Gastrodia Rhizoma, as a precious Chinese materia medica, has attracted the attention of Chinese materia medica experts in the past dynasties for the commercial specification and experimental identification, and has gradually formed a wealth of terms concerning commercial specification and experimental identification. Through combing the literatures of successive dynasties, this paper discussed the change of the commercial specification of the Gastrodiae Rhizoma and formation of its identifying terms. It has found that the Gastrodiae Rhizoma mainly came from the dried rhizomes of the Gastrodia elata f.elata before the Qing Dynasty. Since the Qing Dynasty, G. elata in Yunnan and Guizhou gradually arose and become one of sources of mainstream commodities. After that, G. elata f. glauca and G. elata f. elata were becoming the main sources of Gastrodiae Rhizoma. Before the large-scale cultivation of G. elata in the 1970 s, there is only wild G. elata over the country. In terms of commercial specification, they were often classified into Chunma and Dongma according to their harvest time. With the successful promotion of cultivation technology and the endangered wild resources of G. elata, the Dongma became the mainstream in the market. The adulterants of G. elata increased significantly in the 1960 s and 1970 s, in this period, the terms of experimental identification for G. elata also increased obviously. Experimental identification is distinctive in different times, therefore, studying experimental identification of medicinal materials helps to promote the development of the Chinese materia medica.

Picophytoplankton (<3 µm), comprising picocyanobacteria (PCY) and photosynthetic picoeukaryotes (PPEs), are considerably important in the material circulation and energy flow of aquatic ecosystems. To explore the temporal and spatial variation patterns of picophytoplankton and their correlations with environmental factors in lotic Yangtze-connected lakes, field in-situ investigations were performed on a monthly basis during the wet season (May to August) in 2019 in East Lake Dongting, a Yangtze-connected lake. The results indicated that both the Chla biomass and abundances of picophytoplankton exhibited significant spatial and temporal variability (P<0.05). The picophytoplankton Chla biomass showed an average concentration of 8.52 µg·L-1 and accounted for 41.6% to total phytoplankton on an average. From May to August, Chla biomass of picophytoplankton kept increasing with increasing temperature, especially in the north and south of the lake, and it was the lowest in the east of the lake. PCY dominated picophytoplankton abundance in East Lake Dongting and was 3.4 times the abundance of PPEs on an average. Similar spatial and temporal variation patterns were observed between PCY and PPEs. The abundances of PCY and PPEs both increased first and then decreased during the wet season. Spatially, picophytoplankton showed a trend to migrate from the northern lake to the southern lake from May to July, and the abundance significantly declined in August and peaked mainly in the north of the lake. The analysis results showed that picophytoplankton in East Lake Dongting exhibited significant spatial and temporal variability during the wet season; the water level and N:P ratio were determined to be the most important factors explaining the variation of the abundance proportion of PCY and PPEs.

As a medicine, Heshouwu was first appeared in Kaibao Bencao in Song Dynasty, which was a traditional tonic medicine. In recent years, the hepatotoxicity of Heshouwu has attracted great attention of domestic and foreign scholars. This paper has carried out textual research on illustration, quality evaluation, toxic and side effects of the Heshouwu in the historical Bencao(materia medica) literature. Illustrations of Heshouwu in the historical Bencao literatures are basically Polygonum multiflorum. Only in Taiyixian Zhibencao Yaoxing Daquan and Tujie Bencao, illustrations of Heshouwu may be Pteroxygonum giraldii and Dioscorea bulbifera, respectively. Illustrations of Heshouwu in the historical Bencao literatures are quite different from those of the genus Cynanchum in Asclepiadaceae. In the biography of Heshouwu, the author has noticed the abnormal vascular bundle in the root tuber of Heshouwu, which was inherited by the historical Bencao literatures. This is also one of the evidences to infer that Heshouwu is from Polygonaceae plant P. multiflorum. The description of abnormal vascular bundle in the historical Bencao literatures has been developed and evolved into the term "brocaded patterns", and it is considered that "five petals are good". Furthermore, ancient medical scientists have noticed that there are toxic differences in different characters of Heshouwu, that is, "Heshouwu with abundant parenchyma cells, low fibrous and large is superior in quality", "the hard and fibrous root tuber of P. multiflorum has toxic side effects". In this paper, the germplasm and the theory of assessing the quality by distinguishing features of traditional Chinese medicinal materials of Heshouwu in the historical Bencao literatures are reviewed, which provides the basis of materia medica for the quality evaluation of Heshowu.

OBJECTIVE: To investigate the current status of antibiotic use for very and extremely low birth weight (VLBW/ELBW) infants in neonatal intensive care units (NICUs) of Hunan Province. METHODS: The use of antibiotics was investigated in multiple level 3 NICUs of Hunan Province for VLBW and ELBW infants born between January, 2017 and December, 2017. RESULTS: The clinical data of 1 442 VLBW/ELBW infants were collected from 24 NICUs in 2017. The median antibiotic use duration was 17 days (range: 0-86 days), accounting for 53.0% of the total length of hospital stay. The highest duration of antibiotic use was up to 91.4% of the total length of hospital stay, with the lowest at 14.6%. In 16 out of 24 NICUs, the antibiotic use duration was accounted for more than 50.0% of the hospitalization days. There were 113 cases with positive bacterial culture grown in blood or cerebrospinal fluid, making the positive rate of overall bacterial culture as 7.84%. The positive rate of bacterial culture in different NICUs was significantly different from 0% to 14.9%. The common isolated bacterial pathogens Klebsiella pneumoniae was 29 cases (25.7%); Escherichia coli 12 cases (10.6%); Staphylococcus aureus 3 cases (2.7%). The most commonly used antibiotics were third-generation of cephalosporins, accounting for 41.00% of the total antibiotics, followed by penicillins, accounting for 32.10%, and followed by carbapenems, accounting for 13.15%. The proportion of antibiotic use time was negatively correlated with birth weight Z-score and the change in weight Z-score between birth and hospital discharge (rs=-0.095, -0.151 respectively, P<0.01), positively correlated with death/withdrawal of care (rs=0.196, P<0.01). CONCLUSIONS: Antibiotics used for VLBW/ELBW infants in NICUs of Hunan Province are obviously prolonged in many NICUs. The proportion of routine use of third-generation of cephalosporins and carbapenems antibiotics is high among the NICUs.

Various types of bovine serum albumin (BSA)-protected fluorescent gold nanoclusters (BSA-AuNCs) have been fabricated and applied in various fields. However, the conventional synthesis methods for BSA-AuNCs usually yield a low photoluminescence quantum yield (PLQY) in solution. In this study, we systematically examined the influences of incubation time, temperature, and pH on the formation process of BSA-AuNCs and then developed a novel strategy to synthesize BSA-AuNCs with PLQY (26%), far exceeding that of existing counterparts. Of the three important factors, pH, temperature, and time, pH plays a key role in the formation of BSA-AuNCs with different compositions and fluorescence properties. The matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) results showed that BSA-Au20NCs with high purity can be produced at a pH value of 10 and the correct combination of incubation temperature and reaction time. The advantages of the obtained BSA-Au20NCs, including small size, high PLQY, long lifetime, high purity, as well as facile modification, make them ideal candidates for luminescent probes in imaging and sensing applications.

Through consulting the ancient herbs and medical books, combining with modern literature, this paper makes textual research on herbal medicine, and textual research on the name, origin, position and harvest and processing changes of the medicinal herbs in ancient classical prescriptions. According to research, Cinnamon medicinal materials were first listed in the Shennong's Herbal Classic, as the name of "Jungui" and "Mugui". Among them, Jungui has undergone the evolution of "Qungui-Jungui-Tonggui-Jungui". After the Northern and Southern Dynasties, a half-volume fatty "Gui" was added, but the usage of the three was no different. The names of Cinnamomi Ramulus and Cinnamomi Cortex did not appear until the Tang Dynasty, and they were preferably thick-skinned and with no cork cambium, and they were mostly used in the name of "Guixin"; Since the Song and Yuan Dynasties, the medicinal parts of cassia have gradually separated. Cinnamomi Cortex is the trunk bark of sapling or branch bark, the twig is Cinnamomi Ramulus, and the tenderest twig is Liugui, Song Dynasty unified the name as "Guizhi"; After the Ming and Qing Dynasties, Cinnamomi Cortex was used as the trunk bark, and Cinnamomi Ramulus was used as the tender twig; In modern times, the Chinese Pharmacopoeia stipulates that the Cinnamomi Ramulus is the young branch of C. cassia, which is Cinnamomum, and Cinnamomi Cortex was the dried trunk bark. From the plant morphology recorded in the previous herbals and the drawings, combined with the distribution of the origins described in the previous herbals, the mainstream plant used as a medicine in the past generations should be C. cassia, but there are other sect. Cinnamomum plants that are also used in medicine everywhere, such as C. chekiangensis, C. bejolghota, C. wilsonii, etc. Throughout the ages, different plant morphologies and medicinal traits have been used to distinguish different categories. The origins of the past dynasties are mostly present in Guangdong, Guangxi province and Vietnam, and are regarded as authentic. The methods for the harvest and processing of cinnamon medicinal materials are basically the same from ancient to modern times.

Soybean is one of the most important vegetable oil and protein feed crops. To capture the entire genomic diversity, it is needed to construct a complete high-quality pan-genome from diverse soybean accessions. In this study, we performed individual de novo genome assemblies for 26 representative soybeans that were selected from 2,898 deeply sequenced accessions. Using these assembled genomes together with three previously reported genomes, we constructed a graph-based genome and performed pan-genome analysis, which identified numerous genetic variations that cannot be detected by direct mapping of short sequence reads onto a single reference genome. The structural variations from the 2,898 accessions that were genotyped based on the graph-based genome and the RNA sequencing (RNA-seq) data from the representative 26 accessions helped to link genetic variations to candidate genes that are responsible for important traits. This pan-genome resource will promote evolutionary and functional genomics studies in soybean.

BACKGROUND: Novel coronavirus disease (COVID-19) is spreading globally. Little is known about the risk factors for the clinical outcomes of COVID-19 in children. METHODS: A retrospective case-control study was taken in children with severe acute respiratory syndrome coronary virus-2 infection in Wuhan Children's Hospital. Risk factors associated with the development of COVID-19 and progression were collected and analyzed. RESULTS: Eight of 260 children diagnosed with severe COVID-19 pneumonia were included in the study. Thirty-five children with COVID-19 infection matched for age, sex and date of admission, and who classified as non-severe type, were randomly selected from the hospital admissions. For cases with severe pneumonia caused by COVID-19, the most common symptoms were dyspnea (87.5%), fever (62.5%) and cough (62.5%). In laboratory, white blood cells count was significantly higher in severe children than non-severe children. Levels of inflammation bio-makers such as hsCRP, IL-6, IL-10 and D-dimer elevated in severe children compared with non-severe children on admission. The level of total bilirubin and uric acid clearly elevated in severe children compared with non-severe children on admission. All of severe children displayed the lesions on chest CT, more lung segments were involved in severe children than in non-severe children, which was only risk factor associated with severe COVID-19 pneumonia in multivariable analysis. CONCLUSIONS: More than 3 lung segments involved were associated with greater risk of development of severe COVID-19 in children. Moreover, the possible risk of the elevation of IL-6, high total bilirubin and D-dimer with univariable analysis could identify patients to be severe earlier.

Acute intermittent porphyria (AIP) is a rare inherited disorder, which is caused by the partial deficiency of hydroxymethylbilane synthase (HMBS), an enzyme of the heme biosynthetic pathway. Abdominal pain, neuropsychiatric disturbance and neuropathy are the typical manifestations of the disease. Complications such as posterior reversible encephalopathy syndrome (PRES), a rare type of brain lesion present on MRI, are also observed in patients with AIP. The present study reports on the case of a 36­year­old Chinese female patient with AIP and PRES. Genomic DNA were obtained from peripheral blood leukocytes and genomic regions of the HMBS gene were amplified as 2 fragments, which together contained all the exons and flanking intronic regions. Sanger sequencing of the amplified DNA fragments from the patient and the patient's family revealed a novel frameshift deletion (c.405­406delAA) in exon 8 of the HMBS gene. This mutation leads to a subsequent truncated protein (p.Glu135AspfsX74). The recombinant mutant protein had 62% activity relative to the wild­type protein but similar thermostability. It was confirmed that this novel mutation was the cause of AIP. Accumulation of D­aminolevulinic acid (ALA) due to HMBS dysfunction is a potential mechanism of PRES. The manifestation of PRES may be associated with ALA­induced cytotoxicity and the destruction of the blood­brain barrier. In summary, in the present study, a novel pathogenic HMBS mutation was identified, expanding on the molecular heterogeneity of AIP.

Scutellaria, or skullcaps, are medicinally important herbs in China, India, Japan, and elsewhere. Though Scutellaria is the second largest and one of the more taxonomically challenging genera within Lamiaceae, few molecular systematic studies have been undertaken within the genus; in part due to a paucity of available informative markers. The lack of informative molecular markers for Scutellaria hinders our ability to accurately and robustly reconstruct phylogenetic relationships, which hampers our understanding of the diversity, phylogeny, and evolutionary history of this cosmopolitan genus. Comparative analyses of 15 plastomes, representing 14 species of subfamily Scutellarioideae, indicate that plastomes within Scutellarioideae contain about 151,000 nucleotides, and possess a typical quadripartite structure. In total, 590 simple sequence repeats, 489 longer repeats, and 16 hyper-variable regions were identified from the 15 plastomes. Phylogenetic relationships among the 14 species representing four of the five genera of Scutellarioideae were resolved with high support values, but the current infrageneric classification of Scutellaria was not supported in all analyses. Complete plastome sequences provide better resolution at an interspecific level than using few to several plastid markers in phylogenetic reconstruction. The data presented here will serve as a foundation to facilitate DNA barcoding, species identification, and systematic research within Scutellaria, which is an important medicinal plant resource worldwide.

This study aims to systematically evaluate the efficacy of mindfulness-based intervention (MBI) in improving mental health and quality of life for people with dementia. Comprehensive literature search was performed using the PubMed, EMBASE, Web of Science, Cochrane Library, and CINAHL databases from their inception till June 26, 2019. In total, nine articles met the eligibility criteria and were included. The results of the meta-analysis showed a statistically significant decrease in depressive symptoms (SMD = -0.39, 95% CI: - 0.62 to - 0.15), in people with dementia who were treated with MBI. However, there were no significant improvements in anxiety, stress, or quality of life. These findings suggest that MBI is a promising alternative to conventional interventions in the treatment of depression among dementia patients and warrant further study.

A sodium-ethylenediamine graphite intercalation compound (Na(ethylenediamine)C15: "GIC") made from graphite flakes was used to study the microwave absorption performance of a GIC for the first time. Compared with the pristine graphite flakes, the neighboring layers in this GIC are pillared by Na(ethylenediamine)+ and possess a larger layer distance and improved electrical conductivity. Owing to the electrical conductivity of this GIC, only half of the loading content, compared to graphite flakes, is needed to achieve an outstanding absorption of -75.6 dB at 9.25 GHz (10.0 wt % GIC in paraffin in a 4.0 mm thick sample), but for graphite, 20.0 wt % is required for an absorption of -37.6 dB.

Through consulting the herbs and medical books of past dynasties, combining with modern literature and field investigation, this paper made textual research on the name, origin, producing area, harvesting and processing of Bupleuri Radix used in the Catalogue of Ancient Classical Prescriptions(the first batch).From Han to the Southern and Northern Dynasties, the source of Bupleuri Radix was a mixture of Bupleurum and Peucedanum, it's hard to define the origin of authentic products.In Tang Dynasty, Caryophyllaceae had been used as Bupleuri Radix. In Song Dynasty, it was clear that the genuine production area of Bupleuri Radix was Yinzhou, its origin was B. corzonerifolium and B. yinchowense.After the Ming Dynasty, B. chinense gradually became the mainstream. It has been confirmed that there were differences in the efficacy between Stellaria dichotoma in Caryophyllaceae and Bupleurum in Umbelliferae, however, the use of S. dichotoma is still attributed to Bupleuri Radix. In the Qing Dynasty, S. dichotoma was distinguished from Bupleuri Radix as another medicine. Among the prescriptions containing Bupleuri Radix included in the Catalogue of Classical Prescriptions in Ancient China(the first batch), the mainstream of genuine Bupleuri Radix should be B. scorzonerifolium and B. chinense. It is suggested that the genuine base of Bupleuri Radix should be selected according to the current resources and industrial development of Bupleuri Radix, and it should conform to the Chinese Pharmacopoeia.

The lack of sufficient functional tumor-infiltrating lymphocytes in the tumor microenvironment (TME) is one of the primary indications for the poor prognosis of patients with cancer. In this study, we developed an Erbitux-based IL-21 tumor-targeting fusion protein (Erb-IL21) to prolong the half-life and improve the antitumor efficacy of IL-21. Compared with Erb-IL2, Erb-IL21 demonstrated much lower toxicity in vivo. Mechanistically, Erb-IL21 selectively expanded functional cytotoxic T lymphocytes but not dysfunctional CD8+ T cells in the TME. We observed that the IL-21-mediated antitumor effect largely depended on the existing intratumoral CD8+ T cells, instead of newly migrated CD8+ T cells. Furthermore, Erb-IL21 overcame checkpoint blockade resistance in mice with advanced tumors. Our study reveals that Erb-IL21 can target IL-21 to tumors and maximize the antitumor potential of checkpoint blockade by expending a subset of tumor antigen-specific CD8+ T cells to achieve effective tumor control.

Protein-supported nanoparticles have a great significance in scientific and nanotechnology research because of their "green" process, low cost-in-use, good biocompatibility, and some interesting properties. Ruthenium oxide nanoparticles (RuO2NPs) have been considered to be an important member in nanotechnology research. However, the biosynthetic approach of RuO2NPs is relatively few compared to those of other nanoparticles. To address this challenge, this work presented a new way for RuO2NP synthesis (BSA-RuO2NPs) supported by bovine serum albumin (BSA). BSA-RuO2NPs are confirmed to exert peroxidase-like activity, electrocatalytic activity, in vitro salt resistance (2 M NaCl), and biocompatibility. Results indicate that BSA-RuO2NPs have higher affinity binding for 3,3',5,5'-tetramethylbenzidine or H2O2 than bare RuO2NPs. Moreover, BSA turns out to be a crucial factor in promoting the stability of RuO2NPs. Taking the advantages of these improved properties, we established colorimetric (linear range from 2 to 800 µM, a limit of detection of 1.8 µM) and electrochemical (linear range from 0.4 to 3850 µM, a limit of detection of 0.18 µM) biosensors for monitoring in situ H2O2 secretion from living MCF-7 cells. Herein, this work offers a new biosynthesis strategy to obtain BSA-RuO2NPs and sheds light on the sensitive biosensors to monitor the H2O2 secreted from living cells for promising applications in the fields of nanotechnology, biology, biosensors, and medicine.

Chronic hepatitis B is caused by prolonged infection with the hepatitis B virus (HBV), which can substantially increase the risk of developing liver disease. Despite the development of preventive vaccines against HBV, a therapeutic vaccine inducing an effective antibody response still remains elusive. The preS1 domain of the large HBV surface protein is the major viral attachment site on hepatocytes and thus offers a therapeutic target; however, its poor immunogenicity limits clinical translation. Here, we design a ferritin nanoparticle vaccine that can deliver preS1 to specific myeloid cells, including SIGNR1+ dendritic cells (which activate T follicular helper cells) and lymphatic sinus-associated SIGNR1+ macrophages (which can activate B cells). This nanoparticle vaccine induces a high-level and persistent anti-preS1 response that results in efficient viral clearance and partial serological conversion in a chronic HBV mouse model, offering a promising translatable vaccination strategy for the functional cure of chronic hepatitis B.

In this study, a colorimetric sensing assay of isoniazid based on excellent oxidase-like activity of heparin sodium stabilized platinum nanoparticles (HS-PtNPs) has been demonstrated. The newly prepared HS-PtNPs exhibit a great dispersion with an average size distribution of 4.8 ± 0.6 nm, and maintain more than 90% catalytic activity under strong acid and alkali or long-term storage conditions, indicating a robust nanomaterial with attractive potential. The HS-PtNPs show distinct oxidase-like activity with an ultrahigh affinity (Km = 0.01012 mM) for 3, 3', 5, 5'-tetramethylbenzidine (TMB). More significantly, we found that the pyridine ring of isoniazid has a strong reductive hydrazyl substitution, which can compete with TMB for the catalytic site of HS-PtNPs resulting in a colorless solution. Accordingly, a colorimetric sensing of isoniazid was fabricated. A linear relationship for isoniazid was achieved in 2.5 × 10-6 to 2.5 × 10-4 M (R2 = 0.998) with a low limit of detection 1.7 × 10-6 M (S/N = 3). Recovery experiments in drug tablets show that the standard recovery rates were 95%-103%. The quantitative detection data for isoniazid in drug tablets calculated respectively from the standard method and this method exhibited a high correlation coefficient (a slope of 0.9995), suggesting that high accuracy in isoniazid detection.
