RESUMO

Rationale: Clinical application of doxorubicin (DOX) is limited by its toxic cardiovascular side effects. Our previous study found that toll-like receptor (TLR) 5 deficiency attenuated cardiac fibrosis in mice. However, the role of TLR5 in DOX-induced cardiotoxicity remains unclear. Methods: To further investigate this, TLR5-deficient mice were subjected to a single intraperitoneal injection of DOX to mimic an acute model. Results: Here, we reported that TLR5 expression was markedly increased in response to DOX injection. Moreover, TLR5 deficiency exerted potent protective effects against DOX-related cardiac injury, whereas activation of TLR5 by flagellin exacerbated DOX injection-induced cardiotoxicity. Mechanistically, the effects of TLR5 were largely attributed to direct interaction with spleen tyrosine kinase to activate NADPH oxidase (NOX) 2, increasing the production of superoxide and subsequent activation of p38. The toxic effects of TLR5 activation in DOX-related acute cardiac injury were abolished by NOX2 deficiency in mice. Our further study showed that neutralizing antibody-mediated TLR5 depletion also attenuated DOX-induced acute cardiotoxicity. Conclusion: These findings suggest that TLR5 deficiency attenuates DOX-induced cardiotoxicity in mice, and targeting TLR5 may provide feasible therapies for DOX-induced acute cardiotoxicity.

Paeonia ostii is now being extensively planted for oil extraction in China, which is recognized as a single oil-use tree peony cultivar and commonly called 'Fengdan'. This study investigated the effects of nitrogen fertilizer on oil yield, fatty acid compositions and antioxidant activity of P . ostii . Oil yield (33.46%), oleic acid (25.12%), linoleic acid (29.21%) and a-linolenic acid (43.12%) reached the maximum at N450 treatment, with significant differences compared with other treatments ( P <0.05). Furthermore, strong antioxidant activity with low DPPH IC50 value (19.43±1.91 µg mL -1 ) and large ABTS value (1216.53±30.21 µmoL Trolox g -1 ) and FRAP value (473.57±9.11 µmoL Trolox g -1 ) was also observed at N450. Palmitic acid (5.57%) and stearic acid (2.02%) reached a maximum at N375, but not significant with N450 ( P< 0.05). Nitrogen fertilizer could promote oil yield, fatty acid accumulation and antioxidant activity, and N450 (450kg ha -1 ) is recommended as the optimum application for P. ostii .

Meteorin-like (METRNL) protein is a newly identified myokine that functions to modulate energy expenditure and inflammation in adipose tissue. Herein, we aim to investigate the potential role and molecular basis of METRNL in doxorubicin (DOX)-induced cardiotoxicity. METRNL was found to be abundantly expressed in cardiac muscle under physiological conditions that was decreased upon DOX exposure. Cardiac-specific overexpression of METRNL by adeno-associated virus serotype 9 markedly improved oxidative stress, apoptosis, cardiac dysfunction and survival status in DOX-treated mice. Conversely, knocking down endogenous METRNL by an intramyocardial injection of adenovirus exacerbated DOX-induced cardiotoxicity and death. Meanwhile, METRNL overexpression attenuated, while METRNL silence promoted oxidative damage and apoptosis in DOX-treated H9C2 cells. Systemic METRNL depletion by a neutralizing antibody aggravated DOX-related cardiac injury and dysfunction in vivo, which were notably alleviated by METRNL overexpression within the cardiomyocytes. Besides, we detected robust METRNL secretion from isolated rodent hearts and cardiomyocytes, but to a less extent in those with DOX treatment. And the beneficial effects of METRNL in H9C2 cells disappeared after the incubation with a METRNL neutralizing antibody. Mechanistically, METRNL activated SIRT1 via the cAMP/PKA pathway, and its antioxidant and antiapoptotic capacities were blocked by SIRT1 deficiency. More importantly, METRNL did not affect the tumor-killing action of DOX in 4T1 breast cancer cells and tumor-bearing mice. Collectively, cardiac-derived METRNL activates SIRT1 via cAMP/PKA signaling axis in an autocrine manner, which ultimately improves DOX-elicited oxidative stress, apoptosis and cardiac dysfunction. Targeting METRNL may provide a novel therapeutic strategy for the prevention of DOX-associated cardiotoxicity.

OBJECTIVE: To compare the efficacy between acupoint selection of meridian diagnosis and regular acupoint selection for chronic atrophic gastritis (CAG). METHODS: A total of 70 cases of CAG were randomly divided into an observation group (35 cases, 5 cases dropped off) and a control group (35 cases, 5 dropped off). In the observation group, according to the hand diagnosis of meridians and the results of 80-channels energy determinator, based on the principle of child-mother relation acupoint combination, the luo-connecting point and back-shu points were added for excess syndrome, and the yuan-primary point, front-mu points and he-sea point of foot meridians were added for deficiency syndrome; in addition, the acupoints of the eight extraordinary meridians were added based on the nature of acupoints. In the control group, Zhongwan (CV 12), Neiguan (PC 6), Zusanli (ST 36) and Gongsun (SP 4) were selected as the primary acupoints, and additional acupoints were added according to syndrome differentiation. The two groups were treated twice a week (Tuesday and Thursday, respectively), totally for 6 months. Six months after treatment, the follow-up was conducted. The clinical symptom score, gastroenteropathy patient reported outcomes (PRO) scale score before treatment, after treatment and during follow-up as well as the score of pathological changes of gastric mucosa before and after treatment were compared between the two groups. RESULTS: After treatment and during follow-up, the clinical symptom scores and gastroenteropathy PRO scale scores were decreased in the two groups (P<0.01, P<0.001); at the follow-up, the gastroenteropathy PRO scale score in the observation group was lower than that in the control group (P<0.01). After treatment, the scores of pathological changes of gastric mucosa in the two groups were decreased (P<0.01), and the score in the observation group was lower than that in the control group (P<0.05). CONCLUSION: The acupoint selection of meridian diagnosis is superior to regular acupoint selection for CAG, which has better efficacy, more significant improvement on gastric mucosa pathology, and more stable long-term effect.

BACKGROUND: Literature has demonstrated that diabetes is associated with renal complication and testicular dysfunctions. The current study explored the potential of Tiliacora triandra extract and its major component against diabetic kidney and testicular damages in rats. METHODS: Diabetes was induced by high fat diet/streptozotocin (HFD/STZ) and treated orally with Tiliacora triandra extract (TTE, 100 and 400 mg kg-1 body weight) and its major component, 5,7-dihydroxy-6-oxoheptadecanoic acid (DHA, 25 mg kg-1 body weight) for 30 consecutive days. Testicular activities of testicular enzymes, serum levels of testosterone, luteinizing hormone (LH) and follicle-stimulating hormone (FSH), sperm parameters and urinalysis for protein and albumin levels were evaluated. Renal and testicular biomarkers of oxidative stress and pro-inflammation were analysed along with histology. RESULTS: The experimental diabetes induced significant alterations in the levels and activities of indices evaluated compared to non-diabetic normal rats. The 28-day treatment of diabetic rats with TTE and DHA markedly improved activities of testicular enzymes, restored levels of testosterone, LH and FSH and sperm parameters compared to untreated diabetic rats. TTE and DHA abrogated proteinuria and reversed urine albumin level. Testicular and renal oxidative stress and pro-inflammation were attenuated in diabetic rats treated with TTE and DHA. The diabetes-mediated histopathological damage was alleviated in the kidney and testis. CONCLUSION: The protective effect of TTE and DHA against diabetes induced kidney and testicular damages may be related to its antioxidant and anti-inflammatory activities. © 2020 Society of Chemical Industry.

To further improve the pyrenyl-contained derivatives two-photon absorption (TPA) and third-order nonlinear optical (NLO) properties, three steps of optimization are employed based on experimental molecule PCVS-B: heteroatomic substitution, exchanging the position of double bonds and adding a branch. The contributions of π electrons to localized orbital locators and Mayer bond orders (LOL-π and IABπ) show that the second step can enhance the chemical interaction between pyrenyl and the branched-chain. Two visual methods of charge density difference (CDD) and transition density matrix (TDM) are combined to intuitively analyze the intramolecular charge transfer (ICT) process of one (two) photon absorption; results show that both following two steps can increase the degree of ICT on the conjugated plane of the pyrenyl. The sum over state (SOS) model was used to find out the dominant two-photon transition process. The difference between the dipole moments obtained by the McRae equation is applied to the three-state model, revealing the inherent law of the second static hyperpolarizability.

Biofilm causes considerable technical challenges in agricultural water distribution systems. Electrochemical treatment (ECT) is a potential technique for controlling biofilm in the systems. Given the limited information on how ECT performance changes of irrigation systems and microbial biofilm community shifts. In this study, the effect of anti-biofilm was assessed. Illumina Miseq high-throughput sequencing, combined with molecular ecological network analysis, were applied to detect the effects of ECT on attached biofilm microbial communities. We found that ECT effectively mitigated biofilm formation with the fixed-biofilm biomass reduced by 37.5 %-79.9 %. ECT significantly shifted the bacterial community structures in the biofilm, reduced the communities' diversity, and changed the dominant species. Molecular ecological network analysis showed that the complexity and size of bacterial networks were destabilized under ECT and decreased the interactions among bacterial species. The reconstruction in bacterial community and networks were responsible for the decline in extracellular polymer substances and biofilm biomass. However, chlorine-resistant bacteria were found increased after ECT, and higher relative abundance and low biofilm removal was identified in continuous ECT as compared with intermittent ECT. These results aimed to highlight the opportunity for biofouling mitigation by ECT for irrigation systems, and reveal the potential anti-biofilm microbial mechanisms of ECT.

BACKGROUND: It is well established that retrieved lymph node (RLN) counts were positively correlated with better overall survival in gastric cancer (GC). But little is known about the relationship between RLN count and short-term complications after radical surgery. METHODS: A total of 1487 consecutive GC patients between January 2016 and December 2018 at Nanjing Drum Tower Hospital were retrospectively analyzed. Univariate analyses were performed to elucidate the association between RLN count and postoperative complications. We further identified clinical factors that might affect the RLN count. RESULTS: Among all of the patients, postoperative complications occurred in 435 (29.3%) patients. The mean RLN count was 25.1, and 864 (58.1%) patients were diagnosed with lymph node metastasis. Univariate analyses showed no significant difference between RLN count and postoperative complications (both overall and stratified by CDC grade). Univariate and multivariate analyses further revealed that type of resection, tumor invasion, and lymph node metastasis were associated with RLN count. CONCLUSIONS: The current study demonstrated that RLN count was not associated with postoperative short-term complications following gastrectomy of GC, which provided a rationale for the determination of a proper RLN count of curative gastrectomy.

We investigated if corilagin can ameliorate or reverse atherosclerotic development via the toll-like receptor 4 (TLR4) signaling pathway in vitro and in vivo. Ana-1 cells or mouse peritoneal macrophages (MPMs) were stimulated with oxidized low-density lipoprotein followed by corilagin treatment. TLR4 expression in Ana-1 cells was upregulated by lentiviral transduction and downregulated by small interfering RNA. Peripheral blood mononuclear cells (PBMCs), plasma samples, and femoral arteries were collected from rats exhibiting peripheral artery disease (PAD). mRNA and protein expression of TLR4 and downstream molecules were decreased significantly by corilagin treatment in Ana-1 cells, MPMs, and rat PBMCs, and the reduction remained irrespective of downregulation or upregulation of TLR4 expression in Ana-1 cells. Corilagin also exerted a prominent effect on changes in plasma levels of cytokines and the pathologic manifestation of atherosclerosis in femoral arteries. Corilagin could ameliorate the development of atherosclerotic plaques by inhibiting the TLR4 signaling pathway in monocyte/macrophages and reduce the release of proinflammatory cytokines. This study provides a new therapeutic target and new niche targeting drug to oppose atherosclerosis and reveals the enormous potential of corilagin for control of PAD in humans.

Introduction: Currently in China, many immune checkpoint inhibitors (ICIs) have been approved for the treatment of non-small cell lung cancer (NSCLC). Some patients can not benefit from ICIs, and approximately 50% of patients have immunotherapy-related toxicity. Therefore, it is necessary to monitor carefully the selection of immunotherapy population using biomarkers to maximize the benefit of patients with NSCLC. Methods: A prospective analysis was performed on patients with advanced NSCLC who were treated with ICIS at our hospital from March 2018 to June 2019, up to the follow-up deadline of December 31, 2019. The primary end points were overall survival (OS) and progression-free survival (PFS), and the secondary end points were objective response rate and disease control rate. A lasso regression was used for the univariate analysis, and Cox regression analysis was used for the multivariate analysis. An efficacy prediction line chart was developed. Results: A total of 63 patients were included in the study. The median PFS was 7.0 months (95% CI, 5.0-11.0) and did not reach the median OS. According to the lasso regression, significant univariate factors were smoking index, PD-ligand 1 expression, and neutrophil to lymphocyte ratio (NLR). According to the multivariate analysis, the Cox proportional hazards model showed that smoking index and NLR are independent predictors of PFS in immunotherapy. A model comprised of independent predictors was developed based on a multivariate logical analysis of the main cohort-non-small cell lung cancer immunotherapy prognosis score. This model is shown as a nomogram with a C-index of 0.801 (95% CI, 0.744, 0.858), which has high prediction accuracy. Conclusion: This predictive model, including NLR and smoking index, can achieve a 1-year PFS in immunotherapy of patients. PD-1 inhibitors have been demonstrated to be effective and safe in the clinical treatment of patients with NSCLC.

In this study, a simple and environmentally friendly method was developed for synthesizing 3D porous cellulose supported palladium nanoparticles (PdNPs) catalysts. PdNPs (3.1 ± 1.4 nm) can be synthesized directly by cellulose solution through microwave heating (60 °C, 4 h) and prepared into microspheres by freeze-drying to degrade methylene blue (MB) and Congo red (CR) rapidly. In addition, the reduction ability of cellulose hydroxyl group was proved by XPS and FTIR results. And the contact angle of 38.20° proves its hydrophilic ability. Cellulose-PdNPs microspheres showed high catalytic efficiency achieved 99 % and after multiple cycles it was still higher than 90 %. Therefore, the method proposed in this paper provides some new applications for cellulose, and the cellulose-Pd catalysts prepared by the research also have potential application value in the field of environment.

It is a critical period of fighting against new coronavirus(SARS-CoV-2) disease now,since its outbreak on December 2019 in Wuhan.Even though the front line staffs are thought heroes,the ENT doctors and nurses are also indispensable power in defending the disease.The number of outpatients of ENT is huge.The early stage of SARS-CoV-2 pneumonia(COVID-19) may present pharyngalgia or cough without fever.Thus,the ENT doctors have high risks of being consulted by early stage COVID-19 patients.This paper means to talk about the contributions of ENT doctors and nurses in defending against SARS-CoV-2 virus,as well as the mental status of them.

Reusing biogas slurry (BS) in agricultural drip irrigation systems may provide a solution to deal with the adverse environmental impacts of applying BS. Biofouling and scaling are two leading issues in drip irrigation emitters. This study investigated a practice that applied electromagnetic fields (EMFs) to control biofilms and scales. The bacterial communities and mineral precipitations in the clogging substances of emitters were determined. Results showed that EMFs inhibited the growth of microbes, and influenced BS physicochemical parameters. Consequently, EMFs shifted the bacterial communities with reduced diversities. Network analyses revealed that bacterial species under EMFs treatments showed lower average connectivities and simpler interactions, which were responsible for the decreases of extracellular polymers substances (EPS). Moreover, EMFs treatments not only reduced the carbonates in emitters, but also prevented the depositions of phosphates, silicates, and quartzes. EMFs also had impacts on the lattice parameters and crystal volumes of carbonates. In addition, the changes in bacterial communities and EPS contents were associated with the reductions of various minerals. Accordingly, EMFs effectively mitigated biofilms and scales with the fixed clogging substances reduced by 29.1-53.8 %. These findings demonstrated that applying EMFs is an effective anti-biofouling and anti-scaling treatment with potential applications in BS irrigation systems.

BACKGROUND: Immune checkpoint inhibitors (ICIs) have significant clinical efficacy in the treatment of non-small cell lung cancer (NSCLC); however, the incidence of immune-related adverse events (irAEs) of up to 50% has prevented their widespread use. With the increase in the use of ICIs alone or as combination therapy, clinicians are required to have a better understanding of irAEs and be able to manage them systematically. In this study, we aimed to assess the incidence of irAEs associated with ICIs. METHODS: We searched PubMed, Embase, and the Web of Science databases, and also included relevant literature references to widen our search. The relevant data with inclusion criteria were performed using RevMan 3.6.0 for meta-analysis. We undertook a systematic literature search which included published data up to December 2019. RESULTS: Overall, 147 articles and 23 761 cancer patients with 11 different ICI treatment-related (grade 1-5 and 3-5) irAEs were included in the study. There were 46 articles on pembrolizumab (6598 patients), 27 on nivolumab (3576 patients), 13 on atezolizumab (2787 patients), 12 on avelumab (3213 patients), 10 on durvalumab (1780 patients), 22 on ipilimumab (4067 patients), eight on tremelimumab (1158 patients), three on JS001 (223 patients), four on camrelizumab (SHR-1210) (178 patients), one on sintilimab (96 patients), and one on cemiplimab (85 patients). Grade 1-5 irAEs were: cytotoxic T lymphocyte antigen 4 (CTLA-4) (82.87%), programmed cell death 1 (PD-1) (71.89%), and programmed cell death ligand-1 (PD-L1) (58.95%). Subgroup analysis was: Avelumab (44.53%), durvalumab (66.63%), pembrolizumab (67.25%), atezolizumab (68.77%), nivolumab (76.25%), Ipilimumab (82.18%), and tremelimumab (86.78%). Grade 3-5 irAEs were: CTLA-4 (27.22%), PD-1(17.29%), and PD-L1(17.29%). Subgroup analysis was: Avelumab (5.86%), durvalumab (13.43%), atezolizumab (14.45%), nivolumab (15.72%), pembrolizumab (16.58%), tremelimumab (22.04%), and ipilimumab (28.27%). CONCLUSIONS: This meta-analysis confirmed that anti-PD-1 and anti-PD-L1 inhibitors had a lower incidence of irAEs compared with anti-CTLA-4 inhibitors.

OBJECTIVES: Long non-coding RNAs (lncRNAs) have been demonstrated as crucial regulators in cancer, but whether they are involved in the immune response of cancer cells remains largely undiscovered. GATA3-AS1 is a novel lncRNA that was upregulated in breast cancer (BC) according to online databases. However, its role in triple-negative breast cancer (TNBC) was elusive. METHODS: GATA3-AS1 expression in BC tissues and adjacent normal tissues was obtained from online databases. Loss-of-function assays were designed and conducted to verify the functional role of GATA3-AS1 in TNBC cells. Bioinformatic analysis and mechanism experiments were applied to explore the downstream molecular mechanism of GATA3-AS1. Similarly, the upstream mechanism which led to the upregulation of GATA3-AS1 in TNBC cells was also investigated. RESULTS: GATA3-AS1 was markedly overexpressed in TNBC tissues and cells. Knockdown of GATA3-AS1 suppressed TNBC cell growth and enhanced the resistance of TNBC cells to immune response. GATA3-AS1 induced the deubiquitination of PD-L1 through miR-676-3p/COPS5 axis. GATA3-AS1 destabilized GATA3 protein by promoting GATA3 ubiquitination. CONCLUSION: GATA3-AS1 contributed to TNBC progression and immune evasion through stabilizing PD-L1 protein and degrading GATA3 protein, offering a new target for the treatment of TNBC.

As a new woody oleaginous crop, tree peony is now being widely developed and utilized, which attributed to the outstanding oil-use features regarding the fatty acid profiles and bioactivity. The major fatty acid profiles and bioactivity of seed oils in ten tree peony varieties collected from a common garden were investigated in the present study. The results showed that the oil yields, fatty acid profiles, bioactivity of seed oils existed significant variations among ten tree peony varieties (P<0.05). Considered the application value, 'Fengdan' (FD) and 'Ziban' (ZB) was the optimal resources as the high oil yields, rich unsaturated fatty acids especially high α-linolenic acid of 40.46 %, great antioxidant activity with low IC50 values, high ABTS and FRAP values, and strong antimicrobial activity with high DIZ and low MIC/MBC values. The study also confirmed seed oil of tree peony as the potential raw materials sources in functional food, pharmaceuticals and cosmetics for human health.

Single-phonon modes offer potential applications in quantum phonon optics, but the phonon density of states of most materials consist of mixed contributions from coupled phonons. Here, using theoretical calculations and magneto-Raman measurements, we report two single-phonon vibration modes originating from the breathing and opposite out-of-plane vibrations of InSe layers. These single-phonon vibrations exhibit an anticorrelated scattering rotations of the polarization axis under an applied vertical magnetic field; such an anomalous magneto-optical behavior is due to the reverse bond polarizations of two quantum atomic vibrations, which induce different symmetry for the corresponding Raman selection rules. A 180° (+90° and -90°) integrated scattering rotation angle of two single-phonon modes was achieved when the magnetic field was swept from 0 to 6 T. This work demonstrates new ways to manipulate the magneto-optic effect through phonon polarity-based symmetry control and opens avenues for exploring single-phonon-vibration-based nanomechanical oscillators and magneto-phonon-coupled physics.

The resource centre involves 21 depository institutions in 15 regions of the country, including human parasite and vector depository, animal parasite depository, plant nematode characteristic specimen library, medical insect characteristic specimen library, trematode model specimen library, parasite-vector/snail model specimen library, etc. After nearly 15 years of operation, the resource centre has been built into a physical library with a database of 11 phyla, 23 classes, 1115 species and 117,814 pieces of parasitic germplasm resources, and three live collection bases of parasitic germplasm resources. A variety of new parasite-related immunological and molecular biological detection and identification technologies produced by the resource centre are widely used in the fields of public health responses, risk assessments on food safety, and animal or plant quarantine. The NPRC is the largest and top level resource centre on parasitology in China, and it is a leading technology platform for collecting and identifying parasitic resources.

Both vascular endothelial growth factor (VEGF) and matrix metallopeptidase-9 (MMP-9) are key biomarkers in tumor angiogenesis. Determination of the overexpression of the two biomarkers would provide valuable information on the progression of tumor growth and metastasis, but their simultaneous quantification by a single probe is unprecedented. Here, we develop a triplex DNA-based nanoprobe for simultaneously quantifying VEGF and MMP-9 using an α-hemolysin nanopore. A DNA aptamer is used as the triplex molecular beacon (tMB) loop to bind VEGF, and a stem-forming oligonucleotide modified with a short peptide is used to recognize MMP-9. The sequential presence of VEGF and MMP-9 could also be identified by different patterns of current events. Besides, the characteristic current events generated by the DNA probe possess pH-dependent patterns that can be used to reflect the environmental pH. Success in the construction of such DNA nanoprobes will greatly facilitate the investigation of the mechanisms of different tumor angiogenesis processes and provide a useful approach for cancer diagnosis.
