of the isolated tissues. RNA-seq technology and RT-PCR were employed for gene expression. In the DEN group, 15 rats survived, of which 9 developed liver cirrhosis and 7 developed HCC. In the CSDEN group, all of the 17 surviving rats developed cirrhosis, and 15 showed development of HCC. The incidence of liver cirrhosis and HCC was significantly higher in the CSDEN group than in the DEN group. KEGG pathway analysis of the differentially expressed genes suggested significant upregulation in inflammation-associated pathways. Immunohistochemistry and RT-PCR results showed significant upregulation of hepatic progenitor cell markers (CK19, SOX9, EpCAM) in the CS group compared to the NC group, as well as in the CSDEN group compared to the DEN group. Our study suggests that C. sinensis infection increases risk of HCC in a rat model by stimulating proliferation of hepatic progenitor cells.

Mesenchymal stem cell-derived exosomes (MSC-Exos) are emerging as a promising platform for treating various intractable diseases and organ injuries. Monitoring their migration, homing, and therapeutic capability in vivo is essential to develop exosome-based theranostics. Here, we designed fluorescent semiconductor polymer dots (Pdots) in the second near-infrared window (NIR-II) for bright labeling and tracking of MSC-Exos. Glucose-coated Pdots (Pdots-Glu) were able to label MSC-Exos without changing their biological properties. The NIR-II fluorescent Pdots allow for high labeling brightness and long-term in vivo tracking of MSC-Exos. We investigated the biodistributions and therapeutic functions of these labeled MSC-Exos in liver-resected mice. In vivo and ex vivo imaging demonstrated that the Pdot-labeled MSC-Exos injected via the tail vein mainly accumulated in the residual liver tissue. In terms of the therapeutic effect, MSC-Exos may accelerate postoperative liver function recovery by inhibiting inflammatory responses, promoting cell proliferation, and resisting apoptosis. Our results indicated that MSC-Exos therapeutic systems hold promising applications in liver regenerative medicine.
