RESUMO

The rubber tree [Hevea brasiliensis (Willd. Ex Adr. de Juss.) Muell.-Arg] is the main source of natural rubber in the world. However, in the Amazon region, its production is reduced by biotic and abiotic limitations, which have prompted breeding programs in order to identify desirable agronomic and physiological indicators. The objective of this study was to analyze the temporal dynamics of photosynthetic responses based on the parameters of leaf gas exchange and chlorophyll a fluorescence in 10 rubber tree clones during the immature phase (pre-tapping) in three large-scale clone trials, during daily cycles and under two climatic periods (dry and rainy) in the Caquetá region (Colombian Amazon). The variables A, LT, ΦPSII, ETR and qP were significantly higher in the dry period, where the highest values of PAR, AT and VPD were seen. In San Vicente del Caguán and Florencia, the highest averages were estimated for A, E and gs, as compared with Belén de los Andaquíes. In Florencia, the highest fluorescence parameters of chlorophyll a were recorded. At 9:00 h and 12:00 h, the highest means of A, E, ΦPSII and ETR were observed. The majority of the clones displayed the highest Fv/Fm mean (0.82-0.84) in the dry period. The clones FX 4098, FDR 4575, MDF 180, GU198 and FDR 5788 represent genotypes with the best photosynthetic performance (greater photosynthetic rates and better ability of the photosynthetic apparatus to capture, use and dissipate light energy). These desirable genotypes constitute a promising gene pool for expanding the genetic resource of rubber trees in the Colombian Amazon.

RESUMO

Bacterially expressed proteins used in NMR studies lack glycans, and proteins from other organisms are neither 15N labeled nor glycosylated homogeneously. Here, we add two artificial glycans to uniformly 15N labeled prion protein using a buffer system that evolves over a pH range to accommodate the conflicting pH requirements of the substrate and enzymes without the need to fine-tune buffer conditions. NMR and CD spectroscopy of the protein indicates that the glycans do not influence its fold.

RESUMO

Previously we demonstrated, in rats, that treatment with growth hormone (GH) and rehabilitation, carried out immediately after a motor cortical ablation, significantly improved the motor affectation produced by the lesion and induced the re-expression of nestin in the contralateral motor cortex. Here we analyze cortical proliferation after ablation of the frontal motor cortex and investigate the re-expression of nestin in the contralateral motor cortex and the role of the striatum and thalamus in motor recovery. The rats were subjected to ablation of the frontal motor cortex in the dominant hemisphere or sham-operated and immediately treated with GH or the vehicle (V), for five days. At 1 dpi (days post-injury), all rats received daily injections (for four days) of bromodeoxyuridine and five rats were sacrificed at 5 dpi. The other 15 rats (n = 5/group) underwent rehabilitation and were sacrificed at 25 dpi. GH induced the greatest number of proliferating cells in the perilesional cortex. GH and rehabilitation produced the functional recovery of the motor lesion and increased the expression of nestin in the striatum. In the thalamic ventral nucleus ipsilateral to the lesion, cells positive for nestin and actin were detected, but this was independent on GH. Our data suggest that GH-induced striatal nestin is involved in motor recovery.

RESUMO

The cellular mechanisms underlying the amazing ability of sea cucumbers to regenerate their autotomized intestines have been widely described by us and others. However, the signaling pathways that control these mechanisms are unknown. Previous studies have shown that Wnt homologs are upregulated during early intestinal regenerative stages, suggesting that the Wnt/ß-catenin pathway is active during this process. Here, we used small molecules, putative disruptors of the Wnt pathway, to determine the potential role of the canonical Wnt pathway on intestine regeneration in the sea cucumber Holothuria glaberrima. We evaluated their effects in vivo by using histological analyses for cell dedifferentiation, cell proliferation and apoptosis. We found that iCRT14, an alleged Wnt pathway inhibitor, decreased the size of the regenerating intestine, while LiCl, a presumed Wnt pathway activator, increased its size. The possible cellular mechanisms by which signaling pathway disruptors affect the gut rudiment size were further studied in vitro, using cultures of tissue explants and additional pharmacological agents. Among the tested signaling activators, those that act through GSK-3 inhibition, LiCl, 1-Azakenpaullone, and CHIR99021 were found to increase muscle cell dedifferentiation, while the inhibitor iCRT14 blocked cell dedifferentiation. Differently, cell proliferation was reduced by all GSK-3 inhibitors, as well as by iCRT14 and C59, which interferes with Wnt ligand secretion. The in vivo temporal and spatial pattern of ß-catenin activity was determined using an antibody against phosphorylated ß-catenin and shown to correlate with cell proliferative activity. In vitro treatment using C59 decreased the number of cells immunostained for nuclear phosphorylated ß-catenin. Our results showed that the cell dedifferentiation observed during intestinal regeneration can be decoupled from the cell proliferation event and that these cellular processes can be modulated by particular signaling pathway inhibitors and activators. These results open the door for future studies where the cellular signaling pathways involved at each regeneration stage can be determined.

RESUMO

Background: To establish the prevalence of supernumerary canines (SNC) in a sector of the population of Madrid (Spain), as well possible complications associated with this unusual developmental variation. Study design: This observational study was performed between 2005 and 2017, among 21,615 patients seeking dental treatment at the Faculty of Dentistry, Complutense University of Madrid (Spain), and at the Virgen de la Paloma Hospital, Madrid (Spain); 22 patients with 26 SNCs were diagnosed. These 22 patients underwent clinical and radiological exploration, registering patient data. Results: SNCs presented a prevalence of 0.10% of the study population. The supernumerary teeth (SNT) were located in the upper maxilla more frequently (61.54%) than the mandible (38.46%). 69.23% were found to be impacted, also causing the impaction of the permanent canine in 53.85% of these cases. In 15.38%, follicular expansion > 3mm was observed. SNCs were associated with other SNT in only four patients. Conclusions: Despite of the fact that the SNCs are usually diagnosed casually in the course of radiological explora-tion, in the present study over half of them (53.85%) caused impaction of the permanent canine. Early diagnosis allows optimal patient management and treatment planning, with intervention at an appropriate time to prevent complications in development and so reduce later treatment need

RESUMO

While Helicobacter pylori is a fundamental risk factor, gastric cancer (GC) aetiology involves combined effects of microbial (both H. pylori and non-H. pylori), host and environmental factors. Significant differences exist between the gastric microbiome of those with gastritis, intestinal metaplasia and GC, suggesting that dysbiosis in the stomach is dynamic and correlates with progression to GC. Most notably, a consistent increase in abundance of lactic acid bacteria (LAB) has been observed in GC patients including Streptococcus, Lactobacillus, Bifidobacterium and Lactococcus. This review summarises how LAB can influence GC by a number of mechanisms that include supply of exogenous lactate -a fuel source for cancer cells that promotes inflammation, angiogenesis, metastasis, epithelial-mesenchymal transition and immune evasion-, production of reactive oxygen species and N-nitroso compounds, as well as anti-H. pylori properties that enable colonization by other non-H. pylori carcinogenic pathobionts.

RESUMO

Opioid-binding protein/cell adhesion molecule (OPCML) is expressed in many tissues and localizes to the plasma membrane. It is a putative tumor suppressor that acts on receptor tyrosine kinases (RTKs), influencing cell cycle arrest, apoptosis, and migration. Its expression is silenced epigenetically in cancer, inferring potential diagnostic, prognostic, and therapeutic roles.

RESUMO

OBJECTIVE: To describe patient characteristics, radiological findings and the clinical course of adults with fatal reversible cerebral vasoconstriction syndrome (RCVS). METHODS: A systematic literature search from January 1, 2000, until December 31, 2018, was performed using PubMed, EMBASE, Scopus, Cochrane reviews, LILACS and Scielo. Studies reporting RCVS in adult patients with fatal outcomes were included. RESULTS: 430 studies were initially identified, 179 full-text articles were reviewed, and 9 publications describing 12 subjects were included. The vast majority of the reports were from the U.S. Most of the female cases occurred during postpartum. All patients had a headache on initial presentation, although only 42% had thunderclap headache. A CT scan was performed on 67% of the patients. Imaging results were diverse, with a tendency toward cerebral hemorrhage followed by mixed cases. The main course of treatment included steroids (58% of the patients), with only 42% receiving nimodipine. The time to death ranged from 4 to 14 days, with a median of 9.2 days (SD ±â€¯3.2). CONCLUSION: We found that the majority of fatal cases reported in the literature are most likely related to postpartum angiopathy. We established a tendency in the onset of brain hemorrhage and the combination of infarction and brain hemorrhage. We described various markers for poor prognosis, including focal signs, the presence of hemorrhage and infarct in the first diagnostic image obtained and the need for invasive interventions. The majority of fatal cases in our report occurred in women, with over half of those cases during the puerperium period.

RESUMO

Borderline personality organization (BPO) is a key personality component of some but not all partner-violent men. The study described in this paper examines the psychometric properties of the borderline personality organization scale (BPO Scale; Oldham et al., 1985) in a Spanish sample of 643 men undergoing court-mandated psychological treatment after conviction for episodes of intimate-partner violence. Three confirmatory factor analyses were carried out first, and the three-factor structure of the BPO scale was then tested. Results for concurrent validity show positive and significant correlations between the subscales and the overall BPO scale, and with other instruments that measure borderline and antisocial personality disorders (ASPDs), and impulsivity. The BPO scale also presents evidence of known-groups validity, since BPO scores decrease with age, and of discriminant validity, as the scale discriminates between participants who do and do not exceed the cutoff point on a borderline personality scale. The BPO Scale is a suitable instrument for evaluating BPO in partner-violent men.

Introduction: The peripheral giant cell granuloma (GPCG) is a rare exophytic lesion of the oral cavity, whose appearance is usually associated with the natural tooth and whose etiology is related, among other factors, to the chronic effect over time of various local irritants. With the increasing use of dental implants, the appearance of this type of lesions in the peri-implant mucosa has been observed, which requires studying and understanding its etiology and evolution in order to guarantee the longevity of this type of rehabilitation. Clinical case: We present the case of a 41-year-old female patient, with no medical history of interest, referred to the Oral Surgery Department of the Hospital Virgen de la Paloma, which presents a sessile base tumor associated with an implant in position 47, of 1.2x0.6 cm, painless, firm consistency and nondisplaceable with congestive coloring. A total exeresis treatment of the lesion was carried out, sending the piece to histopathological study, after which the diagnosis of GPCG was obtained. Conclusions: It is vital to maintain an exhaustive control of the various local factors that can be identified as irritants for peri-implant tissues, such as poor oral hygiene, among others, in order to prevent the appearance of injuries that could compromise survival and success of long-term implants

RESUMO

OBJECTIVE: Obsessive-compulsive disorder (OCD) is a debilitating neuropsychiatric disorder with an etiopathophysiology that seems to include immune alterations. Previous studies have suggested that variations in the levels of circulating T cell subpopulations may be involved in psychiatric diseases. However, the role of these cells in OCD remains unexplored. Hence, the present study aimed to examine the levels of T helper 1 (Th1), Th2, Th17 and regulatory T (Treg) cells in patients with early-onset OCD and healthy controls. METHODS: The assessment was performed in 99 children and adolescents with OCD and 46 control subjects. The percentages of circulating Th1, Th2, Th17 and Treg cells were evaluated using flow cytometry. RESULTS: OCD patients had significantly higher levels of Th17 cells and lower percentages of Treg cells than healthy controls (p = 0.001 and p = 0.005, respectively). Furthermore, levels of Th17 cells progressively increased with the duration (p = 0.005) and severity of OCD (p = 0.008), whereas the percentages of Treg cells significantly declined with the duration of the disorder (p = 1.8 × 10-5). CONCLUSIONS: These results provide more evidence of the involvement of immune dysregulation, specifically an imbalance in the levels of circulating T helper and regulatory T cells, in the pathophysiology of early-onset OCD.

Transmigration is defined as the phenomenon in which an unerupted included tooth tran-scends the midline by more than half of its length. It is a rare anomaly and it is even more rare to occur bilaterally. It is presented with greater incidence in the mandibular canines, in the female sex, unilaterally, and the left canine being the most commonly affected. The objective of this study is to describe satisfactory surgical management in a 16-year-old asymptomatic male patient, which presented to the intraoral clinical examination absence of both permanent mandibular canines and temporary biscuspid persistence. Lower left, accompanied by a slight dental malposition. In the radiographic exploration (panoramic), both right and left lower canines were observed, in a horizontal position, under the lower incisor apexes. We proceeded to the surgical extraction of both canines, passing the post-operative period without incidents. Satisfactory periodic and radiographic controls were performed at six months. Keywords: Tooth, impacted; Tooth migration; Tooth, unerupted; Cuspid (source: MeSH NLM).

RESUMO

Rapid response teams have become necessary components of patient care within the hospital community, including for airway management. Pediatric patients with an increased risk of having a difficult airway emergency can often be predicted on the basis of clinical scenarios and medical history. This predictability has led to the creation of airway consultation services designed to develop airway management plans for patients experiencing respiratory distress and who are at risk for having a difficult airway requiring advanced airway management. In addition, evolving technology has facilitated airway management outside of the operating suite. Training and continuing education on the use of these tools for airway management is imperative for clinicians responding to airway emergencies. We describe the comprehensive multidisciplinary, multicomponent Pediatric Difficult Airway Program we created that addresses each component identified above: the Pediatric Difficult Airway Response Team (PDART), the Pediatric Difficult Airway Consult Service, and the pediatric educational airway program. Approximately 41% of our PDART emergency calls occurred in the evening hours, requiring a specialized team ready to respond throughout the day and night. A multitude of devices were used during the calls, obviating the need for formal education and hands-on experience with these devices. Lastly, we observed that the majority of PDART calls occurred in patients who either were previously designated as having a difficult airway and/or had anatomic variations that suggest challenges during airway management. By instituting the Pediatric Difficult Airway Consult Service, we have decreased emergent Difficult Airway Response Team calls with the ultimate goal of first-attempt intubation success.

RESUMO

Background: The Revised Conflict Tactics Scale (CTS-2) is widely used to assess intimate partner violence (IPV). Given the limitations preventing the collection of dyadic data, it is necessary to examine the relationship between the offender’s own self-reporting and the facts established in trial proceedings. This study assesses the relationship between aggression data self-reported by participants via CTS-2 and the data contained in convictions for partner abuse. Method: Two groups of men convicted of abuse against their female intimate partners or ex-partners (1,998 imprisoned offenders and 804 court-referred offenders), and 590 men from the community (general population) participated. The relationship was analyzed between the scores for self-reported CTS-2 items and violent behaviors described by proven facts. An ANCOVA with post-hoc comparisons (Bonferroni) was performed to assess the differences in CTS-2 scores between the three groups. Results: In the two groups of convicted participants, a significant relationship was found between CTS-2 items and the proven facts. Meanwhile, significant differences were found between the three groups in three CTS-2 subscales. Conclusions: The CTS-2 self-reported perpetrated aggression data provided by the convicted participants are related to the factual basis for convictions, and such data are therefore usable in forensic and psychological intervention contexts

Several scales are used in Dating Violence studies assuming cross-cultural invariance and equivalence of the measures without making the proper validation in the intended populations. This study focuses on the importance of adapting existing dating violence psychological instruments (as the widely recognized Modified Version of the Conflict Tactics Scale, M-CTS) in diverse adolescent populations adjusting to international validation procedures that ensure the cultural fit of the instrument and the measurement invariance of the construct. We sought to adapt the M-CTS in Mexican adolescents (N = 1861; 57.5% woman) following the ITC Guidelines for Translating and Adapting Test. We made an analysis of the linguistic and cultural variables, followed by a Confirmatory Factor Analysis, and the evaluation of Construct and Known Groups Validities. We culturally modified six items and verified the four-factorial structure of the questionnaire proposed in previous studies (argumentation, psychological aggression, mild physical aggression, and sever physical aggression). We also found significant correlations in between the scores of the M-CTS and the Aggression Questionnaire (AQ) and the Dominating and Jealous Tactics Scale (DJTS), verifying the Construct Validity of the M-CTS to measure aggressive behaviors. Conclusion: the cultural adaptation of the M-CTS offered adequate reliability and validity scores in Mexican population expanding the possibilities of comparing prevalences of the problem between nations with a reliable instrument based on the same theoretical and methodological perspectives.
