Nowadays, brain natriuretic peptide (BNP-32) is fundamental to early cardiovascular clinical diagnosis, whose accurate assay is of significance by photoelectrochemistry (PEC) for the low background and high precision. Herein, a novel enhanced PEC platform was built by successive deposition of N-doped ZnO nanopolyhedra (N-ZnO NP) and protoporphyrin IX (PPIX). Specifically, the N-ZnO NP with a narrow bandgap of 2.60 eV was synthesized by direct calcination of zeolitic imidazole framework-8 (ZIF-8), and performed as the substrate to enhance the photocurrents of PPIX (as photosensitizer) whose photoelectron transfer pathway and enhanced PEC mechanism were studied in detail. Under such foundation, a label-free PEC aptasensor was developed by deposition of DNA aptamer onto the PEC platform and then ultrasensitive assay of BNP-32 based on a "signal off" model. The biosensor showed a wide linear range (1 pg mL-1- 0.1 µg mL-1) with a limit of detection (LOD) as low as 0.14 pg mL-1. This doping technique of ZnO nanomaterials provides some valuable guidelines for synthesis of advanced PEC probes in bioanalysis.

Recent investigations of fundamental electronic properties (especially the carrier transport mechanisms) of Si nanocrystal embedded in the amorphous SiC films are highly desired in order to further develop their applications in nano-electronic and optoelectronic devices. Here, Boron-doped Si nanocrystals embedded in the amorphous SiC films were prepared by thermal annealing of Boron-doped amorphous Si-rich SiC films with various Si/C ratios. Carrier transport properties in combination with microstructural characteristics were investigated via temperature dependence Hall effect measurements. It should be pointed out that Hall mobilities, carrier concentrations as well as conductivities in films were increased with Si/C ratio, which could be reached to the maximum of 7.2 cm2/V∙s, 4.6 × 1019 cm-3 and 87.5 S∙cm-1, respectively. Notably, different kinds of carrier transport behaviors, such as Mott variable-range hopping, multiple phonon hopping, percolation hopping and thermally activation conduction that play an important role in the transport process, were identified within different temperature ranges (10 K~400 K) in the films of different Si/C ratio. The changes from Mott variable-range hopping process to thermally activation conduction process with temperature were observed and discussed in detail.

Trisomy 21 is a common birth defect in humans. Screening for trisomy 21 is one of the most important tasks in routine prenatal care and robust noninvasive diagnostics are needed in clinical practice. Urinary proteomics offers a new research platform for diagnostics innovation in this context. We report here new biomarker candidates using urinary proteomics profiling. Specifically, we used liquid chromatography-tandem mass spectrometry (LC-MS/MS) to analyze the proteomes of urine samples from 19 pregnant women (aged 28-44 years) carrying fetuses with trisomy 21 and 22 healthy pregnant women (aged 27-42 years) carrying fetuses with normal karyotype. We identified more than 50 differentially expressed proteins between the trisomy 21 group and healthy group, and most of these proteins were associated with the embryonic development. Importantly, tissue inhibitor of metalloproteinases 2 (TIMP2) and lysosomal-associated membrane protein 2 (LAMP2) were further selected as potential urinary protein biomarkers. We found that the combination of TIMP2 and LAMP2 could differentiate fetuses with trisomy 21 from healthy controls with a sensitivity of 74%, a specificity of 82%, and an area under the receiver operating characteristic curves (AUC) value of 0.82 (95% confidence interval, 0.69-0.95). We conclude that TIMP2 and LAMP2 offer promise as biomarker candidates and warrant further clinical research in larger study samples. These findings further our understanding of the pathological processes involved in fetal trisomy 21 and are poised to accelerate the development of new noninvasive potential biomarkers for trisomy 21 prenatal screening.

Intrahepatic cholestasis of pregnancy (ICP) is the most common liver disease during pregnancy. Manifested with pruritus and elevation in bile acids, the etiology of ICP is still poorly understood. Although ICP is considered relatively benign for the mother, increased rates of adverse fetal outcomes including sudden fetal demise are possible devastating outcomes associated with ICP. Limited understanding of the underlying mechanisms restricted treatment options and managements of ICP. In recent decades, evolving evidence indicated the significance of autophagy in pregnancy and pregnancy complications. Autophagy is an ancient self-defense mechanism which is essential for cell survival, differentiation and development. Autophagy has pivotal roles in embryogenesis, implantation, and maintenance of pregnancy, and is involved in the orchestration of diverse physiological and pathological cellular responses in patients with pregnancy complications. Recent advances in these research fields provide tantalizing targets on autophagy to improve the care of pregnant women. This review summarizes recent advances in understanding autophagy in ICP and its possible roles in the causation and prevention of ICP.

N,P-doped carbon quantum dots (N,P-CQDs) are deemed as a promising candidate to environmentally friendly materials owing to the inexpensive, biocompatible nature. TiO2 nanowire is a prospective photocatalyst because of its efficient migration of photoexcited carriers in wastewater treatment. However, the N,P-CQDs-decorated TiO2 nanowire (N,P-CQDs/NW-TiO2) photocatalysts have been rarely reported. In this study, we build N,P-CQDs on the surface of TiO2 nanowires via a simple deposition process. Our investigations demonstrate that N,P-CQDs/NW-TiO2 has a great photocatalytic degradation for methyl blue (MB) under irradiation. The degradation rate of can reach 93.6% within 120 min under proper conditions. The excellent degradation performance of N,P-CQDs/NW-TiO2 is ascribed to the mesoporous structure and high separation rate of photoexcited carriers. In addition, the N,P-CQDs/NW-TiO2 have outstanding recycled photocatalytic capability. After being recycled four times, the N,P-CQDs/NW-TiO2 still maintain 59.9% photocatalytic activity. The fabricated nanosized photocatalyst can be widely utilized in the field of photocatalysis for wastewater treatment.

Our screening data revealed the threat macrolide antibiotics, especially azithromycin (AZN), posed to human health with its increasing occurrence in water environment. The electrochemical sensor based on molecularly imprinted polymer (MIP) is a promising platform that caters for the next generation of intelligent wastewater treatment plants (WWTPs) by virtue of its wide tolerance to water from all sources and in-situ monitoring. However, low initiation potentials of cross-linking monomers contributed by the electron-rich circumstance allowed them to usurp sites designed for functional monomers when electrically stimulated, leading to an unsatisfactory binding capacity. Another uncertainty is that multiple reaction sites of cross-linking monomers granted them complex polymerization routes and made it difficult to ensure the consistency of preparation. Serval monomers had been investigated with electrochemical tools and the performance of sensors constructed with these monomers were compared in this study. Based on the results, we proposed a protocol in which a novel functional monomer possessing a stronger electron-donating group, phenyl, was adopted to compete for the dominance in electropolymerization. Beyond that, the cross-linking monomer was modified with electron-withdrawing groups to raise its initiation potential. A monothiophene with a moderate initiation potential was also recruited as the linker to address the steric hindrance. In this way, polymerization proceeded in a specific order. It is worth mentioning that the Marangoni flow is an ideal tool to deal with the Coffee-ring deposition while drop-casting. The resulting sensor showed good performance with a limitation of detection (LOD) of 0.120 µM for AZN and a satisfactory selectivity, and the design can be applied to constructing sensors for a variety of macrolide antibiotics.

Atherosclerosis is the leading cause of cardiovascular diseases worldwide. Trimethylamine N-oxide (TMAO), a metabolite of intestinal flora from dietary quaternary amines, has been shown to be closely related to the development of atherosclerosis. Previous studies have shown that Enterobacter aerogenes ZDY01 significantly reduces the serum levels of TMAO and cecal trimethylamine (TMA) in Balb/c mice; however, its role in the inhibition of choline-induced atherosclerosis in ApoE-/- mice remains unclear. Here, we demonstrated that E. aerogenes ZDY01 inhibited choline-induced atherosclerosis in ApoE-/- mice fed with 1.3% choline by reducing cecal TMA and modulating CDCA-FXR/FGF15 axis. We observed that E. aerogenes ZDY01 decreased the cecal TMA and serum TMAO levels by utilizing cecal TMA as a nutrient, not by changing the expression of hepatic FMO3 and the composition of gut microbiota. Furthermore, E. aerogenes ZDY01 enhanced the expression of bile acid transporters and reduced the cecal CDCA levels, thereby attenuating the FXR/FGF15 pathway, upregulating the expression of Cyp7a1, promoting reverse cholesterol transport. Taken together, E. aerogenes ZDY01 attenuated choline-induced atherosclerosis in ApoE-/- mice by decreasing cecal TMA and promoting reverse cholesterol transport, implying that E. aerogenes ZDY01 treatment might have therapeutic potential in atherosclerosis.

OBJECTIVE: Polycystic ovary syndrome (PCOS) is a highly heritable disease. Emerging evidence elucidated the elevated prevalence of reproductive abnormalities in first-degree relatives (FDRs) of patients with PCOS. METHODS: Ten databases were searched in December 2020 (PubMed, Embase, Web of Science, Cochrane Central Register of Controlled Trials, ClinicalTrials.gov, CBM, CNKI, VIP and WanFang and WHO international clinical trials registry platform). This study included cohort, case-control, or cross-sectional studies. The Preferred Reporting Items for Systematic Reviews and Meta-Analysis statement was followed. Dichotomous data from each of the eligible studies were combined by Mantel-Haenszel model. Standard mean differences with 95% confidence intervals were assessed. Heterogeneities were assessed using I  2 statistics, and the quality of evidence was evaluated by AHRQ EPC program and GRADE approach. RESULTS: Thirty-eight studies were included. The prevalence of PCOS (0.22; 95%CI 0.16 to 0.29), menstrual irregularities (0.28; 95%CI 0.22 to 0.34, P<0.01) and ovary morphological changes were elevated in female PCOS FDRs. Female FDRs also presented with increased levels of luteinizing hormone, total testosterone (SMD, 0.53; 95%CI 0.28 to 0.78, P<0.01), unconjugated testosterone, free androgen index, dehydroepiandrosterone sulfate (DHEAS), and anti-Mullerian hormone levels. Subgroup analyses indicated that some of these changes begun in pubertal girls. Furthermore, fathers of PCOS had higher risk of premature baldness. The DHEAS level was elevated in male FDRs. CONCLUSIONS: The findings of this analysis suggested that FDRs of patients with PCOS suffered from reproductive endocrinological dysregulations. Thus, more attention should be focused on this population. (PROSPERO-CRD42020183243).

Metal-organic frameworks (MOFs) act as versatile coordinators for the subsequent synthesis of high-performance catalysts by providing dispersed metal-ion distribution, initial coordination condition, dopant atom ratios, and so on. In this work, a crystalline MOF trans-[Cu(NO3)2(Him)4] was synthesized as the novel precursor of a redox-alternating CuxO electrochemical catalyst. Through simple temperature modulation, the gradual transformation toward a highly active nanocomposite was characterized to ascertain the signal enhancing mechanism in H2O2 reduction. Owing to the proprietary structure of the transducer material and its ensuing high activity, a proof-of-principle sensor was able to provide an amplified sensitivity of 2330 µA mM-1 cm-2. The facile one-pot preparation and intrinsic nonenzymatic nature also suggests its wide potentials in medical settings.

RATIONALE: Daporinad is a novel and potent inhibitor of nicotinamide phosphoribosyl transferase with potential antineoplastic and antiangiogenic activities. We aimed to explore the metabolites of daporinad generated from liver microsomes and to propose metabolic pathways. METHODS: The metabolites were generated by individually incubating daporinad (10 µM) with liver microsomes at 37°C for 60 min. The metabolites were identified by ultra-high-performance liquid chromatography/quadrupole-orbitrap mass spectrometry (UPLC/Q-Orbitrap-MS) using electrospray ionization in positive ion mode. They were deduced by accurate MS and MS/MS data. RESULTS: In total, 16 metabolites were found and their identities were characterized. In rat, dog and human, they were minor; in monkey, M11 was the most abundant. Daporinad was metabolized mainly through N-dealkylation, amide hydrolysis, hydrogenation, oxygenation and dehydrogenation. There was no human-specific metabolite. CONCLUSIONS: The current study provided an overview of the metabolism of daporinad, which is helpful in predicting in vivo metabolites and in selecting animal species for toxicology studies.

Studying the relationship between brain-derived neurotrophic factor (BDNF) and post-stroke depression (PSD) may help determine the potential for depression in stroke patients at the earliest stage possible. Current research has identified changes in BDNF levels in PSD patients. Thus, this article was intended as a review of evidence with respect to changes in the expression of BDNF in patients with PSD by integrating extant findings. We conducted a search in the electronic databases PubMed, EMBASE, and PsycINFO (all records from January 1, 2000, through October 20, 2020) using keywords: "brain-derived neurotrophic factor OR BDNF," "post-stroke depression OR PSD," "expression level," "association," and "relationship." Returned articles were considered for inclusion in this review if they were empirical studies investigating the association between BDNF expression and PSD. Seven original papers were selected for review and revealed inconsistent findings. Five out of seven studies reported a significant decrease in BDNF levels in PSD patients at a certain stage (most likely the early stage) of stroke after admission, whereas the other two showed contrasting findings. Overall, this review reveals associations between changes in serum BDNF levels and depression following stroke. Whether serum BDNF levels, especially in the early phase of stroke, can be a potentially effective biomarker for predicting the risk of subsequent PSD development is still open to debate.

PURPOSE: The predictors of trifecta achievement in partial nephrectomy (PN) were poorly inquired and remained a controversial area of discovery. To evaluate predictive factors of trifecta achievement in patients undergoing PN. MATERIALS AND METHODS: A systematic literature search was performed to identify relevant articles. Only studies focusing on postoperative trifecta achievement and exploring its predictor with multivariable analyses were included. The trifecta achievement was defined as negative surgical margins, warm ischemia time <25 minutes, and no complications. Merged odds ratio (OR) and 95% confidence interval (CI) were used to evaluate the predictive effect. RESULTS: Thirteen studies with 7066 patients meeting the inclusion criteria were included. The rate of trifecta achievement ranged from 43.3% to 78.6%. Merged results showed that preoperative eGFR (OR: 1.01, 95% CI: 1.00, 1.02, P=0.02), operative time (OR: 0.99, 95% CI: 0.99, 1.00, P=0.02), estimated blood loss (OR: 1.00, 95% CI: 1.00, 1.00, P <0.001), tumor size (OR: 0.70, 95% CI: 0.58, 0.84, P <0.001), medium (OR: 0.39, 95% CI: 0.18, 0.84, P=0.02) and high PADUA score (OR: 0.23, 95% CI: 0.08, 0.64, P=0.005) were independently associated with trifecta achievement. A publication bias was identified for tumor size. Sensitivity analysis confirmed the stability of result for tumor size. CONCLUSIONS: Larger tumor size, medium and high PADUA score are associated with decreased probability of trifecta achievement. After verifying by further high-quality studies, these variables can be incorporated into tools to predict probability of trifecta achievement during clinical practice.

Background: The effects of different testicular sperm extraction methods on the embryonic development and clinical outcome of azoospermic men in intracytoplasmic sperm injection (ICSI) cycles have not been researched. Our goal was to evaluate the effect of different sperm retrieval methods used for patients with OA or NOA on the embryonic development and clinical outcomes during the ICSI cycles. Methods: This was a retrospective cohort study. A total of 530 azoospermic patients who underwent 570 ICSI cycles met the study criteria. ICSI was performed using testicular sperm by TESA in 282 cycles (TESA group); ICSI with testicular sperm by mTESE was performed due to NOA in 90 cycles (mTESE group); ICSI with testicular sperm by MESA was performed in 198 cycles (MESA group). The embryonic development and clinical outcomes of the three groups were counted. Results: The general characteristics of the three groups were comparable. Our findings showed that the three groups were matched in terms of infertility durations and age. The mean age and the mean BMI of the female partners were similar in the three groups. Also, our findings showed there were no significant differences in the three groups regarding day 3 of the menstrual cycle FSH and days of stimulation. The research results showed that the total dose of FSH and E2 on the HCG administration day was also not statistically different in the three groups. The number of oocytes retrieved had no significant differences in the three groups. However, the number of 2PNs per cycle and the number of cleavages per cycle were higher in the MESA group than in the other two groups; the TESA group and mTESE group were similar. The number of good quality D3 embryos and the number of good quality D5 embryos were significantly decreased in the mTESE group as compared to the other two groups. Good quality D3 embryos and the rate of good quality D5 embryos in the mTESE group were lower than those in the other two groups. Moreover, the clinical pregnancy rates of the TESA group (50.71%) and the MESA group (51.52%) were similar, but both were much higher than that of the mTESE group (32.22%). Conclusions: The mTESE provides a good clinical outcome for NOA patients with severe spermatogenic impairment, including the rate of good quality D3 embryos, the rate of good quality D5 embryos, and the clinical pregnancy rate. However, our data suggested that both the TESA and MESA groups had better embryonic development and clinical outcomes than the mTESE group.

Purpose: Colon cancer (CC) is a serious disease burden. The prognosis of patients with CC is different, so looking for effective biomarkers to predict prognosis is vitally important. Ferroptosis is a promising therapeutic and diagnosis strategy in CC. However, the role of ferroptosis in prognosis of CC has not been studied. The aim of the study is to build a prognosis model related ferroptosis, and provide clues for further therapy of CC. Methods: The RNA-seq data were from TCGA (training group) and GEO (testing group). The R language and Perl language were used to process and analyze data. LASSO regression analysis was used to build the prognosis model. ssGSEA was used to compare the immune status between two groups. Immunohistochemistry was used to detect expression of AKR1C1 and CARS1 in colon cancer tissues and adjacent tissues. Results: The prognosis model consisted of five ferroptosis related genes (AKR1C1, ALOX12, FDFT1, ATP5MC3, and CARS1). The area under curve (AUC) at 1-, 2-, and 3-year were 0.668, 0.678, and 0.686, respectively. The high- and low-risk patients had significant survival probability and could be clearly distinguished by the PCA and t-SNE analysis. The multivariate cox regression analysis also showed the riskscore is an independent prognosis factor. Importantly, we found that the immune status between high- and low-risk patients were different obviously, such as CD8+T cells. And STING, a new promising immune target, was also correlated to our signature genes statistically significantly. Conclusion: Our ferroptosis prognosis signature could predict survival of CC patients to a certain degree. And the crosstalk between ferroptosis and immune, especially STING need further studies.

Enzyme mimetics have attracted wide interest due to their inherent enzyme-like activity and unique physicochemical properties, as well as promising applications in disease diagnosis, treatment and monitoring. Inspired by the attributes of nonheme iron enzymes, synthetic models were designed to mimic their capability and investigate the catalytic mechanisms. Herein, metal-organic gels (Fe-MOGs) with horseradish peroxidase (HRP) like Fe-NX structure were successfully synthesized though the coordination between iron and 1,10-phenanthroline-2,9-dicarboxylic acid (PDA) and exhibited excellent peroxidase-like activity. Its structure-activity relationship and the in-situ electrochemiluminescence (ECL) detection of H2O2 secreted by Hela cells were further investigated. The highly dispersed Fe-NX active sites inside Fe-MOGs were able to catalyze the decomposition of H2O2 into large amounts of reactive oxygen species (ROS) via a Fenton-like reaction under a low overpotential. Due to the accumulation of ROS free radicals, the luminol ECL emission was significantly amplified. A proof-of-concept biosensor was constructed with a detection limit as low as 2.2 nM and a wide linear range from 0.01 to 40 µM. As a novel metal organic gels based enzyme mimetic, Fe-MOGs show great promises in early cancer detection and pathological process monitoring.

ABSTRACT With the continuous improvement of people's material living standards, public health and national fitness has become a social topic, and has been widely concerned by the public, in terms of the traditional physical education and impact of the concept of health. This not only needs the school to carry on the multi-directional adjustment to the physical education teaching, but also needs the physical education teacher to respond positively to this in order to adapt to the development needs of the times and society. This study uses the analytic hierarchy process (AHP) to construct the professionalization model of physical education teachers in Colleges and Universities, and makes decision on the index weight of the model through fuzzy comprehensive evaluation. The results show that the weight of the first-class indicators of P.E. Teachers' professionalization model from large to small is ability > quality > knowledge, and the consistency ratio (CR) of the model indicators is less than 0.1, and the fuzzy score of the model is 85 points. Therefore, the model of P.E. Teachers' professionalization constructed in this study has consistent indicators and shows good applicability. This does not only provide the basis for the professional development evaluation of College PE teachers, but also provides research ideas for the development of the sports industry from the perspective of public health.


RESUMO Com a melhoria contínua dos padrões de vida materiais das pessoas, a saúde pública e a saúde nacional tornaram-se um tema social, e tem sido amplamente discutido pelo público, em termos de educação física tradicional e impacto do conceito de saúde, que depende não só da escola para continuar o ajuste multidirecional para o ensino de educação física, mas também precisa do professor de educação física para dar uma resposta positiva a isso, a fim de se adaptar às necessidades de desenvolvimento dos tempos e da sociedade. Este estudo utiliza o processo hierárquico analítico (AHP) para construir o modelo de profissionalização dos professores de educação física em faculdades e universidades, e toma uma decisão sobre o peso do índice do modelo através de uma avaliação abrangente. Os resultados mostram que o peso dos indicadores de primeira classe do modelo de profissionalização dos professores de grande para pequeno é a capacidade > qualidade > conhecimento, e a razão de consistência dos indicadores do modelo é menor que 0.1, e a pontuação difusa do modelo é 85 pontos. Assim, o modelo de profissionalização dos professores de educação física, construído neste estudo, tem boa consistência de indicadores e mostra boa aplicabilidade. Isto não só proporciona a base para a avaliação do desenvolvimento profissional dos professores universitários de educação física, mas também fornece ideias de investigação para o desenvolvimento da indústria desportiva sob a perspectiva da saúde pública.


RESUMEN Con la mejora continua del nivel de vida material de las personas, la salud pública y la buena condición física nacional se han convertido en un tema social, y han sido motivo de una amplia preocupación en la población, con respecto a la educación física tradicional y su impacto en el concepto de salud. Esto no solo necesita que la escuela lleve a cabo el ajuste multidireccional de la enseñanza de educación física, sino también que el docente de educación física responda positivamente para adaptarse a las necesidades de desarrollo de los tiempos y la sociedad. Este estudio utiliza el proceso de jerarquía analítica (PJA) para construir el modelo de profesionalización de los docentes de educación física en los colegios y universidades, y analiza el peso del índice del modelo a través de una evaluación integral difusa. Los resultados muestran el peso de los indicadores de primera clase del modelo de profesionalización de los docentes, de mayor a menor, es capacidad> calidad> conocimiento, y el índice de consistencia (IC) de los indicadores del modelo es menor a 0.1, y la puntuación difusa del modelo es de 85 puntos. Por tanto, el modelo de profesionalización docente construido en este estudio tiene indicadores consistentes y muestra buena aplicabilidad. Esto no solo proporciona la base para la evaluación del desarrollo profesional de los profesores de educación física universitaria, sino que también ofrece ideas de investigación para el desarrollo de la industria del deporte desde la perspectiva de la salud pública.

BACKGROUND: The objective of this study was to analyze the accuracy of gadolinium-ethoxybenzyl-diethylenetriamine penta-acetic acid enhanced magnetic resonance imaging (Gd-EOB-DTPA-MRI) for predicting microvascular invasion (MVI) in patients with small hepatocellular carcinoma (sHCC) preoperatively. METHODS: A total of 60 sHCC patients performed with preoperative Gd-EOB-DTPA-MRI in the Harbin Medical University Cancer Hospital from October 2018 to October 2019 were involved in the study. Univariate and multivariate analyses were performed by chi-square test and logistic regression analysis. The sensitivity, specificity, accuracy, positive predictive value, and negative predictive value of Gd-EOB-DTPA-MRI were performed by receiver operating characteristic (ROC) curves. RESULTS: Univariate analysis indicated that alanine aminotransferase (≥ 39.00U/L), poorly differentiated pathology, and imaging features including grim enhancement, capsule enhancement, arterial halo sign and hepatobiliary features (tumor highly uptake, halo sign, spicule sign and brush sign) were associated with the occurrence of MVI (p < 0.05). Multivariate analysis revealed that rim enhancement and hepatobiliary spicule sign were independent predictors of MVI (p < 0.05). The area under the ROC curve was 0.917 (95% confidence interval 0.838-0.996), and the sensitivity was 94.74%. CONCLUSIONS: The morphologies of hepatobiliary phase imaging, especially the spicule sign, showed high accuracy in diagnosing MVI of sHCC. Rim enhancement played a significant role in diagnosing MVI of sHCC.

Light-emitting diodes (LED) based on silicon platforms and CdTe quantum dots (QDs) are an important area of research. Due to poor charge balance in QD layers, reduced graphene oxide (RGO) was introduced between the P-Si substrate and CdTe QDs. To further improve the performance of the QDs-LED, Au nanorods (NRs) were incorporated into the hole transport layer. A finite-difference time-domain simulation was used to discuss theoretically the influence of RGO and Au NRs on the LED performance, and investigate the influence of substrate on the localized surface plasmon resonance (LSPR) coupling wavelength. Simulated results demonstrated that the LSPR peaks for Au NRs red shifted with increase in substrate refractive index, and the electric field along the x-y plane was mostly concentrated near the interface. RGO incorporation enhanced the emission of the QDs-LED. This may be attributed to improvement in conductivity and hole mobility in the hole transport layer and reduction in the energy barrier for injection of holes. After introduction of Au NRs, there was a significant enhancement in emission, and light extraction efficiency was enhanced about 10-fold.

RATIONALE: The ß2-adrenoceptor (ß2-AR), a prototypical GPCR (G protein-coupled receptor), couples to both Gs and Gi proteins. Stimulation of the ß2-AR is beneficial to humans and animals with heart failure presumably because it activates the downstream Gi-PI3K-Akt cell survival pathway. Cardiac ß2-AR signaling can be regulated by crosstalk or heterodimerization with other GPCRs, but the physiological and pathophysiological significance of this type of regulation has not been sufficiently demonstrated. OBJECTIVE: Here, we aim to investigate the potential cardioprotective effect of ß2-adrenergic stimulation with a subtype-selective agonist, (R,R')-4-methoxy-1-naphthylfenoterol (MNF), and to decipher the underlying mechanism with a particular emphasis on the role of heterodimerization of ß2-ARs with another GPCR, 5-hydroxytryptamine receptors 2B (5-HT2BRs). METHODS AND RESULTS: Using pharmacological, genetic and biophysical protein-protein interaction approaches, we studied the cardioprotective effect of the ß2-agonist, MNF, and explored the underlying mechanism in both in vivo in mice and cultured rodent cardiomyocytes insulted with doxorubicin, hydrogen peroxide (H2O2) or ischemia/reperfusion. In doxorubicin (Dox)-treated mice, MNF reduced mortality and body weight loss, while improving cardiac function and cardiomyocyte viability. MNF also alleviated myocardial ischemia/reperfusion injury. In cultured rodent cardiomyocytes, MNF inhibited DNA damage and cell death caused by Dox, H2O2 or hypoxia/reoxygenation. Mechanistically, we found that MNF or another ß2-agonist zinterol markedly promoted heterodimerization of ß2-ARs with 5-HT2BRs. Upregulation of the heterodimerized 5-HT2BRs and ß2-ARs enhanced ß2-AR-stimulated Gi-Akt signaling and cardioprotection while knockdown or pharmacological inhibition of the 5-HT2BR attenuated ß2-AR-stimulated Gi signaling and cardioprotection. CONCLUSIONS: These data demonstrate that the ß2-AR-stimulated cardioprotective Gi signaling depends on the heterodimerization of ß2-ARs and 5-HT2BRs.

In traditional coreactant electrochemiluminescence (ECL), the efficiency of the coreactant catalyzed into an active intermediate is one of the dominant factors restricting the luminous intensity. In this work, Co-2-MI-ZnTCPP is designed as a composite material integrating coreaction accelerator (Co-N) and luminophore. Through the catalytic effect of Co-N structures on hydrogen peroxide, the in situ generation and accumulation of active intermediates are achieved, which will react with porphyrin anion radical, thereby bringing out self-enhanced ECL. By adjusting the scanning potential range, the ECL mechanism is thoroughly studied and the contribution of each potential window to the luminescence is obtained. This work provides inspiration for the design of integrated ECL emitters with a coreaction accelerator and luminophore, providing a new way for the construction of a self-enhanced ECL emitter.
