RESUMO

INTRODUCTION: Several natural products exhibit promising antineoplastic activity against bladder cancer cells, including allyl isothiocyanate (AITC). However, the AITC irritates the mucous membranes and induces eczematous or vesicular skin reactions. Thus, pharmaceutical formulations are necessary to overcome these problems. The aim was to develop micellar solutions containing AITC and investigate their antitumoral activity in bladder carcinoma cell lines. METHOD: The micellar solutions were prepared by cold dispersion method. Subsequently, we evaluated cytotoxicity, cell proliferation, cell cycle kinetics and long-term effects of micelles in bladder cancer cells. RESULTS: Cytotoxicity and cell proliferation assays showed there was an increase in AITC activity when it was encapsulated in micelles. We also observed cell cycle arrest in the S phase after treatment with AITC-micelles. Furthermore, the formulation was able to maintain the long-term effects of free AITC. CONCLUSIONS: The micellar solutions developed can become an interesting approach for administration of AITC in the treatment of bladder cancer

Dietary sugar is an important determinant of the development and progression of nonalcoholic fatty liver disease (NAFLD). However, the molecular mechanisms underlying the deleterious effects of sugar intake on NAFLD under energy-balanced conditions are still poorly understood. Here, we provide a comprehensive analysis of the liver lipidome and mechanistic insights into the pathogenesis of NAFLD induced by the chronic consumption of high-sugar diet (HSD). Newly weaned male Wistar rats were fed either a standard chow diet or an isocaloric HSD for 18 weeks. Livers were harvested for histological, oxidative stress, gene expression, and lipidomic analyses. Intake of HSD increased oxidative stress and induced severe liver injury, microvesicular steatosis, and ballooning degeneration of hepatocytes. Using untargeted lipidomics, we identified and quantified 362 lipid species in the liver. Rats fed with HSD displayed increased hepatic levels of triacylglycerol enriched in saturated and monounsaturated fatty acids, lipids related to mitochondrial function/structure (phosphatidylglycerol, cardiolipin, and ubiquinone), and acylcarnitine (an intermediate lipid of fatty acid beta-oxidation). HSD-fed animals also presented increased levels of some species of membrane lipids and a decreased content of phospholipids containing omega-6 fatty acids. These changes in the lipidome were associated with the downregulation of genes involved in fatty acid oxidation in the liver. In conclusion, our data suggest that the chronic intake of a HSD, even under isocaloric conditions, induces lipid overload, and inefficient/impaired fatty acid oxidation in the liver. Such events lead to marked disturbance in hepatic lipid metabolism and the development of NAFLD.

RESUMO

OBJECTIVES: to learn and analyze the structure of nurses' social representations about transvestite people. METHODS: a qualitative research based on the Theory of Social Representations, with 110 nurses enrolled in Graduate Nursing courses, who answered the Free-Association Test, with the stimulus 'transvestite'. Data were processed by the software Ensemble de Programmes Permettant I' Analysedes Évocations. RESULTS: in the central nucleus, the term "prejudice" was the most evoked, followed by "homosexual", "identity" and "female-make-up". Social representation is anchored in the social organization in which transvestite people are still seen and/or associated with homosexuals who make up and assume an identity, without being seen and/or understood as they really are. FINAL CONSIDERATIONS: although prejudice is noteworthy as a central element, terms present in the peripheral system reveal that the group recognizes transvestites as a person with rights, which can translate into health care practices.

RESUMO

This study aims to evaluate the effect of long-term high-sugar diet (HSD) intake and regular physical activity on gut microbiota as well as its health impact. Weaned male Wistar rats were fed with standard chow diet (SSD) or HSD ad libitum and subjected or not to regular swimming training with a workload (2% of body weight) for 15 weeks. Feces samples were used on microbiome analysis using 16S rRNA amplicon sequencing. HSD increased body mass, adipose cushions, and the serum levels of triglycerides and VLDL, also changed the bacteria taxons associated with metabolic disorders (increase taxons belonging to Proteobacteria phylum and decrease Pediococcus genus); the swim training reverted these changes. SSD intake increased the abundance of bacteria associated with metabolization of dietary fiber. Training in association with SSD consumption beneficially modulated the microbiota, increasing the Bacteroidetes, Bacteroidaceae, Porphyromonadaceae, Parabacteroides, and Lactobacillaceae, and decreasing the Firmicute/Bacteroidetes ratio; training was not able to maintain this profile in animals SHD-fed. Physical training modulates the gut microbiota reversing the obesogenic response caused by SHD. However, training itself is not efficient for up-regulating the probiotic bacteria in comparison to its association with a balanced diet.

RESUMO

Scientific advances in recent decades have revealed an incredible degree of plasticity in gene expression in response to various environmental, nutritional, physiological, pathological, and behavioral conditions. Epigenetics emerges in this sense, as the link between the internal (genetic) and external (environmental) factors underlying the expression of the phenotype. Methylation of DNA and histone post-translationa modifications are canonical epigenetic events. Additionally, noncoding RNAs molecules (microRNAs and lncRNAs) have also been proposed as another layer of epigenetic regulation. Together, these events are responsible for regulating gene expression throughout life, controlling cellular fate in both normal and pathological development. Despite being a relatively recent science, epigenetics has been arousing the interest of researchers from different segments of the life sciences and the general public. This review highlights the recent advances in the characterization of the epigenetic events and points promising use of these brands for the diagnosis, prognosis, and therapy of diseases. We also present several classes of epigenetic modifying compounds with therapeutic applications (so-call epidrugs) and their current status in clinical trials and approved by the FDA. In summary, hopefully, we provide the reader with theoretical bases for a better understanding of the epigenetic mechanisms and of the promising application of these marks and events in the medical clinic.

RESUMO

This article investigates sediments collected from the banks of the Subaé River located in Todos os Santos Bay in the state of Bahia, Brazil, in 2018, twenty-five years after the closing of a former lead alloy processing plant. Ten sediment samples were collected at different points of the course of the river and its estuarine region. Chemometric tools were used to determine geochemical correlations between the organic matter content and concentration of sulfides and potentially toxic metals. The inorganic geochemical variables (enrichment factor [EF]) used in this evaluation were concentrations of the Pb, Cd, Cu, Zn, and Ni. Chemical element analyses were performed using ICP-OES. To assess the interaction between metals and sulfide or metals and organic matter, concentrations of Pb, Cd, Cu, Zn, Ni, sulfide, and the silt-clay fraction constituted the organic geochemical parameters selected to characterize the amount of organic matter present in Subaé River sediment samples, determining the carbon content (%TOC) to compose the matrix of the principal component analysis (PCA) and hierarchical cluster analysis. PCA showed that 88.3% of the samples were representative for assessing correlations between geochemical variables. A tendency toward binding was found among Cu, Cd, Ni, and sulfide, as well as the silt-clay fraction. The concentrations (mg kg-1) of lead, zinc, and copper were higher in both collection campaigns, ranging from 4.72 to 31.34, 12.76 to 54.24, and 5.34 to 31.37, respectively. Pb and Zn were presented in elemental form when assessed as a function of the pH and Eh of the environment. Except for Cd (EF: 0.51 to 5.49), the other elements exhibited little or no potential pollution in the aquatic environment of the Subaé River.

RESUMO

Objective Provide a comprehensive view of the events surrounding the sugar consumption, under conditions of energy equivalence; through the analysis of behavioral aspects of intake, and of biochemical, metabolic and physiological parameters, as well as the effect of this nutrient on the plasticity of adipose tissue. Materials and methods Newly weaned male Wistar rats were classified in two groups and subjected to the following normocaloric diets: standard chow diet or to high-sugar diet (HSD) ad libitum for 18 weeks. Results The animals submitted to the HSD were associated with a lower caloric intake during the 18 weeks of experimentation. However, the HSD induced a significant increase in body weight, white adipose tissue weight, adiposity index, Lee index, and the levels of triglycerides and very low-density lipoprotein in the serum. In addition, it induced glucose intolerance, insulin resistance and compensatory increase of insulin secretion by pancreatic ß-cells. Also increased heart rate and induced hyperplasia, and hypertrophy of retroperitoneal visceral adipose tissue. In the liver, the HSD was associated with increased hepatic lipid content (i.e., triglycerides and cholesterol) and hepatomegaly. Conclusion The post-weaning consumption of HSD induces an adaptive response in metabolism; however, such an event is not enough to reverse the homeostatic imbalance triggered by the chronic consumption of this macronutrient, leading to the development of metabolic syndrome, irrespective of caloric intake. These findings corroborate recent evidence indicating that sugar is a direct contributor to metabolic diseases independent of a positive energy balance. Arch Endocrinol Metab. 2020;64(1):71-81.

RESUMO

Scheelea phalerata Mart. ex Spreng (Arecaceae) is a palm tree found in the Brazilian cerrado. There are no topics related to volatile oils from S. phalerata leaves in the literature. This work determines its chemical composition and evaluates the biological activity under two different seasonal conditions (dry and rainy seasons). The dry essential oil yield was 0.034 ± 0.001% and the rainy essential oil yield was 0.011 ± 0.003%. Both essential oils presented different qualitative and quantitative compositions (99.4 and 98.5%). The main constituents of the dry essential oil were phytol (36.7%), nonadecane (9.7%), linolenic acid (9.1%), (Z)-hex-3-en-1-ol (4.2%), and squalene (4.0%). The main constituents of the rainy essential oil were phytol (26.1%), palmitic acid (18.7%), hexan-1-ol (15.6%), (Z)-hex-3-en-1-ol (9.7%), and oleic acid (4.0%). The antileishmanial activity against promastigotes of Leishmania amazonensis was observed only for the rainy season essential oil (IC50 value of 165.05 ± 33.26 µg mL-1). A molecular docking study showed that alcohols exert a paramount efficacy and that the action of some essential oil compounds may be similar to that of amphotericin B. Still, only the essential oil from the dry season showed moderate antibacterial activity against S. sanguinis (MICs 200-400 µg mL-1). The cytotoxicity against Vero cells was identical (>512) for both essential oils. The novel data here for both chemical characterization and biological activity, in particular, evidence that the action of these compounds is similar to that of amphotericin B, provide valuable information to the drug-discovery field.

RESUMO

BACKGROUND AND AIMS: Modulation of the gut microbiota emerges as a therapeutic possibility to improve health. Our objective was to compare the impact of three months of intervention with diet plus nutritional orientation versus only nutritional orientation on the gut microbiota and metabolic-nutritional profile of outpatients with non-alcoholic steatohepatitis. METHODS: It was a randomized clinical trial with 40 outpatients (49.48 ± 10.3 years), allocated in two groups: DIET group (n=20), who received diet (1.651.34 ± 263.25 kcal; 47% carbohydrates, 28% lipids, 25% proteins, 30 g fibers) and nutritional orientation, and control group (n = 20), which received only nutritional orientation. RESULTS: The DIET group, in relation to baseline, presented a reduction in body weight (p<0.001), BMI (p<0.001), waist circumference (p=0.001), percentage of fat (p=0.002), serum aspartate aminotransferase (p<0.001), alanine aminotransferase (p<0.001), γ-glutamyltransferase (p=0.001), glycemia (p=0.003), homeostasis model assessment of insulin resistance (p=0.017), total cholesterol (p=0.014), and triacylglycerols (p=0.008), whereas the control group did not present changes. After intervention, the small intestinal bacterial overgrowth frequency was 30% in the DIET group and 45% in the control group (p=0.327). In the DIET group, an increase in the density of total microorganisms (3.76 ± 7.17 x 10 8 cells g -1 ; p=0.048) was detected, while in the control group reduced Bacteroidetes (-0.77 ± 2.01 x 10 8 cells g -1 , p=0.044) and Verrucomicrobiales (-0.46 ± 0.75 x 10 8 cells g -1 ; p=0.022) were observed. CONCLUSIONS: The results suggest that exclusively dietary modifications contribute to health promotion in non-alcoholic steatohepatitis and should be the basis of nutritional treatment for this condition.

RESUMO

Cutaneous secretions produced by amphibians of the family Bufonidae are rich sources of bioactive compounds that can be useful as new chemical templates for agrochemicals. In crop protection, the use of elicitors to induce responses offers the prospect of durable, broad-spectrum disease control using the plant's own resistance. Therefore, we evaluated the potential of methanolic extracts of cutaneous secretions of two species of amphibians of the family Bufonidae found in the Amazon biome-Rhaebo guttatus (species 1) and Rhinella marina (species 2)-in the synthesis of phytoalexins in soybean cotyledons, bean hypocotyls, and sorghum mesocotyls. Additionally, changes in the enzyme activity of ß-1,3-glucanase, peroxidase (POX), and polyphenol oxidase (PPO) and in the total protein content of soybean cotyledons were determined. In the soybean cultivar 'TMG 132 RR', our results indicated that the methanolic extract of R. guttatus cutaneous secretions suppressed glyceollin synthesis and ß-1,3-glucanase activity and increased POX and PPO activities at higher concentrations and total protein content at a concentration of 0.2 mg/mL. On the other hand, the methanolic extract of R. marina cutaneous secretions induced glyceollin synthesis in the soybean cultivars 'TMG 132 RR' and 'Monsoy 8372 IPRO' at 0.1-0.2 mg/mL and 0.2 mg/mL, respectively. The methanolic extract of R. marina cutaneous secretions also increased the specific activity of POX and PPO in 'Monsoy 8372 IPRO' and 'TMG 132 RR', respectively, and decreased the activity of ß-1,3-glucanases in 'Monsoy 8372 IPRO'. At 0.3 mg/mL, it stimulated phaseolin synthesis. The extracts did not express bioactivity in the synthesis of deoxyanthocyanidins in sorghum mesocotyls. The study in soybean suggests that the bioactivity in defense responses is influenced by cultivar genotypes. Therefore, these results provide evidence that extracts of cutaneous secretions of these amphibians species may contribute to the bioactivity of defense metabolites in plants.

Objective: to understand the content and structure of social representations for health professionals about the rapid test for the detection of sexually transmitted infections in a municipality in Bahia. Method: qualitative research based on the theory of social representations, in its structural approach, carried out in 12 basic health units. 37 higher-level health workers participated. Data collection was performed through a script that included questions related to the professional characterization, plus the inductive expression rapid test. The corpus was organized and processed with the help of the software Ensemble de Programm Espermettant Lanalyse des Evocations. Results: the possible constituent elements of the central core of the representation for these workers, in direct order, were represented by the term HIV and, instead, by fear, HIV, diseases and agility. The senses attributed to the fast test, disclosed in the central core, show discrepancy considering the direct order and the substitution among the representations and, in a certain measure, they characterize them as distinct representations.(AU)

RESUMO

PURPOSE: Nonalcoholic steatohepatitis (NASH) is characterized by steatosis, lobular inflammation, ballooning, and in some cases, fibrosis, which can progress to cirrhosis and carcinoma. The progression of NASH is closely related to oxidative stress. Dietary intake of antioxidants has been suggested in protection against oxidative damage and related clinical complications. Thus, we evaluated the potential association of dietary total antioxidant capacity (TAC) with disease severity in NASH patients, as well as with anthropometric and body composition markers and biochemical parameters. METHODS: Thirty-three outpatients with a mean age of 48.4 ± 1.9 years were evaluated. Dietary TAC was estimated from a quantitative food frequency questionnaire. NASH severity, determined by liver biopsy, lifestyle characteristics, occurrence of comorbidities, anthropometry, body composition, and biochemical parameters were assessed. RESULTS: NASH patients who had a higher dietary TAC had fewer ballooned hepatocytes compared to those with a lower TAC (p = 0.024). The patients with the highest dietary TAC had a reduction of approximately 20% in the risk of having many ballooned hepatocytes (OR 0.791; 95% CI 0.643-0.974; p = 0.027). There was no association of steatosis, lobular inflammation, and fibrosis with dietary TAC. The same occurred for lifestyle characteristics, occurrence of comorbidities, anthropometry, body composition, and biochemical parameters. CONCLUSION: Dietary TAC is higher in patients with lower hepatic injury (ballooning), suggesting a possible role of food intake naturally high in its antioxidant capacity in reducing free radical production and, consequently, oxidative stress.

RESUMO

Objectives: The objectives of the present study are to investigate the association among symptoms of depression and anxiety, sociodemographic variables and personality factors and also verify the predictive power of these variables in cancer outpatients. Method: The sample was composed by 220 individuals, 138 (62.7%) women and 82 (37.3%) men. The instruments used were: Questionnaire on sociodemographic and clinical data; NEO-FFI Personality Inventory Revised (NEO-FFI-R) and Hospital Anxiety and Depression Scale (HADS). The t test and the Spearman correlation were used to verify the relation between sociodemographic and clinic variables and symptoms of anxiety and depression. A Multiple Linear Regression analysis was made to verify the predictive power of sociodemographic and clinic variables and personality factors. Results: The personality factor neuroticism was predictor of anxiety symptoms. The variable previous psychiatric history and the personality factors neuroticism and extraversion were predictors of depressive symptoms. Conclusion: Depression and anxiety symptoms are associated with some sociodemographic variables and personality factors, and these variables can be deemed risk factors for the development of these symptoms. It is important that health professionals who follow-up this population design strategies to collect this information, in order to help patients manage treatment the best way possible

Objetivos: Los objetivos del presente estudio son investigar la asociación entre síntomas de depresión y ansiedad, variables sociodemográficas y factores de personalidad y también verificar el poder predictivo de estas variables en pacientes con cáncer. RESUMO

OBJECTIVES: This work aimed to evaluate the antifungal and cytotoxic activity of the EtOH extract and fractions of Banisteriopsis argyrophylla leaves, and to perform the identification of these bioactive metabolites. METHODS: The EtOAc fraction (EAF) obtained from the ethanolic extract of B. argyrophylla leaves showed better antifungal potential against Candida spp. In this fraction, ten flavonoids have been identified by UHPLC-ESI-MSn . Then, EAF was submitted to column chromatography to give four new fractions (A1-A4). The cytotoxicity was determined against Vero cells. KEY FINDINGS: The EAF showed better antifungal potential against Candida spp. with minimum inhibitory concentrations (MICs) between 31.25 and 93.75 µg/ml. The (-)-catechin (fraction A1) showed a MIC of 2.83 µg/ml against Candida glabrata. Fractions A2, A3 and A4 were rich in quercetins and kaempferols and showed good inhibitory concentrations (5.86-46.87 µg/ml) against C. albicans, C. glabrata and C. tropicalis. CONCLUSIONS: The EtOH extract, fractions and the isolated (-)-catechin showed lower toxicity to Vero cells than cisplatin, used as a positive control. Thus, the leaves of B. argyrophylla are a promising source of antifungal agents.
