RESUMO

Different fertilization regimes can substantially influence soil fungal community composition, yet fewer studies try to control for the effects of nitrogen input. Here, we investigated the impact of fertilization with equal nitrogen upon soil properties and soil fungal diversity and community composition in the North China Plain in a long-term field experiment. Long-term (32 years) fertilization regimes were applied with equal amounts of nitrogen: no chemical fertilizer or organic manure; chemical fertilization only; organic manure fertilization only, and; combination of 1/2 chemical fertilizer and 1/2 organic manure. Then we investigated the influence of these four fertilization regimes to soil properties, fungal diversity and community composition. The results showed that applying organic manure significantly influenced soil properties. Illumina MiSeq sequencing and its analysis revealed that organic manure fertilization significantly changed soil fungal alpha diversity, but chemical fertilization did not. Although soil fungal community composition did not differ significantly among all the fertilization regimes at the phylum and class levels, they did show differences in the abundance of dominant fungi. Yet at the genus level, soil fungal community composition, abundance, and beta diversity was affected by all fertilization regimes. Application of organic manure also reduced the abundance of soil-born fungal pathogens such as Fusarium. Our results suggest that long-term application of organic manure could markedly improve soil properties, altering soil fungal community composition and its diversity. Moreover, organic manure fertilization could limit soil-born fungal diseases, to further contribute to soil ecosystem sustainability.

Right atrial appendage aneurysms (RAAAs) are rare heart malformations, presenting as isolated anomalies or co-existing with other structural heart diseases. We describe a rare case of RAAA complicated with an atrial septal defect (ASD). The diagnosis was established using transthoracic echocardiography and confirmed using cardiac magnetic resonance imaging. To treat the ASD and reduced right atrium volume load, ASD transcatheter closure was performed. On echocardiography performed 3 months post discharge, the RAAA was observed to have reduced in size compared to that presurgery. Six years later, she was in good condition without any adverse events.

X-linked Alport syndrome (XLAS) is a rare form of hereditary nephritis caused by mutations in the COL4A5 gene encoding the type IV collagen α5 chain. A skin biopsy was performed on one female patient with XLAS who carried a heterozygous p.G409S (c. 1225 G > A) mutation in the COL4A5 gene. A human-induced pluripotent stem cell (iPSC) line was generated from dermal fibroblasts using the integrating free Sendai virus technique. The generated iPSC line SHCDNRi001-A offers an efficient resource to research pathogenic mechanisms in XLAS, as well as a cell-based disease model for drug testing or other treatments.

A new andirobin-type limonoid with modified furan ring, khaysenelide K (1), together with a known analogue (2), was isolated from the stem barks of Khaya senegalensis. The structure and absolute configuration of 1 were elucidated by a combination of 1D and 2D NMR, HRESIMS, and single-crystal X-ray diffraction using mirror Cu-Kα radiation. Compound 1 showed moderate NO inhibitory activity in LPS-activated RAW 264.7 macrophages with IC50 value of 27.74 ± 0.68 µM.[Formula: see text].

BACKGROUND: The incidence of dry eye is high after a cataract surgery. However, the clinical efficacy of sodium hyaluronate combined with conventional eye drops for dry eye after cataract surgery remains unclear. The currently available studies were based on small sample sizes, and no systematic review on this topic has been conducted. Thus, a meta-analysis was performed to systematically and accurately evaluate the clinical value of sodium hyaluronate in the treatment of dry eye after cataract surgery. METHODS: English-language databases including PubMed, Cochrane, and Web of Science, and Chineselanguage databases including (CNKI), Wanfang Database, and CQVIP were systematically searched for relevant articles published up to December 31, 2019. The pooled effect size of qualitative data was evaluated with relative risk (RR) and 95% confidence interval (CI), and the overall effect of the quantitative data was estimated by using weighted mean difference (WMD) or standard mean difference (SMD) and 95% CI. Randomized controlled clinical trials concerning the use of sodium hyaluronate combined with conventional treatment for dry eye after cataract surgery were included. The quality of the included literature was evaluated by RevMan5.3 software, and the rest of the statistical data were analyzed using the "meta" package of the R3.5.1 software. RESULTS: A total of 24 articles including 2,177 eyes (1,088 eyes in the combination group and 1089 eyes in the control group) were included. Twelve articles reported the total effective rate after 1 month of treatment (I2 =71%), and the random effects model was used to estimate RR. The total effective rate in the combination group was 1.33 times that of the control group (95% CI: 1.21, 1.47). Ten articles reported the dry eye symptom score after 1 month of treatment (I2 =92%). The SMD was estimated using the random effects model. The dry eye symptom score in the combination group was significantly lower than that in the control group, with an SMD of -2.98 (95% CI: -3.69, -2.27). Seventeen articles reported the tear film break-up time (BUT) after 1 month of treatment. As shown in the random effects model, BUT was significantly longer in the combination group than in the control group, with an MD of 2.06 (95% CI: 1.63, 2.49). Sixteen articles described the scores of a fluorescein (FL) eye stain test after 1 month of treatment. As shown in the random effects model, FL score was significantly lower in the combination group than in the control group, with an SMD of -2.52 (95% CI: -3.23, -1.81). Eight articles reported the results of Schirmer I test (SIt) after 1 month of treatment. As estimated by the random effects model, the wetting length in SIt was significantly higher in the combination group than in the control group, with an MD of 1.50 (95% CI: 0.53, 2.48). In the elderly population, BUT was significantly higher in the combined group than in the control group, with an MD of 2.97 (95%CI: 2.47, 3.47); however, the FL score and SIt showed no such significant difference. CONCLUSIONS: For the patients with dry eye after cataract surgery, sodium hyaluronate on the basis of conventional anti-inflammatory treatment is highly effective as it can improve the dry eye symptom score and the results of 3 tests for dry eye. However, high-quality studies are warranted to further investigate the safety of sodium hyaluronate.

Half-Heusler alloys possess unique and desirable physical properties due to their thermoelectricity, magnetism, superconductivity, and weak antilocalization effects. These properties have become of particular interest since the recent discovery of topological Weyl semimetal state for which the electronic bands are dispersed linearly around one pair of Weyl nodes, with opposite chirality (i.e., chiral anomaly). Here, we report the transport signatures of topological electronic state in a half-Heusler GdPtBi single crystal. We show that the non-trivial  Berry phase, negative magnetoresistance and giant planner Hall effect arise from the chiral anomaly and that the Shubnikov-de Haas (SdH) oscillation frequency in GdPtBi is angle-dependent with an anisotropic Fermi surface (FS). All transport signatures not only demonstrate the topological electronic state in half-Heusler GdPtBi crystals, but also describe the shape of the anisotropy FS.

BACKGROUND: Relaxin is an endogenous protein that has been shown to have antifibrotic properties in various organ systems. There has been no characterization of relaxin's role in the human bladder. Our objective was to characterize relaxin receptor expression in the human bladder and assess relaxin's effect on tissue remodeling/fibrosis pathways in bladder smooth muscle cells. METHODS: Relaxin family peptide receptor 1 (RXFP1) and RXFP2 expression was assessed using quantitative reverse transcriptase-PCR (qRT-PCR) and immunohistochemistry (IHC) on primary bladder tissue. Primary human smooth muscle bladder cells were cultured and stimulated with various concentrations of relaxin. Western blot, qRTPCR, ELISA, and zymogram assays were used to analyze fibrosis/tissue remodeling pathway proteins. RESULTS: There was universal mRNA transcript detection and protein expression of relaxin receptors in primary bladder specimens. Immunohistochemistry demonstrated RXFP1 and RXFP2 localizing to both urothelial and smooth muscle cell layers of the bladder. 24 h of in vitro relaxin stimulation did not affect mRNA expression of selected proteins in human bladder smooth muscle cells. However, 48 h of in vitro relaxin stimulation resulted in upregulation of active (p = 0.004) and latent (p = 0.027) MMP-2 in cell lysate, and upregulation of active MMP-2 in supernatant (p = 0.04). There was a dose dependent relationship with increasing expression of MMP-2 with increasing relaxin concentration. Relaxin stimulation resulted in decreased levels of active and total TGF-ß1 in supernatant and extracellular matrix (p < 0.005 with 100 ng/mL relaxin stimulation). CONCLUSIONS: In the human bladder, relaxin receptors are expressed at the dome and trigone and localize to the urothelium and smooth muscle cell layers. Stimulation of human bladder SMCs with relaxin in vitro affects expression of MMP-2 and TGF-ß1.

Human microbiota play an important role in the health of their human hosts. Recent studies have demonstrated that microbiota exist in seminal plasma. The current study aims to elucidate whether seminal microbiota exist in patients with different types of dysspermatism and whether bacterial biomarkers can be identified for them. A total of 159 study participants were recruited, including 22 patients with oligoasthenospermia, 58 patients with asthenospermia, 8 patients with azoospermia, 13 patients with oligospermia, and 58 matched healthy controls. Seminal microbiota composition was analyzed using 16S rRNA gene-based sequencing. The results showed that the composition of seminal microbiota of patients with dysspermatism differed from those of healthy controls. Comparison of the microbiota composition in semen samples from patients with different types of dysspermatism showed that microbiota in patients with asthenospermia and oligoasthenospermia were distinct from healthy controls in beta diversity (P < 0.05). Characteristic biomarkers, including Ureaplasma, Bacteroides, Anaerococcus, Finegoldia, Lactobacillus and Acinetobacter lwoffii, were identified based on LEfSe analysis. Inferred functional analysis based on seminal microbiome data further indicated the presence of potential pathogenic biomarkers in patients with asthenospermia and oligoasthenospermia. These results provided profiles of seminal microbiota exhibited in different types of dysspermatism, thus providing new insights into their pathogenesis.

Three previously undescribed dammarane triterpenoid glycosides (1-3) along with five known analogues (4-8) were isolated from the leaves of Cyclocarya paliurus. Their structures and configurations were determined on the basis of comprehensive spectroscopic analyses, chemical hydrolysis and DFT GIAO 13C NMR calculation. All the isolates were evaluated cytotoxic activities against seven human cancer cell lines (MCF-7, PC-3, Du145, NCI-H1975, PC-9, SKVO3 and HepG2). Moreover, compound 4 showed a wide spectrum of cytotoxicity against human cancer cells with IC50 values ranging from 11.31 to 29.51 µM.

Artificial light at night has greatly changed the physical environment for many organisms on a global scale. As an energy efficient light resource, light emitting diodes (LEDs) have been widely used in recent years. As LEDs often have a broad spectrum, many biological processes may be potentially affected. In this study, we conducted manipulated experiments in rat-proof enclosures to explore the effects of LED night lighting on behavior of a nocturnal rodent, the Mongolian five-toed jerboa (Allactaga sibirica). We adopted the giving-up density (GUD) method and camera video trapping to study behavioral responses in terms of patch use, searching efficiency and vigilance. With the presence of white LED lighting, jerboas spent less time in patches, foraged less intensively (with higher GUDs) and became vigilant more frequently, while their searching efficiency was higher than under dark treatment. Although both positive and negative effects of LEDs on foraging were detected, the net effect of LEDs on jerboas is negative, which may further translate into changes in population dynamics, inter-specific interaction and community structure. To our knowledge, this is the first field study to explore how LED lighting affect foraging behavior and searching efficiency in rodents. Our results may have potential implications for practices such as pest control.

Psychiatric disorders are a group of complex psychological syndromes whose etiology remains unknown. Previous study suggested that various chemicals contributed to the development of psychiatric diseases through affecting gene expression. This study aims to systematically explore the potential relationships between 5 major psychiatric disorders and more than 11 000 chemicals. The genome-wide association studies (GWAS) datasets of attention deficiency/hyperactive disorder (ADHD), autism spectrum disorder (ASD), bipolar disorder (BD), major depression disorder (MDD), and schizophrenia (SCZ) were driven from the Psychiatric GWAS Consortium and iPSYCH website. The chemicals related gene sets were obtained from the comparative toxicogenomics database (CTD). First, transcriptome-wide association studies (TWAS) were performed by FUSION to calculate the expression association testing statistics utilizing GWAS summary statistics of the 5 common psychiatric disorders. Chemical-related gene set enrichment analysis (GSEA) was then conducted to explore the relationships between chemicals and each of the psychiatric diseases. We observed several significant correlations between chemicals and each of the psychiatric disorders. We also detected common chemicals between every 4 of the 5 major psychiatric disorders, such as androgen antagonists for ADHD (P value = .0098), ASD (P value = .0330), BD (P value = .0238), and SCZ (P value = .0062), and imipramine for ADHD (P value = .0054), ASD (P value = .0386), MDD (P value = .0438), and SCZ (P value = .0008). Our study results provide new clues for revealing the roles of environmental chemicals in the development of psychiatric disorders.

BACKGROUND: Low-dose computed tomography screening has been proved to reduce lung cancer mortality, however, the issues of high false-positive rate and overdiagnosis remain unsolved. Risk prediction models for lung cancer that could accurately identify high-risk populations may help to increase efficiency. We thus sought to develop a risk prediction model for lung cancer incorporating epidemiological and metabolic markers in a Chinese population. METHODS: During 2006 and 2015, a total of 122 497 people were observed prospectively for lung cancer incidence with the total person-years of 976 663. Stepwise multivariable-adjusted logistic regressions with Pentry  = .15 and Pstay  = .20 were conducted to select the candidate variables including demographics and metabolic markers such as high-sensitivity C-reactive protein (hsCRP) and low-density lipoprotein cholesterol (LDL-C) into the prediction model. We used the C-statistic to evaluate discrimination, and Hosmer-Lemeshow tests for calibration. Tenfold cross-validation was conducted for internal validation to assess the model's stability. RESULTS: A total of 984 lung cancer cases were identified during the follow-up. The epidemiological model including age, gender, smoking status, alcohol intake status, coal dust exposure status, and body mass index generated a C-statistic of 0.731. The full model additionally included hsCRP and LDL-C showed significantly better discrimination (C-statistic = 0.735, P = .033). In stratified analysis, the full model showed better predictive power in terms of C-statistic in younger participants (<50 years, 0.709), females (0.726), and former or current smokers (0.742). The model calibrated well across the deciles of predicted risk in both the overall population (PHL  = .689) and all subgroups. CONCLUSIONS: We developed and internally validated an easy-to-use risk prediction model for lung cancer among the Chinese population that could provide guidance for screening and surveillance.

Precartilaginous stem cells (PCSCs) are adult stem cells that can initiate chondrocytes and bone development. In the present study, we explored whether miR-132/212 was involved in the proliferation of PCSCs via Hedgehog signaling pathway. PCSCs were isolated and purified with the fibroblast growth factor receptor-3 (FGFR-3) antibody. Cell viability, DNA synthesis and apoptosis were measured using MTT, BrdU and flow cytometric analysis. The mRNA and protein expression were detected by real-time PCR and Western blot, respectively. The target gene for miR-132/212 was validated by luciferase reporter assay. Results showed that transfection with miR-132/212 mimic significantly increased cell viability and DNA synthesis, and inhibited apoptosis of PCSCs. By contrast, miR-132/212 inhibitor could suppress growth and promote apoptosis of PCSCs. Luciferase reporter assays indicated that transfection of miR-132/212 led to a marked reduction of luciferase activity, but had no effect on PTCH1 3'-UTR mutated fragment, suggesting that Patched1 (PTCH1) is a target of miR-132/212. Furthermore, treatment with miR-132/212 mimics obviously increased the protein expression of Indian hedgehog (Ihh) and parathyroid hormone related protein (PTHrP), which was decreased after treatment with Hedgehog signaling inhibitor, cyclopamine. We also found that inhibition of Ihh/PTHrP signaling by cyclopamine significantly suppressed growth and DNA synthesis, and induced apoptosis in PCSCs. These findings demonstrate that miR-132/212 promotes growth and inhibits apoptosis in PCSCs by regulating PTCH1-mediated Ihh/PTHrP pathway, suggesting that miR-132/212 cluster might serve as a novel target for bone diseases.

In this study, a series of new indole derivatives of ursolic acid bearing different N-(aminoalkyl)carboxamide side chains were designed, synthesized, and evaluated for their in vitro cytotoxic activities against two human hepatocarcinoma cell lines (SMMC-7721 and HepG2) and normal hepatocyte cell line (LO2) via MTT assay. Among them, compound 5f exhibited the most potent activity against SMMC-7721 and HepG2 cells with IC50 values of 0.56 ± 0.08 µM and 0.91 ± 0.13 µM, respectively, and substantially lower cytotoxicity to LO2 cells. A follow-up enzyme inhibition assay and molecular docking study indicated that compound 5f can significantly inhibit the activity of Topoisomerase IIα. Further mechanistic studies performed in SMMC-7721 cells revealed that compound 5f can elevate the intracellular ROS levels, decrease mitochondrial membrane potential, and finally lead to the apoptosis of SMMC-7721 cells. Collectively, compound 5f is a promising Topoisomerase II (Topo II) inhibitor, which exhibited the potential as a lead compound for the discovery of novel anticancer agents.

Vanadium (V) pollution in soil has been widely noted, while knowledge about the effect of soil colloid on migration of V is scarce. Batch adsorption-desorption and transportation of the colloid-adsorbed V in columns packed with quartz sand under various environment conditions were carried out to explore the retention and transportation of V by colloidal kaolinite. Batch adsorption-desorption studies show that the adsorption of V by the colloidal kaolinite was mainly specific adsorption and redox played a limited role in the adsorption process. The maximum adsorption capacity of the colloidal kaolinite was 712.4 mg g-1, and about 5.9-8.7% of the adsorbed V could be desorbed. Both the adsorption-desorption and migration of V with colloidal kaolinite were highly ambient condition dependent. The column studies show that V was highly mobile in the saturated porous media. An easier transfer of V with an increase in pH, IS, and velocity of flow was noted. However, the increase of IS lead to the blockage of the colloidal kaolinite transportation. The recovery rate of the colloidal kaolinite at pH 7 and 9 was 2.0 and 2.1 times that at pH 5, respectively. The migration of colloidal-adsorbed V in sand column preceded that of V ion, but more colloidal-bound V than V ion remained in the column. Lack of consideration of the combination and co-transportation of V and colloidal kaolinite will lead to an overestimation of the risk of V to deeper soil profiles and groundwater. STUDY OBJECTIVES: Insomnia is a common sleep disorder and constitutes a major issue in modern society. We provide new clues for revealing the association between environmental chemicals and insomnia. METHODS: Three genome-wide association studies (GWAS) summary datasets of insomnia (n = 113006, n = 1331010 and n = 453379, respectively) were driven from the UK Biobank, 23andMe and deCODE. The chemical-gene interaction dataset was downloaded from the comparative toxicogenomics database (CTD). First, we conducted a meta-analysis of the three datasets of insomnia using the METAL software. Using the result of meta-analysis, transcriptome-wide association studies (TWAS) were performed to calculate the expression association testing statistics of insomnia. Then chemical related gene set enrichment analysis (GSEA) was used to explore the association between chemicals and insomnia. RESULTS: For GWAS meta-analysis dataset of insomnia, we identified 42 chemicals associated with insomnia in brain tissue (P-value <0.05) by GSEA. We detected five important chemicals such as Pinosylvin (P = 0.0128), Bromobenzene (P = 0.0134), Clonidine (P = 0.0372), Gabapentin (P = 0.0372) and Melatonin (P = 0.0404) which are directly associated with insomnia. CONCLUSION: Our study results provide new clues for revealing the roles of environmental chemicals in the development of insomnia.

PURPOSE: To develop a nonlinear preconditioned total field-inversion algorithm using the MEDI toolbox (MEDInpt) for robust QSM of carotid plaques and evaluate its performance in comparison with a local field-inversion algorithm (STI Suite) previously applied to carotid QSM. METHODS: Numerical simulation and in vivo carotid QSM were performed to compare the MEDInpt and STI Suite algorithms. Multicontrast MRI was used as the reference standard for detecting calcified plaque and intraplaque hemorrhage (IPH). A total of 5 healthy volunteers and 11 patients with at least one significant carotid artery stenosis were enrolled in this study. RESULTS: In the numerical carotid phantom, the relative susceptibility errors for calcified plaque and IPH were reduced from -63.2% and -56.5% with STI Suite to -13.0% and -24.2% with MEDInpt, respectively. In humans, MEDInpt provided a higher QSM quality score and better detection of calcification and IPH than STI Suite. Although all calcifications and IPHs detected on multicontrast MRI could be seen on QSM obtained with MEDInpt, only 50% of calcified plaques and 83% of IPHs could be captured on QSM obtained with STI Suite. CONCLUSION: MEDInpt can resolve calcification and IPH in advanced atherosclerotic carotid plaques. Compared with STI Suite, MEDInpt provided better QSM quality and has the potential to improve the detection of these plaque components.

Tuning the magnetic anisotropy of metal ions remains highly interesting in the design of improved single-molecule magnets (SMMs). We herein report synthetic, structural, magnetic, and computational studies of four mononuclear CoII complexes, namely [Co(hfac)2 (MeCN)2 ] (1), [Co(hfac)2 (Spy)2 ] (2), [Co(hfac)2 (MBIm)2 ] (3), and [Co(hfac)2 (DMF)2 ] (4) (MeCN=acetonitrile, hfac=hexafluoroacetylacetone, Spy=4-styrylpyridine, MbIm=5,6-dimethylbenzimidazole, DMF=N,N-dimethylformamide), with distorted octahedral geometry constructed from hexafluoroacetylacetone (hfac) and various axial ligands. By a building block approach, complexes 2-4 were synthesized by recrystallization of the starting material of 1 from various ligands containing solution. Magnetic and theoretical studies reveal that 1-4 possess large positive D values and relative small E parameters, indicating easy-plane magnetic anisotropy with significant rhombic anisotropy in 1-4. Dynamic alternative current (ac) magnetic susceptibility measurements indicate that these complexes exhibit slow magnetic relaxation under external fields, suggesting field-induced single-ion magnets (SIMs) of 1-4. These results provide a promising platform to achieve fine tuning of magnetic anisotropy through varying the axial ligands based on Co(II) bis(hexafluoroacetylacetonate) complexes.

RESUMO

AIMS: Insomnia, intelligence and neuroticism are three typical traits and dysfunctions mainly regulated by human brain. Our research aimed to explore the potential genetic relationships between brain function related traits and more than 3000 human plasma proteins. MATERIALS AND METHODS: We conducted a large-scale genetic correlation scan of human plasma proteins and three brain function related traits, including insomnia, intelligence and neuroticism. Linkage disequilibrium score regression (LDSC) analysis was performed to estimate the genetic correlations between each of the blood proteins and insomnia, intelligence and neuroticism via utilizing the genome-wide association study summary statistics of plasma proteins and those three traits. RESULTS: LDSC analysis identified 18 specific plasma proteins shown suggestive genetic correlations with insomnia such as Periostin (coefficient=-0.3910, P value = 0.0070). Twenty-one plasma proteins exhibited genetic correlations with intelligence such as Ecto-ADP-ribosyltransferase 3 (coefficient = 0.3066, P value = 0.0013). Six specific plasma proteins shown suggestive genetic correlations with neuroticism, such as CD70 antigen (coefficient = 0.2979, P value = 0.0134). After further comparing the suggestive proteins between insomnia, intelligence and neuroticism, we detected 3 common plasma proteins shared by insomnia and intelligence such as Periostin (coefficient insomnia =-0.3910, Pinsomnia value = 0.0070; coefficient intelligence =0.2673, Pintelligence value = 0.0159) and Neurexin-1 (coefficient insomnia =-0.2913, Pinsomnia value = 0.0197; coefficient intelligence = 0.2399, Pintelligence value = 0.0035). We also detected 2 common plasma proteins shared by intelligence and neuroticism, including CD70 antigen (coefficient intelligence =-0.2092, Pintelligence value = 0.0337; coefficient neuroticism = 0.2979, Pneuroticism value = 0.0134). CONCLUSION: Our results provide novel clues for unveiling the functional relevance of plasma proteins and brain function related traits.
