RESUMO

In this work, a symmetric micro-beam was developed to eliminate horizontal drift of the tip during indent and scratch processes, which was inevitable in the usage of a traditionally one-end-fixed cantilever. Using the finite element simulation, a database involving the maximum stress and spring constant depending on the size parameters (length and inertial moment) of the symmetric beam was established to help in optimizing the beam design. A real symmetric micro-beam was fabricated based on the database, and the loading accuracy was checked through the calibration of the spring constant. The reliability of the symmetric beam was experimentally verified through the fabrication of the micro-channel and micro-dot, showing that the horizontal drift was completely suppressed. This technology can potentially be applied in micro/nanoscale research and manufacturing to increase the accuracy of detection and processing, such as the applications in probe-based lithography to improve the loading accuracy at high load condition and enhance the stability of the beam system at high sliding speed.

The heart is a rare primary site of lymphoma, and cardiac involvement is thought to bring a poorer prognosis. A framework of known clinical presentations, diagnostic features, disease complications, treatments, and outcomes to improve prognostication was constructed by a systematic review in 2011. However, some aspects must be discussed further in light of recent advances in lymphoma research. We collected cardiac lymphoma case reports published from January 2009 to January 2019, collected statistics from each patient, and performed a systematic analysis. The epidemiological characteristics, clinical manifestations, treatments, responses, and survival of primary cardiac lymphoma (PCL) patients are described. We obtained 158 cases of heart lymphoma, of which 101 were defined as PCL. There were more male than female cases. Most cases were diffuse large B-cell lymphoma. Six cases of PCL in cardiac myxomas were described. Patients with arrhythmia had shorter progression-free survival compared those without (HR 0.334, 95% CI 0.112-0.999, log-rank P = 0.042). Surgery did not improve patients' long-term prognosis or reduce the risk of death within 1 month. These data suggest that central nervous system prophylaxis is necessary. The overall survival was longer than that in data from 1949 to 2009.

Melanoma, the most aggressive human skin tumor, has a very short survival time, and there are currently no effective treatments. Alterations in cell metabolism, such as enhanced aerobic glycolysis, have been identified as hallmarks of cancer cells. In the present study, bioinformatics studies using online databases revealed that FOXO3a expression was lower in melanoma tissues compared with normal tissues and nevus. Additionally, Kaplan­Meier analysis showed that high expression of FOXO3a predicted an improved prognosis for patients with melanoma. Furthermore, Pearson correlation analysis indicated that the expression of FOXO3a was positively correlated with SIRT6 expression and negatively correlated with the expression levels of a number of glycolysis­associated genes. Chromatin immunoprecipitation and luciferase assays showed that FOXO3a was enriched in the SIRT6 promoter region and promoted its transcription. Then, SIRT6 was overexpressed in FOXO3a­knockdown MV3 cells and downregulated in FOXO3a­overexpressing MV3 cells by using lentivirus­mediated stable infection. The results showed that SIRT6 knockdown or overexpression rescued the effects of FOXO3a overexpression or knockdown, respectively, on glycolysis, as determined by glucose uptake, glucose consumption and lactate production assays, the expression of glycolytic genes and glucose stress flux tests. SIRT6 overexpression also suppressed FOXO3a knockdown­induced tumor growth in a mouse model. The present findings indicated that the FOXO3a­SIRT6 regulatory axis inhibited glucose metabolism and tumor cell proliferation in melanoma, and provided novel insight into potential therapeutic strategies to treat this disease.

Promoting tourism in China using sustainable practices has become a very important issue. In order to analyze temporal characteristics and spatial regularities of green total factor productivity (GTFP), carbon emissions and the consumption of energy related to tourism in China were estimated using a "bottom-up" method. The construction of a measurement framework (including carbon emissions and energy consumption) of GTFP for the tourism industry was also undertaken. The data envelopment analysis (DEA) model and the Malmquist-Luenberger (ML) index were used to measure and calculate tourism GTFP in China between 2007 and 2018, as well as analyze spatio-temporal differences. Results indicate that: (1) carbon emissions and the consumption of energy are increasing, and they have not yet peaked, with traffic associated with tourism accounting for the largest proportion among tourism sectors; the spatial distribution of carbon emissions and the consumption of energy is not balanced; (2) green development of tourism in China has achieved a good level of performance during the study period, driven by technical efficiency. Since 2014, pure technical efficiency (PE) has been >1, indicating that the tourism industry in China has entered a stage of change and promotion; (3) significant spatial differences exist in tourism GTFP in China. For example, the overall pattern of being strongest in the east and weakest in the west has not changed. Currently, eastern, central, and western regions in China rely on different dynamic mechanisms to promote tourism green development. In addition, some provinces have become the core or secondary growth poles of tourism green development in China.

BACKGROUND: This study is aimed at exploring the correlation of sirtuin 2 (SIRT2) with clinical characteristics as well as overall survival (OS) in breast cancer patients. METHODS: Totally, 296 primary breast cancer patients who underwent surgical resection were retrospectively reviewed in this study, and SIRT2 expression in tumor and adjacent tissues was determined by immunohistochemistry (IHC) and scored by semiquantitative scoring (0-12). Clinicopathological features were retrieved, and OS was calculated. RESULTS: Both SIRT2 IHC semiquantitative score and percentage of SIRT2 high expression by IHC score > 3 were lower in tumor tissues compared with adjacent tissues. Additionally, tumor SIRT2 high expression was associated with lower T stage, decreased N stage, and reduced TNM stage. Kaplan-Meier curves displayed that tumor SIRT2 high expression predicted longer OS. Univariate Cox's regression analysis showed that tumor SIRT2 high expression was associated with prolonged OS, while multivariate Cox's regression analysis displayed that tumor SIRT2 high expression was not an independent predictive factor for OS, which implied that tumor SIRT2 might predict OS indirectly through the interaction of tumor features (such as TNM stage) in breast cancer patients. CONCLUSION: SIRT2 expression is lower in tumor tissues compared with adjacent tissues, and tumor SIRT2 high expression correlates with lower T stage, decreased N stage, reduced TNM stage, and longer OS in breast cancer patients.

Antibiotic resistance genes (ARGs), as emerging environmental contaminants, are becoming a threat to human health. In this study, the combined processes of powdered activated carbon (PAC)/biological PAC (BPAC)-ultrafiltration (UF) were adopted to reduce the levels of ARGs in secondary effluents from a wastewater treatment plant. The removal of dissolved organic carbon (DOC) and the change of normalized flux in the UF process were investigated. In addition, the structural characteristics of the microorganisms of the BPAC were analyzed. The results showed that the appropriate dosage of PAC and BPAC was 40 mg/L. At this dosage, PAC/BPAC-UF combined processes could effectively remove the ARGs in secondary effluents by 1.26-2.69-log and 1.55-2.97-log, respectively; and the removal rates of DOC would be 60.7% and 54.1%, respectively. Relative to the direct UF, the membrane fluxes of the two combined processes were increased by 15.6% and 25.1%, respectively. Significant removal correlations were found between ARGs, intI1, DOC and 16SrDNA. These results revealed that the PAC/BPAC-UF combined process might play a promising role in ARG reduction in secondary effluents from wastewater treatment plants.

The increasing demand for environmental protection has given rise to burgeoning research on green consumption. The present research adds to this expanding literature by investigating a novel predictor of consumer green consumption: awe. As a self-transcendent emotion, awe arises when people encounter perceptually vast stimuli that overwhelm their existing knowledge and mental structures, and, meanwhile, this also elicits a need for accommodation. This research proposes and demonstrates that, compared with happiness and a neutral affective state, experience of awe promotes green consumption via an enhanced psychological ownership of nature. Moreover, this research identifies a condition by showing that, while awe promotes green consumption when interdependent self-construal is activated, this effect diminishes when independent self-construal is activated.

Mucormycosis is an angioinvasive fungal infection with a high mortality rate. Patients with hematological malignancies following voriconazole therapy are at high risk from mucormycosis. Here, the present study reports on a 68-year-old man diagnosed with multiple myeloma and secondary myelodysplastic syndrome, who was infected with disseminated mucormycosis with cerebellum involvement confirmed by mycological culture and histopathological examination. For patients with hematological malignancies who are receiving antifungal therapy, an opportunistic infection of mucormycosis should be considered if a 'breakthrough' infection occurs in the predilection sites (such as the sinuses, lungs, skin, brain and gastrointestinal tract). It is difficult to diagnose mucormycosis because of the limited reliable detection methods, and because mucormycosis often presents with an acute onset and progresses rapidly, particularly in immunocompromised patients. Antifungal therapy with amphotericin B or posaconazole should be started as soon as possible considering the empirical diagnosis.

Multiphoton excitation allows one to access high energy excited states and perform valuable tasks in biological systems using tissue penetrating near-infrared (NIR) light. Here, we describe new photoactive manganese tricarbonyl complexes incorporating the ligand 4'-p-N,N-bis(2-hydroxyethyl)amino-benzyl-2,2':6',2″-terpyridine (TPYOH), which can serve as an antenna for two photon NIR excitation. Solutions of Mn(CO)3(TPYOH)X (X = Br- or CF3SO3-) complexes are very photoactive toward CO release under visible light excitation (405 nm, 451 nm). The same responses were also triggered by multiphoton excitation at 750 and 800 nm. In this context, we discuss the potential applications of these complexes as visible/NIR light photoactivated carbon monoxide releasing moieties (photoCORMs). We also report the isolation and crystal structures of the TPYOH complexes Mn(TPYOH)Cl2 and [Mn(TPYOH)2](CF3SO3)2, to illustrate a possible photolysis product(s).

Ruthenium-catalyzed heteroannulation between enamides and isocyanates has been realized as a complementary approach to conventional strategies for the synthesis of pyrimidin-4-ones. High step- and atom-economy was achieved for the rapid construction of such privileged scaffolds, which are found in a multitude of pharmaceutical compounds. The generality and practicability of this transformation were reflected by the broad scope of substrates with diverse functional groups, large-scale synthesis, and late-stage diversification.

Chemiluminescence (CL) and bioluminescence (BL) imaging technologies, which require no external light source so as to avoid the photobleaching, background interference and autoluminescence, have become powerful tools in biochemical analysis and biomedical science with the development of advanced imaging equipment. CL imaging technology has been widely applied to high-throughput detection of a variety of analytes because of its high sensitivity, high efficiency and high signal-to-noise ratio (SNR). Using luciferase and fluorescent proteins as reporters, various BL imaging systems have been developed innovatively for real-time monitoring of diverse molecules in vivo based on the reaction between luciferin and the substrate. Meanwhile, the kinetics of protein interactions even in deep tissues has been studied by BL imaging. In this review, we summarize in vitro and in vivo applications of CL and BL imaging for biosensing and therapy. We first focus on in vitro CL imaging from the view of improving the sensitivity. Then, in vivo CL applications in cells and tissues based on different CL systems are demonstrated. Subsequently, the recent in vitro and in vivo applications of BL imaging are summarized. Finally, we provide the insight into the development trends and future perspectives of CL and BL imaging technologies.

The objectives of present study were to evaluate the effect of casein kinase 1 (CK1) inhibition D4476 on in vitro maturation (IVM) and developmental competence of bovine oocytes. The cumulus oocyte complexes (COCs) were cultured in maturation medium with D4476 (0, 2, 5, 10, 20 µM) for 24 hr. After IVM and in vitro fertilization, through expansion average scores of cumulus cells (CCs), oocyte maturation efficiency, cleavage rate and blastocyst rate of zygote, we found 5 µM D4476 could increase the development potential of oocytes. After the COCs were treated with 5 µM D4476, the results of quantitative real-time PCR analysis, Lichen red staining and PI staining showed that under without affecting germinal vesicle breakdown and nuclear morphology, D4476 could significantly decrease CK1 and upregulate TCF-4 in oocytes. Furthermore, without influencing the level of Bad and CTSB, D4476 could significantly increase the expression of ß-catenin, TCF-4, Cx43, MAPK, PTGS-2, PTX-3, TGS-6, Bax and Bcl-2 in CCs. Western blot analysis revealed that the addition of 5 µM D4476 during the maturation of COCs resulted in a lower level of Cx43 protein at 12 hr and a higher expression of Cx43 protein at 24 hr compared to the group without D4476. These results indicate that adding optimum D4476 (5 µM) to maturation medium is beneficial to maturity efficiency and development competence of bovine oocytes.

Here, DNA-functionalized nanoscale PCN-224 is established with the aptamer of A549 lung cancer cells. The aptamer was modified with carboxyl and fluorescein at the two terminals. When touching the A549 cells, DOX@PCN-224-DNA can trace tumor cells and present good targeting therapy by the combination of chemotherapy and photodynamic therapy. This facile aptamer functionalization of PCN-224 offers an opportunity to develop metal-organic framework-based target-directed therapy and biosensors.

The friction of hydrogenated diamond-like carbon (H-DLC) films was evaluated under the controlled environments of humid air and vacuum by varying the applied load. In humid air, there is a threshold applied load below which no obvious friction drop occurs and above which the friction decreases to a relatively low level following the running-in process. By contrast, superlubricity can be realized at low applied loads but easily fails at high applied loads under vacuum conditions. Further analysis indicates that the graphitization of the sliding H-DLC surface has a negligible contribution to the sharp drop of friction during the running-in process under both humid air and vacuum conditions. The low friction in humid air and the superlow friction in vacuum are mainly attributed to the formation and stability of the transfer layer on the counterface, which depend on the load and surrounding environment. These results can help us understand the low-friction mechanism of H-DLC film and define optimized working conditions in practical applications, in which the transfer layer can be maintained for a long time under low applied load conditions in vacuum, whereas a high load can benefit the formation of the transfer layer in humid air.

Mechanochemical wear has attracted much attention due to its critical role in micro/nanodevice applications, reliable microscopy, and ultraprecision manufacturing. As a process of stress-associated chemical reactions, mechanochemical wear strongly depends on temperature; however, the impact mechanism is not fully understood at any length scale. Here, we reported different water-temperature dependence of mechanochemical wear on two typical single crystal silicon (Si) surfaces, involving oxide-covered Si partially terminated with Si-OH groups and oxide-free Si fully terminated with Si-H groups. As the water temperature increased from 10 to 80 °C, the mechanochemical wear of the oxide-covered Si underwent a process from no obvious surface damage to significant material removal but that occurring at all temperatures decreased gradually on the oxide-free Si surface. The opposite temperature-dependence was found to have a strong relation to the growth or degeneration of the Si-OH surfacial groups. The mechanochemical wear on the both Si surfaces decreased with the Si-OH coverage rising, which facilitated the growth of strongly hydrogen-bonded ordered water and then suppressed the chemical reaction between the sliding interfaces. These results can provide new insight into the mechanism of the surrounding temperature affecting the reliable micro/nanodevices, manufacturing, and microscopy.

MicroRNAs (miRNAs) are pivotal regulators of the progression of carcinogenesis and negatively regulate the expression of tumour­associated genes. Downregulation of miR­124 expression has been demonstrated in various human cancer tissues, wherein miR­124 serves as a tumour suppressor by targeting oncogenes. However, its function and underlying mechanism of action remain unclear in breast cancer. In the present study, the tissue­specific expression of miR­124 was detected in 10 paired triple­negative breast cancer and normal tissues, and its inhibitory effects on cell growth and invasion were evaluated in vitro and in vivo. Bioinformatics analysis identified signal transducer and activator of transcription 3 (STAT3), a well­known oncogene in breast cancer, as the potential target. Upregulation of miR­124 expression decreased STAT3 mRNA and protein levels in breast cancer cells and the relative luciferase activity. Rescue experiments revealed that the transfection of a STAT3 expression plasmid reversed the inhibitory effect of miR­124 on the proliferation and invasion of MDA­MB­468 cells. These data demonstrate that miR­124 serves vital roles in the suppression of triple­negative breast cancer via inhibition of cell proliferation and invasion through STAT3. These results highlight the potential role of miR­124 as a diagnostic or therapeutic target in patients with breast cancer.

Lipid peroxidation plays a crucial role in early brain injury (EBI) after subarachnoid hemorrhage (SAH), and cystine/glutamate antiporter system Xc- has been proved to be associated with glutathione (GSH) synthesis, which protects cells against oxidative damage. Antioxidant effect of system Xc- is mediated by Beclin 1 (BECN1). Therefore, this study aimed to determine whether administration of BECN1 small interfering RNA (siRNA) could attenuate EBI after SAH experiment, specifically through suppressing lipid peroxidation and increasing system Xc- activity. Endovascular perforation was performed to induce SAH in a rat model and BECN1 siRNA was administered through intracerebroventricular injection. Neurological score, brain edema, lipid peroxidation (malondialdehyde, MDA), and antioxidation system, containing GSH, glutathione peroxidase (GSH-Px), glutathione reductase (GR), and anti-reactive oxygen species (anti-ROS), were examined. The expression of BECN1 and light chain of system Xc- (xCT) was detected by western blot, immunoprecipitation, and immunofluorescence staining. This study confirmed that SAH induced neurological deficits and brain edema, which was accompanied by the increase of BECN1 expression and lipid peroxidation, and the decrease of xCT expression and antioxidative capacity. However, downregulation of BECN1 by siRNA could decrease the formation of the BECN1-xCT complex and lipid peroxidation, enhance antioxidative capacity, and ameliorate neurological deficits and brain edema in SAH rats. The results suggested that inhibition of BECN1 suppresses accumulation of lipid peroxidation by increasing system Xc- activity in EBI after SAH, and BECN1 may be a new effective target for EBI treatment after SAH.

An efficient cobalt-catalyzed ring-opening reaction of bench-stable 1,2-oxazetidines with heteroarenes was unprecedentedly developed. The sustainable Cp *Co(III) catalyst enables a distinctive merger of C-H activation with concomitant N-O and C-C cleavages of 1,2-oxazetidine, leading to site-selective C-H aminomethylation and hydroxymethylation of heteroaromatic compounds containing a broad range of functional groups. Preliminary control experiments unravel some essential mechanistic features of this one-pot transformation.

OBJECTIVE: The incidence of breast cancer in females is gradually increasing. Expression of HER-2 gene and protein is critical for predicting the prognosis of breast cancer. This study examined amplification of HER-2 gene and protein expression in breast cancer patients to analyze their correlation with clinical and pathological features. MATERIALS AND METHODS: Specimens of breast gland tissues were collected from breast cancer patients for measuring HER-2 protein expression by immunohistochemistry (IHC) method. Fluorescent in situ hybridization (FISH) measured HER-2 gene amplification. The consistency of HER-2 protein and gene expression was analyzed in addition to their correlation with clinical and pathological features of patients. RESULTS: Thirty-six percent patients showed negative expression of HER-2 protein, and 9%, 32%, and 23% of them had positive expression to different levels (+, ++, and +++). Forty percent patients were positive for HER-2 gene amplification, including 3, 21, and 14 cases of (+), (++), and (+++) patients. Expression of HER-2 protein was highly correlated with HER-2 gene amplification (r = 1.262; p < 0.05). Both parameters were correlated with tumor size, differentiation grade, lymph node metastasis, and TNM stage (p < 0.05). CONCLUSIONS: Combined assay of IHC and FISH for detecting HER-2 protein or gene amplification in breast cancer tissues showed their correlation with tumor size, differentiation grade, lymph node metastasis, and TNM stage.
