RESUMO

The regulatory pathways required to maintain eukaryotic lipid homeostasis are largely unknown. We developed a systematic approach to uncover new players in the regulation of lipid homeostasis. Through an unbiased mass spectrometry-based lipidomic screening, we quantified hundreds of lipid species, including glycerophospholipids, sphingolipids, and sterols, from a collection of 129 mutants in protein kinase and phosphatase genes of Saccharomyces cerevisiae. Our approach successfully identified known kinases involved in lipid homeostasis and uncovered new ones. By clustering analysis, we found connections between nutrient-sensing pathways and regulation of glycerophospholipids. Deletion of members of glucose- and nitrogen-sensing pathways showed reciprocal changes in glycerophospholipid acyl chain lengths. We also found several new candidates for the regulation of sphingolipid homeostasis, including a connection between inositol pyrophosphate metabolism and complex sphingolipid homeostasis through transcriptional regulation of AUR1 and SUR1. This robust, systematic lipidomic approach constitutes a rich, new source of biological information and can be used to identify novel gene associations and function.

In order to evaluate the effect of postnatal hyperoxia on retinal structure, newborn rats were exposed to different oxygenation intervals (80 +/- 1%) with three interruptions of 21% (30 min each). Four groups of rats were exposed from birth to the 6th, 9th, 12th and 14th postnatal day, respectively and another group was placed under normoxia. After this period all oxygenated groups and the controls remained under normoxia until they were 30 days old for the structural analysis of retina. Retinal histology was carried out using conventional techniques for transmission electron microscopy (TEM). In the ganglion cell layer of the retina from rats exposed for 9 days to hyperoxia, capillaries with large projections toward the lumen, were observed as a possible consequence of cellular edema of endothelium. The most severe damage was observed in rats exposed to hyperoxia during 12 and 14 days, showing mitochondrias swollen up and without crests in the areas surrounding the capillaries, necrosis and apoptosis processes, dense bodies, cells with swollen cytoplasms and rupture of the plasmatic membrane. The results suggest that postnatal hyperoxia causes severe damages to the retina in developing rats with a direct relationship between the time exposed to oxygen and ultra structural damages.

1982 y del incremento que se producía en determinados factores de riesgo asociados a la enfermedad, se consideró la enfermedad tuberculosa como un problema prioritario de salud y se incluyó en el Plan de Salud de Cataluña. Se establecieron tres objetivos de salud y de disminución del riesgo para alcanzar el año 2000. La finalidad de este trabajo es la evaluación de estos objetivos. Población y método: Los datos epidemiológicos de la enfermedad se han obtenido del Registro de Casos de Tuberculosis de Cataluña que, desde el año 1982, recoge la información epidemiológica de todas las formas clínicas de la enfermedad. Las tasas de mortalidad estandarizadas por edad son las del Registro de Mortalidad de Cataluña. Para la información sobre prevalencia de infección y para el cálculo del riesgo anual de infección en niños de 6 a 7 años se han utilizado los estudios tuberculínicos realizados por el Programa de tuberculosis del Departament de Sanitat i Seguretat Social y por el Instituto Municipal de Salud Pública. Resultados: Se han alcanzado los tres objetivos de salud y de disminución de riesgo para el año 2000 que establecía el Plan de Salud de Cataluña sobre tuberculosis. Conclusiones: Las actuaciones ejercidas sobre los distintos factores de riesgo asociados, el descenso de la incidencia del sida y la introducción de los tratamientos directamente observados han provocado un declive de la incidencia de tuberculosis que se ha ido manteniendo durante los últimos años y ha contribuido a alcanzar los objetivos de salud previstos para el año 2000 (AU)


Background and objectives: After analizing the upward trend of tuberculosis since 1982 and the increase of some risk factors associated to this condition, tuberculosis was considered a priority health problem. Therefore, it was included in the Health Plan for Catalonia for the year 2000. Three health objectives were established to achieve in 2000. The aim of this paper will be evaluate this objectives. Subjects and method: The epidemiological data related to the disease are from the Tuberculosis Register of Catalonia. This register includes epidemiological information of all disease variations since 1982. The age standardized mortality rates were extracted from the Register of mortality of Catalonia. In order to get information about the disease prevalence and to work out the annual risk of infection in child ages 6-7, we have utilized the tuberculinic studies carried out by the Tuberculosis Program of the Department of Health and Social Security and by the Municipal Institute of Public Health of Barcelona. Results: All three health and risk-reduction objectives for 2002 were achieved. Conclusions: Interventions applied to risk factors associated, the decrease of AIDS incidence and the introduction of directly observed treatments have lead to a decline of tuberculosis incidence. It has kept constant, so the aims predicted for 2002 are all fulfilled (AU)
