RESUMO

An optimal lipid droplet (LD) content is essential for successful mammalian embryonic development. Salidroside (SAL) is a traditional Chinese medicine and one of the important active components of the Rhodiola plant. SAL possesses antioxidative, anti-aging, and cardiovascular properties. Here, we studied the effects of SAL on in vitro maturation (IVM) of porcine oocytes and the subsequent embryonic development after parthenogenetic activation (PA). We found that 100 µM of SAL had no effect on the extrusion rate of the first polar body of porcine oocytes but significantly improved the subsequent blastocyst formation rate and embryo quality. Our study further revealed that SAL treatment altered the morphology, increased the lipid content in oocytes, increased mitochondrial number. Further analysis revealed that SAL upregulated the expression of genes related to lipid metabolism (FASN, FADS1, HSL, and CPT1a) and the mitochondria function-related genes (PGC-1α). These results suggest that SAL supplementation enhances oocyte maturation and subsequent embryonic development by promoting lipid metabolism, providing the necessary energy for the aforementioned processes.

Dietary phenolic compounds must be released from the food matrix in the gastrointestinal tract to play a bioactive role, the release of which is interfered with by food structure. The release of phenolics (unbound and bound) of cold and hot extruded noodles enriched with phenolics (2.0%) during simulated in vitro gastrointestinal digestion was investigated. Bound phenolic content and X-ray diffraction (XRD) analysis were utilized to characterize the intensity and manner of starch-phenolic complexation during the preparation of extruded noodles. Hot extrusion induced the formation of more complexes, especially the V-type inclusion complexes, with a higher proportion of bound phenolics than cold extrusion, contributing to a more controlled release of phenolics along with slower starch digestion. For instance, during simulated small intestinal digestion, less unbound phenolics (59.4%) were released from hot extruded phenolic-enhanced noodles than from the corresponding cold extruded noodles (68.2%). This is similar to the release behavior of bound phenolics, that cold extruded noodles released more bound phenolics (56.5%) than hot extruded noodles (41.9%). For noodles extruded with rutin, the release of unbound rutin from hot extruded noodles and cold extruded noodles was 63.6% and 79.0%, respectively, in the small intestine phase, and bound rutin was released at a much lower amount from the hot extruded noodles (55.8%) than from the cold extruded noodles (89.7%). Hot extrusion may allow more potential bioaccessible phenolics (such as rutin), further improving the development of starchy foods enriched with controlled phenolics.

This retrospective cohort analysis leveraged vaccination data for BNT162b2, mRNA-1273, and Ad26.COV2.S in the United States from the Komodo Healthcare Map database, the TriNetX Dataworks USA Network, and Cerner Real-World EHR (electronic health record) Data to evaluate rates of adherence to and completion of COVID-19 vaccination series (November 2020 through June 2021). Individuals were indexed on the date they received the first dose of a COVID-19 vaccine, with an adherence follow-up window of 42 days. Adherence/completion rates were calculated in the overall cohort of each database and by month of initiation and stratified by age, race/ethnicity, and urban/rural status. Overall adherence and completion to 2-dose COVID-19 mRNA vaccine schedules ranged from 79.4% to 87.4% and 81.0% to 89.2%, respectively. In TriNetX and Cerner, mRNA-1273 recipients were generally less adherent compared with BNT162b2 across sociodemographic groups. In Komodo, rates of adherence/completion between mRNA-1273 and BNT162b2 were similar. Adherence/completion were generally lower in younger (<65 years) versus older recipients (≥65 years), particularly for mRNA-1273. No other sociodemographic-based gaps in vaccine adherence/completion were identified. These data demonstrate high but incomplete adherence to/completion of multidose COVID-19 vaccines during initial vaccine rollout in the United States. Multidose schedules may contribute to challenges associated with successful global vaccination.

Due to its unique physical and chemical properties, MXene has recently attracted much attention as a promising candidate for wastewater treatment. However, the low water permeation flux of MXene membrane remains a challenge that has not been fully solved. In this study, attapulgite was used to increase the flux of MXene membrane through a facile one-pot method, during which the MXene nanosheets were self-assembled while being intercalated by the attapulgite nanorods to finally form the composite membranes. Under optimal conditions, an increase of water permeation flux of 97.31% could be observed, which was attributed to the broadened nano-channel upon the adequate intercalation of attapulgite nanorods. Its permeation flux and rejection rate for methylene blue (MB) were further studied for diverse applications. In contrast to bare MXene, the permeation flux increased by 61.72% with a still high rejection rate of 90.67%, owing to the size rejection. Overcoming a key technique barrier, this work successfully improved the water permeability of MXene by inserting attapulgite nanorods, heralding the exciting prospects of MXene-based lamellar membrane in dye wastewater treatment.

BACKGROUND: Feminizing hormonal therapy (FHT) and HIV potentially alter cardiovascular disease (CVD) risk in transgender women (TW). METHODS: TW were enrolled in Los Angeles, California and Houston, Texas and frequency-matched to Multicenter AIDS Cohort Study cisgender men (CM) on age, race, substance use, and abacavir use. Biomarkers of CVD risk and inflammation were assessed via ELISA. Wilcoxon rank sum and Fisher's exact tests compared TW and CM. Multivariable linear regression assessed factors associated with biomarker concentrations. RESULTS: TW (HIV+ n  = 75, HIV- n  = 47) and CM (HIV+ n  = 40, HIV- n  = 40) had mean age 43-45 years; TW/CM were 90%/91% non-Hispanic Black, Hispanic, or Multiracial, 26%/53% obese, and 34%/24% current smokers; 67% of TW were on FHT. Among people with HIV (PWH), TW had higher median extracellular newly-identified receptor for advanced glycation end-products (EN-RAGE), lipoprotein-associated phospholipase A2 (LpPLA2), oxidized low-density lipoprotein (oxLDL), soluble tumor necrosis factor receptor type (sTNFR) I/II, interleukin (IL)-8 and plasminogen activator inhibitor (PAI)-1, but lower soluble CD14, von Willebrand factor (vWF) and endothelin (ET)-1 levels than CM. Findings were similar for participants without HIV (all P  < 0.05). In multivariable analysis, TW had higher EN-RAGE, IL-6, IL-8, P selectin, PAI-1, oxLDL and sTNFRI/II concentrations, and lower vWF, independent of HIV serostatus and current FHT use. Both being a TW and a PWH were associated with lower ET-1. CONCLUSIONS: Compared to matched cisgender men, trans women have altered profiles of biomarkers associated with systemic inflammation and CVD. Further work is needed to decipher the contributions of FHT to CVD risk in TW with HIV.

Living with HIV infection is associated with early onset of aging-related chronic conditions, sometimes described as accelerated aging. Epigenetic DNA methylation patterns can evaluate acceleration of biological age relative to chronological age. The impact of initial HIV infection on five epigenetic measures of aging was examined before and approximately 3 years after HIV infection in the same individuals (n=102). Significant epigenetic age acceleration (median 1.9-4.8 years) and estimated telomere length shortening (all p≤ 0.001) were observed from pre-to post-HIV infection, and remained significant in three epigenetic measures after controlling for T cell changes. No acceleration was seen in age- and time interval-matched HIV-uninfected controls. Changes in genome-wide co-methylation clusters were also significantly associated with initial HIV infection (p≤ 2.0 × 10-4). These longitudinal observations clearly demonstrate an early and substantial impact of HIV infection on the epigenetic aging process, and suggest a role for HIV itself in the earlier onset of clinical aging.

Tellurium (Te), the most metallic semiconductor, has been widely explored in recent decades owing to its fantastic properties such as a tunable bandgap, high carrier mobility, high thermal conductivity, and in-plane anisotropy. Many references have witnessed the rapid development of synthesizing diverse Te geometries with controllable shapes, sizes, and structures in different strategies. In all types of Te nanostructures, Te with one-dimensional (1D) hollow internal structures, especially nanotubes (NTs), have attracted extensive attention and been utilized in various fields of applications. Motivated by the structure-determined nature of Te NTs, we prepared a minor review about the emerging synthesis and nanostructure control of Te NTs, and the recent progress of research into Te NTs was summarized. Finally, we highlighted the challenges and further development for future applications of Te NTs.

In non-small cell lung cancer (NSCLC), about 15% of epidermal growth factor receptor (EGFR) mutations are rare. Herein, we report a 39-year-old male with stage IV NSCLC with a rare EGFR M277E mutation and high PD-L1 expression. The patient first underwent gamma-knife treatment for brain metastasis; then, he was started on 40 mg afatinib daily in combination with two cycles of chemotherapy. The clinical effect was stable disease (SD), so the patient underwent intensity-modulated radiation therapy guided by cone beam computed tomography on the lesion in the lower lobe of the right lung. A combination therapy with afatinib and chemotherapy was employed as the first-line therapy. A palliative radiotherapy to the primary pulmonary tumor was added, resulting in a significant response based on the next computerized tomography (CT) scan. To date, this is the first presented case of NSCLC with EGFR p.M277E mutation and its corresponding clinical outcome to combination therapies.

INTRODUCTION: Antifreeze peptides regulate the physiological functions of frozen cells and even their apoptosis; however, the mechanisms by which antifreeze peptides regulate these processes remain unclear, although the interactions between cell membranes and ice are well known to be important in this process. OBJECTIVES: Our study aims to investigate how antifreeze peptides regulate cell physiological functions during the freezing process. METHODS: We investigated the cryoprotective effect of rsfAFP on the physiological functions of S. thermophilus under freezing stress by measuring cellular metabolism activity, intracellular enzyme activity, cell membrane characterization, and cell apoptosis. The mechanism by which rsfAFP impacts S. thermophilus physiological functions under freezing stress was investigated using multispectral techniques and cryo-TEM. RESULTS: We show that a recombinant antifreeze peptide (rsfAFP) interacts with the extracellular capsular polysaccharides and peptidoglycan of Streptococcus thermophilus and ice to cover the outer layer of the membrane, forming a dense protective layer that regulates the molecular structure of extracellular ice crystals, which results in reduced extracellular membrane damage, depressed apoptosis and increased intracellular metabolic activity. This interaction mechanism was indicated by the fact that S. thermophilus better maintained its permeability barrier, membrane fluidity, membrane structural integrity, and cytoplasmic membrane potential during freezing stress with rsfAFP treatment. CONCLUSION: These results provide new insights into the mechanism by which rsfAFP regulates frozen cellphysiological functionsand apoptosis under freezing stress.

The CsSnBr3photodetectors are fabricated by thermal evaporation and 75 °Cin situannealing, and the effect ofin situannealing on the morphology, structure, exciton dynamics and photoresponse of thermally evaporated CsSnBr3films are investigated. Especially, temperature dependent steady-state photoluminescence (PL) and transient PL decaying have been analyzed in details for understanding the exciton dynamics. Meanwhile, effect of annealing on the activation energy for trap sites (Ea), exciton binding energy (Eb), activation energy for interfacial trapped carriers (ΔE), trap densities and carriers mobilities are studied and the annealed (A-CsSnBr3) reveals obviously lowerEband trap density together with notably higher carrier mobility than those of the unannealed (UA-CsSnBr3). Temperature dependence of the integrated PL intensity can be ascribed to the combining effect of the exciton dissociation, exciton quenching through trap sites and thermal activation of trapped carriers. The temperature dependent transient PL decaying analysis indicates that the PL decaying mechanism at low and high temperature is totally different from that in intermediate temperature range, in which combing effect of free exciton and localized state exciton decaying prevail. The beneficial effects of thein situannealing on the photoresponse performance of the CsSnBr3films can be demonstrated by the remarkable enhancement of the optimal responsivity (R) afterin situannealing which increases from less than 1 A W-1to 1350 A W-1as well as dramatically improved noise equivalent power, specific detectivityD* and Gain (G).

BACKGROUND: Mesenchymal stem cells (MSCs) are promising candidates for tissue regeneration and disease treatment. However, long-term in vitro passaging leads to stemness loss of MSCs, resulting in failure of MSC therapy. This study investigated whether the combination of melatonin and human umbilical cord mesenchymal stem cells (hUC-MSCs) was superior to hUC-MSCs alone in ameliorating high-fat diet and streptozocin (STZ)-induced type II diabetes mellitus (T2DM) in a mouse model. METHODS: Mice were divided into four groups: normal control (NC) group; T2DM group; hUC-MSCs treatment alone (UCMSC) group and pretreatment of hUC-MSCs with melatonin (UCMSC/Mel) group. RESULTS: RNA sequence analysis showed that certain pathways, including the signaling pathway involved in the regulation of cell proliferation signaling pathway, were regulated by melatonin. The blood glucose levels of the mice in the UCMSC and UCMSC/Mel treatment groups were significantly reduced compared with the T2DM group without treatment (P < 0.05). Furthermore, hUC-MSCs enhance the key factor in the activation of the PI3K/Akt pathway in T2DM mouse hepatocytes. CONCLUSION: The pretreatment of hUC-MSCs with melatonin partly boosted cell efficiency and thereby alleviated impaired glycemic control and insulin resistance. This study provides a practical strategy to improve the application of hUC-MSCs in diabetes mellitus and cytotherapy. Overview of the PI3K/AKT signaling pathway. α-Ketoglutarate (α-KG) is a metabolite in the tricarboxylic acid cycle. It has a strong antioxidant function and can effectively prevent oxidative damage. Previous studies have shown that α-KG exists in porcine follicles, and its content gradually increases as the follicles grow and mature. However, the potential mechanism of supplementation of α-KG on porcine oocytes during in vitro maturation (IVM) has not yet been reported. The purpose of this study was to explore the effect of α-KG on the early embryonic development of pigs and the mechanisms underlying these effects. We found that α-KG can enhance the development of early pig embryos. Adding 20 µM α-KG to the in vitro culture medium significantly increased the rate of blastocyst formation and the total cell number. Compared with to that of the control group, apoptosis in blastocysts of the supplement group was significantly reduced. α-KG reduced the production of reactive oxygen species and glutathione levels in cells. α-KG not only improved the activity of mitochondria but also inhibited the occurrence of apoptosis. After supplementation with α-KG, pig embryo pluripotency-related genes (OCT4, NANOG, and SOX2) and antiapoptotic genes (Bcl2) were upregulated. In terms of mechanism, α-KG activates the Nrf2/ARE signaling pathway to regulate the expression of antioxidant-related targets, thus combating oxidative stress during the in vitro culture of oocytes. Activated Nrf2 promotes the transcription of Bcl2 genes and inhibits cell apoptosis. These results indicate that α-KG supplements have a beneficial effect on IVM by regulating oxidative stress during the IVM of porcine oocytes and can be used as a potential antioxidant for IVM of porcine oocytes.

To date, flexible pressure sensors built on silver nanowires (AgNWs) have attracted tremendous attention, owing to their versatile applications in wearable, human-interactive, health-monitoring devices. Cellulose and its derivatives, which show great promise in serving flexible pressure sensors as the desired substrate due to their natural abundance, biocompatibility, easy processibility, and low costs. Herein, we reported a rational strategy to design a silver nanowires-dual-cellulose conductive paper. Its morphology, chemical and crystal structures, thermal stability, mechanical performances, and electrical properties were carefully studied. The results suggested that good tensile properties (tensile strength ≤8.10 MPa), high electrical conductivity (≤ 1.74 × 104 S·m-1) with long-term stability, and good adhesion stability (bending cycles over 500) were obtained. Furthermore, the use of such conductive paper as substrate for versatile flexible pressure sensors was demonstrated, which exhibited fast response (~ 0.48 s) and high sensitivity, in response to finger motion, voice recognition, and human pulse, etc.

BACKGROUND: Social determinants of health (SDH), including food insecurity, are associated with depression in the general population. This study estimated the prevalence of depression and food insecurity and evaluated the impact of food insecurity and other SDH on depression in adults with rheumatoid arthritis (RA). METHODS: Adults (≥ 18 years) with RA were identified from the 2013-2014 and 2015-2016 National Health and Nutrition Examination Survey (NHANES). Depression was defined as a score of ≥ 5 (mild depression: 5-9; moderate-to-severe depression: 10-27) using the Patient Health Questionnaire-9 (PHQ-9). Food insecurity was assessed with the 18-item US Household Food Security Survey Module. Adults with household-level marginal-to-very-low food security were classified as experiencing food insecurity. The prevalence of depression and food insecurity among participants with RA were estimated. Weighted logistic regression was used to evaluate the association between depression and participants' characteristics including SDH. Penalized regression was performed to select variables included in the final multivariable logistic regression. RESULTS: A total of 251 and 276 participants from the 2013-2014 and the 2015-2016 NHANES, respectively, had self-reported RA. The prevalence of depression among these participants was 37.1% in 2013-2014 and 44.1% in 2015-2016. The prevalence of food insecurity was 33.1% in 2013-2014 and 43.0% in 2015-2016. Food insecurity was associated with higher odds of having depression (OR 2.17, 95% CI 1.27, 3.72), and the association varied by depression severity. Compared with participants with full food security, the odds of having depression was particularly pronounced for those with very low food security (OR 2.96, 95% CI 1.48, 5.90) but was not significantly different for those with marginal or low food security. In the multivariable regression, being female, having fair/poor health condition, any physical disability, and ≥ 4 physical limitations were significantly associated with depression. CONCLUSIONS: In adults with self-reported RA, the prevalence of depression and food insecurity remained high from 2013 to 2016. We found that depression was associated with SDH such as food insecurity, although the association was not statistically significant once adjusted for behavioral/lifestyle characteristics. These results warrant further investigation into the relationship between depression and SDH among patients with RA.

BACKGROUND: Recent studies indicate that circular RNA (circRNA) serves important roles in the development of intrahepatic cholangiocarcinoma (ICC). However, the role of circRNA reticulon 4 interacting protein 1 (circRTN4IP1) in ICC progression remains unknown. METHODS: Expression of circRTN4IP1, microRNA-541-5p (miR-541-5p), hypoxia inducible factor 1 subunit alpha (HIF1A) and other indicated protein markers was detected by quantitative real-time polymerase chain reaction or Western blot. The functional effects of circRTN4IP1 knockdown in ICC cells were analyzed by cell counting kit-8, cell colony formation, flow cytometry analysis, Western blot, glucose and lactate kit assays. The positive expression rate of HIF1A was detected by immunohistochemistry assay. The interaction between miR-541-5p and circRTN4IP1 or HIF1A was identified by dual-luciferase reporter, RNA immunoprecipitation or RNA pull-down assays. Xenograft mouse model assay was performed to determine the effect of circRTN4IP1 depletion on tumor formation. RESULTS: In contrast, ICC tissues and cells showed high expression of circRTN4IP1 and HIF1A, but low expression of miR-541-5p. Knockdown of circRTN4IP1 led to repression of cell proliferation and glucose metabolism, but promotion of cell apoptosis; however, circRTN4IP1 overexpression had opposite effects. In mechanism, circRTN4IP1 acted as a sponge for miR-541-5p, which was found to target HIF1A. MiR-541-5p inhibitors could remit circRTN4IP1 knockdown-mediated action. Also, HIF1A participated in the regulation of miR-541-5p in ICC progression. In support, circRTN4IP1 depletion impeded tumor formation in vivo. CONCLUSION: CircRTN4IP1 knockdown inhibited ICC cell malignancy by miR-541-5p/HIF1A axis, providing us with a reliable target for the therapy of ICC.

The present study assessed changes in patient management, economic burden, and overall survival (OS) in a contemporary cohort of 2775 US Medicare Advantage beneficiaries aged ≥66 years newly diagnosed with acute myeloid leukemia (AML) between 01 January 2015 and 30 June 2020. Use of venetoclax-based therapy increased and replaced hypomethylating agent (HMA) monotherapy as the most common first-line treatment choice in 2019-2020. In newly diagnosed AML patients aged ≥75 and 66-74 years, mean per-patient 1-year healthcare expenditures were $81,818 and $156,033 (2020 USD) and median OS was 2.3 and 8.5 months, respectively. In addition, 40% of Medicare Advantage patients with newly diagnosed AML continue to receive supportive care alone. These findings indicate that at the population level clinical outcomes remain poor for older adults with AML, pointing to a continuing unmet medical need.

Indoor airborne microplastics fibers (MPFs) are emerging contaminants of growing concern. Nowadays, air conditioners (ACs) are widely used in indoor environments. However, little is known about their impact on the distribution of indoor MPFs. In this study, we first disclosed the prevalence of MPF contamination in filters for indoor split ACs used in living rooms, dormitories, and offices. The average density of microfibers was 1.47-21.4 × 102 items/cm2, and a total 27.7-35.0% of fibers were MPFs. Of these fibers, the majority were polyester (45.3%), rayon (27.8%), and cellophane (20.1%). We further tracked the long-term accumulation of MPFs on AC filters in three types of rooms, and demonstrated that dormitories showed relatively heavy accumulation especially after running for 35-42 days. Furthermore, we found that simulative AC filters which had been lined with PET MPFs could effectively release those MPFs into indoor air, propelling them away from the ACs at varying distances. Statistical analysis showed that the estimated daily intake of MPFs (5-5000 µm length) from AC filters would increase gradually with their usage, with the intake volume reaching up to 11.2 ± 2.2-44.0 ± 8.9 items/kg-BW/day by the 70th day, although this number varied among people of different ages. Altogether, these findings suggest that AC filters can act as both a sink and a source of microplastics fibers. Therefore, AC filters should be evaluated not only for their substantial impact on the distribution of indoor airborne MPFs, but also for their role in the prevalence of the related health risks.

Buckwheat noodles are mainly sold in the form of fresh and dried noodles in China. Among the noodles with varied proportions of extruded buckwheat flour (20% to 80%), the cooking or textural qualities of fresh and dried buckwheat noodles (FBN and DBN, respectively) were significantly different, and FBN showed a lower cooking loss and breakage ratio and were more elastic than DBN. FBN-20% showed the highest sensory score, followed by DBN-50%. The mechanisms causing the quality differences were investigated using water mobility and the internal structures of the noodles were investigated with low-field nuclear magnetic resonance and scanning electron microscopy, respectively. Compared with FBN, DBN showed a denser internal structure, which explained its higher hardness. The water within FBN and DBN was mainly in the form of softly bound water and tightly bound water, respectively. FBN with highly mobile softly bound water (longer T 22) and a more uniform internal structure had a lower breakage ratio, whereas the trends of water relation with texture properties were different for FBN and DBN. The drying process and added extruded buckwheat flour together contributed to the varied cooking and textural properties.

Renewable lignocellulosic biomass can be effectively transformed to value-added products, enabling fast growth of related downstream processing. However, valorization of the by-produced cellulose-poor fraction, which is also in large volumes, is only occasionally reported regarding existing technologies. Here, a simple, general, and effective strategy for fabricating graphene quantum dots (GQDs) from the Miscanthus (MC) biorefinery waste consisting of sugars and depolymerized lignin, is developed. This process involves the fast and selective removal of most lignin and hemicellulose based on mild acid hydrotrope fractionation, with followed hydrothermal carbonization. The as-fabricated MC-derived GQDs (M-GQDs) exhibit several advantages such as few-layer graphene-like single crystalline structure, sulfur and nitrogen co-doping, bright fluorescence, excitation-dependent photoluminescence, and long fluorescence lifetime (11.95 ns). Furthermore, M-GQDs present prominent fluorescence reduction in the presence of Fe3+ with good linearity (≤0.995) and very low detection limit (≥1.41 nM). Later, it is found that the observed high sensitivity for Fe3+ is based on a dynamic quenching mechanism, which is caused by the Fe3+-induced increase in both the energy dissipation and photogenerated electron consumption. This work is anticipated to open new opportunities for promoting the integral valorization of biomass and sensitive fluorometric detection of Fe3+.

A novel recyclable deep eutectic solvent (DES) consisting of p-toluene sulfonic acid (p-TsOH) and choline chloride (ChCl) was developed for efficient woody poplar sawdust (PL) and herbaceous miscanthus (MC) conversion at mild condition. The extraction of leftover lignin on the surface of DES pretreated residues using NaOH solution at room temperature greatly improved the enzymatic hydrolysis efficiency. Near complete cellulose conversion of PL and MC residues were obtained with a degree of delignification and xylan removal over 90% at 100 °C within 40 min. The strong correlations between xylan (R2 = 0.95) and lignin (R2 = 0.82) removal with cellulose conversion were observed in MC as well as positive correlations (R2 > 0.77) in PL. The results demonstrated that the DES system coupling NaOH post-treatment was a promising method to achieve an economically feasible biomass conversion process, which was effective for both woody PL residues and herbaceous MC.
