Fe(III)-oxyhydroxide composites are efficient adsorption materials, however, their detailed geometrical structures and adsorption mechanisms in a broad pH range are still unclear. In this study, the structures of arsenic(V) adsorbed on Fe(III)-oxyhydroxide adsorption complexes were investigated using hybrid density functional theory calculations together with experimental Fourier transform infrared (FT-IR) spectroscopy. Adsorption experiments showed higher arsenic(V) removal rates in the range of pH 5.0-10.0. FT-IR and X-ray diffraction (XRD) revealed that arsenic(V) is stably adsorbed by Fe(III)-oxyhydroxide complexes. The formation of monomeric As(V) appears to be favored in the 1.0-4.0 pH range, while dimeric and trimeric As(V)-Fe(III) oxyhydroxide complexes are preferably formed at higher pH (4.0-10.0). Namely, dimeric and trimeric Fe(III)-oxyhydroxides exhibited good removal efficiencies for arsenic(V) over a wide pH range. Interestingly, the stabilities of the complexes formed between As(V) and monomeric, dimeric and trimeric Fe(III)-oxyhydroxides follow the orders: As(V) trimer > As(V) dimer > As(V) monomer; these complexes are also interconvertible.

Osteosarcoma is the most frequent primary malignant bone tumor, composed of mesenchymal cells producing osteoid and immature bone. The sensitive detection of telomerase plays a pivotal role in the early diagnosis and therapeutic treatment of osteosarcoma. We report here an in vitro strategy for sensitive telomerase activity detection through the integration of rolling circle amplification (RCA) and a clustered regularly spaced short palindrome repeats (CRISPR)-Cas12a system. In the proposed strategy, telomerase substrate (TS) primers are easily controlled to extend five bases (GGGTT) to give short telomerase extension products (TEP) with definite lengths without adding dATP. The resulting short TEPs can then cyclize the padlock through hybridizing with its two terminals and thus initiate the following RCA. To obtain an improved sensitivity, the CRISPR-Cas12a system is attached to collaterally cut surrounding DNA reporter probes after recognizing the target single strand DNA sequence in the RCA products. The highlights of this strategy are as follows: (i) the short TEP triggered strategy is excellent at detecting low telomerase activity and thus contributes to the early diagnosis of malignant tumors; (ii) highly sensitive telomerase activity detection which is easy to operate from RCA initiated CRISPR-Cas12a; (iii) opening up of a new avenue for telomerase activity detection with a CRISPR-Cas12a system. Finally, the proposed strategy exhibited sensitive telomerase activity detection under optimized experimental parameters and has great application potential for the clinical diagnosis of malignant tumors and the development of anti-cancer drugs.

Lung injury caused by cardiopulmonary bypass (CPB) increases the mortality after cardiac surgery. Previous studies have shown that regulatory T cells (Tregs) play a protective role during CPB, but the correlation between Tregs and CPB-induced lung injury remains unclear. Here, we conducted a prospective study about Treg cells in patient receiving CPB. Treg cells were collected from patients before the CPB operation (pre-CPB Tregs), and the effect of pre-CPB Tregs on the occurrence of CPB-induced lung injury was evaluated. Data showed that the baseline level of Treg cells in peripheral blood were lower in patients who developed lung injury after CPB, compared to those who did not develop lung injury after CPB. Function analyses revealed that pre-CPB Tregs from CPB-induced lung injury patients presented decreased ability in suppressing the proliferation and IFN-γ production of CD4 and CD8 T cell. Also, pre-surgery levels of TGF- ß and IL-10 were markedly lower in lung injury patients than in non-lung injury patients. In addition, PD-1 and Tim-3 expression on pre-CPB Tregs were significantly lower in CPB-induced lung injury patients than the CPB patients without lung injury. Above all, we found impaired peripheral Treg responses in CPB-induced lung injury patients, indicating a potential role of Treg cells in the early diagnosis of CPB-induced lung injury.

The accumulation of some harmful elements in plants from intensive production systems pose a serious threat to human health. In this study, seven heavy metals(Cu, Pb, Zn, Cr, Cd, As, and Hg) and their distribution characteristics in the crops, vegetables, and fruits were analyzed alongside single factor evaluation and Nemero index analysis. Combined with dietary recommended consumption data from the Chinese Nutrition Society, the dietary exposure of heavy metals were further analyzed, and a consequent safety risk assessment was conducted. A total of 673 crop, vegetable, and fruit samples were collected from typical intensive production systems in Hainan Province. The results showed that the content of Cu, As, and Hg in the 673 plant samples was below the value of the national food standard. The exceed rates of Pb, Zn, Cr, and Cd were 2.67%, 3.71%, 2.53%, and 3.71%, respectively. The heavy metal comprehensive pollution degree of six species of plants showed the trend of leafy vegetables > tuber crops > non-leafy vegetables > legume crops > fruits > cereals. In particular, Cr in leafy vegetables showed significantly higher hazard quotients(HQ) than that in other types, and exceeded 1, suggesting a high potential health risk via the ingestion of heavy metals through leafy vegetables. The relatively lower hazard index(HI) values of tuber crops, non-leafy vegetables, legume crops, fruits, and cereals suggest that these types of plants are more suitable for intensive production systems.

BACKGROUND: During the process of deep decay, when decay approaches the pulp, an immune response is triggered inside the pulp, which activates the complement cascade. The effect of complement component 5a (C5a) on the differentiation of dental pulp mesenchymal stem cells (DPSCs) is related to dentin reparation. The aim of the present study was to stimulate DPSCs with different concentrations of C5a and evaluate the differentiation of odontoblasts using dentin sialoprotein (DSP). METHODS: DPSCs were divided into the following six groups: (i) Control; (ii) DPSCs treated with 50 ng/ml C5a; (iii) DPSCs treated with 100 ng/ml C5a; (iv) DPSCs treated with 200 ng/ml C5a; (v) DPSCs treated with 300 ng/ml C5a; and (vi) DPSCs treated with 400 ng/ml C5a. Flow cytometry and multilineage differentiation potential were used to identify DPSCs. Mineralization induction, Real-time PCR and Western blot were conducted to evaluate the differentiation of odontoblast in the 6 groups. RESULT: DPSCs can express mesenchymal stem cell markers, including CD105, CD90, CD73 and, a less common marker, mesenchymal stromal cell antigen-1. In addition, DPSCs can differentiate into adipocytes, neurocytes, chondrocytes and odontoblasts. All six groups formed mineralized nodules after 28 days of culture. Reverse transcription-quantitative PCR and western blotting indicated that the high concentration C5a groups expressed higher DSP levels and promoted DPSC differentiation, whereas the low concentration C5a groups displayed an inhibitory effect. CONCLUSION: In this study, the increasing concentration of C5a, which accompanies the immune process in the dental pulp, has demonstrated an enhancing effect on odontoblast differentiation at higher C5a concentrations in vitro.

BACKGROUND: The fall armyworm (FAW), Spodoptera frugiperda (J. E. Smith), is an invasive pest. Maize is one of its preferred host plants. The traditional application of granules has been effective for the control of FAW. Nevertheless, the challenge is to develop an application method that is less labor-intensive and also efficient. RESULTS: Tiny granules in the size range of 0.38-0.55 mm were prepared by the centrifugal granulation method for drone application. The in-swath distribution patterns of granules deposited on sticky tapes were similar to the distribution of aqueous droplets through drone spraying. In the range of 1.5-3.5 m, a relatively higher aircraft height of flight can provide a more uniform distribution pattern of deposited granules, but resulted in less deposit of tiny granules in the whorl of maize plants. Similarly, the deposit of granules in the whorl of maize plants significantly decreased when the flight speed increased from 4 to 6 m s-1 . Granules gathered in the whorl accounted for 50.8-58.7% of total granules deposited in the maize canopy. The field experiment demonstrates granular formulations (containing 0.25% chlorantraniliprole + 0.15% emamectin benzoate, or higher concentrations) can provide better and longer duration of FAW control than the aqueous spray formulation at the whorl stage of maize plants. CONCLUSION: Broadcasting of tiny granules by drone can provide better FAW control than liquid spray. The architecture of the maize plant and the downward airstream of the drone create favorable conditions for the deposition of granules in whorls.

BACKGROUND: The detection of interleukin 33 (IL-33) in pleural effusion may be more sensitive in diagnosing tuberculous pleural effusion (TPE). The present study aimed to assess the accuracy of pleural IL-33 for the diagnosis of TPE by means of meta-analysis and systematic review of relevant studies. METHOD: After retrieving the published studies, the sensitivity, specificity, positive likelihood ratio, negative likelihood ratio, diagnostic odds ratio, and a summary receiver operating characteristic curve were assessed to estimate the usefulness of pleural IL-33 in diagnosing TPE using meta-analysis with a random-effects model. We also performed meta-regression and subgroup analysis. RESULTS: A total of 639 patients from 6 studies were analyzed. The pooled sensitivity, specificity, positive likelihood ratio, negative likelihood ratio, and diagnostic odds ratio were 0.87 (95% confidence interval [CI], 0.82-0.91), 0.76 (95% CI, 0.72-0.80), 6.54 (95% CI, 2.65-16.15), 0.17 (95% CI, 0.10-1.27), and 45.40 (95% CI, 12.83-160.70) respectively. The area under the curve was 0.94. The composition of the included population was the main cause of heterogeneity and subgroup analysis showed that pleural IL-33 had a higher specificity (0.93, 95% CI 0.87-0.96) when used for differential diagnosis between TPE and malignant pleural effusion. CONCLUSION: The detection of IL-33 alone in pleural effusion seems to not be an efficient diagnostic marker for TPE but may serve as a novel biomarker to differentiate between TPE and malignant pleural effusion.

Aim: Coronavirus disease 2019 (COVID-19) is a form of disease triggered by a new strain of coronavirus. This paper proposes a novel model termed "deep fractional max pooling neural network (DFMPNN)" to diagnose COVID-19 more efficiently. Methods: This 12-layer DFMPNN replaces max pooling (MP) and average pooling (AP) in ordinary neural networks with the help of a novel pooling method called "fractional max-pooling" (FMP). In addition, multiple-way data augmentation (DA) is employed to reduce overfitting. Model averaging (MA) is used to reduce randomness. Results: We ran our algorithm on a four-category dataset that contained COVID-19, community-acquired pneumonia, secondary pulmonary tuberculosis (SPT), and healthy control (HC). The 10 runs on the test set show that the micro-averaged F1 (MAF) score of our DFMPNN is 95.88%. Discussions: This proposed DFMPNN is superior to 10 state-of-the-art models. Besides, FMP outperforms traditional MP, AP, and L2-norm pooling (L2P).

Enhancing photocatalytic performance via electronic modulation have attracted much attention for synergetic photocatalytic degradation of antibiotic pollutant. In this study, a new hetero-structured system is raised, which comprises In2S3 coupled to In-MOF and operates as an efficient photocatalyst for RhB degradation. The formation of hetero-structure and occurred electron modulation of In2S3/In-MOF hybrid was confirmed by relevant characterizations. Surprisingly, the In2S3/In-MOF hybrid represented enhanced photocatalytic ability over In-MOF. The photocatalysis of Rhodamine B in presence of In2S3/In-MOF hybrid has achieved 92.2 % degradation.

Insulin resistance (IR) is one of the most common features of polycystic ovary syndrome (PCOS), which is related to obesity. Whether increased anti-Müllerian hormone (AMH) levels in PCOS are involved in the pathogenesis of insulin resistance remains unclear. We investigated serum levels of leptin and AMH along with basic clinical and metabolic parameters in 114 PCOS patients and 181 non-PCOS women. PCOS patients presented higher fasting blood glucose, insulin concentrations and Homeostatic Model Assessment of Insulin Resistance (HOMA-IR) in addition to body mass index (BMI), lipids profiles and hormone levels. HOMA-IR showed a positive correlation with BMI, AMH, leptin, and low-density lipoprotein-cholesterol (LDL-c) levels. Interestingly, AMH is strongly positively correlated with HOMA-IR and insulin concentrations for 1st and 2nd hours of glucose treatment after fasting. Among PCOS women with BMI≥25 kg/m2, high AMH level group showed an increased HOMA-IR when compared to normal AMH level. However, among PCOS women with normal BMI, women with high AMH presented an elevated fasting insulin levels but not HOMA-IR when compared to normal AMH group. In vitro treatment of isolated islet cells with high concentration of leptin (200 ng/ml) or high leptin plus high concentration of AMH (1 ng/ml) significantly enhanced insulin secretion. Importantly, co-treatment of AMH plus leptin upregulates the expression of pro-apoptotic proteins, such as Bax, caspase-3, and caspase-8 after incubating with a high level of glucose. These results suggest that AMH may involve in the pathological process of pancreatic ß-cells in obese PCOS women.

INTRODUCTION: This study aims to determine whether high glucose condition and dynamic O-linked N-acetylglucosamine (O-GlcNAc) modification can promote the proliferation and migration of human retinal microvascular endothelial cells (HRMECs) and whether Runt-related transcription factor 1 (RUNX1) could mediate the glucose and O-GlcNAc-driven proliferation and migration of HRMECs. RESEARCH DESIGN AND METHODS: Western blot analysis was used to detect the O-GlcNAc modification level and RUNX1 level in cells and retina tissues, cell growth was studied by cell counting kit-8 assay, cell proliferation was detected by immunofluorescence staining. Then, cell migration and tube formation were investigated by scratch-wound assay, Transwell assay, and tube-forming assay. The changes of retinal structure were detected by H&E staining. The O-GlcNAc modification of RUNX1 was detected by immunoprecipitation. RESULTS: High glucose increases pan-cellular O-GlcNAc modification and the proliferation and migration of HRMECs. Hence, O-GlcNAc modification is critical for the proliferation and migration of HRMECs. RUNX1 mediates the glucose and O-GlcNAc-driven proliferation and migration in HRMECs. RUNX1 can be modified by O-GlcNAc, and that the modification is enhanced in a high glucose environment. CONCLUSIONS: The present study reveals that high glucose condition directly affects retinal endothelial cells (EC) function, and O-GlcNAc modification is critical for the proliferation and migration of HRMECs, RUNX1 may take part in this mechanism, and maybe the function of RUNX1 is related to its O-GlcNAc modification level, which provides a new perspective for studying the mechanism of RUNX1 in diabetic retinopathy.

Particle manipulation in microfluidic devices is of great significance in biological research. However, currently available inertial capture methods require relatively high flow rates, which will cause damage to biological particles, especially for single-celled organisms that are sensitive to environments. Herein, we demonstrate a label-free, size-based, low shear stress manipulation method using the Viscoelastic Stagnant Region (VSR) to capture sensitive bioparticles. This method uses the deformation of molecular chains in the polymer solution which can generate elastic stresses to form vortices which is called VSR because of the extremely low velocity in the contraction-expansion array (CEA) microchannel. Formation and evolution of VSR was observed experimentally using the Micro-PIV system in polyethylene oxide (PEO) solutions with different concentrations. On this basis, 20 µm and 5 µm polystyrene (PS) particles were confined to a certain area in the microchamber and the trajectory of particles motion in VSR was observed. Both the inertial lift force and the viscoelastic force are affected by the particle size, so the method also presented size selectivity. By quantitatively studying the velocity distribution of the particles on the orbit of motion, it was found that the maximum velocity of the particles in the VSR was about 0.02 m s-1, which is only about 1/100 of that in inertial vortices. Moreover, the spiral motion of particles captured by VSR with variable trajectories was first observed, which is different from the equilibrium orbit in inertial vortices and can provide more motion paths for the particles. This method was further applied to the research of Crypthecodinium cohnii (C. cohnii), which is vulnerable but has high nutritious value. We found that the activity of the C. cohnii decreased slightly after being processed by VSR, but was lost after being processed by the inertial vortices. This research, as a simple and low shear stress particle manipulation method, will provide useful guidance for the manipulation, capture and separation of sensitive biological cells under higher biocompatibility, which is of great significance for the study of cellular and molecular biology.

In vertebrates, 5'-Hoxd genes (Hoxd9), which are expressed in the hindlimb bud mesenchyme, participate in limb growth and patterning in early embryonic development. In the present study, We investigated the mechanisms by which ATRA regulates cultured E12.5 rat embryo hindlimb bud mesenchymal cells (rEHBMCs). Following exposure to ATRA over 24 h, mRNA and protein expression levels of HoxD9 were evaluated by reverse transcription-polymerase chain reaction (RT-PCR), quantitative real-time PCR (qPCR), and western blotting. Flow cytometry was used to detect apoptosis. ATRA inhibited the condensation and proliferation, and promoted the apoptosis rate of the rEHBMCs in a dose-dependent manner. Sox9 and Col2a1 in rEHBMCs were downregulated by ATRA in a dose-dependent manner at both mRNA and protein levels. Similarly, HoxD9 was downregulated by ATRA in a dose-dependent manner, in parallel with the cartilage-specific molecules Sox9 and Col2a1. Both qPCR and western blotting showed that both Shh and Gli3 were downregulated. Overexpression of HoxD9 reversed the effects of ATRA. These results demonstrate that ATRA suppresses chondrogenesis in rEHBMCs by inhibiting the expression of HoxD9 and its downstream protein targets, including Sox9 and Col2a1. This effect may also be correlated with inhibition of the Shh-Gli3 signaling pathway.

This work synthesized a novel CuS/KTa0.75Nb0.25O3 (KTN) heterojunction composite and firstly applied it in photocatalytic and piezocatalytic reduction of N2 to NH3. XRD, Raman, XPS, SEM, and TEM analyses indicate that CuS nanoparticles closely adhered to the surface of KTN nanorods, which facilitates the migration of electrons between the two semiconductors. Mott-Schottky and valence band XPS analysis shows that KNbO3 shows a higher conduction band than CuS, indicating that CuS mainly acts as electron trappers to capture the photogenerated electrons from KTN. Because of the great enhanced spatial separation of photogenerated charge carriers, the CuS/KTN presents much higher performance than pure KNT, which is further confirmed by 1H NMR analysis of the reaction solution. An interesting finding is that synthesized CuS/KTN not only performs well under light irradiation but also can work in an ultrasonic bath, indicating its great potential in photo/piezocatalytic conversion of N2 to NH3. The optimal 10 %CuS/KTN shows an NH3 production rate of 36.2 µmol L-1 g-1 h-1 under ultrasonic vibration, which reaches 7.4 times that of KTN. The electrons generated by KTN through the piezoelectric effect can be captured by CuS, which endows the electrons a longer life to participate in the reaction, thereby improving the catalytic reaction performance.

Semi-supervised learning has always been a hot topic in machine learning. It uses a large number of unlabeled data to improve the performance of the model. This paper combines the co-training strategy and random forest to propose a novel semi-supervised regression algorithm: semi-supervised random forest regression model based on co-training and grouping with information entropy (E-CoGRF), and applies it to the evaluation of depression symptoms severity. The algorithm inherits the ensemble characteristics of random forest, and combines well with co-training. In order to balance the accuracy and diversity of co-training random forests, the algorithm proposes a grouping strategy to decision trees. Moreover, the information entropy is used to measure the confidence, which avoids unnecessary repeated training and improves the efficiency of the model. In the practical application of evaluation of depression symptoms severity, we collect cognitive behavioral data of emotional conflict based on the depressive affective disorder. And on this basis, feature construction and normalization preprocessing are carried out. Finally, the test is conducted on 35 labeled and 80 unlabeled depression patients. The result shows that the proposed algorithm obtains MAE (Mean Absolute Error) = 3.63 and RMSE (Root Mean Squared Error) = 4.50, which is better than other semi-supervised regression algorithms. The proposed method effectively solves the modeling difficulties caused by insufficient labeled samples, and has important reference value for the diagnosis of depression symptoms severity.

The precision and reliability of the synchronous prediction of multi energy consumption indicators such as electricity and coal consumption are important for the production optimization of industrial processes (e.g., in the cement industry) due to the deficiency of the coupling relationship of the two indicators while forecasting separately. However, the time lags, coupling, and uncertainties of production variables lead to the difficulty of multi-indicator synchronous prediction. In this paper, a data driven forecast approach combining moving window and multi-channel convolutional neural networks (MWMC-CNN) was proposed to predict electricity and coal consumption synchronously, in which the moving window was designed to extract the time-varying delay feature of the time series data to overcome its impact on energy consumption prediction, and the multi-channel structure was designed to reduce the impact of the redundant parameters between weakly correlated variables of energy prediction. The experimental results implemented by the actual raw data of the cement plant demonstrate that the proposed MWMC-CNN structure has a better performance than without the combination structure of the moving window multi-channel with convolutional neural network.

OBJECTIVE: To systematically review the safety and effectiveness of tension-free vaginal tape (TVT), transobturator tape (TOT), and transvaginal tension-free vaginal tape-obturator (TVT-O) in the treatment of stress urinary incontinence (SUI). METHODS: The PubMed, Embase, The Cochrane Library, Web of Science, WanFang Data, and CNKI were electronically searched. A total of 22 trials involving 3709 patients were included. RESULTS: The objective cure rate was better with TVT than with TOT and TVT-O. The incidence of postoperative thigh/groin pain was lower with TVT than with TOT and TVT-O, but the incidence of dysuria was higher with TVT than with TVT-O. The incidence of lower urinary tract infection was higher with TVT than with TOT. CONCLUSION: Tension-free vaginal tape may have a better outcome compared to TOT and TVT-O, but the risk of dysuria and lower urinary tract infection is higher with SUI.

BACKGROUND: Ceratocystis fimbriata is a hazardous fungal pathogen able to cause black rot disease on sweet potato. The management of C. fimbriata strongly relies on the use of toxic fungicides, and there is a lack of efficient alternative strategies. RESULTS: The antifungal properties of quinolinic acid (QA) were studied for the first time, indicating that QA shows selective antifungal activity against C. fimbriata. QA inhibited completely the mycelial growth of C. fimbriata at less than 0.8 mg mL-1 concentration (pH 4), and was able to produce alterations in the fungal cell wall, and to impede spore agglutination and mycelium formation. QA significantly reduced the concentration of ergosterol, and was able to associate to iron (II), suggesting that QA may be a lanosterol 14-α demethylase inhibitor. In preventive applications, QA reduced the disease incidence of C. fimbriata on sweet potato by 75%, achieving higher control efficacy in comparison with commercial fungicides prochloraz and carbendazim. CONCLUSIONS: The first selective antifungal agent against C. fimbriata was discovered in this work, and showed suitable antifungal properties for the management of black rot disease. © 2021 Society of Chemical Industry.
