RESUMO

We report a new enantiomeric pair of superatomic silver clusters, R/S-Ag17, prepared from chiral alkynyl ligands. R-Ag17 and S-Ag17 possess C3 symmetry and emit near-infrared (NIR) light with a quantum yield (QY) of 8.0% under ambient condition as well as NIR circularly polarized luminescence (CPL) as a result of the chirality of the excited states. Both experiments and theoretical calculations indicate for the first time that the CPL originates from transitions between superatomic 1Pz (along the C3 axis) and 1S orbitals. This work opens a new avenue for CPL-active metal nanoclusters by utilizing chiral alkynyl ligands and enlightens the chirality transfer from chiral protecting ligands to superatomic states in metal clusters.

OBJECTIVE: The early symptoms of nasopharyngeal carcinoma (NPC) are not obvious, and it is difficult to make early diagnosis. A case-control study was conducted to identify potential biomarkers and established a diagnosis model for nasopharyngeal carcinoma. METHODS: Plasma samples of 131 cases of NPC and 132 cases of healthy individuals were incubated with the Ray Biotech Human Lung Cancer IgG Autoantibody Detection Array G1, and signal values were used to develop a risk prediction model for NPC diagnosis. RESULTS: Of the 30 autoantibodies, high expression of MAGE-A4, NY-ESO-1, HuD, Survivin, IMDH2, Ubiquilin-1, IMP1, PGP9.5, IMP3, C-Myc and low expression of Cyclin B1 were potential biomarkers for NPC diagnosis (p <0.05), among which Survivin, MAGE-A4 and IMP3 shows higher AUC of 0.674, 0.652 and 0.650 respectively, the specificity of them was 89.39% (95% CI: 82.85-94.08%), 90.15% (95% CI: 83.75-94.65%) and 88.64% (81.95-93.50%).The risk probability analysis for NPC diagnosis based on the panel of Cyclin B1, NY-ESO-1, Survivin, and IMP3 displayed the best diagnosis performance with an AUC of 0.779, p (Yi = 1) = 1/(1+EXP[8.316+1.672*CyclinB1-1.152*NY-ESO-1-2.052*Survivin-0.950*IMP3]), the specificity of that was 86.36% (95% CI: 79.31-91.71%). CONCLUSIONS: Our findings demonstrated that the panel of Cyclin B1, NY-ESO-1, Survivin, and IMP3 has a good performance in the detection of NPC, and all 11 autoantibodies may also have a certain significance for the prognosis of NPC.

In this study, a series of green, interference-free fluorimetric detection methods of the excitation-emission matrix coupled with the second-order calibration methods were proposed for the determination of ibrutinib and pralatrexate in various complicated biological fluids. The second-order advantage of the proposed method can overcome the problem of poor selectivity caused by the wide spectra of the fluorescence method. Even in the presence of uncalibrated interferences and severe peak overlap, the signal of pure substance and accurate quantitative results were still obtained. The average recoveries of the three methods were 94.5-104.9% for Alternating Trilinear Decomposition (ATLD) algorithm, 95.5-105.8% for Alternating Normalization Weighted Error (ANWE) algorithm and 94.4-105.7% for Parallel Factor Analysis (PARAFAC) algorithm, respectively. For ATLD, ANWE and PARAFAC, the relative standard deviations (RSD) were lower than 9.2%, 6.8% and 9.2%, and the RMSEPs were less than 8.1, 8.4 and 8.6 ng mL-1, respectively. In addition, the elliptic joint confidence region (EJCR) was adopted to further prove the accuracy of the three methods. The results showed that the three methods can accurately be quantified without significant difference. Good figures of merit parameters were also obtained. Among them, the limit of detection (LOD) and limit of quantification (LOQ) of ibrutinib and pralatrexate were in the range of 0.11-0.76 ng mL-1 and 0.21-1.12 ng mL-1, respectively, which were lower than the corresponding blood concentrations. These results indicate that the proposed method provides a promising, alternative and universal analysis strategy for clinical drug monitoring.

Atmospheric deposition of cadmium (Cd) and lead (Pb) was investigated together with the accumulation, distribution and health risks from potentially toxic metals in soils, vegetables and human hair at a mining area in southwest China. Annual atmospheric deposition of Cd and Pb were 41.1 and 192 g ha- 1, respectively, and consisted mainly of dry deposition. Agricultural soils experienced high levels of metal pollution around the mine, with 66.4% and 57.3 % of vegetable samples grown on these polluted fields exceeding maximum permissible Cd and Pb concentrations, particularly the leafy vegetables. Residents living near the mining area had high Cd (0.75 mg kg- 1) and Pb (6.87 mg kg- 1) concentrations in their hair, and the maximum values occurred in occupationally exposed individuals. Long-term mining activities have resulted in high health risks to the local population due to Cd and Pb deposition and accumulation from the atmosphere, soils and vegetables.

Japanese encephalitis virus (JEV) is a mosquito-borne flavivirus, which causes the most commonly diagnosed viral encephalitis named Japanese encephalitis (JE) in the world with an unclear pathogenesis. Axl, a receptor tyrosine kinase from TAM family, plays crucial role in many inflammatory diseases. We have previously discovered that Axl deficiency resulted in more severe body weight loss in mice during JEV infection, which we speculate is due to the anti-inflammatory effect of Axl during JE. Currently, the role of Axl in regulating the neuroinflammation and brain damage during JE has not been investigated yet. In this study, by using Axl deficient and heterozygous control mice, we discovered that Axl deficient mice displayed accelerated JE progression and exacerbated brain damage characterized by increased neural cell death, extended infiltration of inflammatory cells, and enhanced production of pro-inflammatory cytokines, in comparison to control mice. Additionally, consistent with our previous report, Axl deficiency had no impact on the infection and target cell tropism of JEV in brain. Taken together, our results suggest that Axl plays an anti-inflammatory and neuroprotective role during the pathogenesis of JE.

Quadcopters are widely used in a variety of military and civilian mission scenarios. Real-time online detection of the abnormal state of the quadcopter is vital to the safety of aircraft. Existing data-driven fault detection methods generally usually require numerous sensors to collect data. However, quadcopter airframe space is limited. A large number of sensors cannot be loaded, meaning that it is difficult to use additional sensors to capture fault signals for quadcopters. In this paper, without additional sensors, a Fault Detection and Identification (FDI) method for quadcopter blades based on airframe vibration signals is proposed using the airborne acceleration sensor. This method integrates multi-axis data information and effectively detects and identifies quadcopter blade faults through Long and Short-Term Memory (LSTM) network models. Through flight experiments, the quadcopter triaxial accelerometer data are collected for airframe vibration signals at first. Then, the wavelet packet decomposition method is employed to extract data features, and the standard deviations of the wavelet packet coefficients are employed to form the feature vector. Finally, the LSTM-based FDI model is constructed for quadcopter blade FDI. The results show that the method can effectively detect and identify quadcopter blade faults with a better FDI performance and a higher model accuracy compared with the Back Propagation (BP) neural network-based FDI model.

H2S is actual an endogenous signaling gas molecule and involved in a range of cell physiological processes. However, the mechanism of endogenous H2S regulating autophagy and apoptosis has not been thoroughly investigated. Here, we try to address this issue by using a H2S probe, (E)-2-(4-(4-(7-(diethylamino)-2-oxo-2H-chromene-3-carbonyl)-piperazin-1-yl)-styryl)-1, 3, 3-trimethyl-3H-indol-1-ium iodide (CPC), which could react with endogenous H2S. Herein, we reported that CPC inhibited autophagy and decreased the expression and activity of NF-E2-related factor 2 (Nrf2), then induced cell apoptosis. CPC inhibited autophagy and promoted apoptosis by inhibiting Nrf2 activation, which was H2S dependent. Furthermore, we found that CPC inhibited Nrf2 nucleus translocation by inhibiting glutathionylation of Kelch-like ECH-associated protein 1 (Keap1) at the Cys434 residue. CPC also inhibited various cancer cell growth, but had no effect on normal cell growth in vitro, and inhibited A549 cancer growth, but did not affect normal angiogenesis in vivo. Therefore, we not only found a new inhibitor of autophagy and Nrf2, but also suggested a novel mechanism that endogenous H2S could regulate autophagy, apoptosis and Nrf2 activity through regulating glutathionylation of Keap1 at the Cys434 residue.

Protein deglycase DJ-1 (DJ-1) is a multifunctional protein involved in various biological processes. However, it is unclear whether DJ-1 influences atherosclerosis development and plaque stability. Accordingly, we evaluated the influence of DJ-1 deletion on the progression of atherosclerosis and elucidate the underlying mechanisms. We examine the expression of DJ-1 in atherosclerotic plaques of human and mouse models which showed that DJ-1 expression was significantly decreased in human plaques compared with that in healthy vessels. Consistent with this, the DJ-1 levels were persistently reduced in atherosclerotic lesions of ApoE-/- mice with the increasing time fed by western diet. Furthermore, exposure of vascular smooth muscle cells (VSMCs) to oxidized low-density lipoprotein down-regulated DJ-1 in vitro. The canonical markers of plaque stability and VSMC phenotypes were evaluated in vivo and in vitro. DJ-1 deficiency in Apoe-/- mice promoted the progression of atherosclerosis and exaggerated plaque instability. Moreover, isolated VSMCs from Apoe-/- DJ-1-/- mice showed lower expression of contractile markers (α-smooth muscle actin and calponin) and higher expression of synthetic indicators (osteopontin, vimentin and tropoelastin) and Kruppel-like factor 4 (KLF4) by comparison with Apoe-/- DJ-1+/+ mice. Furthermore, genetic inhibition of KLF4 counteracted the adverse effects of DJ-1 deletion. Therefore, our results showed that DJ-1 deletion caused phenotype switching of VSMCs and exacerbated atherosclerotic plaque instability in a KLF4-dependent manner.

Fractionation of the ethanol extract of a marine fungus, Arthrinium sp., afforded a new pyridone alkaloid (arthpyrone L (1)), the structure with absolute configuration of which was established by comprehensive spectroscopic analyses. In vitro cell viability assays revealed that compound 1 showed antiproliferative effects toward human A549 (lung), MG63, U2OS (bone), MCF-7 and MDA-MB-231 (breast) cancer cells. MG63 cell lines were chosen for further biological evaluations and presented apoptosis and cell cycle arrest (G0/G1 phase) upon treatment of 1. Subsequent mechanism studies demonstrated that the growth inhibition of 1 against MG63 cells was via activation of caspase-modulated apoptotic pathway and inhibition of PI3K/Akt pathway.

Structured light with more controllable degrees-of-freedom (DoFs) is an exciting topic with versatile applications. In contrast to conventional vector vortex beams (VVBs) with two DoFs of orbital angular momentum (OAM) and polarization, a hybrid ray-wave structure was recently proposed [Optica 7(7), 820-831 (2020)], which simultaneously manifests multiple DoFs such as ray trajectory, coherent state phase, trajectory combination, besides OAM and polarization. Here we further generalize this exotic structure as the astigmatic hybrid VVB by hatching a new DoF of astigmatic degree. Importantly, the transverse topology varies with propagation, e.g. a linearly distributed hybrid trajectory pattern can topologically evolve to a circularly polygonal star shape, where the number of singularity changes from zero to multiple in a single beam. The propagation-dependent evolution can be easily controlled by the astigmatic degree, including as a vector vortex state such that different astigmatic trajectories have different polarizations. We experimentally generate such beams from a simple laser with a special astigmatic conversion by combined spherical and cylindrical lenses, and the results agree well with our theoretical simulation. With our new structured light, the propagation-multiplexing multi-DoF patterns can be controlled in a single beam, which can largely extend related applications such as high-dimensional large-capacity optical communication, laser machining, and particle trapping.

Individuals suffering from traumatic brain injury (TBI) often experience the activation of the immune system, resulting in declines in cognitive and neurological function after brain injury. Despite decades of efforts, approaches for clinically effective treatment are sparse. Evidence on the association between current therapeutic strategies and clinical outcomes after TBI is limited to poorly understood mechanisms. For decades, an increasing number of studies suggest that the gut-brain axis (GBA), a bidirectional communication system between the central nervous system (CNS) and the gastrointestinal tract, plays a critical role in systemic immune response following neurological diseases. In this review, we detail current knowledge of the immune pathologies of GBA after TBI. These processes may provide a new therapeutic target and rehabilitation strategy developed and used in clinical treatment of TBI patients.

Due to the rapid growth of electronic information technology, the need for the higher stability of crystal oscillators has increased. The temperature-compensated X'tal (crystal) oscillator (TCXO), a type of crystal oscillator with high frequency stability, has been widely used in communications, sensor networks, automotive electronics, industrial control, measuring devices, and other equipment. The traditional TCXO only performs frequency compensation based on the current temperature, without considering the error caused by thermal hysteresis. As the frequency stability of the TCXO improves, the thermal hysteresis of the crystal oscillator has a negligible influence on the frequency stability of the crystal oscillator. This study measured different compensation tables for hysteresis curves at different temperatures and used a microprocessor to store the historical information of crystal temperature changes. Furthermore, corresponding algorithms were designed to select the correct values, according to the temperature change history, to compensate for the thermal hysteresis of the crystal oscillator error. Experiments show that this method can reduce the hysteresis error of the crystal oscillator from 700 to 150 ppb (-40 to 80 °C).

In this work, kaolin/steel slag particle electrodes (KSPEs) were synthesized using a calcination method, and they were used to degrade norfloxacin (NOR) wastewater in three-dimensional (3D) reactor. Characterization methods used by KSPEs included SEM, XRF, XRD and BET. The effects of cell voltage, initial pH, KSPEs dosage and initial NOR concentration on NOR degradation were studied in the optimization experiment of operating parameters. The NOR degradation rate and COD removal rate can reach 96.02% and 93.45% under the optimal parameters within 30 min, and energy consumption is 0.99 kWh m-3. As a result, KSPEs shows excellent catalytic performance and cycling, and still has high electrocatalytic activity after 10 cycles. Finally, the degradation mechanism and degradation pathways of KSPEs to treat NOR are proposed.

Despite significant progress achieved in the preparation of chiral nanoparticles, the enantioseparation of racemates still presents a big challenge in nanomaterial research. Herein, we report the synthesis and structural characterization of racemic anisotropic nanocluster Ag30(C2B10H9S3)8Dppm6 (Ag 30 -rac), which is protected by mixed carboranetrithiolate and phosphine ligands. Spontaneous self-resolution of the racemates was realized through conglomerate crystallization in dimethylacetamide (DMAc). The homochiral nanoclusters in the racemic conglomerates adopt enantiomeric helical self-assemblies (R/L-Ag 30 ). Diverse noncovalent interactions as the driving force in directing superstructure packing were elucidated in detail. Furthermore, the nanoclusters show red luminescence in both solid and solution states, and the racemic conglomerates display a mirror-image CPL response. This work provides atom-precise helical nanoparticle superstructures that facilitate an in-depth understanding of the helical-assembly mechanism.

A series of (E)-N-2(5H)-furanonyl sulfonyl hydrazone derivatives have been rationally designed and efficiently synthesized by one-pot reaction with good yields for the first time. This green approach with wide substrate range and good selectivity can be achieved at room temperature in a short time in the presence of metal-free catalyst. The cytotoxic activities against three human cancer cell lines of all newly obtained compounds have been evaluated by MTT assay. Among them, compound 5 k exhibits high cytotoxic activity against MCF-7 human breast cancer cells with an IC50 value of 14.35 µM. The cytotoxic mechanism may involve G2/M phase arrest pathway, which is probably caused by activating DNA damage. Comet test and immunofluorescence results show that compound 5 k can induce DNA damage in time- and dose-dependent manner. Importantly, 5 k also can effectively inhibit the proliferation of MCF-7 cells and angiogenesis in the zebrafish xenograft model. It is potential to further develop N-2(5H)-furanonyl sulfonyl hydrazone derivatives as potent drugs for breast cancer treatment with higher cytotoxic activity by modifying the structure of the compound.

Structure evolution has become an effective way to assemble novel monolayer-protected metal nanomolecules. However, evolution with alkynyl-stabilized metal clusters still remains rarely explored. Herein, we present a carboranealkynyl-protected gold nanocluster [Au28 (C4 B10 H11 )12 (tht)8 ]3+ (Au28 , tht=tetrahydrothiophene) possessing an open-shell electronic structure with 13 free electrons, which was isolated by a facile self-reduction method with 9-HC≡C-closo-1,2-C2 B10 H11 as the two-in-one reducing and protecting agent. Notably, Au28 undergoes a complete transformation in methanol into a stable and smaller-sized nanocluster [Au23 (C4 B10 H11 )9 (tht)6 ]2+ (Au23 ) bearing 12 valence electrons and crystal-field-like split superatomic 1D orbitals. The transformation process was systematically monitored with ESI-MS and UV/Vis absorption spectra. Au28 and Au23 both display optical absorption covering the UV/Vis-NIR range and NIR emission, which facilitates their potential application in the biomedical and photocatalytic fields.

Dengue virus (DV) is the most rapidly spreading arbovirus in the world. Our previous studies indicated that Rac1, a kind of Rho GTPase, was related with the increased vascular permeability in DV infection. However, the molecular mechanisms that regulate the activity of the Rac1 pathway during DV infection is not fully understood yet. Recently, Rho-specific guanine nucleotide dissociated inhibitors (Rho GDIs), as a pivotal upstream regulator of Rho GTPase, attract our attention. To identify the role of GDI-1 in DV2 infection, the expression of GDI in Eahy926 cells was detected. Moreover, a GDI-1 down-regulated cell line was constructed to explore the correlation between GDI-1 and Rac1 and to further evaluate the function of GDI in DV life cycle. Our results indicated that DV2 infection could up-regulate GDI-1 expression, and down-regulation of GDI enhanced the activity of Rac1. In addition, down-regulated GDI-1 significantly inhibited all steps of DV2 replication cycle. GDI-1 plays an important role in DV2 infection via negatively regulating the activation of the Rac1-actin pathway. These results not only contribute to our further understanding of the pathogenesis of severe dengue but also provide further insight into the development of antiviral drugs.

BACKGROUND: Hypopigmented mycosis fungoides (HMF) is an uncommon variant of mycosis fungoides. AIMS: To study the clinical and histopathology presentation in children with HMF. METHOD: We reviewed 9 children diagnosed with HMF. The clinical data were collected and analyzed. RESULT: Eight boys and 1 girl were included, with a median onset age of 7.4 year old and median age of diagnosis of 10.5 year old. Multiple hypopigmented patches were observed in all patients, and 5 patients exhibited multiple scaly erythema at the center of hypopigmented patches. Histopathology showed atypical lymphocytes with hyperchromatic, irregular, and cerebriform nuclei, infiltrated in the epidermis and dermis. Pautrier's microabscesses was noted in 6 of 9 patients, and papillary dermal fibroplasia was noted in 6 of 9 patients. CD8 predominance was detected in 4 of 6 patients. Four patients were simultaneously subjected to skin biopsy on hypopigmented patches and scaly erythema simultaneously. Compared with hypopigmented specimens, erythema biopsy detected deeper and denser infiltration of atypical lymphoid cells in 3 of 4 patients, higher CD4/CD8 ratio in 4 of 4 patients, more CD5 loss in 2 of 4 patients, and more CD7 loss in 2 of 4 patients. TCR gene monoclonal rearrangement was detected in 2 of 5 patients. Narrowband ultraviolet B phototherapy was applied in 7 patients. One of 7 patients achieved complete response, and 6 of 7 patients achieved partial response. No recurrence was noted with the median follow-up period of 6 months. CONCLUSION: HMF could occur in young patients, with indolent and benign course. HMF could gradually seem as scaly erythema based on hypopigmented patches. The histopathology indicated a more advanced stage of the scaly erythema lesions than hypopigmented patches.

AIM: Our previous study has shown that leucine-rich repeat containing GPCR-4 (LGR4, or GPR48) LGR4 plays a role in cell migration, invasion, proliferation and apoptosis of prostate cancer (PCa). In this study, we aimed to explore whether LGR4 would affect radiation response in PCa. MATERIALS AND METHODS: LGR4 expression was silenced by shRNA transfection. qRT-PCR was employed to determine mRNA expression of LGR4 and DNA damage repair genes. Western blot was used to evaluate protein expression of LGR4, RSPO1-4, androgen receptor (AR), cyclic AMP response-element binding protein (CREB1), γH2A.X, and H2A.X. Cell proliferation was detected by CCK-8 assay and apoptosis was assayed by flow cytometry. Additionally, a xenograft model was also established to validate the role of LGR4 in PCa cells after radiation. KEY FINDINGS: LGR4 expression was enhanced in PCa cells by radiation treatment in dose- and time-dependent means. RSPO1-4 were also upregulated post-radiation. Furthermore, LGR4 knockdown exacerbated apoptosis, reduced cell viabilities and strengthened nuclear γH2A.X staining in AR positive PCa cells but not in AR negative cells in the presence of radiation. Likewise, LGR4 ablation diminished AR and CREB1 expression induced by radiation. In contrast, RSPO1 stimulation augmented cell viabilities, promoted AR and CREB1 expression, and upregulated DNA repair gene expression, which could be reversed by enzalutamide, except for AR expression. Additionally, LGR4 knockdown further suppressed tumor growth and AR/CREB1 expression but enhanced γH2A.X expression in xenografts. SIGNIFICANCE: In all, our study suggested that LGR4 might serve as an important regulator of radiation sensitivity in PCa.
