RESUMO

Doxorubicin is associated with cardiotoxicity, and physical exercise seeks to minimize the toxic effects of doxorubicin through physiological cardiac remodeling, as well as the reduction of oxidative stress, evidenced by previous studies. This study aimed to analyze whether running training before treatment with doxorubicin influences tolerance to physical exertion and cardiotoxicity. Thirty-nine male Wistar rats, aged 90 days and weighing between 250 and 300 g, were divided into 4 groups: Control (C), Doxorubicin (D), Trained (T), and Trained + Doxorubicin (TD). Animals in groups T and DT were submitted to treadmill running for 3 weeks, 5 times a week at 18 m/min for 20-30 min before treatment with doxorubicin. Animals in groups D and DT received intraperitoneal injections of doxorubicin hydrochloride three times a week for two weeks, reaching a total cumulative dose of 7.50 mg/kg. Our results show an increase in total collagen fibers in the D group (p = 0.01), but no increase in the TD group, in addition to the attenuation of the number of cardiac mast cells in the animals in the TD group (p = 0.05). The animals in the TD group showed maintenance of tolerance to exertion compared to group D. Therefore, running training attenuated the cardiac damage caused by the treatment with doxorubicin, in addition to maintaining the tolerance to exertion in the rats.

RESUMO

Several studies have been developed using the Gallus gallus embryo as an experimental model to study the toxicity of drugs and infections. Studies that seek to standardize the evaluated parameters are needed to better understand and identify the viability of CEs as an experimental model. Therefore, we sought to verify whether macroscopic, histopathological, blood count, metabolites and/or enzymes changes and oxidative stress in CE of different ages are specific to the model. To achieve this goal, in ovo assays were performed by injecting a virus (Gammacoronavirus) and two drugs (filgrastim and dexamethasone) that cause known changes in adult animals. Although congestion and inflammatory infiltrate were visible in the case of viral infections, the white blood cell count and inflammation biomarkers did not change. Filgrastim (FG) testing did not increase granulocytes as we expected. On the other hand, CE weight and red blood cell count were lower with dexamethasone (DX), whereas white blood cell count and biomarkers varied depended on the stage of CE development. Our work reinforces the importance of standardization and correct use of the model so that the results of infection, toxicity and pharmacokinetics are reproducible.

RESUMO

This study aimed to evaluate the influence of age and sex on the blood biochemical constituents of broiler breeders during the egg production stage. The analysis was performed in an industrial broiler breeder farm, and blood samples were collected from males and females at five different ages. Biochemical elements analysed in each serum sample were total proteins, albumin, globulins, uric acid, total cholesterol and triglycerides, alanine aminotransferase (ALT) and aspartate aminotransferase (AST), gamma-glutamyltransferase (GGT), creatine kinase (CK), alkaline phosphatase (PAL), calcium and phosphorus, beside the glycaemic status. At most ages, females had higher values of total proteins, albumin, globulins, triglycerides, cholesterol, glucose, calcium, phosphorus, Ca/P (calcium and phosphorus) ratio and gamma-glutamyltransferase (GGT). The values of uric acid, aspartate aminotransferase (AST), creatine kinase (CK) and alkaline phosphatase (PAL) were higher in males. The lowest protein values were found at 28 and 60 weeks old. The mean albumin values were significantly higher at 44 and 52 weeks old in males and females. A trend of increasing globulin values with increasing age up to 52 weeks old was observed. Although calcium and phosphorus did not vary according to age, the Ca/P ratio was lowest at 36 weeks old. Comparing the means of both sexes, the AST and GGT values were significantly higher in 60-week-old birds. The highest serum levels of CK occurred at 28 and 52 weeks old. The physiological levels of serum biomarkers presented in this work are important for evaluating productivity performance, welfare and disease indication in breeding flocks.

RESUMO

The objective of this work was to evaluate the proteins, minerals, metabolites, and serum enzymes profile in Quarter Mile equines subjected to training for Team Penning competitions by comparing the results before and after training. The animals were from the Agricultural Exhibition Park of Uberlândia (CAMARU), in Uberlândia in the State of Minas Gerais, Brazil. These animals perform constant periodic training with an average duration of two and a half hours per training. Blood samples from twenty male and female equines were collected at the beginning and end of a 45-day interval and then analyzed. A 5 mL blood sample from each animal was collected before and after training by a jugular external venipuncture, and the samples were placed in a tubewithout anticoagulant and containing separator gelto obtain the serum. The components analyzed were: total proteins, albumin, globulins, albumin to globulin ratio (A:G), total and ionized calcium, phosphorus, calcium to phosphorus ratio (Ca:P), cholesterol, triglycerides, creatinine, urea, alkaline phosphatase, ɤ-glutamyl transferase, lactate dehydrogenase (LDH), aspartate amino transferase (AST), alanine aminotransferase (ALT) and creatine kinase (CK). Statistical differences were observed with higher values after exercise for total calcium, ionized calcium, Ca: P ratio, creatinine and ALT and decreased concentrations were found for globulins, phosphorus, cholesterol and triglycerides. Despite the changes observed in the serum concentrations of some elements after exercises, it was concluded that the animals were in adequate physical condition to perform the proposed physical exercises.

Ophidic accidents are among the problems of public health in Brazil. The components from bothropic venom are responsible for many systemic clinical complications resulting from envenomation. The present work aimed to analyse the systemic changes induced in mice after intraperitoneal administration of BmooTX-I, a myotoxic acidic phospholipase A2 isolated from Bothrops moojeni venom. Urinalysis was performed and the following plasma biochemical markers were documented: urea, creatinine and uric acid (renal function); glucose and amylase (pancreatic function); alanine aminotransferase, alkaline phosphatase and gamma-GT (intra- and extrahepatic function); creatine kinase and enzymatic lactate (muscle function). Our results showed that after the intraperitoneal injection of BmooTX-I the urine of these animals showed glycosuria, proteinuria, haematuria, bacteriuria, bilirubinuria, polyuria and nitrite. The plasma biochemical analysis showed alterations in levels of urea, creatinine and uric acid. Amylase concentration was not altered significantly, but the plasma glucose increased significantly compared to controls. The plasma levels of alanine aminotransferase and alkaline phosphatase decreased and increased, respectively, in these same animals. On the other hand, the plasma γGT concentration did not undergo significant modification compared to the control group. The plasma concentration of CK increased, while the enzymatic lactate concentration decreased after the injection of the BmooTX-I. Therefore, in mice BmooTX-I is capable of causing systemic alterations which manifest as renal, muscular, hepatic and pancreatic impairment.

RESUMO

The objectives of this study were to evaluate the serum biochemical profile in Quarter Horse mares in their peripartum, and neonates on their birth date, and to compare the values obtained for male and female foals, as well as values obtained for foals and mares on the day of delivery. Forty one adult, pregnant mares and their respective offspring on the day of delivery were analyzed, totaling 82 animals. Two samples of blood were collected from the mares, seven days before and on the day of delivery. From the foals, blood collection was performed on the day of their birth about 12 h after colostrum ingestion. The samples were centrifuged and the serum stored at -20 ºC for analyses. Each sample was used to determine the serum concentrations proteins, metabolites, minerals and enzymes. Physiological changes caused by peripartum led to higher serum total protein, albumin, and globulins on the day of delivery. Serum concentration of triglycerides and calcium /phosphorus ratio were greater seven days before delivery; iron concentration was higher than reference ranges in both moments of evaluation. In foals, albumin serum concentrations, albumin/globulins ratio, and aspartate aminotransferase remained below reference ranges. In foals, there was no influence of sex in the serum biochemical profile. When results obtained for foals on their birth date were compared with those obtained for the mares on the delivery day, cholesterol, triglycerides, phosphorus, iron, alkaline phosphatase, and gamma glutamyltransferase were higher in foals, and total protein, albumin, globulins, urea nitrogen, calcium/phosphorus ratio, and aspartate aminotransferase were higher in mares. Results showed variations in serum biochemistry in peripartum mares and neonates, as well as in the comparison between foals and mares on the day of delivery. Clinically, serum biochemistry values for peripartum mares and neonates should be considered in the analysis of laboratory results of tests carried out in this period.

Platinosomosis affects felines and it is caused by a trematode of the genus Platynosomum sp. The parasitized animals can be symptomatic or asymptomatic and the disease can evolve severely, killing the animal. Due to the similarity between clinical signs and changes of platinosomosis and those of other pathologies caused by the parasite and having in mind that the parasite occasionally appears on the stool, the search for trematodes and eggs in material coming from bile aspirate is a more efficient way to diagnose the parasitic disease, what helps to distinguish it from other liver diseases. Thus, the clinical veterinarian is able to perform the treatment correctly. The aim was to report a case of cystic liver disease related to platinosomosis in a domestic feline which has been undergoing treatment of liver disease for over a year. During the clinical evaluation, the animal was apathetic and with hepatomegaly. A complete blood count (CBC) revealed a discreet lymphopenia. The enzymes alkaline phosphatase and alanine transaminase were above the normal range for the species. The parasitological examination of the biliary material indicated eggs from the trematode Platynosomum sp. whereas the histopathology examination of liver masses indicated cystic structures covered by connective tissue and multifocal mononuclear inflammatory infiltrate. In conclusion, liver cysts in felines with chronic liver disease can be associated with cases of diseases caused by the trematode helminths.

ABSTRACT Objective This study investigated the possible blood changes in wistar rats elderly with and without treatment with anabolic steroids submitted physical training. Materials and methods Elderly rats (32) were divided into four groups: normal (N), treated normal (NT), diabetic (D) and treated diabetic (DT). They were submitted to 20 sessions of swimming with overload (5% body weight), 40 min/day for four weeks. The NT and DT groups received application of testosterone twice a week. At the end of the sessions, the animals were subjected to swimming until exhaustion and then killed for removal of blood and visceral fat. We evaluated maximum swim time, weight of visceral fat, erythrogram, leukogram, lipidogram and serum levels of glucose, lactate, aspartate aminotransferase and creatine kinase. The results were compared using one-way ANOVA followed by the post hoc Tukey test. Results In elderly diabetic rats, the use of anabolic associated with physical training in older rats resulted in improvement in erythrogram, lipidogram and physical performance for high-intensity aerobic exercise. However, it was related to changes in leukocyte count, probably associated with inflammation. Conclusion The combination of the use of testosterone with physical training, followed by maximal effort test caused changes hematological and biochemical can be associated with improvement in physiological characteristics, with increase of the swimming time and decrease of visceral fat levels, improvement in aerobic metabolism of fatty acids and glucose in normal and diabetic animals.

Tendon lesions may involve the partial or total section of the common calcaneal tendon and cause postural changes of the member. This study evaluated, after 45 and 90 postoperative days (PO), the repair of the tendon of gastrocnemius muscle of rabbits with topical application of autologous platelet concentrate. Twelve adult rabbits were divided into two groups (n = 6) undergoing cardiac puncture and collection of 10 ml of blood to obtain platelet rich-plasma (PRP). Animals of both groups had a transverse tenotomy in the middle third of the lateral belly of the gastrocnemius tendon and muscle that was approximated with modified Kessler suture and nylon thread. In the animals of the treated group it was applied the average of 490.644 platelets / uL of PRP, per animal over the tendon synthesis. The treated group showed a higher amount of collagen fibers than the control one, and at 90 PO days the intensity of collagen was higher than at 45 days with more fibroblasts in the control than in treated one. The administration of plasma autogenous platelet concentrate in the repair of the gastrocnemius tendon of rabbits stimulates and organizes the repair process and causes early production of collagen fibers.

RESUMO

Potamotrygonidae is a family of freshwater stingray endemic in South America. We determined the hematological and serum biochemistry parameters of Potamotrygon motoro and Potamotrygon falkneri from Parana Basin and Potamotrygon orbignyi and Potamotrygon scobina from the Amazon Basin. Blood was collected from 55 specimens of Potamotrygon sp., and these parameters were evaluated: red blood cell count, hematocrit percent, hemoglobin concentration, leucocytes count, trombocytes count, total protein concentration, albumin, uric acid, urea, creatinine, triglycerides, cholesterol, high-density lipoprotein HDL, calcium, phosphorus, calcium/phosphorus relationship, creatine kinase CK, gamma-glutamyl transpeptidase GGT, Aspartate transaminase AST, alanine transaminase ALT, globulin, albumin/globulin relationship, alkaline phosphatase AP, sodium, chloride, potassium, triiodothyronine T3, and thyroxine T4 hormones. The hematological indices were similar for all species, except for levels of heterophils in P. orbignyi and P. scobina that were higher. Calcium, phosphorus, calcium/phosphorus relationship, sodium, potassium, chloride, AST, ALT, GGT, CK, albumin/globulin relationship, uric acid, creatinine, urea, triglycerides, HDL, T3, T4 levels had no significant difference between the species. These results suggest that there is low influence of habitat and feeding habits. AP, total protein, albumin, globulin, glucose, and total cholesterol had significant differences between the studied species. These results support the theory that stingrays migrated from the Atlantic Ocean, adapting to different conditions over time and placed themselves geographically distant from each other.

We evaluated the polypeptide profiles, inhibition of human salivary α-amylase activity, and hemagglutination properties of a commercial phaseolamin sample. We also performed an in vivo assay to investigate the effects of a commercial phaseolamin treatment (100, 500, or 1500 mg/kg) over 20 days on the glycemia, body weight, and serum biochemical parameters (total cholesterol, triglycerides, alanine aminotransferase, and aspartate aminotransferase) of nondiabetic and streptozotocin-induced diabetic rats. The in vitro evaluation showed defined protein profiles, low hemagglutination activity, and high α-amylase inhibition. None of the experimental groups treated with phaseolamin or acarbose showed decreases in body weight. Our data demonstrate that phaseolamin inhibits amylase activity in vitro, reduces blood glucose levels, decreases or attenuates some of the renal and hepatic effects of diabetes in streptozotocin-induced rats, and could therefore have therapeutic potential in the treatment or prevention of the complications of diabetes.

Leucine is a regulator of protein metabolism in vivo, and there is little information regarding its action on cardiac hypertrophy induced by experimental hyperthyroidism and its relationship to serum creatine kinase. The study aimed to verify the effect of leucine in cardiac hypertrophy and serum creatine kinase in rats with hyperthyroidism. We used 20 animals were divided into four groups and these control, hormone, hormone + leucine and leucine. Hyperthyroidism was induced by administration of daily 20µg/100 grams of levothyroxine sodium in aqueous suspension by gavage. Leucine was supplemented by adding 5% of the amino conventional diet. Blood was collected by cardiac puncture and analyzes made in kit CK-NAC, CK-MB and TSH. At the end of the trial period of seven days the hearts were removed and weighed. Subsequently, the left ventricle was separated together with the interventricular septum and weighed. In statistical comparison was used analysis of variance (ANOVA) and two-way post-Tukey test, considering p values <0.05. Serum concentrations of serum CK-MB were lower in leucine (p <0.05) and hormone + leucine (p <0.05) compared to control. The hormone group, showed greater relative heart weight than the control group (p <0.05) and absolute heart weight greater than hormone + leucine (p <0.05). In conclusion, leucine appears to reduce cardiac hypertrophy and serum concentration of creatine kinase by unknown mechanisms.

The objective of this study was to compare serum concentrations of the hormones triiodothyronine (T3) and thyroxine (T4) in pigs on commercial line beginning and end of growing-finishing phase, correlating them with productive performance and quality characteristics of carcass and meat. Blood samples were collected from 48 animals at the beginning and end of growing-finishing phase by puncturing the jugular vein, around the same time (08:00 to 10:00a.m.). Measurements of serum thyroid hormones were made by enzyme-immunoassay. It was evaluated the performance and quality of carcass and meat. At the beginning and end of the housing, serum T3 concentrations were 1.85 and 1.32nmol L-1, respectively, and T4 of 100.33 and 86.53nmol L-1, respectively. There was negative correlation between T3 and low-end and average daily weight gain, final weight and backfat thickness. Inicial T4 was moderately and moderately negatively correlated with initial weight. The values of T3 and T4 in pigs early in the growing-finishing phase are larger than in termination. The thyroid hormones are associated with the initial and final weight, backfat thickness at the last lumbar, pH45min and pH24h of growing-finishing pigs.

Aiming to establish the hepatic and proteic biochemical profile in dogs with visceral leishmaniasis in Uberlândia - MG, blood of 40 seropositive dogs in indirect immunofluorescence tests and ELISA was collected by jugular venipuncture tube without anticoagulant to obtain serum and measurement of albumin, alkaline phosphatase (ALP), total protein (TP), globulins, alanine aminotransferase (ALT) and gamma-glutamyltransferase (GGT) in an automatic analyzer (ChemWell ®). For data analysis, 40 animals were divided into three groups classified as asymptomatic (4 animals), oligosymptomatic (19) and symptomatic (17 animals). Hypoalbuminemia and hypergammaglobulinemia were noted in all groups. However, significant differences in serum albumin level (1.48 ± 0.49 g/dL) occurred in the symptomatic group compared to other groups. The mean values of FA, PT, ALT and GGT remained within the reference standards. Thus, visceral leishmaniasis can lead to hypoalbuminemia and hyperglobulinemia independent of symptomatology presented by dogs or not, and the other parameters, the hepatic protein may remain within the reference standards.

RESUMO

Ehrlichiosis and leptospirosis are frequent diseases in tropical countries, although they are not always considered as differential diagnosis. The purpose of this work was to evaluate clinical signs and hematological alterations in dogs reactive to leptospirosis, among them those that presented morulae of Ehrlichia spp. in peripheral blood, and to recognize the frequency of Leptospira serovars by means of the microscopic agglutination test. Blood samples were used from 50 dogs presenting morulae of Ehrlichia spp. in blood extensions collected from the ear marginal capillaries. The clinical signs frequently observed were apathy (100.00%), anorexia/dysorexia (100.00%), adypsia (83.33%), ocular secretion (66.66%), reactive lymph nodes (66.66%), diarrhea (58.33%), vomiting, anemia and fever (41.66%). Albeit unspecific, anemia and thrombocytopenia are frequent hematological findings in canine ehrlichiosis and are present in both diseases. Of the 50 dogs with ehrlichiosis, 12 (24.00%) were reactive to leptospirosis, and the most frequent serovars were Pomona (33.33%), followed by Canicola (27.77%) and Icterohaemorrhagiae (22.22%). The results suggest the need to establish a differential diagnosis between these diseases that may occur concomitantly.

Canine hepatozoonosis is a tick-borne disease caused by protozoans of the genus Hepatozoon. Several tick species have been implicated as potential vectors. Therefore, extensive studies are needed to determine the 'natural' endemic cycle of this parasite. This paper presents the first report of the presence of Hepatozoon canis oocysts in Rhipicephalus (Boophilus) microplus collected from an infected dog.

The canine distemper is a systemic infectious disease with high mortality rate in dogs caused by a Morbillivirus. Affected animals often develop nervous manifestations. The main of transmission is by aerosolization of respiratory exudate containing the microorganism, but transplacental infection has been documented in domestic dogs. The presence of intracytoplasmic Lentz bodies in leukocytes is found in the viremia phase of the disease. This inclusion is considered a tool for early diagnosis of distemper. It was routed at the Veterinary Hospital, Federal University of Uberlândia a 10-day-old puppy, male, mixed breed and weighed 500 grams. It presented seizures of 20 to 20 minutes. At the hemogram, it was found intracytoplasmic Lentz body. The animal with only 10-day-old, reports a high viremia by the presence of intracytoplasmic corpuscle. Therefore, occurred a transplacental infection due to the incubation period of canine distemper virus is seven days, and the animal presents a young age.

Variations in hematologic parameters are used to assess the degree of training or clinical state of the animal. The hematologic evaluation of horses at rest has been an object of study in order to establish a correlation with training or athletic capacity. The purpose of this study was to evaluate the hematologic profile of horses subjected to Team Penning competitions, correlating sex and frequency of physical activity. Two milliliters of blood were drawn through a puncture made in the external jugular vein from 29 horses, 18 males and 11 females, at rest (Moment I) and after exercising (Moment II). The blood samples were processed in an ABC VET automated veterinary hematology analyzer (Horiba ABX Diagnostics). The animals were divided into Group A, B, C and D according to the number of times they participated in the competition. The values of globular volume, hemoglobin, erythrocytes, rod-shaped segmented neutrophils and monocytes increased after the physical exercise, unlike the number of lymphocytes and eosinophils, which decreased. A comparison of the before/after exercise correlations showed no significant differences (p<0.05) between males and females. In addition, it was found that the value of the MI/MII ratio for the globular volume, hemoglobin and number of erythrocytes varied according to the frequency of the exercise. It was concluded that the Team Penning competition produces hematologic alterations in horses, which are affected by the frequency of exercising, regardless of sex.

RESUMO

The occurrence of Hepatozoon sp. infection in dogs was evaluated in the urban area of Uberlândia, Minas Gerais, Brazil. The study involved 300 animals, 120 from the Veterinary Hospital of the Federal University of Uberlândia's Faculty of Veterinary Medicine, 80 from private clinics, and 100 from the Animal Protective Association (APA). Among these animals, 7.66% presented Hepatozoon sp. gamonts inside neutrophils. No statistically significant difference was found among the diagnoses based on capillary and venous blood smears, but a statistical significance was identified in age range and breed. The main signs and symptoms the animals presented were pale mucous membranes, swollen lymph nodes, ocular discharge and pain in response to renal palpation. All the ticks collected were of the species Rhipicephalus sanguineus and no oocyst of the parasite was found in the hemolymph of the ixodidae. The findings of this study reinforce the presence of Hepatozoon sp. infecting dogs in Uberlândia, MG, with age and breed possibly related to the infection. The symptoms presented by the animals were consistent with those reported by other researchers.
