RESUMO

Pig leg weakness not only causes huge economic losses for producers but also affects animal welfare. However, genes with large effects on pig leg weakness have not been identified and suitable methods to study porcine leg weakness are urgently needed. Bone mineral density (BMD) is an important indicator for determining leg soundness in pigs. Increasing pig BMD is likely to improve pig leg soundness. In this study, porcine BMD was measured using an ultrasound bone densitometer in a population with 212 Danish Landrace pigs and 537 Danish Yorkshires. After genotyping all the individuals using GeneSeek Porcine 50K SNP chip, genetic parameter estimation was performed to evaluate the heritability of BMD. Genome-wide association study and haplotype analysis were also performed to identify the variants and candidate genes associated with porcine BMD. The results showed that the heritability of BMD was 0.21 in Landrace and 0.31 in Yorkshire. Five Single Nucleotide Polymorphisms (SNP) on chromosome 6 were identified associated with porcine BMD at suggestive significance level. Two candidate Quantitative Trait Loci ( QTL ) (74.47- 75.33Mb; 80.20- 83.83Mb) and three potential candidate genes (ZBTB40, CNR2, and Lin28a ) of porcine BMD were detected in this study.

We present an IoT-based monitoring system for healthcare that allows for long-term measurements of blood pressure (BP), heart rate (HR), and body weight (BW), as well as near-infrared spectroscopy (NIRS) for measurement of prefrontal cortex (PFC) activity. To verify the applicability of the system, it was set up in a local fitness gym for a preliminary study. A total of 39 subjects, selected from members of the gym, participated in the study. We analyzed the BP, HR, and BW data, collected from the subjects over one half-year. In addition, to assess the degree of mental stress of the subjects, we analysed left-right asymmetry of the PFC activity using the laterality index at rest (LIR) of the NIRS parameter. Results show that the subjects were able to measure their physiological data by themselves when they visited the gym, after being instructed how to perform the measurements. Furthermore, the results also indicate that ordinary people can continuously monitor physiological functions such as brain function in a non-medical facility, such as a fitness gym.

We present an unobtrusive cuff-less sphygmomanometer based on contact-type and optical pulse sensors for continuous and minimally invasive monitoring of blood pressure (BP). We developed a cuff-less sphygmomanometer that utilizes the pulse arrival time (PAT) to estimate continuous BP. To assess its accuracy, we recruited 10 healthy subjects in whom we carried out BP studies using the cuff-less sphygmomanometer compared with a standard cuff-type device in a stationary sitting patient. Preliminary results showed that the mean difference (MD) of estimated systolic blood pressure and diastolic blood pressure were 0.96 ± 9.6 (mean ± SD) mmHg and 1.14 ± 7.5 mmHg, respectively, compared to the control. The corresponding correlation between the estimated BP values and controls were 0.78 for systolic blood pressure (p < 0.01) and 0.69 for diastolic blood pressure (p < 0.01); thus, there were significant correlations. These results suggest that the developed cuff-less sphygmomanometer has the potential for continuous BP monitoring. Finally, we conducted a preliminary study of simultaneous monitoring of cuff-less BP and near-infrared spectroscopy to evaluate the potential for assessment of autonomic nervous system functions during mental stress tasks.

AIM: This study was carried out to investigate the effects of dietary calcium pyruvate supplementation on growth performance and intestinal health of weaned piglets fed low-protein diets. METHODS AND RESULTS: After a 7-day adaptation period, 60 individually housed piglets (Duroc × Yorkshire-Landrace) weaned at 28 days of age were randomly assigned to receive one of three treatments (20 pigs/treatment) for 28 days: control diet (20·0% crude protein [CP]), low-protein diet (15·5% CP), and experimental (15·5% CP + 1·8% calcium pyruvate). At the end of the experiment, six piglets from each diet group were slaughtered and blood and tissue samples were collected. Compared with the control group, feeding piglets with 15·5% CP decreased the daily body weight gain; lengths of the duodenum, jejunum and ileum; and weights of the stomach, duodenum, jejunum and ileum (P < 0·05), while 15·5% CP + 1·8% calcium pyruvate supplementation removed those differences (P > 0·05). Compared with the control group, the diarrhoea incidence and relative richness of Firmicutes in the colon contents of piglets in both the 15·5% CP and 15·5% CP + 1·8% calcium pyruvate groups was decreased. The relative richness of Bacteriodetes in the colon contents of piglets was higher in the 15·5% CP + 1·8% calcium pyruvate group than in the control and 15·5% CP groups (P < 0·05). CONCLUSION: Calcium pyruvate supplementation for four weeks removed the negative effects of a low-protein diet on the gastrointestinal tract development and daily body weight gain of weaned piglets. SIGNIFICANCE AND IMPACT OF THE STUDY: This study showed that supplementing a low-protein diet with calcium pyruvate, an effective alternative metabolic fuel to amino acids, was beneficial in improving the intestinal health and maximizing the growth of newly weaned piglets.

BACKGROUND: Adipose-derived stem cells have been considered as a promising therapy for erectile dysfunction. However, the therapeutic efficacy of adipose-derived stem cell-based therapy requires improvement. OBJECTIVE: To determine whether the inhibition of phosphodiesterase type 5 in adipose-derived stem cells would improve stem cell therapy for rats with diabetes-induced erectile dysfunction. MATERIALS AND METHODS: A phosphodiesterase type 5 siRNA was incorporated into lentiviral vectors and transduced into adipose-derived stem cells. The mRNA and protein levels of phosphodiesterase type 5 were evaluated. Three days after transduction, the adipose-derived stem cell supernatant was collected to determinate the levels of insulin-like growth factor 1 and vascular endothelial growth factor. Streptozotocin-induced diabetic rat models were established and used for comparative analysis of 1- and 2-week treatment regimens with intracavernosal injection of adipose-derived stem cells or Lv-siPDE5-modified adipose-derived stem cells. RESULTS: Lv-siPDE5-ADSCs secreted more insulin-like growth factor 1 and vascular endothelial growth factor in supernatants than unmodified adipose-derived stem cells. Preconditioned Adipose-derived stem cells-treated diabetic rats showed consistently superior erectile function when compared with non-preconditioned adipose-derived stem cells after 2 weeks of treatment. Lv-siPDE5-ADSCs provided additional benefits in recovery of cavernous structures with rapid effects (1 week) when compared to plain adipose-derived stem cells. These features were associated with the significantly increased levels of insulin-like growth factor 1 and vascular endothelial growth factor in Lv-siPDE5-ADSC-treated diabetic rats. CONCLUSIONS: Adipose-derived stem cell therapy could serve as an alternate approach for diabetes-induced erectile dysfunction, albeit with a long onset period. In vitro preconditioning of adipose-derived stem cells could accelerate the functional and structural recovery in vivo, indicating that preconditioning by inhibition of phosphodiesterase type 5 may improve adipose-derived stem cells therapy following diabetes-induced erectile dysfunction.

AIM: The aim of this study was to investigate the effects of dietary supplementation of Rhodotorula mucilaginosa solid-state fermentation product (RSFP) on the laying performance, egg quality and intestinal microbial flora of hens. METHODS AND RESULTS: In this study, 40-week-old Roman laying hens (n = 216) were randomly assigned to one of the four groups: the control (CON) group, fed 87.5% basal diet +12.5% fermentation substrate; the 0.5% RSFP group, fed 87.5% basal diet +12.0% fermentation substrate +0.5% RSFP; the 2.5% RSFP group, fed 87.5% basal diet +10.0% fermentation substrate +2.5% RSFP; and the 12.5% RSFP group, fed 87.5% basal diet +12.5% RSFP. The effect of each treatment was analysed in six replicates of nine hens. The experimental period was 31 days, which included a 3-day adaptation period. After 31 days of feeding, one hen from each replicate was randomly selected and killed, and the jejunum and digesta in the cecum were collected for the determination of the intestinal morphology and microbial flora respectively. Daily egg mass in the 2.5 and 12.5% RSFP groups and egg production and feed conversion ratio in the 12.5% RSFP group were higher than those in control group (P < 0.05). The yolk colour was improved in hens fed RSFP-supplemented diets (P < 0.05). Hens fed RSFP-supplemented diet showed a decrease in the relative abundances of Proteobacteria, Bacteroides, Helicobacteraceae, Helicobacter and Lachnospiraceae UCG-002, but an increase in the relative abundance of Prevotellaceae UCG-001 in the cecum (P < 0.05). CONCLUSION: Dietary RSFP supplementation improved the laying performance, egg quality and intestinal microflora of hens. SIGNIFICANCE AND IMPACT OF THE STUDY: Dietary supplementing diet with Rhodotorula mucilaginosa solid-state fermentation product, which is rich in carotenoids, improved the yolk colour and increased the carotenoid content, thereby improving the intestinal health of hens.

OBJECTIVES: The HIV prevalence among Chinese men who have sex with men (MSM) is high, while the HIV testing rate is low. HIV self-testing (HIVST) is a promising means of scaling up HIV testing among MSM in China, as the ability to test in private is appealing to many individuals. We evaluated the feasibility of promoting oral HIVST via the internet in the MSM population. METHODS: From April 2013 to April 2014, MSM in two major cities in China were recruited for an observational study with assessment at baseline and follow-up. Data were collected via questionnaire, oral HIVST, and clinic-based HIV confirmatory testing. RESULTS: A total of 510 MSM were recruited at baseline and 279 (54.7%) returned for a clinic follow-up visit. Compared to MSM recruited via peer referral, those recruited via the internet were better educated, had a higher monthly income, sought and met male sex partners online more frequently, had less often tested or orally tested for HIV before, and being more likely to seek HIV knowledge or HIV intervention information online. The sensitivity of oral HIVST among MSM recruited via the internet was 92.9%, and the specificity was 96.8%. A total of 19.0% of MSM sought both male sex partners and HIV intervention programmes online. The associated factors were > 10 past male sex partners and ever testing for HIV using an oral self-test kit. All MSM who were followed up re-administered oral HIVST, yet fewer MSM recruited via the internet accepted blood retesting than other MSM. CONCLUSIONS: Promoting oral HIVST via the internet could be a feasible and promising approach to facilitate HIV testing among MSM in China.

OBJECTIVE: To explore the effects from the thread shapes of custom-made root-analogue implant (RAI) on distributions of von Mises stress around the peri-implant bone. METHODS: Five one-stage RAI three-dimensional finite element (FE) models with different thread shapes (V-shaped design, square design, buttress design, reverse buttress design and none thread design) and congruent bone were created through reverse engineering technology. The data of the five models were imported into the FE analysis software to calculate. A force of 100 N was applied parallelly and of 45° to the implant axis respectively. Analysis was performed to evaluate the von Mises stress distributions at the peri-implant regions with the help of the Ansys 16 software. RESULTS: The von Mises stresses distributed mostly at the implant cervical regions and the tip ends of the threads on the cortical bone under oblique loading, while on the cancellous bone, the stresses concentrated mostly on the implant lateral cervical regions, the tip ends of the threads and the apical regions. When under vertical loading, the von Mises stresses distributed mostly at the implant cervical regions on the cortical bone while at the tip ends of the threads and the lateral apical regions on the cancellous bone. The von Mises stresses were better distributed on the thread groups under both kinds of loadings compared with no thread design. But there was no obvious difference among the different thread groups. The concentrations of the von Mises stresses on the cancellous bone in the thread groups were mostly at the tip ends of the threads while less in the apical area. The von Mises stresses were better distributed on the cancellous bone on the other three thread designs than on square design. CONCLUSION: Thread designs are advocated for the reason that adding thread designs to the RAI standard design will have a positive effect on stress distributions at the peri-implant regions and it will reduce the concentrations of von Mises stresses on the cortical bone. From the standpoint of the stress distribution, V-shaped design, buttress design and reverse buttress design are more suitable for RAI than square design. There is no difference of the distributions of the von Mises stresses in the RAI between different thread designs.

Objective: To investigate the influence of different variable combinations of the carrying system on gait and muscle fatigue during weight-bearing walking on both shoulders and back, and to optimize daily carrying experience. Methods: From September to December, 2018, 6 male college students were selected as subjects; the electromyographic signal of the anterior tibial muscle and plantar pressure were measured during weight-bearing walking under six different variable combinations of the carrying system (with the three variables of gravity center, single or double shoulders, and waist cushion), and a subjective evaluation test was performed for the degree of fatigue. The electromyographic data and plantar pressure data were processed and compared to evaluate behavior and fatigue. Results: Different variable combinations of the carrying system had significant influence on the degree of fatigue and gait (P<0.05). Both subjective and objective analyses found that the carrying system with different heights of gravity center and single-or double-shoulder carrying mode had great influence on human fatigue and walking gait, while the hardness of waist cushion had little influence. Compared with the other groups, the group with a high gravity center, a double-shoulder carrying mode, and a soft waist cushion had a significantly smaller slope of average electromyographic amplitude, a significantly larger slope of median frequency, and a significantly lower degree of plantar pressure curve disorder. Conclusion: The carrying system with a relatively high gravity center, a double-shoulder carrying mode, and a soft waist cushion can significantly relieve fatigue, with low influence on walking gait.

OBJECTIVE: Long non-coding RNAs (lncRNAs) have been confirmed to play important roles in the progression of different cancers. The aim of this study was to detect the expression level of lncRNA CEBPA-AS1 in liver cancer and to study its influence on cell proliferation, invasion and prognosis. PATIENTS AND METHODS: Quantitative Real Time-Polymerase Chain Reaction (qRT-PCR), MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide) assay, transwell assay, Western blot, Kaplan-Meier survival curve and Cox regression were used to evaluate lncRNA CEBPA-AS1 expression, cell proliferation, invasion, epithelial-mesenchymal transition (EMT)-related molecules expression and prognosis, respectively. RESULTS: The expression of lncRNA CEBPA-AS1 increased significantly in liver cancer tissues (p<0.05). Meanwhile, CEBPA-AS1 expression was associated with tumor size, portal vein tumor thrombus and invasion and metastasis (p<0.05). In vitro experiments indicated that downregulation of lncRNA CEBPA-AS1 could effectively reduce cell proliferation, invasion and EMT process. CONCLUSIONS: LncRNA CEBPA-AS1 acts as an oncogene in liver cancer, which may be a novel biomarker in liver cancer progression.

OBJECTIVE: To evaluate the sinus bone gains after sinus floor elevation procedures with or without grafts when implants were placed simultaneously. METHODS: The research included 26 edentulous patients in the maxillary posterior region, who were divided into 2 different groups according to the sinus lift procedures employed: group of osteotome sinus floor elevation with bone grafts (group A) and group of osteotome sinus floor elevation without bone grafts (group B). The implants were placed simultaneously, and were followed for a period of 30-50 months after sinus floor elevation as routine and then were observed for sinus bone change. RESULTS: There were 27 implants (13 implants for group A and 14 implants for group B) and the following time was 40.23 (36.20, 48.07) months. The residual bone height (RBH) was (6.64±1.21) mm for group A and ( 6.96±1.36) mm for group B; the difference between the two groups was not statistically significant (P=0.459).The sinus bone gain (SBG) when followed was (2.20±1.71) mm for group A, and 1.77 (0.94, 2.05) mm for group B; the difference between the two groups was not statistically significant (P = 0.583). The quality of apical bone around implant apex was divided into 3 categories which were excellent, fine and fair.The quality of apical bone around implant apex was excellent and fine for 9 implants in group A and 8 implants in group B, and the difference between the two groups was not statistically significant (Fisher exact test, P=0.695).The implant apex when followed was (0.09±1.32) mm below the new sinus floor for group A and (0.03±0.91) mm for group B, and the difference between the two groups was not statistically significant (P = 0.898). The SBG for the total was 1.85 (1.10, 2.20) mm. The SBG was statistically significantly only related to the protrusion height of the implant apex (r=0.383, P=0.049). CONCLUSION: Sinus floor elevation procedures with or without grafts can all gain good results. The SBG is related to the protrusion height of the implant apex significantly.

OBJECTIVES: During surveillance, we found a new type of Vibrio parahaemolyticus named 'O4:KUT-recAin' and studied the phenotypic, pathogenic and epidemiological characteristics of O4:KUT-recAin. METHODS: V. parahaemolyticus were isolated from acute diarrhoeal patients in coastal hospitals of China. Serum agglutination test, specific PCR assay, growth curves under different conditions and rabbit diarrhoeal models were using to characterize O4:KUT-recAin. RESULTS: The O4:KUT-recAin strain has a new type of K antigen and a 25 043-bp-large fragment encoding 20 proteins inserted in the housekeeping gene recA. Retrospective analysis found that only one O4:KUT-recAin strain was detected in 563 V. parahaemolyticus strains in 2014; then the proportion increased rapidly and reached 17.8% (105/590) in 2016 and 31.1% (224/721) in 2017, making O4:KUT-recAin the second dominant serotype following O3:K6. O4:KUT-recAin strains (100%, 14/14) exhibited increased acid resistance and could reproduce in medium at pH 4.9, while 92.9% (13/14) of the O3:K6 strains could not grow at this pH value. O4:KUT-recAin could cause diarrhoea and small intestinal tissue lesions in infant rabbits, but its diarrhoeal (93.1%, 27/29) and mortality (78.6%, 22/28) rates were slightly lower than those of O3:K6 (100% 16/16, 100% 16/16). Based on diarrhoea patients, there were no significant differences in the two groups for most clinical symptoms and laboratory results, except media age, haemoglobin and the number of red blood cells in stool samples. CONCLUSIONS: O4:KUT-recAin had enhanced acid resistance, was capable of causing infectious diarrhoea in both rabbits and humans, and has become widespread during a short period of time in China.

LncRNAs comprise a vast majority of the RNAs transcribed from non-coding regions of mammalian genomes. Recently, lncRNAs have emerged as key regulators of a wide range of biological processes. In this study, we identified lncRNAs expressed in swine ovary tissue from Yorkshire and Meishan pigs. We obtained 3827 lncRNAs expressed from 4973 transcripts in swine ovary tissue, of which 510 were ovary specific, 192 were differentially expressed between Yorkshire and Meishan pigs and 38 were both ovary specific and differentially expressed. Furthermore, we randomly selected several differentially expressed and ovary-specific lncRNAs for verification by qRT-PCR and RT-PCR respectively. We also predicted the functions of the lncRNAs by analysing their nearest neighbouring protein-coding genes. Together, our results identify a series of ovary-associated lncRNAs for further experimental investigation of the functions of these genes in ovary development or for selection of candidate genes for breeding.

Abstract: Objective To analyze the genetic phenotypes of Y-chromosome STR and SNP in Han male population of Wujiang area, Suzhou City and explore the genetic structure of population of Wujiang area for further examination of regional-specific Y-SNP genetic markers ancestor haplogroups. Methods Blood samples of 472 Wujiang area Han males were randomly collected and genotyped by YfilerTM Plus PCR Amplification Kit. The allele frequencies and haplotype frequencies of each locus were obtained using the direct calculation method. Y-SNP haplogroups of each sample were estimated using Y-Predictor software and verified through experiments by amplification refractory mutation system-polymerase chain reaction (ARMS-PCR). Results A total of 453 haplotypes were found in the 27 Y-STR genetic markers in 472 Han males of Wujiang area. The haplotype diversity (HD) was 0.997 696 93, among which, the highest gene diversity (GD) value was DYF387S1a/b (GD=0.953 1) and the lowest was DYS438 (GD=0.321 8). Based on genotyping data of 27 Y-STRs and 472 samples, 132 haplogroups from C, D, N, O and Q, etc downstream Y-SNP haplogroups were estimated and then verified through experiments. Conclusion This study is based on Y-chromosome STR haplotypes, and predicts Y-SNP haplogroups by Y-Predictor software, then uses ARMS-PCR to verify. Y-SNP genetic markers were introduced to achieve precise analysis of the genetic structure of male families in population of three towns in Wujiang area.

OBJECTIVE: Acute lung injury (ALI) is the most common complication of severe acute pancreatitis (SAP) in the early stage, which causes systemic inflammatory response and organ damage. Human runt-associated transcription factor 3 gene (RUNX3) has been reported to participate in various inflammatory diseases. However, the exact role of RUNX3 in SAP and its-related ALI remains unclear. MATERIALS AND METHODS: To establish the model of SAP, rats were retrogradely injected with 5% sodium taurocholate (1 mg/kg body weight) into the biliary-pancreatic duct. Cytokine level in serum was measured by ELISA, and the polymorphonuclear neutrophil (PMN) was isolated from rat's blood 12 h-post SAP induction. RESULTS: We found RUNX3 expression was significantly decreased with the progression of SAP. Both pancreas damages and cytokine production abilities were reduced in RUXN3-overexpressed SAP rats compared with control rats. Moreover, SAP-associated ALI was also improved upon RUNX3 overexpression in SAP rats. RUNX3 upregulation enhanced PMN apoptosis and inhibited Janus kinase 2/signal transducer and activator of transcription 3 (JAK2/STAT3) phosphorylation. CONCLUSIONS: Our study indicates that RUNX3 protects against SAP and SAP-associated ALI through controlling PMN apoptosis and regulating JAK2/STAT3 signaling pathway. RUNX3 could be regarded as a potent therapeutic target in SAP for future studies.

Objective: This study was conducted to evaluate the effects of tannins and cellulase on growth performance, nutrient digestibility, blood profiles, intestinal morphology and carcass characteristics in Hu sheep. Methods: A total of 48 three-month-old meat Hu sheep (25.05 ± 0.9 kg) were blocked based on body weight, and randomly allotted to 4 treatments with 3 replicates of 4 sheep each. The experiment lasted for 80 d, and dietary treatments were as follows: (1) CON, control diet; (2) TAN, CON + 0.1% tannins; (3) CEL, CON + 0.1% cellulase; (4) TAN+ CEL, CON + 0.1% tannins and 0.1% cellulase. Results: Compared with CON, CEL and TAN+CEL had greater (P<0.05) final birth weight (FBW) and average daily gain but lower (P<0.05) F/G, while FBW of TAN+CEL was lower (P<0.05) than that of CEL. The apparent total tract digestibility (ATTD) of DM in TAN, CEL and TAN+CEL groups were higher (P<0.05) than that in CON. CEL and TAN+CEL groups had greater (P<0.05) ATTD of CF compared with TAN and CON, while TAN group had lower (P<0.05) ATTD of CP than other treatments. TAN, CEL and TAN+CEL groups increased (P<0.05) serum globulin and alkaline phosphatase but decreased (P<0.05) A/G. Serum total protein was greatest for TAN+CEL, intermediate for TAN and CEL and least for CON (P<0.05). TAN+CEL group increased (P<0.05) dressing percentage compared with CON, while the backfat thickness of CEL was lower (P<0.05) than that of CON. The villus height of jejunum and ileum in CEL and TAN+CEL groups were greater (P<0.05) than that in CON, and the crypt depth and villus height: crypt depth of jejunum were increased (P<0.05) in TAN, CEL and TAN+CEL groups. Conclusion: The addition of tannins and cellulase together promoted nutrient digestion, liver protein synthesis and intestinal development and thus improved growth performance and carcass characteristics.

Intrahepatic cholangiocarcinoma(ICC) is the second common primary liver cancer originated from epithelium of the sub-branches of intrahepatic bile ducts with extremely poor prognosis and lack of effective treatment.The prognosis of ICC is mostly affected by the origin,the type and the size of the tumor as well as the intrahepatic metastasis(satellite lesion) and lymph node metastasis etc.Surgical resection remains the first choice of treatment to patients with ICC.However, there are multiple issues in surgical treatment of ICC, which have not been reached a consensus.Among them, the value of systematic lymphadenectomy during hepatic resection for ICC patient remains one of the hot spot issues.Given the heterogeneity of ICC,we recommend planning the procedure of the radical resection and lymphadenectomy personally, according to the type and origin of the tumor, the number and locationof the lesion.The pre-operation imaging examination and the intra-operation lymph node tracing technique could provide valuable information to help the surgeon decide the range of systematic lymphadenectomy.Routine systematic lymphadenectomy is recommended in the surgical treatment of ICC patients by experienced surgeons even without evidence of lymph node metastasis.The resected lymph tissue should be labeled by the provenance for further study.
