WHAT IS KNOWN AND OBJECTIVE: B vitamin therapy is a common treatment for diabetic pain and neuropathy, yet its use remains controversial in patients lacking B vitamin deficiencies. The aim of this review was to summarize the current evidence for the efficacy of B vitamin therapy in diabetic patients with neuropathy. COMMENT: We screened the English literature for clinical studies evaluating B vitamins as a therapy for pain and neuropathy in diabetic patients. We selected 43 relevant studies for qualitative analysis based on our selection criteria. Our survey of the literature revealed substantive heterogeneity with respect to efficacies of reported outcomes, as well as study design. Most beneficial outcomes were reported against baseline measures, with few positive comparisons against placebo. This highlights the need for larger, placebo-controlled studies. WHAT IS NEW AND CONCLUSION: B vitamins should be considered a plausible therapy for diabetic neuropathy, but its overall efficacy remains uncertain and requires further study.

Street dust is an important contributor to heavy metal exposure in street sweepers. In this work, the levels of cadmium (Cd), lead (Pb), and arsenic (As) in street dust were evaluated, and the corresponding health implications for street sweepers in the smelting district of Huludao city northeast China were assessed. The contributions of dietary sources and dust to total metal exposure in street sweepers were compared. Because street sweepers are exposed to street dust both during work and nonwork hours, the health risks faced by street sweepers are uncertain. Therefore, variable exposure duration was considered using a deterministic model. A probabilistic risk assessment model was developed to explore the health effects of street dust on street sweepers via Monte Carlo simulation. The various exposure parameters that affect risk were analyzed using sensitivity analysis. The average Cd, Pb, and As levels in the hair of street sweepers were 2.04, 20.12, and 0.52 mg·kg-1, respectively. These values were higher than those for residents (i.e., not street sweepers) of Huludao. Strong correlations were found between the logarithms (base 10) of the Cd, Pb, and As contents in dust and hair (rCd = 0.581, p < 0.01; rPb = 0.428, p < 0.01; rAs = 0.378, p < 0.01; n = 62). Based on analysis using deterministic models, the maximum exposures to Cd and Pb via dust through the alimentary canal were nearly three and six times higher than the dietary exposures, respectively. Sensitivity analysis indicated that exposure duration is an important parameter.

Oligomers, are, in general, unknown components of the polymer. These oligomers can migrate from the polymer into the food and become a non-intentionally added substance to the food. In this work, ion mobility time-of-flight mass spectrometry has been used to identify oligomers migrating from kitchenware. The structure elucidation of oligomers from polyamide 6 and polyamide 66 was achieved through the analysis of accurate m/z values of adducts and collision cross section values of precursor ions together with high-energy fragmentation patterns. Additionally, a method to extract oligomers from sunflower oil, cooked beans, soup and whole milk has been developed. Extraction recoveries ranged from 87 to 102% and limits of detection were from 0.03 to 0.11 mg/kg. It was observed that the migration from kitchenware to real food was below the specified migration limit of 5 mg/kg. However, this limit was exceeded for food simulants, which therefore overestimated the oligomer migration.

Ultra-thin two-dimensional nanosheets have attracted increasing attention due to their great application prospects in nanomaterial science and biomedicine. Herein, we report the preparation of exfoliated raw and oxidized 4-layer Ti7O13 (O-Ti7O13) and their ability to produce reactive oxygen species (ROS). The results show that O-Ti7O13 nanosheets can effectively produce ROS induced by X-ray irradiation. The 4-layer nanosheets can quickly load doxorubicin (DOX) within 5 min with a high loading rate to obtain a novel nanodrug system through their electrostatic adsorption capacity, and they exhibit a sustained release behavior. In this way, chemotherapy, radiation therapy and photodynamic therapy effectively combine for cancer synergistic treatment. We evaluated the cytotoxicity, cellular uptake and intracellular location of the O-Ti7O13 nanosheet-based drug delivery system in A549 lung cancer cells. Our results show that the O-Ti7O13/DOX complex is more cytotoxic to A549 cells than free DOX since a low concentration of loaded DOX (10 µg/mL) with a low dose of X-rays can cause the complete apoptosis of tumor cells. This work reveals that the therapeutic effect of DOX-loaded O-Ti7O13 nanosheets is strongly dependent on their loading mode, and the effects of chemotherapy and photodynamic therapy are enhanced under X-ray irradiation, which allows O-Ti7O13 nanosheet use as a photo-activated drug carrier. This work provides a new strategy for preparing 2D metal oxide nanosheets toward biomedical applications.

Herein, small Co nanoparticles (NPs) encapsulated in N-doped double-shelled carbon nanocages grafted with thin carbon nanotubes (Co@CNTs@DSCNCs) were synthesized from yolk-shell bimetallic zeolitic imidazolate framework (BMZIF). For HER electrocatalysis, they exhibit higher activity (η10 = 214 mV) and more favorable kinetics than Co@CNTs@PC (PC = porous carbon) with thick CNTs and large Co NPs derived from solid BMZIF cubes.

A new schiff base cobalt(III) complex [N,N'-bis(2'-hydroxyphenylacetone)-o-ethanediamine] cobalt(III) (M3) has been synthesized and characterized by single X-ray crystallography. The cytotoxicity of complex M3 was evaluated against HeLa, LoVo, A549, A549/cis cancer cell lines, and the normal cell lines LO2 by MTT assays. The IC50 is in the range of 6.27-22.68 µM, which is somewhat lower than cisplatin on the basis of platinum molar concentration. Furthermore, anticancer mechanistic studies showed that the complex M3 inhibited cell proliferation by blocking DNA synthesis and then acted on nuclear division of HeLa cells over time. Moreover, western blot analysis indicated M3 dramatically decreased the target protein c-Myc and KLF5 expression levels, and activated many signaling pathways including ER stress, apoptosis, cell cycle and DNA damage in HeLa. M3 did not affect proteasomal activity.

It is challenging to develop highly stable lanthanide luminescent sensors for detecting heavy metal ions and nitroaromatics in view of the human health and environmental security. To this end, two water stable Ln-MOFs with the chemical constitution of {[Ln(HL)]·3DMF·3H2O}n (Ln = Eu, LZG-Eu and Ln = Tb, LZG-Tb) have been developed solvothermally using a multidentate ligand (H4L) with the central phenyl backbone bisubstituted by 2,6-pyridine-dicarboxylic acid at the para-position, H4L = 1,4-bis(2',2'',6',6''-tetracarboxy-1,4':4,4''-pyridyl)benzene. Single crystal analysis demonstrates that two novel Ln-MOFs feature 4,4,4-connected nets with an unprecedented topology symbol of {42·6·83}2{42·62·82}{42·84} and contain two kinds of one-dimensional channels. Powder X-ray diffraction as well as the luminescence determination results indicate that they retain their crystallinity and structural integrity in harsh acidic and basic conditions with pH in the range of 4-11. Moreover, they are highly luminescent, which makes them excellent chemical sensors for detecting Cu2+ and 4-NP (4-nitrophenol) with high selectivity and sensitivity in aqueous media such as deionized water, tap water, and river water based on distinct quenching effects. To the best of our knowledge, their detection limits are lower than those documented so far. In addition, the quenching efficiency of 4-NP was retained in the presence of interfering ions even after the compounds were used for five cycles, which makes them attractive, reliable, visual, and recyclable luminescent Ln-MOF sensor materials for 4-NP. The recognition mechanism for Cu2+ could be attributed to the dissociation of the main framework induced by Cu2+ and the subsequent formation of a Cu2+ coordination species and that for 4-NP is considered to be multi-quenching mechanisms dominated by competition absorption.

Microplastics (MPs) are a class of emerging contaminants with diverse sizes. They influence the behavior of pollutants in the environment and cause harmful effects on organisms. To date, the size effects of MPs on the accumulation of organic pollutants by terrestrial invertebrates remain unclear. Here, we study the impacts and mechanisms of polystyrene MPs on the accumulation and elimination of phenanthrene in earthworms. Results showed that larger-size MPs (10 and 100 µm) facilitated the accumulation of phenanthrene by earthworms in the first week, whereas 100 nm MPs inhibited the elimination of phenanthrene in earthworms afterwards. Higher genotoxicity to earthworms was observed for co-exposure of micron-size MPs and phenanthrene, and 10 µm MPs were detected at the highest concentration and caused the most serious DNA damage to earthworm coelomocytes. Biomarkers and their mRNA gene expression levels suggested that larger-size MPs caused severer damage to earthworms, thus leading to increased accumulation of phenanthrene by earthworms at the beginning. Moreover, high-throughput 16S rRNA gene sequencing indicated that nano-size MPs significantly inhibited phenanthrene-degrading bacteria in earthworms, resulting in the highest residual concentration of phenanthrene. This study highlights the size effects of MPs and their impacts on the accumulation of organic pollutants by terrestrial organisms.

The MSWI fly ash (FA) is classified as hazardous waste and electrolytic manganese residue (EMR) as the harmful industrial waste. FA, water-washed FA (WFA), EMR and coal fly ash (CFA) were co-recycled to form lightweight MFCE ceramisites. The effects of FA, WFA and mixed MSWI fly ash on ceramisites were discussed. The approach to mixing FA and WFA increased the recycling amount of MSWI fly ash. The optimal mixture of 34.5% EMR, 24.1% CFA, 20.7% FA and 20.7% WFA sintered at 1160 °C for 12 min with a procedural heating rate (10 °C/min) and belonged to Class 800 artificial lightweight aggregate (GB/T 17431.1-2010); the quantity of MSWI fly ash in ceramisite was as high as 41.4%. Volatilization rates of Cd, Pb, Cu, Zn, Mn and Cr for ceramisite were higher 75.0, 24.2, 62.7, 133, 343 and 764% than FA respectively, attributed to the co-existence of chlorides and sulfates. The remained Zn, Cu, Pb, Mn and Cr were exchanged with Mg2+/Ca2+/Al3+ of diopside and wollastonite to form residual fractions. Our findings provided a feasibility method of co-recycling MSWI fly ash and electrolytic manganese residue to produce green lightweight aggregates.

Triple-negative breast cancer (TNBC) remains with highest incidence and mortality rates among females, and a critical bottleneck lies in rationally establishing potent therapeutics against TNBC. Here, the self-assembled micellar nanoarchitecture of heavy-atom-modulated supramolecules with efficient cytoplasmic translocation and tunable photoconversion is shown, for potent suppression against primary, metastatic, and recurrent TNBC. Multi-iodinated boron dipyrromethene micelles yield tunable photoconversion into singlet oxygen and a thermal effect, together with deep penetration and subsequent cytoplasmic translocation at the tumor. Tetra-iodinated boron dipyrromethene micelles (4-IBMs) particularly show a distinctly enhanced cooperativity of antitumor efficiency through considerable expressions of apoptotic proteins, potently suppressing subcutaneous, and orthotopic TNBC models, together with reduced oxygen dependence. Furthermore, 4-IBMs yield preferable anti-metastatic and anti-recurrent efficacies through the inhibition of metastasis-relevant proteins, distinct immunogenic cell death, and re-education of M2 macrophages into tumoricidal M1 phenotype as compared to chemotherapy and surgical resection. These results offer insights into the cooperativity of supramolecular nanoarchitectures for potent phototherapy against TNBC.

In many species, sexual differentiation is a vital prelude to reproduction, and disruption of this process can have severe fitness effects, including sterility. It is thus interesting that genetic systems governing sexual differentiation vary among-and even within-species. To understand these systems more, we investigated a rare example of a frog with three sex chromosomes: the Western clawed frog, Xenopus tropicalis. We demonstrate that natural populations from the western and eastern edges of Ghana have a young Y chromosome, and that a male-determining factor on this Y chromosome is in a very similar genomic location as a previously known female-determining factor on the W chromosome. Nucleotide polymorphism of expressed transcripts suggests genetic degeneration on the W chromosome, emergence of a new Y chromosome from an ancestral Z chromosome, and natural co-mingling of the W, Z, and Y chromosomes in the same population. Compared to the rest of the genome, a small sex-associated portion of the sex chromosomes has a 50-fold enrichment of transcripts with male-biased expression during early gonadal differentiation. Additionally, X. tropicalis has sex-differences in the rates and genomic locations of recombination events during gametogenesis that are similar to at least two other Xenopus species, which suggests that sex differences in recombination are genus-wide. These findings are consistent with theoretical expectations associated with recombination suppression on sex chromosomes, demonstrate that several characteristics of old and established sex chromosomes (e.g., nucleotide divergence, sex biased expression) can arise well before sex chromosomes become cytogenetically distinguished, and show how these characteristics can have lingering consequences that are carried forward through sex chromosome turnovers.

The benzylic C-H group of α,α-dicyanoolefins from 3-substituted 1-indanones could be significantly activated via transmission along the aromatic system, thus enabling dynamic kinetic resolution via a traditional reversible deprotonation-protonation process. Enantioenriched 9-substituted 9H-fluorene frameworks were finally constructed through an asymmetric vinylogous Michael addition to nitroolefins, followed by a cascade cyclization and oxidative aromatization process, under the catalysis of a chiral bifunctional thiourea-tertiary amine.

An important aspect of visual object recognition is the ability to perceive object shape. Two basic components of complex shapes are straight and curved contours. A large body of evidence suggests a modular hierarchy for shape representation progressing from simple and complex orientation in early areas V1 and V2, to increasingly complex stages of curvature representation in V4, TEO, and TE. Here, we reinforce and extend the concept of modular representation. Using intrinsic signal optical imaging in Macaque area V4, we find sub-millimeter sized modules for curvature representation that are organized from low to high curvatures as well as domains with complex curvature preference. We propose a possible 'curvature hypercolumn' within V4. In combination with previous studies, we suggest that the key emergent functions at each stage of cortical processing are represented in systematic, modular maps.

OBJECTIVE: Patients hesitate to consent to Electroconvulsive Therapy (ECT) because of the fear of memory impairment. The mechanisms underlying this impairment are unclear, but several observations suggest hippocampal alterations may be involved. We investigated whether ECT-induced change in hippocampal volume correlates with memory impairment. METHODS: Using a 3T MRI-scanner, we acquired brain images and assessed cognitive performance in 22 severely depressed patients at three time-points: (1) before ECT series, (2) within one week after the series, and (3) at six-month follow-up. The hippocampus was segmented into subregions using Freesurfer. The dentate gyri (DG) were the primary regions of interest (ROIs) and major hippocampal subregions secondary ROIs. Cognitive performance was assessed using the Screen for Cognitive Impairment in Psychiatry and verbal memory using the Verbal Learning subtest. The linear mixed model and the repeated measures correlation were used for statistical analyses. RESULTS: ECT induced an increase in the right and left DG volume with co-occurring worsening in verbal memory, and these changes were negatively correlated (right DG, rrm =-.85, df=18, p=.0000002; left DG, rrm =-.58, df=18, p=.008). At a six-month follow-up, the volume of both DG decreased with a co-occurring improvement in verbal memory, and these changes were negatively correlated in the right DG (rrm =-.64, df=15, p=.005). Volume increases in 14 secondary ROIs were also negatively correlated with memory impairment. CONCLUSION: ECT-related transient increases in the volume of major hippocampal subregions are associated with memory impairment. Hippocampal alterations following ECT should be the focus in searching for causes of the cognitive side-effects.

Introduction of an α-vinyl group into enal substrates can prohibit the traditional [3 + 2] cycloaddition with N-2,2,2-trifluoroethyl isatin imines catalysed by a chiral secondary amine, but give ß,γ'-regioselective [4 + 3] annulation products via cascade iminium ion-dienamine catalysis. A spectrum of CF3-containing spirooxindoles incorporating an azepane motif were constructed with good to excellent enantioselectivities. In addition, asymmetric [4 + 2] annulations between α-vinylenals and α,α-dicyanoalkenes were disclosed through a similar catalytic strategy, generally affording complex tricyclic frameworks with outstanding enantioselectivities.

Metal-organic frameworks (MOFs) as drug carriers have many advantages than traditional drug carriers and have received extensive attention from researchers. However, how to regulate the microstructure of MOFs to improve the efficiency of drug delivery and sustained release behaviour is still a big problem for the clinical application. Herein, the authors synthesise surfactant-modified ZIF-8 nanoparticles with different microstructures by using different types of surfactants to modify ZIF-8. The surfactant-modified ZIF-8 nanoparticles have the larger specific surface area and total micropore volumes than the original ZIF-8, which enables doxorubicin (DOX) to be more effectively loaded on the drug carriers and achieve controlled drug sustained release. Excellent degradation performance of ZIF-8 nanoparticles facilitates the metabolism of drug carriers. The formulation was evaluated for cytotoxicity, cellular uptake and intracellular location in the A549 human non-small-cell lung cancer cell line. ZIF-8/DOX nano drugs exhibit higher cytotoxicity towards cells in comparison with free DOX, suggesting the potential application in nano drugs to cancer chemotherapy.

BACKGROUND: Myocardial ischemia and reperfusion injury (MI/RI) is a complex pathophysiological process, which can lead to severe myocardial injury. The long noncoding RNA alpha-2-macroglobulin antisense RNA 1 (A2M-AS1) has been revealed to be abnormally expressed in MI, However, its function in MI and the potential mechanism are still unclear. OBJECTIVE: To evaluate the functional role of A2M-AS1 in hypoxia/reoxygenation (H/R)-induced neonatal cardiomyocytes and its potential molecular mechanism. METHODS: Dataset GSE66360 was obtained from GEO database for analyzing the RNA expression of A2M-AS1 and interleukin 1 receptor type 2 (IL1R2). KEGG pathway enrichment analysis of the genes that co-expressed with A2M-AS1 was performed. Human neonatal cardiomyocytes were subjected to H/R to construct in vitro models. QRT-PCR and Western blot were adopted to test the levels of mRNA and protein. The viability and apoptosis of cardiomyocytes were tested by CCK-8 and flow cytometry assays, respectively. RESULTS: The expression of A2M-AS1 was notably downregulated in H/R-treated cardiomyocytes. Overexpression of A2M-AS1 can notably enhance the cell viability of H/R-damaged cardiomyocytes, whereas knockdown of A2M-AS1 showed the opposite outcomes. Besides, a negative correlation was showed between A2M-AS1 and IL1R2 expression. In H/R-treated cardiomyocytes, overexpression of IL1R2 weakened the promoting proliferation and anti-apoptosis effects caused by overexpressing A2M-AS1, however, IL1R2-knockdown abolished the anti-proliferation and pro-apoptosis effects caused by silencing A2M-AS1. CONCLUSION: This study demonstrates the potential regulatory role of A2M-AS1/ IL1R2 axis in cardiomyocytes suffered from H/R, and provides insight into the protection of MI/RI.

OBJECTIVE: Pulmonary nodules (PNs) are one of the imaging manifestations of early lung cancer screening, which should receive more attention. Traditional Chinese medicine believes that voice changes occur in patients with pulmonary diseases. The purpose of this study is to explore the differences in phonetic characteristics between patients with PNs and able-bodied persons. METHODS: This study explores the phonetic characteristics of patients with PNs in order to provide a simpler and cheaper method for PN screening. It is a case-control study to explore the differences in phonetic characteristics between individuals with and without PNs. This study performed non-parametric statistics on acoustic parameters of vocalizations, collected from January 2017 to March 2018 in Shanghai, China, from these two groups; it explores the differences in third and fourth acoustic parameters between patients with PNs and a normal control group. At the same time, computed tomography (CT) scans, course of disease, combined disease and other risk factors of the patients were collected in the form of questionnaire. According to the grouping of risk factors, the phonetic characteristics of the patients with PNs were analyzed. RESULTS: This study was comprised of 200 patients with PNs, as confirmed by CT, and 86 healthy people that served as a control group. Among patients with PNs, 43% had ground glass opacity, 32% had nodules with a diameter ≥ 8 mm, 19% had a history of smoking and 31% had hyperlipidemia. Compared with the normal group, there were statistically significant differences in pitch, intensity and shimmer in patients with PNs. Among patients with PNs, patients with diameters ≥ 8 mm had a significantly higher third formant. There was a significant difference in intensity, fourth formant and harmonics-to-noise ratio (HNR) between smoking and non-smoking patients. Compared with non-hyperlipidemia patients, the pitch, jitter and shimmer of patients with PNs and hyperlipidemia were higher and the HNR was lower; these differences were statistically significant. CONCLUSION: This measurable changes in vocalizations can be in patients with PNs. Patients with PNs had lower and weaker voices. The size of PNs had an effect on the phonetic formant. Smoking may contribute to damage to the voice and formant changes. Voice damage is more pronounced in individuals who have PNs accompanied by hyperlipidemia.

Introduction: Dry eye disease (DED) prevalence is estimated at 9.3% of the US adult population, although diagnosed rate is much lower. This study examined real-world incidence rates (IR) and prevalence rates (PR) of DED in adults using continuous positive airway pressure (CPAP) or nasal mask therapy (NMT) devices to treat sleep apnea. Methods: Using IBM MarketScan Commercial and Medicare Supplemental claims databases, this study identified adults with ≥1 claim of CPAP or other NMT device between January 1, 2014 and June 30, 2018, ≥1 diagnosis of sleep apnea during a 12-month pre-index period, and continuous benefit enrollment ≥12 pre- and post-index date. The date of the first CPAP or NMT device claim was considered the index date. Descriptive analyses included PR, IR, and IR per 100-person years (100PY) for the overall population and subgroups including age, sex, and baseline comorbidities. Results: The 1-, 2-, and 3-year PR of DED was 6.2%, 10.0%, and 13.0%, while the IR of DED was 4.0%, 7.3%, and 10.3%, respectively. Females had a higher IR of DED compared to males: 5.8%, 10.8%, and 15.1% vs 3.0%, 5.4%, and 7.9%, respectively. DED increased with age with a 1-, 2-, and 3-year PR for patients aged 18-24 years of 2.2%, 3.4%, and 5.0% vs 17.6%, 25.8%, and 32.1% in patients aged ≥75, respectively. Overall, IR per 100PY of DED was 3.68, higher for females than males (5.51 vs 2.73). PR and IR of DED were high among patients with comorbid inflammatory or metabolic conditions. Conclusion: The PR and IR of DED in CPAP or NMT users were higher than the reported prevalence of DED in the general population. CPAP/NMT users who were female, older, or had comorbid inflammatory or metabolic conditions may experience a higher incidence and prevalence of DED.

Objective: Our study aimed to investigate the effect of anti-differentiation noncoding RNA (ANCR) on hepatocellular carcinoma (HCC) and its potential molecular mechanisms. Methods: The expression of ANCR was detected by qRT-RCR in both HCC tissues and HCC cells. Moreover, the relationship between ANCR expression and clinical parameters in HCC patients was investigated. The proliferation, cell clones, migration, invasion and apoptosis of MHCC97H and HCCLM3 cells were measured by MTT assay, colony formation assay, transwell assay and flow cytometry, respectively. The expressions of N-cadherin, vimentin, E-cadherin, cleaved caspase-3, Bax, Bcl-2, Wnt1, ß-catenin and GSK-3ß in MHCC97H and HCCLM3 cells were measured by Western blot. Results: Our results showed that ANCR was lowly expressed in both HCC tissues and HCC cells. ANCR expression was closely associated with tumor size, tumor-node-metastasis (TNM) stages and vascular invasion in HCC. ANCR could dramatically inhibit cell proliferation, migration and invasion, as well as promote apoptosis in MHCC97H and HCCLM3 cells. ANCR could significantly increase the expression of cleaved caspase-3, Bax, E-cadherin and GSK-3ß but reduce the expression of Bcl-2, N-cadherin, vimentin, Wnt1 and ß-catenin in MHCC97H and HCCLM3 cells. In addition, Wnt/ß-catenin pathway inhibitor (IWP-2) partially reversed the effects of silencing ANCR on the proliferation, migration, invasion and apoptosis of HCCLM3 cells. Conclusion: Our study demonstrated that ANCR can suppress cell proliferation, migration and invasion, as well as promote apoptosis of HCC cells via modulation of the Wnt/ß-catenin signaling pathway.
